Abstract:Cells are highly advanced microreactors that form the basis of all life. Their fascinating complexity has inspired scientists to create analogs from synthetic and natural components using a bottom-up approach. The ultimate goal here is to assemble a fully man-made cell that displays functionality and adaptivity as advanced as that found in nature, which will not only provide insight into the fundamental processes in natural cells but also pave the way for new applications of such artificial cells. In this Account, we highlight our recent work and that of others on the construction of artificial cells. First, we will introduce the key features that characterize a living system; next, we will discuss how these have been imitated in artificial cells. First, compartmentalization is crucial to separate the inner chemical milieu from the external environment. Current state-of-the-art artificial cells comprise subcompartments to mimic the hierarchical architecture of eukaryotic cells and tissue. Furthermore, synthetic gene circuits have been used to encode genetic information that creates complex behavior like pulses or feedback. Additionally, artificial cells have to reproduce to maintain a population. Controlled growth and fission of synthetic compartments have been demonstrated, but the extensive regulation of cell division in nature is still unmatched. Here, we also point out important challenges the field needs to overcome to realize its full potential. As artificial cells integrate increasing orders of functionality, maintaining a supporting metabolism that can regenerate key metabolites becomes crucial. Furthermore, life does not operate in isolation. Natural cells constantly sense their environment, exchange (chemical) signals, and can move toward a chemoattractant. Here, we specifically explore recent efforts to reproduce such adaptivity in artificial cells. For instance, synthetic compartments have been produced that can recruit proteins to the membrane upon an external stimulus or modulate their membrane composition and permeability to control their interaction with the environment. A next step would be the communication of artificial cells with either bacteria or another artificial cell. Indeed, examples of such primitive chemical signaling are presented. Finally, motility is important for many organisms and has, therefore, also been pursued in synthetic systems. Synthetic compartments that were designed to move in a directed, controlled manner have been assembled, and directed movement toward a chemical attractant is among one of the most life-like directions currently under research. Although the bottom-up construction of an artificial cell that can be truly considered "alive" is still an ambitious goal, the recent work discussed in this Account shows that this is an active field with contributions from diverse disciplines like materials chemistry and biochemistry. Notably, research during the past decade has already provided valuable insights into complex synthetic systems with life-like properties. In the future, artificial cells are thought to contribute to an increased understanding of processes in natural cells and provide opportunities to create smart, autonomous, cell-like materials.

Abstract:Artificial cells have generated much interest since the concept was introduced by Aleksandr Oparin in the 1920s, and they have had an impact on the pharmaceutical and biotechnology industry in various areas, including potential therapeutic applications. Here, we discuss the development of small-scale, bio-inspired artificial cell components that recreate the function of key cellular and physiological systems. We describe artificial cells, selected current applications and how small-scale biology could be used to provide what might be a next-generation approach in this area. We believe that this type of work is in its infancy and that exploiting small-scale biological inspiration in the field of artificial cells has great potential for successes in the future.

Abstract:Building an artificial cell is a research area that is rigorously studied in the field of synthetic biology. It has brought about much attention with the aim of ultimately constructing a natural cell-like structure. In particular, with the more mature cell-free platforms and various compartmentalization methods becoming available, achieving this aim seems not far away. In this review, we discuss the -various types of artificial cells capable of hosting several cellular functions. Different compartmental boundaries and the mature and evolving technologies that are used for compartmentalization are examined, and exciting recent advances that overcome or have the potential to address current challenges are discussed. Ultimately, we show how compartmentalization and cell-free systems have, and will, come together to fulfill the goal to assemble a fully synthetic cell that displays functionality and complexity as advanced as that in nature. The development of such artificial cell systems will offer insight into the fundamental study of evolutionary biology and the sea of applications as a result Although several challenges remain, emerging technologies such as artificial intelligence also appear to help pave the way to address them and achieve the ultimate goal.

Abstract:Artificial cells are developed to redesign novel biological functions in a programmable and tunable manner. Although it aims to reconstitute living cell features and address 'origin of life' related questions, rapid development over the years has transformed artificial cells into an engineering tool with huge potential in applied biotechnology. Although the application of artificial cells was introduced decades ago as drug carriers, applications in other sectors are relatively new and could become possible with the technological advancement that can modulate its designing principles. Artificial cells are non-living system that includes no prerequisite designing modules for their formation and therefore allow freedom of assembling desired biological machinery within a physical boundary devoid of complex contemporary living-cell counterparts. As stimuli-responsive biomimetic tools, artificial cells are programmed to sense the surrounding, recognise their target, activate its function and perform the defined task. With the advantage of their customised design, artificial cells are being studied in biosensing, drug delivery, anti-cancer therapeutics or artificial photosynthesis type fields. This mini-review highlights those advanced fields where artificial cells with a minimalistic setup are developed as user-defined custom-made microreactors, targeting to reshape our future 'life'.

Abstract:Artificial cells are constructed to imitate natural cells and allow researchers to explore biological process and the origin of life. The construction methods for artificial cells, through both top-down or bottom-up approaches, have achieved great progress over the past decades. Here we present a comprehensive overview on the development of artificial cells and their properties and applications. Artificial cells are derived from lipids, polymers, lipid/polymer hybrids, natural cell membranes, colloidosome, metal-organic frameworks and coacervates. They can be endowed with various functions through the incorporation of proteins and genes on the cell surface or encapsulated inside of the cells. These modulations determine the properties of artificial cells, including producing energy, cell growth, morphology change, division, transmembrane transport, environmental response, motility and chemotaxis. Multiple applications of these artificial cells are discussed here with a focus on therapeutic applications. Artificial cells are used as carriers for materials and information exchange and have been shown to function as targeted delivery systems of personalized drugs. Additionally, artificial cells can function to substitute for cells with impaired function. Enzyme therapy and immunotherapy using artificial cells have been an intense focus of research. Finally, prospects of future development of cell-mimic properties and broader applications are highlighted.

Abstract:<p>Artificial cells that mimic the architectural and functional characteristics of living cells not only shed light on the physical principle of life but also facilitate development in the areas such as cell engineering and biomedicine. Cell‐free systems carry out the central dogma that refers to the transcription and translation in the absence of living cells, offering a simplified and controllable platform to study cellular biological processes and design principle. Many efforts have been devoted to construct artificial cells by using cell‐free systems with mimicking cellular structures and characteristics, such as molecular crowding, compartments, gene noise, network, dynamic behavior, and cell communication. Here, the recent emerging advances of cell‐free system toward artificial cells are summarized and discussed. It is notable that this area involves multidisciplinary fields such as material chemistry, nanotechnology, and molecular biology. Although there are still much difficulties and challenges for bottom‐up designing systems with desirable properties for cells, it is firmly believed that with the development of synthetic biology, artificial cells based on the cell‐free systems will provide more possibilities to mimic the complex structures and behaviors of cells.</p>

Abstract:One of the main goals of synthetic biology is to build artificial cells in a bottom-up manner, which not only facilitates the deep understanding of the origin of life and cell function but also plays a critical role in the research fields such as the development of artificial cell chassis, tissue models, engineering drug delivery systems, and drug screening tools. However, achieving this goal is extremely challenging. The complexity of cell structures and the miniaturization and diversity of basic modules pose high requirements for the construction methods. The microfluidic chip, as an advanced microanalysis system, serves as an effective tool for building artificial cells. It can accurately control the structure and local microenvironment of artificial cells, becoming the preferred approach for the current research on synthetic life. This article reviewed the methods of constructing, manipulating, and analyzed artificial cells based on microfluidic chips, emphasized the importance of the microenvironment for life systems and artificial self-sustaining systems. In addition, this article demonstrated the wide applications of artificial cells in multiple critical biomedical fields. Exploring the advantages, disadvantages, and application performance of different microfluidic methods can enrich our knowledge about artificial cell research. Finally, we made an outlook on the development of artificial cell research based on microfluidics, expecting that this field can achieve greater breakthroughs and progress.

Abstract:The construction of artificial cells from inanimate molecular building blocks is one of the grand challenges of our time. In addition to being used as simplified cell models to decipher the rules of life, artificial cells have the potential to be designed as micromachines deployed in a host of clinical and industrial applications. The attractions of engineering artificial cells from scratch, as opposed to re‐engineering living biological cells, are varied. However, it is clear that artificial cells cannot currently match the power and behavioural sophistication of their biological counterparts. Given this, many in the synthetic biology community have started to ask: is it possible to interface biological and artificial cells together to create hybrid living/synthetic systems that leverage the advantages of both? This article will discuss the motivation behind this cellular bionics approach, in which the boundaries between living and non‐living matter are blurred by bridging top‐down and bottom‐up synthetic biology. It details the state of play of this nascent field and introduces three generalised hybridisation modes that have emerged.

Abstract:A cell, the fundamental unit of life, contains the requisite blueprint information necessary to survive and to build tissues, organs, and systems, eventually forming a fully functional living creature. A slight structural alteration can result in data misprinting, throwing the entire life process off balance. Advances in synthetic biology and cell engineering enable the predictable redesign of biological systems to perform novel functions. Individual functions and fundamental processes at the core of the biology of cells can be investigated by employing a synthetically constrained micro or nanoreactor. However, constructing a life-like structure from nonliving building blocks remains a considerable challenge. Chemical compartments, cascade signaling, energy generation, growth, replication, and adaptation within micro or nanoreactors must be comparable with their biological counterparts. Although these reactors currently lack the power and behavioral sophistication of their biological equivalents, their interface with biological systems enables the development of hybrid solutions for real-world applications, such as therapeutic agents, biosensors, innovative materials, and biochemical microreactors. This review discusses the latest advances in cell membrane-engineered micro or nanoreactors, as well as the limitations associated with high-throughput preparation methods and biological applications for the real-time modulation of complex pathological states.

Abstract:Abstract Artificial cells are synthetic constructs that mimic the architecture and functions of biological cells. Artificial cells are designed to replicate the fundamental principles of biological systems while also have the ability to exhibit novel features and functionalities that have not been achieved before. Mainly, Artificial cells are made up of a basic structure like a cell membrane, nucleus, cytoplasm and cellular organelles. Nanotechnology has been used to make substances that possess accurate performance in these structures. There are many roles that artificial cells can play such as drug delivery, bio-sensors, medical applications and energy storage. An additional prominent facet of this technology is interaction with biological systems. The possibility of synthetic cells being compatible with living organisms opens up the potential for interfering with specific biological activities. This element is one of the key areas of research in medicine, aimed at developing novel therapies and comprehending life processes. Nevertheless, artificial cell technology is not exempt from ethical and safety concerns. The interplay between these structures and biological systems may give rise to questions regarding their controllability and safety. Hence, the pursuit of artificial cell research seeks to reconcile ethical and safety concerns with the potential advantages of this technology.

Abstract:This study highlights the achievements and help researchers to understand the current status of artificial cells constructed by microfluidics from simple droplets, vesicles, polymeric microparticles to sophisticated cell‐inspired systems. Various applications of artificial cells are also showcased. Furthermore, associated challenges and future directions of the field are outlined. Artificial cells are synthetic constructs that emulate natural cells, with potential applications in areas of energy science, environmental treatment, and the study of life's origins. Nevertheless, the construction of artificial cells is a formidable undertaking, given the intricate nature of natural cells in structures, functions, and working mechanisms. With precise control, high automation, and excellent uniformity, microfluidics has emerged as a promising approach for the construction of artificial cells. This review summarizes the latest microfluidic techniques utilized to construct artificial cells, ranging from simple droplets to sophisticated cell‐inspired systems. These include the generation of droplets, the production of vesicles (lipid‐based and polymer‐based vesicles), the fabrication of polymeric microparticles with various compartments, shapes, and microstructures, as well as the manufacture of sophisticated cell‐inspired systems. The characteristics of different methods for the construction of artificial cells are discussed in detail. Furthermore, the wide‐ranging applications of artificial cells are also showcased. Finally, contemporary obstacles and forthcoming advancements are discussed in the field of microfluidic‐based artificial cells. This review is supposed to stimulate research in the construction of more functional and natural‐like artificial cells, as well as works in the fields of material, biology, environment, medicine, and energy.

Abstract:An artificial cell is a simplified model of a living system, bringing breakthroughs into both basic life science and applied research. The bottom-up strategy instructs the construction of an artificial cell from nonliving materials, which could be complicated and interdisciplinary considering the inherent complexity of living cells. Although significant progress has been achieved in the past 2 decades, the area is still facing some problems, such as poor compatibility with complex bio-systems, instability, and low standardization of the construction method. In this review, we propose creating artificial cells through the integration of different functional modules. Furthermore, we divide the function requirements of an artificial cell into four essential parts (metabolism, energy supplement, proliferation, and communication) and discuss the present researches. Then we propose that the compartment and the reestablishment of the communication system would be essential for the reasonable integration of functional modules. Although enormous challenges remain, the modular construction would facilitate the simplification and standardization of an artificial cell toward a natural living system. This function-based strategy would also broaden the application of artificial cells and represent the steps of imitating and surpassing nature.

Abstract:Synthetic biology, as a new life science discipline, involves structured construction of biological systems using modular and standardized engineering concepts. It enables engineering biological parts, devices, and systems for versatile applications such as medical therapeutics, medical diagnostics, bioenergy production, and understanding biology. Considering the complexity, variability, and redundancy of living cellular systems, a view comes from scientists who focus on the engineering of biosystems in vitro from the bottom up. It is like solution biochemistry for better applications and opens up a new understanding about biology. Therefore, an enabling technology called cell-free synthetic biology has been rapidly adopted and developed.

Abstract:A major goal of synthetic biology is to understand the transition between non-living matter and life. The bottom-up development of an artificial cell would provide a minimal system with which to study the border between chemistry and biology. So far, a fully synthetic cell has remained elusive, but chemists are progressing towards this goal by reconstructing cellular subsystems. Cell boundaries, likely in the form of lipid membranes, were necessary for the emergence of life. In addition to providing a protective barrier between cellular cargo and the external environment, lipid compartments maintain homeostasis with other subsystems to regulate cellular processes. In this Review, we examine different chemical approaches to making cell-mimetic compartments. Synthetic strategies to drive membrane formation and function, including bioorthogonal ligations, dissipative self-assembly and reconstitution of biochemical pathways, are discussed. Chemical strategies aim to recreate the interactions between lipid membranes, the external environment and internal biomolecules, and will clarify our understanding of life at the interface of chemistry and biology.

Abstract:Artificial cell mimicry is extremely valuable in basic and applied biotechnological research. However, the existing construction of artificial cell compartments is typically static, creating a distinction between them and the dynamic natural cells. Herein, we describe a programmable dynamic artificial cell model using swarm nanorobotics and cell-free synthetic transcription-translation solution chemistry. Under the physical control of two- or three-dimensional magnetic fields by varying the direction, strength, and frequency, diverse compartmentation configurations in clusters, chains, and sheets were produced with DNAs and magnetic particles of different morphologies. These diverse dynamic artificial cell architectures exhibit distinct transcription and translation activities. Using swarm robotics and synthetic life chemistry could provide a new paradigm to construct imaginative artificial cells and provide significant insights into the fundamental life processes and biological applications.

Abstract:From the top-down view, an organism is constituted by an assembly of cells where a network of molecules interacts to create a regulated complex system. Cells are a universe of molecular interactions maintained under non-equilibrium regimes. The complex network of interactions drives life where DNA and epigenetic signals are considered as the memory of a cell. The full understanding of these interactions is the first step toward the design of artificial synthetic pathways controlling a program. In our review, we present some insights into the different methods and designs applicable for the development of synthetic artificial cells. A self-replicative process of these cells, thermodynamic and kinetic features, as well as equilibrium systems of artificial cells, will enable scientists to have an in-depth knowledge of this domain. Furthermore, the cascade of encoded molecular chemical interactions orchestrated by the genetic program DNA, biomolecules, epigenetic signals, and nano-factory modulation will be discussed in terms of their application in the entire set of interactomes within a cell.

Abstract:Fabrication of artificial biomimetic materials has attracted abundant attention. As one of the subcategories of biomimetic materials, artificial cells are highly significant for multiple disciplines and their synthesis has been intensively pursued. In order to manufacture robust “alive” artificial cells with high throughput, easy operation, and precise control, flexible microfluidic techniques are widely utilized. Herein, recent advances in microfluidic‐based methods for the synthesis of droplets, vesicles, and artificial cells are summarized. First, the advances of droplet fabrication and manipulation on the T‐junction, flow‐focusing, and coflowing microfluidic devices are discussed. Then, the formation of unicompartmental and multicompartmental vesicles based on microfluidics are summarized. Furthermore, the engineering of droplet‐based and vesicle‐based artificial cells by microfluidics is also reviewed. Moreover, the artificial cells applied for imitating cell behavior and acting as bioreactors for synthetic biology are highlighted. Finally, the current challenges and future trends in microfluidic‐based artificial cells are discussed. This review should be helpful for researchers in the fields of microfluidics, biomaterial fabrication, and synthetic biology.

This limitation highlights the need for a new chassis combining the engineering simplicity of synthetic materials and the functionalities of natural cells. Here, we describe a protocol to assemble a synthetic polymeric network inside bacterial cells, rendering them incapable of cell division and allowing them to resist environmental stressors such as high pH, hydrogen peroxide and cell-wall-targeting antibiotics that would otherwise kill unmodified bacteria. This cellular bioengineering protocol details how bacteria can be transformed into single-lifespan devices that are resistant to environmental stressors and possess programable functionality. We designate the modified bacteria as cyborg bacterial cells. This protocol expands the synthetic biology toolset, conferring precise control over living cells and creating a versatile cell chassis for biotechnology, biomedical engineering and living therapeutics. The protocol, including the preparation of gelation reagents and chassis strain, can be completed in 4 d. The implementation of the protocol requires expertise in microbiology techniques, hydrogel chemistry, fluorescence microscopy and flow cytometry. Further functionalization of the cyborg bacterial cells and adaptation of the protocol requires skills ranging from synthetic genetic circuit engineering to hydrogel polymerization chemistries.
