Abstract:A novel capsule system composed of sodium cellulose sulfate (NaCS), carboxymethyl cellulose (CMC) and poly[dimethyl(diallyl)ammonium chloride] (PDMDAAC) was prepared for improving the properties of NaCS/PDMDAAC capsules. The process parameters, such as CMC concentration (0, 2, 4, 6 and 8g/L), NaCS concentration (20, 25, 30, 35 and 40g/L), PDMDAAC concentration (20, 30, 40, 50, 60, 70 and 80g/L), reaction time and temperature were investigated to understand their effects on the diameter, membrane thickness and mechanical strength of capsules. The optimum operation conditions for preparing NaCS–CMC/PDMDAAC capsules were determined as 6–8g/L CMC, 35–40g/L NaCS, 60g/L PDMDAAC and polymerization for 30–40min. Diffusion of substances with low molecular weight into capsules was investigated, and diffusion coefficients were calculated according to the developed model. The yeast of Candida krusei was chosen as representative cell to evaluate the effects of different cell loading on capsule mechanical strength. Meanwhile the encapsulated osmophilic C. krusei cells were cultured in 250mL shaking flasks for 72h to determine the cell leaking properties in short and long term.

Abstract:Novel capsules loaded with lactoferrin (LF) were prepared using polyelectrolyte complexes that were formed by water soluble chitosan (WSC), sodium cellulose sulfate (NaCS) and sodium polyphosphate (PPS). Normal chitosan (soluble in acidic conditions) was chosen as a control to prepare similar capsules with NaCS and PPS. 1H NMR and FTIR spectra analysis showed that WSC was in a form of chitosan hydrochloride which can be directly dissolved and protonated in acid-free water. SEM results showed that the capsules had a typical wall-capsule structure with a regular spherical shape and an average diameter of 1.97mm. TGA studies revealed that the thermal stability of the capsules were enhanced and the moisture content of the drug-free/loaded capsules were 6.3% and 3.2%. SDS-PAGE results showed that the primary structures of the processed LF in the capsules were unchanged. Drug loading (LE%) and encapsulation efficiency (EE%) analysis showed that the capsules had a higher LE% (45.6%) and EE% (70.7%) than that of the control. In vitro release studies showed that the capsules had a regular and sustainable release profiles in simulated colonic fluid. All of these results indicated that the capsules prepared could be used as a candidate protein drug carrier for colon.
Fabrication and formation studies on single-walled CA/NaCS-WSC microcapsules

Abstract:The micron-sized calcium alginate/sodium cellulose sulfate-water soluble chitosan (CA/NaCS-WSC) microcapsules were prepared by membrane emulsification method using sodium alginate (NaAlg), NaCS and WSC as raw materials. The CA/NaCS microspheres prepared dispersed well and held spherical shape with an emulsifier volume ratio of 7:3 (Span 80:Tween 80) and a concentration of cross-linking agent of 1.5% (w/v) calcium chloride and 5% (w/v) sodium chloride. The CA/NaCS-WSC microcapsules had a spherical shape with average diameter of 62.36 +/- 13.87 pm. A fluorescent ring could be seen obviously on the surface of CA/NaCS-WSC microcapsules under confocal microscope, when WSC was labeled by fluorescein isothiocyanate. The discussion on the formation studies implied that Ca2+ could diffuse into the droplets of NaAlg/NaCS forming CA/NaCS microspheres, while NaCS.could react with WSC forming a polyelectrolyte complexes film. The microcapsules prepared with typical wall-capsule/core structure could be used to develop micron-sized drug delivery carriers. (C) 2015 Elsevier B.V. All rights reserved.

Abstract:Chitosan, the cationic polysaccharide derived from the natural polysaccharide chitin, has been studied as a biomaterial for more than two decades. As a polycationic polymer with favorable properties, it has been widely used to form polyelectrolyte complexes with polyanions for various applications in drug delivery fields. In recent years, a growing number of studies have been focused on the preparation of polyelectrolyte complexes based on chitosan and its water soluble derivatives. They have been considered well-suited as biomaterials for a number of vital drug carriers with targeted/controlled release profiles, e.g., films, capsules, microcapsules. In this work, an overview highlights not only the favorable properties of chitosan and its water soluble derivatives but also the good performance of the polyelectrolyte complexes produced based on chitosan. Their various types of applications as drug carriers are reviewed in detail.
Preparation of Sustained Release Capsules by Electrostatic Dry Powder Coating, Using Traditional Dip Coating As Reference

Abstract:Lately, a great deal of attention is being paid to capsule coating, since the coat protects active pharmaceutical ingredients (APIs) from damage, as is in the case of tablet and pellet. However, moisture and heat sensitivity of gelatin shells make it challenging to coat capsules using the conventional aqueous coating techniques. In an effort to overcome this challenge, the present study aims to coat capsules using two different coating techniques: electrostatic dry powder coating (EDPC) and dip coating (DC). Both capsule coatings and free films were prepared by these two coating techniques, and the effects of coating formulations and processing conditions on the film quality were investigated. The corresponding drug in vitro release and mechanisms were characterized and compared. The results of dissolution tests demonstrated that the drug release behavior of both EDPC and DC coated capsules could be optimized to a sustained release of 24 h, following the Fick's diffusion law. The results of this study suggest that EDPC method is better than DC method for coating capsules, with respect to the higher production efficiency and better stability, indicating that this dry coating technology has promised in gelatin capsule coating applications.

Abstract:Hybrid CaCO3 particles were used as templates to fabricate two kinds of "micelles enhanced" polyelectrolyte capsules with three different polyel ectrolytes such as poly( allylamine hydrochloride), poly( sodium styrenesulfonate), and poly (di-allyldlmethylammonium chloride) via LbL technique. Both of (PAH/PSS)(6) and (PADA/PSS)(6) capsules had good spherical shape and good dispersion by CLSM. In contrast with conventional polyelectrolyte capsules, these novel micelles enhanced capsules had thick and compact capsule wall. Meanwhile, fine FESEM images showed that there was a significant difference in microstructure and topology between the two kinds of capsules made of different component polyelectrolytes.
Synthesis and Characterization of Fucoidan/chitosan-Coated Nanoliposomes for Enhanced Stability and Oral Bioavailability of Hydrophilic Catechin and Hydrophobic Juglone

Abstract:This study aimed to improve the thermodynamic performance of nanoliposomes (NLs) using fucoidan (F) as the second-layer coating biopolymer along with chitosan (CS), to control the delivery and bioavailability of catechin (C) and juglone (J). The stabilized liposomal carrier of F/CS-conjugated JC-NL (F-CS-JC-NL) was developed with optimum concentrations of CS (0.09 wt%) and F (0.10 wt%), with the highest encapsulation efficiency of juglone (95.47%) and catechin (90.88%). Physicochemical characterization revealed that F-CS-JC-NL disclosed improved stability under different pH and ionic strengths, with the maximum juglone/catechin retention under thermal, oxidative and storage conditions. In vitro digestion revealed that NL double-coating (F-CS-JC-NL) significantly reduced compound leakage in the gastrointestinal tract, providing a controlled release and better bioavailability of juglone/catechin compared to CS-JC-NL and JC-NL. Conclusively, this study provides a novel NL-based de-livery carrier with enhanced physicochemical stability and controlled release that might have promising use in delivering functional ingredients.

Abstract:A novel biocompatible shell-crosslinked nanocapsule system was developed based on nanoemulsion templates stabilized by a class of food proteins. The nanoemulsion templates were prepared using a combination of mechanical mixing and high-pressure homogenization, while the nanocapsule shell formed simultaneously through calcium ions-induced crosslinking of the food proteins. These core–shell structured nanocapsules with a particle size of about 200nm showed high drug-loading capacity and well improved stability in comparison with their nanoemulsion counterpart. The nanocapsule suspension can be freeze-fried directly; and the solidified nanocapsules can be well reconstituted in water, retaining their original particle size. It is concluded that the nanoemulsion-templated core–shell structured nanocapsules can be used as novel drug delivery systems with high loading capacity for poorly water-soluble drugs as well as well improved long-term and storage stability. Furthermore, the presence of surface food proteins introduces carboxyl and amine moieties, which enables the nanocapsules to anchor ligands, suggesting its potential application in targeting drug delivery, bioimaging and therapeutics.

Abstract:Plant-based hard capsules have gained considerable attention because of their great properties. Accordingly, designing and developing of these kinds of capsules will be a difficult task. Herein, an innovative pullulan-based hard capsule formulation was prepared for the first time. A series of characterization approaches, including Fourier transform infrared, field emission scanning electron microscope, and rheology analysis, were utilized to figure out the straightforward preparation of a designed hard capsule. Many tests and experiments were performed to achieve the optimum capsule formulation. Based on the obtained results, specifications such as uniform downfall and non-desirable adhesion, and other ideal characteristics of the capsule display the critical function. The gelling promoter of divalent cationic salts is more beneficial than its single-valent counterparts. With respect to the key role of gelling promoter, the presence of chosen MgSO4.7H2O salt and the source of selected carrageenan are important parameters to achieve optimal formulation. Moreover, field emission scanning electron microscope images illustrate that the weight ratio of 3.5 (gelling agent to salt) displays uniform surface morphology without any impurities or other foreign materials. Likewise, the outcomes of the rheology test also illustrated that the weight ratio of 3.5 is preferable. Considering the different weight ratios, the benefits of a weight ratio of 3.5 outweigh the other investigated ratios. Overall, the current research addresses substantial information about developing pullulan-based hard capsules for target usage.
Crosslinked Polysaccharide Nanocapsules: Preparation and Drug Release Properties.

Abstract:Crosslinked polysaccharide and composite polysaccharide capsules with diameters ranging from 200nm to several microns and wall thicknesses of several tens of nanometers have been fabricated by interfacial polymerization of methacrylated N,N-diethylaminoethyl dextran (DdexMA) and DdexMA-vinyl terminated polylactide macromonomers (PLAM). In this method, chloroform droplets or PLAM-containing chloroform droplets were dispersed in water, on which water soluble DdexMA was polymerized to form closed shell structure. Their hollow nature was confirmed by confocal laser scanning microscopy and transmission electron microscopy. Dynamic light scattering revealed that these capsules possess good stability against coagulation during storage. Fourier transform infrared and elemental analysis found that the DdexMA capsules were actually composed of crosslinked DdexMA, while the DdexMA–PLAM capsules were composed of the crosslinked DdexMA–PLAM copolymers and PLAM. By dissolution of ibuprofen in the chloroform droplets, drug-loaded capsules were also fabricated. It was found that the loaded drug could be released again in a sustained manner for up to 100h. The capsule walls had a prominent effect in slowing down the drug release rate, particularly for the DdexMA–PLAM capsules.
A Novel System for Water Soluble Protein Encapsulation with High Efficiency: “micelles Enhanced” Polyelectrolyte Capsules

Abstract:Novel "micelles enhanced" polyelectrolyte (PE) capsules based on functional templates of hybrid calcium carbonate were fabricated. Evidences suggested that the structure of capsule wall was different from that of conventional PE capsules, and the wall permeability of these PE capsules changed significantly. Lysozyme, a positively charged protein in neutral solution, was studied as a model protein to be encapsulated into the "micelles enhanced" PE capsules. Confocal laser scanning microscope was used to observe the entrapping process in real time, while UV-Vis spectroscope and scanning force microscope measurements suggested the high efficiency of encapsulation. In addition, the fluorescence recovery after photobleaching technique was employed to determine the existence form of deposited molecules. Further studies showed even negatively charged water-soluble peptides or proteins can be encapsulated into these hybrid capsules by modulating the pH value in bulk solution under its isoelectronic point as well.
Sodium alginate/carboxymethyl starch/κ-carrageenan enteric soft capsule: Processing, characterization, and rupture time evaluation

Abstract:Although gelatin has good characteristics in preparing soft capsules, its noticeable shortcomings force researchers to further develop substitutes for gelatin soft capsules. In this paper, sodium alginate (SA), carboxymethyl starch (CMS) and κ-carrageenan (κ-C) were used as matrix materials, and the formula of the co-blended solutions was screened through rheological method. In addition, films of the different blends were characterized by thermogravimetry analysis, SEM, FTIR, X-ray, water contact angle and mechanical properties. The results showed that κ-C had strong interaction with CMS and SA, and the mechanical properties of capsule shell were greatly improved by the addition of κ-C. When the ratio of CMS/SA/κ-C was 2:0.5:1.5, the microstructure of the films was more dense and uniform. In addition, this formula had the best mechanical properties and adhesion properties, and was more suitable for the production of soft capsules. Finally, a novel plant soft capsule was successfully prepared by dropping method, and its appearance and rupture properties met the requirements of enteric soft capsules. In simulated intestinal fluid, the soft capsules were almost completely degraded within 15 min, and they were superior to the gelatin soft capsules. Therefore, this study provides an alternative formula for preparing enteric soft capsules.
Novel Nanoparticles Composed of Chitosan and Β-Cyclodextrin Derivatives As Potential Insoluble Drug Carrier

Abstract:This research was aim to develop novel cyclodextrin/chitosan (CD/CS) nanocarriers for insoluble drug delivery through the mild ionic gelation method previously developed by our lab. A series of different beta-cyclodextrin (beta-CD) derivatives were incorporated into CS nanoparticles including hydroxypropyl-beta-cyclodextrin (HP-beta-CD), sulphobutylether-beta-cyclodextrin (SB-beta-CD), and 2,6-di-O-methy-beta-cyclodextrin (DM-beta-CD). Various process parameters for nanoparticle preparation and their effects on physicochemical properties of CD/CS nanoparticles were investigated, such as the type of CD derivatives, CD and CS concentrations, the mass ratio of CS to TPP (CS/TPP), and pH values. In the optimal condition, CD/CS nanoparticles were obtained in the size range of 215-276 nm and with the zeta potential from 30.22 mV to 35.79 mV. Moreover, the stability study showed that the incorporation of CD rendered the CD/CS nanocarriers more stable than CS nanoparticles in PBS buffer at pH 6.8. For their easy preparation and adjustable parameters in nanoparticle formation as well as the diversified hydrophobic core of CD derivatives, the novel CD/CS nanoparticles developed herein might represent an interesting and versatile drug delivery platform for a variety of poorly water-soluble drugs with different physicochemical properties. Building a polysaccharide hydrogel capsule delivery system for control release of ibuprofen

Abstract:Development of a delivery system which can effectively carry hydrophobic drugs and have pH response is becoming necessary. Here we demonstrate that through preparation of β-cyclodextrin polymer (β-CDP), a hydrophobic drug molecule of ibuprofen (IBU) was incorporated into our prepared β-CDP inner cavities, aiming to improve the poor water solubility of IBU. A core-shell capsule structure has been designed for achieving the drug pH targeted and sustained release. This delivery system was built with polysaccharide polymer of Sodium alginate (SA), sodium carboxymethylcellulose (CMC) and hydroxyethyl cellulose (HEC) by physical cross-linking. The drug pH-response control release is this hydrogel system's chief merit, which has potential value for synthesizing enteric capsule. Besides, due to our simple preparing strategy, optimal conditions can be readily determined and the synthesis process can be accurately controlled, leading to consistent and reproducible hydrogel capsules. In addition, phase-solubility method was used to investigate the solubilization effect of IBU by β-CDP. SEM was used to prove the forming of core and shell structure. FT-IR and 1H-NMR were also used to perform structural characteristics. By the technique of UV determination, the pH targeted and sustained release study were also performed. The results have proved that our prepared polysaccharide hydrogel capsule delivery system has potential applications as oral drugs delivery in the field of biomedical materials.

Carboxyl-modified nanocellulose (cNC) enhances the stability of cNC/pullulan bio-nanocomposite hard capsule against moisture variation

Abstract:The quality of polysaccharide-based films and hard capsules is often affected by changes in relative humidity, manifesting as unstable water content, and changes in mechanical strength that make them brittle or soft. Herein, carboxyl-modified nanocellulose (cNC) was prepared and used as a new component to successfully improve the moisture resistance of cNC/pullulan/high-acyl gellan bio-nanocomposite hard capsules (NCPGs). Homogenously dispersed cNC in the pullulan/high-acyl gellan matrix could render the formation of more hydrogen bonds that provided additional water-binding sites and limited the free movement of pullulan and high-acyl gellan molecular chains within NCPGs. This contributed to a decreased amount of pooling adsorption water and an increased amount of Langmuir adsorption water in NCPGs, as compared to pullulan/high-acyl gellan hard capsules (PGs) without cNC. Therefore, the equilibrium moisture content (EMC) values of NCPGs decreased at 83 % relative humidity and increased at 23 % relative humidity compared to those of PGs. Together with enhanced mechanical and barrier properties, NCPGs effectively protected encapsulated amoxicillin and probiotic powder from changes in the outside humidity. Additionally, NCPGs exhibited faster drug release. This study presents a new mechanism and strategy for fabricating films and hard capsules with enhanced stability against moisture variation.

Abstract:A novel microcapsule system composed of sodium cellulose sulfate (NaCS) and chitosan was prepared by layer-by-layer (LbL) self-assembly technique. As a potential drug delivery system, it had several advantages in biocompatibility and biodegradation due to its use of natural polysaccharides. Some parameters in the preparation of the microcapsule, such as layers in the LbL process (up to 20 layers), chitosan viscosities (50, 100, and 200 mPas), outmost layer materials (NaCS or chitosan), and NaCl concentrations (0 M, 0.5 M, 1 M), were investigated to better understand their effects on the LbL process. A novel method for removing the core templates PLA and CaCO3 from the formed core-shell structure was proposed by addition of N-methyl pyrrolidone or EDTA-Na. This preparation process was well observed by a microscope-camera system. The hollow microcapsules were tested by scanning electron microscopy (SEM), confocal laser scanning microscopy (CLSM) and scanning probe microscopy (SPM) to characterize their size, morphology, and surface. The results showed that the shell thickness of microcapsules was about 6 nm, and the average thickness of one layer of NaCS/chitosan polyelectrolyte complex was 1.5 nm. The molecular weight cutoff in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) showed about 70 kDa by using of diffusion test with the cell lysate of Escherichia coli.

Abstract:β-cyclodextrin (CD) grafted N-maleoyl chitosan (CD-g-NMCS) with two different degrees of substitution (DS) of N-maleoyl (DS = 21.2% and 30.5%) were synthesized from maleic anhydride and chitosan bearing pendant cyclodextrin (CD-g-CS). CD-g-NMCS based nanoparticles were prepared via an ionic gelation method together with chitosan and CD-g-CS nanoparticles. The size and zeta potential of prepared CD-g-NMCS nanoparticles were 179.2~274.0 nm and 36.2~42.4 mV, respectively. In vitro stability test indicated that CD-g-NMCS nanoparticles were more stable in phosphate-buffered saline compared with chitosan nanoparticles. Moreover, a poorly water-soluble drug, ketoprofen (KTP), was selected as a model drug to study the obtained nanoparticle's potentials as drug delivery carriers. The drug loading efficiency of CD-g-NMCS20 nanoparticles were 14.8% for KTP. MTT assay showed that KTP loaded CD-g-NMCS nanoparticles were safe drug carriers. Notably, in vitro drug release studies showed that KTP was released in a sustained-release manner for the nanoparticles. The pharmacokinetic of drug loaded CD-g-NMCS20 nanoparticles were evaluated in rats after intravenous administration. The results of studies revealed that, compared with free KTP, KTP loaded CD-g-NMCS20 nanoparticles exhibited a significant increase in AUC0→24h and mean residence time by 6.6-fold and 2.9-fold, respectively. Therefore, CD-g-NMCS nanoparticles could be used as a novel promising nanoparticle-based drug delivery system for sustained release of poorly water-soluble drugs. The carboxylic acid groups of the CD-g-NMCS molecule provide convenient sites for further structural modifications including introduction of tissue- or disease- specific targeting groups.
Cellulose nanofiber/nanocrystal reinforced capsules: a fast and facile approach toward assembly of liquid-core capsules with high mechanical stability

Abstract:Liquid-core capsules of high mechanical stability open up for many solid state-like applications where functionality depending on liquid mobility is vital. Herein, a novel concept for fast and facile improvement of the mechanical properties of walls of liquid-core capsules is reported. By imitating nature's own way of enhancing the mechanical properties in liquid-core capsules, the parenchyma plant cells found in fruits and vegetables, a blend of short cellulose nanofibers (<1 μm, NFC) and nanocrystals (CNC) was exploited in the creation of the capsule walls. The NFC/CNC blend was prepared from a new version of the classical wood pulp hydrolysis. The capsule shell consisted of a covalently (by aromatic diisocyanate) cross-linked NFC/CNC structure at the outer capsule wall and an inner layer dominated by aromatic polyurea. The mechanical properties revealed an effective capsule elastic modulus of 4.8 GPa at 17 wt % NFC/CNC loading, about six times higher compared to a neat aromatic polyurea capsule (0.79 GPa) and 3 orders of magnitude higher than previously reported capsules from regenerated cellulose (0.0074 GPa). The outstanding mechanical properties are ascribed to the dense nanofiber structure, present in the outer part of the capsule wall, that is formed by oriented NFC/CNC of high average aspect ratio (L/d ∼ 70) and held together by both covalent (urethane bonds) and physical bonds (hydrogen bonds).
Nanoengineering of Polymeric Capsules with A Shell-In-Shell Structure

Abstract:Polymeric capsules possessing a shell-in-shell structure were fabricated by assembling layers of polyelectrolytes with interlayers of silica nanoparticles onto microspheres of melamine formaldehyde. Subsequently the cores were removed with hydrochloric acid and the silica layers with hydrofluoric acid. Removal of the silica layers leaves an aqueous phase of a poly(allylamine hydrochloride) solution between an outer and an inner polyelectrolyte shell. The interlayer thickness could be varied by the diameter of silica particles or by the number of deposition steps. The double-shell structure of the microcapsules was determined by scanning force microscopy, confocal laser scanning microscopy, and transmission electron microscopy on ultramicrotome sections in detail. In contrast to one-shell capsules of polyelectrolytes, the permeability for small molecules does not change with time and temperature. In addition, the shell-in-shell capsules exhibit a remarkably higher mechanical stability as shown by experiments of osmotically induced deformations.
