Abstract:Nature uses combined covalent (chemical bonds) and non‐covalent (physical bonds) synthesis in a highly, controlled multi‐step fashion to create functional materials with different mechanical and dynamic properties out of similar building blocks. Surprisingly, this control in fully synthetic systems remains elusive, even though the effects of formulation pathways on the assembly processes have been emphasized—highlighting the importance of and relationship between energy landscapes and function in synthetic systems. Here, we control multiple, coherent supramolecular assembly processes (fiber formation and crosslinking) to formulate hydrogels with tunable mechanical and dynamic properties. Hydrogels are prepared via two different formulation methods using similar building blocks (monofunctional and bifunctional supramolecular monomers), including (1) the mixing of the supramolecular monomers under basic conditions (Fbasic) and (2) the mixing of the supramolecular monomers under neutral conditions (Fneutral). In Fbasic, network formation is induced via simultaneous fiber formation and crosslinking, yielding homogeneously mixed networks which are dense, stiff (~10 kPa) and robust. In Fneutral, network formation is induced through sequential fiber formation and crosslinking, yielding heterogenous, soft (~2 kPa) and dynamic networks. With these results, we advance towards the controlled, multistep, non‐covalent synthesis to create larger hierarchical, functional structures, similar to nature.

Abstract:Hydrogels are a versatile class of materials, often exhibiting stimuli‐responsiveness. Herein the high‐throughput synthesis and screening of methacrylate‐based, photodegradable hydrogels on a sub‐microliter scale, using droplet microarrays is reported. The generated data is used to train a machine learning model that helped us to significantly improve the target hydrogel properties through Bayesian optimization. Due to the large chemical space, the design of functional and responsive soft materials poses many challenges but also offers a wide range of opportunities in terms of the scope of possible properties. Herein, an experimental workflow for miniaturized combinatorial high‐throughput screening of functional hydrogel libraries is reported. The data created from the analysis of the photodegradation process of more than 900 different types of hydrogel pads are used to train a machine learning model for automated decision making. Through iterative model optimization based on Bayesian optimization, a substantial improvement in response properties is achieved and thus expanded the scope of material properties obtainable within the chemical space of hydrogels in the study. It is therefore demonstrated that the potential of combining miniaturized high‐throughput experiments with smart optimization algorithms for cost and time efficient optimization of materials properties.

Abstract:Additive fabrication of biocompatible 3D structures out of liquid hydrogel solutions has become pivotal technology for tissue engineering, soft robotics, biosensing, drug delivery, etc. Electron and X-ray lithography are well suited to pattern nanoscopic features out of dry polymers, however, the direct additive manufacturing in hydrogel solutions with these powerful tools is hard to implement due to vacuum incompatibility of hydrated samples. In this work, we resolve this principal impediment and demonstrate a technique for in-liquid hydrogel 3D-sculpturing separating high vacuum instrumentation and volatile sample with ultrathin molecularly impermeable membranes transparent to low-energy electrons and soft X-rays. Using either scanning focused electron or synchrotron soft X-ray beams, the principle of the technique, particularities of the in-liquid crosslinking mechanism and factors affecting the ultimate gel feature size are described and validated through the comparison of experiments and simulations. The potential of this technique is demonstrated on a few practical examples such as encapsulation of nanoparticles and live-cell as well as fabrication of mesoscopic 3D-hydrogel structures via modulation of the beam energy
In Situ Synthesis of Poly(Acrylic Acid) Physical Hydrogels from Silica Nanoparticles

Abstract:This paper introduces a versatile method to synthesize physical hydrogels by silica nanoparticle (SN) surface in situ polymerization. A series of poly(acrylic acid) chains were covalently grafted from the SNs that were treated with γ-methacryloxypropyl trimethoxy silane. The entanglements among the grafted chains constructed the hydrogel networks and the SNs played the role of ‘analogous crosslinking points’. The SNs were encapsulated by the grafted polymer chains and formed a ‘core–shell’ structure. It was found that the molecular weight of the grafted chains and the content and diameter of the SNs affected the hydrogel’s properties. In comparison with chemically crosslinked hydrogels, the hybrid hydrogels exhibited excellent mechanical properties and could endure stress as high as 3.16 MPa and an elongation of 440% at a water content of 100 wt%. The gel–sol phenomenon confirmed that this hybrid hydrogel belonged to the physically crosslinked type.
Additive-Free Method for Enhancing the Volume Phase Transition Rate in Light-Responsive Hydrogels: A Study of Micro-Nano Bubble Water on PNIPAM-co-AAc Hydrogels

Abstract:Light-responsive hydrogels containing light-thermal convertible pigments have received interest for their possible applications in light-responsive shutters, valves, drug delivery systems, etc. However, their utility is limited by the slow response time. In this study, we investigated the use of micro-nano bubble water as a preparation solvent to accelerate the volume phase transition kinetics of poly(N-isopropylacrylamide-co-acrylic acid) (PNIPAM-co-AAc) hydrogels. The hydrogels were characterized by dynamic light scattering (DLS) and dissolved oxygen (DO) measurements. The mechanical properties, surface morphology, and chemical composition of the hydrogels were analyzed by Young's modulus measurements, scanning electron microscopy (SEM), and Fourier transform infrared (FT-IR) spectroscopy, respectively. The results showed that hydrogels prepared with bubble water changed the volume transition rate by more than two orders of magnitude by simply changing the standing time of the bubble water for only a few hours. The cooperative diffusion coefficients obtained from the light-induced volume transition kinetics correlated linearly with Young's modulus and metastable state swelling ratio. Our results suggest that bubbles act as efficient water channels, thereby modulating the response rate and providing a simple, additive-free method for preparing hydrogels with a wide range of response rates.

Abstract:Hydrogels are the most relevant biochemical scaffold due to their tunable properties, inherent biocompatibility, and similarity with tissue and cell environments. Over the past decade, hydrogels have developed from static materials to "smart" responsive materials adapting to various stimuli, such as pH, temperature, chemical, electrical, or light. Light stimulation is particularly interesting for many applications because of the capability of contact-free remote manipulation of biomaterial properties and inherent spatial and temporal control. Moreover, light can be finely adjusted in its intrinsic properties, such as wavelength and intensity (i.e., the energy of an individual photon as well as the number of photons over time). Water is almost transparent for light in the photochemically relevant range (NIR-UV), thus hydrogels are well-suited scaffolds for light-responsive functionality. Hydrogels' chemical and physical variety combined with light responsiveness makes photoresponsive hydrogels ideal candidates for applications in several fields, ranging from biomaterials, medicine to soft robotics. Herein, the progress and new developments in the field of light-responsive hydrogels are elaborated by first introducing the relevant photochemistries before discussing selected applications in detail.

Abstract:Light-responsive hydrogel, as a typical functional hydrogel, is composed of crosslinked hydrophilic polymer chains, photosensitive molecules and water. Under illumination, the photosensitive molecules exhibit a transition in hydrophilic/ hydrophobic properties, leading to migration of water molecules and deformation of the hydrogel based on the temporal and spatial distribution of light. In this study, we propose a nonequilibrium thermodynamic framework to study the photo-chemo-mechano behaviors of light-responsive hydrogel. We firstly describe the light propagation in the hydrogel and the photochemical reaction kinetics. New free energy functions to expound the relationship between the photochemical reaction and thermodynamic process are established, and the constitutive equations are derived. Subsequently, we implant the model into a multi-field coupling analysis software, COMSOL, to conduct a spatio-temporal analysis of the hydrogel's response under uniform light exposure. Finally, we simulate the inhomogeneous deformation of light-responsive hydrogel strips under different illumination conditions and compare the results with experiments. The results highlight the importance of the photochemical reaction rate and the redistribution of light field caused by deformation. The present research holds potential for the precise manipulation and optimal design of light-responsive hydrogel in prospective applications, and offers insights for the synthesis of similar materials as well as the design of pertinent devices.

Abstract:Living organisms operate far from equilibrium in order to maintain their adaptive structures and functions by continuous energy consumption. The development of synthetic out-of-equilibrium systems using light as an input fuel will allow for remote and noninvasive control over a range of important chemical/biological processes. Here, we report on the rational design of light-fueled DNA hydrogels that exhibit life-like nonequilibrium and dissipative features. Such hydrogels are formed through a two-component mixing strategy, where merocyanine photoacid bearing a robust pH-adjustment capability delicately imparts photoinactive but pH-sensitive DNA components with an adaptive response to light. The transient hydrogel spontaneously dissipates to its sol state in the dark, enabling a highly reversible sol–gel transition that is fine-tuned by irradiation conditions. We construct reprogrammable DNA hydrogel shapes/patterns by alternating the photopatterning/dark-erasing processes using a single recyclable ink. Our work provides a general waste-free approach to developing other light-fueled out-of-equilibrium hydrogels for applications in smart materials and soft robotics.

Abstract:Surfactant‐free colloidal syntheses in aqueous media are attractive to develop nanomaterials relevant for various applications, e.g. catalysis or medicine. However, controlled green syntheses without surfactants of metal nanoparticles in aqueous media remain scarce. Here, room temperature syntheses of gold (Au) nanoparticles (NPs) that require only HAuCl4, alkaline water and an alcohol, i.e. relatively benign chemicals and mild reaction conditions, are developed. The findings of a comprehensive multi‐parameters screening performed in small volumes (< 3 mL) over 1000+ experiments pave the way to greener high throughput screenings of large parametric spaces and lead to scalable (100 mL) synthetic strategies. A rational selection of the alcohol is proposed. The influence of lights with defined wavelengths (222‐690 nm) is investigated. It is found that lights with lower wavelengths favor the formation of smaller 5 nm NPs. Different kinetics and formation pathways are observed for different alcohols and for lights with different wavelengths. The sensitivity to various experimental parameters increases in the order glycerol < ethylene glycol < ethanol < methanol. New strategies for a rational fine size control, and to some extend shape control, are identified. The results lead to more sustainable and reproducible surfactant‐free colloidal syntheses of NPs.

Abstract:A facile route for the fabrication of surface-attached hydrogel thin films with well-controlled chemistry and tailored architecture on wide range of thickness from nanometers to micrometers is reported. The synthesis, which consists in cross-linking and grafting the preformed and ene-reactive polymer chains through thiol--ene click chemistry, has the main advantage of being well-controlled without the addition of initiators. As thiol--ene click reaction can be selectively activated by UV-irradiation (in addition to thermal heating), micropatterned hydrogel films are easily synthesized. The versatility of our approach is illustrated by the possibility to fabricate various chemical polymer networks, like stimuli-responsive hydrogels, on various solid substrates, such as silicon wafers, glass, and gold surfaces. Another attractive feature is the development of new complex hydrogel films with targeted architecture. The fabrication of various architectures for polymer films is demonstrated: multilayer hydrogel films in which single-networks are stacked one onto the other, interpenetrating networks films with mixture of two networks in the same layer, and nanocomposite hydrogel films where nanoparticles are stably trapped inside the mesh of the network. Thanks to its simplicity and its versatility this novel approach to surface-attached hydrogel films should have a strong impact in the area of polymer coatings

Abstract:The growing demand for drug carriers and green-technology-based tissue engineering materials has enabled the fabrication of different types of micro- and nano-assemblies. Hydrogels are a type of material that have been extensively investigated in recent decades. Their physical and chemical properties, such as hydrophilicity, resemblance to living systems, swelling ability and modifiability, make them suitable to be exploited for many pharmaceutical and bioengineering applications. This review deals with a brief account of green-manufactured hydrogels, their characteristics, preparations, importance in the field of green biomedical technology and their future perspectives. Only hydrogels based on biopolymers, and primarily on polysaccharides, are considered. Particular attention is given to the processes of extracting such biopolymers from natural sources and the various emerging problems for their processing, such as solubility. Hydrogels are catalogued according to the main biopolymer on which they are based and, for each type, the chemical reactions and the processes that enable their assembly are identified. The economic and environmental sustainability of these processes are commented on. The possibility of large-scale processing in the production of the investigated hydrogels are framed in the context of an economy aimed at waste reduction and resource recycling.

Three-dimensional Manipulation of Hydrogel Microparticles by Positional Change of Light Focus in Stop Flow Lithography

Abstract:Hydrogel microparticles have garnered substantial attention in the fields of drug delivery, tissue engineering, and biosensing due to their properties of high biocompatibility, hydrophilicity, and tunability. Stop Flow Lithography (SFL), a microfluidic-based photopolymerization technique, has emerged as one of the leading methods by which these particles are synthesized due to its high throughput of controlled geometrically defined particles and ease of use. Previous research assumes that light distributes evenly throughout the SFL flow channel due to the almost instantaneous speed of light (3*108 m/s), synthesizing uniformly shaped hydrogels. In this study, the position of UV-light focus in SFL was manipulated using microscope focus, to which three-dimensional differences in hydrogel shapes were observed, disproving this presumption and ultimately suggesting that light intensity is not uniformly distributed. In observing these three-dimensional differences, we also discovered a technique by which SFL, normally confined to two-dimensional control, can be used to manipulate three-dimensional hydrogel shape. Furthermore, we explored potential applications of this technique in the fields of multiplex immunoassays, tissue engineering, and drug delivery. Our results indicate that the ability to manipulate hydrogels in a three-dimensional manner significantly enhanced all three fields, demonstrating promising application of this technique in biomedical and clinical fields.

Abstract:Aims & Scope: The natural polymers, extracted from the natural products or formed from the biochemical reaction or photosynthesis in natural world, including the polymers derived from plants, such as cellulose, starch, lignin, natural rubber and alginate, etc., the polymers from the animals, including chitin, chitosan, hyaluronic acid and protein, etc., and the polymer from microbial fermentation, such as polylactic acid, polycaprolactone and poly (glutamic acid) etc.. This kind of materials is the precious natural resources endowed by our mother nature. They are inexhaustible in supply and always available for use. By the way, almost all of the natural polymers are biocompatibility, biodegradability and environmental-friendly, and they are widely used in medicinal fields. Hydrogels are crosslinked polymeric networks prepared from hydrophilic polymers, which can absorb and retain high content of water while maintaining their three-dimensional integrity. As the diversified properties of the natural polymers, they are the ideal skeleton for preparation of the hydrogels. The hydrogels prepared from the natural polymers could show interesting stimuli-responsiveness, good biodegradability, biocompatibility, and good flexibility similar to natural tissue. As the above properties, nowadays hydrogels are widely used in the medicinal fields. They can be used as the wound dressing, the carrier for drug delivery, the substrate for cell culture and scaffold for tissue engineering. They are also widely used as the critical materials for microfluidic chips, medical examination, photodynamic therapy, and gene therapy, etc.. In the last 2-3 decades there are a lot of reported papers that highlight the hydrogels based on natural polymers and applications in the medicinal fields. In this theme issue, we’ll bring together a number of top-ranked international scientists to discuss the recent advances about the advancements of natural polymer based hydrogels in synthetic pathways, properties and applications in medicinal fields. This special issue is aiming for a broad audience of readers in the field of material sciences, chemistry, medicine and pharmaceutics.

Abstract:Soft matter integration of photosensitizers and catalysts provides promising solutions to developing sustainable materials for energy conversion. Particularly, hydrogels bring unique benefits as spatial control and 3D‐accessiblity of molecular units as well as recyclability. Herein, we report the preparation of polyampholyte hydrogels based on poly(dehydroalanine) (PDha). Chemically‐crosslinked PDha with bis‐epoxy poly(ethylene glycol) lead to a transparent, self‐supporting hydrogel. Due to the ionizable groups on PDha, this 3D polymeric matrix can be anionic, cationic, or zwitterionic depending on the pH, and its high density of dynamic charges has a potential for electrostatic attachment of charged molecules. We realized the integration of the cationic molecular photosensitizer [Ru(bpy)3]2+ (bpy = 2,2′‐bipyridine), which is a reversible process controlled by pH, leading to light harvesting hydrogels. They were further combined with either a thiomolybdate catalyst ([Mo3S13]−2) for hydrogen evolution reaction (HER) or a cobalt polyoxometalate catalyst (Co4POM = [Co4(H2O)2(PW9O34)2]10−) for oxygen evolution reaction (OER). Under the optimized condition, the resulting hydrogels showed catalytic activity in both case upon visible light irradiation. In case of OER, we observed higher photosensitizer stability compared to homogeneous systems, as the polymer environment seems to influence decomposition pathways. Abstract:Surfactant-free colloidal syntheses in aqueous media are attractive to develop nanomaterials relevant for various applications, e.g. catalysis or medicine. However, controlled green syntheses without surfactants of metal nanoparticles in aqueous media remain scarce. Here, room temperature syntheses of gold (Au) nanoparticles (NPs) that require only HAuCl4, alkaline water and an alcohol, i.e. relatively benign chemicals and mild reaction conditions, are developed. The findings of a comprehensive multi-parameters screening performed in small volumes (< 3 mL) over 1000+ experiments, that pave the way to greener high throughput screening of large parametric spaces and leads to scalable (100 mL) synthetic strategies, are summarized. A rational selection of the alcohol is proposed. The influence of lights with defined wavelengths (222-690 nm) is investigated. It is found that lights with lower wavelengths favor the formation of smaller 5 nm NPs. Different kinetics and formation pathways are observed for different alcohols and for lights with different wavelengths. The sensitivity to various experimental parameters increases in the order glycerol < ethylene glycol < ethanol < methanol. New strategies for a rational fine size control, and to some extend shape control, are identified. The results lead to more sustainable and reproducible surfactant-free colloidal syntheses of NPs.
