Effects of fungicide application on physiological and molecular responses of grapevine (Vitis vinifera L.): a comparison between copper and sulfur fungicides applied alone and in combination with novel fungicides

Abstract:Background: Chemical products against fungi and oomycetes pose serious environmental issues. In the last decade, the use of less impacting active ingredients was encouraged to reduce chemical inputs in viticulture. In this study, the effect of different antifungal compounds on grapevine agronomic, physiological, and molecular responses in the vineyard was evaluated in addition to protection against powdery and downy mildews. Results: In 2 years and in two Vitis vinifera cultivars (Nebbiolo and Arneis), a conventional crop protection approach, based on traditional fungicides (sulfur and copper), was compared to combined strategies. A well-known resistance inducer (potassium phosphonate), Bacillus pumilus strain QST 2808 and calcium oxide, both active ingredients whose biological interaction with grapevine is poorly characterized, were applied in the combined strategies in association with chemical fungicides. Despite a genotype effect occurred, all treatments optimally controlled powdery and downy mildews, with minimal variations in physiological and molecular responses. Gas exchange, chlorophyll content and photosystem II efficiency increased in treated plants at the end of season, along with a slight improvement in the agronomic performances, and an activation of molecular defense processes linked to stilbene and jasmonate pathways. Conclusion: The disease control strategies based on potassium phosphonate, Bacillus pumilus strain QST 2808 or calcium oxide combined with traditional chemical compounds did not cause severe limitations in plant ecophysiology, grape quality, and productive yields. The combination of potassium phosphonate and calcium oxide with traditional fungicides can represent a valuable strategy for reducing copper and sulfur inputs in the vineyards, including those organically managed. © 2023 The Authors. Abstract:Efficient grapevine downy mildew control necessitates the implementation of anti-resistance strategies to ensure the ongoing efficacy of available substances and optimal disease control. With the gradual disappearance of multi-site fungicides from the market, reliance on single-site fungicides poses a long-term risk of selecting strains resistant to multiple modes of action. Challenges in disease management encompass selecting optimal spray programs and monitoring field population sensitivity. This study evaluated the efficacy of anti-resistance strategies, including two single-site fungicides (mandipropamid and oxathiapiprolin), on disease control and fungicide sensitivity through a combination of field trials and laboratory tests for the biological and molecular characterization of the pathogen populations over a three-year period (2019–2021). Mandipropamid, a cellulose synthase inhibitor, is used since a long time for downy mildew control, while oxathiapiprolin, an OxySterol Binding Protein homologue Inhibitor, was introduced recently. Field trials demonstrated effective disease control, even in the presence of mandipropamid-resistant strains (with G1105S/V mutations in PvCesA3 ) and revealed a pronounced selection and spread of resistance to both fungicides in the vineyard where disease pressure was higher. Characterizing pathogen strains remained a significant obstacle in sensitivity monitoring, hindering precise determination of resistance frequencies related to fungicide programs. Traditional techniques, in fact, lack the resolution required for high-throughput isolation and characterization of resistant individuals. To address this challenge, we propose utilizing flow cytometry and fluorescence-activated cell sorting on field sporangia populations, a method able to determine both the number of resistant isolates and isolate pathogen strains in a single assay.

Increasing vineyard sustainability: innovating a targeted chitosan-derived biocontrol solution to induce grapevine resistance against downy and powdery mildews

Abstract:The European Green Deal aims to reduce the pesticide use, notably by developing biocontrol products to protect crops from diseases. Indeed, the use of significant amounts of chemicals negatively impact the environment such as soil microbial biodiversity or groundwater quality, and human health. Grapevine ( Vitis vinifera ) was selected as one of the first targeted crop due to its economic importance and its dependence on fungicides to control the main damaging diseases worldwide: grey mold, downy and powdery mildews. Chitosan, a biopolymer extracted from crustacean exoskeletons, has been used as a biocontrol agent in many plant species, including grapevine, against a variety of cryptogamic diseases such as downy mildew ( Plasmopara viticola ), powdery mildew ( Erysiphe necator ) and grey mold ( Botrytis cinerea ). However, the precise molecular mechanisms underlying its mode of action remain unclear: is it a direct biopesticide effect or an indirect elicitation activity, or both? In this study, we investigated six chitosans with diverse degrees of polymerization (DP) ranging from low to high DP (12, 25, 33, 44, 100, and 470). We scrutinized their biological activities by evaluating both their antifungal properties and their abilities to induce grapevine immune responses. To investigate their elicitor activity, we analyzed their ability to induce MAPKs phosphorylation, the activation of defense genes and metabolite changes in grapevine. Our results indicate that the chitosans with a low DP are more effective in inducing grapevine defenses and possess the strongest biopesticide effect against B. cinerea and P. viticola . We identified chitosan with DP12 as the most efficient resistance inducer. Then, chitosan DP12 has been tested against downy and powdery mildews in the vineyard trials performed during the last three years. Results obtained indicated that a chitosan-based biocontrol product could be sufficiently efficient when the amount of pathogen inoculum is quite low and could be combined with only two fungicide treatments during whole season programs to obtain a good protection efficiency. On the whole, a chitosan-based biocontrol product could become an interesting alternative to meet the chemicals reduction targeted in sustainable viticulture.

Abstract:After being accidentally introduced from the USA at the end of the 19th century, downy mildew caused by Plasmopara viticola (Berk. et Curt.) Berlese et De Toni became one of the most damaging diseases affecting Vitis vinifera in Europe. Downy mildew causes both direct and indirect losses and can lead to severe reduction of yield. Our understanding of the life cycle and epidemiology of P. viticola has been recently altered by molecular studies that revealed that the overwintering inoculum (i.e., the oospores) does more than initiate disease, as was previously thought. A mechanistic model was developed for predicting the entire chain of processes leading to primary infections, and this primary infection model was linked to other models of secondary infection cycles. The model for primary infections defines the length of the primary inoculum season and a seasonal oospore dose consisting of several cohorts of oospores that progressively mature. The model was evaluated by means of Bayesian analysis in both Italy and eastern Canada, and showed high sensitivity, specificity, and accuracy both for potted plants and vineyards. Fungicide applications are necessary to control downy mildew because preventive agronomic practices are not very effective, including host resistance. The use of warning systems based on weather-driven models leads to a reduction in the use and cost of chemicals and a reduction in their environmental impact.

Abstract:Downy mildew, caused by the oomycete Plasmopara viticola, is one of the diseases causing the most severe economic losses to grapevine (Vitis vinifera) production. To date, the application of fungicides is the most efficient method to control the pathogen and the implementation of novel and sustainable disease control methods is a major challenge. RNA interference (RNAi) represents a novel biotechnological tool with a great potential for controlling fungal pathogens. Recently, a candidate susceptibility gene (VviLBDIf7) to downy mildew has been identified in V. vinifera. In this work, the efficacy of RNAi triggered by exogenous double-stranded RNA (dsRNA) in controlling P. viticola infections has been assessed in a highly susceptible grapevine cultivar (Pinot noir) by knocking down VviLBDIf7 gene. The effects of dsRNA treatment on this target gene were assessed by evaluating gene expression, disease severity, and development of vegetative and reproductive structures of P. viticola in the leaf tissues. Furthermore, the effects of dsRNA treatment on off-target (EF1α, GAPDH, PEPC, and PEPCK) and jasmonic acid metabolism (COI1) genes have been evaluated. Exogenous application of dsRNA led to significant reductions both in VviLBDIf7 gene expression, 5 days after the treatment, and in the disease severity when artificial inoculation was carried out 7 days after dsRNA treatments. The pathogen showed clear alterations to both vegetative (hyphae and haustoria) and reproductive structures (sporangiophores) that resulted in stunted growth and reduced sporulation. Treatment with dsRNA showed signatures of systemic activity and no deleterious off-target effects. These results demonstrated the potential of RNAi for silencing susceptibility factors in grapevine as a sustainable strategy for pathogen control, underlying the possibility to adopt this promising biotechnological tool in disease management strategies.

Abstract:Grapevine downy mildew (GDM) is one of the most serious diseases of grapevines. With limitations in the use of copper-based products imposed for organic agriculture by the European Union, research for alternatives is encouraged. The aim of this research was to follow a 2-year trial to evaluate the control of GDM using some alternative compounds, and to determine their effects on shoot growth, plant photosynthesis, and grape quality and quantity. Under low disease pressure, Bordeaux mixture, copper hydroxide, laminarin combined with low copper, and 0.5 and 0.8% chitosan had the lowest GDM incidence, reduced on leaves by 96, 95, 75, 56, and 81%, respectively, compared with the untreated control in the last survey. With high disease pressure, Bordeaux mixture, laminarin combined with Saccharomyces extracts, and 0.5 and 0.8% chitosan had the lowest GDM incidence, reduced on grape by 86, 37, 66, and 75%, respectively, compared with the untreated control in the survey of mid-July. Chitosan at 0.8% lowered net photosynthesis, due to reduced stomatal conductance, leaf area, and dry weight, with no negative effects observed on the quantity of the grape berries and the quality parameters of their juice. Among the alternatives to copper, chitosan provided the best GDM protection and reduced the vigor of the vegetation, inducing physiological changes without negative effects on grape production.

Abstract:Plasmopara viticola (Berk.et Curtis) Berl. Et de Toni is the pathogen that causes grape downy mildew, which is an airborne disease that severely affects grape yield and causes huge economic losses. The usage of effective control methods can reduce the damage to plants induced by grape downy mildew. Biocontrol has been widely used to control plant diseases due to its advantages of environmental friendliness and sustainability. However, to date, only a few comprehensive reviews on the biocontrol of grape downy mildew have been reported. In this review, we summarize the biological characteristics of P. viticola and its infection cycle, followed by a detailed overview of current biocontrol agents, including bacteria and fungi that could be used to control grape downy mildew, and their control effects. Furthermore, potential control mechanisms of biocontrol agents against grape downy mildew are discussed. Lastly, suggestions for future research on the biocontrol of grape downy mildew are provided. This review provides the basis for the application of grape downy mildew biocontrol.

Abstract:Several pests and diseases have grapevine as their favourite host and the vineyard as preferred environment, so an intensive pesticide schedule is usually required to meet qualitative and quantitative production standards. The need to prevent the negative impact of synthetic chemical pesticides on human health and the environment and the consumer expectations in term of chemical residues in food stimulated the research of innovative tools and methods for sustainable pest management. The research project PURE (www.pure-ipm.eu) was a Europe-wide framework, which demonstrated that several solutions are now available for the growers and evaluated several new alternatives that are under development or almost ready for being applied in practice. Although the use of resistant/tolerant varieties is not yet feasible in several traditional grape growing areas, at least part of the synthetic chemical pesticides can be substituted with biocontrol agents to control pests and pathogens and/or pheromone mating disruption, or the number of treatments can be reduced by the use of decision support systems, which identify the optimal timing for the applications. This review presents the state of the art and the perspectives in the field of grapevine protection tools and strategies.

Abstract:Background: Downy mildew, caused by Plasmopara viticola, is one of the most severe diseases of grapevine and is commonly controlled by fungicide treatments. The beneficial microorganism Trichoderma harzianum T39 (T39) can induce resistance to downy mildew, although the molecular events associated with this process have not yet been elucidated in grapevine. A next generation RNA sequencing (RNA-Seq) approach was used to study global transcriptional changes associated with resistance induced by T39 in Vitis vinifera Pinot Noir leaves. The long-term aim was to develop strategies to optimize the use of this agent for downy mildew control. Results: More than 14.8 million paired-end reads were obtained for each biological replicate of T39-treated and control leaf samples collected before and 24 h after P. viticola inoculation. RNA-Seq analysis resulted in the identification of 7,024 differentially expressed genes, highlighting the complex transcriptional reprogramming of grapevine leaves during resistance induction and in response to pathogen inoculation. Our data show that T39 has a dual effect: it directly modulates genes related to the microbial recognition machinery, and it enhances the expression of defence-related processes after pathogen inoculation. Whereas several genes were commonly affected by P. viticola in control and T39-treated plants, opposing modulation of genes related to responses to stress and protein metabolism was found. T39-induced resistance partially inhibited some disease-related processes and specifically activated defence responses after P. viticola inoculation, causing a significant reduction of downy mildew symptoms. Conclusions: The global transcriptional analysis revealed that defence processes known to be implicated in the reaction of resistant genotypes to downy mildew were partially activated by T39-induced resistance in susceptible grapevines. Genes identified in this work are an important source of markers for selecting novel resistance inducers and for the analysis of environmental conditions that might affect induced resistance mechanisms.

Abstract:The persistent use of copper-based fungicides to control grapevine downy mildew (GDM) still represents risks to both human health and the environment, especially in organic production. This heavy metal can affect wine quality due to residues' presence on grape bunches. Implementing sustainable viticultural practices to control diseases is crucial for the wine industry. This work aimed to evaluate chitosan effectiveness toward GDM on a large scale, over a period of three years (2019–2021), and in three distinct vineyards with different environmental conditions. Three GDM management strategies were tested: application of 0.5% chitosan individually; in alternation with copper; and during the last year, in combination with low rate of copper, using conventional strategies and untreated plants as controls. Over three consecutive seasons, chitosan treatments effectively reduced GDM McKinney Index on leaves and bunches under different environmental conditions. On bunches, chitosan alone was less effective than copper, when applied after copper had the same effectiveness, and combined with copper was more effective than copper standard. In 2020 and 2021, the analysis of cooper amount detected in berries of two cultivars showed that all strategies allowed to decrease the amount of copper residue in berries. The application of chitosan alone or in alternation with copper was able to reduce this amount as untreated control. This large scale investigation suggests chitosan as alternative that can integrate and eventually replace copper application against GDM. Further studies to validate these strategies in different environments, disease pressure conditions and cultivar are fundamental towards achieving green and sustainable solutions for GDM management strategies.

Abstract:This study examined the potential use of three bacterial strains-Paraburkholderia sp. strain CRV74, Pseudomonas sp. strain CRV21, and Acinetobacter sp. strain CRV19-as biocontrol agents of Botrytis cinerea in grapevine. These strains were selected for their ability to inhibit B. cinerea growth in vitro and used in field conditions for the control of grey mould symptoms in 'Glera' grapes. To this end, after inoculating these microorganisms onto plants sprayed with B. cinerea spores, the final yield, the physicochemical characteristics of the must, disease incidence, and the possible influence on the expression of plant-defence proteins were evaluated. Strain CRV21 resulted as being the most effective in combating grey mould (-20% of disease incidence). Although yield was not affected, significantly different values of total soluble solids content was observed. Additionally, a significant up-regulation of the genes PR-1, PR-5, β-1,3-glucanase, and class III chitinase was observed. These findings highlight the potential application of strains with anti-botrytis activity as sustainable alternatives to chemical defence for the control of this pathogen.
A new digital technology to reduce fungicide use in vineyards

Abstract:There is a growing demand for technologies able to decrease the environmental impact of agricultural activities without penalizing quali-quantitative characteristics of productions. In the case of viticulture, one of the key problems is represented by the spray drift during fungicide treatments. The diffusion in operational farming contexts of technologies based on variable-rate and recycling tunnel sprayers is often limited by their cost and, for the latter, by their size and lower maneuverability, representing clear disadvantages especially in case of small farms or in hilly and mountain areas. We present a new digital technology implemented in a mobile app that supports the reduction of both the number of treatments and the amount of fungicide distributed per treatment. The technology is based (i) on an alert system that prevents unneeded treatments in case of no risk of infection and (ii) on the quantification of the optimal amounts of active ingredients and dilution water based on the sprayer type/settings and on leaf area index values estimated with a common smartphone. An internal database allows to adjust (in case of need) the active ingredient dose to assure full compliance with product's legal requirements. In case of heterogeneity in leaf area index values inside the vineyard, prescription maps are generated. Results from a 2-year case study in a vineyard in northern Italy are shown, where the system allowed to reduce by 26.4 % and 27.4 % (mean of two years), respectively, the seasonal amounts of fungicides and dilution water, and by 43.8 % the copper content in must. The high usability of the technology proposed (just a common smartphone is needed) and the fact that it does not require updating the farm machine park highlights the suitability of the proposed solution for operational farming conditions, including premium wine production districts often characterized by small farms in hilly areas.

In vitro evaluation of the efficacy of 8-hydroxyquinoline derivatives for the control of Phaeomoniella chlamydospora, the causative agent of Petri disease in grapevines

Abstract:Aim: This study evaluates the in vitro efficacy of 8-hydroxyquinoline (8HQ) derivatives in controlling the phytopathogenic fungus Phaeomoniella chlamydospora. Methods and results: The in vitro tests assessed the susceptibility to the minimum inhibitory concentration (MIC), checkerboard assay, mycelial growth (MG) inhibition, and EC50 determination. Among the seven agricultural fungicides tested, tebuconazole (TEB) displayed the lowest MIC, 1.01 µg mL-1, followed by captan (CAP), thiophanate methyl (TM), and mancozeb with MICs of 4.06, 5.46, and 10.62 µg mL-1, respectively. The 8HQ derivatives used in this study were clioquinol and PH 151 (PH) with MICs of 1.09 and 2.02 µg mL-1, respectively. PH associated with TEB and CAP showed synergism and inhibited 95.8% of MG at the highest dose. TEB inhibited 100% of MG at the three highest doses, while associated with PH exhibited the lowest EC50 (0.863 + 0.0381 µg mL-1). Conclusions: We concluded that the 8HQ derivatives tested controlled effectively the P. chlamydospora in vitro. PH associated with CAP and TEB exhibited a synergistic effect. The association between PH and TM was considered indifferent. Impact statement: This study expands the list of active ingredients tested against P. chlamydospora, with the PH 151 and clioquinol derivatives being tested for the first time. The in vitro efficacy and synergistic action with other fungicides suggest a potential use as a grapevine wound protectant. This association makes it possible to reduce doses and increase the potency of both drugs, reducing the risk of resistance development and harm to humans and the environment.
A Plant Extract Acts Both as a Resistance Inducer and an Oomycide Against Grapevine Downy Mildew

Abstract:Protecting vineyards from cryptogamic diseases such as downy mildew, caused by Plasmopara viticola, generally requires a massive use of phytochemicals. However, the issues on unintentional secondary effects on environment and human health, and the occurrence of P. viticola resistant strains, are leading to the development of alternative strategies, such as the use of biocontrol products. In this paper, we evidenced the ability of a plant extract to protect grapevine from P. viticola. Further experiments carried out both on cell suspensions and on plants revealed that plant extract activates typical defense-related responses such as the production of H2O2, the up-regulation of genes encoding pathogenesis-related proteins and stilbene synthase, as well as the accumulation of resveratrol or its derivative piceid. We also brought to light a strong direct effect of PE on the release and motility of P. viticola zoospores. Furthermore, we found out that PE application left dried residues on leaf surface, impairing zoospores to reach stomata. Altogether, our results highlight the different modes of action of a new biocontrol product able to protect grapevine against downy mildew.

Quantification of grapevine yield losses as a function downy mildew severity on foliage and cluster

Abstract:To quantify the relationship between grapevine disease severity and crop losses at the plant scale, we conducted a three-year field experiment at two sites near Bordeaux (France), surveying mildews and rots on both foliage and clusters. A first set of analysis indicated that only downy mildew (DM) significantly affects yield (mass of grape clusters harvested) in our experimental conditions. We then leverage this situation to model the relationship between DM severity (measured by standardized AUDPC) and yield losses at vine stock scale. For AUDPC ranging from 0 to 10%, an increase of the severity of DM of 1% on the clusters decrease yield by 2% regardless of years and sites. These values ranges from 1.1% to 9% when considering the severity of DM on the foliage, depending on sites and years. This variability was correlated with a moisture risk index calculated between crop stages inflorescences visibles to pre-ripening of the berries. An increase of the severity of DM of 1% on the foliage decreases yield by 1.2% during dry year (quantile 10% of the index), by 3.3% during intermediate year (median the index) and by 7.3% during wet year (quantile 90% of the index). Regarding quality variables measuring grape ripeness on the must, we found that an increase of the severity of DM of 1% on the foliage decrease the potential alcohol content from 0.014 to 0.18 % vol depending on years while few effects on the pH were detected.

Abstract:Black rot (Guignardia bidwellii) and downy mildew (Plasmopara viticola) are two major grapevine diseases against which the development of efficient biocontrol solutions is required in a context of sustainable viticulture. This study aimed at evaluating and comparing the efficacy and modes of action of bacterial culture supernatants from Bacillus velezensis Buz14 and B. ginsengihumi S38. Both biocontrol agents (BCA) were previously demonstrated as highly effective against Botrytis cinerea in grapevines. In semi-controlled conditions, both supernatants provided significant protection against black rot and downy mildew. They exhibited antibiosis against the pathogens by significantly decreasing G. bidwellii mycelial growth, but also the release and motility of P. viticola zoospores. They also significantly induced grapevine defences, as stilbene production. The LB medium, used for the bacterial cultures, also showed partial effects against both pathogens and induced plant defences. This is discussed in terms of choice of experimental controls when studying the biological activity of BCA supernatants. Thus, we identified two bacterial culture supernatants as new potential biocontrol products exhibiting multi-spectrum antagonist activity against different grapevine key pathogens and having a dual mode of action.
Effect of fungicides and biological products in controlling rust and gray mold in grapevines

Abstract:Grape rust and gray mold are diseases in grapevines with high destructive potential. In this study, the effectiveness of different fungicides and biological products in controlling these diseases was evaluated using the cultivar “Niágara Rosada” in the municipality of Louveira-SP, from February to April 2017. A randomized block design was used, with 10 treatments and four replicates and five plants per plot. Each treatment was applied nine times at weekly intervals, with application volumes in the range of 400–1000 L/ha. The evaluation criterion for rust was the severity, based on the percentage of leaf area affected by the disease (0 to 100%), whereas gray mold was evaluated using the incidence of diseased bunches (0 to 100%). All treatments were effective in controlling both diseases and significantly reduced the disease incidence in the experimental plot compared with that in the control plots. The best rust control was obtained by treating the plants with boscalide+pyraclostrobin (1 L c.p./ha), which was superior to fluxapyroxade+pyraclostrobin (0.5 L) + Bacillus amyloliquefaciens (0.5 kg), B. amyloliquefaciens (0.5 kg), Melaleuca alternifolia essential oil (1.5 L), and dimoxystrobin+boscalide (1 L). The plants treated with fluxapyroxad+pyraclostrobin (0.5 L) + B. amyloliquefaciens (0.5 kg) had a lower incidence of gray mold than did those treated with dimoxystrobin+boscalide (1 L), boscalide+kresoxim methyl (0.5 L), and piremethanil (1.5 L). These results suggest that the use of M. alternifolia essential oil and B. amyloliquefaciens may represent an important tool for the management of rust and gray mold in grapevines.
Integration of mathematical modeling and target-based application of biocontrol agents for the control of Botrytis cinerea in vineyards

Abstract:Background: Biocontrol agents (BCAs) are alternatives to synthetic fungicides with low risk to the environment and human health. Although several studies on the biocontrol of gray mold in vineyards have been performed, it is necessary to improve the usage of BCAs in fields conditions. Therefore, in the present study, BCAs were used both in calendar-based [based on four growth stages (GSs), i.e., flowering, pre-bunch closure, veraison, and before harvest] and predictive model-based strategies (only when Botrytis cinerea infection risk was predicted by the model). The BCAs applied during the seasons were selected considering the grapevine GSs. Treatments performed with BCAs were compared with synthetic fungicide treatments and an untreated control. The trials were conducted in three experimental vineyards with four epidemics. To evaluate the level of gray mold control of each treatment, disease severity was assessed at harvest and the presence of latent infection was evaluated. Results: The integrative use of the predictive model and BCAs provided satisfactory levels of gray mold control, with gray mold severity levels significantly lower (P < 0.001) than those of the untreated control, which had severity values (< 7%) similar to those observed with synthetic fungicides following both calendar and model-based strategies. Conclusions: The integrative use of the predictive model and BCAs represents a valid alternative to conventional methods of gray mold control in vineyards, with more than 75% reduction in fungicide usage. © 2024 Society of Chemical Industry.
