Immune and Protective Effects of Subunit Vaccines from S-domain or P-domain in Capsid Protein Against Nervous Necrosis Virus in Pearl Gentian Grouper

Abstract:Capsid protein (CP), only structural protein of nervous necrosis virus (NNV), is a major target for viral encephalopathy and retinopathy (VER) prevention and virus detection. It is also crucial for precise vaccine design. In the present study, CP and its S-domain, P-domain were recombinantly expressed as subunit vaccines candidates. Then the recombinant proteins were immunized to pearl gentian grouper (Epinephelus lanceolatus x Epinephelus fuscoguttatus). After immunization, the vaccine-induced humoral and cellular immunity of grouper was measured. After challenge with 106.1, 107.1and 108.1 TCID50/fish of NNV, respectively, the immune protection provoked by the vaccines was also evaluated. The results showed that the expressed recombinant S-domain (rS), P-domain (rP), and CP (rCP) were proved to be high immunogenicity. After immunization, the secretory immunoglobulins (sIgM)+ lymphocytes proliferated significantly and peaked at the 3rd week, with the percentage in the head kidney was 25.1 +/- 2.1% in the rS group, 22.1 +/- 2.2% in rP group, 29.9 +/- 3.0% in the rCP group, in spleen it was 18.4 +/- 1.9% in rS group, 17.3 +/- 2.3% in rP group, 21.5 +/- 1.7% in rCP group. Total antibodies peaked at 4th week and were highest in rCP group, rCP-specific antibodies in serum peaked at 3rd week and were highest in rP group and neutralizing antibodies at 3rd week were highest in rCP group. Six immune-related genes were significantly up-regulated in the spleen and head kidney at the 1st/3rd/5th days in all groups. For protection results with three titers of virus (106.1, 107.1and 108.1 TCID50/fish), the relative percent survival rate (RPS) was 46.66%, 40% and 33.33% in rS group, 60%, 55% and 50% in rP group, and 86.66%, 75% and 70% in rCP group, respectively. The percentage of sIgM+ lymphocytes in spleen and head kidney decreased at 7th day post-challenge compared to pre-challenge in all groups and increased significantly at 14th post challenge day, the level of antibodies in the serum has decreased, and the virus copies in brain and eye on 7th day showed significantly lower than control group. In addition, the vaccines had no side effects on the growth of fish. In conclusion, the results showed S-domain and P-domain induced strong immune effect and good protection for grouper, the data provided possibility for precise and concise vaccine design and production against NNV.
