Expression of Vp2e and Vp3 Genes from Marine Birnavirus (MABV) in Insect Cells

Abstract:Marine birnavirus (MABV), a RNA virus with two segments of double stranded RNA in genome, may cause serious ascites disease for many marine organisms. The cDNA of fragment A from MABV Y-6 strain was amplified by RT-PCR. A 714 bp fragment of structural protein (VP3) and a 642 bp fragment (vp2e) of capsid protein VP2e, in which all epitopes were concentrated, were cloned. These two fragments were subsequently constructed into recombinant virus expression vectors through transposition, and transfected into insect cells of Tn-5B1-4 for expression of the fusion proteins, EGFP-VP2e and EGFP-VP3. SDS-PAGE and film scanning showed that VP2e and VP3 were expressed efficiently and the percentages of the fusion proteins were accumulated to 15.8% and 8.2%, respectively. VP2e fusion protein was confirmed by Western blot using His-tag monoantibody and polyclonal antibody of MABV. Also, VP3 fusion protein was confirmed by Western blotting with His-tag monoantibody and polyclonal antibody against VP3. Western blottig assay indicated they were expressed correctly and had an excellent property as antigen. These two fusion proteins were also purified using the His Ni-NTA column. This experiment lays a basis for the further studies of the MABV structural proteins and the development of vaccines against MABV.
