Abstract:Synthetic Biology is a growing field which holds great promise in several areas of application. However, it also poses serious risks for human health and the environment that must be anticipated in order to develop effective prevention and management measures. Here, the current situation on biosafety and biosecurity in this scientific field is reviewed. Biosafety concerns mainly relate to the damaging effects for workers and the environment that could result from accidental interactions with dangerous biological agents. On the other hand, biosecurity risks refer to the potential misuses of Synthetic Biology, such as bioterrorism, biowarfare or bioattacks that could derive from the genetic engineering of organisms. In this paper, the specific challenges posed by Synthetic Biology are discussed, and perspectives in the development of measures to guide the safe advance of this discipline are presented.

Abstract:In 2014, an international group of scholars from various fields analysed the “societal dimensions" of synthetic biology in an interdisciplinary summer school. Here we report and discuss the biologists’ observations on the general perception of synthetic biology by non-biologists who took part in this event. Most attendees mainly associated synthetic biology with contributions from the best-known public figures of the field, rarely mentioning other scientists. Media extrapolations of those contributions appeared to have created unrealistic expectations and irrelevant fears that were widely disconnected from current research in synthetic biology. Abstract:This structured literature analysis aims to map the current, emerging, and predicted future of synthetic biology (SB) by putting the focus on the implied conceptual, societal, and ethical challenges. The central objective of the analysis is to provide an initial systematization of the ethical and socio-scientific debate on SB by structuring and categorizing widely discussed issues within the debate in recent years. Starting with the quest for possible definitions, issues of biosafety and biosecurity are emphasized. Furthermore, the focus is on the more conceptual challenges of SB, including the relationship between natural and synthetic, or concepts of life and living. From the very beginning, one specific characteristic of SB has been a strong entanglement with different forms of public participation. In some respects SB has already taken a leading position in claiming and orchestrating itself as an integrative and participatory discipline. After addressing SB as an emerging biotechnology at the interface between science and society, a venture is initiated to focus on the possible regulatory and governmental challenges which are entangled in SB.

Abstract:Synthetic biology has immense potential to ameliorate widespread environmental damage. The promise of such technology could, however, be argued to potentially risk the public, industry or governments not curtailing their environmentally damaging behavior or even worse exploit the possibility of this technology to do further damage. In such cases, there is the risk of a worse outcome than if the technology was not deployed. This risk is often couched as an objection to new technologies, that the technology produces a moral hazard. This paper describes how to navigate a moral hazard argument and mitigate the possibility of a moral hazard. Navigating moral hazard arguments and mitigating the possibility of a moral hazard will improve the public and environmental impact of synthetic biology.

Abstract:Synthetic biology is designing and creating biological tools and systems for useful purposes. It uses knowledge from biology, such as biotechnology, molecular biology, biophysics, biochemistry, bioinformatics, and other disciplines, such as engineering, mathematics, computer science, and electrical engineering. It is recognized as both a branch of science and technology. The scope of synthetic biology ranges from modifying existing organisms to gain new properties to creating a living organism from non-living components. Synthetic biology has many applications in important fields such as energy, chemistry, medicine, environment, agriculture, national security, and nanotechnology. The development of synthetic biology also raises ethical and social debates. This article aims to identify the place of ethics in synthetic biology. In this context, the theoretical ethical debates on synthetic biology from the 2000s to 2020, when the development of synthetic biology was relatively faster, were analyzed using the systematic review method. Based on the results of the analysis, the main ethical problems related to the field, problems that are likely to arise, and suggestions for solutions to these problems are included. The data collection phase of the study included a literature review conducted according to protocols, including planning, screening, selection and evaluation. The analysis and synthesis process was carried out in the next stage, and the main themes related to synthetic biology and ethics were identified. Searches were conducted in Web of Science, Scopus, PhilPapers and MEDLINE databases. Theoretical research articles and reviews published in peer-reviewed journals until the end of 2020 were included in the study. The language of publications was English. According to preliminary data, 1,453 publications were retrieved from the four databases. Considering the inclusion and exclusion criteria, 58 publications were analyzed in the study. Ethical debates on synthetic biology have been conducted on various issues. In this context, the ethical debates in this article were examined under five themes: the moral status of synthetic biology products, synthetic biology and the meaning of life, synthetic biology and metaphors, synthetic biology and knowledge, and expectations, concerns, and problem solving: risk versus caution.

Abstract:In our view, synthetic biology is an extension of the continuum of genetic science that has been used safely for more than 40 years by the biotechnology industry in the development of commercial products. Examples of synthetic biology use by biotechnology companies illustrate the potential to substantially reduce research and development time and to increase speed to market. Improvements in the speed and cost of DNA synthesis are enabling scientists to design modified bacterial chromosomes that can be used in the production of renewable chemicals, biofuels, bioproducts, renewable specialty chemicals, pharmaceutical intermediates, fine chemicals, food ingredients, and health care products. Regulatory options should support innovation and commercial development of new products while protecting the public from potential harms.

The whack-a-mole governance challenge for AI-enabled synthetic biology: literature review and emerging frameworks

Abstract:AI-enabled synthetic biology has tremendous potential but also significantly increases biorisks and brings about a new set of dual use concerns. The picture is complicated given the vast innovations envisioned to emerge by combining emerging technologies, as AI-enabled synthetic biology potentially scales up bioengineering into industrial biomanufacturing. However, the literature review indicates that goals such as maintaining a reasonable scope for innovation, or more ambitiously to foster a huge bioeconomy don't necessarily contrast with biosafety, but need to go hand in hand. This paper presents a literature review of the issues and describes emerging frameworks for policy and practice that transverse the options of command-and control, stewardship, bottom-up, and laissez-faire governance. How to achieve early warning systems that enable prevention and mitigation of future AI-enabled biohazards from the lab, from deliberate misuse, or from the public realm, will constantly need to evolve, and adaptive, interactive approaches should emerge. Although biorisk is subject to an established governance regime, and scientists generally adhere to biosafety protocols, even experimental, but legitimate use by scientists could lead to unexpected developments. Recent advances in chatbots enabled by generative AI have revived fears that advanced biological insight can more easily get into the hands of malignant individuals or organizations. Given these sets of issues, society needs to rethink how AI-enabled synthetic biology should be governed. The suggested way to visualize the challenge at hand is whack-a-mole governance, although the emerging solutions are perhaps not so different either.

Abstract:Background: The integration of Artificial Intelligence (AI) with synthetic biology is driving unprecedented progress in both fields. However, this integration introduces complex biosecurity challenges. Addressing these concerns, this article proposes a specialized biosecurity risk assessment process designed to evaluate the incorporation of AI in synthetic biology. Methods: A set of tailored tools and methodology was developed for conducting biosecurity risk assessments of AI language models used for synthetic biology. These resources were developed to guide risk management professionals through a systematic process of identifying, evaluating, and mitigating potential risks. Results: The tools and methodology provided offer a structured approach to risk assessment, enabling risk management professionals to comprehensively analyze the biosecurity implications of AI applications in synthetic biology. They facilitate the identification of potential risks and the development of effective mitigation strategies. An example of a risk assessment performed on the large language model "ChatGPT 4.0" is provided here. Conclusion: AI's role in synthetic biology is rapidly expanding; thus, establishing proactive and secure practices is crucial. The biosecurity risk assessment tools and methodology presented here are the first provided in the literature and will be instrumental steps toward the responsible integration of AI in synthetic biology. By adopting these resources, the biorisk management community can effectively navigate and manage the biosecurity challenges posed by AI, ensuring its responsible and secure application in the field of synthetic biology.

Abstract:Researchers in xenobiology, a subdiscipline of synthetic biology, aim to build a 'second nature' with nucleic acid analogues, termed Xeno-nucleic acids (XNA). They promise biosafe technologies, based on the impossibility of transferring genetic material to other organisms and controlling the proliferation of genetically modified microorganisms. Proponents of xenobiology have employed metaphors and narratives that represent the separation of synthetic life from DNA-based, constituting a safer space for the exploration and navigation of virtual biological worlds. Based on interviews with synthetic biologists and participant observation in a synthetic biology laboratory, I argue that the reconfiguration of nature that xenobiologists seek is inspired by the vision of design and governance laid out in the 1975 Asilomar conference, so normative aims of safety are co-produced with visions of unnaturalness. I interrogate the types of limits that xenobiologists aim to cross, to propose that they conceive limits as pushing beyond what is biologically plausible, finding the challenge motivating. I show that the division between the natural and the unnatural is not clearly established as xenobiologists portray. In giving priority to safety as the determinant of the permissibility of new technologies, who gets to define nature and its limits remains restricted to scientists.

Abstract:Synthetic biology enthusiasts often tout the emerging field for its present and future potential to revolutionize the life sciences. In the biosecurity arena, which has received considerable government and non-government attention, many are concerned that synthetic biology may prove to be an easier and cheaper way to conduct bioterrorism. To evaluate these claims, this article will focus on contrasting two different frameworks that have been used for understanding the development, diffusion and adoption of synthetic biology. In contrasting these frameworks, I will draw on examples from biotechnology and information technology because they are often used as analogies in synthetic biology discussions. I conclude that the critical elements for successful development, transfer, and use of synthetic biology methodologies and tools for harm are not purely material or technical, but involve important social dimensions that underpin technical work, requiring time, teams of experts, appropriate political, legal, and funding structures, and the development of new (still unknown) techno-organizational processes. To date, there have been few studies that have explored these socio-technical mechanisms of synthetic biology diffusion through in-depth examination at a micro and macro level. However, by having a more nuanced understanding of various synthetic biology approaches and how they are (or are not) able to travel easily to new settings, one can create a more refined spectrum of factors shaping threats from state and non-state actors related to synthetic biology. This article ends by outlining new research agendas important to support and pursue in order to improve biosecurity policymaking.

Abstract:The Safe-by-Design approach in synthetic biology holds the promise of designing the building blocks of life in an organism guided by the value of safety. This paves a new way for using biotechnologies safely. However, the Safe-by-Design approach moves the bulk of the responsibility for safety to the actors in the research and development phase. Also, it assumes that safety can be defined and understood by all stakeholders in the same way. These assumptions are problematic and might actually undermine safety. This research explores these assumptions through the use of a Group Decision Room. In this set up, anonymous and non-anonymous deliberation methods are used for different stakeholders to exchange views. During the session, a potential synthetic biology application is used as a case for investigation: the Food Warden, a biosensor contained in meat packaging for indicating the freshness of meat. Participants discuss what potential issues might arise, how responsibilities should be distributed in a forward-looking way, who is to blame if something would go wrong. They are also asked what safety and responsibility mean at different phases, and for different stakeholders. The results of the session are not generalizable, but provide valuable insights. Issues of safety cannot all be taken care of in the R&D phase. Also, when things go wrong, there are proximal and distal causes to consider. In addition, capacities of actors play an important role in defining their responsibilities. Last but not least, this research provides a new perspective on the role of instruction manuals in achieving safety.

Abstract:The basic chemical constitution of all living organisms in the context of carbon-based chemistry consists of a limited number of small molecules and polymers. Until the twenty-first century, biology was mainly an analytical science and has now reached a point where it merges with engineering science, paving the way for synthetic biology. One of the objectives of synthetic biology is to try to change the chemical compositions of living cells, that is, to create an artificial biological diversity, which in turn fosters a new sub-field of synthetic biology, xenobiology. In particular, the genetic code in living systems is based on highly standardized chemistry composed of the same "letters" or nucleotides as informational polymers (DNA, RNA) and the 20 amino acids which serve as basic building blocks for proteins. The universality of the genetic code enables not only vertical gene transfer within the same species but also horizontal gene transfer across biological taxa, which require a high degree of standardization and interconnectivity. Although some minor alterations of the standard genetic code are found in nature (e.g., proteins containing non-conical amino acids exist in nature, and some organisms use alternated coding systems), all structurally deep chemistry changes within living systems are generally lethal, making the creation of artificial biological system an extremely difficult challenge.In this context, one of the great challenges for bioscience is the development of a strategy for expanding the standard basic chemical repertoire of living cells. Attempts to alter the meaning of the genetic information stored in DNA as an informational polymer by changing the chemistry of the polymer (i.e., xeno-nucleic acids) or by changes in the genetic code have already yielded successful results. In the future this should enable the partial or full redirection of the biological information flow to generate "new" version(s) of the genetic code derived from the "old" biological world.In addition to the scientific challenges, the attempt to increase biochemical diversity also raises important ethical and philosophical issues. Although promotors of this branch of synthetic biology highlight the many potential applications to come (e.g., novel tools for diagnostics and fighting infection diseases), such developments could also bring risks affecting social, political, and other structures of nearly all societies.

Abstract:The convergence of artificial intelligence (AI) and synthetic biology is rapidly accelerating the pace of biological discovery and engineering. AI techniques, such as large language models and biological design tools, are enabling the automated design, build, test, and learning cycles for engineered biological systems. This convergence promises to democratize synthetic biology and unlock novel applications across domains from medicine to environmental sustainability. However, it also poses significant risks around reliability, dual use, and governance. The opacity of AI models, the deskilling of workforces, and the outdated nature of current regulatory frameworks present challenges in ensuring responsible development. Urgent attention is needed to update governance structures, integrate human oversight into increasingly automated workflows, and foster a culture of responsibility among the growing community of bioengineers. Only by proactively addressing these issues can we realize the transformative potential of AI-driven synthetic biology while mitigating its risks.

Abstract:The ability to manipulate living organisms is at the heart of a range of emerging technologies that serve to address important and current problems in environment, energy, and health. However, with all its complexity and interconnectivity, biology has for many years been recalcitrant to engineering manipulations. The recent advances in synthesis, analysis, and modeling methods have finally provided the tools necessary to manipulate living systems in meaningful ways and have led to the coining of a field named synthetic biology. The scope of synthetic biology is as complicated as life itself-encompassing many branches of science and across many scales of application. New DNA synthesis and assembly techniques have made routine customization of very large DNA molecules. This in turn has allowed the incorporation of multiple genes and pathways. By coupling these with techniques that allow for the modeling and design of protein functions, scientists have now gained the tools to create completely novel biological machineries. Even the ultimate biological machinery-a self-replicating organism-is being pursued at this moment. Biosecurity Assessments for Emerging Transdisciplinary Biotechnologies: Revisiting Biodefense in an Age of Synthetic Biology

Abstract:Introduction: Rapid advances in biotechnologies and transdisciplinary research are enhancing the ability to perform full-scale engineering of biology, contributing to worldwide efforts to create bioengineered plants, medicines, and commodities, which promise sustainability and innovative properties. Objective: This rapidly evolving biotechnology landscape is prompting focused scrutiny on biosecurity frameworks in place to mitigate harmful exploitation of biotechnology by state and non-state actors. Concerns about biosafety and biosecurity of engineering biology research have existed for decades as views about how advances in this and associated fields might provide new capabilities to malicious actors. This article considers biosecurity concerns using examples of research advances in engineering biology. Methods: The authors explore risk assessment and mitigation of transdisciplinary biotechnology research and development, using the framework developed in the National Academies' study on Biodefense in an Age of Synthetic Biology. Results: The Synthetic Biology Assessment Framework focuses on risks of using advanced approaches and technologies to enhance or create novel pathogens and toxins. The field of engineering biology continues to advance at a pace that challenges current risk assessment frameworks. Conclusions: This framework likely is sufficient to assess new science and technology advances affecting conventional biological agents. However, the risk assessment framework may have limited applicability for technologies that are not usable with conventional biological agents and result in economic or broader national security concerns. Finally, the vast majority of discourse has been focused primarily on risks rather than benefits, and analyzing both in future evaluations is critical to balancing scientific progress with risk reduction.

Abstract:Tremendous advances in the field of synthetic biology have been witnessed in multiple areas including life sciences, industrial development, and environmental bio-remediation. However, due to the limitations of human understanding in the code of life, any possible intended or unintended uses of synthetic biology, and other unknown reasons, the development and application of this technology has raised concerns over biosafety, biosecurity, and even cyberbiosecurity that they may expose public health and the environment to unknown hazards. Over the past decades, some countries in Europe, America, and Asia have enacted laws and regulations to control the application of synthetic biology techniques in basic and applied research and this has resulted in some benefits. The outbreak of the COVID-19 caused by novel coronavirus SARS-CoV-2 and various speculations about the origin of this virus have attracted more attention on bio-risk concerns of synthetic biology because of its potential power and uncertainty in the synthesis and engineering of living organisms. Therefore, it is crucial to scrutinize the control measures put in place to ensure appropriate use, promote the development of synthetic biology, and strengthen the governance of pathogen-related research, although the true origin of coronavirus remains hotly debated and unresolved. This article reviews the recent progress made in the field of synthetic biology and combs laws and regulations in governing bio-risk issues. We emphasize the urgent need for legislative and regulatory constraints and oversight to address the biological risks of synthetic biology.

A Systematic Review of the Criminogenic Potential of Synthetic Biology and Routes to Future Crime Prevention

Abstract:Synthetic biology has the potential to positively transform society in many application areas, including medicine. In common with all revolutionary new technologies, synthetic biology can also enable crime. Like cybercrime, that emerged following the advent of the internet, biocrime can have a significant effect on society, but may also impact on peoples' health. For example, the scale of harm caused by the SARS-CoV-2 pandemic illustrates the potential impact of future biocrime and highlights the need for prevention strategies. Systematic evidence quantifying the crime opportunities posed by synthetic biology has to date been very limited. Here, we systematically reviewed forms of crime that could be facilitated by synthetic biology with a view to informing their prevention. A total of 794 articles from four databases were extracted and a three-step screening phase resulted in 15 studies that met our threshold criterion for thematic synthesis. Within those studies, 13 exploits were identified. Of these, 46% were dependent on technologies characteristic of synthetic biology. Eight potential crime types emerged from the studies: bio-discrimination, cyber-biocrime, bio-malware, biohacking, at-home drug manufacturing, illegal gene editing, genetic blackmail, and neuro-hacking. 14 offender types were identified. For the most commonly identified offenders (>3 mentions) 40% were outsider threats. These observations suggest that synthetic biology presents substantial new offending opportunities. Moreover, that more effective engagement, such as ethical hacking, is needed now to prevent a crime harvest from developing in the future. A framework to address the synthetic biology crime landscape is proposed.
