Abstract:Gene synthesis is an emerging field which has widespread implications in synthetic biology and molecular biology. The field is constantly evolving which has led to key advances in oligonucleotide synthesis and gene synthesis technologies, with simplicity, cost effectiveness and high throughput. The miniaturization, multiplexing, microfluidic processing and the integrated microchip engineering will drive down cost and increase productivity without compromising DNA synthesis fidelity, whereas the gigantic amount of genome information provides infinite source of DNA elements and genes as raw material for synthetic biology. This article describes some of the recent patents on oligonucleotide synthesis and gene synthesis.

Abstract:The ability to manipulate living organisms is at the heart of a range of emerging technologies that serve to address important and current problems in environment, energy, and health. However, with all its complexity and interconnectivity, biology has for many years been recalcitrant to engineering manipulations. The recent advances in synthesis, analysis, and modeling methods have finally provided the tools necessary to manipulate living systems in meaningful ways and have led to the coining of a field named synthetic biology. The scope of synthetic biology is as complicated as life itself-encompassing many branches of science and across many scales of application. New DNA synthesis and assembly techniques have made routine customization of very large DNA molecules. This in turn has allowed the incorporation of multiple genes and pathways. By coupling these with techniques that allow for the modeling and design of protein functions, scientists have now gained the tools to create completely novel biological machineries. Even the ultimate biological machinery-a self-replicating organism-is being pursued at this moment. The word synthetic biology hides a big world, ready to be explored by interdisciplinary research collaborations. The purpose of this investigation is to reveal the what, where, when of the current situation in this emerging field. A keyword search string for the field was constructed and applied in the Web of Science and in the Derwent Innovations Index. In particular, we calculated year based h-type indices for high-frequent keywords.

Abstract:The topic synthetic biology appears still as an 'empty basket to be filled'. However, there is already plenty of claims and visions, as well as convincing research strategies about the theme of synthetic biology. First of all, synthetic biology seems to be about the engineering of biology - about bottom-up and top-down approaches, compromising complexity versus stability of artificial architectures, relevant in biology. Synthetic biology accounts for heterogeneous approaches towards minimal and even artificial life, the engineering of biochemical pathways on the organismic level, the modelling of molecular processes and finally, the combination of synthetic with nature-derived materials and architectural concepts, such as a cellular membrane. Still, synthetic biology is a discipline, which embraces interdisciplinary attempts in order to have a profound, scientific base to enable the re-design of nature and to compose architectures and processes with man-made matter. We like to give an overview about the developments in the field of synthetic biology, regarding polymer-based analogs of cellular membranes and what questions can be answered by applying synthetic polymer science towards the smallest unit in life, namely a cell.

Abstract:Synthetic biologists have made great progress over the past decade in developing methods for modular assembly of genetic sequences and in engineering biological systems with a wide variety of functions in various contexts and organisms. However, current paradigms in the field entangle sequence and functionality in a manner that makes abstraction difficult, reduces engineering flexibility, and impairs predictability and design reuse. Functional Synthetic Biology aims to overcome these impediments by focusing the design of biological systems on function, rather than on sequence. This reorientation will decouple the engineering of biological devices from the specifics of how those devices are put to use, requiring both conceptual and organizational change, as well as supporting software tooling. Realizing this vision of Functional Synthetic Biology will allow more flexibility in how devices are used, more opportunity for reuse of devices and data, improvements in predictability, and reductions in technical risk and cost.

Abstract:In our view, synthetic biology is an extension of the continuum of genetic science that has been used safely for more than 40 years by the biotechnology industry in the development of commercial products. Examples of synthetic biology use by biotechnology companies illustrate the potential to substantially reduce research and development time and to increase speed to market. Improvements in the speed and cost of DNA synthesis are enabling scientists to design modified bacterial chromosomes that can be used in the production of renewable chemicals, biofuels, bioproducts, renewable specialty chemicals, pharmaceutical intermediates, fine chemicals, food ingredients, and health care products. Regulatory options should support innovation and commercial development of new products while protecting the public from potential harms.

Synthetic biology is involved in many aspects of human life, such as processing information, manipulating chemicals, providing food, fabricating materials and structures, producing energy, as well as maintaining and enhancing human health and our environment. Advances in omics technologies have profound effects on synthetic biology research, pushing synthetic biology research toward rational data-driven and model-driven science. The ultimate goals of synthetic biology are to design and build engineered biological systems. Furthermore, synthetic biology has a broad area for applications, including sustainable food, sustainable energy, sustainable environment, etc. In this chapter, we will introduce the history, techniques, applications, and future challenges of synthetic biology.

Abstract:Synthetic biology is a rising discipline that combines biology, chemistry, computer science, engineering, and physics. In the early twentieth century, French physical chemist Stephane Leduc [1] put forward the idea that life can be simplified into a chemical reaction in his book The Mechanism of Life; however, because people’s understanding has stayed at the early biological research stage, the level of understanding of molecular biology is insufficient, and synthetic biology has not been developed. Until 1962, Francois Jacob and Jacques Monod [2] proposed an operon model for E. coli gene expression, which was favoured by researchers for its precise regulation. With the rapid development of recombinant DNA technology in the 1970s and high-throughput sequencing in the 1980s, the construction of artificial biological systems has gradually changed from idea to reality, and people’s understanding of synthetic biology has gradually deepened. In 1980, German scientist Barbara H-bomb [3] defined synthetic biology as a gene for bacteria using recombinant DNA technology in his long-form paper “Gene Surgery: On the Threshold of Synthetic Biology”. In January 2000, Nature published two studies on the construction of the first artificial bistable gene regulatory network and synthetic gene oscillator in E. coli [4, 5]. So far, synthetic biology remains a new field. In the same year, Eric Kool and other spokespersons reintroduced the concept of synthetic biology at the American Chemical Society, defining synthetic biology as genetic engineering based on systems biology, from artificial base DNA molecules, gene fragments, gene regulatory networks with signal transduction pathways, to artificial design and synthesis in cells. There are many different opinions on the definition of synthetic biology; nowadays, scholars generally recognize that the use of engineering concepts rationally synthesizes complex, biologically meaningful systems of different levels, from individual biomolecules, to whole cells, tissues, and organs. Importantly, these biological systems can perform functions not found in nature.

Abstract:Synthetic biology breakthroughs have facilitated genetic circuit engineering to program cells through novel biological functions, dynamic gene expressions, as well as logic controls. SynBio can also participate in the rapid development of new treatments required for the human lifestyle. Moreover, these technologies are applied in the development of innovative therapeutic, diagnostic, as well as discovery-related methods within a wide range of cellular and molecular applications. In the present review study, SynBio applications in various cellular and molecular fields such as novel strategies for cancer therapy, biosensing, metabolic engineering, protein engineering, and tissue engineering were highlighted and summarized. The major safety and regulatory concerns about synthetic biology will be the environmental release, legal concerns, and risks of the engineered organisms. The final sections focused on limitations to SynBio.

Abstract:The convergence of advances in chemistry, physics, mathematics, computer science, and engineering into life science research gives rise to synthetic biology. Synthetic biology adopts the concept and strategy of engineering science research, aiming at redesigning and reprogramming the existing biological systems, designing and constructing new bio-bricks such as enzymatic parts, genetic circuits, and chassis cells, or even creating non-natural functions of "artificial life". Synthetic biology promotes the leap from understanding of life to design of life, and is revolutionizing biotechnology and sustainable development of bioeconomy. Via this retrospective review of Tianjin Institute of Industrial Biotechnology, Chinese Academy of Sciences, the most representative research entity focusing on "build-for-use" of synthetic biology in China, this article summarizes the important scientific and technological breakthroughs and industry impacts in the past decade, and prospects future development of synthetic biology in China.

Abstract:Synthetic Biology is a growing field which holds great promise in several areas of application. However, it also poses serious risks for human health and the environment that must be anticipated in order to develop effective prevention and management measures. Here, the current situation on biosafety and biosecurity in this scientific field is reviewed. Biosafety concerns mainly relate to the damaging effects for workers and the environment that could result from accidental interactions with dangerous biological agents. On the other hand, biosecurity risks refer to the potential misuses of Synthetic Biology, such as bioterrorism, biowarfare or bioattacks that could derive from the genetic engineering of organisms. In this paper, the specific challenges posed by Synthetic Biology are discussed, and perspectives in the development of measures to guide the safe advance of this discipline are presented.

Abstract:Synthetic biology is the design and construction of novel proteins, genetic circuits, and metabolic networks using information from a variety of disciplines, including molecular biology, engineering, mathematics, and physics. In unicellular and multicellular systems, we are currently heading toward the construction of complex networks rather than the early development stages' goal of changing the behaviour of individual constituents. Synthetic biologists can be broadly divided into two groups, one of which seeks to produce artificial life using unnatural substances. Another instance involves taking replaceable components from biological systems and integrating them into systems that function in an artificial way. Innovative behaviours like biostability, oscillations, proteins for biosensing, and drug production have recently been developed. Biological principles and informatics are a commonly accepted combination to make processing more accurate and faster. In this paper we give a deeper insight into the synthetic biology methods, few of its applications and also the discussing various bioinformatics tools for synthetic biology.

Abstract:Synthetic biology is a nascent field that emerged in earnest only around the turn of the millennium. It aims to engineer new biological systems and impart new biological functionality, often through genetic modifications. The design and construction of new biological systems is a complex, multistep process, requiring multidisciplinary collaborative efforts from "fusion" scientists who have formal training in computer science or engineering, as well as hands-on biological expertise. The public has high expectations for synthetic biology and eagerly anticipates the development of solutions to the major challenges facing humanity. This article discusses laboratory practices and the conduct of research in synthetic biology. It argues that the fusion science approach, which integrates biology with computer science and engineering best practices, including standardization, process optimization, computer-aided design and laboratory automation, miniaturization, and systematic management, will increase the predictability and reproducibility of experiments and lead to breakthroughs in the construction of new biological systems. The article also discusses several successful fusion projects, including the development of software tools for DNA construction design automation, recursive DNA construction, and the development of integrated microfluidics systems.

Abstract:Synthetic biology is designing and creating biological tools and systems for useful purposes. It uses knowledge from biology, such as biotechnology, molecular biology, biophysics, biochemistry, bioinformatics, and other disciplines, such as engineering, mathematics, computer science, and electrical engineering. It is recognized as both a branch of science and technology. The scope of synthetic biology ranges from modifying existing organisms to gain new properties to creating a living organism from non-living components. Synthetic biology has many applications in important fields such as energy, chemistry, medicine, environment, agriculture, national security, and nanotechnology. The development of synthetic biology also raises ethical and social debates. This article aims to identify the place of ethics in synthetic biology. In this context, the theoretical ethical debates on synthetic biology from the 2000s to 2020, when the development of synthetic biology was relatively faster, were analyzed using the systematic review method. Based on the results of the analysis, the main ethical problems related to the field, problems that are likely to arise, and suggestions for solutions to these problems are included. The data collection phase of the study included a literature review conducted according to protocols, including planning, screening, selection and evaluation. The analysis and synthesis process was carried out in the next stage, and the main themes related to synthetic biology and ethics were identified. Searches were conducted in Web of Science, Scopus, PhilPapers and MEDLINE databases. Theoretical research articles and reviews published in peer-reviewed journals until the end of 2020 were included in the study. The language of publications was English. According to preliminary data, 1,453 publications were retrieved from the four databases. Considering the inclusion and exclusion criteria, 58 publications were analyzed in the study. Ethical debates on synthetic biology have been conducted on various issues. In this context, the ethical debates in this article were examined under five themes: the moral status of synthetic biology products, synthetic biology and the meaning of life, synthetic biology and metaphors, synthetic biology and knowledge, and expectations, concerns, and problem solving: risk versus caution.

Abstract:Tremendous advances in the field of synthetic biology have been witnessed in multiple areas including life sciences, industrial development, and environmental bio-remediation. However, due to the limitations of human understanding in the code of life, any possible intended or unintended uses of synthetic biology, and other unknown reasons, the development and application of this technology has raised concerns over biosafety, biosecurity, and even cyberbiosecurity that they may expose public health and the environment to unknown hazards. Over the past decades, some countries in Europe, America, and Asia have enacted laws and regulations to control the application of synthetic biology techniques in basic and applied research and this has resulted in some benefits. The outbreak of the COVID-19 caused by novel coronavirus SARS-CoV-2 and various speculations about the origin of this virus have attracted more attention on bio-risk concerns of synthetic biology because of its potential power and uncertainty in the synthesis and engineering of living organisms. Therefore, it is crucial to scrutinize the control measures put in place to ensure appropriate use, promote the development of synthetic biology, and strengthen the governance of pathogen-related research, although the true origin of coronavirus remains hotly debated and unresolved. This article reviews the recent progress made in the field of synthetic biology and combs laws and regulations in governing bio-risk issues. We emphasize the urgent need for legislative and regulatory constraints and oversight to address the biological risks of synthetic biology.
