Abstract:Highly flexible nanocomposite hydrogels were prepared by using silica nanoparticles (SNPs) as fillers and multi-functional cross-links to graft hydrophilic poly(acrylic acid) (PAA) by free radical polymerization from an aqueous solution. The SNPs were collected by neighboring polymer chains and dispersed uniformly within a PAA matrix. The mechanical properties of the nanocomposite hydrogels were tailored by the concentration of SNPs according to the percolation model. It was proposed that covalent bonds of adsorbed chains on the filler surface resulted in the formation of a shell of an immobilized glassy layer and trapped entanglements, where the glassy polymer layer greatly enhanced the elastic modulus and the release of trapped entanglements at deformation contributed to the viscoelastic properties.
The robust hydrogel hierarchically assembled from a pH sensitive peptide amphiphile based on silk fibroin.

Abstract:Supramolecular polymers can be formed by self-assembly of designed subunits to yield highly ordered materials. In this paper, hierarchically structured materials, from molecules to nanofibers to macroscopical hydrogel, were fabricated by pH-induced assembly of C-12-GAGAGAGY, a peptide amphiphile (PA) based on silk fibroin. Due to the different acid dissociation constants of the carboxyl and phenolic hydroxyl groups on tyrosine residue (Y), the PM showed unique pH sensitive assembly and aggregation behaviors. It was found that not only the molecular-scale assemblies of these PM gradually changed from cylindrical nanofibers to nanoribbons with the decreasing of pH value from 11 to 8 but also most of nanoribbons aggregated into parallel bundles in such a case. Further decrease of pH value resulted in a hierarchically structured robust and plastic hydrogel, of which the rheological moduli reached around 10(5) Pa. Moreover, noodle-like hydrogel fibers with bundles of nanoribbons aggregated parallel along the long axis in them could be steadily prepared under shear force. Taking the pH-sensitive reversible sol-gel transition, high modulus and plasticity into account, the hydrogel is believed to have significant potential applications in tissue engineering or as the biocompatible adhesives.
Directed Nanoscale Self-Assembly of Low Molecular Weight Hydrogelators Using Catalytic Nanoparticles.

Abstract:The work presented here shows that the growth of supramolecular hydrogel fibers can be spatially directed at the nanoscale by catalytic negatively charged nanoparticles (NCNPs). The NCNPs with surfaces grafted with negatively charged polymer chains create a local proton gradient that facilitates an acid-catalyzed formation of hydrogelators in the vicinity of NCNPs, ultimately leading to the selective formation of gel fibers around NCNPs. The presence of NCNPs has a dominant effect on the properties of the resulting gels, including gelation time, mechanical properties, and network morphology. Interestingly, local fiber formation can selectively entrap and precipitate out NCNPs from a mixture of different nanoparticles. These findings show a new possibility to use directed molecular self-assembly to selectively trap target nano-objects, which may find applications in therapy, such as virus infection prevention, or engineering applications, like water treatment and nanoparticle separation.
Self-assembly/condensation interplay in nano-to-microfibrillar silicified fibrin hydrogels

Abstract:Fibrin-based gels are used in clinics as biological glues but their application as 3D cellularized scaffolds is hindered by processing and stability issues. Silicification of fibrin networks appears as a promising strategy not only to address these limitations but also to take advantage of the bioactivity of Si. However, it raises the question of the influence of silica sources on fibrin self-assembly. Here tetraethoxysilane, aminopropyltriethoxysilane and silica nanoparticles were used to design hybrid and nanocomposite fibrin-based hydrogels. By varying the concentration in silica source, we could evidence two regimes of interactions that depend on the extent of inorganic condensation. These interactions modulated the fibrillar structure of the fibrin network from more than 500 nm to less than 100 nm. These nanofibrillar hydrogels could exhibit higher mechanical properties than pure fibrin while preserving their capacity to support proliferation of myoblasts, opening promising perspectives for the use of fibrin-silica constructs in tissue engineering.

Abstract:Hydrogels are of intense recent interest in connection with biomedical applications ranging from 3-D cell cultures and stem cell differentiation to regenerative medicine, controlled drug delivery and tissue engineering. This prototypical form of soft matter has many emerging material science applications outside the medical field. The physical processes underlying this type of solidification are incompletely understood and this limits design efforts aimed at optimizing these materials for applications. We address this general problem by applying multiple techniques (e.g., NMR, dynamic light scattering, small angle neutron scattering, rheological measurements) to the case of a peptide derivative hydrogelator (molecule 1, NapFFKYp) over a broad range of concentration and temperature to characterize both the formation of individual nanofibers and the fiber network. We believe that a better understanding of the hierarchical self-assembly process and control over the final morphology of this kind of material should have broad significance for biological and medicinal applications utilizing hydrogels.

Abstract:This paper introduces a versatile method to synthesize physical hydrogels by silica nanoparticle (SN) surface in situ polymerization. A series of poly(acrylic acid) chains were covalently grafted from the SNs that were treated with γ-methacryloxypropyl trimethoxy silane. The entanglements among the grafted chains constructed the hydrogel networks and the SNs played the role of ‘analogous crosslinking points’. The SNs were encapsulated by the grafted polymer chains and formed a ‘core–shell’ structure. It was found that the molecular weight of the grafted chains and the content and diameter of the SNs affected the hydrogel’s properties. In comparison with chemically crosslinked hydrogels, the hybrid hydrogels exhibited excellent mechanical properties and could endure stress as high as 3.16 MPa and an elongation of 440% at a water content of 100 wt%. The gel–sol phenomenon confirmed that this hybrid hydrogel belonged to the physically crosslinked type.
Hybrid assembly of polymeric nanofiber network for robust and electronically conductive hydrogels

Abstract:Abstract Electroconductive hydrogels have been applied in implantable bioelectronics, tissue engineering platforms, soft actuators, and other emerging technologies. However, achieving high conductivity and mechanical robustness remains challenging. Here we report an approach to fabricating electroconductive hydrogels based on the hybrid assembly of polymeric nanofiber networks. In these hydrogels, conducting polymers self-organize into highly connected three dimensional nanostructures with an ultralow threshold (~1 wt%) for electrical percolation, assisted by templating effects from aramid nanofibers, to achieve high electronic conductivity and structural robustness without sacrificing porosity or water content. We show that a hydrogel composed of polypyrrole, aramid nanofibers and polyvinyl alcohol achieves conductivity of ~80 S cm −1 , mechanical strength of ~9.4 MPa and stretchability of ~36%. We show that patterned conductive nanofiber hydrogels can be used as electrodes and interconnects with favorable electrochemical impedance and charge injection capacity for electrophysiological applications. In addition, we demonstrate that cardiomyocytes cultured on soft and conductive nanofiber hydrogel substrates exhibit spontaneous and synchronous beating, suggesting opportunities for the development of advanced implantable devices and tissue engineering technologies.

Abstract:One of the most prominent properties of hydrogels is their excellent hydrolubrication that derives from the strong hydration of the gel network. However, excessive hydration makes hydrogels exhibit a very poor mechanical property, which limits their practical applications. Here, we prepared a novel composite surface of hydrogel nanofibers embedded in an anodic aluminum oxide substrate which exhibited both excellent lubrication and a high load-bearing capacity. Through the copolymerization of acrylic acid and 3-sulfopropyl methacrylate potassium salt, the gel network swelled sufficiently in aqueous solution and caused high osmotic pressure repulsion to bear heavy loads and hence exhibited excellent aqueous lubrication (μ ≈ 0.01). Notably, the friction coefficient of gels showed no dependence on the load in the experiment, whereas it was strongly influenced by the sliding velocity. Additionally, both electrolyte solution and ionic surfactants affect the conformation of the polymer chains, which results in a significant impact on the friction properties of hydrogel fibers.
Stretchable Sponge-like Hydrogels with a Unique Colloidal Network Produced by Polymerization-Induced Microphase Separation

Abstract:We report a series of sponge-like hydrogels prepared by polymerizing an aqueous solution of methacrylic acid (MAAc) in the presence of N,N,N',N'- tetramethylethylenediamine (TMEDA), which results in hydrogen bond complexation, microphase separation, and formation of a unique colloidal network. The microstructure of the gels is determined by the coupling of microphase separation and gelation, which are influenced by the feeding concentrations of MAAc and TMEDA. Colloidal gels are only obtained at a relatively low concentration of MAAc in the presence of a certain amount of TMEDA. The structural evolution during the polymerization and the formation mechanism for the colloidal network are revealed by a combination of dynamic light scattering measurements and optical microscopy. It reveals that hydrogen bond complexes are formed between poly(methacrylic acid) (PMAAc) and TMEDA, leading to the formation of colloidal particles that assemble into clusters and then the interconnected colloidal network. The physical hydrogels with a colloidal network are stable in water and possess high stretchability and good selfrecovery ability due to the robustness of colloidal particles joined by adhesive PMAAc chains. These sponge-like hydrogels are applied to remove dye molecules for water purification and devised into a solar vapor generation device to produce clean water. The strategy by harnessing reaction-induced phase separation should be applicable for other systems to engineering the microstructure and properties of functional materials toward specific applications.

Abstract:Hydrogels have been widely used in water purification. However, there is not much discussion and comparison about the effects of different nanofillers on the reinforcement and adsorption performances of hydrogels, which can be subjected to rapid water flow and possess strong adsorption ability. In this work, polyacrylamide (PAAM)-sodium alginate (SA) interpenetrating polymer network-structured hydrogels were prepared by in situ polymerization. PAAM formed the first flexible network and SA constructed the second rigid network. Three kinds of inorganic nanoparticles including carbon nanotubes (CNTs), nanoclays (NCs), and nanosilicas (NSs) were incorporated into a PAAM-SA matrix via hydrogen bond. The obtained hydrogels exhibited a macroporous structure with low density (≈1.4 g/cm3) and high water content (≈83%). Compared with neat PAAM-SA, the hydrogels with inorganic nanoparticles possessed excellent mechanical strengths and elasticities, and the compression strength of PAAM-SA-NS reached up to 1.3 MPa at ε = 60% by adding only 0.036 g NS in a 30 g polymer matrix. However, CNT was the best filler to improve the adsorption capacity owing to its multi-walled hollow nanostructure, and the adsorption capacity of PAAM-SA-CNT was 1.28 times higher than that of PAAM-SA. The prepared hydrogels can be potential candidates for use as absorbents to treat wastewater.

Abstract:Most natural supporting tissues possess both exceptional mechanical strength, a significant amount of water, and the anisotropic structure, as well as nanoscale assembly. These properties are essential for biological processes, but have been challenging to emulate in synthetic materials. In an effort to achieve simultaneous improvement of these trade-off features, a hydrogen bonding-induced self-assembly strategy was introduced to create nanoporous plastic-like polymer hydrogels. Multiple hydrogen bonding-mediated networks and nanoporous orientation structures endow transparent hydrogels with remarkable mechanical robustness. They exhibit Young's modulus of up to 223.7 MPa and a breaking strength of up to 10.3 MPa, which are superior to those of most common polymer hydrogels. The uniform porous nanostructures of hydrogen-bonded hydrogels contribute to a significantly larger specific surface area compared to conventional hydrogels. This allows for the retention of high mechanical properties in environments with a high water content of 70 wt %. A rubbery stage is observed during the heating process, which can reverse and reshape the manufacture of objects with various desired 2D or 3D shapes using techniques such as origami and kirigami. Finally, as a proof-of-concept, the outstanding mechanical properties of poly(MAA-co-AA-co-NVCL) hydrogel, combined with its high water content, make it suitable for applications such as smart temperature monitors, multilevel information anticounterfeiting, and artificial muscles.

Abstract:Today, the challenge of enhancing the strength and adhesion of the polyacrylamide hydrogel network is still a major concern in many applications. This study investigates the effect of silica nanoparticle concentration on the cross-linked polyacrylamide hydrogel's gelation, viscoelastic, structural, and adhesion properties. The hybrid hydrogels were evaluated using DSC, XRD, SEM, and rheological analysis to determine their enthalpy of cross-linking, crystallinity, morphology, and elastic properties. The results of kinetic analysis confirmed the catalytic effect of silica nanoparticles on the gelation kinetics. A deviation from the elastic behavior of the unfilled hydrogel was found from the rheological studies. Using the Weibull Model to describe the creep test results showed a significant increase in retardation time with increasing silica nanoparticles content up to the percolation threshold of 0.24 wt.%. This confirmed the development of a more viscous behavior due to the formation of a physical network of nano-silica particles. Based on the DSC energy of hydrogels, both chemical and physical tie points (approximately 19 covalent crosslinking points) were calculated. The hydrogel containing 9 wt.% of silica nanoparticles demonstrated significant improvement in density, and adhesion to the metal surface by about 100% and 350%, respectively. As a result of this study, the hydrogel containing 9 wt% of silica nanoparticles (H 9 ) is suggested for various field studies, such as oil well plugging industries.

Self-assembly of multiscale anisotropic hydrogels through interfacial polyionic complexation

Abstract:Polysaccharides are explored for various tissue engineering applications due to their inherent cytocompatibility and ability to form bulk hydrogels. However, bulk hydrogels offer poor control over their microarchitecture and multiscale hierarchy, parameters important to recreate extracellular matrix-mimetic microenvironment. Here, we developed a versatile platform technology to self-assemble oppositely charged polysaccharides into multiscale fibrous hydrogels with controlled anisotropic microarchitecture. We employed polyionic complexation through microfluidic flow of positively charged polysaccharide, chitosan, along with one of the three negatively charged polysaccharides: alginate, gellan gum, and kappa carrageenan. These hydrogels were composed of microscale fibers, which in turn were made of submicron fibrils confirming multiscale hierarchy. Fibrous hydrogels showed strong tensile mechanical properties, which were further modulated by encapsulation of shape-specific antioxidant cerium oxide nanoparticles (CNPs). Specifically, hydrogels with chitosan and gellan gum showed more than eight times higher tensile strength compared to the other two pairs. Incorporation of sphere-shaped cerium oxide nanoparticles in chitosan and gellan gum further reinforced fibrous hydrogels and increased their tensile strength by 40%. Altogether, our automated hydrogel fabrication platform allows fabrication of bioinspired biomaterials with scope for one-step encapsulation of small molecules and nanoparticles without chemical modification or use of chemical crosslinkers.

Abstract:Bionanomaterial based hydrogels originated from natural biopolymer have drawn much attention for advanced applications. However, nanosilk-based hydrogels derived from top-down approaches remain in their infancy. First, nanosilks based on existing methods fail to prepare hydrogels; second, both nanosilk extraction and surface modification remain a challenge due to high crystallinity and sophisticated hierarchical structures. To produce nanosilk-based hydrogels, pretreatment and oxidation are necessary. In this work, pretreatments were conducted first to loosen the sophisticated structures of natural silk fibers, NaClO oxidation was utilized in succession to introduce carboxyl groups onto silk fibroin. Combined with moderate mechanical disintegration, silk nanocrystals with additional carboxyl groups were prepared facilely. Finally, silk nanocrystal-based hydrogels were prepared successfully through gas phase coagulation. An optimization of pretreatment approaches and oxidation conditions was carried out. The morphologies, chemical and crystalline structures of original, pretreated and oxidized silk fibroin as well as nanofibrillated silk were investigated. In addition, the silk nanocrystal-based hydrogel exhibited outstanding mechanical properties compared to those of dissolved and regenerated silk fibroin-based hydrogels. Moreover, silk nanocrystal-based aerogels present highly porous, interconnected, and crisscrossed network nanostructures, which are ideal candidates for tissue regeneration and provide new prospects as porous scaffolds for bioengineering applications.

Acrylic acid (AA) and octylphenol polyoxyethylene acrylate with 10 ethoxyl units (OP-10-AC) as basic monomers in situ graft from the SNs surface to build poly(acrylic acid) hydrophilic backbone chains with randomly distributed OP-10-AC hydrophobic side chains. The entanglements among grafted backbone polymer chains and hydrophobic branch architecture lead to the SNs and HAMDs play the role of physical cross-links for the hydrogels network structure. The rheological behavior and polymer concentration for gelation process are measured to examine the critical gelation conditions. The correlation of the polymer dual cross-linked networks with hydrogels swelling behavior, gel-to-sol phase transition, and mechanical strength are addressed, and the results imply that the unique dual cross-linking networks contribute the hydrogels distinctive swelling behavior and excellent tensile strength. The effects of SNs content, molecular weight of polymer backbone, and temperature on hydrogels properties are studied, and the results indicate that the physical hydrogel network integrity is depended on the SNs and HAMDs concentration.
