Abstract:The application of immobilized enzymes in medicine is the main objective of this book. The author reviews natural and synthetic carriers for enzyme immobilization, chemistry of enzyme binding, and in-vitro and in-vivo properties of immobilized enzymes. Four chapters are dedicated to clinical use of immobilized enzymes.

Abstract:Abstract Background This article is a comprehensive review of the events recorded in the history of enzymology from twentieth century to the recent work of author’s groups on different biochemical aspects of the immobilized enzymes. The review differs from those presented before on the basis that it is not limited to one or few aspects. It extends its spectrum to a large number of aspects from the definition to biomedical applications of immobilized enzymes. Main body The author and his associates have proposed modification of classification of immobilized enzymes by Enzyme Engineering Conference 1971. His groups have worked on cell bound proteases of medicinal seeds and have given them the name of “naturally immobilized enzymes”. Thus, the author proposes that the enzymes may be basically classified into “naturally immobilized enzymes” and “artificially immobilized enzymes”. The artificially immobilized enzymes may be further sub-classified as done in 1971. Conclusion The classification suggested above sounds logical and thus acceptable to the author and his associates. The author and his associates also suggest some applications on the basis of their results on naturally immobilized enzymes.

Abstract:Immobilized enzymes are used in organic syntheses to fully exploit the technical and economical advantages of biocatalysts based on isolated enzymes. Immobilization enables the separation of the enzyme catalyst easily from the reaction mixture, and can lower the costs of enzymes dramatically. This is true for immobilized enzyme preparations that provide a well-balanced overall performance, based on reasonable immobilization yields, low mass transfer limitations, and high operational stability. There are many methods available for immobilization which span from binding on prefabricated carrier materials to incorporation into in situ prepared carriers. Operative binding forces vary between weak multiple adsorptive interactions and single attachments through strong covalent binding.Which of the methods is the most appropriate is usually a matter of the desired applications. It is therefore the intention of this paper to outline the common immobilization methods and reaction technologies to facilitate proper applications of immobilized enzymes.

Abstract:Reports on chemical immobilization of proteins and enzymes first appeared in the 1960s. Since then, immobilized proteins and enzymes have been widely used in the processing of variety of products and increasingly used in the field of medicine. Here, we present a review of recent developments in immobilized enzyme use in medicine. Generally speaking, the use of immobilized enzyme in medicine can be divided into two major categories: biosensors and bioreactors. A brief overview of the evolution of the biosensor and bioreactor technology, of currently existing applications of immobilized enzymes, of problems that researchers encountered, and of possible future developments will be presented.

Abstract:Enzymes are outstanding (bio)catalysts, not solely on account of their ability to increase reaction rates by up to several orders of magnitude but also for the high degree of substrate specificity, regiospecificity and stereospecificity. The use and development of enzymes as robust biocatalysts is one of the main challenges in biotechnology. However, despite the high specificities and turnover of enzymes, there are also drawbacks. At the industrial level, these drawbacks are typically overcome by resorting to immobilized enzymes to enhance stability. Immobilization of biocatalysts allows their reuse, increases stability, facilitates process control, eases product recovery, and enhances product yield and quality. This is especially important for expensive enzymes, for those obtained in low fermentation yield and with relatively low activity. This review provides an integrated perspective on (multi)enzyme immobilization that abridges a critical evaluation of immobilization methods and carriers, biocatalyst metrics, impact of key carrier features on biocatalyst performance, trends towards miniaturization and detailed illustrative examples that are representative of biocatalytic applications promoting sustainability.

Abstract:Therapeutic enzymes play important roles in modern medicine due to their high affinity and specificity. However, it is very expensive to use them in clinical medicine because of their low stability and bioavailability. To improve the stability and effectiveness of therapeutic enzymes, immobilization techniques have been employed to enhance the applications of therapeutic enzymes in the past few years. Reported immobilization techniques include entrapment, adsorption, and covalent attachment. In addition, protein engineering is often used to improve enzyme properties; however, all methods present certain advantages and limitations. For carrier-bound immobilization, the delivery and release of the immobilized enzyme depend on the properties of the carrier and enzyme. In this review, we summarize the advantages and challenges of the current strategies developed to deliver therapeutic enzymes and provide a future perspective on the immobilization technologies used for therapeutic enzyme delivery.

Abstract:Publisher This chapter reviews the history of immobilized cells. Enzymes are protein biocatalysts that participate in the chemical reactions that occur in living organisms. The utilization of enzymes began with human history and has gradually expanded into a variety of fields, such as brewing, food production, textiles, tanning, and medicine. The techniques that started with immobilized enzymes have been adapted to immobilize microbial cells, to immobilize living microbial cells, and, further, to immobilize plant and animal cells. Microbial enzymes can be classified into two groups: extracellular (that is, excreted from the cells into the broth) and intracellular (that is, retained in the cells during cultivation). These immobilized systems are applied in a variety of fields, and they play a very important role in the field of biotechnology. Genetic engineering and enzyme engineering are two technologies in which immobilized biocatalysts play the main role.

Abstract:Immobilized enzymes are becoming increasingly popular as analytical reagents because of their reusability, stability, and sensitivity to many inhibitors that would seriously interfere in assays using soluble enzymes. In this article, some of the kinetic and catalytic effects of immobilized enzymes in analysis will be discussed. The shift of the activity-pH profile curves on immobilization, the changes in temperature dependence. the inhibitor constants (K1). Michaelis constants (K m ), and the maximum velocity (Vmax). plus others, will be discussed. Finally, the use of these immobilized enzymes in fluorometric and electrochemical monitoring systems will be shown, and the future of these reagents in various areas will be discussed. A survey of enzyme electrodes will be presented as an example of the use of immobilized enzymes. Application of immobilized enzyme technology to the assay of BUN, glucose, uric acid, amino acids, ethanol. and other metabolites will be discussed.

Abstract:Although the history of immobilized enzymes and their applications in different fields are traced back to the second half of the twentieth century, their importance in bioreactors and biosensors highlighted at the turn of the current century is under active consideration in these days for broad-spectrum applications in different medical fields. Thus, this article presents a review of the literature concerning the current and future role of the immobilized enzymes in different medical fields. As the author and his supervised research groups have also been actively involved in research on immobilized enzymes, he has also made some input with a recommendation to revise the current classification of immobilized enzymes basically into “Naturally Immobilized” and “Artificially Immobilized Enzymes”, keeping the sub-classification intact. He has also suggested the development of some medical products based on the results of his research groups.

Abstract:The current demands of the world's biotechnological industries are enhancement in enzyme productivity and development of novel techniques for increasing their shelf life. Compared to free enzymes in solution, immobilized enzymes are more robust and more resistant to environmental changes. More importantly, the heterogeneity of the immobilized enzyme systems allows an easy recovery of both enzymes and products, multiple reuse of enzymes, continuous operation of enzymatic processes, rapid termination of reactions, and greater variety of bioreactor designs. This review summarizes immobilization definition, different immobilization methods, advantages and disadvantages of each method. In addition, it covers some food industries, protein purification, human nutrition, biodiesel production, and textile industry. In these industries, the use of enzymes has become an inevitable processing strategy when a perfect end product is desired. It also can be used in many other important industries including health care and pharmaceuticals applications. One of the best uses of enzymes in the modern life is their application in diagnose and treatment of many disease especially when used in drug delivery system or when used in nanoform.

Abstract:In recent years, enzyme immobilization technology has been developed, and studies on immobilized enzyme materials have become very prominent. With the immobilization technique, enzymes and compatible carrier materials are combined or enzyme crystals/aggregates are used in a carrier-free fashion, by physical, chemical, or biochemical methods. As a kind of biocatalyst, immobilized enzymes can catalyze certain chemical reactions with high selectivity and high efficiency under relatively mild reaction conditions and eliminate pollution to the environment. Considering the current status and applications of immobilized enzyme technology and materials emerging in the last 5 years, this mini-review introduces the advantages and disadvantages of various enzyme immobilization techniques with carriers as well as the pros and cons of different materials for immobilization. The future prospects of immobilization technology and carrier materials are outlined, aiming to provide a reference for further research and applications of sustainable technology.

Abstract:Enzymes play vital roles in all organisms. The enzymatic process is progressively at its peak, mainly for producing biochemical products with a higher value. The immobilization of enzymes can sometimes tremendously improve the outcome of biocatalytic processes, making the product(s) relatively pure and economical. Carrier-free immobilized enzymes can increase the yield of the product and the stability of the enzyme in biocatalysis. Immobilized enzymes are easier to purify. Abstract:Publisher This chapter introduces several techniques for biocatalyst immobilization that include inclusion, adsorption, cross-linking, or covalent grafting. One of the key parameters with regard to the feasibility of a biotechnological process is the combination of efficiency and stability of the biocatalyst used. Immobilization very often results in a greatly increased resistance to various denaturation factors: extreme pH and temperature values, high ionic strength, denaturating reagents, and proteases. Another approach to stabilization is to modify the biocatalyst microenvironment by the use of additives, such as salts, polyols, sugars, and various polymers. It has thus been possible to obtain enzyme preparations with a highly increased resistance to denaturation factors, for example, certain protease preparations used in the manufacture of detergents. The chapter discusses the possibilities of biocatalyst stabilization, on the one hand by immobilization and on the other hand by immobilization plus additives, with the help of a few significant examples. Direct comparison of the operational stability—that is, in the presence of a continuous supply of substrate—of an immobilized enzyme with that of a free enzyme is possible in the ultrafiltration reactor.

Abstract:The current demands of the world’s biotechnological industries are enhancement in enzyme productivity and development of novel techniques for increasing their shelf life. These requirements are inevitable to facilitate large-scale and economic formulation. Enzyme immobilization provides an excellent base for increasing availability of enzyme to the substrate with greater turnover over a considerable period of time. Several natural and synthetic supports have been assessed for their efficiency for enzyme immobilization. Nowadays, immobilized enzymes are preferred over their free counterpart due to their prolonged availability that curtails redundant downstream and purification processes. Future investigations should endeavor at adopting logistic and sensible entrapment techniques along with innovatively modified supports to improve the state of enzyme immobilization and provide new perspectives to the industrial sector.

Abstract:In spite of their excellent catalytic properties, enzyme properties have to be usually improved before their implementation at industrial scale (where many cycles of high yield processes are desired). Generally, soluble enzymes have to be immobilized to be reused for long times in industrial reactors and, in addition to that, some other critical enzyme properties have to be improved like stability, activity, inhibition by reaction products, selectivity towards non-natural substrates. Some strategies to improve these enzyme properties during the performance of tailor-made enzyme immobilization protocols are here reviewed. In this way, immobilized enzymes may also exhibit much better functional properties than the corresponding soluble enzymes by very simple immobilization protocols. For example, multipoint and multisubunit covalent immobilization improve the stability of monomeric or multimeric enzymes. Moreover, enantioselectivity of different enzymes (e.g., lipases) may be also dramatically improved (from E=1 to >100) by performing different immobilization protocols of the same enzyme. In all cases, enzyme engineering via immobilization techniques is perfectly compatible with other chemical or biological approaches to improve enzyme functions and the final success depend on the availability of a wide battery of immobilization protocols.
