Abstract:Enzymes are biological catalysts that play an immense role in the food industry. However, the desirable characteristics of enzymes are hindered by their long-term operational stability, shelf storage life, recovery, and reuse. These drawbacks can be overcome by enzyme immobilization, which enhances the functional efficiency and reproducibility while requiring less labor input and reducing contamination. Therefore, this boosts the stability of the products. The primary immobilization of the enzyme was amino cyclase of Aspergillus oryzae for the production of L-amino acids from Japan. Key enzymes such as pectinase and cellulases are immobilized on suitable carriers and are useful in the food industry (baking, dairy products, jams, and jellies), beverage processing (wine, beer, fruit, and vegetable juices), etc. There are various techniques to immobilize an enzyme, including adsorption (static process, dynamic process, reactor loading, electrodeposition), Covalent bonding (diazoation, peptide bond polyfunctional reagents), entrapment (inclusion in the gels, fibers, and microcapsules), and copolymerization and encapsulation (microencapsulation, nanoencapsulation). Nanotechnology is one of the most promising fields that attempts to provide an efficient technique that could answer several challenges in terms of economy and industrial development in food processing.

Abstract:Immobilized enzymes have been widely used in the processing of variety of products. New strategies are continuously emerging for the formation of diverse immobilized enzymes having superior efficiency and usage. Immobilized enzymes have biomedical and industrial applications and for this reason, this area has continued to develop into an ever-expanding and multidisciplinary field during the last couple of decades. This study is a review of the latest literatures on different enzymes immobilized on various supporting materials. Immobilized enzymes for multiplicity of applications ranging from wine, sugar and fish industry through organic compounds removal from wastewaters to sophisticated biosensors for both in situ measurements of environmental pollutants and metabolite control are reviewed. Generally speaking, immobilized enzymes have immense potential in the analysis of clinical, industrial and environmental samples. Immobilized enzymes and proteins have been tremendously used in antibiotic production, drug metabolism, food industry, biodiesel production and bioremediation. The success of the vast usage of immobilized enzymes lies in the fact that they prove to be environmental friendly, cheaper and easy to use when compared to other parallel technologies.

Abstract:Abstract Background This article is a comprehensive review of the events recorded in the history of enzymology from twentieth century to the recent work of author’s groups on different biochemical aspects of the immobilized enzymes. The review differs from those presented before on the basis that it is not limited to one or few aspects. It extends its spectrum to a large number of aspects from the definition to biomedical applications of immobilized enzymes. Main body The author and his associates have proposed modification of classification of immobilized enzymes by Enzyme Engineering Conference 1971. His groups have worked on cell bound proteases of medicinal seeds and have given them the name of “naturally immobilized enzymes”. Thus, the author proposes that the enzymes may be basically classified into “naturally immobilized enzymes” and “artificially immobilized enzymes”. The artificially immobilized enzymes may be further sub-classified as done in 1971. Conclusion The classification suggested above sounds logical and thus acceptable to the author and his associates. The author and his associates also suggest some applications on the basis of their results on naturally immobilized enzymes.

Abstract:The current demands of the world’s biotechnological industries are enhancement in enzyme productivity and development of novel techniques for increasing their shelf life. These requirements are inevitable to facilitate large-scale and economic formulation. Enzyme immobilization provides an excellent base for increasing availability of enzyme to the substrate with greater turnover over a considerable period of time. Several natural and synthetic supports have been assessed for their efficiency for enzyme immobilization. Nowadays, immobilized enzymes are preferred over their free counterpart due to their prolonged availability that curtails redundant downstream and purification processes. Future investigations should endeavor at adopting logistic and sensible entrapment techniques along with innovatively modified supports to improve the state of enzyme immobilization and provide new perspectives to the industrial sector.

Abstract:Enzymes are outstanding (bio)catalysts, not solely on account of their ability to increase reaction rates by up to several orders of magnitude but also for the high degree of substrate specificity, regiospecificity and stereospecificity. The use and development of enzymes as robust biocatalysts is one of the main challenges in biotechnology. However, despite the high specificities and turnover of enzymes, there are also drawbacks. At the industrial level, these drawbacks are typically overcome by resorting to immobilized enzymes to enhance stability. Immobilization of biocatalysts allows their reuse, increases stability, facilitates process control, eases product recovery, and enhances product yield and quality. This is especially important for expensive enzymes, for those obtained in low fermentation yield and with relatively low activity. This review provides an integrated perspective on (multi)enzyme immobilization that abridges a critical evaluation of immobilization methods and carriers, biocatalyst metrics, impact of key carrier features on biocatalyst performance, trends towards miniaturization and detailed illustrative examples that are representative of biocatalytic applications promoting sustainability.

Abstract:Reports on chemical immobilization of proteins and enzymes first appeared in the 1960s. Since then, immobilized proteins and enzymes have been widely used in the processing of variety of products and increasingly used in the field of medicine. Here, we present a review of recent developments in immobilized enzyme use in medicine. Generally speaking, the use of immobilized enzyme in medicine can be divided into two major categories: biosensors and bioreactors. A brief overview of the evolution of the biosensor and bioreactor technology, of currently existing applications of immobilized enzymes, of problems that researchers encountered, and of possible future developments will be presented.

Compared to free enzymes in solution, immobilized enzymes are more robust and more resistant to environmental changes. More importantly, the heterogeneity of the immobilized enzyme systems allows an easy recovery of both enzymes and products, multiple reuse of enzymes, continuous operation of enzymatic processes, rapid termination of reactions, and greater variety of bioreactor designs. This review summarizes immobilization definition, different immobilization methods, advantages and disadvantages of each method. In addition, it covers some food industries, protein purification, human nutrition, biodiesel production, and textile industry. In these industries, the use of enzymes has become an inevitable processing strategy when a perfect end product is desired. It also can be used in many other important industries including health care and pharmaceuticals applications. One of the best uses of enzymes in the modern life is their application in diagnose and treatment of many disease especially when used in drug delivery system or when used in nanoform.

Abstract:Enzyme immobilization refers to the process of attaching or confining enzymes onto a solid support or within a matrix, often made of polymers or other materials. This immobilization creates a stable and controlled environment for the enzyme to interact with substrates and perform catalysis. The primary goal of enzyme immobilization is to enhance enzyme stability, reusability, and activity under specific conditions, making them more practical and efficient for various biotechnological, industrial, and medical applications. Immobilization methods can vary widely, including physical adsorption, covalent bonding, entrapment within matrices, encapsulation, crosslinking, and more. These methods provide a means to control the interactions between the enzyme and the surrounding environment, affecting factors such as substrate accessibility, enzyme orientation, and stability. Due to their ease of fabrication and superior structural adaptability, polymer compounds in a variety of physical forms, including beads, films, fibers,and coatings,have become popular as supportive materials for enzyme immobilization. For enzyme immobilization, a number of natural polymers, including agar, agarose, alginate, dextran, chitosan,and carrageenan, as well as synthetic polymers, such as polyamides, polystyrene, and polyacrylamide, are often employed as a carrier system. The immobilization offers a cost-effective system for various applications in biotechnology, industry, and research.

Abstract:Immobilized enzymes are used in organic syntheses to fully exploit the technical and economical advantages of biocatalysts based on isolated enzymes. Immobilization enables the separation of the enzyme catalyst easily from the reaction mixture, and can lower the costs of enzymes dramatically. This is true for immobilized enzyme preparations that provide a well-balanced overall performance, based on reasonable immobilization yields, low mass transfer limitations, and high operational stability. There are many methods available for immobilization which span from binding on prefabricated carrier materials to incorporation into in situ prepared carriers. Operative binding forces vary between weak multiple adsorptive interactions and single attachments through strong covalent binding.Which of the methods is the most appropriate is usually a matter of the desired applications. It is therefore the intention of this paper to outline the common immobilization methods and reaction technologies to facilitate proper applications of immobilized enzymes.

Abstract:Enzyme-based catalysis has become one of the most important disciplines in organic synthesis and plays a noteworthy role in the establishment of many chemical industries, e.g. fine chemicals, food or energy, textiles, agricultural, cosmeceutical, medicinal and pharmaceutical industries. However, pristine enzymes fail to demonstrate requisite functionalities for an industrial setting where extremely specific and stable catalysts are required. Immobilization enhances the catalytic stability and activity of enzymes and trims the overall cost burden of the enzyme. Therefore, it widely endeavours for proficient, sustainable, and environmentally responsive catalytic processes. Amongst several immobilization strategies, e.g. (1) supports-assisted, i.e. physical or covalent coupling and (2) supports-free techniques, i.e. cross-linked enzyme crystals (CLECs) or aggregates are the most promising ones and widely pursued for enzyme immobilization purposes. This perspective review focuses on up-to-date developments in the area of enzyme immobilization and presents their potentialities to upgrade and/or modify enzyme properties. Both types of immobilization strategies, i.e. supports-assisted and supports-free techniques are discussed with particular reference to CLECs or aggregates and protein-coated microcrystals. Also, several useful traits achieved after immobilization are also discussed in the second half of the review.

Abstract:Enzyme immobilization is an essential technology for commercializing biocatalysis. It imparts stability, recoverability, and other valuable features that improve the effectiveness of biocatalysts. While many avenues to join an enzyme to solid phases exist, protein-mediated immobilization is rapidly developing and has many advantages. Protein-mediated immobilization allows for the binding interaction to be genetically coded, can be used to create artificial multienzyme cascades, and enables modular designs that expand the variety of enzymes immobilized. By designing around binding interactions between protein domains, they can be integrated into functional materials for protein immobilization. These materials are framed within the context of biocatalytic performance, immobilization efficiency, and stability of the materials. In this review, supports composed entirely of protein are discussed first, with systems such as cellulosomes and protein cages being discussed alongside newer technologies like spore-based biocatalysts and forizymes. Protein-composite materials such as polymersomes and protein-inorganic supraparticles are then discussed to demonstrate how protein-mediated strategies are applied to many classes of solid materials. Critical analysis and future directions of protein-based immobilization are then discussed, with a particular focus on both computational and design strategies to advance this area of research and make it more broadly applicable to many classes of enzymes.

Abstract:Enzyme immobilization has been developing since the 1960s and although many industrial biocatalytic processes use the technology to improve enzyme performance, still today we are far from full exploitation of the field. One clear reason is that many evaluate immobilization based on only a few experiments that are not always well-designed. In contrast to many other reviews on the subject, here we highlight the pitfalls of using incorrectly designed immobilization protocols and explain why in many cases sub-optimal results are obtained. We also describe solutions to overcome these challenges and come to the conclusion that recent developments in material science, bioprocess engineering and protein science continue to open new opportunities for the future. In this way, enzyme immobilization, far from being a mature discipline, remains as a subject of high interest and where intense research is still necessary to take full advantage of the possibilities.

Abstract:Engineering enzymes with improved catalytic properties in non-natural environments have been concerned with their diverse industrial and biotechnological applications. Immobilization represents a promising but straightforward route, and immobilized biocatalysts often display higher activities and stabilities compared to free enzymes. Owing to their unique physicochemical characteristics, including the high-specific surface area, exceptional chemical, electrical, and mechanical properties, efficient enzyme loading, and multivalent functionalization, nano-based materials are postulated as suitable carriers for biomolecules or enzyme immobilization. Enzymes immobilized on nanomaterial-based supports are more robust, stable, and recoverable than their pristine counterparts, and are even used for continuous catalytic processes. Furthermore, the unique intrinsic properties of nanomaterials, particularly nanoparticles, also confer the immobilized enzymes to be used for their broader applications. Herein, an effort has been made to present novel potentialities of multi-point enzyme immobilization in the current biotechnological sector. Various nano-based platforms for enzyme/biomolecule immobilization are discussed in the second part of the review. In summary, recent developments in the use of nanomaterials as new carriers to construct robust nano-biocatalytic systems are reviewed, and future trends are pointed out in this article.

Abstract:The instability of several macromolecules (e.g., enzymes and essential oils) under such factors as pH, temperature, and organic solvents may render their application in industrial and biotechnological processes nonviable. To overcome these drawbacks, some alternatives have been proposed, which includes immobilization. This technique corresponds to the confinement of a compound of interest within/to a support or matrix through different mechanisms (e.g., adsorption, entrapment, and covalent attachment). Although this methodology has been widely explored for innovative and multifaceted applications, it does have some limitations such as additional costs and loss or reduction of activity of the immobilized compound. However, depending on the immobilization technique used, some advantages such as ability to protect compounds, catalyst reuse, and easier product separation may outweigh these disadvantages. In this regard, new techniques and the immobilization varied materials of interest are in development, and contribute to a substantial growth in the number of publications in this field of study. Due to the large increase of research, several new articles are published daily; therefore, reviews that summarize this field are important. This review focuses on the use of immobilization techniques and their advantages in bioprocesses based on three main groups and related applications: (1) immobilized enzymes for biodiesel, flavoring agent, and antimicrobial compound production; (2) immobilized microorganisms for ingredient production, wastewater treatment, and probiotic protection; (3) immobilized essential oils for food preservation and as ingredients in pharmaceuticals and cosmetics. Furthermore, conclusions and future perspectives are discussed, aiming at improvement of existing processes and the release of new technologies for industrial use with less costs.

Abstract:The current demands of sustainable green methodologies have increased the use of enzymatic technology in industrial processes. Employment of enzyme as biocatalysts offers the benefits of mild reaction conditions, biodegradability and catalytic efficiency. The harsh conditions of industrial processes, however, increase propensity of enzyme destabilization, shortening their industrial lifespan. Consequently, the technology of enzyme immobilization provides an effective means to circumvent these concerns by enhancing enzyme catalytic properties and also simplify downstream processing and improve operational stability. There are several techniques used to immobilize the enzymes onto supports which range from reversible physical adsorption and ionic linkages, to the irreversible stable covalent bonds. Such techniques produce immobilized enzymes of varying stability due to changes in the surface microenvironment and degree of multipoint attachment. Hence, it is mandatory to obtain information about the structure of the enzyme protein following interaction with the support surface as well as interactions of the enzymes with other proteins. Characterization technologies at the nanoscale level to study enzymes immobilized on surfaces are crucial to obtain valuable qualitative and quantitative information, including morphological visualization of the immobilized enzymes. These technologies are pertinent to assess efficacy of an immobilization technique and development of future enzyme immobilization strategies.
