Abstract:Repeat breeder (RB) cows are clinically healthy animals with regular oestrous cycles that do not become pregnant after three or more services. This syndrome has an incidence ranging between 10.1% and 24%. Repeat breeder syndrome (RBS) in dairy cows leads to economic losses to dairy farmers by increasing the calving interval and consequently reducing milk and calf production. RBS has a complex oetiopathogenesis as many factors are involved in its onset. The causes can be grouped into two categories: causes leading to fertilisation failure and factors leading to early embryonic death. Accurate identification of the cause and early diagnosis of RBS is essential to minimise the problems that this issue brings to the farm, but is not always possible. Hypothesising the underlying aetiology of the syndrome is also crucial for targeted therapy, whether pharmacological or managerial. The aim of this review is to report the different therapies, proposed in the literature, for the treatment of RBS in cattle, based on the knowledge of possible aetiological causes.
Prevalence, risk factors for and impact of subclinical endometritis in repeat breeder dairy cows

Abstract:The aim of this study was to determine the prevalence and risk factors for subclinical endometritis (SE) and its effects on fertility in repeat breeder dairy cows. Dairy cows of parity 1 to 5 that were artificially inseminated (AI) 3 or more times (selected cows were artificially inseminated an average of 3.9 times) were examined at 190 ± 40 days in milk, and clinically normal cows (n = 77) were selected based on the absence of abnormal discharges on external inspection and the absence of abnormal findings on transrectal palpation and ultrasonographic examination. Endometrial samples were collected from the uterus by using the lavage technique in the luteal phase of the estrus cycle. Collected samples were centrifuged and a drop of sediment was streaked on to a clean microscopic slide and stained with Giemsa. The percentage of polymorphonuclear cells (neutrophils) was counted for each specimen. The analysed data showed that the average amount of neutrophils was 3.1% (0-9) in the selected cows. Abnormal calving (dystocia, twin births, and abortion), retained placenta, and postpartum uterine infections were associated with an increase in prevalence of SE. Subsequently, SE was significantly associated with a decrease in conception rate in the next AI. Conception rate in the next AI was 5% for cows (n = 38) with SE (≥ 3% neutrophil), and 47% for cows (n = 34) without SE (< 3% neutrophil) (P = 0.001). The prevalence of cytologically diagnosed SE (≥ 3% neutrophil) was 52.7% (n = 38). In conclusion, abnormal calving, retained placenta, and postpartum uterine infections may be associated with an increase in prevalence of SE and subsequently, SE may decrease reproductive performance and increase the incidence of repeat breeder syndrome.

Abstract:Improving the historically low reproductive performance of beef cattle in smallholder systems is essential for its productivity and profitability. Therefore, identifying and addressing risk factors associated with low performance in this system present an opportunity for improvement. The study aimed to evaluate the effect of animal and management risk factors on pregnancy rate (PR), fetal and calf loss (FC), calving interval (CI) and days open (DO) in smallholder beef cattle farms. A multi-stage selection approach was conducted in five provinces of South Africa. Cow records (3694) collected from 40 smallholder herds over two years (2018-2019) were analysed. Data on animal and herd management factors including body condition score (BCS), cow age class, breed type, lactation status, culling old/non-productive cows, record-keeping, and breeding and calving seasons were recorded. The GLIMMIX procedure was computed to determine risk factors associated with performance indicators (PR, FC, CI and DO). Risk factors highly associated with performance were breed type, BCS, cow-age class and breeding/calving season (P ≤ 0.05). Indicators FC, DO and CI increased with decreased BCS, autumn calving seasons, and in first calvers and aged cows. Whereas, PR increased with increasing odds of BCS and breeding seasons between December-March, November-February and January-March. Optimizing reproductive performance in smallholder beef cattle herds may involve managing cow body condition, breeding with adapted genotypes and synchronizing breeding with favourable seasons. Therefore, record keeping and regular monitoring of herd nutrition, climate and breed performance may be significant in improving reproductive performance in smallholder herds.

It considers key ART techniques including estrus synchronization for artificial insemination, and ovulation induction, highlighting their role in improving fertility and overall herd productivity. However, it also addresses common postpartum inflammatory and functional reproductive disorders, discussing their diagnosis and treatment protocols, stressing their impact on the overall reproductive outcome in buffalo farming.

Repeat breeding: prevalence and potential causes in dairy cows at different milk pocket areas of Bangladesh

Abstract:The objective of this study was to figure out the prevalence and probable causes of repeat breeding (RB) in dairy cows. Hence, a cross-sectional study was conducted on randomly selected 265 dairy farms in Sirajganj, Bogura, Rangpur, Satkhira, and Munshiganj districts of Bangladesh from December 2018 to February 2019. Data were collected through a direct interview method using a survey questionnaire. The reproductive organs of repeat breeder cows were examined for pathological, infectious, and functional reasons, and genital tract abnormalities. Additionally, the influence of nutrition, season, and age on the frequency of RB was recorded. The prevalence of RB was 28% among the 3824 cows investigated. Among the total repeat breeder cases, 72.54% of RB cases were found in Holstein-Friesian crossbred, 23.90% in Jersey crossbred, 1.50% in Sahiwal crossbred, and 2.06% in indigenous cows. The prevalence of RB was significantly highest (P < 0.01) in Satkhira (44.35%) and lowest in the Munshiganj district (15.87%). Data indicated that a major proportion of cows significantly (P < 0.05) faced RB problems due to functional causes (34.18%), followed by pathological causes (28.01%), genital tract abnormalities (21.32%), and infectious causes (16.49%). Furthermore, the cows were remarkably (P < 0.001) affected in RB during the summer season and nutritional deficient diseases like milk fever (70%). Age (3-7 years) had a significant (P < 0.001) effect on the RB occurrence (90%) in crossbred cows. However, particular focus should be given to systematic breeding, balanced nutrition, artificial inseminator efficiency, and hygienic inseminating tools to reduce RB incidence in high-yielding crossbred cows.
Evaluation of the effect of sustained-release progesterone injection on the expression of interferon-related genes in repeat-breeder dairy cows

Abstract:Background: Repeat-breeder cows repeatedly fail to conceive after at least three attempts and return to oestrus at apparently normal intervals. Repeat-breeder cows cause economic losses in dairy farms in different ways. Objective: In the present study, we investigated the effect of sustained-release progesterone injection in two different doses on the expression of interferon-related genes in repeat-breeder dairy cows. Methods: A total of 96 repeat-breeder primiparous and multiparous cows were assigned among three groups: control group, inseminated and do not receive progesterone treatment; P400 and P600 groups, inseminated and received a single-intramuscular injection of 400 and 600 mg slow-release progesterone 5 days after insemination, respectively. Blood sampling was carried out on Day 20 after AI for progesterone measurement and evaluation of gene expression for ISG15, MX1 and MX2 genes. Results: One injection of sustained-release progesterone increased the expression of ISG15, MX1 and MX2 genes with differences between two different progesterone concentrations. For all three genes, the level of gene expression was higher in progesterone-supplemented group than in control group, when P400 and P600 groups considered together. The level of MX2 gene expression was significantly higher in pregnant cows than non-pregnant cows. There was a significant positive correlation between expression level of all three genes and blood progesterone concentration. The expression level of ISG15 gene showed a significant positive correlation with MX1 and MX2 gene expression. Conclusion: The use of this sustained-release progesterone is simple and can be used in repeat-breeder cows to improve fertility.

Abstract:The purpose of this study was to evaluate the effect of retained placenta (RP) and clinical mastitis (CM) on the reproductive efficiency of crossbred dairy cows during the postpartum period and the effect in some innate immune system indicators. For this, two experiments were carried out. In the first, a total of 232 cows were evaluated and divided as: healthy control (n = 184), RP (n = 22), and CM (n = 26) groups. The RP and CM was evaluated until 30 days postpartum (DPP) and reproductive rates were measured. In experiment 2, cows were divided in control (n = 10), RP (n = 10), and CM (n = 30) groups. Between 40 and 50 DPP, clinical, gynecological examination and endometrial cytobrush were performed to evaluate subclinical endometritis (SE) and gene expression of interleukins 1β (IL-1β) and 6 (IL-6), chemokine ligand 5 (CCL5), estrogen α (ESR1), and progesterone (PGR) receptors by qRT-PCR analysis. In experiment 1, the conception rate at 1st artificial insemination (AI) was lower in RP and CM groups and pregnancy rate at 150 days decreased in CM group. Calving-to-1st AI interval and days open were shorter in healthy cows. In experiment 2, the occurrence of SE was 26.7% and higher in RP and CM groups. The expression of IL-1β increased in RP and CM groups, while IL-6 was less expressed in RP group. The CCL5, ESR1, and PGR were similar between groups. In conclusion, cows with RP and CM had their reproductive efficiency negatively affected and had they initial pro-inflammatory response improved by the increase of IL-β.

Abstract:Objective: A study examined the relationship between somatic cell count (SCC) and milk yield Theoretical Framework: The sustainable development of dairy farms will be key to stopping the growing agricultural frontier. In comparison to breeds from temperate regions, tropical bovine production is low in terms of milk kilos, composition, or udder health. Method: We enrolled one hundred fifty cows (Primiparous, PM, 75 and Multiparous, MP 75) in early lactation (days in milk, PM =134 ± 3; MP = 136 ± 5), milk production (9,88 kg/d, on average) of the creole breed Gyr lechers. Prior to the assignment to each treatment, the SCC values were lower than 220,000 cells/mL, on average. All cows were maintained to graze daily on Megathyrsus maximus and supplemented with Morus alba ad libitum, being hand-milking at 0700 daily. Before analysis, the SCC was logarithmically trans-formed (log10). Then, PROC Mixed from SAS version 9.4 was used to evaluate all measurements. Results and Discussion: The MP had higher milk yields than PM cows (10.83 vs. 9.18 ± 0.38 kg/d; P = 0.003). Similar results were observed for fat-corrected milk (8.26 vs. 6.80 ± 0.34; P = 0.002), although the fat values did not differ between both groups (P = 0.86) being lower than referential values for these breeds (2.46 ± 0.16, on average). Additionally, no differences were observed in the other milk components (P = 0.65 to 0.85). Despite that, the somatic cell count (SCC) values indicated a statistical tendency in PM than in MP (1.89 vs. 2.13 ± 0.05; P = 0.07). Research Implications: Low-fat contents were observed in both groups, possibly due to the low quality of foods used in ruminant feeding. Correspondingly, the parity and advanced lactation conditioned the SCC contents. Therefore, further studies may be relevant in order to identify complementary factors that possibly could be more determinants. Originality/Value: The relevance and value of this research are very importance due to the main activity in this zone is livestock cattle.
Recent Possibilities for Diagnosis and Treatment of Post Parturient Uterine Diseases in Dairy Cow

Abstract:The successful genetic selection for higher milk production caused a dramatic decline in the reproductive performance of dairy cows all over the world during the last decades. Achievement of optimum herd reproductive performance (calving interval of 12 months or 13 months with the first calf born at 24 months of age) requires concentrated management activities especially during the first 100 days following calving. The following management activities are needed to pursue during the early postpartum period to reach or approach the optimal reproductive performance such as careful surveillance and assistance at calving, prevention of post parturient metabolic diseases, early diagnosis and treatment of post parturient uterine diseases, accurate detection of oestrus, correct timing of insemination, reducing the effect of heat stress and early pregnancy diagnosis. Among these main activities only early diagnosis and treatment of post parturient uterine diseases and their effects on reproductive performance are discussed. Clinical metritis and clinical endometritis (or recently used purulent vaginal discharge: PVD) can be accurately diagnosed in the field, however it is very important to remark that the presence of PVD alone is not sufficient to diagnose clinical endometritis because cervicitis and/or vaginitis can also cause PVD. Both diseases have detrimental effect on reproduction therefore it is important to diagnose as soon as possible and treat accordingly. Different treatment protocols that have changed in the recent years for those diseases, as well as recently performed therapies like ozone therapy or vaccination are also discussed. Although subclinical endometritis has also a detrimental effect on reproduction, it is complicated to diagnose in the field.

Abstract:The microbiota plays an important role in numerous physiological processes, pathogenesis, development, and metabolism in different animal species. In humans, several studies have demonstrated an association between the vaginal microbiota and fertility rates, and even success in assisted reproduction techniques. In the context of cattle reproduction, although few studies have addressed the microbiota in a healthy state (which is not associated with diseases that affect the reproductive tract of cows), changes in its composition also seem to influence fertility. This review aims to explain the importance of the reproductive microbiota in female bovines and what is available in the literature regarding its possible role in increasing fertility. What are the challenges involved in this process? Future perspectives on its use and manipulation as a selection or intervention tool. Will it be possible to one day extrapolate the findings to reality and apply them in the field? In short, understanding the role of the reproductive microbiota of female bovines can signal the prospect of increasing production, whether of milk or meat, from the same number of animals, as it can optimize reproductive efficiency and perhaps become an allied tool for the economic profitability and sustainability of livestock farming.
Differential diagnosis of chronic infertility in high-yield cows

Abstract:The infertility of cows and heifers has been the most pressing problem in the livestock sector. The genitals and mammary gland have an increased functional load associated with pregnancy, childbirth, and lactation. The purpose of this study was to determine the prevalence of symptoms of chronic irreversible infertility in Ayrshire cows culled as a result of multiple artificial inseminations. The study employed analytical, structural-comparative, and statistical methods, as well as a modified differential palpation diagnosis of the clinical state of the reproductive organs in the ovary+oviduct area with lateral localisation was consistently applied in two stages. The findings of the study showed that cows with in vivo diagnosed symptoms of chronic adherent salpingitis and ovosalpingitis (52.17%), i.e., irreversible form of infertility, had significantly higher (+28.05%; P < 0.01) milk production and better lactation compared to those without these pathologies. Furthermore, in the group of cows culled due to numerous ineffective inseminations, related gonadopathies were detected: from 4.35% to 10.87% of ovarian hypotrophy, 4.35% of gonadal sclerosis symptoms and 17.39% of cystic follicular degeneration cases. In 73.91% of females, the presence of sexual cyclicity with unchanged ovarian function in the follicular phase and in 84.78% – in the luteal phase of the cycle was established. The use of differential palpation diagnostics in production conditions allows in vivo predicting the tendency to loss of genetic resources (the ability of cow ovaries to maintain generative and secretory functions) in highly productive cows due to chronic inflammatory gynecological pathologies, which will contribute to the improvement of drug and biotechnological schemes of therapeutic measures for the prevention of infertility in the dairy herd

Genome-wide association study for reproduction traits in Colombian Creole Blanco Orejinegro cattle

Abstract:The profitability of the beef cattle production system relies heavily on reproductive traits. Unfortunately, certain traits, such as age at first calving (AFC), calving interval (CI), and gestation length (GL), can pose challenges in traditional breeding programs because of their low heritability (0.01-0.12) and sex-limited characteristics. Another important aspect is the conservation of the genetic resources of animals adapted to the Colombian regions, which implies the preservation and rational use of the creole breeds in the country market. Therefore, this study aimed to identify genomic regions in the creole cattle breed Blanco Orejinegro (BON) that influence the reproductive traits in females. The dataset comprised 439 animals and 118,116 single-nucleotide polymorphisms' (SNPs) markers. The GS3 program was used to identify the SNP effects employing the BAYES Cπ methodology. The number of SNPs with effect for AFC was 25, 1527 for CI, and 23 for GL. Some of the genes found associated with reproductive and growth traits as well as immune response and environmental adaptation ECE1, EPH, EPHB2, SMARCAL1, IGFBP5, IGFBP2, FCGRT, EGFR, MUL1, PINK1, STPG1, CNGB1, TGFB1, OXTR, IL22RA1, MYOM3, OXTR, CNR2, HIVEP3, CTPS1, CXCL8, FCGRT, MREG, TMEM169, PECR, and MC1R. Our results evidenced a high contribution of the genetic architecture of the Colombian creole cattle breed Blanco Orejinegro that may impact should be included in implementing genetic improvement and conservation programs.
The Effect of Maternal Behavior around Calving on Reproduction and Wellbeing of Zebu Type Cows and Calves

Abstract:The behaviors associated with domestic cattle such as maternal care are quite similar to those behaviors observed in wild ungulates. These behaviors allow the cow to bond with her calf, protect and provide it with nourishment and ultimately reduce the bond at weaning. Although maternal behavior is an important factor influencing the survival and early development of the newborn calf, Zebu type cows around calving have not been studied extensively. Herein, we consider the four main aspects of maternal behavior in cattle and particularly Bos indicus cows and calves. Firstly, we provide a brief description of the behavior of cows around parturition and the behavior of the first stages of the calves’ lives. In the second part, the protective behavior of the mother is analyzed. Subsequently, examples of animal welfare implications followed by an analysis of some factors that affect calf survival, including mother experience and weather conditions, are discussed, and in the last part, reproduction along with some peculiarities of reproductive behavior, and the wellbeing of mother and calves are examined. We concluded that knowledge of maternal behavior of Zebu type cows around calving and interactions with calves might contribute to an enhanced reproductive efficiency of the mother and the welfare of the calf.

Relationships between milk production, ovarian function and fertility in high-producing dairy herds in north-eastern Spain

Abstract:In the dairy industry worldwide, reproductive disorders are a major cause of economic losses and a challenge to scientists and technicians. In recent decades, declining fertility and increasing milk production have been widely reported in dairy cattle. In this article, the relationships between milk production, ovarian disorders and fertility in high-producing dairy herds are briefly described. We carried out a retrospective study of 23 204 lactations included in a reproductive control programme in north-eastern Spain, a geographical area experiencing both warm and cool conditions. The data were collected between 1991 and 2007 and refer to cows first inseminated or examined 45-80 days postpartum in five well-managed, commercial, Holstein-Friesian high-producing dairy herds. Ovarian disorders were classified as ovarian inactivity or hypofunction, cystic ovarian disease, sub-oestrus or silent ovulation and sub-luteal function. Ovarian hypofunction and milk production increased throughout the study period and there was a decrease in the pregnancy rate to first artificial insemination (AI). Cows suffering ovarian hypofunction were efficiently treated using combined progestagen-prostaglandin treatments. The incidence of ovarian cysts showed little variation with time. Treatment of this syndrome may include different GnRH-based treatments or manual rupture. During the last 5 years, sub-oestrus was the predominant dysfunction (42.1%) compared with the cystic (6.3%) and ovarian hypofunction (12%) forms. Response of sub-oestrous cows to treatment with luteolitic agents was usually higher than 60%. Ovarian function and fertility were dramatically impaired during the warm period. However, during the later years of the study, the inclusion of fans and water sprinklers for the warm season appeared to overcome the seasonal effect on fertility.
Improving decision-making in complicated or rare cases-An approach towards Evidence-based Veterinary Medicine in small animal reproduction

Abstract:The concepts of Evidence-based Veterinary Medicine (EBVM) provide a methodological and systematic approach to include the best evidence from research into clinical decision-making. These concepts include steps as the search and assessment of relevant research findings and consideration of individual aspects. In addition, owners and other persons involved in animal health care should be included in shared decision-making. Some breeders have good basic knowledge concerning breeding management and characteristics of diseases and concerning advantages and disadvantages of different therapeutic approaches, while others are notable to understand complex medical interrelations or emergency situations. All these aspects need to be addressed when communicating and discussing different diagnostic, prophylactic and therapeutic options. In special fields, such as small animal reproduction, veterinarians often see animals with rare diseases or complex conditions so that an application of standard therapies and well-established textbook recommendations is not possible. To learn more about cases too rare for successful systematic research, the case collection tool REPROCASES (www.evssar.org/reprocases) is now available. The aim of this project is to gather information from specialists on small animal reproduction in a multicentre approach. Even if this approach cannot completely replace standardized clinical trials, the idea is to gather more information on effects, prognosis, side effects and long-term fertility for specific conditions.

Abstract:The domestic buffalo is an indispensable livestock resource to millions of smallholder farmers in developing countries, particularly in Asia. Although its reproductive biology is basically similar to that of cattle, there are important differences and unique characteristics that need to be considered in order to apply modern reproductive technologies to improve its productivity. Under most smallholder production systems, the reproductive efficiency of buffalo is compromised by factors related to climate, management, nutrition and diseases. However, when managed and fed properly, buffalo can have good fertility and provide milk, calves and draught power over a long productive life. The basic technical problems associated with artificial insemination in buffalo were largely overcome two decades ago, but the technology has not had the expected impact in some developing countries, because largely of infrastructural and logistic problems. Approaches involving the use of hormones for treating anoestrus and for synchronizing oestrus have had varying rates of success, depending on the protocols used and the incidence of underlying problems that cause infertility. Embryo technologies such as multiple ovulation embryo transfer, in vitro embryo production, cryopreservation and cloning are being intensively studied but have had far lower success rates than in cattle. Improving the productivity of buffalo requires an understanding of their potential and limitations under each farming system, development of simple intervention strategies to ameliorate deficiencies in management, nutrition and healthcare, followed by judicious application of reproductive technologies that are sustainable with the resources available to buffalo farmers.
Fertility outcomes in cows with subclinical endometritis after clinical cure of clinical endometritis

Abstract:Clinical endometritis (CE) is common in post-partum dairy cows and is associated with impaired reproductive performance. The aim of the study was to evaluate the effect of subclinical endometritis (SE) in cows clinically cured of CE on their fertility. The study was performed on 215 Holstein Friesian cows with CE diagnosed by vaginoscopy and ultrasound between 21 and 28 days after parturition. All cows were clinically examined three times at an interval of 2 weeks. Cows without signs of CE were considered cured, and endometrial samples from the uteri were collected by cytobrush to diagnose SE using cytological evaluation of polymorphonuclear neutrophils (PMNs) percentage. The threshold for SE was set at ≥ 5% PMNs. Intervals calving to oestrus and calving to conception, first AI pregnancy rate, pregnancy rate 200 days after artificial insemination (AI), the number of AI per pregnancy (AI/P), pregnancy loss, and culling rate were calculated. SE was diagnosed in 40.9% of cows clinically cured of CE. There were significant differences in the AI/P (3.2 vs. 2.6; p < 0.027) and the pregnancy loss (18.2% vs. 4.7%; p < 0.002) between cows with SE and without SE. Cows with SE showed a tendency towards longer interval calving to conception, lower pregnancy rate 200 days after AI, and higher culling rate. In conclusion, SE after a clinical cure of CE may reduce fertility in dairy cows.

Abstract:Female fertility is the foundation of the cow–calf industry, impacting both efficiency and profitability. Reproductive failure is the primary reason why beef cows are sold in the U.S. and the cause of an estimated annual gross loss of USD 2.8 billion. In this review, we discuss the status of the genomics, transcriptomics, and systems genomics approaches currently applied to female fertility and the tools available to cow–calf producers to maximize genetic progress. We highlight the opportunities and limitations associated with using genomic and transcriptomic approaches to discover genes and regulatory mechanisms related to beef fertility. Considering the complex nature of fertility, significant advances in precision breeding will rely on holistic, multidisciplinary approaches to further advance our ability to understand, predict, and improve reproductive performance. While these technologies have advanced our knowledge, the next step is to translate research findings from bench to on-farm applications.

Abstract:Much progress has been made in the reproductive efficiency of lactating dairy cows across the USA in the past 20years. The standardisation of evaluation of reproductive efficiency, particularly with greater focus on metrics with lesser momentum and less lag-time such as 21-day pregnancy rates (21-day PR), and the recognition that subpar reproductive efficiency negatively impacted profitability were major drivers for the changes that resulted in such progress. Once it became evident that the genetic selection of cattle for milk yield regardless of fertility traits was associated with reduced fertility, geneticists raced to identify fertility traits that could be incorporated in genetic selection programs with the hopes of improving fertility of lactating cows. Concurrently, reproductive physiologists developed ovulation synchronisation protocols such that after sequential treatment with exogenous hormones, cows could be inseminated at fixed time and without detection of oestrus and still achieve acceptable pregnancy per service. These genetic and reproductive management innovations, concurrently with improved husbandry and nutrition of periparturient cows, quickly started to move reproductive efficiency of lactating dairy cows to an upward trend that continues today. Automation has been adopted in Israel and European countries for decades, but only recently have these automated systems been more widely adopted in the USA. The selection of dairy cattle based on genetic indexes that result in positive fertility traits (e.g. daughter pregnancy rate) is positively associated with follicular growth, resumption of ovarian cycles postpartum, body condition score and insulin-like growth factor 1 concentration postpartum, and intensity of oestrus. Collectively, these positive physiological characteristics result in improved reproductive performance. Through the use of automated monitoring devices (AMD), it is possible to identify cows that resume cyclicity sooner after calving and have more intense oestrus postpartum, which are generally cows that have a more successful periparturient period. Recent experiments have demonstrated that it may be possible to adopt targeted reproductive management, utilising ovulation synchronisation protocols for cows that do not have intense oestrus postpartum and relying more heavily on insemination at AMD-detected oestrus for cows that display an intense oestrus postpartum. This strategy is likely to result in tailored hormonal therapy that will be better accepted by the public, will increase the reliance on oestrus for insemination, will improve comfort and reduce labour by reducing the number of injections cows receive in a lactation, and will allow for faster decisions about cows that should not be eligible for insemination.
Oocyte competence and gene expression in parthenogenetic produced embryos from repeat breeder and normally fertile buffaloes (Bubalus bubalis) raised in sub-humid tropical climate

Abstract:The reproductive management of the buffalo species still faces several unresolved problems, which directly affect the productivity of the herd, one of them being the presence of repeat breeder females. Given this scenario, this study aimed to verify the developmental competence of oocytes obtained from repeat breeder females and submitted to parthenogenetic activation. In addition, embryo gene expression was compared to normally fertile females. Murrah buffaloes were divided into two groups: repeat breeder (RB, n = 8) and normally fertile or control (CR, n = 7). Cumulus-oocyte complexes (COCs) were aspirated by transvaginal ovum pick-up from estrus synchronized females. The COCs were submitted to IVM for 24 h, and subsequently, the oocytes were activated using ionomycin, followed by 6-DMAP. Afterwards, the presumptive parthenotes were cultured for six or seven days in a microenvironment of 5 % CO2, 5 % O2, and 90 % N2 at 38.5 °C. The expression of OCT4, GLUT1, BCL2 and TFAM genes from blastocysts was evaluated. The overall COCs recovery rate was 70.9 % (190/268). The maturation (57.8 vs 71.1), cleavage (45.2 vs 62.2) and blastocyst (30.1 vs 45.9) rates did not differ (P > 0.05) between RB and CR females, respectively. Similarly, no significant difference (P > 0.05) was observed for the expression of studied genes in both RB and CR females. In conclusion, oocytes obtained from RB were as developmentally competent as those collected from CR females, with similar energy metabolism and in vitro development capacity. Thus, the low fertility rate of repeat breeder buffaloes, when compared to normal cyclic females, must be due to subsequent events to the blastocyst stage.
