Abstract:Elasmobranchs (sharks and rays) show an amazing diversity of reproductive modes and behaviors. Multiple paternity (MP) has been identified in all species where more than 1 litter has been investigated; yet neither direct nor indirect benefits from MP have been determined in elasmobranchs. This has led to the suggestion that MP in this group may simply be a product of convenience polyandry with variation in the frequency of MP driven by differences in mate encounter rates. Here, we use molecular markers to investigate polyandry and MP in 2 closely related and commercially important species of shark, Mustelus antarcticus and Mustelus lenticulatus. In total, 328 M. antarcticus embryos originating from 29 different mothers and 75 M. lenticulatus embryos originating from 19 different mothers were genotyped using 8 microsatellite loci. We find that MP occurs in both species. However, in both species, the majority of litters were sired by a single father. Our results do not support increased fecundity per se as a driver of MP. Further, our results do not suggest that high population densities with resulting high mate encounter rates generated by breeding aggregations necessarily lead to high frequencies of MP. Importantly, we note evidence of reproductive skew within polyandrous litters, which is a predicted outcome of postcopulatory mechanisms.
Perspective on physiological/endocrine and nutritional factors influencing fertility in post-partum dairy cows

Abstract:Increasing reproductive performance of post-partum lactating dairy cows is a multi-factorial challenge involving disciplines of production medicine, nutrition, physiology and herd management. Systems of programmed timed insemination have been fine-tuned to achieve pregnancy per artificial inseminations (AI) approximating 45%. Systems have optimized follicle development, integrated follicle development with timing of induced corpus luteum regression and fine-tuned sequential timing of induced ovulation and AI. Use of programmes for insemination have identified occurrence of anovulatory ovarian status, body condition, uterine health and seasonal summer stress as factors contributing to reduced herd fertility. Furthermore, programmes of timed insemination provide a platform to evaluate efficacy of nutritional and herd health systems targeted to the transition and post-partum periods. The homeorhetic periparturient period, as cows deal with decreases in dry matter intake, results in a negative energy balance and is associated with a period of immunosuppression. Cows that transition well will cycle earlier and have a greater risk of becoming pregnant earlier post-partum. The innate arms of the immune system (acute and adaptive) are suppressed during the periparturient period. Cows experiencing the sequential complex of disorders such as dystocia, puerperal metritis, metritis, endometritis and subclinical endometritis are subsequently less fertile. Targeted strategies of providing specific nutraceuticals that provide pro- and anti-inflammatory effects, such as polyunsaturated fatty acids (e.g., linoleic, eicosapentaenoic/docosahexaenoic, conjugated linoleic acid), sequential glycogenic and lipogenic enrichment of diets, and organic selenium appear to differentially regulate and improve the immune and reproductive systems to benefit an earlier restoration of ovarian activity and increased fertility.
Impact of oocyte donor age and breed on in vitro embryo production in cattle, and relationship of dairy and beef embryo recipients on pregnancy and the subsequent performance of offspring: A review

Abstract:Genomic selection combined with in vitro embryo production (IVEP) with oocytes from heifer calves provides a powerful technology platform to reduce generation interval and significantly increase the rate of genetic gain in cattle. The ability to obtain oocytes with developmental competence from calves has been recognised for more than 60years. There is renewed interest in the potential of this reproductive technology as the beef and dairy industries seek to identify and multiply animals that satisfy consumer demand for efficient utilisation of natural resources, with minimal environmental impact and high product quality. Heifer calves show a good follicular response to FSH stimulation and oocytes can be recovered by ovum pick-up (OPU). However, the efficiency of OPU/IVEP remains lower for calves compared with peripubertal heifers and cows, in both indicus (Zebu, Bos indicus ) and taurus (Bos taurus ) breeds. Indicus cattle generally have more follicles, yield a greater number of oocytes, and have a better IVEP outcome, compared with taurus cattle. The treatment of prepubertal heifers with bovine somatotrophin (bST) and propylene glycol before OPU/IVEP has yet to show a major improvement in embryo production. Holstein (taurus) dairy heifers derived from different reproductive technologies (AI, MOET, OPU/IVEP) showed a similar age at puberty and first-lactation milk production. OPU/IVEP Holstein embryos transferred to beef or dairy cows likewise yielded heifers with the same performance. The gains in productivity that can be achieved with strategic use of OPU/IVEP in heifer calves make this a relevant and highly important reproductive technology in cattle breeding. Ongoing optimisation of the technology is needed for the potential of OPU/IVEP in young donors to be fully realised.

Abstract:The reproductive efficiency of the herd is correlated with higher productivity in livestock. Reproduction biotechniques, such as ovulation synchronization protocols, are important to optimize production and accelerate genetic profit in beef and dairy herds. The objective of this review is to describe the evolution over the last 40 years of the artificial insemination (AI) and the timed-AI (TAI) protocols in cattle from a Brazilian perspective. TAI protocols are based on synchronizing emergence of the wave of follicular growth, controlling circulating progesterone (P4) concentrations, stimulating the final growth of the follicle and inducing a synchronized ovulation. Hormonal alternatives that optimize the response at the end of the protocol and strategies to induce final follicle growth and ovulation in categories of females with low expression of estrus are described. Furthermore, the potential positive effect of previous exposure to injectable P4 on fertility of Bos indicus and Bos taurus cows is also discussed.
Reproductive physiological outcomes of dairy cows with different genomic merit for fertility: Biomarkers, uterine health, endocrine status, estrus features, and response to ovarian synchronization

Abstract:Our overarching objective was to characterize associations between genomic merit for fertility and the reproductive function of lactating dairy cows in a prospective cohort study. In this manuscript, we present results of the association between genomic merit for fertility and indicators of metabolic status and inflammation, uterine health, endocrine status, response to synchronization, and estrous behavior in dairy cows. Lactating Holstein cows entering their first (n = 82) or second (n = 37) lactation were enrolled at parturition and fitted with an ear-attached sensor for automated detection of estrus. Ear-notch tissue samples were collected from all cows and submitted for genotyping using a commercial genomic test. Based on genomic PTA values for daughter pregnancy rate (gDPR), cows were classified into a high (Hi-Fert; gDPR >0.6; n = 36), medium (Med-Fert; gDPR -1.3 to 0.6; n = 45), or low (Lo-Fert; gDPR <-1.3; n = 38) group. At 33 to 39 DIM, cohorts of cows were enrolled in the Presynch-Ovsynch protocol for synchronization of estrus and ovulation. Body weights, BCS, and uterine health measurements (i.e., vaginal discharge, uterine cytology) were collected from parturition to 60 DIM, and milk yield was collected through 90 DIM. Blood samples were collected weekly through 3 wk of lactation for analysis of BHB, nonesterified fatty acids (NEFA), and haptoglobin plasma concentrations. Body weight, BCS, NEFA, BHB, and haptoglobin were not associated with fertility groups from 1 to 9 wk after parturition. The proportion of cows classified as having endometritis at 33 to 36 DIM tended to be greater for the Lo-Fert than the Hi-Fert group. The proportion of cows that resumed cyclicity did not differ at any time point evaluated, and there were no significant associations between probability or duration and intensity of estrus with fertility group. Cows of superior genetic merit for fertility were more likely to ovulate, have a functional corpus luteum, have greater circulating progesterone, and have larger ovulatory size than cows of inferior fertility potential at key time points during synchronization of estrus and ovulation. Despite observing numerical differences with potential performance consequences for the proportion of cows that responded to synchronization of ovulation and were both cyclic and responded to the Ovsynch portion of the synchronization protocol, we did not observe significant differences between fertility groups. Although not consistent and modest in magnitude, the collective physiological and endocrine differences observed suggested that cows of superior genetic fertility potential might have improved reproductive performance, at least in part, because of modestly improved endocrine status, uterine health, and ability to ovulate.

Abstract:INTRODUCTION Fertility is considered a complex trait influenced by many physiological and disease-related variables. These is considerable scientific evidence to support the view that fertility performance is influenced by environment (E), genetics (G) and the interaction between G by E (GxE). Many hypothesis have been proposed to explain this including genetics, physiology, nutrition nad management, and these factors have been investigated at the animal, organ and celluar level at critical time points of the productive life of dairy cows ( Walsh et al. , 2011). It is largely known that, fertility in dairy cows strongly decreased over the last decades as milk production per cows has highly increased. Hence, the reproductive efficiency is became an high priority in all systems and it is considered higher in seasonal calving systems as the opportunity for cow to calve and become pregnant is time limited to ensure a calf per cow per year in synchrony with grass growth ( Dillon et al., 2006). Over the last 30 years, genetic selection for increased milk production, particularly within the North American Holstein-Friesian genotype, has been very successful. Between 1985 and 2003, the rate of phenotypic gain in milk production per cow per year has been 193 kg for the United States, 131 kg for the Netherlands, 35 kg for the New Zealand and 46 kg for Ireland ( Dillon et al. , 2006). In Italy, the rate of the phenotypic gain in milk production per cow per year has been 112 kg in Holstein-Friesian with an average increase of the calving interval of 1.4 d per year ( Cassandro and Penasa , 2010). Despite these countries having diverse production systems, genetic selection criteria and climatic conditions, they all report a sensible decline in reproductive performance during the same period inducing, in recent years, the emphasis within selection indices for Holstein-Friesian has shifted from predominantly production to funcional nonproduction traits associated with improved health and fertility ( Miglior et al. , 2005). Poor reproductive performace often leads to premature culling and decreased productive career of dairy cows. The association between the declines in fertility and milk production in the last decades, is evident in the Holstein population, as reported in Figure 1 (USDA-ARS AIPL , 2007).

Abstract:With the development of in vitro technologies, embryos can be produced using oocytes retrieved directly from the ovaries, i.e., regardless of ovulation. This has allowed the use of different animal categories as oocyte donors, including prepubertal cattle. The advantages of using this strategy to shorten the generation interval and accelerate genetic gain over time were soon recognized, and the first offspring generated using oocytes collected from calves were born in the early 1990s. Nevertheless, embryo production from calves and prepubertal heifers remains a challenge. The oocytes collected before puberty present low in vitro developmental potential, and the subsequent blastocyst rates are consistently lower than those from pubertal females. The acquisition of developmental competence by the oocytes occurs progressively throughout the prepubertal period, which can be subdivided into an early, intermediate, and late prepubertal (or peripubertal) phases, each characterized by different physiological and endocrine features. Therefore, embryo yield increases with age but will only achieve its maximum after puberty. The most common strategy to improve oocyte developmental potential before puberty is the use of gonadotrophic stimulation prior to oocyte retrieval. The results with superstimulation, however, vary among studies, depending on the source, dose, and length of FSH treatment, as well as the age and breed of the donors. The use of calves and prepubertal heifers as oocyte donors should also consider the possible impacts of the oocyte retrieval technique (LOPU or OPU) and the use of exogenous hormones on their subsequent fertility and productive life.

Abstract:The productivity of the beef and dairy industries depends directly on the reproductive efficiency and genetic gain of the herd, which are directly associated with the appropriate use of Assisted Reproductive Technologies (ARTs). The objective of this review is to show from a Brazilian perspective the evolution over the last 40 years of ARTs related to ovulation resynchronization programs and embryo transfer in cattle. Despite significant improvements and high fertility obtained in timed artificial insemination (TAI) protocols (Sales et al., 2024 - Part I), the improvement of the use of in vitro-produced embryos, development of resynchronization programs, and the advance in Doppler ultrasonography (Doppler-US) for reproductive assessments of bovine females were the ARTs that presented the greatest relevance on reproductive effectiveness in cattle. In the last seven years, the embryo transfer (ET) technology using in vitro-produced (IVP) embryos took over the conventional ET of in vivo produced embryos after donor's superovulation. Also, procedures and pregnancy rates after ET of IVP embryos were improved in dairy and beef operations. The Doppler-US allows the identification of non-pregnant females at an early stage based on the evaluation of blood perfusion of the corpus luteum. Recent studies in beef and dairy cows indicate satisfactory accuracy when Doppler-US is used at 20-22 days after TAI. Consequently, super-early resynchronization programs have been developed and are being implemented in commercial programs, thereby facilitating earlier conception through the use of semen from superior bulls, providing genetic and economic improvements in herds. Likewise, the assessment of luteal function by Doppler-US allows the selection of embryo recipients with greater receptivity, and consequently may increase the effectiveness of timed ET programs.

Abstract:Assisted reproductive technologies (ART) are fundamental for cattle breeding and sustainable food production. Together with genomic selection, these technologies contribute to reducing the generation interval and accelerating genetic progress. In this paper, we discuss advancements in technologies used in the fertility evaluation of breeding animals, and the collection, processing, and preservation of the gametes. It is of utmost importance for the breeding industry to select dams and sires of the next generation as young as possible, as is the efficient and timely collection of gametes. There is a need for reliable and easily applicable methods to evaluate sexual maturity and fertility. Although gametes processing and preservation have been improved in recent decades, challenges are still encountered. The targeted use of sexed semen and beef semen has obliterated the production of surplus replacement heifers and bull calves from dairy breeds, markedly improving animal welfare and ethical considerations in production practices. Parallel with new technologies, many well-established technologies remain relevant, although with evolving applications. In vitro production (IVP) has become the predominant method of embryo production. Although fundamental improvements in IVP procedures have been established, the quality of IVP embryos remains inferior to their in vivo counterparts. Improvements to facilitate oocyte maturation and development of new culture systems, e.g. microfluidics, are presented in this paper. New non-invasive and objective tools are needed to select embryos for transfer. Cryopreservation of semen and embryos plays a pivotal role in the distribution of genetics, and we discuss the challenges and opportunities in this field. Finally, machine learning (ML) is gaining ground in agriculture and ART. This paper delves into the utilization of emerging technologies in ART, along with the current status, key challenges, and future prospects of ML in both research and practical applications within ART.
Analysis of reproductive performance of lactating cows on large dairy farms using machine learning algorithms

Abstract:The fertility of lactating dairy cows is economically important, but the mean reproductive performance of Holstein cows has declined during the past 3 decades. Traits such as first-service conception rate and pregnancy status at 150 d in milk (DIM) are influenced by numerous explanatory factors common to specific farms or individual cows on these farms. Machine learning algorithms offer great flexibility with regard to problems of multicollinearity, missing values, or complex interactions among variables. The objective of this study was to use machine learning algorithms to identify factors affecting the reproductive performance of lactating Holstein cows on large dairy farms. This study used data from farms in the Alta Genetics Advantage progeny-testing program. Production and reproductive records from 153 farms were obtained from on-farm DHI-Plus, Dairy Comp 305, or PCDART herd management software. A survey regarding management, facilities, labor, nutrition, reproduction, genetic selection, climate, and milk production was completed by managers of 103 farms; body condition scores were measured by a single evaluator on 63 farms; and temperature data were obtained from nearby weather stations. The edited data consisted of 31,076 lactation records, 14,804 cows, and 317 explanatory variables for first-service conception rate and 17,587 lactation records, 9,516 cows, and 341 explanatory variables for pregnancy status at 150 DIM. An alternating decision tree algorithm for first-service conception rate classified 75.6% of records correctly and identified the frequency of hoof trimming maintenance, type of bedding in the dry cow pen, type of cow restraint system, and duration of the voluntary waiting period as key explanatory variables. An alternating decision tree algorithm for pregnancy status at 150 DIM classified 71.4% of records correctly and identified bunk space per cow, temperature for thawing semen, percentage of cows with low body condition scores, number of cows in the maternity pen, strategy for using a clean-up bull, and milk yield at first service as key factors.

Abstract:Female fertility is the foundation of the cow–calf industry, impacting both efficiency and profitability. Reproductive failure is the primary reason why beef cows are sold in the U.S. and the cause of an estimated annual gross loss of USD 2.8 billion. In this review, we discuss the status of the genomics, transcriptomics, and systems genomics approaches currently applied to female fertility and the tools available to cow–calf producers to maximize genetic progress. We highlight the opportunities and limitations associated with using genomic and transcriptomic approaches to discover genes and regulatory mechanisms related to beef fertility. Considering the complex nature of fertility, significant advances in precision breeding will rely on holistic, multidisciplinary approaches to further advance our ability to understand, predict, and improve reproductive performance. While these technologies have advanced our knowledge, the next step is to translate research findings from bench to on-farm applications.

Efficiency of Two Protocols of Resynchronization of Estrus and Ovulation in High-Producing Dairy Cows at Peak Lactation

Abstract:Abstract The reproductive efficiency of the cows was monitored after two resynchronization protocols: Ovsynch (OVS) and Double Ovsynch (DOS). The research initially included 70 HF cows who entered the first synchronization protocol – Presynch. Cows that did not conceive after the first synchronization were divided into two groups and introduced to two resynchronization protocols. In the first group of cows (n=35), the DOS protocol began with the application of GnRH on day 22 after the Presynch TAI (Timed Artificial Insemination), and seven days later pregnancy check-up was done and PGF 2α was applied only to non-pregnant cows (n=23), which remained in the study. In the second group of cows, the OVS protocol started on day 32 after Presynch TAI only in non-pregnant animals (n=20). Progesterone (P4) concentration was determined at the time of application of GnRH1, PGF 2α and GnRH2 in both groups of cows, and then 30 days after Resynch TAI, ultrasound pregnancy diagnosis was done. A higher percentage of pregnant cows were recorded in the OVS group compared to the DOS group (45% and 35%, respectively). The concentration of P4 in the serum of cows in the DOS group during the first measurement (GnRH1) was significantly higher than the value in cows that did not conceive (p<0.05), while in the third measurement (GnRH2) the average concentration of P4 in conceiving cows was significantly lower (p<0.001) compared to cows that did not coincive. The open days period was significantly longer in pregnant cows that were resynchronized using the DOS protocol compared to cows from the OVS protocol. In conclusion, the OVS protocol of estrus resynchronization in dairy cows proved to be more successful than the DOS protocol. However, considering the advantages the OVS, it is needed to determine which day of the sexual cycle is the best to start resynchronization.

Abstract:Female infertility represents a major challenge for improving the production efficiency in the dairy industry. Historically, fertility has declined whereas milk yield has increased tremendously due to intensive genetic selection. In vivo evidence reveals about 60% pregnancy loss takes place during the first month following fertilization. Meanwhile, early embryo development is significant for somatic cell nuclear transfer in cattle as a large proportion of cloned embryos fail to develop beyond peri-implantation stage. Oocyte quality is of utmost importance for the early embryo to develop to term for both fertilized and cloned embryos. Epigenetic reprogramming is a key process occurring after fertilization and critical roles of epigenetic modifiers during preimplantation development are now clear. Incomplete epigenetic reprogramming is believed to be a major limitation to cloning efficiency. Treatment of cloned embryos with epigenetic modifying drugs (e.g., Trichostatin A) could greatly improve cloning efficiency in both mice and cattle. Recently, the rapid progress in high-throughput sequencing technologies has enabled detailed deciphering of the molecular mechanisms underlying these events. The robust efficiency of genomic editing tools also presents an alternative approach to the functional annotation of genes critical to early development.

Abstract:The objective of this randomized controlled experiment was to evaluate the effect of reproductive management programs that prioritized artificial insemination (AI) at detected estrus (AIE) or timed AI (TAI) during the first lactation on the economic performance of dairy cows of different genomically enhanced predicted transmitting ability for fertility. Lactating primiparous Holstein cows from 6 commercial farms were stratified into high, medium, and low fertility groups based on a reproduction index value calculated from multiple genomically enhanced predicted transmitting abilities to predict the number of days to achieve pregnancy. Within herd and fertility group, cows were randomly assigned either to a program that prioritized AIE (P-AIE; n = 1,416) and used TAI for cows not AIE for all AI services or another that prioritized TAI and had an extended voluntary waiting period for first service and prioritized TAI for second and greater AI services (P-TAI; n = 1,338). Cash flow (CF) per cow accumulated for the experimental (first) and second calving interval (CIN) and cash flow per slot per 28 mo after calving in the experimental lactation were calculated. Market and rearing heifer cost values were used for estimating CF. For cows in the high fertility group, a positive effect of delayed pregnancy on milk income during the first lactation was observed (+$248 for P-TAI) but was insufficient to generate significant differences in CF between treatments mainly because of milk income compensation in the second lactation (+$125 for P-AIE) and minor reductions in reproductive cost and gains in calf value for the P-AIE treatment. In this regard, CF for 2 CIN was greater for the P-TAI treatment by $61 and $86 for market and rearing replacement heifer cost, respectively. Similarly, CF per slot was favorable to the P-TAI treatment but only by $13 and $47 for market and rearing replacement heifer cost, respectively. For cows in the low fertility group, CF was numerically in favor of the P-AIE treatment due to a pregnancy and herd exit dynamics that resulted in gains in milk income over feed cost during the first ($29) and second ($113) lactation. Differences in CF for the 2 CIN were $58 and $47 for market or rearing heifer value, respectively, and $77 and $19 for market and rearing heifer values, respectively for the slot analysis. Differences in CF between cows of different genetic merit for fertility were consistent across treatment and estimation method. Of note, cows in the low fertility group had greater CF than cows in the high fertility group in all comparisons, ranging from $198 per cow for 2 CIN to as much as $427 per slot. For the low fertility group, greater milk production contributed directly (milk income over feed cost) and indirectly (reduced culling) to increased CF. We concluded that genetic merit for fertility and CF are associated because cows of inferior genetic potential for fertility had greater CF than cows of superior genetic for fertility despite some increased costs and reduced revenues. Also, the magnitude of the CF differences observed for cows of different genetic merit for fertility managed with the P-AIE or P-TAI program may be valuable to commercial dairy farms but did not allow to conclusively support the choice of a type of reproductive management strategy for cows of different genetic merit for fertility.
Control of pig reproduction in a breeding programme

Abstract:Important improvements in the management of sows have been made over the past 20 years in Europe and the U.S.A. Nevertheless, annual productivity varied between 15 and 25 piglets weaned per sow per year in France for 1983. Reasons for such a difference in productivity have been analysed quantitatively. Marked differences exist between herds in the genotype of pig used and in the age of puberty. Transportation acts as a stress stimulus and stimulates puberty attainment. However, there is variability in the occurrence of oestrus amongst different purchased batches of pigs. Recent endocrine results led to four phases in sexual maturation of gilts being distinguished (perinatal, infancy, activation and resting) and permitted a better understanding of the variation in the onset of puberty. After a lactation of 3 weeks or more, about 25% of primiparous sows did not return to oestrus within 1 week of weaning. Fertility of these sows with a delayed oestrus was 10% lower than in sows having a normal oestrus after weaning. The past 3 decades of research on control of ovulation have yielded positive results. However, there is no satisfactory method for the induction of puberty. One method used to regulate the oestrous cycle in gilts is treatment with a progestagen. This method is still available commercially in France. The close synchronization obtained with a progestagen treatment has led to the use of AI at fixed days (Days 6 and 7) in crossbred gilts but not for purebred gilts. Some environmental factors (e.g. male stimulation, age of gilts at treatment, diet and month of AI) have been analysed to evaluate their effects on farrowing rate and litter size. After weaning, a short treatment with a progestagen (20 mg/day) tends to improve the synchronization of oestrus (3-9 days), especially in primiparous sows. Farrowing rate and litter size are similar in treated sows and control sows. However, this technique does not permit fixed-time AI without detection of oestrus. Diagnosis of pregnancy was successful with ultrasound scanning. This technique can be performed 20 days after insemination if the sow is tied up during the examination and at 22 days if the sow is restrained in a retention box. From 22 days after mating, errors made for positive and negative diagnoses were less than 5%. These techniques contribute to a better control of reproduction in different management systems.
Altered release of growth hormone and thyrotropin induced by endotoxin in the rat.

Abstract:Plasma growth hormone (GH) and thyrotropin (TSH) levels were measured in freely behaving rats for 6 h on three consecutive days. On the 1st and 3rd days there was no treatment, and on the 2nd day endotoxin (150 mg/kg) was administered intravenously. Endotoxin causes a biphasic temperature response: an initial hypothermia persisting for 1 h, followed by hyperthermia evident from 5 to 8 h after injection. Normal pulsatile GH release observed on the 1st day was abolished on the 2nd day by the endotoxin treatment. On the 3rd day, however, GH secretion was greater than on the initial control day. TSH release was also suppressed by endotoxin and showed a rebound release on the subsequent day. The suppression of GH secretion by endotoxin was reversed in all animals by antisomatostatin serum, and the suppression of TSH secretion by endotoxin was reversed in some animals. These results suggest that endotoxin is a potent stimulus for hypothalamic somatostatin release in the rat. Endotoxin profoundly alters adenohypophyseal hormone release.
Artificial insemination and optimization of the use of seminal doses in swine

Abstract:The optimization of processes associated with artificial insemination (AI) is of great importance for the success of the pig industry. Over the last two decades, great reproductive performance has been achieved, making further significant progress limited. Optimizing the AI program, however, is essential to the pig industry's sustainability. Thus, the aim is not only to reduce the number of sperm cells used per estrous sow but also to improve some practical management in sow farms and boar studs to transform the high reproductive performance to a more efficient program. As productivity is mainly influenced by the number of inseminated sows, guaranteeing a constant breeding group and with healthy animals is paramount. In the AI studs, all management must ensure conditions to the health of the boars. Some strategies have been proposed and discussed to achieve these targets. A constant flow of high-quality and well-managed breeding groups, quality control of semen doses produced, more reliable technology in the laboratory routine, removal of less fertile boars, the use of intrauterine AI, the use of a single AI with control of estrus and ovulation (fixed-time AI), estrus detection based on artificial intelligence technologies, and optimization regarding the use of semen doses from high genetic-indexed boars are some strategies in which improvement is sought. In addition to these new approaches, we must revisit the processes used in boar studs, semen delivery network, and sow farm management for a more efficient AI program. This review discusses the challenges and opportunities in adopting some technologies to achieve satisfactory reproductive performance and efficiency.
Influences of nutrition and metabo lism on fertility of dairy cows

Abstract:During early postpartum, high-producing dairy cows undergo a period of extensive tissue catabolism because of negative nutrient balance. Homeorrhetic controls assure that nutrients are partitioned to favor lactation at the same time that homeostasis secures survival. However, unrestrained metabolic disturbances often lead to diseases which, in turn, dramatically decrease both productive and reproductive performance. Negative nutrient balance has been associated with compromised immune and reproductive functions in dairy cows. Low circulating concentrations of glucose and insulin associated with elevated concentrations of non-esterified fatty acids and ketone bodies postpartum have disruptive and detrimental effects on the oocyte, granulosa and immune cells. Negative nutrient balance is associated with changes in the pattern of ovarian follicle growth which can indirectly affect oocyte quality. Some of this disruption seems to be the result of endocrine and biochemical changes that alter the microenvironment of the growing and maturing oocyte. In addition, cows under negative nutrient balance have extended periods of anovulation. Postpartum anestrus, as well as infertility, is magnified by losses of body condition during the early postpartum period. The underlying mechanism for resumption of ovulatory cycles seems to be associated with metabolic signals and regulatory hormones primarily insulin and insulinlike growth factor (IGF)-1, which link nutritional status with gonadotropin secretion, recoupling of the growth hormone-IGF system, and follicle maturation and ovulation. Feeding diets that promote increases in plasma glucose and insulin may improve the metabolic and endocrine status of cows in early lactation. Furthermore, fertility in postpartum cows is also determined by uterine health. Reductions in circulating concentrations of Ca and antioxidant vitamins around parturition are also linked with impaired immune competence and result in greater risk of uterine diseases that impair reproduction. Specific nutrients and dietary ingredients have been implicated to affect reproduction in cattle. Excess intake of dietary protein has been suggested as detrimental to fertility, although feeding excess of dietary protein can no longer be justified. Addition of moderate amounts of supplemental fat to the diet improves caloric intake, modulates prostaglandin F2 secretion by the uterus, affects ovarian dynamics, enhances luteal function and embryo quality, and has moderate positive effects on fertility. More specifically, some fatty acids might impact fertilization rate and embryo quality in dairy cows. On the contrary, some dietary ingredients, such as gossypol, when ingested in large quantities decrease fertility of dairy cows because of its negative effects on embryo quality and pregnancy maintenance.

Economic impact of different strategies to use sex-sorted sperm for reproductive management in seasonal-calving, pasture-based dairy herds

Abstract:To maximize efficiency, profitability, and societal acceptance of modern dairy production, it is important to minimize the production of male dairy calves with poor beef merit. One solution involves using sex-sorted sperm (SS) to generate dairy replacements and breeding all other cows to an easy-calving, short-gestation bull with good beef merit. We used the Pasture Based Herd Dynamic Milk Model to investigate the effect of herd fertility and use of SS on farm net profit in a herd of 100 cows. This was completed by simulating herds with differing fertility performance (good, average, poor), and differing farm reproductive management [conventional semen (CONV) or SS with varying pregnancy per artificial insemination (P/AI) relative to CONV (i.e., relative P/AI 100%, 85%, and 70%)]. As an additional consideration, the method of allocating SS to cows was also examined. The first option used SS on random heifers and cows (S). The second option used SS on heifers and targeted high-fertility cows (SSel). The final option was similar to SSel, but used a fixed-time artificial insemination (AI) protocol to facilitate AI on the farm mating start date (SSync). For CONV, dairy breed semen was used for AI until 50 animals were pregnant (50% chance of a female calf), whereas for S, SSel, or SSync the target number of animals successfully conceiving with SS was set at 28 (based on assumed 90% chance of a female calf from pregnancies derived from SS). Beef breed semen was used on all other dams. The results indicated that the biggest effect on farm net profit was not based on whether or not SS was used, but instead was most affected by the overall fertility performance of the herd. Total farm profit decreased by 10% between the good and average fertility herds, and decreased by a further 12% between the average and poor fertility herds. In almost all situations, when the relative P/AI with SS was ≥85%, use of SS led to an overall increase of the farm net profit. There was an economic benefit of using either SSel or SSync compared with S for the average and poor fertility herds but not for the good fertility herd, highlighting an interaction between SS P/AI and overall herd fertility as well as management practices. If the relative P/AI with SS was <70%, the use of SS led to a decrease in profitability in all simulations except for SSync, highlighting the importance of a good management strategy for use of SS. The findings in this study indicated that SS has significant potential to help facilitate greater integration between the dairy and beef production sectors, as well as increase farm profitability when used appropriately.
