Abstract:Excessive or unfolded proteins accumulation in endoplasmic reticulum (ER) will cause ER stress, which has evolved to involve in various metabolic diseases. In particular, ER stress plays an important role in the pathogenesis of diabetes. Both ER stress and course of diabetes accompany oxidative stress and production of reactive oxygen species (ROS), among which superoxide anion (O2•-) is the first produced ROS and has been recognized as cell signaling mediator involved in the physiological and pathological process of diabetes. Hence, the development of effective monitoring methods of O2•- in live cells and in vivo is of great importance for ascertaining the onset and progress of related diseases. Herein, a new endoplasmic reticulum-targeted two-photon fluorescent probe termed ER-BZT is designed and synthesized for imaging of O2•-. The probe ER-BZT shows high sensitivity, selectivity, stability, and low cytotoxicity. Based on these superior properties, the rise of O2•- levels in endoplasmic reticulum induced with different stimuli is visualized by one- and two-photon fluorescence imaging. Most importantly, by utilizing ER-BZT, the two-photon fluorescence imaging results demonstrate that the endogenous O2•- concentration in abdominal or hepatic tissue of diabetic mice is higher than that in normal mice. Meanwhile, after treated with metformin, a broad-spectrum antidiabetic drug, the diabetic mice exhibit depressed O2•- level. The proposed two-photon probe, ER-BZT might serve as perfect tool to image the O2•- fluctuations and study the relevance between O2•- and various diseases in live cells and in vivo.
Fluorescent Probes with Dual-Targeting Organelles Monitor Polarity in Non-Alcoholic Fatty Liver Disease.

Abstract:Non-alcoholic fatty liver disease (NAFLD) becomes a world health issue due to its rising prevalence and lack of a definitive pathogeny. However, the excessive accumulation of fat droplets has been recognized as a crucial characteristic of NAFLD, accompanied with endoplasmic reticulum stress in its onset and progression as well. Therefore, real-time monitoring the dynamic of lipid droplets (LDs) and endoplasmic reticulum (ER) within cells is paramount. In this regard, four D-A-π-D structural fluorescent probes COB1-COB4 were designed and synthesized wherein coumarin connected with carbazole acted as precursors while the side chains attached to carbazole groups are different. Here, probes COB1-COB4 exhibited high sensitivity towards polarity, while COB2 was chosen for further study attributing to its excellent anti-interference property. Cell imaging demonstrated that COB2 could accurately target both LDs and ER at the same time and monitor the changes of the two organelles under different physiological conditions. Notably, probe COB2 also exhibited the ability to distinguish normal liver from fatty liver at the tissue level. The above results lay an experimental foundation for developing novel dual-targeted probes with potential for early diagnosis of non-alcoholic fatty liver.
Visualization of Polarity Changes in Endoplasmic Reticulum (ER) Autophagy and Rheumatoid Arthritis Mice with Near-Infrared ER-targeted Fluorescent Probe

Abstract:The crucial cellular activities for maintaining normal cell functions heavily rely on the polarity of the endoplasmic reticulum (ER). Understanding how the polarity shifts, particularly in the context of ER autophagy (ER-phagy), holds significant promise for advancing knowledge of disorders associated with ER stress. Herein, a polarity-sensitive fluorescent probe CDI was easily synthesized from the condensation reaction of coumarin and dicyanoisophorone. CDI was composed of coumarin as the electron-donating moiety (D), ethylene and phenyl ring as the π-conjugation bridge, and malononitrile as the electron-accepting moiety (A), forming a typical D-π-A molecular configuration that recognition in the near-infrared (NIR) region. The findings suggested that as the polarity increased, the fluorescence intensity of CDI decreased, and it was accompanied by a redshift of emission wavelength at the excitation wavelength of 524 nm, shifting from 641 nm to 721 nm. Significantly, CDI exhibited a notable ability to effectively target ER and enabled real-time monitoring of ER-phagy induced by starvation or drugs. Most importantly, alterations in polarity can be discerned through in vivo imaging in mice model of rheumatoid arthritis (RA). CDI has been proven effective in evaluating the therapeutic efficacy of drugs for RA. ER fluorescent probe CDI can be optically activated in lysosomes, providing a sensitive tool for studying ER-phagy in biology and diseases.

Abstract:The reduced coenzymes (NADH and NADPH) are an important product in energy metabolism and closely related to the occurrence and development of cancer. So it is necessary to use a powerful detection tool to visualize NAD(P)H in energy metabolism of tumor cells and find a new strategy to improve cancer treatment based on NAD(P)H. Herein, a novel multifunctional probe (Cy-N) is synthesized with good near-infrared fluorescence (NIRF) response to NAD(P)H and the photoacoustic (PA) and photothermal properties are successfully activated by NAD(P)H. The probe is successfully applied in visualizing NAD(P)H in energy metabolism of tumor cells and imaging NAD(P)H in bacteria. Moreover, the probe can be used to image NAD(P)H in energy metabolism of tumor-bearing mice by dual-modal imaging (NIRF and PA). More importantly, in terms of the role of NAD(P)H in energy metabolism, the photothermal therapy (PTT) is activated by NAD(P)H and a novel strategy of enhanced PTT is proposed by injecting glucose. As far as we know, this is the first probe to detect NAD(P)H in energy metabolism through dual-modal imaging, and also the first probe to activate PTT based on NAD(P)H, which will provide important information of the diagnosis and treatment of cancer.

Abstract:Diabetes is one of the metabolic diseases marked by hyperglycemia and is often accompanied by the occurrence of some complications. As a biomarker of oxidative stress, hydrogen peroxide (H2O2) has close association with the occurrence and development of diabetes and its complications. Unfortunately, there is no fluorescent probe reported for imaging H2O2 in diabetic mice. Here, a novel near-infrared (NIR) fluorescent probe named QX-B was designed and synthesized to detect H2O2. For the probe, the quinolinium-xanthene dye is used as the fluorophore and borate ester is chosen as the response group. After the addition of H2O2, a strong MR fluorescence signal at 772 nm is observed. The probe not only shows high sensitivity with 10-fold enhancement but also displays excellent selectivity to H2O2 over other possible interfering species. In the meantime, the possible response mechanism of QX-B toward H2O2 was proposed and verified by the high-performance liquid chromatography (HPLC) experiment, mass spectra (MS) experiment, and density functional theory (DFT) calculation. Furthermore, based on the low cell cytotoxicity of QX-B, it has been applied in imaging exogenous and endogenous H2O2 in HeLa cells, HCT116 cells, 4T1 cells, and zebrafish successfully. More importantly, inspired by the performance of NIR fluorescence, QX-B has been used in monitoring H2O2 in diabetic mice for the first time. This provides very important information for the diagnosis and treatment of diabetes and its complications.

Abstract:Tracking signaling H2S in live mice demands responsive imaging with fine tissue imaging depth and low interferences from tissue scattering/autofluorescence and probe concentration. With complementary advantages of fluorescence and photoacoustic (PA) imaging, optical/PA dual-modality imaging was suggested for in/ex vivo H2S imaging. Therefore, a meso-benzoyloxyltricarboheptamethine cyanine, HS-CyBz, was prepared as the first ratiometric optical/PA dual-modality probe for H2S, profiting from a keto-enol transition sensing mechanism. Tail intravenous injection of this probe leads to probe accumulation in the liver of mice, and the endogenous H2S upregulation triggered by S-adenosyl-l-methionine has been verified by ratiometric optical/PA imaging, suggesting the promising potential of this ratiometric dual-modality imaging.
Visualizing Detection of Diabetic Liver Injury by a Biomarker-Activatable Probe via NIR-II Fluorescence Imaging

Abstract:Diabetes characterized by hyperglycemia is a chronic metabolic disease with increasing prevalence worldwide, and over time it leads to damage to various organs including the liver. As the liver is the major organ for glucose metabolism, patients with diabetes are prone to liver injuries or damages; and without timely and effective treatment, diabetic liver injury may further deteriorate into serious or life-threatening complications including liver cancer or liver failure. Hence, the accurate detection of diabetic liver injury is of great significance for diabetic patients in terms of improving quality of life. Herein, a fluorescent probe (TTX-P) has been developed for in situ detection and imaging of diabetic liver injury in the near-infrared second-window (NIR-II) region. In routine clinical practice, alkaline phosphatase (ALP) is one of the most commonly assayed enzymes, and thus serving as an important diagnostic biomarker for liver dysfunction or injury. The probe TTX-P responds in situ to the overexpressed ALP in liver and thus giving out strong NIR-II fluorescence as the reporting signals for detection and imaging. The NIR-II probe TTX-P consists of the NIR-II chromophore TTX-OH as the fluorophore and phosphate group as the responsive unit. Without the presence of ALP, the probe TTX-P displays weak fluorescence because the electron-withdrawing phosphate group weakens the electron-pushing capability of the electron-donor side of the NIR-II fluorophore. While in the presence of ALP, the phosphate group is cleaved by the enzyme and consequently the fluorophore emits evident NIR-II fluorescence. The probe TTX-P has been applied in imaging liver injury in diabetic mice. The approach provides a utilitarian means for visualizing detection of diabetic liver injury in mice with NIR-II fluorescence imaging.

Abstract:AbstractReversible imaging probes that allow for the dynamic visualization of the redox cycle between hydroxyl radical (⋅OH) and hydrogen sulfide (H2S) are vital to probe the redox imbalance‐involved pathological process in vivo. Herein, we report a reversible ratiometric photoacoustic (PA) imaging nanoprobe (1‐PAIN) for the real‐time imaging of ⋅OH/H2S redox cycle in vivo.1‐PAIN displays a low PA ratio between 690 and 825 nm (PA690/PA825), which significantly increases by ≈5‐fold upon oxidation by ⋅OH, and is switched back to the initially low PA690/PA825value upon reduction by H2S.1‐PAIN could dynamically report on the hepatic ⋅OH production in mice during the lipopolysaccharide (LPS)‐induced liver inflammation process, and visualize hepatic H2S generation during theN‐acetyl cysteine (NAC)‐induced anti‐inflammation process.1‐PAIN can act as a useful tool to probe the redox state in living biology, beneficial for the study of redox imbalance‐related diseases.
Polarity-sensitive Fluorescent Probe Based on Coumarin Derivatives for Imaging Lipid Droplets and Diagnosis of Non-Alcoholic Fatty Liver

Abstract:Aberrant alterations in the polarity of lipid droplets (LDs) are closely linked to disorders of lipid metabolism, which can result in LDs dysfunction and the onset of various diseases, including non-alcoholic fatty liver disease (NAFLD) and specific types of cancer. Therefore, it is imperative to comprehensively comprehend and accurately diagnose associated diseases by monitoring changes in LDs polarity in real time. In this paper, we designed and synthesized four novel fluorescent probes XOT1-XOT4 by using coumarin as a polarity-sensitive group and incorporate various triphenylamine derivatives as electron-donating groups, enabling specific targeting of LDs. All four probes possess high sensitivity towards minute polarity changes. Among them, XOT3 exhibited excellent photophysical properties and demonstrated a linear fluorescence change (R-2 = 0.98596) within the polarity range of Delta f = 0.023496 similar to 0.047464. Moreover, probe XOT3 could be effectively utilized for visualizing intracellular LDs by precisely targeting them without requiring any washing steps. Importantly, probe XOT3 has been successfully utilized to monitor alterations in the polarity of LDs during apoptosis and ferroptosis as well as the quantity, size, and fluorescence intensity of LDs stimulated by oleic acid. Furthermore, probe XOT3 was capable of effectively visualizing LDs in tissues and organs in mouse models of non-alcoholic fatty liver disease (NAFLD), which provides a powerful tool for the early diagnosis of NAFLD.

Abstract:Recent investigations have revealed the tight connections between carboxylesterase (CE) and the lipid metabolism-associated disorders. In this work, based on the previous reports, a photoacoustic (PA) probe Hcy-CE was developed for the in vivo imaging of CE in two lipid metabolism disorders. The reaction with CE led to the departure of the carbamate quencher together with the linkage, thus to release of the reporter Hcy to exhibit the turn-on signals of PA reporting accompanied with the absorbance and fluorescence modes. The responses in all three modes showed high consistency. In the PA mode, the linear correlation range was CE level 0–10 U/L, while the limit of detection value was 0.52 U/mL. The optical performance of the probe Hcy-CE in the solution system was practical with high steadiness and selectivity. Moreover, Hcy-CE exhibited potential capability of monitoring the CE level in the induced foam cells. In particular, Hcy-CE realized the in vivo imaging of the CE level from multiple views and continuous cross-sections in the model mice of different groups. The combination of the PA imaging with the blood biochemical tests and immunohistochemically staining methods might help distinguish the three statuses of different mice from each other. This work provided meaningful information for the accurate diagnosis and regulation of the lipid metabolism-related disorders.

Abstract:Drug-induced liver injury (DILI) poses a severe threat to public health. Endoplasmic reticulum (ER) stress contributes significantly to DILI pathogenesis, with peroxynitrite (ONOO-) identified as a pivotal indicator. However, the temporal and spatial fluctuations of ONOO- associated with ER stress in the pathogenesis of DILI remain unclear. Herein, a novel ER-specific near-infrared (NIR) probe (QM-ONOO) with aggregation-induced emission (AIE) features for monitoring ONOO- fluctuations in DILI was elaborately constructed. QM-ONOO exhibited excellent ER-targeting specificity, a large Stoke's shift, and a low detection limit (26.9 nM) toward ONOO-. QM-ONOO performed well in imaging both exogenous and endogenous ONOO- in HepG2 cells. Furthermore, molecular docking calculations validated the ER-targeting mechanism of QM-ONOO. Most importantly, using this probe allowed us to intuitively observe the dynamic fluctuations of ONOO- during the formation and remediation processes of DILI in the acetaminophen (APAP)-induced mouse model. Consequently, this work provides a promising tool for in-depth research of ONOO- associated pathological processes in DILI.
A Bright Two‐Photon Lipid Droplets Probe with Viscosity‐Enhanced Solvatochromic Emission for Visualizing Lipid Metabolic Disorders in Deep Tissues

Abstract:A two‐photon lipid droplets probe displaying aggregation‐induced emission, high polarity sensitivity, high brightness, and superior tissue permeability is facilely developed for monitoring lipid metabolic disorders in deep tissues. The results demonstrate that the lipids deposition as well as their polarity distribution in hyperlipoidemia mouse tissues are clearly different from tissues of normal mouse. The polarity of lipid droplets (LDs) plays an important role in pathological processes associated with abnormal lipid metabolism. Monitoring the variation of LDs polarity in cells and tissues is of great importance in biomedical research and clinical diagnosis. However, developing fluorescent LDs‐specific probes with high polarity sensitivity, brightness, and permeability for deep tissue imaging is still challenging. Herein, a push–pull fluorescent luminogen (DPBT) with aggregation‐induced emission, strong solvatochromism, large Stokes shift, high solid‐state fluorescence efficiency and superior two‐photon absorption is facilely developed. The lipophilic DPBT can specifically stain LDs with high biocompatibility and good photostability. The viscosity‐enhanced solvatochromic emission property enables DPBT to visualize LDs polarity with high brightness and imaging contrast, and deep penetration depth under two‐photon microscopy. DPBT can specifically stain lipids in various mouse tissues (atherosclerotic plaque, liver, and mesenteric adipose tissues) and map their polarity distribution to reflect lipid metabolic states within those tissues. It is found that the lipids deposition as well as their polarity distribution in tissues of hyperlipoidemia mouse are clearly different from the tissues of the normal mouse. Its excellent properties make DPBT a promising candidate for investigating LDs‐associated physiological and pathological processes in live biological samples.

Abstract:Dynamic variations in the concentration and abnormal distribution of endogenous biomarkers are strongly associated with multiple physiological and pathological states. Therefore, it is crucial to design imaging systems capable of real-time detection of dynamic changes in biomarkers for the accurate diagnosis and effective treatment of diseases. Recently, ratiometric imaging has emerged as a widely used technique for sensing and imaging of biomarkers due to its advantage of circumventing the limitations inherent to conventional intensity-dependent signal readout methods while also providing built-in self-calibration for signal correction. Here, the recent progress of ratiometric probes and their applications in sensing and imaging of biomarkers are outlined. Ratiometric probes are classified according to their imaging mechanisms, and ratiometric photoacoustic imaging, ratiometric optical imaging including photoluminescence imaging and self-luminescence imaging, ratiometric magnetic resonance imaging, and dual-modal ratiometric imaging are discussed. The applications of ratiometric probes in the sensing and imaging of biomarkers such as pH, reactive oxygen species (ROS), reactive nitrogen species (RNS), glutathione (GSH), gas molecules, enzymes, metal ions, and hypoxia are discussed in detail. Additionally, this Review presents an overview of challenges faced in this field along with future research directions.

Liver cancer can develop from viral hepatitis, alcoholic liver, and fatty liver, thus making the above diseases share common features such as elevated viscosity, reactive oxygen species, and reactive nitrogen species. Therefore, accurate differentiation between other liver diseases and liver cancer is both a paramount practical need and challenging. Numerous fluorescent probes have been reported for the diagnosis of liver cancer by detecting a single biomarker, but these probes lack specificity for liver cancer in complex biological systems. Obviously, using multiple liver cancer biomarkers as the basis for judgment can dramatically improve diagnostic accuracy. Herein, we report the first fluorescent probe, LD-TCE, that sequentially detects carboxylesterase (CE) and lipid droplet polarity in liver cancer cells with high sensitivity and selectivity, with linear detection of CE in the range of 0-6 U/mL and a 65-fold fluorescence enhancement in response to polarity. The probe first reacts with CE and releases weak fluorescence, which is then dramatically enhanced due to the decrease in lipid droplet polarity in liver cancer cells. This approach allows the probe to enable specific imaging of liver cancer with higher contrast and accuracy. The probe successfully achieved the screening of liver cancer cells and the precise identification of liver cancer in mice. More importantly, it is not disturbed by liver fibrosis, which is a common pathological feature of many liver diseases. We believe that the LD-TCE is expected to be a powerful tool for early diagnosis of liver cancer.

Abstract:Nowadays, it has been proven that lipid droplets (LDs) not only maintain the fundamental cellular functions, but also play an essential role in the pathogenesis of numerous diseases. Non-alcoholic fatty liver disease (NAFLD) is among these diseases. In this work, we designed two polarity sensitive fluorescent probes TST and TSO with D-π-A-D structure by introducing different electron acceptor groups according to the low polarity of LDs. The experimental discovered that probe TST exhibited the characteristics of near-infrared emission, high selectivity towards polarity, large Stokes shift, rapid targeting ability of LDs, and robust wash-free biological imaging capability. Confocal images illustrated that probe TST has been successfully applied in monitoring LDs polarity during ferroptosis, as well as visualizing changes in LDs polarity at both tissue and organ levels in fatty liver conditions. With these exceptional properties, probe TST was anticipated to make further contributions to the field of LDs research.

Abstract:Drug-induced liver injury (DILI) is a frequent cause of hepatic dysfunction as well as the single most frequent reason for removing approved medications from the market, and multispectral optoacoustic tomography (MSOT) is an emerging and noninvasive imaging modality for diagnosing and monitoring diseases. Herein, we report an activatable optoacoustic probe for imaging DILI through detecting the activity of leucine aminopeptidase (LAP). In this probe, an N-terminal leucyl moiety serving as the LAP recognition element is linked with a chromene-benzoindolium chromophore via 4-aminobenzylalcohol group. The elevated expression of hepatic LAP as a result of DILI cleaves the leucyl moiety and causes the red-shift of the probe's absorption band, thereby generating prominent optoacoustic signals for MSOT imaging. During this process, the probe also exhibits prominent NIR fluorescence, which can be utilized for fluorescent imaging. More importantly, by rendering stacks of cross-sectional images as maximal intensity projection (MIP) images, we could precisely locate the focus of drug-induced liver injury in mice. This probe is expected to serve a powerful tool for studying physiological and pathological processes related to LAP.
