Abstract:Drug-induced liver injury (DILI) not only poses a serious health threat to patients, but is also a major obstacle to the development of new drugs. The liver toxicity of many drugs is not detected until late in the development process or even after marketing, which significantly increases the cost of new drug development. This may be limited by the lack of tools to detect DILI. Studies have shown that abnormal changes in peroxynitrite (ONOO-) levels in the endoplasmic reticulum of liver cells may be an early manifestation of DILI. Therefore, the development of fluorescent probes to detect ONOO- in the endoplasmic reticulum may provide assistance in the detection of DILI and the screening of new drugs. Herein, we report a fluorescent probe, ER-N, capable of detecting ONOO- in the endoplasmic reticulum with high selectivity and sensitivity. The probe selects di(trifluoromethyl)benzene as the targeting group of the endoplasmic reticulum, which possesses excellent targeting to the endoplasmic reticulum, and provides a new design method for the establishment of other endoplasmic reticulum-targeting probes. In addition, the probe ER-N successfully realized the detection of endogenous and exogenous ONOO- in cells and zebrafish. More importantly, the probe ER-N was able to be used for tracing changes in the levels of ONOO- in the endoplasmic reticulum of cells and tissues under drug stimulation. Together, these results suggest that the probe ER-N has the potential to be a tool for DILI detection and new drug screening.

Abstract:Background: Over-consumption of drugs can result in drug-induced liver damage (DILI), which can worsen liver failure. Numerous studies have shown the significant role ferroptosis plays in the pathophysiology of DILI, which is typified by a marked imbalance between the generation and breakdown of lipid reactive oxygen species (ROS). The content of peroxynitrite (ONOO-) rapidly increased during this process and was thought to be a significant marker of early liver injury. Therefore, the construction of fluorescence probe for the detection and imaging of ONOO- holds immense importance in the early diagnosis and treatment of ferroptosis-mediated DILI. Results: We designed a probe DILI-ONOO based on the ICT mechanism for the purpose of measuring and visualizing ONOO- in ferroptosis-mediated DILI processes and associated studies. This probe exhibited significant fluorescence changes with good sensitivity, selectivity, and can image exogenous and endogenous ONOO- in cells with low cytotoxicity. Using this probe, we were able to show changes in ONOO- content in ferroptosis-mediated DILI cells and mice models induced by the intervention of acetaminophen (APAP) and isoniazid (INH). By measuring the concentration of ferroptosis-related indicators in mice liver tissue, we were able to validate the role of ferroptosis in DILI. It is worth mentioning that compared to existing alanine transaminase (ALT) and aspartate aminotransferase (AST) detection methods, this probe can achieve early identification of DILI prior to serious liver injury. Significance: This work has significant reference value in researching the relationship between ferroptosis and DILI and visualizing research. The results indicate a strong correlation between the progression of DILI and ferroptosis. Additionally, the use of DILI-ONOO shows promise in investigating the DILI process and assessing the effectiveness of medications in treating DILI.

Abstract:Drug-induced liver injury (DILI) poses a significant risk to human health. Increasing evidence indicates that the superoxide anion (O2•-), as the precursor of the other reactive oxygen species, is key in the pathological processes associated with DILI. Nonetheless, understanding of the mechanisms of DILI is difficult due to the lack of an imaging tool for monitoring the fluctuation of O2•- levels during the progression of DILI. Herein, we developed an upconversion nanoprobe (Rbh-UCNs) for in vivo ratiometric tracking of endogenous O2•- in DILI. In this design, the addition of O2•- triggers the luminescent resonance energy transfer between Rbh and UCNs, which significantly enhances absorption centered at 534 nm and translates into a distinct decrease of the UCL emission at 543 nm, while the UCL emission peak at 654 nm and 800 nm are not significantly affected, offering a ratiometric UCL signal for the quantitative detection of O2•-. In addition, Rbh-UCNs could effectively visualize endogenous O2•- in living cells, zebrafish, and liver tissues upon stimulation with PMA or cisplatin. More importantly, tissue imaging of the liver region of mice revealed that the fluctuation of O2•- levels is associated with DILI and the protective effect of L-carnitine against DILI. Altogether, this study provides an available method for a deeper comprehension of the mechanisms underlying DILI and accelerating the development process of hepatoprotective medicines.

Abstract:Drug-induced liver injury (DILI) is the most common cause for acute liver failure in the USA and Europe. However, most of DILI cases can recover or be prevented if treatment by the offending drug is discontinued. Recent research indicates that peroxynitrite (ONOO − ) can be a potential indicator to diagnose DILI at an early stage. Therefore, the establishment of an assay to detect and track ONOO − in DILI cases is urgently needed. Here, a FRET-based ratiometric nano fluorescent probe CD–N-I was developed to detect ONOO − with high selectivity and excellent sensitivity. This probe consists of carbon dots and a naphthalimide–isatin peroxynitrite sensing system assembled based on electrostatic interactions. Using CD–N-I we were able to detect exogenous ONOO − in live cells and endogenous ONOO − in APAP-induced liver injury of HepG2 cells.

Abstract:Peroxynitrite (ONOO-) is a potential biomarker of drug-induced liver injury (DILI) and is involved in the process of DILI. Therefore, developing a reliable detection method for ONOO- will greatly contribute to ensuring drug safety and improving treatment efficiency. Here, based on the previous work, two kinds of NIR fluorescence probes PN and SPN were developed with phenyl-hydrazine as the ONOO- recognition group, which based on two fluorophores RN and SRN that are stable to ONOO-. A sensitive NIR probe SPN with good water solubility, low detection limit and good biocompatibility was selected through in vitro spectral property screening. Further experimental results show that there is a good linear relationship between the response intensity of probe SPN to ONOO- and the concentration of ONOO-, and the detection limit can reach 19.7 nM. At the cellular level, probe SPN can achieve a good and specific response to endogenous and exogenous ONOO-. Also, the probe SPN can be used for imaging and detection of DILI in zebrafish level and small animal level, indicating that probe SPN can be used as a powerful tool for diagnosis of DILI and efficacy evaluation of therapeutic drugs.

Abstract:Hepatic lipid droplets (LDs) and peroxynitrite (ONOO-) levels are closely related to nonalcoholic fatty liver disease (NAFLD). Additionally, some drug-induced liver injury (DILI) is often associated with ONOO-. Here, we constructed and screened the quasi-LDs-targeted and ONOO--responsive fluorescent probe MBDP-Py+ to investigate the interaction of NAFLD with DILI. By monitoring the upregulation of the ONOO- levels and the accumulation of LDs, MBDP-Py+ was more sensitive and efficient than tissue staining and serum markers detection in evaluating the early toxicity of NAFLD and diagnosing the anticancer-DILI. More importantly, the sensitive enhancement of fluorescence signals demonstrated that in different stages of NAFLD, the dominant element of liver injury was different in the NAFLD combined with DILI mice models. As the degree of NAFLD deepens, the synergistic effect of the two will lead to more serious liver damage.

Abstract:Liver injury is usually caused by different sources (such as drugs, food, and alcohol), which is regarded as a serious threat to human health. Since the current indicators are not well defined, it is crucial to develop novel probes to characterize the kinetic process of liver injury. In this paper, a novel probe with fluorescence signals, Lyso-ONOO, was synthesized to detect the fluctuating peroxynitrite (ONOO−) level in various acute injury model mice. With the addition of ONOO−, the fluorescence response of Lyso-ONOO exhibited a large Stokes shift of 105 nm under 450 nm excitation. The reaction had the merits of high sensitivity, high selectivity, rapid reaction, and stability under various pH conditions. In addition, Lyso-ONOO had prominent lysosome-targeting ability. It achieved monitoring the exogenous and endogenous ONOO− in human hepatic stellate cells. Further, Lyso-ONOO could display endogenous ONOO− in models of 4T1-xenograft and inflammation in mice. More importantly, Lyso-ONOO could gauge the changing levels of ONOO− in three types (induced by CCl4, APAP, and alcohol) of acute liver injury models and treatments (silybin and bicyclol) of CCl4-induced liver injury mice. Both in vivo imaging and depth-of-penetration fluorescence imaging demonstrated that Lyso-ONOO could characterize the developmental and therapeutic status during the liver injury process by measuring the dynamic change of ONOO−. The monitoring of Lyso-ONOO appeared to be useful in inferring the order of severity in diverse acute liver injury processes and taking into account the efficacy of treatment regimens, which might provide important information for corresponding research.

Abstract:Drug-induced liver injury (DILI) is a serious public health issue related to oxidative stress. Peroxynitrite (ONOO−) is considered to be a direct biomarker of DILI. Recently, the Golgi apparatus has become a new choice for the research and treatment of DILI. A probe that can target Golgi to detect ONOO− is therefore of great value for the study of DILI. However, no such probe has been reported. Herein, we report a Golgi-targetable fluorescence probe MG-ONOO for ONOO−, which was prepared by linking a sulfanilamide moiety to a conjugated system of coumarin and hemi-cyanine. The probe shows low cytotoxicity, high sensitivity and selectivity, and rapid response for ONOO− with distinct ratiometric fluorescence changes. Unexpectedly, MG-ONOO shows unique targeting properties in living cells. The probe can target and label the cell membrane first (≤ 30 min incubation) and then the Golgi (≥ 60 min incubation), making it capable of detecting ONOO− not only in Golgi but also on cell membranes. Imaging experiments showed that MG-ONOO could be applied to monitor ONOO− under the oxidative stress of Golgi and in DILI (using mice models). Overall, this work provides a new tool for exploring Golgi oxidative stress and evaluating DILI.

Abstract:Peroxynitrite (ONOO-) has been proven to participate in various physiological and pathological processes, and may also be a contributing factor in many diseases. In view of this, there is a need to develop detection tools for unambiguously tracking a small amount of endogenous ONOO- to reveal its exact mechanisms. In this paper, a colorimetric and red-emitting fluorescent probe Red-PN, based on a rhodamine-type fluorophore and hydrazide reactive site is described. The probe Red-PN possesses the advantages of rapid response (within 5 s), visual color change (from colorless to pink), preeminent sensitivity (detection limit = 4.3 nM) and selectivity. Because of these outstanding performances, it was possible to accurately detect endogenous ONOO-. It was encouraging that the probe Red-PN could be used effectively for tracking the relatively low levels of endogenous and exogenous ONOO-in living cells and zebrafish. Thus, it is envisioned that the probe Red-PN would have promising prospects in applications for imaging ONOO- in a variety of biological settings.

Abstract:Drug-induced hepatotoxicity represents an important challenge for safety in drug development. The production of peroxynitrite (ONOO-) is proposed as an early sign in the progression of drug-induced hepatotoxicity. Currently, reported ONOO- probes mainly emit in the visible range or the first NIR window, which have limited in vivo biosensing application due to the autofluorescence and photon scattering. Herein, we developed a peroxynitrite activatable second near-infrared window (NIR-II) molecular probe for drug-induced hepatotoxicity monitoring, based on the fusion of an NIR-II fluorescence turn-on benzothiopyrylium cyanines skeleton and the phenyl borate. In the presence of ONOO-, the probe IRBTP-B can turn on its NIR-II fluorescence by yielding its fluorophore IRBTP-O and display good linear response to ONOO-. Tissue phantom study confirmed reliable activated signals could be acquired at a penetration depth up to 5 mm. Using this probe, we disclose the upregulation of ONOO- in a preclinical drug-induced liver injury model and the remediation with N-acetyl cysteine (NAC) in vivo. We expect that this strategy will serve as a general method for the development of an activatable NIR-II probe based on the hydroxyl functionalized reactive sites by analyte-specific triggering.

Abstract:Background: Drug-induced liver injury (DILI) is the most important standard for the entrance of clinical drugs into the pharmaceutical market. The elevation of superoxide anion (O2•-) during drug metabolism can mediate apoptosis of hepatocytes and further generation of liver damage. Therefore, developing an effective imaging method for evaluating O2•- levels during DILI is of great importance. However, current reported O2•- fluorescent probes either use short excitation wavelengths or a single intensity detection system, limiting the accurate quantification of O2•- in deep tissue in vivo. Results: We developed a NIR-excited ratiometric nanoprobe (CyD-UCNPs) by assembly of O2•--sensitive hemicyanine dyes (CyD) on the surface of Tm/Er-codoped upconversion nanoparticles (UCNPs) with the assistance of α-cyclodextrin, which exhibited a robust "turn-on" ratiometric sensing signal. In vitro experiments indicated that CyD-UCNPs respond well to O2•- with high selectivity. Furthermore, by taking advantage of the outstanding optical properties produced by the luminescent resonance energy transfer between the UCNPs and CyD upon the excitation of 980 nm, the ratiometric upconversion luminescence signal of CyD-UCNPs was successfully utilized to monitor the fluctuation of O2•- levels under phorbol-12-myristate-13-acetate (PMA)/cisplatin-induced oxidative stress in living cells, liver tissues, and zebrafish. More importantly, endogenous change in O2•- levels in the liver sites of mice during DILI and its prevention with L-carnitine was visualized using CyD-UCNPs. Significance: This study provides a ratiometric NIR-excited imaging strategy for investigating the correlation between O2•- levels and DILI and its prevention, which is significant for early diagnosis of DILI and preclinical screening of anti-hepatotoxic drugs in vivo.

Abstract:Hydrogen peroxide (H2O2), as one of important reactive oxygen species (ROS), can be endogenously produced in the liver, and used as a biomarker for predicting drug-induced liver injury (DILI). So, it is important to construct an efficient method for monitoring H2O2 in the liver to assess DILI. Herein, a hepatocyte-targeted fluorescent probe (GCP) for ratiometric detection of H2O2 is developed. GCP consists of two main parts: coumarin-pyran-based fluorophore and galactose-based hepatocyte-targetable group. The probe itself shows near-infrared (NIR) emission owing to its extended p-conjugation. Meanwhile, a large emission shift from 706 to 482 nm (224 nm) is observed after the reaction with H2O2. Taking advantage of the spectroscopic property, GCP exhibits high sensitivity toward H2O2 with 19-fold fluorescence ratio enhancement and is highly selective over other ROS. Moreover, the probe possesses a good hepatocyte-targeting ability and is applied for ratiometric monitoring endogenous H2O2 in HepG2 cells. Furthermore, the probe can trace H2O2 level during DILI in HepG2 cells. Most importantly, GCP can locate in the liver of zebrafish and detect H2O2 level in the liver during DILI, which will be a potentially useful tool to assess DILI.

Abstract:Herein, an activable fluorescent probe for peroxynitrite (ONOO-), named NOP, was constructed for the accurate imaging in the processes of formation and inhibition of drug-induced liver injury induced by Acetaminophen (APAP). During the in-solution tests on the general optical properties, the probe showed advantages including good stability, wide pH adaption, high specificity and sensitivity in the monitoring of ONOO-. Subsequently, the probe was further applied in the model mice which used APAP to induce the injury and used inhibiting agents (GSH, Glu, NAC) to treat the induced injury. The construction of the liver injury model was confirmed by the pathological staining and the serum indexes including ALT, AST, ALP, TBIL as well as LDH. During the formation of the drug-induced liver injury, the fluorescence in the red channel enhanced in both time-dependent and dose-dependent manners. In inhibition tests, the inhibition of the liver injury exhibited the reduction of the fluorescence intensity. Therefore, NOP could achieve the accurate imaging in the processes of formation and inhibition of drug-induced liver injury. The information here might be helpful for the early diagnosis and the screening of potent treating candidates in liver injury cases.

Due to the luminescence resonance energy transfer (LRET) between UCNPs and the cyanine dye (P-cy7), the irradiation changed from visible light at 660 nm to near infrared (NIR) light at 980 nm; therefore, the issues of poor photostability and severe photobleaching of cyanine dye can be effectively solved. After injecting via the tail vein, the nanoprobes are rapidly concentrated in the liver. Since the level of ONOO- is up-regulated during the drug-induced liver injury, the LRET between UCNPs and P-cy7 is disrupted to release the upconversion luminescence at 656 nm, while the upconversion luminescence at 800 nm remains constant, thus achieving the ratio-fluorescent imaging (RFLI) of ONOO- in the liver to calibrate the influence of the environment. In addition, the reduction in the absorption of nanoprobes in the presence of ONOO- allows for sensitive photoacoustic imaging (PAI). Based on the RFLI and PAI of the liver, the real-time monitoring and accurate diagnosis of different degrees of hepatotoxicity using the model of Acetaminophen (APAP) induction was achieved successfully, providing a new approach for the clinical evaluation of drug-induced hepatotoxicity.
Rational design of a ratiometric fluorescent probe for selective visualizing peroxynitrite in mitochondria and evaluation of organ injury

Abstract:Abnormal peroxynitrite (ONOO−) expression is closely associated with the development of various pathological processes. In this study, we devised a series of fluorescent probes, namely DHU-DCM-1, DHU-DCM-2, and DHU-DCM-3, tailored specifically for monitoring ONOO− through rationally molecular design. By modulating the electron-donating site within the fluorophore structure, the optimal probe DHU-DCM-3 exhibited excellent ratiometric detecting performance. DHU-DCM-3 displayed high selectivity and sensitivity towards ONOO−, effectively distinguishing it from other reactive oxygen species. Notably, DHU-DCM-3 was capable of labeling mitochondrial ONOO−, producing an enhanced fluorescence ratio (F656/F550), thereby allowing for the visualization of ONOO− fluctuations in both exogenous and endogenous contexts within living cells. The utility of DHU-DCM-3 was further demonstrated in various applications. For instance, through detecting elevated levels of ONOO−, senile C. elegans was successfully identified by utilizing DHU-DCM-3. Moreover, relying on its ratiometric fluorescence, DHU-DCM-3 was proved to be valuable in accurately diagnosing liver injury in mice with varying degrees of severity. Overall, we have constructed a ratiometric fluorescent probe for detecting and imaging ONOO− in both subcellular organelle and in vivo, highlighting its potential significance in various biomedical applications.
