Abstract:Skin-interfaced electrochemical sensing devices are widely developed for biochemical sensing at molecular levels. However, the sensing electrodes fabricated with photolithography or printing technique are hard to achieve high stretchability without special designs like serpentine shape. Also, most of them require wired connections with external electrochemical workstations for data acquisition or on-board batteries to power the circuits, which largely restrain the flexibility, simplification, and miniaturization of the devices. Here, a battery-free, wireless, and epidermal electrochemical system is presented for in situ biochemical sensing. The sensing component of the system is a stretchable electrode array all-printed with soft polymer and conductive silver nanowires. The electrodes show stable electrical properties within strain range of 0-50%, without serpentine designs. The electronic component of the system is a fully integrated flexible circuit board with near field communication module, which enables wireless energy harvesting and data transmission with smartphones. The system demonstrates good performance in real-time on-body sweat analysis for simultaneous quantitative detections of glucose, hydrogen, sodium, and potassium. This battery-free and wireless epidermal electrochemical system provides a simplified, miniaturized, and flexible solution for a wide range of biochemical platforms, including wearable and implanted bioelectronics.

Abstract:Monitoring blood glucose concentration is crucial in diagnosing and predicting clinical diabetes progression. However, the invasion of blood sampled by venipuncture and the informative delay of alternative biofluids inevitably affect dynamic and accurate monitoring. Here, we proposed a skin-patchable, noninvasive, and wireless blood glucose biosensing system composed of a flexible electromagnetic resonant circuit and a wireless data acquisition unit, correlating the S parameter shifts and glucose level-inducing blood dielectric property alteration. Ex vivo and volunteer hypo-to hyperglycemic monitoring validations achieved supered fidelity and correlation with high sensitivity > 0.0626°/(mg/dl). The correlation coefficient values were 0.996 and 0.945 for ex vivo mammalian skin and healthy volunteers’ oral glucose tolerance tests, comparable to commercial blood glucose meters. Emerging noninvasive and wireless modalities would strive to bridge gaps between minimal discomfort and reliable measurements, spurring the widespread adoption of this portable blood glucose monitoring system.
Smartphone-Based Electrochemical Systems for Glucose Monitoring in Biofluids: A Review

Abstract:As a vital biomarker, glucose plays an important role in multiple physiological and pathological processes. Thus, glucose detection has become an important direction in the electrochemical analysis field. In order to realize more convenient, real-time, comfortable and accurate monitoring, smartphone-based portable, wearable and implantable electrochemical glucose monitoring is progressing rapidly. In this review, we firstly introduce technologies integrated in smartphones and the advantages of these technologies in electrochemical glucose detection. Subsequently, this overview illustrates the advances of smartphone-based portable, wearable and implantable electrochemical glucose monitoring systems in diverse biofluids over the last ten years (2012-2022). Specifically, some interesting and innovative technologies are highlighted. In the last section, after discussing the challenges in this field, we offer some future directions, such as application of advanced nanomaterials, novel power sources, simultaneous detection of multiple markers and a closed-loop system.
A Touch-Actuated Glucose Sensor Fully Integrated with Microneedle Array and Reverse Iontophoresis for Diabetes Monitoring.

Abstract:The development of non-invasive biosensor for monitoring glucose in interstitial fluid (ISF) is still challenging, because ISF extraction through classical reverse iontophoresis (RI) is limited by low extraction flux and consistency. Here, we developed a touch-actuated biosensor for monitoring glucose in ISF. The biosensor is composed of three main components: 1) the solid microneedle array (MA) for painless skin penetration; 2) the RI unit for ISF extraction through the MA-created microchannels; and 3) the sensing unit for glucose monitoring. The sensing strategy of this biosensor is "skin penetration-RI extraction-electrochemical detection". Compared with RI extraction only, the reported skin penetration-RI extraction sampling strategy obviously increased the glucose extraction flux by ∼1.6 times not only in vitro but also in vivo. Moreover, we developed a wearable glucose monitoring system by incorporating this touch-actuated biosensor, a wireless electrochemical detector, and a smartphone application. In vivo experiments using healthy and diabetic rats revealed a high correlation between the results measured by the reported wearable system and commercially blood glucometer. This sampling strategy which combined skin penetration and RI extraction paves the way to develop wearable platforms for not only glucose monitoring but also various ISF biomarkers without the need of painful finger-stick blood sampling.

Abstract:We report on a nonenzymatic electrochemical sensor for wearable glucose monitoring in interstitial fluid. The sensor exhibited acceptable selectivity and reliability for continuous glucose detection for up to 30 days. The sensor tip is coated with polyurethane, and the biocompatibility of the tip is investigated by tissue staining. A fully integrated wearable glucose monitoring system is developed with a wireless connection with a smartphone. The test results are in agreement with reference methods. So, we believe the sensor is promising for the development of a continuous glucose monitoring system and diabetes management.

Abstract:To enhance personalized diabetes management, there is a critical need for non-invasive wearable electrochemical sensors made from flexible materials to enable continuous monitoring of sweat glucose levels. The main challenge lies in developing glucose sensors with superior electrochemical characteristics and high adaptability. Herein, we present a wearable sensor for non-enzymatic electrochemical glucose analysis. The sensor was synthesized using hydrothermal and one-pot preparation methods, incorporating gold nanoparticles (AuNPs) functionalized onto aminated multi-walled carbon nanotubes (AMWCNTs) as an efficient catalyst, and crosslinked with carboxylated styrene butadiene rubber (XSBR) and PEDOT:PSS. The sensors were then integrated onto screen-printed electrodes (SPEs) to create flexible glucose sensors (XSBR-PEDOT:PSS-AMWCNTs/AuNPs/SPE). Operating under neutral conditions, the sensor exhibits a linear range of 50 μmol/L to 600 μmol/L, with a limit of detection limit of 3.2 μmol/L (S/N = 3), enabling the detection of minute glucose concentrations. The flexible glucose sensor maintains functionality after 500 repetitions of bending at a 180° angle, without significant degradation in performance. Furthermore, the sensor exhibits exceptional stability, repeatability, and resistance to interference. Importantly, we successfully monitored changes in sweat glucose levels by applying screen-printed electrodes to human skin, with results consistent with normal physiological blood glucose fluctuations. This study details the fabrication of a wearable sensor characterized by ease of manufacture, remarkable flexibility, high sensitivity, and adaptability for non-invasive blood glucose monitoring through non-enzymatic electrochemical analysis. Thus, this streamlined fabrication process presents a novel approach for non-invasive, real-time blood glucose level monitoring.

Abstract:Glucose is one of the most significant biomarkers due to its association with numerous diseases related to blood glucose concentration. In this study, a novel electrochemical microneedle sensor (EMS) for highly sensitive, high spatiotemporal resolution, real time glucose monitoring is developed. Initially, the sensor is modified by electrochemically depositing gold nanoparticles to enhance its conductivity. Subsequently, a platinum sensing layer is deposited onto the sensor to realize the detection of hydrogen peroxide. Glucose oxidase (GOx) is then immobilized onto the sensor by immersing it in an O-phenylenediamine solution and electro-polymerizing O-phenylenediamine. A polyurethane-based biological protective film is applied to protect the electrode from interference by large proteins and cell deposition in body fluid. The prepared EMS demonstrates excellent selectivity for glucose detection (0.818 μA/mM) over a broad range (0 to 20 mM). Furthermore, clinical serum glucose testing experiments verified the sensor's performance, showcasing its potential for biomedical research and clinical practice. This EMS offers a less invasive and more convenient method for highly sensitive, high spatiotemporal resolution, real time glucose monitoring. Moreover, this electrochemical implantable sensor provides a minimally invasive and convenient solution for continuous monitoring of other biomarkers.

Abstract:We demonstrate a novel minimally invasive flexible sensor for continuous in vivo glucose monitoring. This sensor is based on a gold-coated polyimide microneedle array (MNA) electrode, which is modified with glucose oxidase. Owing to the spiky micro-needles, flexible substrate, and biocompatible materials, the enzymatic surface is able to penetrate the outer skin, and directly detect the subcutaneous glucose with high biosafety. The current results show that our device performs good response, linearity, and stability to the change of glucose concentration.

Abstract:Wearable non-invasive glucose sensors that can provide human a painless and portable means to monitor their blood glucose and manage their health condition draw great attentions, recently. Non-invasive human glucose sensors by detecting glucose in interstitial fluid (ISF) extracted through a reverse iontophoresis (RI) approach have been widely investigated, but the current challenges are their complex structure and instability for continuous monitor. Herein, we demonstrate a simple two-electrode non-invasive blood glucose sensor, which is fabricated by using graphene/carbon nanotubes/glucose oxidase composite textile and graphene/carbon nanotube/silver/silver chloride composite textile as the working electrode and counter electrode, respectively. By using one single device, extraction of ISF through RI process is firstly conducted by loading a certain electric current between two electrodes, then the glucose concentration in the ISF is detected through an amperometric approach by using the same two electrodes. The feasibility of these non-invasive glucose sensors is validated on porcine skin, nude mice and human. The blood glucose concentration calculated according to the response currents of the two-electrode sensors is highly consistent with that measured by commercial glucose meter. Furthermore, the used textile-like electrodes provide the non-invasive blood glucose sensors with excellent flexible and wearable properties, which make them promising to be integrated with other electronic units for monitor and management of human health.

Abstract:Current advances in non-invasive fluid diagnostics highlight unique benefits for monitoring metabolic diseases. However, the low concentrations and complex compositions of biomarkers in fluids such as sweat, urine, and saliva impose stringent demands on the sensitivity and stability of detection technologies. Here, we developed a high-sensitivity, low-cost instantaneous electrochemical sensor based on the superadditive effect mechanism of Cu-TCPP(Fe)/Mxene (MMs Paper-ECL Sensor), which has been successfully applied for the simultaneous real-time detection of glucose and uric acid. Strong interfacial interactions between Mxene and Cu-TCPP(Fe) were revealed through precise simulation calculations and multi-dimensional characterization analysis, significantly enhancing the sensor's electrocatalytic performance and reaction kinetics. Experimentally, this exceptional electrocatalytic activity was demonstrated in its unprecedented high sensitivity and wide linear detection range for glucose and uric acid, with a non-invasive linear range from 0.001 nM to 5 mM, 0.025 nM-5 mM, detection limits as low as 1.88 aM and 5.80 pM, and stability extending up to 100 days. This represents not only a breakthrough in sensitivity and stability but also provides an effective, low-cost solution that overcomes the limitations of existing electronic devices, enabling multi-channel simultaneous detection. The universality of this sensor holds vast potential for application in the field of non-invasive fluid diagnostics.

Abstract:Currently, there is no commercially-available noninvasive method for glucose monitoring in the interstitial fluid (ISF) mainly due to the measurement accuracy, sensitivity and/or the weak blood correlation. Here, we present an electrochemical biosensor based on PEDOT:PSS conductive hydrogel incorporated with Prussian blue nanoparticles (PBNPs) for the noninvasive and continuous monitoring of glucose on body. The hydrogel was prepared with the addition of dimethyl sulfoxide (DMSO) and Zonyl FS-300, which helped to improve the expanding of the polymer chains for the formation of better conductive and nanoporous networks. The hydrogel-based biosensor showed a low detection limit of 0.85 μM for glucose detection and high sensitivity up to 340.1 μA·mM−1·cm−2, which was about 10-fold higher than the counterparts based on agarose and hydroxyethyl methacrylate (HEMA) hydrogels. Furthermore, it provided good accuracy with commercial glucometer for the glucose detection in serum. More importantly, incorporated with the reverse iontophoresis (RI), the hydrogel was applied as a skin patch on human subject for the in vivo noninvasive and continuous monitoring of ISF-borne glucose. The results showed good correlation with that measured by blood glucometer. We believe such glucose biosensor paves the way to clinically wearable noninvasive glucose monitoring in diabetics.

Abstract:Diabetes remains a great threat to human beings' health and its world prevalence is projected to reach 9.9% by 2045. At present, the detection methods used are often invasive, cumbersome and time-consuming, thus increasing the burden on patients. In this paper, we propose a novel noninvasive and low-cost biosensor capable of detecting glucose in human sweat using enzyme-based electrodes for point-of-care uses. Specifically, an electrochemical method is applied for detection and the electrodes are covered with multilayered films including ferrocene-polyaniline (F-P), multi-walled carbon nanotubes (MWCNTs) and glucose oxidase (GOx) on Cu substrates (GOx/MWCNTs/F-P/Cu). The coated layers enhance the immobilization of GOx, increase the conductivity of the anode and improve the electrochemical properties of the electrode. Compared with the Cu electrode and the F-P/Cu electrode, a maximum peak current is obtained when the MWCNTs/F-P/Cu electrode is applied. We also study its current response by cyclic voltammetry (CV) at different concentrations (0–2.0 mM) of glucose solution. The best current response is obtained at 0.25 V using chronoamperometry. The effective working lifetime of an electrode is up to 8 days. Finally, to demonstrate the capability of the electrode, a portable, miniaturized and integrated detection device based on the GOx/MWCNTs/F-P/Cu electrode is developed. The results exhibit a short response time of 5 s and a correlation coefficient R2 of 0.9847 between the response current of sweat with blood glucose concentration. The LOD is of 0.081 mM and the reproducibility achieved in terms of RSD is 3.55%. The sweat glucose sensor is noninvasive and point-of-care, which shows great development potential in the health examination and monitoring field.

Abstract:Abstract Integrated electrochemical sensing platforms in wearable devices have great prospects in biomedical applications. However, traditional electrochemical platforms are generally fabricated on airtight printed circuit boards, which lack sufficient flexibility, air permeability, and conformability. Liquid metals at room temperature with excellent mobility and electrical conductivity show high promise in flexible electronics. This paper presents a miniaturized liquid metal-based flexible electrochemical detection system on fabric, which is intrinsically flexible, air-permeable, and conformable to the body. Taking advantage of the excellent fluidity and electrical connectivity of liquid metal, a double-layer circuit is fabricated that significantly miniaturizes the size of the whole system. The linear response, time stability, and repeatability of this system are verified by resistance, stability, image characterization, and potassium ferricyanide tests. Finally, glucose in sweat can be detected at the millimolar level using this sensing system, which demonstrates its great potential for wearable and portable detection in biomedical fields, such as health monitoring and point-of-care testing.

Abstract:In this study, we present the development of an innovative electrochemical biosensor integrated into a microneedle-based system for non-invasive and sensitive quantification of cholesterol levels in interstitial fluid (ISF). The biosensor employs a graphene-based electrode with a polyelectrolyte interlayer to immobilize cholesterol oxidase (ChOx), enabling selective cholesterol detection. Graphene oxide is electrochemically reduced to form a conductive layer, and PANI is chosen as the optimal polyelectrolyte for ChOx immobilization. The biosensor's performance is thoroughly evaluated, demonstrating excellent sensitivity, stability, and selectivity. Furthermore, the biosensor is successfully applied to skin-mimicking agarose gel and porcine skin, showcasing its potential for real-world interstitial fluid extraction and cholesterol monitoring. The integrated microneedle-based system offers a promising approach for non-invasive monitoring of cholesterol levels, with implications for personalized healthcare diagnostics.

Abstract:Based on reverse iontophoresis principle, a non-invasive blood glucose detection system was built up. Flexible screen printed solid electrodes, using Prussian blue as the medium, combined with glucose oxidase,formed the flexible glucose biosensor.The performance of the biosensor was evaluated by chronoamperometry.The glucose in vitro quantitative detection system was established by optimizing each of modified conditions.Within the scope of low concentration,the linear range of glucose concentration was 05-65 mmol/L,the correlation coefficient ( R) was 09933 and detection limit was 01 mmol/L,and within the scope of relatively high concentration, the linear range was 65-190 mmol/L and the correlation coefficient was 09923. A 3D printed device was designed to simulate reverse iontophoresis on pig skin,thus the quantitative detection system of subcutaneous glucose was established. The response of the sensor had a good correlation with the subcutaneous glucose concentration in the range of 20-140 mmol/L (R=09929) of daily blood glucose testing.The results found the basis for the development of a practical non-invasive blood glucose meter.

Abstract:Considering the demand of long-term blood glucose monitoring,a novel MEMS sensor system was developed,which extracts tissue fluid in vivo by ultrafiltration and monitors glucose in vitro.The construction and properties of this sensor system for tissue fluid ultrafiltration sampling and on-line glucose monitoring were presented.It comprised a glucose sensor chip integrated on the microfluidics base.The microfluidics base consisted of PDMS microchannel and SU-8 check valve fabricated by MEMS technology.This sensor system could harvest tissue fluid under negative pressure for glucose monitoring and discharge the used tissue fluid after each measurement.The 3-electrode glucose sensor chip based on electrochemical mechanism was fabricated by silicon processing method,and it had a silicon etched cavity on the top side.Pores were etched into the membrane at the cavity ground,so the sensor chip was provided with diffusion control of the analyte through the membrane into cavity containing agarose immobilised enzyme.Experimental results show that this system can realize the flexible tissue fluid sampling,and its signal response time at increasing glucose concentrations is 5 s;under the voltage of 0.4 V the linear range is large enough to cover the required broad area of blood glucose(0.2—20 mmol / L),the sensitivity is 9.76 nA/(mmol·L-1),and the correlation coefficient is 0.995 4;the coefficient of variation for measuring 5 mmol/L glucose solution is 3.48%.It can be seen that the glucose sensor chip has good reliability and stability,and it can be easily integrated into the sensor system for tissue fluid sampling and subsequent glucose monitoring because of its small dimension.

Abstract:A microneedle-based skin patch system allows the minimally invasive extraction of skin interstitial fluid, which offers hope for the realization of quantitative and non-invasive diagnosis/monitoring of biological/physiological signals (biomarkers) in the human body. This work describes a nanometallic conductive composite-hydrogel core-shell microneedle skin patch that can realize minimally invasive real-time monitoring of physiological signals. The microneedle sensing system contains an inner conductive silver paste core and an outer bioactive hydrogel layer. The inner core is coated with biomarker-specific enzymes while the outer hydrogel layer extracts the biomarkers from the skin interstitial fluid. This patch can be integrated with the commercial signal processing and transmission modules and enable real-time monitoring. Taking glucose as a model biomarker, we confirm the function and potential application of this core-shell microneedle patch for transdermal diagnosis. It is proven that the core-shell microneedle patch can quickly extract skin interstitial fluid within 30 seconds and has a fast and linear response to glucose concentrations from 0 to 21 mM with a correlation coefficient of 0.9970, which may pave the way for the future development of smart wearable devices with minimally invasive transdermal biosensors.

Abstract:Wearable sensors, specifically microneedle sensors based on electrochemical methods, have expanded extensively with recent technological advances. Today's wearable electrochemical sensors present specific challenges: they show significant modulus disparities with skin tissue, implying possible discomfort in vivo, especially over extended wear periods or on sensitive skin areas. The sensors, primarily based on polyethylene terephthalate (PET) or polyimide (PI) substrates, might also cause pressure or unease during insertion due to the skin's irregular deformation. To address these constraints, we developed an innovative, wearable, all-fiber-structured electrochemical sensor. Our composite sensor incorporates polyurethane (PU) fibers prepared via electrospinning as electrode substrates to achieve excellent adaptability. Electrospun PU nanofiber films with gold layers shaped via thermal evaporation are used as base electrodes with exemplary conductivity and electrochemical catalytic attributes. To achieve glucose monitoring, gold nanofibers functionalized by gold nanoflakes (AuNFs) and glucose oxidase (GOx) serve as the working electrode, while Pt nanofibers and Ag/AgCl nanofibers serve as the counter and reference electrode. The acrylamide-sodium alginate double-network hydrogel synthesized on electrospun PU fibers serves as the adhesive and substance-transferring layer between the electrodes. The all-fiber electrochemical sensor is assembled layer-by-layer to form a robust structure. Given the stretchability of PU nanofibers coupled with a high specific surface area, the manufactured porous microneedle glucose sensor exhibits enhanced stretchability, superior sensitivity at 31.94 μA (lg(mM))-1 cm-2, a broad detection range (1-30 mM), and a significantly low detection limit (1 mM, S/N = 3), as well as satisfactory biocompatibility. Therefore, the novel electrochemical microneedle design is well-suited for wearable or even implantable continuous monitoring applications, thereby showing promising significant potential within the global arena of wearable medical technology.
