Abstract:This study investigates the neural representation of speech in complex listening environments. Subjects listened to a narrated story, masked by either another speech stream or by stationary noise. Neural recordings were made using magnetoencephalography (MEG), which can measure cortical activity synchronized to the temporal envelope of speech. When two speech streams are presented simultaneously, cortical activity is predominantly synchronized to the speech stream the listener attends to, even if the unattended, competing-speech stream is more intense (up to 8 dB). When speech is presented together with spectrally matched stationary noise, cortical activity remains precisely synchronized to the temporal envelope of speech until the noise is 9 dB more intense. Critically, the precision of the neural synchronization to speech predicts subjectively rated speech intelligibility in noise. Further analysis reveals that it is longer-latency (similar to 100 ms) neural responses, but not shorter-latency (similar to 50 ms) neural responses, that show selectivity to the attended speech and invariance to background noise. This indicates a processing transition, from encoding the acoustic scene to encoding the behaviorally important auditory object, in auditory cortex. In sum, it is demonstrated that neural synchronization to the speech envelope is robust to acoustic interference, whether speech or noise, and therefore provides a strong candidate for the neural basis of acoustic-background invariant speech recognition.
Temporal Modulations Reveal Distinct Rhythmic Properties of Speech and Music

Abstract:Speech and music have structured rhythms, but these rhythms are rarely compared empirically. This study, based on large corpora, quantitatively characterizes and compares a major acoustic correlate of spoken and musical rhythms, the slow (0.25-32 Hz) temporal modulations in sound intensity. We show that the speech modulation spectrum is highly consistent cross 9 languages (including languages with typologically different rhythmic characteristics, such as English, French, and Mandarin Chinese). A different, but similarly consistent modulation spectrum is observed for Western classical music played by 6 different instruments. Western music, including classical music played by single instruments, symphonic, jazz, and rock music, contains more energy than speech in the low modulation frequency range below 4 Hz. The temporal modulations of speech and music show broad but well-separated peaks around 5 and 2 Hz, respectively. These differences in temporal modulations alone, without any spectral details, can discriminate speech and music with high accuracy. Speech and music therefore show distinct and reliable statistical regularities in their temporal modulations that likely facilitate their perceptual analysis and its neural foundations.

Abstract:Speech and music have structured rhythms. Here we discuss a major acoustic correlate of spoken and musical rhythms, the slow (0.25-32 Hz) temporal modulations in sound intensity and compare the modulation properties of speech and music. We analyze these modulations using over 25 h of speech and over 39 h of recordings of Western music. We show that the speech modulation spectrum is highly consistent across 9 languages (including languages with typologically different rhythmic characteristics). A different, but similarly consistent modulation spectrum is observed for music, including classical music played by single instruments of different types, symphonic, jazz, and rock. The temporal modulations of speech and music show broad but well-separated peaks around 5 and 2 Hz, respectively. These acoustically dominant time scales may be intrinsic features of speech and music, a possibility which should be investigated using more culturally diverse samples in each domain. Sensitivity to Temporal Modulation Rate and Spectral Bandwidth in the Human Auditory System: Fmri Evidence

Abstract:Hierarchical models of auditory processing often posit that optimal stimuli, i.e., those eliciting a maximal neural response, will increase in bandwidth and decrease in modulation rate as one ascends the auditory neuraxis. Here, we tested how bandwidth and modulation rate interact at several loci along the human central auditory pathway using functional MRI in a cardiac-gated, sparse acquisition design. Participants listened passively to both narrowband (NB) and broadband (BB) carriers (1/4- or 4-octave pink noise), which were jittered about a mean sinusoidal amplitude modulation rate of 0, 3, 29, or 57 Hz. The jittering was introduced to minimize stimulus-specific adaptation. The results revealed a clear difference between spectral bandwidth and temporal modulation rate: sensitivity to bandwidth (BB > NB) decreased from subcortical structures to nonprimary auditory cortex, whereas sensitivity to slow modulation rates was largest in nonprimary auditory cortex and largely absent in subcortical structures. Furthermore, there was no parametric interaction between bandwidth and modulation rate. These results challenge simple hierarchical models, in that BB stimuli evoked stronger responses in primary auditory cortex (and subcortical structures) rather than nonprimary cortex. Furthermore, the strong preference for slow modulation rates in nonprimary cortex demonstrates the compelling global sensitivity of auditory cortex to modulation rates that are dominant in the principal signals that we process, e.g., speech.
Cortical encoding of acoustic and linguistic rhythms in spoken narratives

Abstract:Speech contains rich acoustic and linguistic information. Using highly controlled speech materials, previous studies have demonstrated that cortical activity is synchronous to the rhythms of perceived linguistic units, for example, words and phrases, on top of basic acoustic features, for example, the speech envelope. When listening to natural speech, it remains unclear, however, how cortical activity jointly encodes acoustic and linguistic information. Here we investigate the neural encoding of words using electroencephalography and observe neural activity synchronous to multi-syllabic words when participants naturally listen to narratives. An amplitude modulation (AM) cue for word rhythm enhances the word-level response, but the effect is only observed during passive listening. Furthermore, words and the AM cue are encoded by spatially separable neural responses that are differentially modulated by attention. These results suggest that bottom-up acoustic cues and top-down linguistic knowledge separately contribute to cortical encoding of linguistic units in spoken narratives.
Robust Cortical Entrainment to the Speech Envelope Relies on the Spectro-Temporal Fine Structure

Abstract:Speech recognition is robust to background noise. One underlying neural mechanism is that the auditory system segregates speech from the listening background and encodes it reliably. Such robust internal representation has been demonstrated in auditory cortex by neural activity entrained to the temporal envelope of speech. A paradox, however, then arises, as the spectro-temporal fine structure rather than the temporal envelope is known to be the major cue to segregate target speech from background noise. Does the reliable cortical entrainment in fact reflect a robust internal "synthesis" of the attended speech stream rather than direct tracking of the acoustic envelope? Here, we test this hypothesis by degrading the spectro-temporal fine structure while preserving the temporal envelope using vocoders. Magnetoencephalography (MEG) recordings reveal that cortical entrainment to vocoded speech is severely degraded by background noise, in contrast to the robust entrainment to natural speech. Furthermore, cortical entrainment in the delta-band (1-4Hz) predicts the speech recognition score at the level of individual listeners. These results demonstrate that reliable cortical entrainment to speech relies on the spectro-temporal fine structure, and suggest that cortical entrainment to the speech envelope is not merely a representation of the speech envelope but a coherent representation of multiscale spectro-temporal features that are synchronized to the syllabic and phrasal rhythms of speech.

Abstract:Auditory cortical activity is entrained to the temporal envelope of speech, which corresponds to the syllabic rhythm of speech. Such entrained cortical activity can be measured from subjects naturally listening to sentences or spoken passages, providing a reliable neural marker of online speech processing. A central question still remains to be answered about whether cortical entrained activity is more closely related to speech perception or non-speech-specific auditory encoding. Here, we review a few hypotheses about the functional roles of cortical entrainment to speech, e.g., encoding acoustic features, parsing syllabic boundaries, and selecting sensory information in complex listening environments. It is likely that speech entrainment is not a homogeneous response and these hypotheses apply separately for speech entrainment generated from different neural sources. The relationship between entrained activity and speech intelligibility is also discussed. A tentative conclusion is that theta-band entrainment (4-8 Hz) encodes speech features critical for intelligibility while delta-band entrainment (1-4 Hz) is related to the perceived, non-speech-specific acoustic rhythm. To further understand the functional properties of speech entrainment, a splitter's approach will be needed to investigate (1) not just the temporal envelope but what specific acoustic features are encoded and (2) not just speech intelligibility but what specific psycholinguistic processes are encoded by entrained cortical activity. Similarly, the anatomical and spectro-temporal details of entrained activity need to be taken into account when investigating its functional properties.

Abstract:Complex sounds, especially natural sounds, can be parametrically characterized by many acoustic and perceptual features, one among which is temporal modulation. Temporal modulations describe changes of a sound in amplitude (amplitude modulation, AM) or in frequency (frequency modulation, FM). AM and FM are fundamental components of communication sounds, such as human speech and species-specific vocalizations, as well as music. Temporal modulations are encoded in at least two ways, temporal coding and rate coding. Magnetoencephalography (MEG), with its high temporal resolution and simultaneous access to multiple auditory cortical areas, is a non-invasive tool that can measure and describe the temporal coding of auditory modulations. We refer to the neural temporal encoding of temporal acoustic modulations as "modulation encoding". For simple, individually presented, acoustic modulations, modulation encoding is well described by a simple modulation transfer function (MTF). Even in this simple case, however, the MTF may depend strongly on the type of modulation being encoded (e.g. AM vs. FM, narrowband vs. broadband) or the context in which the modulation is heard (e.g. attended vs. unattended). Here we present a range of different types of modulation encoding employed by human auditory cortex. The simplest examples are for sinusoidally amplitude modulated carriers of a range of bandwidths (with special emphasis on those modulation rates relevant to speech and other natural sounds: below a few tens of Hz). We provide evidence that the modulation transfer functions are lowpass in shape and relatively independent of bandwidth. When several modulations are applied concurrently however, the modulation encoding typically, but not always, becomes non-linear: the auditory modulations are at the rates of the acoustic modulations but also at the rates of cross-modulation frequencies. The physiological occurrence, or not, of these cross terms seem be in accord with the psychophysical concept of modulation filterbanks.
Auditory-motor synchronization and perception suggest partially distinct time scales in speech and music

Abstract:Speech and music might involve specific cognitive rhythmic timing mechanisms related to differences in the dominant rhythmic structure. We investigate the influence of different motor effectors on rate-specific processing in both domains. A perception and a synchronization task involving syllable and piano tone sequences and motor effectors typically associated with speech (whispering) and music (finger-tapping) were tested at slow (~2 Hz) and fast rates (~4.5 Hz). Although synchronization performance was generally better at slow rates, the motor effectors exhibited specific rate preferences. Finger-tapping was advantaged compared to whispering at slow but not at faster rates, with synchronization being effector-dependent at slow, but highly correlated at faster rates. Perception of speech and music was better at different rates and predicted by a fast general and a slow finger-tapping synchronization component. Our data suggests partially independent rhythmic timing mechanisms for speech and music, possibly related to a differential recruitment of cortical motor circuitry.

Abstract:Synchronizing motoric responses to metrical sensory rhythms is key to social activities, e.g., group singing and dancing. It remains elusive, however, whether there is a common neural network for motoric synchronization to metrical rhythms from different sensory modalities. Here, we separate sensorimotor responses from basic sensory responses by combining a metrical sensorimotor synchronization task with frequency-domain magnetoencephalography (MEG) analysis. A common frontal-temporal network, not including visual cortex, is observed during both visual- and auditory-motor synchronization, and the network remains in congenitally deaf participants during visual-motor synchronization, suggesting the network is formed by intrinsic cortical connections instead of auditory experience. Furthermore, activation of the left and right frontal-temporal areas, as well as the ipsilateral white matter connection, separately predict the precision of auditory and visual synchronization. These results reveal a common but lateralized frontal-temporal network for visual- and auditory-motor synchronization, which is generated based on intrinsic cortical connections.

Abstract:We investigated the role of the cerebellum in differential aspects of temporal control of rhythmic auditory motor synchronization using positron emission tomography (PET). Subjects tapped with their right index finger to metronome tones at a mean frequency of .8 Hz during 5 conditions: (1) an isochronous rhythm condition, (2) random changes in interval durations, and while the duration of rhythmic intervals was continuously time-modulated following a cosine-wave function at (3) 3%, (4) 7%, and (5) 20% of base interval. Anterior lobe cerebellar neuronal populations showed similar motor-associated activity across all conditions regardless of rhythmic time structure in vermal and hemispheric parts ipsilateral to the movements. Neuronal populations in bilateral anterior posterior lobe, especially in the simple lobule, increased their activity stepwise with each increase in tempo modulation from a steady beat. Neuronal populations in other parts of the posterior lobe showed an increase of activity only during the 20% condition, which involved conscious monitoring of rhythmic pattern synchronization, especially on the left side contralateral to the movements. Differential cerebellar activation patterns correspond to those in contralateral primary (primary sensorimotor), ipsilateral secondary (inferior parietal close to the intraparietal sulcus) and bilateral tertiary (dorsolateral prefrontal cortex) sensorimotor areas of the cerebral cortex, suggesting that distinct functional cortico-cerebellar circuits subserve differential aspects of rhythmic synchronization in regard to rhythmic motor control, conscious and subconscious response to temporal structure, and conscious monitoring of rhythmic pattern tracking.

Abstract:Cortical oscillations have been proposed to play a functional role in speech and music perception, attentional selection, and working memory, via the mechanism of neural entrainment. One of the properties of neural entrainment that is often taken for granted is that its modulatory effect on ongoing oscillations outlasts rhythmic stimulation. We tested the existence of this phenomenon by studying cortical neural oscillations during and after presentation of melodic stimuli in a passive perception paradigm. Melodies were composed of ~60 and ~80 Hz tones embedded in a 2.5 Hz stream. Using intracranial and surface recordings in male and female humans, we reveal persistent oscillatory activity in the high- band in response to the tones throughout the cortex, well beyond auditory regions. By contrast, in response to the 2.5 Hz stream, no persistent activity in any frequency band was observed. We further show that our data are well captured by a model of damped harmonic oscillator and can be classified into three classes of neural dynamics, with distinct damping properties and eigenfrequencies. This model provides a mechanistic and quantitative explanation of the frequency selectivity of auditory neural entrainment in the human cortex. SIGNIFICANCE STATEMENT It has been proposed that the functional role of cortical oscillations is subtended by a mechanism of entrainment, the synchronization in phase or amplitude of neural oscillations to a periodic stimulation. One of the properties of neural entrainment that is often taken for granted is that its modulatory effect on ongoing oscillations outlasts rhythmic stimulation. Using intracranial and surface recordings of humans passively listening to rhythmic auditory stimuli, we reveal consistent oscillatory responses throughout the cortex, with persistent activity of high- oscillations. On the contrary, neural oscillations do not outlast low-frequency acoustic dynamics. We interpret our results as reflecting harmonic oscillator properties, a model ubiquitous in physics but rarely used in neuroscience.

Abstract:Natural sounds, including vocal communication sounds, contain critical information at multiple time scales. Two essential temporal modulation rates in speech have been argued to be in the low gamma band (similar to 20-80 ms duration information) and the theta band (similar to 150-300 ms), corresponding to segmental and diphonic versus syllabic modulation rates, respectively. It has been hypothesized that auditory cortex implements temporal integration using time constants closely related to these values. The neural correlates of a proposed dual temporal window mechanism in human auditory cortex remain poorly understood. We recorded MEG responses from participants listening to non-speech auditory stimuli with different temporal structures, created by concatenating frequency-modulated segments of varied segment durations. We show that such non-speech stimuli with temporal structure matching speech-relevant scales (similar to 25 and similar to 200 ms) elicit reliable phase tracking in the corresponding associated oscillatory frequencies (low gamma and theta bands). In contrast, stimuli with non-matching temporal structure do not. Furthermore, the topography of theta band phase tracking shows rightward lateralization while gamma band phase tracking occurs bilaterally. The results support the hypothesis that there exists multi-time resolution processing in cortex on discontinuous scales and provide evidence for an asymmetric organization of temporal analysis (asymmetrical sampling in time, AST). The data argue for a mesoscopic-level neural mechanism underlying multi-time resolution processing: the sliding and resetting of intrinsic temporal windows on privileged time scales.
Prominence of delta oscillatory rhythms in the motor cortex and their relevance for auditory and speech perception

Abstract:In the motor cortex, beta oscillations (∼12-30 Hz) are generally considered a principal rhythm contributing to movement planning and execution. Beta oscillations cohabit and dynamically interact with slow delta oscillations (0.5-4 Hz), but the role of delta oscillations and the subordinate relationship between these rhythms in the perception-action loop remains unclear. Here, we review evidence that motor delta oscillations shape the dynamics of motor behaviors and sensorimotor processes, in particular during auditory perception. We describe the functional coupling between delta and beta oscillations in the motor cortex during spontaneous and planned motor acts. In an active sensing framework, perception is strongly shaped by motor activity, in particular in the delta band, which imposes temporal constraints on the sampling of sensory information. By encoding temporal contextual information, delta oscillations modulate auditory processing and impact behavioral outcomes. Finally, we consider the contribution of motor delta oscillations in the perceptual analysis of speech signals, providing a contextual temporal frame to optimize the parsing and processing of slow linguistic information.

Entrainment of traveling waves to rhythmic motor acts

Abstract:We hypothesized that rhythmic motor acts entrain neural oscillations in speech production brain regions. We tested this hypothesis in an experiment where a subject produced consonant-vowel (CV) syllables in a rhythmic fashion, while we performed ECoG recordings. Over the ventral sensorimotor cortex (vSMC) we detected significant concentration of phase across trials at the specific frequency of speech production. We also observed amplitude modulations. In addition we found significant coupling between the phase of brain oscillations at the frequency of speech production and their amplitude in the high-gamma range (i.e., phase-amplitude coupling, PAC). Furthermore, we saw that brain oscillations at the frequency of speech production organized as traveling waves (TWs), synchronized to the rhythm of speech production. It has been hypothesized that PAC is a mechanism to allow low-frequency oscillations to synchronize with high-frequency neural activity so that spiking occurs at behaviorally relevant times. If this hypothesis is true, when PAC coexists with TWs, we expect a specific organization of PAC curves. We observed this organization experimentally and verified that the peaks of high-gamma oscillations, and therefore spiking, occur at the same times across electrodes. Importantly, we observed that these spiking times were synchronized with the rhythm of speech production. To our knowledge, this is the first report of motor actions organizing (a) the phase coherence of low-frequency brain oscillations, (b) the coupling between the phase of these oscillations and the amplitude of high-frequency oscillations, and (c) TW. It is also the first demonstration that TWs induce an organization of PAC so that spiking across spatial locations is synchronized to behaviorally relevant times.

Entrainment echoes in the cerebellum

Abstract:Historically, researchers have considered the cerebellum a coordinator of motor programs that ensures precise timing of movements and their adaptation to external events . However, it has become increasingly clear that this role is not restricted to the motor system . Rather, the cerebellum seems to play an important role in temporal prediction in general, as shown in its involvement in multiple functions that rely on precise event timing . Although previous work suggested that the cerebellum exclusively predicts the interval between two events , rather than tracking a global rhythm, it is also active when a rhythmic stimulus in rate . The latter finding is in line with a cerebellar role in speech processing that entails frequent rate changes . Neural mechanisms underlying the cerebellum’s involvement in speech processing, however, remain poorly understood. Moreover, there is a lack of studies contrasting speech and non-speech stimuli to establish speech-specificity of the observed effects . In a re-analysis of magnetoencephalography (MEG) data , we found that activity in the cerebellum aligned to rhythmic sequences of noise-vocoded speech, irrespective of its intelligibility. We then tested whether these “entrained” responses persist, and how they interact with other brain regions, when the rhythmic stimulus stopped and temporal predictions had to be updated. We found that only intelligible speech produced rhythmic responses in the cerebellum that outlasted the stimulus. During this “entrainment echo”, but not during rhythmic speech itself, cerebellar activity was coupled with that in the left inferior frontal gyrus (IFG), and specifically at rates corresponding to the preceding stimulus rhythm. This finding represents unprecedented evidence for specific cerebellum-driven temporal predictions in speech processing and their relay to cortical regions.

Abstract:Many decades after Penfield's (1937) classic depiction of the motor homunculus, it remains unclear how spatially contiguous and interconnected representations within human sensorimotor cortex might separate their activities to achieve the directed and precise control of distinct body regions evident in activities as different as typing and speaking. One long-standing but relatively neglected explanation draws from models of simple physical systems (like swinging pendulums) to posit that small differences in the oscillatory properties of neuronal populations (termed 'frequency detuning') can result in highly effective segregation of their activities and outputs. We tested this hypothesis by comparing the peak frequencies of beta-band (13-30 Hz) motor rhythms measured in a magnetoencephalographic neuroimaging study of finger and speech movements in a group of healthy adults and a group of typically developing children. Our results confirm a peak frequency task difference of about 1.5 Hz in the beta motor rhythms of both left and right hemispheres in adults. A comparable task difference was obtained in children for the left but not for the right hemisphere. These results provide novel support for the role of frequency detuning in the functional organisation of the brain and suggest that this mechanism should play a more prominent role in current models of bodily representations and their development within the sensorimotor cortex.

Abstract:Music makes people move. This human propensity to coordinate movement with the rhythm of music requires multiscale temporal integration, allowing fast sensory events composing rhythmic input to be mapped onto slower internal templates such as periodic beats. Relatedly, beat perception has been shown to involve a selective enhancement of the beat periodicities in the neural response to rhythm. However, the extent to which this ability to move to the beat, and the related periodized neural representation, are shared across the senses beyond audition remains unknown. Here we addressed this question using acoustic and tactile rhythms, while recording separately the electroencephalography (EEG) responses and finger tapping to these rhythms in healthy volunteers. Consistent with previous studies, EEG responses to the acoustic rhythm featured significant enhancement of the beat, and this periodized neural representation was specifically concentrated within a low-frequency range below 15 Hz. In contrast, the same rhythm conveyed with tactile inputs elicited responses over a broader frequency range, up to 25Hz, with no significant periodization, and resulted in less stable tapping. Together, these findings indicate that low-frequency neural activity preferentially supports behavior-relevant internal representation of rhythmic input. However, this neural representation is not necessarily shared across the senses, as well as the ability to move to the beat, corroborating multimodal differences in beat perception. This low-frequency neural representation may thus reflect a process of multiscale temporal integration allowing the auditory system to go beyond mere tracking of onset timing and support higher-level internal representation and motor entrainment to rhythm.

Abstract:Music and speech encode hierarchically organized structural complexity at the service of human expressiveness and communication. Previous research has shown that populations of neurons in auditory regions track the envelope of acoustic signals within the range of slow and fast oscillatory activity. However, the extent to which cortical tracking is influenced by the interplay between stimulus type, frequency band, and brain anatomy remains an open question. In this study, we reanalyzed intracranial recordings from thirty subjects implanted with electrocorticography (ECoG) grids in the left cerebral hemisphere, drawn from an existing open-access ECoG database. Participants passively watched a movie where visual scenes were accompanied by either music or speech stimuli. Cross-correlation between brain activity and the envelope of music and speech signals, along with density-based clustering analyses and linear mixed effect modeling, revealed both anatomically overlapping and functionally distinct mapping of the tracking effect as a function of stimulus type and frequency band. We observed widespread left-hemisphere tracking of music and speech signals in the Slow Frequency Band (SFB, band-passed filtered low-frequency signal between 1-8Hz), with near zero temporal lags. In contrast, cortical tracking in the High Frequency Band (HFB, envelope of the 70-120Hz band-passed filtered signal) was higher during speech perception, was more densely concentrated in classical language processing areas, and showed a frontal-to-temporal gradient in lag values that was not observed during perception of musical stimuli. Our results highlight a complex interaction between cortical region and frequency band that shapes temporal dynamics during processing of naturalistic music and speech signals.

Abstract:Speech production and perception involve complex neural dynamics in the human brain. Using magnetoencephalography (MEG), our study explores the interaction between cortico-cortical and cortico-subcortical connectivities during these processes. Our connectivity findings during speaking revealed a significant connection from the right cerebellum to the left temporal areas in low frequencies, which displayed an opposite trend in high frequencies. Notably, high-frequency connectivity was absent during the listening condition. These findings underscore the vital roles of cortico-cortical and cortico-subcortical connections within the speech production and perception network. The results of our new study enhance our understanding of the complex dynamics of brain connectivity during speech processes, emphasizing the distinct frequency-based interactions between various brain regions.
