Abstract:Magnesium alloys as biodegradable metal implants in orthopaedic research received a lot of interest in recent years. They have attractive biological properties including being essential to human metabolism, biocompatibility, and biodegradability. However, magnesium can corrode too rapidly in the high-chloride environment of the physiological system, loosing mechanical integrity before the tissue has sufficiently healed. Hydroxyapatite (HAp) coating was proposed to decrease the corrosion rate and improve the bioactivity of magnesium alloy. Apatite has been cathodically deposited on the surface of Mg alloy from solution that composed of 3 mM Ca(H 2 PO 4 ) 2 and 7 mM CaCl 2 at various applied potentials. The growing of HAp was confirmed on the surface of the coatings after immersion in SBF solution for 7 days. The coating obtained at −1.4 V showed higher corrosion resistance with bioactive behaviors.

Abstract:Magnesium (Mg) alloys have been promised for biomedical implants in orthopedic field, however, the fast corrosion rate and mode challenge their clinical application. To push Mg alloys materials into practice, a composite coating with biodegradable and high compatible components to improve anticorrosion property of an Mg alloy (i.e., AZ31) is designed and fabricated. The inner layer is micro-nano structured Mg(OH)2 through hydrothermal treatment. Then stearic acid (SA) is introduced to modify Mg(OH)2 for better reducing the gap below a surface-degradation polymer layer of poly(1,3-trimethylene carbonate). Benefited by the SA modification effect, this sandwiched coating avoids corrosive medium penetration via enhancing the adhesion strength at the interface between outer and inner layers. Both in vitro and in vivo tests indicate that the composite coating modified AZ31 perform a better anticorrosion behavior and biocompatibility compared to bare AZ31. Strikingly, a 1.7-fold improvement in volume of newly formed bone is observed surrounding the composite coating modified implant after 12 week implantation. The sandwiched biocompatible coating strategy paves a hopeful way for future translational application of Mg alloys orthopedic materials in clinics.

Abstract:The market for orthopedic implant alloys has seen significant growth in recent years, and efforts to reduce the carbon footprint of medical treatment (i.e., green medicine) have prompted extensive research on biodegradable magnesium-based alloys. Magnesium alloys provide the mechanical strength and biocompatibility required of medical implants; however, they are highly prone to corrosion. In this study, Mg-9Li alloy was immersed in cell culture medium to simulate degradation in the human body, while monitoring the corresponding effects of the reaction products on cells. Variations in pH revealed the generation of hydroxyl groups, which led to cell death. At day-5 of the reaction, a coating of MgCO 3 (H 2 O) 3 , HA, and α -TCP appeared on sample surfaces. The coating presented three-dimensional surface structures (at nanometer to submicron scales), anti-corrosion effects, and an altered surface micro-environment conducive to the adhesion of osteoblasts. This analysis based on bio-simulation immersion has important implications for the clinical use of Mg alloys to secure regenerated periodontal tissue.

Abstract:Magnesium alloys as bioresorbable materials with good biocompatibility have raised a growing interest in the past years in temporary implant manufacturing, as they offer a steady resorption rate and optimal healing in the body. Magnesium exhibits tensile strength properties similar to those of natural bone, which determines its application in load-bearing mechanical medical devices. In this paper, we investigated the biodegradation rate of Mg-Zn-Mn biodegradable alloys (ZMX410 and ZM21) before and after coating them with hydroxyapatite (HAP) via the electrophoretic deposition method. The experimental samples were subjected to corrosion tests to observe the effect of HAP deposition on corrosion resistance and, implicitly, the rate of biodegradation of these in simulated environments. X-ray diffraction (XRD), scanning electron microscopy (SEM) and energy-dispersive spectroscopy (EDS) provided detailed information on the quality, structure, and morphology of the HAP coating. The obtained results demonstrate that coating of Mg-Zn-Mn alloys by HAP led to the improvement of corrosion resistance in simulated environments, and that the HAP coating could be used in order to control the biodegradation rate.

Abstract:Magnesium is a candidate metal for biodegradable implant applications for its biodegradation tendency and excellent biocompatibility. Unfortunately, the high degradation rate of magnesium and also its localized degradation in physiological conditions are the main issues for its successful implant applications. The degradation rate of magnesium has been reduced to some degree via alloying, but the localized degradation susceptibility is a great concern. For many years, hydroxyapatite (HAp), a biocompatible ceramic material, has been extensively used for bio-implant applications. Recently, a substantial amount of research has been carried out on coating HAp on magnesium-based materials for improved degradation resistance in particular and also to enhance the biocompatibility. This review article focuses on the different methods of HAp coating on magnesium-based materials and also the recent cutting-edge advancements made in the coating process for improved degradation resistance and biocompatibility. The mechanical stability of the HAp coated magnesium-based materials is also discussed.

Abstract:Magnesium and its alloys have been introduced as innovative orthopedic implants due to their potential in serving lightweight, bio-active, degradable, and biocompatible properties. Mg and its alloys corrode rapidly in a biological environment and result in losing mechanical properties. However, to enhance corrosion resistance and mechanical properties, Mg alloy was plasma-sprayed with pure hydroxyapatite (HA) and HA-reinforced with strontium (Sr) powder at three levels (4, 8, and 12 wt.%). Surface parameters such as microhardness, surface roughness, and wettability were examined. The electrochemical technique was used to study the corrosion behavior in Ringer's solution. The outcomes confirmed that with the rise in Sr content in pure HA coatings, the surface properties as well as the corrosion resistance improved significantly. The contact angle of substrates under examination exhibits hydrophilic nature. Collectively, the findings of this study signify HA/Sr reinforced coatings are a promising approach to improve surface properties and corrosion behavior of Mg alloys for future bone implant applications.

Abstract:In order to solve the problem of excessive degradation rate and insufficient biocompatibility of magnesium‐based bone implants, a polyphenol (EGCG) induced hydroxyapatite (HA) coating was prepared on the surface of AZ31 alloy. The physical and chemical properties and corrosion resistance of the coating were analysed in depth, and its biocompatibility was preliminarily explored in vitro. The results showed that the polyphenol (EGCG) conversion coating constructed on the AZ31 could successfully induce the formation of HA by complexing the phenolic hydroxyl group with calcium ions. The electrochemical and long‐term immersion experiments showed that the corrosion resistance of EGCG/HA composite coating was significantly improved. The self‐corrosion current density, hydrogen evolution and the increase of pH value of AZ31‐EGCG/HA were significantly lower than those of AZ31. On the basis of inhibiting the excessive corrosion of the substrate, the composite coating significantly improves the compatibility of pre‐osteoblasts, supports the adhesion and spreading and effectively reduces the haemolysis rate to less than 5%. The preparation method of the coating is simple, low cost and suitable for complex shape surfaces, which can significantly improve the corrosion resistance and biocompatibility of the AZ31 substrate. It is expected to provide a solution for the surface modification of magnesium‐based bone implants.

Abstract:Magnesium (Mg) and its alloys have emerged as a favored candidate for bio-regenerative medical implants due to their superior biocompatibility, biodegradability and the elastic modulus close to that of human bone. Unfortunately, the rapid and uncontrollable degradation rate of Mg alloys in chloride-rich body microenvironments limits their clinical orthopedic applications. Recently, Calcium Phosphate (Ca-P) biomaterials, especially Hydroxyapatite (HA), have been broadly applied in the surface functional modification of metal-based biomaterials attributed to their excellent bioactivity and biocompatibility. Hydrothermal modification of Ca-P coatings on Mg alloys has been extensively exploited by researchers for its significant superiorities in controlling coating structure and improving interfacial bonding strength for better osseointegration and corrosion resistance. This work focuses on the up-to-the-minute advances in Ca-P coatings on the surface of Mg and its alloys via hydrothermal methods, including the strategies and mechanisms of hydrothermal modification. Herein, we are inclined to share some feasible and attractive hydrothermal surface modification strategies. From the perspectives of hydrothermal manufacturing technique innovation and coating structure optimization, we evaluate how to foster the corrosion resistance, coating bonding strength, osseointegration and antibacterial properties of Mg alloys with Ca-P coatings synthesized by hydrothermal method. The challenges and future perspectives on the follow-up exploration of Mg alloys for orthopedic applications are also elaborately proposed.

In vitro corrosion and cytocompatibility of Mg-Zn-Ca alloys coated with FHA

Abstract:In the field of orthopedics, it's crucial to effectively slow down the degradation rate of Mg alloys. This study aims to improve the degradation behavior of Mg-Zn-Ca alloys by electrodepositing fluorohydroxyapatite (FHA). We investigated the microstructure and bond strength of the deposition, as well as degradation and cellular reactions. After 15-30 days of degradation in Hanks solution, FHA deposited alloys showed enhanced stability and less pH change. The strong interfacial bond between FHA and the Mg-Zn-Ca substrate was verified through scratch tests (Critical loads: 10.73 ± 0.014 N in Mg-Zn-0.5Ca alloys). Cellular studies demonstrated that FHA-coated alloys exhibited good cytocompatibility and promoted the growth of MC3T3-E1 cells. Further tests showed FHA-coated alloys owed improved early bone mineralization and osteogenic properties, especially in Mg-Zn-0.5Ca. This research highlighted the potential of FHA-coated Mg-Zn-0.5Ca alloys in orthopedics applications.
Pure Mg–Al Layered Double Hydroxide Film on Magnesium Alloys for Orthopedic Applications

Abstract:Mg alloys are promising biodegradable orthopedic implants in the future. However, poor corrosion resistance and biocompatibility limit their wide applications. In this study, a pure Mg–Al layered double hydroxide (Mg–Al LDH) film on AZ31 was prepared through combining hydrofluoric acid pretreatment and hydrothermal treatment. Electrochemical analysis and the immersion test suggested that the as-prepared Mg–Al LDH-coated sample exhibited significantly enhanced corrosion resistance. The in vitro cell culture revealed that the Mg–Al LDH film was favorable for the alkaline phosphatase activity, collagen secretion, and osteogenesis-related gene expression of MC3T3-E1. Furthermore, the LDH-coated sample was beneficial for the migration, vascular endothelial growth factor secretion, and angiogenesis-related gene expression of human umbilical vein endothelial cells. The subcutaneous implantation test demonstrated that the Mg–Al LDH film could protect the substrate from corrosion and induce milder inflammation. The femur implantation demonstrated that the Mg–Al LDH sample showed better bone regeneration and osseointegration than bare AZ31. In summary, the as-prepared pure Mg–Al LDH film is able to enhance the in vitro and in vivo performances of AZ31, indicating a promising application in the orthopedic field.This article has not yet been cited by other publications.

Polydopamine mediated assembly of hydroxyapatite nanoparticles and bone morphogenetic protein-2 on magnesium alloys for enhanced corrosion resistance and bone regeneration

Abstract:Magnesium alloys have the great potential to be used as orthopedic implants due to their biodegradability and mechanical resemblance to human cortical bone. However, the rapid degradation in physiological environment with the evolution of hydrogen gas release hinders their clinical applications. In this study, we developed a novel functional and biocompatible coating strategy through polydopamine mediated assembly of hydroxyapatite nanoparticles and growth factor, bone morphogenetic protein-2 (BMP-2), onto the surface of AZ31 Mg alloys. Such functional coating has strong bonding with the substrate and can increase surface hydrophilicity of magnesium alloys. In vitro electrochemical corrosion and hydrogen evolution tests demonstrate that the coating can significantly enhance the corrosion resistance and therefore slow down the degradation of AZ31 Mg alloys. In vitro cell culture reveals that immobilization of HA nanoparticles and BMP-2 can obviously promote cell adhesion and proliferation. Furthermore, in vivo implantation tests indicate that with the synergistic effects of HA nanoparticles and BMP-2, the coating does not cause obvious inflammatory response and can significantly reduce the biodegradation rate of the magnesium alloys and induce the new bone formation adjacent to the implants. © 2017 Wiley Periodicals, Inc. Optimization of the in Vitro Biodegradability, Cytocompatibility, and Wear Resistance of the AZ31B Alloy by Micro-Arc Oxidation Coatings Doped with Zinc Phosphate

Abstract:As implanted bone fixation materials, magnesium (Mg) alloys have significant advantages because the density and elastic modulus are closest to those of the human bone and they can bio-degrade in the physiological environment. However, Mg alloys degrade too rapidly and uncontrollably thus hampering clinical adoption. In this study, a highly corrosion-resistant zinc-phosphate-doped micro-arc oxidation (MAO) coating is prepared on the AZ31B alloy, and the degradation process is assessed in vitro. With increasing zinc phosphate concentrations, both the corrosion potentials and charge transfer resistance of the AZ31B alloy coated with MAO coatings increase gradually, while the corrosion current densities di-minish gradually. Immersion tests in the simulated body fluid (SBF) reveal that the increased zinc phos-phate concentration in MAO coating decreases the degradation rate, consequently reducing the release rates of Mg2+ and OH - in the physiological micro-environment, which obtains the lowest weight loss of only 5.22% after immersion for 56 days. Effective regulation of degradation provides a weak alkaline environment that is suitable for long-term cell growth and subsequent promotion of bone proliferation, differentiation, mineralization, and cytocompatibility. In addition, the zinc-phosphate-doped MAO coat-ings show an improved wear resistance as manifested by a wear rate of only 3.81 x 10 -5 mm3 N -1 m -1 . Abstract:Magnesium alloys as biodegradable materials have received great attention for orthopedic application as a result of their good biocompatibility, bioactivity, and mechanical properties. However, the clinical use of Mg alloys is restricted by high degradation rate. In order to reduce the degradation rate, TiO2 incorporated micro-arc oxidation (TM) coatings were prepared on Mg- Ca alloy using micro-arc oxidation (MAO). Subsequently, zinc-doped hydroxyapatite (ZH) coating was deposited by electrophoretic deposition (EPD) on the MAO coating. The electrochemical test results demonstrated that the deposition of ZH composite coatings on Mg alloy significantly reduces its corrosion rate and improves its charge transfer resistance. Antibacterial activity of the coating against Escherichia coli (E. coli) was studied using disk-diffusion and spread plate methods. The results revealed that the inhibition zone amplified after deposition of TM and ZH coatings on Mg alloy, whereas more inhibition zone was found around ZH coating. In addition, the number of E. coli colonies reduces to 92% after ZH coating implying its good antibacterial properties. The cytotoxicity test indicated that cell viability of MG63 osteoblast cells cultured with ZH extracts was higher compared to the TM coating and bare Mg alloy. These results confirm that Mg alloy coated by TM/ZH exhibits high corrosion resistance, antibacterial activity and favorable bioactivity and cytocompatibility, indicating their substantial potentials for biomedical applications.

Abstract:Recently, magnesium (Mg) and its alloys have attracted more attention because of their biodegradability and fascinating mechanical properties in the medical field. However, their low corrosion resistance and high degradability in the body have a great effect on mechanical stability and cytocompatibility, which hinders its clinical applications. Therefore, here we introduce a bifunctional composite coating composed of polycaprolactone and synthesized hydroxyapatite nanoparticles (HA-NPs) loaded with simvastatin deposited on the AZ31 alloy via electrospinning technique. The synthesized HA-NPs and composite nanofibers layer were characterized using TEM, FE-SEM, FTIR, and XRD to understand the physiochemical properties of the composite nanofibers compared to pristine polymer and bare alloy. Corrosion resistance was evaluated electrochemically using potentiodynamic polarization and EIS measurements, and biodegradability was evaluated in terms of pH and Mg ions release in SBF solution. The as-prepared coating was found to retard the corrosion and increased the osteocompatibility as resulted in cell culture test, a higher cell attachment and proliferation on the implant biointerface, in addition to releasing simvastatin in a controlled platform.

Construction of bio-functional Mg/HA composite layered coating for orthopedic application

Abstract:Magnesium was considered as a revolutionary biodegradable implant material for orthopedic application. Concerning the weakness of intrinsic strength and corrosion behavior, a novel strategy of Mg/metal hybrid system was proposed for extension of orthopedic application, especially at load-bearing site. In this work, an Mg and HA composite layered coating was constructed on titanium by means of chemical conversion and vapor deposition. The HA transition interlayer was introduced to enhance the bonding between Mg film and Ti substrate. Compared with the bare Mg coating, the Mg/HA coating presented good interface bonding, which avoided the occurrence of Mg film peeling off from the substrate. The Mg/HA coating showed a uniform degradation and kept integrity after immersion of 14 d. The Mg ions release by degradation played a crucial role in osteopromotion and antibacterial effect. Incubation of MC3T3-E1 osteoblasts with the Mg/HA coating showed significant promotion on osteogenic differentiation according to ALP activity and Alizarin Red staining assays. Meanwhile the degradation of Mg exhibited strong suppression of bacteria proliferation. It was believed that this novel Mg/HA composite layered coating could be potentially applied in further development of bio-functional hybrid orthopedic implants.

Abstract:Magnesium-based implants are subjected to complicated stresses during implantation in the human body. In this case, they undergo stress-induced corrosion and usually lead to early breakage or disintegration of implants. Calcium phosphate compounds have been used as coatings, bone cement and drug carrier in orthopedic application. Their success has been expanded to Mg alloys. Calcium phosphate-based coatings succeeded in corrosion control and osteointegration of Mg alloys in a big extent. However, their protective effect under stress condition is questionable considering the brittle nature. With the assistance of high pH microenvironment near Mg alloy surface, calcium phosphate coatings have the self-repair potential via the dissolution-precipitation mechanism. Inspired by this phenomenon, strontium doped hydroxyapatite (Sr-HA) coatings with higher solubility have been prepared on the surface of Mg alloys. Sr-HA coatings served as not only a physical barrier effect to postpone pitting formation, but also provide ion supplement to seal the cracks initiated during tensile deformation. The results obtained in slow strain rate tensile test (SSRT) demonstrated improved corrosion and mechanical behaviors of Sr-HA coated Mg alloys. Specifically, the elongation of Sr-HA coated alloy increased by 56.7 and 35.2% as compared to the bare and HA coated one in DMEM. In ovariectomized rats, the implants exhibited only 2.7–5.6% reduction of implant volume after 12 weeks implantation. Sr-HA coating further increased early new bone formation than HA coated implants. 12 weeks post-implantation, a large amount of bony tissue with higher trabecular thickness and hardness was formed adjacent to the Sr-HA coated implants. This study demonstrated the efficiency of a rapid dissolved Sr-HA coating on the corrosion behavior of Mg alloys under mechanical loading and the osteogenesis in ovariectomized rats. It therefore holds a promising practical significance for the future coating design strategy for Mg alloys when trying to expand their application range to locations with higher loading conditions.

Abstract:To slow down the corrosion rate and increase the bioactivity of magnesium alloys, biodegradable hydroxyapatite/magnesium (HA/Mg) composite coating was deposited onto AZ91D alloy substrate by high-velocity suspension flame spray technique using nano-sized HA and micron-sized Mg powders. Microstructures and phase constitutions of HA/Mg composite coating were analyzed by scanning electron microscopy and x-ray diffraction. Corrosion resistance of composite coating was also evaluated by potentiodynamic polarization test and electrochemical impedance spectroscopy in Hanks’ balanced salt solution (HBSS). The result shows that the HA/Mg composite coating mainly consisted of Mg and HA in addition to a few of MgO phases. The coating showed a rough surface morphology with homogeneous elemental distribution and a lamellar structure in cross section as well as well-bonded coating/substrate interface. Mg particles presented superior deposition during spraying due to their higher melting degree compared to HA particles. AZ91D alloy substrate was significantly protected from corrosion in HBSS by the HA/Mg composite coating with Mg dissolution on surfaces of the composite coating. This strategy can offer a new route to fabricate HA/Mg composite coatings on other metal substrates for use in biodegradable implant fields.

Abstract:A newly-developed degradable Mg-1Ca (mass fraction%) alloy was modified with porous ceramic coatings through Micro-arc oxidation (MAO) technique in alkaline silicate electrolyte. The sparking behavior and growth characteristics of oxide coatings were discussed. The influence of working voltage and treatment time on the the resulting coatings' structure and composition were investigated by SEM-EDS, XPS and XRD. Potentiodynamic polarization test was carried out to understand corrosion behavior of the coated Mg-1Ca in Kokubo's SBF solution, in which the optimal MAO treatment at 360 V for 5 min was determined.

Abstract:As potential biological materials for degradable temporary implants, magnesium (Mg) and its alloys have get extensive attentions in recent years. However, magnesium alloys are easily corroded in the physiological environment, so their wide application is limited in the biological field. In this work, the super-hydrophobic coating composed by the mixture of magnesium compounds and zinc substance is fabricated by a simple wet chemical method. The coating possesses the three-dimensional nano-sheets structure, exhibits excellent anti-contamination and super-hydrophobic property with a water contact angle of 162.04°. The super-hydrophobic characteristic of this coating results in that the corrosion resistance of Mg alloys of in the phosphate buffer saline (PBS) is improved and the adhesion of blood platelet on the surface is inhibited. In addition, the super-hydrophobic film exhibits both the low hemolysis ratio and the high cell toxicity, which may be anticipated as potential medical implant materials.

Abstract:Biodegradable magnesium (Mg) alloys exhibit excellent biocompatibility, adequate mechanical properties, and osteogenic effect. They can contribute to complete recovery of damaged tissues without concerns about a second surgery and have achieved clinical applications in orthopedic and cardiovascular fields. Porous scaffolds can provide functions such as bone integration and adjustable mechanical properties, thus widely used for bone repair. Additive manufacturing (AM) offers the advantages of design freedom and high precision, enabling the reliable production of porous scaffolds with customized structures. The combination of biodegradable Mg alloys, porous scaffolds, and AM processes has created tremendous opportunities for the precision treatment of bone defects. This article reviews the current development in the additive manufacturing process and design of Mg alloy biodegradable orthopedic implants, focusing on chemical compositions, structural design, surface treatment, and their effects on mechanical properties, degradation behavior, and biocompatibility.
