Abstract:The ultimate objective of hybrid miniemulsion polymerization is to produce a water-based crosslinkable coating through in situ grafting of a free radical growing acrylic polymer with an unsaturated resin. Certain authors have reported low grafting while others have reported higher. This article explores the factors that influence the grafting tendencies of these systems. Methacrylates such as methyl methacrylate (MMA) have a sterically hindered radical center that lowers its reactivity toward unsaturated resin. This steric hindrance from the methyl group forces grafting of this type of monomer to occur by abstraction of a hydrogen allylic to a resinous double bond. This chain transfer produces a relatively inactive radical on the resin that reduces the grafting efficiency. The transfer process also inherently produces some degree of terminated PMMA polymer within the particle. Grafting occurs in this type of system through termination of living PMMA chains with that radical produced on the resin. For relatively water-soluble monomers such as MMA, grafting efficiency is further lessened by homogeneous nucleation resulting from the monomer hydrophilicity. These newly created particles cannot contain alkyd due to its hydrophobicity and thus inability to transport across the aqueous phase, and hence cannot produce grafted polymer. Nonetheless, degree of grafting of nearly 50% was observed in these systems. For hybrid systems involving an acrylate monomer such as butyl acrylate (BA), virtually complete grafting with alkyd was observed. This is due to the uninhibited BA radical center allowing the molecule to add directly through a resin double bond. This process offers the possibility for complete grafting. Homogeneous nucleation is not involved in this system due to the insolubility of BA in the aqueous phase. Grafting through Method for Implanting of Lysozyme Enzyme in Molecular Brush for Improved Biocatalytic Activity and Thermal Stability

Abstract:We report a “grafting through” conjugation strategy to improve lysozyme catalytic activity and thermal stability by the synthesis of a synthetic polymer–enzyme hybrid. The lysozyme was first conjugated with glycidyl methacrylate via ring-opening reaction between epoxy groups and lysine residues of the enzyme to synthesize the enzyme macromonomer. The conjugation was followed by free radical copolymerization of the macromonomer with poly(ethylene glycol) methyl ether acrylate (PEGMEA) (M n = 5000 g/mol) and poly(ethylene glycol) methyl ether methacrylate (PEGMEMA) (M n = 500 g/mol). We demonstrated that implanting of a single lysozyme molecule in the molecular brush polymer chain resulted a significant improvement of the thermal stability up to 90 °C and 9-time extended half-life for this synthetic enzyme structure. The improved enzyme performance is explained by the crowding effect provided by molecular brush architecture of the synthetic hybrid.

Abstract:Effective utilization of abundant industrial lignin has growingly attracted much attention besides potential environmental issues. Although chemical graft polymerization modification is one facile strategy for extending its applications, it remains an intractable challenge to select a highly efficient initiation system. We herein have attempt to understand the CaCl2–H2O2 system in initiating the graft polymerization of acrylic monomers onto acetic acid lignin (AAL) and biobutanol lignin (BBL). The initiation system is found to be highly efficient and selective, as proved by the successful graft of polyacrylates onto them, and a possible mechanism is also proposed. Thermal analysis shows that the graft modification results in a higher glass transition temperature and higher thermal stability of lignin. The graft modification make both AAL and BBL become more hydrophobic than before modification. Abstract:Precision remodeling of glycans in their native environments is pivotal for understanding glycan-mediated biological events and has important biotechnological implications in fields of clinical diagnosis, glyco-immune checkpoint therapy, and so forth. However, the influence of aglycone-steric diversity on the selectivity of glycan remodeling has been largely overlooked, limiting the application in complex biological scenarios. Here, we report the achievement of aglycone sterics-selective enzymatic glycan remodeling by controlled grafting of functional polymers from glycoenzyme. Through tuning polymer length, a series of enzyme-polymer composites with varying substrate permeability are prepared, which afford an activity pattern-based differentiation strategy for aglycone sterics. This leads to the implementation of glycolipid's partner screening, and aglycone sterics-selective glycan remodeling in a complex biological environment. We further orchestrate the polymer length adjustment with external cues to regulate aglycone-steric selectivity in a multi-faceted fashion, resulting in an unexpected enhancement of glycolipid remodeling, and temporal control of glycan remodeling on live cells.

Abstract:The on‐demand building copolymer structures, from sequence to architecture, is crucial in understanding the relation between polymer structure and property, meanwhile motivating the innovation of polymer hierarchy. However, the challenge is conspicuous for complicated polymer structures from inherently intricate polymerization. In this work, copolymers with tailored grafting density and distributions were achieved using successive latent monomer and grafting‐from strategies. The hydroxyl group functionalized furan/maleimide adduct (FMOH) was selected as the latent monomer for RAFT polymerization of an array of copolymers with tailored localization of hydroxyl group along the main chain. The hydroxyl group further initiated the ring opening polymerization (ROP) of L‐lactide or ε‐caprolactone, resulting in a library of multicomponent copolymers via grafting‐from strategy. The initiating efficiency reached to ~100% with variable molecular weight (21,300 – 58,600 Da) and narrow distributions (ÐM < 1.25), indicating that such graft copolymers possessed controlled density and distribution of side chains as its linear template. The investigation on thermal properties of the well‐defined graft copolymers implied that the precise tailoring over copolymer structures at the molecule level could lead to tunable chemical/physical properties. This work bridged polymer from sequence to architecture, unveiled a new method in creating graft copolymers with programable structures and provided the insight into the structure/property relationship.This article is protected by copyright. Synthesis of starch graft-copolymers via reactive extrusion: Process development and structural analysis

Abstract:To promote the large-scale production of starch-based biomaterials, we developed a method of synthesis based on reactive extrusion that combines the benefits of continuous manufacturing with the use of green chemistry principles. This paper describes the grafting of four different types of starches with acrylamide monomers via free radical copolymerization using twin-screw extrusion technology. The elemental analysis confirmed the success of this method, with an average monomer conversion of 80% and grafting efficiency of ˜74% across all samples. The 13C-NMR/1H-NMR and ATR-FTIR analysis revealed that the type of starch substrate used strongly influenced the mechanism of the grafting reaction. Thermal analysis (TG/DTG-DSC) indicated that the graft-copolymers thermal stability was influenced by the amylose-amylopectin content ratio. Swelling tests suggested that cationic modification of the substrate is a promising approach to produce stimuli-responsive graft-copolymers via reactive extrusion. The proposed method has proved to be a viable alternative for the production of starch-copolymers.

Abstract:A novel approach was developed to prepare cellulose acetoacetate-graft-copolymers via horseradish peroxidase (HRP)-mediated polymerization. Cellulose acetoacetate (CAA), the macroinitiator, was obtained by transesterification between tert-butyl acetoacetate (t-BAA) and cellulose in ionic liquid. The CAA-graft-polyacrylamide (CAA-g-PAM) was synthesized using a ternary initiator system consisting of CAA, HRP and hydrogen peroxide (H2O2). The results of surface-initiated polymerization showed that the formation of free polymers could be inhibited in this process. The kinetics exhibited that this method is a powerful tool for obtaining graft copolymers. The effects of H2O2 concentration and reaction temperature were investigated to explore the optimal conditions for the process of enzymatic initiation. The synthesized copolymers were characterized by FT IR, NMR, TGA and elemental analysis to confirm the successful grafting of polyacrylamide chains onto the cellulose backbones. Furthermore, generality of the HRP-mediated graft polymerization approach was demonstrated using various monomers, including 2-hydroxyethyl methacrylate (HEMA), methyl methacrylate (MMA) and sulfobetaine methacrylate (SBMA). Abstract:Molecules adsorbed or attached on a surface is a quite basic phenomenon in numerous chemical or biological systems. Grafting-onto is considered as a feasible way to achieve it. The grafting reaction is essentially controlled by the diffusion of the molecules, thus it is more likely a physical issue, instead of a chemical issue. Because of the experimental difficulty in measuring the properties of surface-attached molecules (e.g., the polymeric molecules), the surface-bound molecules are often assumed as with the same properties as that of the start feeding ones in solution. This assumption was even used to guide further characterization, while it is proved to be invalid by different quantifying methods. Consequently, an effective prediction for the properties of surface-bound molecules is still lacking. Based on a microscopic level and a dynamic perspective, the grafting process onto a flat substrate with polydisperse feeding polymeric molecules is investigated in-depth by coarse-grained Brownian dynamics simulation as well as model analysis. We find from simulations that for the final grafting density σg and the mean chain length of start feeding molecules <N0>, the dependence of σg-<N0>γ with the constant exponential factor γ may be a determined rule for one-end functionalized flexible linear polymer chains grafting on the flat substrate. Since grafting-onto is a multiple interplayed process, our simulation study indicates that there is an optimized initial concentration of start feeding molecules for achieving high grafting density of surface-bound polymers. We also propose a correctional equation to quantitatively predict the molecular weight distribution (MWD) of surface-bound polymeric molecules, which may be effective for predicting the MWD of the surface-bound ones in specific conditions. This simulation study helps to better understand the kinetics of grafting-onto process, and serves as a theoretical guide to achieve the precise design of surface modification materials via grafting-onto strategy.

Abstract:A novel approach to uniformly bulk graft modification of pre-existing polymer materials without ungrafted homopolymers was achieved at relatively low temperature by means of gamma (γ)-rays pre-irradiation-induced “grafting-from” polymerization with supercritical carbon dioxide (scCO2) both as solvent and as swelling agent. The polymer substrates were first irradiated with γ-rays originated from cobalt-60 resource under nitrogen atmosphere at ambient temperature, and thereby leading to uniform formation of trapped radicals on polymer backbone. Then, the produced polymer-trapped radicals were utilized to initiate graft polymerization of vinyl monomers dissolved in scCO2 within polymer substrates. A combination of transmission electron microscopy, scanning electron microscopy and elemental analysis shows that side graft chains covalently bonded to polymer substrate backbone in nanometer scale are uniformly dispersed within polymer membranes with a thickness of 5mm. Altering the experimental conditions can easily control the grafting yield with regard to side graft-chain length. This method can also be applied to various functional polymer chains covalently attached to different polymer substrates, even with high viscosity or limited solubility. The novel graft-polymer materials produced by this method are not formed in the absence of scCO2 and are impossible to prepare by conventional methodologies.

Abstract:The capability to graft synthetic polymers onto the surfaces of live cells offers the potential to manipulate and control their phenotype and underlying cellular processes. Conventional grafting-to strategies for conjugating preformed polymers to cell surfaces are limited by low polymer grafting efficiency. Here we report an alternative grafting-from strategy for directly engineering the surfaces of live yeast and mammalian cells through cell surface-initiated controlled radical polymerization. By developing cytocompatible PET-RAFT (photoinduced electron transfer-reversible addition-fragmentation chain-transfer polymerization), synthetic polymers with narrow polydispersity (Mw/Mn < 1.3) could be obtained at room temperature in 5 minutes. This polymerization strategy enables chain growth to be initiated directly from chain-transfer agents anchored on the surface of live cells using either covalent attachment or non-covalent insertion, while maintaining high cell viability. Compared with conventional grafting-to approaches, these methods significantly improve the efficiency of grafting polymer chains and enable the active manipulation of cellular phenotypes.

Abstract:Protein-polymer conjugates combine properties of biopolymers and synthetic polymers, such as specific bioactivity and increased stability, with great benefits for various applications from catalysis to biomedicine. Furthermore, polymer conjugation can mimic important posttranslational modifications of proteins such as glycosylation. There are typically two approaches to create protein-polymer conjugates: the protein is functionalized in advance with an initiator for a grafting-from method or a previously produced polymer is conjugated to the protein via a grafting-to method. In this study, we present a new approach that uses native proteins and allows for direct grafting-from using a thiol-induced, light-activated controlled radical polymerization (TIRP) that is initiated at thiols from specific cysteine residues of the protein. This straightforward method is employed to introduce polymers onto proteins and enzymes without any prior protein modifications, it works in aqueous buffer and maintains the protein's native structure and activity. The resulting protein-polymer conjugates exhibit high molar masses and low dispersities. We demonstrate the versatility of this approach by introducing different types of polymers such as hydrophilic poly(2-hydroxyethyl acrylate) (pHEAA), temperature-responsive poly(N-isopropylacrylamide) (pNIPAM) as well as glycopolymers mimicking the natural protein glycosylation and enabling selective interactions. We present successful combinations of the protein and polymer functions e.g., temperature-induced aggregation leading to an increase in enzyme activity and the introduction of artificial glycosylation inducing specific protein-protein cluster formation and giving straightforward access to glycosurfaces. Based on this straightforward, potentially scalable yet highly controlled synthesis of protein-polymer conjugates, various areas of applications are envisioned ranging from biomedicine to material sciences.

Abstract:Through the graft polymerization of acrylic monomers onto starch, materials with interesting new properties can be synthesized. Fenton's chemistry, Fe2+/H2O2, is considered to be attractive for the initiation of graft polymerization with the monomer acrylic acid since it is cheap and reacts quickly at ambient conditions and should therefore be easy to scale up. However, the selectivity of the grafting versus the homopolymerization reaction poses a challenge with this monomer and this type of initiator. In the present review paper, we investigate why data from the literature on grafting systems with other monomers and initiation systems tend to show higher graft selectivity. A scheme is presented, based on reaction engineering principles, that supports an explanation for these observed differences. It is found that more selective activation of starch is a factor, but perhaps even more important is a low monomer-to-starch ratio at the starting sites of graft reactions. Since water is the most common solvent, monomers that are less water-soluble have an advantage in this respect. Based on the proposed scheme, methods to improve the graft selectivity with Fenton's initiator and acrylic acid are evaluated. Most promising appears to be a method of gradual monomer dosage. With gelatinized cassava starch in a batch reactor, both the grafting percentage (17 => 29%) and graft selectivity (18 => 31%) could be improved. This can be considered a principal breakthrough. Still, more research and development would be needed to refine the method and to implement the idea in a continuous reactor at a larger scale.

Abstract:Transferring the biological function of one protein to another is a key issue in understanding the structure and function relationship of proteins. We have developed a strategy for grafting protein-protein interaction epitopes. As a first step, residues at the interface of the ligand protein which strongly interact with the receptor protein were identified. Then protein scaffolds were docked onto receptor protein based on geometric complementarity. Only high docking score matches were saved. For each saved match, the scaffold protein was accepted if it had suitable positions for grafting key interaction residues of the ligand protein. These candidate residues were mutated to corresponding residues in the ligand protein at each relevant position and the mutated scaffold protein was co-minimized with receptor protein. Finally, the minimized complexes were evaluated by a scoring function deduced from statistical analysis of rigid binding data sets. As a test case, the binding epitope of barstar, the inhibitor of barnase, was grafted onto smaller proteins. Pheromone Er-1 (PDB entry 1erc) has been found to be a good scaffold. The calculated binding free energy for mutated Pheromone Er-1 is equivalent to that of barstar.
Novel Photoinduced Grafting-Chemical Reaction Sequence for the Construction of A Glycosylation Surface

Abstract:Carbohydrates play a major role in many recognition events, such as blood coagulation, immune response, fertilization, cell growth, embryogenesis, and cellular signal transfer, which are essential for the survival of living entities. Synthetic carbohydrate-based polymers, so-called glycopolymers, are emerging as important well-defined tools for investigating carbohydrate-based biological processes and for simulating various functions of carbohydrates. In this work, we present a facile strategy for the formation of glycopolymer tethered on polypropylene microporous membrane surface. Acrylamide was grafted onto the polypropylene microporous membrane surface by photoinduced graft polymerization in the presence of benzophenone. The amide groups of grafted poly(acrylamide) were then transformed to primary amine groups by the Hofmann rearrangement reaction. Quantificational evaluation of the rearrangement reaction was carried out by ninhydrin method and mass weighting. Sugar moieties were coupled with the grafted functional layer to form glycopolymer by the reaction between primary amine groups and carbohydrate lactones. The grafting of acrylamide, the conversion of amide groups to amine groups, and the coupling of sugar moieties were confirmed by Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy combined with surface morphology observation by scanning electron microscopy.
Activation and stabilization of a lipase nanogel using GMA for acryloylation

Abstract:A lipase nanogel has been prepared by aqueous in situ polymerization, with initial acryloylation to introduce vinyl groups onto the lipase surface for subsequent polymerization with acrylamide monomers. Activation of lipase was observed using glycidyl methacrylate (GMA) as the acryloylation agent, leading to an increase of activity yield from 78 +/- 4% (obtained using NAS for acryloylation) to 122 +/- 14%. The acryloylation ratio was also improved, from 8 +/- 5% to 20 +/- 8%, which favored the subsequent polymerization. In addition, the overall activity yield of the lipase nanogel was increased from 44 +/- 3% to 105 +/- 2%. The lipase nanogel prepared with GMA for acryloylation displayed a lower K-m and a higher k(cat) in comparison to its native counterpart, as well as the nanogel obtained using NAS for acryloylation. The half-life of the lipase nanogel at 60 degrees C was extended from 1.63 h (using NAS for acryloylation) to 5.61 h, while the half-life of the native lipase was 1.41 h. Molecular dynamics simulations and experiments further suggested that the activation effect was due in part to interactions between the lipase and the GMA, which reinforced the 'open' configuration of the lipase, thereby facilitating mass transport to and from the modified lipase in the free and encapsulated forms. The above mentioned activation effect of the enzyme nanogel illustrated the power of chemical modification by tailored design of the nanostructure of the enzyme catalyst, potentially expanding the applications of enzyme catalysis.

Abstract:Protein engineering techniques have vastly expanded their domain of impact, notably following the success of antibodies. Likewise, smaller peptide therapeutics have carved an increasingly significant niche for themselves in the pharmaceutical landscape. The concept of grafting such peptide onto larger protein scaffolds, thus harvesting the advantages of both, has given rise to a variety of protein engineering strategies that are reviewed herein. We also describe our own "Lasso‐Grafting" approach, which combines traditional grafting concepts with mRNA display to streamline the production of multiple grafted drug candidates for virtually any target.

Abstract:The considerable utility of glycoside phosphorylases (GPs) has led to substantial efforts over the past two decades to expand the breadth of known GP activities. Driven largely by the increase of available genomic DNA sequence data, the gap between the number of sequences in the carbohydrate active enzyme database (CAZy DB) and its functionally characterized members continues to grow. This wealth of sequence data presented an exciting opportunity to explore the ever-expanding CAZy DB to discover new GPs with never-before-described functionalities. Utilizing an in silico sequence analysis of CAZy family GH94, we discovered and then functionally and structurally characterized the new GP β-1,3-N-acetylglucosaminide phosphorylase. This new GP was sourced from the genome of the cell-wall-less Mollicute bacterium, Acholeplasma laidlawii and was found to synthesize β-1,3-linked N-acetylglucosaminide linkages. The resulting poly-β-1,3-N-acetylglucosamine represents a new, previously undescribed biopolymer that completes the set of possible β-linked GlcNAc homopolysaccharides together with chitin (β-1,4) and PNAG (poly-β-1,6-N-acetylglucosamine). The new biopolymer was denoted acholetin, a combination of the genus Acholeplasma and the polysaccharide chitin, and the new GP was thus denoted acholetin phosphorylase (AchP). Use of the reverse phosphorolysis action of AchP provides an efficient method to enzymatically synthesize acholetin, which is a new biodegradable polymeric material.The identification of a novel glycoside phosphorylase enzyme led to the discovery of a new biodegradable polysaccharide named acholetin.This article has not yet been cited by other publications.

Abstract:Graft polymers are gaining increasing interest because of their unique architectural characteristics. We recently reported a novel type of depolymerizable graft polymer based on poly( trans -cyclobutane fused cyclooctene), in an effort to address the trade-off between depolymerizability and controlled grafting-through polymerization. In this work, we examine the thermal, mechanical, and morphological properties of a graft copolymer thermoplastic material prepared by copolymerizing poly(L-lactide) and margaric acid-based macromonomers. A copolymerization kinetics study reveals that the two macromonomers are incorporated almost randomly and that the domain spacing measured from small-angle X-ray scattering is consistent with the random distribution. An investigation of the crystallization behavior suggests that proper thermal treatment is required to maximize, or to even observe crystallinity. The physical states of the soft and hard domains, whether melt, glassy, or semicrystalline, significantly impact the tensile properties of the resulting copolymer materials. Finally, the rheological properties and morphological features are discussed.

Abstract:Lipase from Rhizopus oryzae (ROL) exhibits remarkable sn-1,3 stereoselectivity and catalytic activity, but its poor thermostability limits its applications in the production of 1,3-dioleoyl-2-palmitoyl glycerol (OPO, a high-quality substitute for human milk fat). In this work, a semirational method was proposed to engineer the thermostability and catalytic activity of 4M (ROL mutant in our previous study). First, a computer-aided design is performed using 4M as a template, and N-glycosylation mutants are then recombinantly expressed and screened in Pichia pastoris, the optimal mutant N227 exhibited a half-life of 298.8 h at 45 °C, which is 7.23-folds longer than that of 4M. Its catalytic activity also reached 1043.80 ± 61.98 U/mg, representing a 29.2% increase compared to 4M (808.02 ± 47.02 U/mg). Molecular dynamics simulations of N227 suggested that the introduction of glycan enhanced the protein rigidity, while the strong hydrogen bonds formed between the glycan and the protein stabilized the lipase structure, thereby improving its thermostability. The acidolysis reaction between oleic acid (OA) and glycerol tripalmitate (PPP) was successfully carried out using immobilized N227, achieving a molar conversion rate of 90.2% for PPP. This engineering strategy guides the modification of lipases, while the glycomutants obtained in this study have potential applications in the biosynthesis of OPO.
