Abstract:Plant regeneration from protoplasts has proved a very useful technique for crop genetic improvement programs,genetic transformation,and somatic hybridization etc.Now reliable procedures are available for protoplast isolation and culture from a range of high plants including cotton.There were many successful reports on plant regeneration from protoplast.Great progresses was also made for cotton somatic hybridization via protoplast fusion,several inter-specific hybrids were created.As is known,there were 47 wild species,which have advantageous properties such as disease and herbicide resistance that the genetic improvements need.Somatic hybridization is a unique useful way to transfer both nuclear and cytoplasmic genes of different wild species simultaneously in comparison to chromosomal and gene engineering.The success of protoplast fusion in cotton was based on the effective system of cotton protoplast culture,and plant regeneration via somatic embryogenesis from protoplasts was a key step to somatic hybridization.So it is necessary to establish a highly reproducible system for efficient plant regeneration from protoplasts or fused protoplasts in cotton.Advances on fundamental techniques,research status,problems,and promising development of cotton protoplast culture,as well as the somatic hybridization were reviewed,in order to offer beneficial references for cotton gene and chromosome engineering.
Stadies on Long-term Preservation Techniques of the Intergeneric Hybrid-Common Wheat×Eremopyrum Orientale and Its Somaclonal Stability

Abstract:Based on studies of the totally sterile intergeneric hybrid of common wheat (T. aestivum)×Er. orientale and its somatic culture, especially the suitable components of the media, the relationship between inducing rate and length of the explant and the culture methods, a longterm preservation system was established. 1.1～3.0 cm long immature infioreseses of 2n=35 F1 plants were used as explants. They Were cut into pieces as small as possible before culture. Modified MS medium(inorganic elements of MS+organic elements of W14+400 mg/L LH+150 mg/L Asp+2 mg/L 2,4-D)were used as inducing medium, with varied content of 2,4-D to adjust the growth state of calli untill embryonic calli appeared. After subcultured for 4～5 months, the calli were transferred to the differentiation medium(N6+400 mg/L LH+150 mg/L Asp+0.5 mg/L IAA)to differentiate into plantlets. By using N6+400 mg/L LH+150 mg/L Asp+10 mg/L IAA as strong sprout medium and separating tillers in vitro, a large number of healthy seedlings were obtained. Studies on the somaclonal variation revealed that 37 percent of the plantlets showed morphological variation and the chromosome numbers of the F1 plants varied between 32～35. 2n=32 plants accounted for 19.6 percent, 2n=33 plants accounted for 6.5 percent and the rest were all 2n=35. A detailed survey of some 2n=32 variants showed that their plant height and tillering ability decreased by 49.1% and 49. 90% respectively and the flowering date became later while the number of spiklelets increased and the embryo-forming rate raised by 9.1 times when backerossed with common wheat. The results indicated that long term preservation of the F1 hybrid-common wheat×Er. orientale was feasible and its characteristics of 2n=35 could be maintained. At the same time, some kinds of useful variation could be selected out.

Abstract:Much attention is being denoted to somatic cell hybridization in higher plants scince the first example was shown by Carlson et al. (1972) on hybridization of Nicotiana. In order to develop the somatic cell hybridization successfully, good conditions for inducing protoplast aggregation was looked for. Some substances which promote the aggregation have been noticed such as sodium nitrate (Power et al. 1979), gelatin (Kameya 1973) and polyethylene glycol (Kao and Michayluk 1974). Hartman et al. (1973) induced the aggregation with an immunological method. In the previous work, the writer found that gelatin or high molecular gelatin derivatives enhance the aggregation of protoplasts and subsequent their fusion (Kameya 1973). This finding led the writer to an assumption that water soluble and high molecular substances might promote the protoplast aggregation. The present studies were, therefore, carried out to investigate the effect of dextran sulfate, which is water soluble and high molecules on protoplast aggregation. Next problem comes how to mark cells, and how to make somatic cell fusion and hybrid plants. Here the writer would like to present one case of hybrid plant production through fusion between haploid cells.

Plant protoplasts: status and biotechnological perspectives

Abstract:Plant protoplasts ("naked" cells) provide a unique single cell system to underpin several aspects of modern biotechnology. Major advances in genomics, proteomics, and metabolomics have stimulated renewed interest in these osmotically fragile wall-less cells. Reliable procedures are available to isolate and culture protoplasts from a range of plants, including both monocotyledonous and dicotyledonous crops. Several parameters, particularly the source tissue, culture medium, and environmental factors, influence the ability of protoplasts and protoplast-derived cells to express their totipotency and to develop into fertile plants. Importantly, novel approaches to maximise the efficiency of protoplast-to-plant systems include techniques already well established for animal and microbial cells, such as electrostimulation and exposure of protoplasts to surfactants and respiratory gas carriers, especially perfluorochemicals and hemoglobin. However, despite at least four decades of concerted effort and technology transfer between laboratories worldwide, many species still remain recalcitrant in culture. Nevertheless, isolated protoplasts are unique to a range of experimental procedures. In the context of plant genetic manipulation, somatic hybridisation by protoplast fusion enables nuclear and cytoplasmic genomes to be combined, fully or partially, at the interspecific and intergeneric levels to circumvent naturally occurring sexual incompatibility barriers. Uptake of isolated DNA into protoplasts provides the basis for transient and stable nuclear transformation, and also organelle transformation to generate transplastomic plants. Isolated protoplasts are also exploited in numerous miscellaneous studies involving membrane function, cell structure, synthesis of pharmaceutical products, and toxicological assessments. This review focuses upon the most recent developments in protoplast-based technologies.
An Improved System to Establish Highly Embryogenic Haploid Cell and Protoplast Cultures from Pollen Calluses of Maize (zea Mays L.)

Abstract:Regenerable, embryogenic haploid cell suspensions were initiated and established from type II pollen calluses of two selected Chinese maize genotypes (No 592 Y and 592.A2 LY). The induction frequency of friable, embryogenic callus (type II) was highly dependent on three factors: genotype, medium, cold pretreatment, and on their interactions. Repeated callus and cell selection during the culture procedure led to stable haploid suspensions consisting of fine clusters each containing 20–50 cells. The selected cell lines were able to maintain their morphogenic ability during long-term subculture (2 years). Protoplasts were successfully isolated from subcultured, friable, embryogenic pollen calluses and cultured on N6BM and N6K media using a feeder layer, obtained from 2-day-old suspension culture. Healthy plants were regenerated from protoplast-derived calluses.

Abstract:The wild cotton species of G.tomentosum has some outstanding characters such as the higher resistance to pests and tolerance to drought.However,the upland cotton germplasm with the trait has not been obtained through sexual hybridization between G.hirsutum and G.tomentosum due to the distant relationship between the wild species and upland cotton.Protoplast fu-sion technique which depends on the success in the somatic culture of two different species can avoid sexual incompatibility.The callus inducing and somatic embryo germination of G.tomentosum were studied through the regulation of culture conditions such as plant growth regulators(PGRs),PEG,CuSO4and AgNO3.The results indicated that under the same condition the frequency of somatic embryo germination and the plant regeneration ability of G.tomentosum were lower and the plant regeneration was more difficult than Coker 201.But through the regulation of culture conditions,we can get many embryoids and few of them converted to plantlets.The callus of G.tomentosum induced in the medium with the hormone combination of 0.1 mg.L-1 KT + 0.1 mg.L-1 2,4-D held good for cell differentiation,and it was better for proliferation of the embryogenic callus in the medium with 0.5 mg.L-1 IBA + 0.2 mg.L-1 KT.The medium supplied with PEG was not helpful for initiation of somatic embryos.But the medium of MSB5 with 3.5 mg.L-1 CuSO4+0.5 mg.L-1 IBA + 0.2 mg.L-1 KT,or 3 mg.L-1 AgNO3+0.5 mg.L-1 IBA+ 0.2 mg.L-1 KT were good for the proliferation of embryogenic callus and germination of somatic embryo.This report provides an useful technique for somatic culture of the wild cotton species,and also ensures the protoplast fusion feasibly between these wild cotton species and other cotton species to centralize several good traits into a new germplasm.However,it is needed to study further about the various culture factors and conditions for building a quick and high efficient regeneration technique of G.tomentosum.

Abstract:The Brassica oleracea L. species embrace important horticultural crops, such as broccoli, cauliflower, and cabbage, which are highly valued for their beneficial nutritional effects. However, the complexity of flower emasculation in these species has forced breeders to adopt biotechnological approaches such as somatic hybridization to ease hybrid seed production. Protoplasts entail a versatile tool in plant biotechnology, supporting breeding strategies that involve genome editing and hybridization. This review discusses the use of somatic hybridization in B. oleracea L. as a biotechnological method for developing fusion products with desirable agronomic traits, particularly cytoplasmic male sterile (CMS) condition. These CMS lines are critical for implementing a cost-effective, efficient, and reliable system for producing F1 hybrids. We present recent studies on CMS systems in B. oleracea L. crops, providing an overview of established models that explain the mechanisms of CMS and fertility restoration. Additionally, we emphasize key insights gained from protoplast fusion applied to B. oleracea L. breeding. Key steps including pre-treatments of donor plants, the main tissues used as sources of parental protoplasts, methods for obtaining somatic hybrids and cybrids, and the importance of establishing a reliable plant regeneration method are discussed. Finally, the review explores the incorporation of genome editing technologies, such as CRISPR-Cas9, to introduce multiple agronomic traits in Brassica species. This combination of advanced biotechnological tools holds significant promise for enhancing B. oleracea breeding programs in the actual climate change context.

Obstacles and Perspectives in Protoplast Research

Abstract:A list of obstacles in using protoplasts in basic studies of plant physiology and genetics will be long if we expected the isolated plant protoplast to behave as a unicellular micro-organism in culture. A protoplast is a component of a very complex developmental system and in addition it is, in general, cultured under conditions which induce heavy stress reactions. It is today known that different types of stress can affect gene regulation that genes are turned on or off. Therefore, it is not surprising that there are difficulties in inducing normal growth with consecutive divisions, new wall formation, and normal hormone metabolism in cultured protoplasts. It has also been reports of increased RNase activity and lower amount of mRNA available in protoplasts to mention a few problems. Some of these problems can be partly overcome by pretreatment of the doner plant or by using other tissues like meristems and stam segments or by modifying the procedure of protoplast isolation and culture.

Abstract:The progress in the development of the technologies of plant tissue and cell culture over the past four decades has been remarkable. This article covers my personal reflections on the various topics and is based on my involvement in the field during that period. There are three fundamental technologies which constitute most of what is referred to as plant in vitro technologies or tissue culture. The origin and some of the key persons involved in the development of each of these procedures will be discussed. The technology that is most common is growing plant tissue on gel-solidified nutrient media. That technology is being used in the most vital procedures, namely the regeneration of plants from cultured cells. The culture of plant cells in liquid suspension was developed very shortly after that, and has become a very effective technology for plant regeneration by somatic embryogenesis. The method of meristem culture arose out of a need for developing plants that were virus-free. In many species the technique is now being used to produce virus-free crop plants. Another important technology is the culture of anthers and microspores for producing haploid and homozygous plants. Included with plant tissue culture is the development of the plant protoplast and cell fusion technologies for the production of new plant hybrids. The final aspect of the development concerns the integration of tissue culture with molecular genetics, which has developed into the rapidly expanding field of biotechnology.

An Introduction to Plant Cell, Tissue, and Organ Culture: Current Status and Perspectives

Abstract:Plant cell, tissue, and organ cultures (PCTOC) have been used as experimental systems in basic research, allowing gene function demonstration through gene overexpression or repression and investigating the processes involved in embryogenesis and organogenesis or those related to the potential production of secondary metabolites, among others. On the other hand, PCTOC has also been applied at the commercial level for the vegetative multiplication (micropropagation) of diverse plant species, mainly ornamentals but also horticultural crops such as potato or fruit and tree species, and to produce high-quality disease-free plants. Moreover, PCTOC protocols are important auxiliary systems in crop breeding crops to generate pure lines (homozygous) to produce hybrids for the obtention of polyploid plants with higher yields or better performance. PCTOC has been utilized to preserve and conserve the germplasm of different crops or threatened species. Plant genetic improvement through genetic engineering and genome editing has been only possible thanks to the establishment of efficient in vitro plant regeneration protocols. Different companies currently focus on commercializing plant secondary metabolites with interesting biological activities using in vitro PCTOC. The impact of omics on PCTOC is discussed.

Abstract:With the development and operation of industrial-scale plant cell culture processes, cell culture technology may be said to have “come of age.” One or two processes operating commercially do not, however, “make an industry.” Much yet still remains to be done if plant cell culture technology is to have a major impact upon process technology in the chemical industry. With this in mind, and with the wide range of papers presented at the Giessen Symposium, it was felt appropriate to review developments in plant cell culture and to provide a perspective of the challenges to be faced in the years ahead. To this end a small group of people were asked to put on paper their ideas, concerns and visions relating to the future of plant cell culture. The following text is an edited version of their thoughts.
Plant Tissue Culture Techniques for Breeding

Abstract:Several tissue culture techniques can be used to generate genetic variation in plants. These in vitro procedures have been applied successfully in many species and have been used to create plants that are disease and insect resistant and tolerant to water stress and demonstrate enhanced growth and improved yields and much more. Traditional plant breeding is enhanced with techniques such as embryo culture to rescue recalcitrant hybrids. Technologies as protoplast fusion and somaclonal variation are used to exploit existing genetic diversity or to produce new genetic variability. Once new plants are developed, micropropagation can be used to efficiently clonally propagate large numbers for the commercial market. Genetic engineering would not be possible without the involvement of in vitro plant tissue culture regeneration procedures.

Abstract:Advances in cell fusion technology have propelled breeding into the realm of somatic hybridization, enabling the transfer of genetic material independent of sexual reproduction. This has facilitated genome recombination both within and between species. Despite its use in plant breeding for over fifty years, somatic hybridization has been limited by cumbersome procedures, such as protoplast isolation, hybridized-cell selection and cultivation, and regeneration, particularly in woody perennial species that are difficult to regenerate. This review summarizes the development of somatic hybridization, explores the challenges and solutions associated with cell fusion technology in woody perennials, and outlines the process of protoplast regeneration. Recent advancements in genome editing and plant cell regeneration present new opportunities for applying somatic hybridization in breeding. We offer a perspective on integrating these emerging technologies to enhance somatic hybridization in woody perennial plants.

Abstract:The growing demand for native planting material in ecological restoration and rehabilitation for agro-silvo-pastoral ecosystems has resulted in a major global industry in their sourcing, multiplication, and sale. Plant tissue culture is used for producing high-quality, disease-free, and true-to-type plants at a fast rate. Micropropagation can help to meet the increasing demand for planting material and afforestation programs. However, in vitro plant propagation is an expensive technique compared to conventional methods using suckers, seeds, and cuttings. Therefore, adopting measures to lower production costs without compromising plant quality is essential. This can be achieved by improving the culture media composition. Incorporating organic growth additives can stimulate tissue growth and increase the number of shoots, leaves, and roots in culture media. Organic growth supplementation speeds up the formation and development of cultures and yields vigorous plants. Plant regeneration from meristems (shoot tips and axillary buds) is a reliable way to produce true-to-type plants compared with callus and somatic embryogenesis regeneration, but in vitro culture environments can be mutagenic. Therefore, detecting somaclonal variations at an early stage of development is considered crucial in propagating plants. The genetic stability of in vitro regenerated plants needs to be ascertained by using DNA-based molecular markers. This review aims to provide up-to-date research progress on incorporating organic growth additives to enhance in vitro tissue culture protocols and to emphasize the importance of using PCR-based molecular markers such as RAPD, ISSR, SSR, and SCoT. The review was assessed based on the peer-reviewed works published in scientific databases including Science Direct, Scopus, Springer, JSTOR, onlinelibrary, and Google Scholar.

Abstract:Major advances in the achievement of complete protoplast systems for deciduous angiosperm trees and shrubs have occurred in the last 3 years. At least 12 different species of woody trees and shrubs representing 6 different families have now been regenerated to plants from protoplasts. More importantly, two-thirds of these examples utilized non-seedling source plants, thus allowing the application of protoplast-based techniques to mature, highly selected genotypes. In general, two strategies for the development of complete protoplast systems were utilized: (1) the isolation of leaf protoplasts from in vitro shoot cultures with subsequent shoot regeneration via organogenesis, often from nodular callus; (2) isolation of protoplasts from embryonic tissue sources (cotyledons or embryogenic cultures) with subsequent shoot regeneration via somatic embryogenesis. The actual utilization of these protoplast techniques for genetic improvement or other research objectives has not yet occurred.

Abstract:Methods are described for the enzymatic release of protoplasts from leaves of Petunia hybrida and for the utilization of protoplasts in studies in plant developmental biology. As a result of spontaneous fusion during cell wall degradation of leaf material, fresh preparations can contain a high proportion of multinucleate protoplasts. This level can be dramatically reduced by a gradual plasmolysis of the material prior to enzyme incubation.Leaf protoplasts maintained in liquid media are seen to undergo cell wall synthesis, “budding,” and limited regenerated cell division sometimes associated with anthocyanin production. Under such conditions, multinucleate cells are formed as a result of mitosis without cytokinesis.Protoplasts, plated out in a fully defined medium, undergo cell wall synthesis followed by sustained progeny cell division with eventual cell colony production. Cell colonies, derived from individual mesophyll protoplasts, grow rapidly upon subculture, to produce callus capable of shoot differentiation and ultimately whole plant formation. Protoplasts isolated from varieties of P. hybrida were found to differ in their cultural requirements.
