Abstract:Bioelectronic devices must be fast and sensitive to interact with the rapid, low-amplitude signals generated by neural tissue. They should also be biocompatible and soft, and should exhibit long-term stability in physiologic environments. Here, we develop an enhancement-mode, internal ion-gated organic electrochemical transistor (e-IGT) based on a reversible redox reaction and hydrated ion reservoirs within the conducting polymer channel, which enable long-term stable operation and shortened ion transit time. E-IGT transient responses depend on hole rather than ion mobility, and combine with high transconductance to result in a gain–bandwidth product that is several orders of magnitude above that of other ion-based transistors. We used these transistors to acquire a wide range of electrophysiological signals, including in vivo recording of neural action potentials, and to create soft, biocompatible, long-term implantable neural processing units for the real-time detection of epileptic discharges. E-IGTs offer a safe, reliable and high-performance building block for chronically implanted bioelectronics, with a spatiotemporal resolution at the scale of individual neurons.

Abstract:Organic electrochemical transistors (OECTs) and OECT-based circuitry offer great potential in bioelectronics, wearable electronics and artificial neuromorphic electronics because of their exceptionally low driving voltages (<1 V), low power consumption (<1 µW), high transconductances (>10 mS) and biocompatibility 1 – 5 . However, the successful realization of critical complementary logic OECTs is currently limited by temporal and/or operational instability, slow redox processes and/or switching, incompatibility with high-density monolithic integration and inferior n-type OECT performance 6 – 8 . Here we demonstrate p- and n-type vertical OECTs with balanced and ultra-high performance by blending redox-active semiconducting polymers with a redox-inactive photocurable and/or photopatternable polymer to form an ion-permeable semiconducting channel, implemented in a simple, scalable vertical architecture that has a dense, impermeable top contact. Footprint current densities exceeding 1 kA cm −2 at less than ±0.7 V, transconductances of 0.2–0.4 S, short transient times of less than 1 ms and ultra-stable switching (>50,000 cycles) are achieved in, to our knowledge, the first vertically stacked complementary vertical OECT logic circuits. This architecture opens many possibilities for fundamental studies of organic semiconductor redox chemistry and physics in nanoscopically confined spaces, without macroscopic electrolyte contact, as well as wearable and implantable device applications.

Abstract:Conducting polymers is not the only way, inorganic oxides can make electrochemical transistors too. It is shown that ultrathin, flexible, ITO electrolyte‐gated transistors are designed for bioelectronics. These transistors demonstrate high transconductance, excellent on/off ratios, and mechanical flexibility. Via surface passivation strategies are used to enhance the electrochemical stability of ITO, making these devices promising candidates for future in vivo and in vitro bioelectronic applications. Electrolyte‐gated field effect transistors and electrochemical transistors have emerged as powerful components for bioelectronic sensors and biopotential recording devices. A set of parameters must be considered when developing devices to amplify weak electrophysiological signals. These include maximum transconductance values, cut‐off frequencies, and large on/off current ratios. Organic polymer‐based devices have recently dominated the field, especially when considering flexibility as a key factor. Oxide semiconductors may also offer these features, as well as advantages like higher mobility. Herein, flexible, ultrathin, indium tin oxide (ITO) electrolyte‐gated transistors are reported. These accumulation‐mode devices combine n‐type operation with μe = 9.5 cm2 Vs−1, high transconductance (gm = 44 mS), and on/off ratios (105) as well as optically transparent layouts. While oxides are normally considered brittle, mechanically flexible ITO layers are obtained by room temperature deposition of amorphous layers onto parylene C. This process results in low strain, producing devices that survive bending. ITO electrochemically degrades, however, with cycling. To overcome this, the surface is passivated with high dielectric constant inert capping layers of Ta2O5 or Ta2O5/AlN. This greatly improves stability while preserving low gate voltages. Based on their overall performance, ITO‐based EGFETs are promising for bioelectronics.

Abstract:By tuning annealing temperature, internal ion‐gated organic electrochemical transistors (IGT) can be optimized to demonstrate high transconductance and high speed. Re‐arrangement of polymer chains into structures favorable for hole conduction is attributed to enhanced crystallization and partial water removal of ion‐embedded conducting polymer. The as‐optimized IGT demonstrates its potential in high‐frequency applications, i.e. 50 kHz fluid assessment for dialysis management. Internal ion‐gated organic electrochemical transistor (IGT) demonstrates volume‐dependent transconductance with the unprecedented advantages of high speed and self‐(de)doping capability among ion‐based transistors. The novel characteristics have albeit rendered IGT a promising platform for integrated bioelectronics, its potential in high‐frequency applications has yet been fully harnessed. Moreover, a study from a material's point of view is especially needed for this recently emerged platform as the necessity of maintaining the internal ion reservoir has posed difficulties in processing hydrated poly(3,4‐ethylenedioxythiophene) doped with poly(styrene sulphonate) (PEDOT:PSS) with electronically favorable morphologies. Herein, a comprehensive investigation of the structural and functional properties of ion‐embedded PEDOT:PSS modified by different annealing temperatures is performed and correlated to the IGT performance. A short‐time high‐temperature annealing treatment is found effective in facilitating the formation of compact microstructures without significantly influencing film hydration. The structural improvement enhances the film's conductivity and hole mobility, with the corresponding IGTs exhibiting higher gain, higher conductance, and high cut‐off frequency consistently in a batch. This study also successfully demonstrates the first use of electrochemical transistors like IGTs in high‐frequency applications through proof‐of‐concept experiments simulating fluid estimation in 50 kHz bioimpedance analysis. This work contributes to the development of high‐performance IGTs for extensive biological applications.

Ion Exchange Gels Allow Organic Electrochemical Transistor Operation with Hydrophobic Polymers in Aqueous Solution

Abstract:Conjugated-polymer-based organic electrochemical transistors (OECTs) are being studied for applications ranging from biochemical sensing to neural interfaces. While new polymers that interface digital electronics with the aqueous chemistry of life are being developed, the majority of high-performance organic transistor materials are poor at transporting biologically relevant ions. Here, the operating mode of an organic transistor is changed from that of an electrolyte-gated organic field-effect transistor (EGOFET) to that of an OECT by incorporating an ion exchange gel between the active layer and the aqueous electrolyte. This device works by taking up biologically relevant ions from solution and injecting more hydrophobic ions into the active layer. Using poly[2,5-bis(3-tetradecylthiophen-2-yl) thieno[3,2-b]thiophene] as the active layer and a blend of an ionic liquid, 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide, and poly(vinylidene fluoride-co-hexafluoropropylene) as the ion exchange gel, four orders of magnitude improvement in device transconductance and a 100-fold increase in kinetics are demonstrated. The ability of the ion-exchange-gel OECT to record biological signals by measuring the action potentials of a Venus flytrap is demonstrated. These results show the possibility of using interface engineering to open up a wider palette of organic semiconductors as OECTs that can be gated by aqueous solutions.

Abstract:Electrolyte-gated transistors (EGTs), capable of transducing biological and biochemical inputs into amplified electronic signals and stably operating in aqueous environments, have emerged as fundamental building blocks in bioelectronics. In this Primer, the different EGT architectures are described with the fundamental mechanisms underpinning their functional operation, providing insight into key experiments including necessary data analysis and validation. Several organic and inorganic materials used in the EGT structures and the different fabrication approaches for an optimal experimental design are presented and compared. The functional bio-layers and/or biosystems integrated into or interfaced to EGTs, including self-organization and self-assembly strategies, are reviewed. Relevant and promising applications are discussed, including two-dimensional and three-dimensional cell monitoring, ultra-sensitive biosensors, electrophysiology, synaptic and neuromorphic bio-interfaces, prosthetics and robotics. Advantages, limitations and possible optimizations are also surveyed. Finally, current issues and future directions for further developments and applications are discussed.

Abstract:Conjugated polymer-based organic electrochemical transistors (OECTs) are being studied for applications ranging from biochemical sensing to neural interfaces. While new conjugated polymers are being developed that can interface digital electronics with the aqueous chemistry of life, the vast majority of high-performance, high-mobility organic transistor materials developed over the past decades are extremely poor at taking up biologically-relevant ions. Here we incorporate an ion exchange gel into an OECT, demonstrating that this structure is capable of taking up biologically-relevant ions from solution and injecting larger, more hydrophobic ions into the underlying polymer semiconductor active layer in multiple hydrophobic conjugated polymers. Using poly[2,5-bis(3-tetradecylthiophen-2-yl) thieno[3,2-b]thiophene] (PBTTT) as a model semiconductor active layer and a blend of the ionic liquid 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide (BMIM TFSI) and poly(vinylidene fluoride-co-hexafluoropropylene) (PVDF-HFP) as the ion exchange gel, we demonstrate more than a four order of magnitude improvement in OECT device transconductance and a one hundred-fold increase in ion injection kinetics. We demonstrate the ability of the ion exchange gel OECT to record biological signals by measuring the action potentials of a Venus flytrap plant. These results show the possibility of using interface engineering to open up a wider palette of organic semiconductor materials as OECTs that can be gated by aqueous solutions.

Abstract:Organic electrochemical transistors (OECTs) are presently a focus of intense research and hold great potential in expanding the horizons of the bioelectronics industry. The notable characteristics of OECTs, including their electrolyte-gating, which offers intimate interfacing with biological environments, and aqueous stability, make them particularly suitable to be operated within a living organism (in vivo). Unlike the existing in vivo bioelectronic devices, mostly based on rigid metal electrodes, OECTs form a soft mechanical contact with the biological milieu and ensure a high signal-to-noise ratio because of their powerful amplification capability. Such features make OECTs particularly desirable for a wide range of in vivo applications, including electrophysiological recordings, neuron stimulation, and neurotransmitter detection, and regulation of plant processes in vivo. In this review, a systematic compilation of the in vivo applications is presented that are addressed by the OECT technology. First, the operating mechanisms, and the device design and materials design principles of OECTs are examined, and then multiple examples are provided from the literature while identifying the unique device properties that enable the application progress. Finally, one critically looks at the future of the OECT technology for in vivo bioelectronic applications.

Anisotropic Ion Conducting Particulate Composites for Bioelectronics

Abstract:Acquisition, processing, and manipulation of biological signals require transistor circuits capable of ion to electron conversion. However, use of this class of transistors in integrated sensors or circuits is limited due to difficulty in patterning biocompatible electrolytes for independent operation of transistors. It is hypothesized that it would be possible to eliminate the need for electrolyte patterning by enabling directional ion conduction as a property of the material serving as electrolyte. Here, the anisotropic ion conductor (AIC) is developed as a soft, biocompatible composite material comprised of ion-conducting particles and an insulating polymer. AIC displays strongly anisotropic ion conduction with vertical conduction comparable to isotropic electrolytes over extended time periods. AIC allows effective hydration of conducting polymers to establish volumetric capacitance, which is critical for the operation of electrochemical transistors. AIC enables dense patterning of transistors with minimal leakage using simple solution-based deposition techniques. Lastly, AIC can be utilized as a dry, anisotropic interface with human skin that is capable of non-invasive acquisition of individual motor action potentials. The properties of AIC position it to enable implementation of a wide range of large-scale organic bioelectronics and enhance their translation to human health applications.
Dual-liquid-gated electrochemical transistor and its neuromorphic behaviors

Abstract:Organic electrochemical transistors (OECTs) are attracting great interest in the field of bioelectronics due to their low operating voltage, flexibility, and biocompatibility. Tunability of the static and transient characteristics renders OECTs with flexible electrical responses and versatile functionalities. However, existing tuning methods are known by changing the structure or composition of OECTs, which are empirical due to the lack of accurate structure-function relationships. Here, we report a post-fabrication and facile tuning method by using a dual-liquid-gate configuration. Based on this, critical parameters of OECT, e.g., threshold voltage (VTH), gate bias for the peak transconductance (VG(g*m)), electric hysteresis (Vhys), minimum of the subthreshold swing (SS*), and response time (τ), can be readily tuned over a range of 0.52 V, 0.48 V, 0.20 V, 0.38 V/decade and 7.2 ms, respectively. We have also developed corresponding mathematical analyses based on the dual-liquid-gating process. Detailed studies on the transient electrical properties demonstrate that auxiliary-gate biases influence the electrochemical doping/de-doping state of the semiconducting channel during the main-gate bias sweeping. Furthermore, typical neuromorphic behaviors of paired-pulse depression and decay time were successfully controlled by varying the auxiliary-gate bias. The proposed dual-liquid-gating is ready for precise engineering on OECT, which is beneficial as an effective tool for conducting an in-depth theoretical study on OECT, constructing multifunctional sensors, and developing more plasticizable neuromorphic devices.

Abstract:This work presents a scalable photolithographic process for large‐scale and simple microfabrication of OECTs. The method uses a two‐layer photoresist with controlled cross‐linking for precise polymer channel patterning and electrodes encapsulation. The proposed process yields reproducible and high‐performance OECTs, enabling their integration with polarizable gates and ion detection with sensitivity normalized to supply voltage of up to 6550 mV V−1 dec−1, opening opportunities for advanced bioelectronics and in situ neuromorphic biosensing. In bioelectronics, organic electrochemical transistors (OECTs) are pivotal in bridging electronic devices and biological systems, especially in sensing, neuromorphic interfacing, and biological monitoring. Current OECT fabrication methods face challenges due to conductive polymers incompatibility with photoresist solvents and the complexity of multi‐step parylene‐C coatings. This work introduces a scalable photolithographic fabrication process for high‐performance OECTs using the prototypical conductive polymer poly(3,4‐ethylenedioxythiophene) doped with poly(styrene sulfonate) (PEDOT:PSS). The method employs a two‐layer photoresist approach with controlled cross‐linking, incorporating (3‐glycidyloxypropyl)trimethoxysilane to pattern polymeric channel and encapsulate electrodes. This process yields high‐performance OECTs with highly reproducible characteristics, typical ON/OFF current modulation of 5 103, and transconductance normalized to channel thickness > 200 S cm−1. To avoid cytotoxic Ag/AgCl pellets, the impact of scaling polarizable gates is analyzed on device performance, including bare Au, protein‐functionalized, and PEDOT:PSS gates. The analysis provides design rules to tailor OECT performance for diverse applications. The effectiveness of the approach is demonstrated by integrating OECTs with polarizable gates in current‐driven circuit configuration, allowing ion detection at a supply voltage as low as 0.4 V with a sensitivity of up to 2620 mV dec−1, the highest ever reported. These advancements open opportunities for next‐generation integrated bioelectronics and neuromorphic biosensing.

Abstract:Biocompatible field-effect-transistor-based biosensors have drawn attention for the development of next-generation human-friendly electronics. High-performance electronic devices must achieve low-voltage operation, long-term operational stability, and biocompatibility. Herein, we propose an electrolyte-gated thin-film transistor made of large-area solution-processed indium-gallium-zinc oxide (IGZO) semiconductors capable of directly interacting with live cells at physiological conditions. The fabricated transistors exhibit good electrical performance operating under sub-0.5 V conditions with high on-/off-current ratios (>107) and transconductance (>1.0 mS) over an extended operational lifetime. Furthermore, we verified the biocompatibility of the IGZO surface to various types of mammalian cells in terms of cell viability, proliferation, morphology, and drug responsiveness. Finally, the prolonged stable operation of electrolyte-gated transistor devices directly integrated with live cells provides the proof-of-concept for solution-processed metal oxide material-based direct cellular interfaces.
In-Liquido Computation with Electrochemical Transistors and Mixed Conductors for Intelligent Bioelectronics

Abstract:Next-generation implantable computational devices require long-term stable electronic components capable of operating in, and interacting with, electrolytic surroundings without being damaged. Organic electrochemical transistors (OECTs) emerged as fitting candidates. However, while single devices feature impressive figures of merit, integrated circuits (ICs) immersed in a common electrolytes are hard to realize using electrochemical transistors, and there is no clear path forward for optimal top-down circuit design and high-density integration. The simple observation that two OECTs immersed in the same electrolytic medium will inevitably interact hampers their implementation in complex circuitry. The electrolyte's ionic conductivity connects all the devices in the liquid, producing unwanted and often unforeseeable dynamics. Minimizing or harnessing this crosstalk has been the focus of very recent studies. In this Perspective, we discuss the main challenges, trends, and opportunities for realizing OECT-based circuitry in a liquid environment that could circumnavigate the hard limits of engineering and human physiology. We analyze the most successful approaches in autonomous bioelectronics and information processing. Elaborating on the strategies to circumvent and harness device crosstalk proves that platforms capable of complex computation and even machine learning can be realized in-liquido using mixed ionic-electronic conductors.

Empowering Bioelectronics with Supramolecular Nanoarchitectonics: PEDOT-Based Organic Electrochemical Transistors with Tunable Electronic Properties

Abstract:Organic bioelectronics has the potential to unlock the utmost innovation in medicine and healthcare through the combination of biological and digital realms. Particularly, organic electrochemical transistors (OECTs) are a promising class of bioelectronics transducer. Nevertheless, a fabrication strategy is needed to lower the access barrier to the OECTs, thereby expediting product development and innovation. In this work, we present a supramolecular approach with simple equipment to prepare conductive films based on poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate), PEDOT:PSS, integrated with cationic molecular blocks for OECTs manufacturing. By employing the layer-by-layer (LbL) self-assembly technique, we facilely prepared transistor channels of PEDOT:PSS integrated with either the surfactant cetyltrimethylammonium bromide, CTAB, or the polyelectrolyte poly(diallyldimethylammonium chloride), PDADMAC, with nanometric precision. Both cationic blocks feature positively charged quaternary amines but possess different mesogenic and surfactant features. The PEDOT:PSS/CTAB system yields an electrical conductivity of 275 S m–1, which is 4 orders of magnitude higher than those integrated with PDADMAC (0.0531 S m–1). This enhancement is attributed to CTAB integration, which boosts the PEDOT:PSS charge transport, while PDADMAC diminishes it. The electronic performance indicators of OECTs (current at the on-state, I max, threshold potential, V TH, transconductance, g m) are easily tuned by adjusting the thickness of the transistor channel film. The cycling stability of the transistor channel is 8-fold enhanced by coating it with a protective layer using nonelectroactive polymers. These OECTs exhibit a g m of 2.21 mS, a μC* product (μ is the hole mobility, and C* is the capacitance per unit of volume) of 0.23 F cm–1 V–1 s–1, and on- and off-switching times, τ, of 24.2 and 12.3 ms, respectively. Their performance was comparable to or better than OECTs with channels prepared using techniques based on equipment of higher complexity and cost. Finally, we demonstrate the utility of these facilely prepared OECTs in biosensing the neurotransmitter dopamine with an exceptional sensitivity of 279 mV/decade and good operation range (1–300 μM) and reversibility.

Abstract:Biocompatible, biodegradable, and solid‐state electrolyte‐based organic transistors are demonstrated. As the electrolyte is composed of all edible materials, which are levan polysaccharide and choline‐based ionic liquid, the organic transistor fabricated on the electrolyte can be biocompatible and biodegrable. Compared to the other ion gel based electrolytes, it has superior electrical and mechanical properties, large specific capacitance (≈40 µF cm−2), non‐volatility, flexibility, and high transparency. Thus, it shows mechanical reliability by maintaining electrical performances under up to 1.11% of effective bending strain, 5% of stretching, and have low operation voltage range when it is utilized in organic transistors. Moreover, the biodegradable electrolyte‐based organic transistors can be applied to bio‐integrated devices, such as electrocardiogram (ECG) recordings on human skin and the heart of a rat. The measured ECG signals from the transistors, compared to signals from electrode‐based sensors, has a superior signal‐to‐noise ratio. The biocompatible and biodegradable materials and devices can contribute to the development of many bioelectronics.

Abstract:Realization of biological synapses using electronic devices is regarded as the basic building blocks for neuromorphic engineering and artificial neural network. With the advantages of biocompatibility, low cost, flexibility, and compatible with printing and roll-to-roll processes, the artificial synapse based on organic transistor is of great interest. In this paper, the artificial synapse simulation by ion-gel gated organic field-effect transistors (FETs) with poly(3-hexylthiophene) (P3HT) active channel is demonstrated. Key features of the synaptic behaviors, such as paired-pulse facilitation (PPF), short-term plasticity (STP), self-tuning, the spike logic operation, spatiotemporal dentritic integration, and modulation are successfully mimicked. Furthermore, the interface doping processes of electrolyte ions between the active P3HT layer and ion gels is comprehensively studied for confirming the operating processes underlying the conductivity and excitatory postsynaptic current (EPSC) variations in the organic synaptic devices. This study represents an important step toward building future artificial neuromorphic systems with newly emerged ion gel gated organic synaptic devices.
