Abstract:4-Aryl-3,4-dihydrocoumarins(neoflavones) represent a minor class of natural compounds which based on the C6—C3—C6 skeleton with an characteristic aryl group in the 4 position.Structurally,4-aryl-3,4-dihydrocoumarins can also be regarded as the hybrids of isoflavones and coumarins,thus being expected to possess corresponding bioactivity.In this paper,ten 4-aryl-3,4-dihydrocoumarins were synthesized by two steps.The first condensation of substituted benzaldehydes and malonic acids in the presence of pyridine and piperidine at 70-90 ℃ gave the expected cinnamic acids in good yields.Then condensation of substituted cinnamic acids with appropriate phenols in the presence of BF3-Et2O and POCl3 gave the target molecule at room temperature.Their structures were confirmed by EI-MS,IR and 1H-NMR.The antioxidant and antitumor activity of these compounds were evaluated by DPPH and MTT assay,respectively.Some of 4-aryl-3,4-dihydrocoumarin derivatives showed strong scavenging activities against 1,1-diphenyl-2-picrylhydrazyl radical:(±)-7,8-dihydroxy-4-(3-hydroxy-4-methoxyphenyl)-3,4-dihydrocoumarin(3c) EC50=(2.69±0.09) μmol · L-1,(±)-7,8-dihydroxy-4-(4-methoxyphenyl)-3,4-dihydrocoumarin(3g) EC50=(2.32±0.10) μmol · L-1 and weak antitumor in vitro activity against BGC-823 cell line[(±)-7,8-dihydroxy-4-(3-hydroxy-4-methoxyphenyl)-3,4-dihydrocoumarin(3c) IC50=(136±21) μmol · L-1].The results showed that the 4-arylcoumarins which possessing the catechol moiety may be a beneficial scaffold for therapeutic purpose.

Abstract:A new natural coumarin, angepubebisin (1), and a new furan, angepubefurin (2), together with the five known compounds, umbelliferone, angelol B (3), ulopterol (4), peucedanol (5), and scopoletin, were isolated from the roots of Angelica pubescens Maxim. f. biserrata Shan et Yuan. The structures of angepubebisin (1) and known compounds were determined by spectroscopic methods, including IR, UV, EI-MS, HR-FTICR-MS, 1D-, and 2D-NMR spectral analyses, and angepubefurin (2) was determined by HR-FTICR-MS and X-ray diffraction analyses.

Abstract:An efficient, oxidant and metal-free synthesis of polycyclic heteroaromatic coumarins was developed. H-Furo[2',3':3,4]naphtho[2,1-c]chromen-4-one (2a-2f), 1H-benzofuro[2',3':3,4]naphtho[2,1-c]chromen-1-one (2g-2j), and 4H-thieno[2',3':3,4]naphtho[2,1-c]chromen-4-one (2k-2s) derivatives were obtained by the irradiation of 4-phenyl-3-heteroarylcoumarin in EtOH-H2O (9:1, v/v) using a high-pressure Hg lamp as the light source, at room temperature and under an Ar atmosphere. Owing to the expansion of the π-conjugation system, 2a-2s showed strong fluorescence emissions in ethanol solution ( ΦF = 0.40-0.83).

Abstract:A method for the synthesis of 4-methyl-2-oxo-9-phenyl-2H-furo[2,3-h]chromene-8-carboxylic acid amides combining in the structure the angelicin core with heterocyclic fragments at the amide nitrogen atom was developed. The cytotoxic activity of the obtained amides against the MCF7 (breast adenocarcinoma), A549 (non-small cell lung cancer), VA-13 (embryonic lung fibroblast cells) and HEK293T (embryonic kidney cells) cell lines was studied. In contrast to the previously described highly toxic angelicin derivatives with aryl- and hetarylcarbonyl substituents at position 8, the synthesized amides demonstrated in in vitro experiments in these cell lines the IC50abs values ranging within 2–100 µmol L−1. The cytotoxicity and selectivity of the action of this group of derivatives were found to be significantly affected by the substituent at position 5′ of the angelicin moiety.

Abstract:Seventeen new flavone derivatives substituted at the 4′-OH position were designed, synthesized and evaluated for their anticancer and antibacterial activities. Among them, compounds 3, 4, 6f, 6e, 6b, 6c and 6k demonstrated the most potent antiproliferative activities against a human erythroleukemia cell line (HEL) and a prostate cancer cell line (PC3). The results also showed that the IC50 value of compounds 3, 4, 6f, 6e, 6b, 6c and 6k were close to that of the anticancer drug cisplatin (DDP) and lower than that of apigenin. All of the derivatives did not present antibacterial activities. The structure–activity relationships evaluation showed that the configuration of methyl amino acid might affect their biological activities.

Abstract:3,4-Fused pyrrolocoumarins, synthetically prepared or naturally occurring, possess interesting biological properties. In this review, the synthetic strategies for the synthesis of the title compounds are presented along with their biological activities. Two routes are followed for that synthesis. In one, the pyrrole ring is formed from coumarin derivatives, such as aminocoumarins or other coumarins. In the other approach, the pyranone moiety is built from an existing pyrrole derivative or through the simultaneous formation of coumarin and pyrrole frameworks. The above syntheses are achieved via 1,3-dipolar cycloaddition reactions, Michael reaction, aza-Claisen rearrangement reactions, multi-component reactions (MCR), as well as metal-catalyzed reactions. Pyrrolocoumarins present cytotoxic, antifungal, antibacterial, α-glucosidase inhibition, antioxidant, lipoxygenase (LOX) inhibition, and fluorescent activities, as well as benzodiazepine receptor ability.

Abstract:Seven new dimeric furanocoumarins, dahuribiethrins A-G (1-7), were isolated from the roots of Angelica dahurica. Their structures were determined by chemical derivatization and extensive spectroscopic techniques, including (1)H NMR, (13)C NMR, HSQC, (1)H-(1)H COSY, HMBC, and NOESY experiments. Compounds 2, 3, 4, and 5 exhibited significant inhibition of nitric oxide production in the lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells with IC50 values in the range of 8.8-9.8 μM.

Abstract:Background: Condensation of 3-acetyl-2H-chromen-2-one (1) with 2‐(4-methoxybenzylidene) malononitrile (2) in ammonium acetate/acetic acid or absolute ethanol/piperidine affords pyridine (3) and chromen-2-one (4) derivatives, respectively. background: Synthesis of a novel series of chromen-3-yl-pyridine derivatives were elucidated using spectroscopic techniques and elemental analysis. antimicrobial activities of compounds were then evaluated in vitro. Compound (E)-3-(3-(4-methoxyphenyl)acryloyl)-2H-chromen-2-one (4), was more effective against all five microorganisms and nicotinonitrile derivative 3 and 1,8-naphthyridin-2-yl)-2H-chromen-2-one derivative 14, while compounds 9, 10a-c, 11, 12 and 13 were moderate activity for antimicrobial activities. Methods: In this study, the reaction of 3 with an electrophilic reagent, namely, formic acid, acetic anhydride and formamide afforded the pyridopyrimidinone and pyridine derivatives (6- 9). Also, treatment of 3 with different aromatic aldehydes, carbon disulfide, NaN3/NH4Cl, 2- (4-methoxybenzylidene)malononitrile and acetophenone afforded pyridopyrimidinone, carbamodithioic acid, tetrazol-5-yl-chromen-2-one, 2H-chromen-2-one, and pyridine derivatives (10-14), respectively. Results: This study synthesized coumarins with antimicrobial activity and verified the structure and purity of the synthesized compounds using spectral data. Conclusion: The new compounds were evaluated in vitro for their antimicrobial activity against gram-positive bacteria (Staphylococcus aureus, Methicillin-resistant Staphylococcus aureus) and gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa), in addition to fungus (Candida albicans). The investigated substances 3 and 14 presented good activity against MRSA, E. coli, P. aeruginosa and C. albicans, while compounds 9, 10a-c, 11, 12 and 13 exhibited good to moderate activity.

Abstract:Many organic and medicinal chemists are interested in the coumarin compounds due to their potential for use in pharmaceuticals and other industries. Coumarin compounds are a significant class of biologically active chemicals. Here, we outline the steps forproducing coumarin derivatives quickly and effectively from commercially accessible substrates via multicomponent condensation processes involving pyrone and benzene derivatives. These substances were examined for their potential anti-tumor effects, including their ability to block carbonic anhydrase, activate the cell apoptosis protein, and coumarin and its derivatives are the primary oral anticoagulants. Candida albicans, Aspergillus fumigatus, and Fusarium solani are three distinct fungus species that are resistant to the antifungal properties of synthetic coumarins. Long known are the coumarins' antibacterial due to its biological activities against some kinds of bacteria such as Zeitschrift für Naturforschung C, as well as their antifilarial, antiulcerogenic, anti-inflammatory, and antioxidant effects for its capacity to inhibit their growth activities. Coumarin has long been valued as a major raw element in the fragrance sector due to its distinct sweet aroma and stability. When mixed with organic essential oils like lavender, citrus, rosemary, and oak moss, it is used as an odour-enhancer to produce a long-lasting impact. It has been utilized as a bleaching or brightening agent in detergents when it revealed a remarkable brightening effect with excellent overall stability on fibers of synthetic origin. Materials such as liquid crystalline, organic/inorganic composites, as well as light harvesting, energy transferring compounds, and electro-optics materials are all employed as a dye in organic photo-redox catalysis and as potent photo-sensitizers reagent. These materials polymerize epoxy-silicones, under near-UV and visible light. Coumarin has also found useful as metal-free sensitizers for solar cells.

Abstract:Four novel coumarin derivatives (1-4) were synthesized via a one-pot three-component reaction by condensing 4-hydroxycoumarin, aromatic aldehydes and ethyl cyanoacetate in the presence of 4-(dimethylamino) pyridine (DMAP) as a highly efficient homogenous catalyst. The structures of the synthesized compounds have been deduced from IR, H-1 NMR, HRMS, and single crystal X-ray crystallography. The local anesthetic effect of these derivatives was assessed in comparison to lidocaine as a standard using a rabbit corneal and mouse tail anesthesia model. The results showed that compared with compounds 1-3, compound 4 with thiophene ring exhibited better anesthetic activity.
Novel heterocyclic coumarin annulates: synthesis and figuring their roles in biomedicine, bench-to-bedside investigation

Abstract:This study revealed the first synthesis of seven novel coumarins annulated with heterocycle composed of four heteroatoms, one sulfur, and three oxygen atoms in a linear pattern. This is an effort to find readily available coumarin frameworks with a broad variety of biological properties that are both adjustable and easy to get. The synthetic annulates' structural framework was confirmed by means of spectroscopic techniques, which included 1 H-NMR, 13 C-NMR, and FTIR. The synthesized annulates were investigated in vitro for their biomedical potential as antioxidative stress, anti-inflammatory, antidiabetic, anticancer, and antimicrobial agents. In addition, their biosafety toward nontumor cells and commensal bacterial strains was also assessed in vitro. Computer-aided programs were employed to explore the toxicity and pharmacokinetic profiles of the synthesized annulates. Based on the findings that were obtained, the authors stated the following main outcomes. There have been promising and far-reaching biological effects of the synthesized heterocyclic coumarin annulates. HC1 demonstrated strong anti-inflammatory potential through the lipoxygenase-dependent route. Moreover, HC1 exhibited significant antifungal efficacy, surpassing that of nystatin. HC2 held great promise as an antioxidative stress, anticancer, and biosafe candidate. HC3 exhibited a strong antibacterial potential against all tested aerobic bacterial strains, demonstrating a potency equivalent to that of ciprofloxacin. In addition, all of the synthesized annulates, especially HC3 , exhibited a noteworthy biosafety profile against the commensal bacterial strains. The strong inhibitory capabilities of HC6 and HC7 toward glucosidase and amylase indicate that they possess great promise as antidiabetic agents. Finally, the synthesized annulates showed favorable toxicity and oral bioavailability properties. It can be inferred that these annulates have the potential to be useful frameworks for developing new drugs with a broad spectrum of bioactivity in the coming years.

Abstract:Twelve angular oxazole-fused coumarin derivatives were designed, synthesised and characterised by 1H NMR, 13C NMR and HRMS. The structure of compound 4a was further confirmed by X-ray single-crystal diffraction. The bioassay experiment results indicated that compounds 4f and 4l have high antifungal activity on the mycelium growth of 4 plant disease fungi. Especially, compound 4l has a stronger antifungal activity compare to the commercial fungicide, Carbendazim. The herbicidal activity experiment showed that 4a and 4b can significantly inhibit the taproot and caulis development of Chenopodium album seedling and have better activities than the commercial herbicide, Acetochlor.

Abstract:As a novel analogue of furocoumarin,2H-4,8-dimethylfuro[2��, 3��: 5,6]naphtho[1, 2-b] pyran-2-one 6 was sythesized through Pechmann condensation, etherification, Claisen rear rangement, acetylation, bromine addition and cyclization of 1, 5-naphthalenediol. The sructures of 6 and its precusors were fully characterized by 1H NMR, MS, IR and elemental analysis. The DNA-intercalate activity of 6 and 7 was investigated in DMSO-Tris HCl system and their apparent Scatchard binding constants were calculated. It was found that 7 intercalate into DNA more efficiently than 6 due to the different substituents on the furan ring.
