Abstract:Current medical adhesives based on cyanoacrylates typically exhibit cellular toxicity. In contrast, fibrin adhesives are non-toxic but have poor adhesive properties. To overcome these drawbacks we designed a simple and scalable adhesive precursor inspired by marine mussel adhesion that functioned with strong adhesion in wet conditions and with low cytotoxicity. Dopamine, an-amine derivative of an amino acid abundantly present in mussel adhesive proteins, was co-polymerised with a tri-functional vinyl monomer, to form a hyperbranched poly(β-amino ester) polymer termed poly(dopamine-co-acrylate) (PDA). A variety of molecular weights and crosslinking methods were analysed using an ex vivo porcine skin model and an almost 4 fold increase in wet adhesion strength was observed compared to TISSEEL® fibrin sealant. With a fast curing time, degradable properties and low cytotoxicity, PDA is highly attractive for medical purposes and could have a broad impact on surgeries where surgical tissue adhesives, sealants, and haemostatic agents are used.

Abstract:In order to replace conventional sutures in wound closing applications, favorable hydrogels with strong wet tissue adhesion and biocompatibility have attracted considerable attention. Herein, inspired by mussel adhesive protein, a series of dopamine modified carboxymethyl cellulose (CMC-DA) hydrogels were prepared in situ using enzymatic crosslinking in the presence of horseradish peroxidase (HRP) and H2O2. The biomimetic CMC-DA hydrogel exhibited about 6-fold enhanced wet tissue adhesion strength (28.5 kPa) over the commercial fibrin glue. In addition, the gelation time, swelling ratio and rheological property of the hydrogel can be simply controlled by changing the concentrations of HRP, H2O2, and CMC-DA polymer. The gels also exhibited good biodegradation and biocompatibility in vitro. The overall results show that the CMC-DA hydrogel with enhanced wet adhesiveness will be a promising tissue adhesive material.
Injectable Dopamine-Modified Poly(ethylene glycol) Nanocomposite Hydrogel with Enhanced Adhesive Property and Bioactivity

Abstract:A synthetic mimic of mussel adhesive protein, dopamine-modified four-armed poly(ethylene glycol) (PEG-D4), was combined with a synthetic nanosilicate, Laponite (Na(0.7+)(Mg5.5Li0.3Si8)O20(OH)4)(0.7-)), to form an injectable naoncomposite tissue adhesive hydrogel. Incorporation of up to 2 wt % Laponite significantly reduced the cure time while enhancing the bulk mechanical and adhesive properties of the adhesive due to strong interfacial binding between dopamine and Laponite. The addition of Laponite did not alter the degradation rate and cytocompatibility of PEG-D4 adhesive. On the basis of subcutaneous implantation in rat, PEG-D4 nanocomposite hydrogels elicited minimal inflammatory response and exhibited an enhanced level of cellular infiltration as compared to Laponite-free samples. The addition of Laponite is potentially a simple and effective method for promoting bioactivity in a bioinert, synthetic PEG-based adhesive while simultaneously enhancing its mechanical and adhesive properties.

Abstract:Commercial or clinical tissue adhesives are currently limited due to their weak bonding strength on wet biological tissue surface, low biological compatibility, and slow adhesion formation. Although catechol-modified hyaluronic acid (HA) adhesives are developed, they suffer from limitations: insufficient adhesiveness and overfast degradation, attributed to low substitution of catechol groups. In this study, we demonstrate a simple and efficient strategy to prepare mussel-inspired HA hydrogel adhesives with improved degree of substitution of catechol groups. Because of the significantly increased grafting ratio of catechol groups, dopamine-conjugated dialdehyde-HA (DAHA) hydrogels exhibit excellent tissue adhesion performance (i.e., adhesive strength of 90.0 +/- 6.7 kPa), which are significantly higher than those found in dopamine-conjugated HA hydrogels (similar to 10 kPa), photo-cross-linkable HA hydrogels (similar to 13 kPa), or commercially available fibrin glues (2-40 kPa). At the same time, their maximum adhesion energy is 384.6 +/- 26.0 J m(-2), which also is 40-400-fold, 2-40-fold, and similar to 8-fold higher than those of the mussel-based adhesive, cyanoacrylate, and fibrin glues, respectively. Moreover, the hydrogels can gel rapidly within 60 s and have a tunable degradation suitable for tissue regeneration. Together with their cytocompatibility and good cell adhesion, they are promising materials as new biological adhesives.
Injectable Dopamine-Polysaccharide In Situ Composite Hydrogels with Enhanced Adhesiveness

Abstract:Polysaccharide bio-adhesives used for non-invasive repair often show weak mechanical strength and tissue adhesion, even when covalently modified with dopamine (DA) from mussel proteins and its derivatives. Low cohesion of the polysaccharide adhesives and easy oxidation of DA may result in the low adhesion properties of the polysaccharide-DA adhesives. In this work, we aimed to prepare a series of injectable hydrogel adhesives to improve their cohesion and adhesion by in situ mixing DA with the polysaccharide without covalent modification. The injectable and rapid curing adhesives were prepared by mixing oxidized dextran (ODE) and chitosan (CS) through a Schiff base reaction in the presence (or absence) of DA. The gelation time of the adhesive was customized to be less than 20 s by controlling the amount of ODE, regardless of the amount of DA. Multi-cross-linked (MC) hydrogels were further prepared by adding cross-linking agents such as sodium periodate (NaIO4) and ferric trichloride (FeCl3), and their sol-gel transitions were easily adjusted by changing the amounts of the cross-linking agents. The MC-FeCl3 hydrogel adhesive displayed good tissue adhesion with a lap shear adhesion strength of 345 kPa, which was 43 times that of fibrin glue. Results from Raman spectra, texture profile analyses, and atomic force microscopy images confirmed the enhanced adhesion induced by a higher cohesion of MC-FeCl3, owing to the coordination of Fe3+ and DA and non-covalent and covalent bonds of DA. Moreover, the adhesives showed good biodegradability and biocompatibility. These results demonstrate that the injectable and sticky hydrogels with good adhesion are promising materials for tissue repair.

Abstract:Recently, cellulose-based hydrogels have been widely used in biomedical applications. However, obtaining hydrogels with integrated tissue adhesion, sensing, haemostatic and wound healing capabilities remains challenging. In this study, we created a catechol-functionalised hydrogel based on multiple hydrogen bonds and π–π stacking using dopamine-modified dialdehyde carboxymethyl cellulose (DCMC-DA) and polyacrylic acid (PAA). The DCMC-DA/PAA hydrogel exhibited strong tissue adhesion, excellent biocompatibility, high sensing performance, effective haemostatic and wound-healing-inducive properties. The abundant aldehyde and catechol moieties of the hydrogels enhanced their adhesion to various substrates. The ions formed by catechol and carboxyl groups endowed the hydrogels with the ability to detect physiological activity signals. Furthermore, the hydrogel demonstrated haemostatic capability, reducing blood loss by 61.1

Abstract:The non-invasive tissue adhesives with strong tissue adhesion and good biocompatibility are ideal for replacing traditional wound treatment methods such as sutures and needles. The self-healing hydrogels based on dynamic reversible crosslinking can recover their structure and function after damage, which is suitable for the application scenario of tissue adhesives. Herein, inspired by mussel adhesive proteins, we propose a facile strategy to achieve an injectable hydrogel (DACS hydrogel) by grafting dopamine (DOPA) onto hyaluronic acid (HA) and mixing it with carboxymethyl chitosan (CMCS) solution. The gelation time and rheological and swelling properties of the hydrogel can be controlled conveniently by adjusting the substitution degree of the catechol group and the concentration of raw materials. More importantly, the hydrogel exhibited rapid and highly efficient self-healing ability and excellent biodegradation and biocompatibility in vitro. Meanwhile, the hydrogel exhibited ~4-fold enhanced wet tissue adhesion strength (21.41 kPa) over the commercial fibrin glue. This kind of HA-based mussel biomimetic self-healing hydrogel is expected to be used as a multifunctional tissue adhesive material.

Abstract:The need for wound closure or surgical procedures has been commonly met by application of sutures. Unfortunately, these are often invasive or subject to contamination (and consequent infection and negative impact on patients). Alternative solutions are offered by surgical adhesives that can be applied and set without major disruption; a new class of supramolecular‐based adhesives provide potential solutions to some of these challenges. In this study a series of polymers utilising dopamine as a self‐assembling unit were synthesized. It was found that these motifs act as extremely effective adhesives, with control over the mechanical strength of the adhesion and materials' tensile properties enabled by changing monomer feed ratios and levels of crosslinking. These materials significantly outperform commercially available bio‐adhesives, showing yield strengths after adhesion at least 2 times higher than that of BioGlue and Tisseel, as well as the ability to re‐adhere with significant recovery of adhesion strength. Promisingly, the materials were shown to be non‐cytotoxic, with cell viability > 90%, and able to perform in aqueous environments without significant loss in strength. Finally, the removal of the materials, after use as an adhesive, was possible using benign organic solvents such as ethanol, ensuring recyclability. These properties all demonstrate the effectiveness of the materials as potential bio‐adhesives, with potential advantages for use in surgery. This article is protected by copyright. Abstract:Gelatin, widely employed in hydrogel dressings, faces limitations when used in high fluid environments, hindering effective material adhesion to wound sites and subsequently reducing treatment efficacy. The rapid degradation of conventional hydrogels often results in breakdown before complete wound healing. Thus, there is a pressing need for the development of durable adhesive wound dressings. In this study, 3-glycidoxypropyltrimethoxysilane (GPTMS) was utilized as a coupling agent to create gelatin-silica hybrid (G-H) dressings through the sol-gel method. The coupling reaction established covalent bonds between gelatin and silica networks, enhancing structural stability. Dopamine (DP) was introduced to this hybrid (G-H-D) dressing to further boost adhesiveness. The efficacy of the dressings for wound management was assessed through in-vitro and in-vivo tests, along with ex-vivo bioadhesion testing on pig skin. Tensile bioadhesion tests demonstrated that the G-H-D material exhibited approximately 2.5 times greater adhesion to soft tissue in wet conditions compared to pure gelatin. Moreover, in-vitro and in-vivo wound healing experiments revealed a significant increase in wound healing rates. Consequently, this material shows promise as a viable option for use as a moist wound dressing.

Abstract:Adhesive hydrogels are attractive biomaterials for various applications, such as electronic skin, wound dressing, and wearable devices. However, fabricating a hydrogel with both adequate adhesiveness and excellent mechanical properties remains a challenge. Inspired by the adhesion mechanism of mussels, we used a two-step process to develop an adhesive and tough polydopamine-clay-polyacrylamide (PDA-clay-PAM) hydrogel. Dopamine was intercalated into clay nanosheets and limitedly oxidized between the layers, resulting in PDA-intercalated clay nanosheets containing free catechol groups. Acrylamide monomers were then added and in situ polymerized to form the hydrogel. Unlike previous single-use adhesive hydrogels, our hydrogel showed repeatable and durable adhesiveness. It adhered directly on human skin without causing an inflammatory response and was easily removed without causing damage. The adhesiveness of this hydrogel was attributed to the presence of enough free catechol groups in the hydrogel, which were created by controlling the oxidation process of the PDA in the confined nanolayers of clay. This mimicked the adhesion mechanism of the mussels, which maintain a high concentration of catechol groups in the confined nanospace of their byssal plaque. The hydrogel also displayed superior toughness, which resulted from nanoreinforcement by clay and PDA-induced cooperative interactions with the hydrogel networks. Moreover, the hydrogel favored cell attachment and proliferation, owning to the high cell affinity of PDA. Rat full-thickness skin defect experiments demonstrated that the hydrogel was an excellent dressing. This free-standing, adhesive, tough, and biocompatible hydrogel may be more convenient for surgical applications than adhesives that involve in situ gelation and extra agents.
A functional hydrogel of dopamine-modified gelatin with photothermal properties for enhancing infected wound healing

Abstract:Infected skin wound has gradually become a prevalent injury that affects overall health. Currently, biomaterials with good adhesion, efficient antibacterial properties, and angiogenesis are considered as a suitable way to effectively heal infected wound. Herein, a multifunctional hydrogel comprising gelatin, dopamine (DA), and ferric ions (Fe3+) was developed for infected wound healing. The modified gelatin-dopamine (Gel-DA) enhanced adhesive capability. Subsequently introducing ferric ions (Fe3+) to form Gel-DA-Fe3+ hydrogels by Fe3+ and catechol coordination bonds. The designed hydrogels demonstrated multifaceted functionality, encompassing photothermal antibacterial, angiogenesis, and so on. The introduction of DA enhanced the adhesion of Gel-DA-Fe3+ to the skin surface and might serve as a physical barrier to seal wound. Meanwhile, DA and Fe3+ jointly endowed good photothermal effects to composite hydrogels, which could eliminate over 95 % of bacteria. In vitro results revealed that Gel-DA-Fe3+ hydrogels had good biocompatibility and promoted HUVECs migration and tube formation. Furthermore, in vivo studies confirmed that Gel-DA-Fe3+ hydrogels markedly expedited the wound healing of rats through eradicating bacteria, accelerating the deposition of collagen, and promoting angiogenesis. What's more, Gel-DA-Fe3+ hydrogels under near-infrared laser had a more pronounced ability for wound healing. Therefore, Gel-DA-Fe3+ hydrogels had great potential for application in bacteria-infected wound healing.

Abstract:Hydrogels' exceptional mechanical strength and skin-adhesion characteristics offer significant advantages for various applications, particularly in the fields of tissue adhesion and wearable sensors. Herein, we incorporated a combination of metal-coordination and hydrogen-bonding forces in the design of stretchable and adhesive hydrogels. We synthesized four hydrogels, namely PAID-0, PAID-1, PAID-2, and PAID-3, consisting of acrylamide (AAM), N,N′-methylene-bis-acrylamide (MBA), and methacrylic-modified dopamine (DA). The impact of different ratios of iron (III) ions to DA on each hydrogel's performance was investigated. Our results demonstrate that the incorporation of iron–dopamine complexes significantly enhances the mechanical strength of the hydrogel. Interestingly, as the DA content increased, we observed a continuous and substantial improvement in both the stretchability and skin adhesiveness of the hydrogel. Among the hydrogels tested, PAID-3, which exhibited optimal mechanical properties, was selected for adhesion testing on various materials. Impressively, PAID-3 demonstrated excellent adhesion to diverse materials and, combined with the low cytotoxicity of PAID hydrogel, holds great promise as an innovative option for biomedical engineering applications.

Abstract:Despite recent progress in hydrogel wound dressings, joint wound treatment remains an ongoing challenge due to their poor adhesive and stretchable properties. Herein, inspired by protein secretion-curing process of natural marine mussels, an adhesive nanofibrous hydrogel is developed via a sequential crosslinking strategy. Firstly, to avoid uncontrolled self-polymerization of dopamine (DA), DA grafted gelatin (GT) nanofibers are fabricated via electrospinning under pH value around 4. Further, the fibrous hydrogels are in-situ crosslinked under pH value of about 8 to strengthen the intermolecular interaction within fibers. The DA crosslinked GT fibrous hydrogels via Michael addition and Schiff base reaction exhibit superior adhesive and tensile properties which are 1.5 times and 3.1 times higher than those of GT fibrous hydrogels, respectively. The in vitro cytocompatibility tests and in vivo wound healing study further prove that GT grafted DA nanofibrous hydrogels are able to accelerate wound healing. Taken together, we demonstrate a simple and versatile strategy for generating adhesive and stretchable nanofibrous hydrogel, showing a great potential for joint wound care.

Abstract:Natural collagen has good biocompatibility and ability to promote tissue regeneration and repair, but the poor mechanical properties and intolerance of degradation of natural collagen limit its applications in the biomedical field. In this research, we synthesized a skin wound repair hydrogel with good biological activity, high strength and excellent water absorption properties. Inspired by the theory of wet healing, dopamine was introduced into the side chain of the water-absorbing polymer polyglutamic acid to synthesize a cross-linking agent (PGAD) with both water absorption and cell adhesion ablities, and then it was introduced into collagen/polyvinyl alcohol (PVA-COL) system to form a double network hydrogel. Scanning electron microscope observation of the morphological characteristics of the hydrogel showed that after the introduction of PGAD, the hydrogel formed an obvious pore structure, and the swelling rate showed that the introduction of PGAD significantly improved the water absorption rate of the hydrogel.In addition, PVA-COL-PGAD hydrogel has good mechanical properties and water absorption behavior.In vitro experimental results revealed that the hydrogel has good biocompatibility. In vivo wound healing experiments showed that hydrogel can promote wound healing process.These results indicated that our hydrogel has great potential as a medical wound dressing.

Abstract:Dopamine was grafted to gelatin, and dextran was oxidized, then the functional hydrogel dressings were fabricated using dopamine‐grafted gelatin and oxidized dextran with incorporation of dipotassium glycyrrhizinate. Hydrogel have been widely used to promote skin tissue regeneration and accelerate wound healing. The development of injectable hydrogels with multi‐functional integration is a priority, especially with antibacterial properties and cytocompatibility. Here, we reported a simple, and versatile method to fabricate a functional hydrogel with potential for wound dressing applications. The dopamine‐grafted gelatin (GT‐DA) and oxidized dextran (oxyglucan, ODex) were synthesized, and dipotassium glycyrrhizinate (DG) was in situ incorporated to fabricate the composite hydrogels (GDO) through the Schiff base reaction. SEM images revealed that hydrogel has a porous structure. By increasing the ratio of GT‐DA to ODex from 1 to 4, the storage modulus (G') and hydrogel network stability nearly doubled, reaching 900 Pa and 220% strain, respectively. The enhancement in mechanical properties could be attributed to the higher cross‐linking density resulting from increased Schiff base bonds in the GDO hydrogels. Furthermore, the GDO hydrogel exhibited antioxidant activity and antibacterial effects on Escherichia coli and Staphylococcus aureus. Moreover, the hydrogel demonstrated cytocompatibility after culturing with human umbilical vein endothelial cells (HUVECs). The results proved that the fabricated GDO hydrogels have the potential to be used as wound dressings.

Abstract:Favorable adhesion and on-demand detachment are the key characteristics for evaluating the effectiveness of diabetic wound dressings in clinical. Herein, a novel adhesive hydrogel is constructed based on dual dynamic covalent (Schiff base and borate ester bond) cross-linking among carboxymethyl chitosan (CMCS), 2-formylphenylboronic acid (2-FPBA) and epigallocatechin-3-gallate (EGCG) as a multifunctional dressing material for diabetic wound healing. The CMCS/2-FPBA/EGCG (CFE) adhesive hydrogels not only display good mechanical strengths (adhesive, injectable, moldable and self-healing), considerable pH-responsive EGCG releasing and photothermal properties, but also excellent antimicrobial, anti-oxidant, anti-inflammatory and angiogenetic activity. Interestingly, CFE adhesive hydrogels display on-demand detachable property while spraying natural organic acid (citric acid, salicylic acid, and vitamin C), which could avoid secondary injury during changing the wound dressings. Therefore, this study provides a promising and comprehensive strategy with great potential for application in the diabetic wound treatment.

Abstract:In this study, a silk fibroin‐dopamine composite is introduced that yields easily deployable hydrogel fibers possessing adhesive properties that can be released on demand to capture and retrieve loads from a distance. The dope combines the functional attributes of silk fibroin (i.e., hydrogel formation) and dopamine (i.e., adhesion) to instantly generate adjustable hydrogel fibers displaying a sticky behavior. B. mori silk has been extensively utilized to create Regenerated Silk Fibroin solutions (RSF) for a wide range of technological applications, including tissue engineering, drug delivery, biomaterials, and adhesives. In this study, an RSF‐dopamine (DA) composite is introduced that yields easily deployable hydrogel fibers possessing adhesive properties that can be released on demand to capture and retrieve loads from a distance. The RSF‐DA serves as an artificial dope, combining the functional attributes of silk fibroin (i.e., hydrogel formation) and dopamine (i.e., adhesion) to instantly generate adjustable hydrogel fibers displaying a sticky behavior. The mechanical strength and adhesive characteristics of this material are assessed using tensile and lap‐shear tests. Furthermore, the possibility of tuning these properties is demonstrated by adding chitosan (Ch) and borate ions (BB), leading to remarkable mechanical and adhesive performances up to 107 MPa and 280 kPa, respectively, which allows the retrieval of objects from the ejected structure. This process can be finely tuned to achieve a controlled fabrication of instantaneously formed adhesive hydrogel fibers for manifold applications, mimicking living organisms' ability to eject tunable adhesive functional threads.

Abstract:Herein, a novel dopamine‐based bioadhesive hydrogel is developed through simple in situ oxidative free‐radical polymerization. This method avoids the self‐polymerization of dopamine and results in the Gel/DMA bioadhesive displaying both excellent light transmittance and adhesion behavior. As an ocular bioadhesive, it can reduce the inflammatory response and fibrosis formation induced by suture surgical trauma. Ocular injuries and their complications represent the most common causes of visual impairment. For ocular surgery, there is an unmet need for highly transparent bioadhesives with superior adhesion, biocompatibility, and regenerative properties. Herein, a novel high‐transparent bioadhesive hydrogel composed of gelatin methacryloyl (GelMA) and dopamine methacrylamide (DMA) is developed by in situ oxidative free‐radical polymerization. This bioadhesive hydrogel overcomes the fundamental weakness of mussel‐inspired adhesive copolymers in clinical practice by combining multiple favorable properties, including high light transmission, mechanical strength, adhesive strength, and biocompatibility. DMA significantly enhances corneal epithelial cells adhesion, proliferation, and migration on GelMA, and prevents the accumulation of reactive oxygen species (ROS) in corneal epithelial cells. In rabbit models of corneal and conjunctiva transplantation, the bioadhesive is able to decrease the inflammatory response and fibrosis formation induced by suture surgical trauma. In addition, the rabbit corneal stromal defect model reveals that the Gel/DMA bioadhesive could effectively seal corneal defects, accelerates corneal re‐epithelialization, and promotes wound healing. Thus, given the advantages of high bioactivity and simple preparation, the Gel/DMA bioadhesive represents a promising strategy for suture‐free ocular repair.

Abstract:Whether used for underwater adhesion or wound hemostasis, adhesives are required to have strong wet adhesion. However, traditional adhesives are difficult to break through the blockade of the hydration interface, so they cannot achieve strong underwater adhesion, and their biological toxicities are also greater, not suitable for wound hemostasis. In this work, small molecule polyethylene glycol diacrylate (PEGDA-200) and dopamine (DA) with good biocompatibility are used as raw materials to synthesize a long chain polymer (PDDA) with hydrophobic backbone and hydrophilic group through the Michael addition reaction. Based on the hydrogen bond interaction between PDDA and silicotungstic acid (SiW) and the solvent exchange between dimethyl sulfoxide and water, the two are successfully caused to agglomerate and form a viscous aggregate (PDDA-SiW). PDDA-SiW not only has strong underwater adhesion to various substrates, but also has strong adhesion and readhesion to fresh pigskins. In addition, PDDA-SiW also has very low hemolysis and cytotoxicity and strong antibacterial properties, and it is a very good underwater adhesion and wound hemostatic material.

Abstract:Inspired by mussel foot proteins, catechol derivatives with a long aliphatic side chain (≈10 atoms in length) are synthesized to prepare tough wet/underwater tissue adhesives. The adhesive with excellent wet tissue adhesion and on‐demand detachment has potential applications in emergency healthcare and the field of sensors and wearable devices. Wet adhesion is critical in cases of wound closure, but it is usually deterred by the hydration layer on tissues. Inspired by dopamine‐mediated underwater adhesion in mussel foot proteins, wet tissue adhesives containing catechol with 2–3 carbons side chains are reported mostly. To make wet adhesion of this type of adhesives much tougher, catechol derivatives with a long aliphatic side chain (≈10 atoms length) are synthesized. Then, a series of strong wet tissue adhesive hydrogels are prepared through photoinduced copolymerization of acrylic acid with synthetic monomers. The adhesive hydrogel has a high cohesion strength, that is, tensile strength and strain, and toughness of ≈1800 kPa, ≈540%, and ≈4100 kJ m−3, respectively. Its interfacial toughness on wet and underwater porcine skin is respectively ≈1300 and ≈1100 J m−2, and its adhesion strength to wet porcine skin is ≈153 kPa. These values are much higher than those of dopamine‐based adhesives in the same conditions, demonstrating that the long aliphatic side chain on catechol can greatly improve the wet tissue‐adhesion. Additionally, the tough interfacial adhesion can be broken on demand with 5 wt.% aqueous urea solution. This adhesive hydrogel is highly promising in safe wound closure.
