Abstract:Man's inhumanity to man is expressed through a plethora of tools of modern warfare and terror. The use of chemical and biological weapons with the goals of assault, demoralisation and lethality has been documented in recent history, both on the battlefield and in urban terror against civilians. A general review of a few of the currently employed chemical weapons and biological toxins, along with a look at potential chemical weapons and tools of counter-terrorism, follows. While these weapons are fearsome elements, the dangers should be viewed in the context of the widespread availability and efficacy of conventional weapons.

Abstract:Abstract Occurrence of large-scale biochemical wars is of little possibility today, but the international chemical security situation is still not optimistic. Effective decontamination of chemical warfare agents has received close attention, becoming a global necessity. In this paper, data in the Web of Science Core Collection database were selected to systematically investigate the relevant literature about the research progress on decontamination of chemical warfare agents from 1990 to 2020. Origin Pro 2019b and CiteSpace were conducted for multi-dimensional bibliometric analysis on literature type distribution, number of annual publications, countries, institutions, journals, authors, literature co-citation and citation bursts. By August 1, 2020, there are 2,602 papers related have been published and the number of annual publications shows a linear growth tendency. Amongst all 71 countries, United States is the country with the most publications as well as the largest centrality value. US ARMY ranks fourth in the number of publications while with the highest institution-based cooperation. “Nerve agents and blister agents”, “metal-organic frameworks” and “applications of metal-organic frameworks” are prevailing in recent years, which highlights the latest development direction and research frontiers. This article promotes scientific predictions and impact assessments of decontamination of chemical warfare agents.

Abstract:Nuclear arms control remains a priority for the foreseeable future for many stakeholders, and proposals have emerged to focus on capping nuclear warheads of the main nuclear-weapon states. However, delivery vehicles are another source of instability and arms race dynamics. Whether they are coupled with weapons of mass destruction or considered exclusively in the context of their use with conventional weapons, missiles are increasingly transferred, produced, modernized, and used in military conflicts. The development of offensive capabilities can also lead to a negative regional or global spiral with the increased deployment of defensive systems, and in response, new efforts to procure offensive weapons. It is therefore useful to keep thinking about ways to limit the destabilizing effect of these weapon systems. Some legal instruments currently exist in unilateral, bilateral or multilateral forums. Their focus may be limited to nonproliferation or they may cover a broader range of issues and address the behavior of states acquiring these delivery vehicles. This article will discuss ways in which these instruments can evolve to better respond to current trends and dynamics regarding missiles, but also will suggest new initiatives, particularly confidence-building measures, that could be useful to reduce the destabilizing effect of these systems.

Abstract:This article offers a Chinese perspective on reducing the threat of nuclear weapons due to nuclear weapons proliferation and its deterrent gestures, long employed since the Cold War. It is pointed out that nuclear weapons are increasingly irrelevant to nuclear weapons states. However, some non-nuclear weapons states may view this differently. Nuclear abolition has presented a visionary opportunity for all sides to relinquish those weapons increasingly unnecessary for nuclear weapons states. Before such an idealistic world is attained, efforts of nuclear non-proliferation are indispensable along with a process of nuclear disarmament.

Abstract:AbstractSince the insight to fuse Fenton chemistry and nanomedicine into cancer therapy, great signs of progress have been made in the field of chemodynamic therapy (CDT). However, the exact mechanism of CDT is obscured by the unique tumor chemical environment and inevitable nanoparticle‐cell interactions, thus impeding further development. In this Scientific Perspective, the significance of CDT is clarified, the complex mechanism is deconstructed into primitive chemical and biological interactions, and the mechanism research directions based on the chemical kinetics and biological signaling pathways are discussed in detail. Moreover, beneficial outlooks are presented to enlighten the evolution of next‐generation CDT. Hopefully, this Scientific Perspective can inspire new ideas and advances for CDT and provide a reference for breaking down the interdisciplinary barriers in the field of nanomedicine.

Abstract:To the Editor — Dual-use research of concern (DURC) can be defined as research, mainly in the life sciences, that has the potential to be misapplied for harmful purposes 1 . Key examples are the synthesis of mousepox 2 , the synthesis of poliovirus 3 , the generation of the 1918 influenza virus 4 , gain-of-function studies with H5N1 in ferrets 5 and the synthesis of horsepox, the viral cousin of smallpox 6 . These seminal events involved the physical synthesis of a biological agent. However, the time may have come to also consider dual-use risk of the development of toxic agents in silico, in the light of the alarming results of a computational experiment we recently performed for a biennial arms control conference 7 ; we used a generative artificial intelligence (AI) approach previously developed for drug discovery applications, and found it could easily design a range of nerve agents including VX. The experiment demonstrated the alarming speed and ease with which such software — based on open-source tools and datasets from the public domain — could be used for bad purposes. Our experiment was subsequently covered widely in the media, reaching a network of scientists, experts and lay people alike 8 , and its implications were recognized at the highest levels of governments within a matter of days. The level of interest was probably amplified owing to the enfolding war in Ukraine, with Russia's invasion and the threat of biochemical weapons use.

Disaster Medicine and Public Health Preparedness

Abstract:Although chemical and radiological agents cause toxicity through different mechanisms, the multiorgan injuries caused by these threats share similarities that convene on the level of basic biological responses. This publication will discuss these areas of convergence and explore "multi-utility" approaches that could be leveraged to address common injury mechanisms underlying actions of chemical and radiological agents in a threat-agnostic manner. In addition, we will provide an overview of the current state of radiological and chemical threat research, discuss the US Government's efforts toward medical preparedness, and identify potential areas for collaboration geared toward enhancing preparedness and response against radiological and chemical threats. We also will discuss previous regulatory experience to provide insight on how to navigate regulatory paths for US Food and Drug Administration (FDA) approval/licensure/clearance for products addressing chemical or radiological/nuclear threats. This publication follows a 2022 trans-agency meeting titled, "Overlapping Science in Radiation and Sulfur Mustard Exposures of Skin and Lung: Consideration of Models, Mechanisms, Organ Systems, and Medical Countermeasures," sponsored by the National Institute of Allergy and Infectious Diseases (NIAID), a part of the National Institutes of Health (NIH). Discussions from this meeting explored the overlapping nature of radiation and chemical injury and spurred increased interest in how preparedness for one threat leads to preparedness for the other. Herein, subject matter experts from the NIAID and the Biomedical Advanced Research and Development Authority (BARDA), a part of the Administration for Strategic Preparedness and Response (ASPR), summarize the knowledge gained from recently funded biomedical research, as well as insights from the 2022 meeting. These topics include identification of common areas for collaboration, potential use of biomarkers of injury to identify injuries caused by both hazards, and common and widely available treatments that could treat damage caused by radiological or chemical threats.

Abstract:The United States now knows that it is vulnerable to terrorist attacks. In Countering Terrorism, experts from such disparate fields as medicine, law, public policy, and international security discuss institutional changes the country must make to protect against future attacks. In these essays, they argue that terrorism preparedness is not just a federal concern, but one that requires integrated efforts across federal, state, and local governments.The authors focus on new threats -- biological attacks, dirty bombs containing radioactive materials, and cyberattacks that would disrupt the computer networks we rely on for communication, banking, and commerce -- and argue that US institutions must make fundamental changes to protect against them. They discuss not only the needed reorganization of government agencies but such institutional issues as establishing legal jurisdiction to respond to new threats, preparing health workers for attacks involving mass casualties, and equipping police, fire, and other emergency workers with interoperable communications systems. The final essays examine how Israel, Japan, and the United Kingdom have dealt with domestic terrorism, and what the United States can learn from their examples.
The bioterrorism threat and dual-use biotechnological research: an Israeli perspective

Abstract:Israel has a long history of concern with chemical and biological threats, since several hostile states in the Middle East are likely to possess such weapons. The Twin-Tower terrorist attacks and Anthrax envelope scares of 2001 were a watershed for public perceptions of the threat of unconventional terror in general and of biological terror in particular. New advances in biotechnology will only increase the ability of terrorists to exploit the burgeoning availability of related information to develop ever-more destructive bioweapons. Many areas of modern biological research are unavoidably dual-use by nature. They thus have a great potential for both help and harm; and facilitating the former while preventing the latter remains a serious challenge to researchers and governments alike. This article addresses how Israel might best (1) prevent hostile elements from obtaining, from Israel's biological research system, materials, information and technologies that might facilitate their carrying out a biological attack, while (2) continuing to promote academic openness, excellence and other hallmarks of that system. This important and sensitive issue was assessed by a special national committee, and their recommendations are presented and discussed. One particularly innovative element is the restructuring and use of Israel's extensive biosafety system to also address biosecurity goals, with minimal disruption or delay.

It provides an overview of the status of compliance with the norm against chemical weapons, and sets out the compliance mechanisms that are available under the Convention, outside the Convention, and that can be used by individual States. Finally, the authors suggest six concrete ways in which the different tools for management and enforcement of compliance can be improved.

Abstract:This Commentary delves into the current progress and challenges on ongoing research on medical countermeasures (MCs) for chemical, biologic, radiologic, and nuclear (CBRN) threats. CBRN agents pose a serious risk to human health and safety, with the potential for mass casualties in both military and civilian settings. Chemical threats are toxic compounds that could be used in a terrorist attack, an accidental release, or chemical warfare. They include nerve agents, organophosphates, pulmonary agents, metabolic/cellular agents, vesicants, ocular toxicants, and opioid agents. Developing effective MCs is crucial for mitigating the acute and chronic effects of exposure to CBRN agents. The papers in this special issue of JPET highlights the latest advancements in MC research, showcasing insightful outcomes on experimental models, mechanisms, and translational research on MCs for CBRN threats. They portray several notable contributions, including the development of neurosteroid and combination anticonvulsant therapies for nerve agent poisoning, the exploration of chronic impacts and diagnostic tracers for OP neurotoxicity, the establishment of innovative pediatric OP models, the identification of novel molecules for ocular, pulmonary and vesicant injuries, and the repurposing of existing drugs for the treatment of botulism, cyanide, and OP poisoning. These crucial outcomes underscore the breadth of current research covering a variety of chemical threats. Overall, this collection of articles highlights the importance of ongoing research and development in the field of MCs, emphasizing the potential of these countermeasures to effectively treat and mitigate the effects of toxicant exposures and thereby enhance our preparedness for mass casualty incidents. SIGNIFICANCE STATEMENT CBRN agents pose a significant threat to public health. Effective MCs exist for certain chemical threats, but there is a need for new and improved MCs for many others. The research presented in this special issue of JPET highlights the latest advancements in MCs for CBRN threats. This research has the potential to lead to the development of new and repurposed MCs that are more effective, broad-spectrum, and easier to administer to mitigate acute and long-term consequences of chemical exposures.

Abstract:<p>Although 193 states have committed to the Chemical Weapons Convention and 98% of the declared chemical weapons stockpiles have been destroyed so far, nerve agent poisoning remains a lingering threat. The recent dissemination of sarin in Syria, the assassination of Kim Jong‐Nam in Malaysia, and the assault on Sergei Skripal in the United Kingdom underline the need for effective treatment. The current therapeutic options of a muscarinic receptor antagonist, an oxime, and an anticonvulsant have been unchanged for decades. Therefore, new therapeutic strategies, for example, bio scavengers and receptor‐active substances, are promising concepts that have to be examined for their benefits and limitations. In order to facilitate rapid diagnosis in challenging clinical situations, point‐of‐care diagnostics and detection are of importance. Therapeutic guidance concerning the duration and success of the current oxime therapy via determination of the cholinesterase status can contribute to an optimal use of resources. In summary, the challenges of current and future therapies for nerve agent poisoning and key diagnostic devices will be discussed.</p>

Abstract:With the discovery of chemical weapons, mankind has faced a great threat. These weapons are also called weapon of mass destruction and cause mass human deaths in the region where they are used, regardless of whether they are soldiers or civilians. Countries have focused on chemical defense activities because of the easy production and development of chemical weapons and negative psychological effects on the public. Many people were killed or injured due to the use of chemical weapons during World War I. Nerve agents started to be used in making chemical weapons before World War II began. Thus, the threat of chemical weapons on humanity has reached a much more critical point. Especially during World War II and the Cold War, the development of chemical weapons and the production of nerve agents increased more than ever.In this review, various sensor systems developed for detecting nerve agents have been investigated and these sensors have been compared in terms of operating principles and detection limits.

Abstract:The end of the Cold War has brought about an important shift in emphasis concerning the world's submarine forces. Rather than investing in nuclear submarines, many emerging naval powers are looking to cheaper diesel craft as a key to their future naval strategy. Diesel submarines have some major disadvantages when it comes to operations in the world's oceans, but they are ideally suited to a defensive role in coastal waters, especially deterring an amphibious operation. Widespread fear of US military intervention in regional conflicts has encouraged many states to concentrate on defending their coastal areas, where the West may now find itself at a disadvantage.
Repurposing FDA-approved drugs to treat chemical weapon toxicities: Interactive case studies for trainees

Abstract:The risk of a terrorist attack in the United States has created challenges on how to effectively treat toxicities that result from exposure to chemical weapons. To address this concern, the United States has organized a trans-agency initiative across academia, government, and industry to identify drugs to treat tissue injury resulting from exposure to chemical threat agents. We sought to develop and evaluate an interactive educational session that provides hands-on instruction on how to repurpose FDA-approved drugs as therapeutics to treat toxicity from exposure to chemical weapons. As part of the Rutgers Summer Undergraduate Research Fellowship program, 23 undergraduate students participated in a 2-h session that included: (1) an overview of chemical weapon toxicities, (2) a primer on pharmacology principles, and (3) an interactive session where groups of students were provided lists of FDA-approved drugs to evaluate potential mechanisms of action and suitability as countermeasures for four chemical weapon case scenarios. The interactive session culminated in a competition for the best grant "sales pitch." From this interactive training, students improved their understanding of (1) the ability of chemical weapons to cause long-term toxicities, (2) impact of route of administration and exposure scenario on drug efficacy, and (3) re-purposing FDA-approved drugs to treat disease from chemical weapon exposure. These findings demonstrated that an interactive training exercise can provide students with new insights into drug development for chemical threat agent toxicities.
