Abstract:Photo-sensitive probes based on coumarin deserve extensive attention because of their high biocompatibility, quantum yield, and stability. The synthesis of coumarin and its derivatives is mainly based on four classical methods: Perkin synthesis, Knoevenagel synthesis, Wittig synthesis, and Pechmann synthesis. As everyone knows, fluorescent probes consist of a recognition group, a bridging group, and a luminescent group. Mechanism of fluorescent probes can be roughly summarized into four models: Chelation-induced enhanced fluorescence (CHEF), intramolecular charge transfer (ICT), photoinduced electron transfer (PET), and aggregation-induced luminescence (AIE), which can instruct the design of coumarin-based photo-sensitive probes. Since the fluorescence-based probe was first reported, probes based on coumarin have found widespread application in analytical chemistry, proving to be a selective, efficient, and convenient tool in ion monitoring, molecular monitoring, cell imaging, drug delivery, and many other fields. In this critical review, the synthetic routes of these compounds and the design for fluorescent probes were reported. Furthermore, the recent researches on the employment of coumarin in analytical chemistry were also introduced. Finally, the future research trends and application foreground of coumarin derivatives were forecasted herein.

In recent 20 years, research attention has been paid particularly to synthesis and separation of fluorescein derivatives having various linker groups. This mini-review briefly summarizes synthesis of fluorescein derivatives for improving their chemical, fluorescent and biological properties and conjugation suitability.

Abstract:Compact fluorophores are desired for bioconjugation because they minimally perturb the native biological functions of proteins or organelles. However, their absorption and emission wavelengths are typically short and not ideal for practical applications. For example, coumarin is one of the smallest fluorophores with excellent fluorescence properties. However, its potentials in biological applications are limited by its relatively short excitation wavelengths in the UV spectral region. It is the aim of this manuscript to develop coumarin analogs absorbing and emitting in the visible range through rational molecular engineering. We designed and synthesized C-coumarin dyes, and found that their maximal absorption wavelengths fall well within the visible region, rendering them superior for live cell imaging than O-coumarins. The other promising properties of C-coumarins include larger Stokes’ shifts, low cytotoxicity, and high chemostability.

Abstract:Coumarin-based fluorogenic probes have emerged as preferred imaging tools to real-time and non-invasively monitor target analyte(s) in biological systems, due to their inherent merits of tunable optical properties, facile synthesis, excellent cell-membrane permeability, and favorable biocompatibility. Compared to natural coumarins, coumarin-based near-infrared probes (CBNPs) display improved signal-to-noise ratio and higher spatial imaging resolution. Over the past few decades, multifunctional coumarin scaffolds have been used to construct practical CBNPs for dynamically tracking cellular microenvironments, metal ions, small molecules, and biomacromolecules of interest, as well as for drug screening and disease diagnosis. Herein, the advances in the development of CBNPs from molecular engineering to biomedical applications in the last decade are reviewed. Firstly, the structure-fluorescence relationships (SFRs) of coumarin derivatives, along with diverse structural types, as well as sensing mechanisms of diverse high-performance CBNPs, are well-summarized. Next, a wide array of state-of-the-art CBNPs with various recognition fragments and sensing mechanisms, together with their biomedical applications in native living systems, are illustrated. Meanwhile, the applications of CBNPs for sensing or visualizing disease biomarkers, drug screening, assessing drug efficacy in vivo, and photothermal/photodynamic therapy are also covered. Finally, the existing challenges and future perspectives in this field are highlighted. Collectively, the information and knowledge presented here provide valuable strategies and practical cases for constructing multifunctional CBNPs, which will be very helpful for researchers to devise more versatile CBNPs tools for both fundamental and applied studies.

Abstract:Two novel chlorinated fluoresceins 2′,4′,5′,7′-tetrachloro-6-(5-carboxypentyl)-4,7-dichlorofluorescein succinimidyl ester and 2′,4′,5′,7′-tetrachloro-6-(3-carboxypropyl)-4,7-dichloro fluorescein succinimidyl ester were synthesized as fluorescent probes for labeling proteins. The investigation in immunofluorescence histochemistry showed them had strong fluorescence, high photostability and good biocompatibility.
A Coumarin-Based Fluorescent Probe for Biological Thiols and Its Application for Living Cell Imaging.

Abstract:In this work, compound 1 has been rationally designed and synthesized as a new fluorescent probe for biological thiols. Notably, probe 1 has almost no background fluorescence (Phi(f) < 0.0001) in aqueous solutions; however, it exhibited fluorescence turn-on response to thiols with high sensitivity (a 246-fold fluorescence enhancement and a low detection limit of 0.22 mu M for Cys). Moreover, probe 1 showed excellent thiol specificity over other biologically relevant species. The kinetic studies indicated that the probe responded to thiols rapidly, and the pseudo-first-order rate constants of probe 1 reaction with Cys, Hcy, and GSH were determined to be 1.85842, 0.67656, and 0.51519 min(-1), respectively. A possible detection mechanism was proposed to involve the Michael addition of the thiol to the alpha,beta-unsaturated ketone, followed by a cleavage of the hemiketal group, thereby leading to the formation of a fluorescent 7-hydroxyl coumarin derivative. Furthermore, the optical responses of probe 1 to thiols were studied by TD-DFT calculations. Finally, probe 1 has been successfully applied to the detection of biological thiols in human blood serum. And the intracellular imaging applications established that probe 1 can be used to detect different concentrations of intracellular thiols in living cells.

Abstract:In this study, a novel low-molecular-weight coumarin fluorescent dye, 3,4-dicyano-7-diethylaminocoumarin, was designed. This dye features a donor-π-bridge-acceptor (D-π-A) conjugated structure, which confers superior fluorescence properties.The dye was synthesized using a simple, efficient, and environmentally friendly two-step process, facilitating the direct conversion of the key intermediate into the final product via simultaneous addition and elimination reactions. A comprehensive structural characterization of the dye was undertaken, and its UV absorption and fluorescence emission properties were extensively analyzed using spectroscopic techniques, with structural explanations of the fluorescence activities provided by the Density Functional Theory (DFT) method. Experimental results revealed that this coumarin dye emits strong fluorescence, characterized by a long wavelength, a large Stokes shift, and high fluorescence intensity. It exhibits excellent fluorescent performance in various solvents. Despite experiencing fluorescence quenching in its solid state, the formation of an inclusion complex with β-cyclodextrin restores strong red fluorescence, presenting new possibilities for biofluorescence imaging. Notably, this coumarin dye also demonstrates a significant solvatochromic effect. This effect enables rapid visual discrimination among multiple sets of structurally and chemically similar organic solvents. Consequently, the dye holds promise as a versatile solvatochromic fluorescent probe for a broad spectrum of applications.

Abstract:A series of Cu 2+ probe which contains 9 probes have been synthesized and established. All the probes were synthesized using Rhodamine B as the fluorophore, conjugated to various differently substituted cinnamyl aldehyde with C=N Schiff base structural motif as their core moiety. The structure-property relationships of these probes have been investigated. The change of optical properties, caused by different electronic effect and steric effect of the recognition group, has been analyzed systematically. DFT calculation simulation of the Ring-Close and Ring-Open form of all the probes have been employed to illuminate, summarize and confirm these correlations between optical properties and molecular structures. In addition, biological experiment demonstrated that all the probes have a high potential for both sensitive and selective detection, mapping of adsorbed Cu 2+ both in vivo and environmental microbial systems. This approach provides a significant strategy for studying structure-property relationships and guiding the synthesis of probes with various optical properties.

Abstract:Flavonols, named as 3-hydroxyl-2-phenyl-1-benzopyran-4-one, are important natural botanical functional pigments widely distributing in fruits, vegetables, and flowers. Owing to intramolecular hydrogen bond, the flavonols can undergo the excited-state intramolecular proton transfer (ESIPT) process upon irradiation, leading to unique environmental-sensitive dual emissions, which makes flavonol an ideal fluorophore skeleton for probe design. In this paper, we summarized the recent progress of flavonol-based small-molecule fluorescent probes for diverse sensing applications, including detection of anions, cations, reactive biological species, proteins, DNAs, environmental hazards, and microenvironmental factors. This review highlighted the basic characteristics of flavonol probes in the respect of design principles, photophysical processes, sensing mechanism, sensing performances, and biological applications. The present review article aims to offer a comprehensive summary for researchers to inspire new probe design, and push forward the advancement of flavonol-based fluorescent probes.

Abstract:Xanthene dyes are a type of fluorescent dye with a π conjugated system and a unique ring-opening mechanism. Fluorescent probes with xanthene dyes as fluorescent groups have the advantages of high molar extinction coefficient, high fluorescence quantum yield, good stability, and good water solubility, making them suitable for medical imaging and biological imaging diagnosis. Therefore, this article reviews the research progress of xanthene fluorescent probes in recent years in terms of pH value, reactive oxygen species, metal ions, anions, enzymes, thiol derivatives, etc., summarizes their design ideas, detection performance, and applications, and points out the development trend of xanthene fluorescent probes, providing a reference for the subsequent development and utilization of xanthene fluorescent probes.

Abstract:Bright and hydrosoluble fluorescent dyes with large Stokes shifts are widely used as molecular probes and light emitting markers in chemistry and biology. In this report, one-step synthesis of a bright green-emitting coumarin (Phi = 0.52, lambda(em) = 505 nut) was achieved from available reagents under neat conditions. The novel dye 1 showed very large Stokes shift (Delta lambda = 138 nm) due to strong intramolecular charge transfer (ICT) effect, which was confirmed by density functional theory (DFT) calculations. Dye 1 was hydrosoluble and displayed strong emission at a wide range of pH from 3 to 8. Importantly, the secondary amine in 1 made it extremely convenient for futher bioconjugation. Representative derivatives were showcased for various successful applications of fluorogenic enzyme assay, biolabelling and mitochondrial staining.

Abstract:Bioanalysis based on optical imaging has gained significant progress in the last few decades. Luminescence probes are capable of detecting, monitoring, and tracing particular biomolecules in complex biological systems to figure out the roles of these molecules in organisms. Considering the rapid development of luminescence probes for bio-applications and their promising future, we have attempted to explore the working principles and recent advances in bio-applications of luminescence probes, in the hope of helping readers gain a detailed understanding of luminescence probes developed in recent years. In this review, we first focus on the current widely used luminescence probes, including fluorescence probes, bioluminescence probes, chemiluminescence probes, afterglow probes, photoacoustic probes, and Cerenkov luminescence probes. The working principles for each type of luminescence probe are concisely described and the bio-application of the luminescence probes is summarized by category, including metal ions detection, secretion detection, imaging, and therapy.

Abstract:Fluorescein derivatives are important fluorescent probes which can be used for detection and optical imaging. Fluorescein derivatives are usually constructed by introducing aldehyde groups or esterified onto fluorescein xanthene ring and benzene moiety. Typically, the research direction of connecting amino groups with fluorescein monoaldehyde is in hot. Because of their high activity, these derivatives can be complexed with the analytes to produce changes of colors and the increase or decrease of fluorescence intensity. This article reviewed fluorescein probes in the past two years according to classification of different analytes including metal ions, anions, small molecules and biological macromolecules. The synthesis methods, optical properties, possible mechanisms and applications of fluorescein probes are summarized. This article provides a reference for the screening of fluorescein probes with high sensitivity and biological detection and can propel their further applications in sensing and detection of analytes.

Abstract:Coumarin-based chromophores with azo and pyrazoline moieties were synthesized and their structures and properties elucidated using spectroscopy. Abstract:The discovery of a simple and efficient detection method for biothiols would be scientifically significant due to the crucial role of them in various physiological processes. Recently, a simple fluorescent probe, DEMCA-NBSC, based on coumarin fragments, was developed by Ding et al., and provided an efficient way for real-time sensing of biothiols both in vivo and vitro. Theoretical insights to the fluorescence sensing mechanism of the probe were provided in this work. Details of the electron transfer process in the probe under optical excitation and the fluorescent character of the probe were analyzed using a quantum mechanical method. All these theoretical results could inspire the development of a highly convenient and efficient fluorescent probe to sense biothiols both in vivo and vitro.
