Abstract:Good sense: A highly sensitive and selective ratiometric fluorescent probe for Fe2+ was developed by linking an Fe2+-insensitive fluorophore to an Fe2+-sensitive reporter–fluorophore fragment. The fluorescence intensity ratio increased linearly with the concentration of Fe2+ (see scheme). The probe was applied to the ratiometric imaging of intracellular and adscititious fluctuations of Fe2+ in live HL-7702 and HepG2 cells.

Abstract:A novel coumarin-derived Schiff base fluorescent probe was reported to detect Cu 2+ ions in human cells. It was synthesized by the reaction between 7-(diethylamino)coumarin-3-carbohydrazide and pyridoxal 5′-phosphate. The probe has good stability and it exhibited obviously decreasing fluorescent response at λ 490 nm upon addition of Cu(II) ions. The fluorescent probe showed high selectivity for Cu(II) ions compared to other metal ions. The probe has been successfully applied to detect Cu(II) ions in human cells.

Abstract:Mercury (Hg) is a heavy metal with high toxicity and easy migration; it can be enriched through the food chain, and cause serious threats to the natural environment and human health. So, the development of a method that can be used to detect mercury ions (Hg2+) in the environment, in cells, and in organisms is very important. Here, a new 7-hydroxycoumarin-derived carbonothioate-based probe (CC-Hg) was designed and synthesized for detection of Hg2+. After addition of Hg2+, a large fluorescence enhancement was observed due to the formation of 7-hydroxyl, which reinforced the intramolecular charge transfer process. The CC-Hg probe had good water solubility and selectivity. Moreover, the probe was able to detect Hg2+ quantitatively over the concentration range 0-2 mu M and with a detection limit of 7.9 nM. Importantly, we successfully applied the probe to detect Hg2+ in water samples, in living cells, and in zebrafish. The experimental results demonstrated its potential value in practical applications.

Abstract:A novel coumarin-derived Schiff base fluorescence probe (CTB) has been successfully designed and synthesized through exploiting tris-(2-aminothyl)-amine moiety as a recognition unit for the highly selective and sensitive detection of Cd2+. It is based on CN isomerization and the photo-induced electron transfer (PET) mechanism. The investigation into the sensing processes showed that CTB exhibited an excellent selectivity for Cd2+. The sensitivity exceeded that of other competing metal ions, and had a high sensitivity, a detection limit of 1.16 × 10-7 M with the association constants of 1.37 × 1011 M-2. The experiments including Job's plot, UV-Vis titration, 1H NMR titration and ESI-MS spectrum established that the probe CTB binds to Cd2+ in a 1:2 ratio. Further studies also demonstrated that probe CTB can be successfully applied to the fluorescence imaging of Cd2+ in HepG-2 cells.

Abstract:A novel coumarin-based pyrazoline fluorescent probe A was synthesized through a four-step reaction. The structure was characterized by 1H NMR(Nuclear magnetic resonance), 13C NMR and HR-ESI-MS(High resolution electrospray ionization mass spectrometry). The optical properties of the fluorescent probe was investigated by UV–vis absorption and fluorescence emission spectra. The results showed that the addition of Fe3+ makes the fluorescence emission spectra of A quenched and displayed a linear fluorescence response to Fe3+ with a detection limit of 1.01 × 10−7 M. It also shows good reversibility upon addition of EDTA(Ethylene diamine tetraacetic acid). The structure of the fluorescent probe was optimized by DFT(Density functional theory) calculations, and the binding mechanism between the fluorescent probe and the recognition ion was explained. Moreover, the probe also has practical application in cell imaging.

Abstract:In this paper, a novel coumarin-derived fluorescent probe NY was designed and synthesized. NY displayed a significant ratiometric fluorescence response towards Cu 2+ in PBS buffer (10 mM, pH = 7.4), with the emission wavelength blue-shifted from 580 to 495 nm, and a fluorescence change from orange to green was evident under a 365 nm UV light. Meanwhile, NY had the advantages of high selectivity, short response time (5 min), low detection limit (1.3 × 10 -8 M) and large binding constant (1.45 × 10 5 M -1 ) towards Cu 2+ . The binding mechanism between NY and Cu 2+ was elucidated by FT-IR, 1 H NMR titration, TOF-MS and Job's plot analysis. In addition, NY was successfully employed in the detection of Cu 2+ within environmental water and vegetable samples with satisfactory results. Laser confocal microscopy imaging results showed that NY could easily penetrate HeLa cells membrane to target mitochondria and image Cu 2+ in living cells. Furthermore, NY demonstrated mechanochromic properties by exhibiting orange-red fluorescence when subjected to mechanical grinding.

Abstract:Photo-sensitive probes based on coumarin deserve extensive attention because of their high biocompatibility, quantum yield, and stability. The synthesis of coumarin and its derivatives is mainly based on four classical methods: Perkin synthesis, Knoevenagel synthesis, Wittig synthesis, and Pechmann synthesis. As everyone knows, fluorescent probes consist of a recognition group, a bridging group, and a luminescent group. Mechanism of fluorescent probes can be roughly summarized into four models: Chelation-induced enhanced fluorescence (CHEF), intramolecular charge transfer (ICT), photoinduced electron transfer (PET), and aggregation-induced luminescence (AIE), which can instruct the design of coumarin-based photo-sensitive probes. Since the fluorescence-based probe was first reported, probes based on coumarin have found widespread application in analytical chemistry, proving to be a selective, efficient, and convenient tool in ion monitoring, molecular monitoring, cell imaging, drug delivery, and many other fields. In this critical review, the synthetic routes of these compounds and the design for fluorescent probes were reported. Furthermore, the recent researches on the employment of coumarin in analytical chemistry were also introduced. Finally, the future research trends and application foreground of coumarin derivatives were forecasted herein.

The construction of a near-infrared fluorescent probe with dual advantages for imaging carbon monoxide in cells and in vivo

Abstract:As a kind of toxic gas, carbon monoxide (CO) can hinder uptake of oxygen in humans. However, more and more studies have shown that CO is an important gaseous messenger in the body and playing an indispensable role in intracellular signaling pathways. So, it is necessary to develop an analytical method to study CO in living organisms. Although there are many CO-responsive probes, most of them have the disadvantages of a small Stokes shift or short emission wavelength. In order to address the above issue, a novel probe (FDX-CO) with a large Stokes shift (190 nm) and long emission wavelength (770 nm) was firstly synthesized to detect CO. The probe shows high sensitivity and superior selectivity toward CO. Moreover, the probe was successfully used for visualizing exogenous and endogenous CO in cells by fluorescence imaging, 3D quantification analysis and flow cytometric analysis. More importantly, FDX-CO could excellently detect CO in mice, which suggests that this probe has the potential ability to image CO in vivo. This probe can be viewed as a useful tool in the research of CO.

Abstract:A novel near-infrared fluorescent probe, IR-736, was synthesized and applied to living cells. The probe exhibited good response fluorescent characteristic, and cells experiments showed the probe had high affinity and without apparent cytotoxicity. Fluorescent image experiments in living L929 cells, MCF-7 cells, HepG2 cells, further demonstrated its potential applications in biological systems. The probe effectively prevented the influence of autofluorescence and native cellular species in biological systems. It also exhibited good photostability, and excellent cell membrane permeability.

Abstract:Carbon monoxide (CO), as one of significant gas transmitter, is closely associated with a variety of physiological and pathological processes. Although plenty of fluorescent probes have been prepared for detecting CO, most of them suffer from water-soluble fluorophores and short emission wavelength, which tends to diffuse and is limited to apply in vivo. Herein, a novel water-soluble fluorescent probe (HPQ-MQ-CO) is prepared to detect CO by releasing a precipitating fluorochrome (HPQ-MQ-OH), which is developed by introducing the 1-ethyl-2-methylquinoline group into HPQ to obtain long emission wavelength and good diffusion resistant ability. Allyl formate, as the identification unit of CO, has good water solubility and quenches the fluorescence of HPQ-MQ-CO. When the probe reacts with CO and Pd2+, an long-emission and solid-state fluorescence signal at 650 nm can be observed, which is based on excited-state intramolecular proton transfer (ESIPT) mechanism. When the concentration of CO is raised to 100.0 μM, the fluorescence is increased 29 times, indicating the sensitivity of the probe. Moreover, this probe shows prominent selectivity for CO compared with other interfering species. Given these advantages, HPQ-MQ-CO can be used for CO detection in HepG2 cells and zebrafish by in-situ and long-term fluorescence imaging. In addition, this probe can monitor the up-regulation of CO in HepG2 cells and zebrafish during drug-induced liver injury (DILI).

Abstract:In recent years, targeting drugs made by physical loading or chemical bonding of drugs on small molecular carriers have shown a very wide application prospect in the field of tumor and cancer treatment. How to achieve the release of drugs in cancer cells has become the core of this research. One of the most important bases for drug localization is to use the difference of small molecular biothiol concentration between cancer cells and normal cells. Details of the changes of biothiol levels in the growth and reproduction of cancer cells are still poorly understood, and the main reason is the lack of sensitive real-time imaging tools for biothiols in cancer cells. In this work, we reasonably designed and synthesized the combination of 4-hydroxy-1,8-naphthalimide and NBD-Cl as a concise fluorescent probe HN-NBD for imaging biothiols in live cells and zebrafish. In addition, due to the advantages of HN-NBD design, it is sufficiently sensitive to biothiols, and further imaging can distinguish cancer cells from normal cells. Probe HN-NBD would be of great significance to biomedical researchers for the study of biothiol-related diseases, the screening of new anticancer drugs, and the early diagnosis and treatment of cancers.

Abstract:Carbon monoxide (CO) is an important gas transporter and is closely associated with a series of physiological and pathological processes. So far, it is clear that plenty of fluorescent probes have been prepared, but most of them still have some disadvantages and is unsuitable for imaging in vivo. Herein, a novel fluorecent probe named HPQ-BI-CO is synthesized and used to detect CO. In this probe, the benindole group is introduced to extend the wavelength. This probe possess a variety of advantages such as long emission wavelength, high sensitivity and selectivity, and rapid response capability. Moreover, the probe possesses a low cytotoxicity, which make it apply in the imaging of exogenous and endogenous CO in living 4T1 cells. Furthermore, this probe can be utilized as a promising tool for fluorescent bioimaging of CO in vivo like zebrafish.

Abstract:Six coumarin-based fluorescent probes were designed and synthesized for the detection of sulfite. The probes were constructed based on "fluorophore-receptor/linker-steric group" mode with N,N-diethyl-7-aminocoumarin (DEA) or 2,3,5,6-1H,4H-tetrahydro-quinolazino-(9,9a,1-gh)coumarin (THQ) as the fluorophore, 2-naphthyl ketone (N) or phenyl ketone (B) as the steric group and vinyl (1) or 1,3-butadienyl (2) as the receptor/linker. The effects of the fluorophore, the linker and the steric group on the photophysical properties and sensing performance were systematically studied. Among all the probes, DEA-N2 exhibited the most excellent spectral responses toward sulfite: upon addition of sulfite, the absorption and emission maxima shifted from 488 nm and 684 nm to 405 nm and 484 nm, respectively. I 484 /I 688 displayed good linear relationship with sulfite concentration in the range of 3-50 μM, with a detection limit of 0.31 μM. The experimental results demonstrate that DEA-N2 has potential in ratiometric fluorescent imaging of mitochondrial sulfite in living cells.

Abstract:A novel photoconvertible fluorescent probe, which can be activated by intracellular thiols, has been synthesized. Such a molecular probe comprises three parts: a 7-aminocoumarin phototrigger, a thiol-removable energy acceptor, and a caged fluorescein scaffold with intracellular thiols reactivity as the fluorescent reporter. Extracellularly, the energy acceptor blocks the emission of the coumarin that regulates the photocleavage and photoactivation of the fluorescein. Intracelluarly, the high concentration of thiols releases the energy acceptor, thus activating the S1 state of the phototrigger, which emits coumarin blue fluorescence for pre-visualization and liberates the caged green-fluorescent fluorescein to highlight the specific cell upon illumination. Compared to traditional photoactivated organic dyes, the intracellular thiols activated probe requires double activations: one by intracellular thiols and the other by light activation. The dual activations restrict fluorescence precisely inside live cells and at the particular spatial region of light activation, thus a probe with precise spatial accuracy in live cells.

Abstract:Calcium is an essential element for living systems and involved in numerous intracellular signaling cascades. In this work, a novel fluorescent probe based on a coumarin and pyrazoline scaffold was designed and synthesized for the detection of Ca2+ by a photoinduced electron transfer (PET) strategy. The probe showed good selectivity for Ca2+ compared to other substances in ethanol with a low detection limit (1.37 × 10−7 M). Meanwhile, following the addition of Ca2+ ions, the probe showed an obvious "turn on" with a rapid response of less than 5 s and 1:1 binding with an association constant of 7.97 × 103 M−1. Furthermore, the fluorescence imaging of the probe was also performed in the presence of Ca2+ in living A549 cells.
