Abstract:The intricate part of the non-covalent associates in numerous realms covering from chemistry, biology, catalysis, material science, to medicinal chemistry inspires a continuous motivation towards the discovery of new synthons that stabilize the supramolecular systems. In this work the co-crystallization between 2-amino-4,6-dimethoxypyrimidine (admpym) and eight organic acids resulted in the manufacturing of eight new crystalline salts, which were further fully characterized via the single crystal X-ray diffraction (SCXRD), spectroscopic (FT-IR) and elemental analysis methods. Their melting points were also measured. All the fabricated salts exhibited the identical stoichiometry regarding the number of the acidic unit and the admpym, namely with the 1:1 ratio at the monacids and 2 : 1 ratio in the diacids. Among the eight crystals, the H-shift from the acid to pyrimidine ring N atom has occurred, yet the NH 2 group was not protonated ever. In all the crystals, the respective molecular fragments are permanently tethered to each other via the energetic H-bonds as the N pyrimidinium -O phenolate/carboxylate type. The various weaker intermolecular linkages, embodying CH···O/CH 3 ···O, CH···CH, CH 3 ···N, CH···Br, CH 3 ···CH, CH 3 ···Cl, Cl···O, CH···π/CH 3 ···π, C···π, O···π and π···π also contributed to the stabilization of the crystal structures in the respective conglomerates. The percentage contribution of the significant non-covalent contacts were calculated via the Hirshfeld surface analysis. The R 2 2 (8) graph was enclosed in all the systems when an acid pairs with an admpym. The job ensures that there engender both principle contact modes between the admpym and the acids, which appear in the relative higher frequency, linear heterotetramer (LHT), and cyclic heterotetramer (CHT). LHT is dominant as it appears in 5 cases, CHT in 2 ones, with the rest one structure building a rare heterodimer. The heterotrimer (HT) was never occurred here.

Abstract:The complex role of the non-covalent contacts in a wide kind of areas ranging from chemistry, biology, catalysis, material science, to medicinal chemistry has aroused a continuous motivation towards the discovery of new synthons that build the supramolecular systems. In this job, the co-crystallization between triphenylmethylamine, 3-nitrophthalic acid, and its position isomer 5-nitroisophthalic acid resulted in the manufacturing of two new anhydrous crystalline salts, which were further fully characterized via the single crystal X-ray diffraction (SCXRD) in detail, spectroscopic (FT-IR) and elemental analysis methods. Their melting points were also measured. Both the fabricated salts exhibited the identical M stoichiometry of 1:1 ratio, forming the hydrogenphthalate salt. In both the salts, the respective molecular fragments are permanently tethered with each other via the energetic hydrogen bonds as the N triphenylmethylaminium -O carboxylate type. The other extensive non-covalent associates of CH-O, CH-π, π-π, CH-CH, and O-π also act great functions in the space extension in the relevant conglomerates. Analysis by the Hirshfeld surface affords extra insights into the prevalence of the various short contacts in the crystal structure. The synthons R 1 2 (7), R 2 1 (7), R 2 2 (7), R 2 2 (9), R 3 2 (8), R 3 2 (12), R 3 3 (10), R 3 3 (13), and R 4 2 (8) were noted at the salts. For the combination of the classical hydrogen bonds plus the various non-covalent contacts, both salts took the 3D net graphs.

Preparation and Structure Analysis of Non-covalent Interactions Mediated 2D-3D Supramolecular Adducts from 6-methylnicotinamide and Carboxylic Acids

Abstract:6-methylnicotinamide was utilized to create co-crystal/salts for the first time. By introducing it into the co-crystallization system based on different carboxylic acids of mono- to dicarboxylic acids, a total of six novel adducts including one cocrystal (6) and five salts (1–5) were engendered. They have been featured by SCXRD, IR and EA, their m.p. were also surveyed. Their structural and supramolecular portions are fully examined. The outcome unveils that in all salts the pyridine N is the only protonated location. The crystal piling is stitched by the strong NH···O, OH···N and OH···O H-bonds. The CONH2 dimerized in 5 and 6 with the help of twain NH···O H-bonds. Deep view of the crystal packing opened that a distinct set of subsidiary CH2C, CHN, CH···O/CH3···O, CH-S, OC, OO, O···π, π···C, S-π, CH···π/CH2-π/CH3-π and π···π connections conduce to the steadiness and growth of the overall 2D-3D structures. For the exquisite equilibrium of the various non-covalent links there embedded the homo/hetero supramolecular synthons or both, the universal R22(7)/R22(8) graphs have been the top encountered ones. The present findings lay the seat for burgeoning new solids of 6-methylnicotinamide, and furnish some new views into its co-crystals/salts with the carboxylic acids.

Abstract:A new cocrystal (ACP − SA) of 4 − acetylpyridine (ACP) with succinic acid (SA) has been synthesized by slow evaporation method and characterized. The single crystal X-ray diffraction technique confirmed the coexistence of ACP and SA molecules in a 1:0.5 ratio in an asymmetric unit. The ACP − SA crystallizes in a monoclinic space group P 2 1 / n , with a = 9.0185(14) Å, b = 8.8955(14) Å, c = 10.9044(18) Å, β = 93.017(3)°, V = 873.6(2) Å 3 , Z = 4. Classical O − H⋯N and non classical C − H⋯O hydrogen bonds aid in stabilizing the supramolecular architecture of ACP − SA by creating D (2) and R44(22) motifs. In addition, density functional theory (DFT) computations were performed to get insights into the structure and intermolecular interactions of ACP − SA cocrystal. The TGA/DSC thermogram revealed that the cocrystal began to degrade at temperatures over 78 °C and retained its crystallinity up to 106 °C. The nature of the interactions and their contribution to the crystal packing is quantitatively measured using Quantum topological atoms in molecule (QTAIM), 3D Hirshfeld surfaces analysis and 2D fingerprint plots. The maximum percentage contribution of H⋯H (40%), O⋯H (32%) and C⋯H (11.6%) interactions were identified from Hirshfeld surfaces analysis. With the aid of both experimental and computational methods, the crystal packing and hydrogen bonding were examined.

Abstract:The crystal structures and the supramolecular architectures of a series of novel salts originating from 2,4-diaminopyrimidine and four different chain dicarboxylic acids are reported. For this purpose, 2,4-diaminopyrimidin-1-ium 2,2′-thio(acetic)acetate (1), 2,4-diaminopyrimidin-1-ium monoglutarate (2), 2,4-diaminopyrimidin-1-ium 3,3′-dithio(propionic)propionate (3) and 2,4-diaminopyrimidin-1-ium suberate (4) were synthesized in good to high yields from 2,4-diaminopyrimidine and appropriate dicarboxylic acids (2,2′-thiodiacetic acid, glutaric acid, 3,3′-dithiodipropionic acid and suberic acid, respectively). Each of the compounds were formed as a monohydrate and compound 4 additionally co-crystallized with the suberic acid molecule. Despite the similar structures of compounds 1 and 2 as well as 3 and 4, subtle but important differences are observed in their crystal packing and H-bonding patterns, especially between 3 and 4. Supramolecular self-assemblies can be distinguished through different interactions considering anions, leading to diverse H-bonding motifs, which also include sulphur atoms in 1 and 3, at the upper level of supramolecular architecture. Notably, the basic motif is always the same—2,4-diaminopyrimidine-based homosynthon R22(8) via N-H∙∙∙N interactions. The impact of diverse types of intermolecular interactions was evaluated by Hirshfeld analysis, while the propensity of atom pairs of elements to build interactions was calculated using enrichment ratios. Although compounds 1 and 3 contain S-atoms, the percentage of S-derived interactions is rather low. In 1, the contribution of S∙∙∙H/H∙∙∙S, S∙∙∙C/C∙∙∙S, S∙∙∙N/N∙∙∙S intermolecular contacts is 5.7%. In 2, the contribution of S∙∙∙H/H∙∙∙S accounts for only 0.6%.

Abstract:We have synthesized and thoroughly analyzed many intermolecular interactions present in (E)-1-(((3-nitrophenyl)amino)methylene)naphthalen-2(1H)-one ( a ), (E)-5-(diethylamino)-2-(((3-nitrophenyl)imino)methyl)phenol( b ) and (E)-2-(((3-nitrophenyl)imino)methyl)phenol ( c ). Compounds a (keto-amine tautomeric form) and b, which have one molecule in the asymmetric unit, crystallize in the monoclinic crystal system with space groups P21/n and C2/c, respectively. Compound C, on the other hand, crystallizes in an orthorhombic crystal structure with the space group Pca21. The structural analysis showed that the construction of ring motifs to stabilize the crystal packing involves several interactions, including pi stacking interactions, NH...O, and CH...O interactions. Hirshfeld surface analysis has been used to examine their solid-state structures, including the evaluation of several energy frameworks. The results show that hydrogen bonds play a major role in the formation of the molecular sheets, and that electrostatic energy contribution dominates stability. Using fingerprint plots, it was possible to examine the variations and commonalities in the target compounds' relative contributions from noncovalent interactions. In addition, B3LYP hybrid functional with 6- 311 G (d, p) level basis set was used in the density functional theory computations to optimize the structural coordinates. Through the examination of the molecular electrostatic potential surface, the chemically active areas of the target imine molecules were located. Additionally, using the internal ribosome entry site of the Hepatitis C virus ED42 (PDB-ID 3T4B), molecular docking of the title compounds was performed based on PASS analysis and literature findings.

Abstract:A series of novel salts based on aromatic polyamines and 2,3-pyrazinedicarboxylic acid, such as C10H12N6O5 (1), C10H9ClN6O4 (2), C11H10N8O4 (3), and C14H17N16O5.5 (4) or 3,4-thiophenedicarboxylic acid, such as C10H10N4O4S (5), C10H9ClN4O4S (6), and C10H10N4O4S2 (7), were synthesized and characterized by single-crystal X-ray diffraction. All compounds crystallize in a monoclinic space group. The structure was subjected to complex Hirshfeld surface analysis, molecular electrostatic potential, enrichment ratio, and energy framework calculations. The influence of different cations on the packing of 3-carboxypyrazine-2-carboxylate and 4-carboxythiophene-3-carboxylate anions in the crystal lattice was studied. O...H/H...O interactions are the main contributor in all crystals. In addition, in a series of pyrazine-containing structures, N(C)...H/H...N(C) interactions have relevance, while in a series of thiophene-based compounds, C...H/H...C and S...H(O)/H(O)...S. In addition, Cl-based interactions are observed in compound 2. According to the enrichment ratio calculations, O...H/H...O and C...C are the most preferable interactions in all structures. The energy frameworks are dominated by the dispersive contribution, only in compound 3 is the electrostatic term dominant. The analyzed structures reveal intra- and intermolecular recurrent supramolecular synthons. In both series of crystals, the robust H-bonded centrosymmetric dimer R22(8) as homo- or as heterosynthon (in compounds 2, 3, 6, and 7) and the intramolecular synthon S(7) generated by O-H...O interactions (in compounds 2, 6, and 7) are present. The supramolecular patterns formed by π...π (C...C) and C-O(Cl,S)...C are also noticeable. Notably, a dual synthon linking the supramolecular chain via π...π interactions and the homosynthon R22(8) via N-H...N interactions is visible in both series of new salts. A library of H-bonding motifs at diverse levels of supramolecular architecture is provided. We extended the analysis of intramolecular H-bonding motifs to similar structures deposited in the Cambridge Structural Database. Another important feature is the existence of an intramolecular O...H...O bridge between two neighboring carboxylic groups as substituents in anions in compounds 3 and 5. In this context, we performed quantum theory of atoms-in-molecule calculations to reveal more details.

Abstract:A series of novel salts of heterocyclic polyamines with 5-sulfosalicylic acid (C4H7N4+)(C7H5O6S−)∙2(H2O) (1), (C4H6ClN4+)(C7H5O6S−)∙H2O (2), (C5H8N3+)(C7H5O6S−)∙H2O (3), (C5H7N6+)(C7H5O6S−)∙H2O (4), (C6H14N22+)(C7H4O6S2−)∙H2O (5), and (C14H19N2+)(C7H5O6S−) (6) have been successfully synthesized. Their crystal structures have been determined by single-crystal X-ray diffraction. Overall, compounds adopt a layered structure with aminium cations and 5-sulfosalicylic anions linked via water molecules. The solid-state architectures of these compounds are dominated by O(N,H)-H⋯O and N-H⋯N hydrogen bonds and stabilized by weak interconnects. C-Cl⋯π and S-O⋯π interactions, apart from π⋯π and C-H(O)⋯π, were reported. Diverse approaches were used to study the effect of substituents in the polyamines in solid-state arrangement. A Hirshfeld surface analysis, with associated 3D Hirshfeld surface maps and 2D fingerprint plots, molecular electrostatic potential, and energy frameworks were used to comprehensively investigate the nature and hierarchy of non-covalent interactions and inspect supramolecular differences. The contact enrichment ratio calculations provided deeper insight into the propensity of interconnects to influence crystal packing. The evaluation of the effects of H-bonding synthons resulting from different substituents in the polyamines on self-assemblies is also presented. In the context of crystal engineering, a specific intramolecular synthon via O-H⋯O observed in nearly all crystals can be employed in the pseudo-cyclic replacement strategy in the design of new molecules.

Abstract:The current work is about the modification of primary amine functionalized drugs, pyrimethamine and 4-amino- N -(2,3-dihydrothiazol-2-yl)benzenesulfonamide, via condensation reaction with 2-hydroxy-1-naphthaldehyde to produce new organic zwitterionic compounds ( E )-1-(((4-( N -(2,3-dihydrothiazol-2-yl)sulfamoyl)phenyl)iminio)methyl)naphthalen-2-olate ( DSPIN ) and ( E )-1-(((4-amino-5-(4-chlorophenyl)-6-ethylpyrimidin-2-yl)iminio)methyl)naphthalen-2-olate ( ACPIN ) in methanol as a solvent. The crystal structures of both compounds were confirmed to be imine-based zwitterionic products via single-crystal X-ray diffraction (SC-XRD) analysis which indicated that the stabilization of both crystalline compounds is achieved via various noncovalent interactions. The supramolecular assembly in terms of noncovalent interactions was explored by the Hirshfeld surface analysis. Void analysis was carried out to predict the crystal mechanical response. Compound geometries calculated in the DFT (Density Functional Theory) study showed reasonably good agreement with the experimentally determined structural parameters. Frontier molecular orbital (FMO) analysis showed that the DSPIN HOMO/LUMO gap is by 0.15 eV smaller than the ACPIN HOMO/LUMO gap due to some destabilization of the DSPIN HOMO and some stabilization of its LUMO. The results of the charge analysis implied formation of intramolecular hydrogen bonds and suggested formation of intermolecular hydrogen bonding and dipole–dipole and dispersion interactions.

Abstract:Quaternary salts d -π- A + X - having a large π-conjugated system ( I ) 2-((Z)-1-cyano-2-[4-(dimethylaminophenyl]ethenyl)-1-methylpyridin-1-ium iodide, ( II ) 3-((Z)-1-cyano-2-[4-(dimethylaminophenyl]ethenyl)-1-methylpyridin-1-ium iodide, and ( III ) 4-((Z)-1-cyano-2 iodide -[4-(dimethylaminophenyl]ethenyl)-1-methylpyridin-1-ium iodide, were synthesized straightforwardly and easily and characterized by single-crystal X-ray diffraction, IR, UV–Vis, 1HNMR spectroscopy, DSC, and TGA. Quantum chemical calculations were performed to obtain electronic structure by using DFT. The salts I - III exhibited a hyperchromic shift and a strong bathochromic effect of 100 nm compared to the precursors, which indicates charge transfer (CT) due to ionic interactions. The Hirshfeld, MEPS, QTAIM, and NCI-RDG analyses showed that the -CN group and the counterion I (-) atom play major roles in the intermolecular interaction for crystal packing. The significance of non-covalent interactions for the stability of the molecules in their solid state is also revealed by natural bond orbital analysis.

Abstract:The structural identity of multi-component crystals as a salt or cocrystal is dictated by the proton transfer state between the molecular components. In pharmaceutical drugs, solid-state forms such as salt or cocrystal can have significantly distinct stability, dissolution and solubility profiles. The accurate location of proton positions is a formidable task using conventional X-ray crystallography as the atomic scattering factors are based on spherical electron density models. Herein, we demonstrate that the X-ray quantum crystallographic technique of Hirshfeld Atom Refinement (HAR) based on aspherical atomic scattering factors can be effectively employed to resolve this riddle. Our HAR models accurately located the proton positions, thus distinguishing salts, co-crystals, and continuum crystal structures, which are substantiated by the N-1s binding energies from X-ray photoelectron spectroscopy (XPS) corresponding to the base components in a series of crystals. The quantum crystallography models reveal the subtle features of electron localization and bonding around the double well potential in the intermolecular proton-transfer regions in these crystals.

Efficient Synthesis of Imine-Carboxylic Acid Functionalized Compounds: Single Crystal, Hirshfeld Surface and Quantum Chemical Exploration

Abstract:Two aminobenzoic acid based crystalline imines (HMBA and DHBA) were synthesized through a condensation reaction of 4-aminobenzoic acid and substituted benzaldehydes. Single-crystal X-ray diffraction was employed for the determination of structures of prepared Schiff bases. The stability of super molecular structures of both molecules was achieved by intramolecular H-bonding accompanied by strong, as well as comparatively weak, intermolecular attractive forces. The comparative analysis of the non-covalent forces in HMBA and DHBA was performed by Hirshfeld surface analysis and an interaction energy study between the molecular pairs. Along with the synthesis, quantum chemical calculations were also accomplished at M06/6-311G (d, p) functional of density functional theory (DFT). The frontier molecular orbitals (FMOs), molecular electrostatic potential (MEP), natural bond orbitals (NBOs), global reactivity parameters (GRPs) and natural population (NPA) analyses were also carried out. The findings of FMOs found that Egap for HMBA was examined to be smaller (3.477 eV) than that of DHBA (3.7933 eV), which indicated a greater charge transference rate in HMBA. Further, the NBO analysis showed the efficient intramolecular charge transfer (ICT), as studied by Hirshfeld surface analysis.

Abstract:Cocrystallization of the widely available simplest voluminous primary amine, tert‑butylamine, with a set of acids got a total of 14 salts, which have been featured by XRD technique, Infrared spectra and elemental analysis. Their melting points were also surveyed. Their structural and supramolecular aspects are analyzed in detail. All compounds are ionic, with H-transferring to the NH2 of the tert‑butylamine. The results unveil that among all the studied crystals the NH2 in L are protonated when the acids are deprotonated, the crystal packing is interpreted via the strong NH⋯O H-bond from the NH3 + and the deprotonated acids. Apart from the NH⋯O H-bond, the OH···O H-bonds were also existed at 6 and 12. Salts 1 - 2 had the NH⋯S H-bond. The NH···Cl H-bond existed in 7. Further view of the crystal packing told that a different array of additional CH3···S, O···Cl, S···S, CH3···C, CH3···CH3, CH···O/CH3···O, CH2···Cl, CH···N, CH3···π and π···π contacts help the stabilization and expansion of the whole structures. For the synergism of the various nonbonding contacts these structures had the hetero supramolecular synthons. Some classical synthons of R2 2(8), R4 2(8), R4 3(10) and R4 4(12), commonly present in solids of organic acids with amine, were again shown to be created in making these H-bonding nets.

Abstract:An unprecedented non‐AAIPEX protocol has been developed to access diverse monosubstituted cationic polycyclic heteroaromatic compounds (cPHACs) from the readily available azolium salts and phenacyl bromides via Ru(II)‐catalyzed tandem annulation cum aromatization. This atom‐economic protocol executes a range of intermediate steps e.g. double C‐H activation, nucleophilic addition, annulation, and dehydration cum aromatization in one‐pot manner under the generation of H2O as the sole byproduct. Moreover, the systematic tunability of photo‐physical and electrochemical properties of these new class of cPHACs can be authenticated from the DFT calculated frontier molecular orbital energies that might be beneficial for their potential applications in optoelectronics and DNA intercalation.

Abstract:While X-ray crystallography routinely provides structural characterization of kinetically stable pre-catalysts and intermediates, elucidation of the structures of transient reactive intermediates, which are intimately engaged in bond-breaking and -making during catalysis, is generally not possible. Here, we demonstrate in crystallo synthesis of Rh2 nitrenoids that participate in catalytic C–H amination, and we characterize these transient intermediates as triplet adducts of Rh2. Further, we observe the impact of coordinating substrate, which is present in excess during catalysis, on the structure of transient Rh2 nitrenoids. Abstract:The molecule of anti-epileptic drug lamotrigine [LAM; 3,5-diamino-6-(2,3-dichlorophenyl)-1,2,4-triazine] is capable of the formation of multicomponent solids. Such an enhanced tendency is related to the diverse functionalities of the LAM chemical groups able to form hydrogen bonds. Two robust synthons are recognized in the supramolecular structure of LAM itself formed via N—H...N hydrogen bond: homosynthon, so-called aminopyridine dimer or synthon 1 [R22(8)] and larger homosynthon 2 [R32(8)]. The synthetic procedures for a new hydrate and 11 solvates of LAM (in the series: with acetone, ethanol: two polymorphs: form I and form II, 2-propanol, n-butanol, tert-butanol, n-pentanol, benzonitrile, acetonitrile, DMSO and dioxane) were performed. The comparative solid state structural analysis of a new hydrate and 11 solvates of LAM has been undertaken in order to establish robustness of the supramolecular synthons 1 and 2 found in the crystal structure of LAM itself as well as LAM susceptibility to build methodical solid state supramolecular architecture in the given competitive surrounding of potential hydrogen bonds. The aminopyridine dimer homosynthon 1 [R22(8)] has been switched from para-para (P-P) topology to ortho-ortho (O-O) topology in all crystal structures, except in LAM:n-pentanol:water solvate where it remains P-P. Homosynthon 2 [R32(8)] of the LAM crystal structure imitates in the LAM solvates as a heterosynthon by replacing the triazine nitrogen proton acceptor atoms of LAM with the proton acceptors of solvates molecules.

Abstract:In this study, we present a series of Zn(II) mono- and Cu(II) binuclear salen complexes synthesized and assessed for their effectiveness in the Cα-alkylation reaction. Through systematic experimentation, it was observed that the introduction of a methoxy group at position 3 of the phenyl group in the salicylidene ligand led to a notable enhancement in asymmetric yield, while an allyl group reduced yield. Computational DFT calculations supported the involvement of the binuclear complex in the transition state of the reaction, elucidating the underlying mechanisms governing the observed catalytic behavior. A newly synthesized binuclear complex exhibited significantly higher catalytic activity compared to its mononuclear counterpart which could potentially be explained by increased intramolecular rigidity. This comprehensive investigation not only advances our understanding of structure-activity relationships in chiral salen complexes but also provides valuable insights for the rational design and optimization of catalysts for the asymmetric Cα-alkylation reaction.

Abstract:Three new compounds, namely, 5-fluorocytosine–urea (2/1), 2C 4 H 4 FN 3 O·CH 4 N 2 O, (I), 5-fluorocytosine–5-fluorocytosinium 3,5-dinitrosalicylate–water (2/1/1), 2C 4 H 4 FN 3 O·C 4 H 5 FN 3 O + ·C 7 H 2 N 2 O 7 − ·H 2 O, (II), and 2-amino-4-chloro-6-methylpyrimidine–6-chloronicotinic acid (1/1), C 6 H 4 ClNO 2 ·C 5 H 6 ClN 3 , (III), have been synthesized and characterized by single-crystal X-ray diffraction. In compound (I), 5-fluorocytosine (5FC) molecules A and B form two different homosynthons [ R 2 2 (8) ring motif], one formed via N—H...O hydrogen bonds and the second via N—H...N hydrogen bonds. In addition to this interaction, a sequence of fused-ring motifs [ R 2 1 (6), R 3 3 (8), R 2 2 (8), R 4 3 (10) and R 2 2 (8)] are formed, generating a supramolecular ladder-like hydrogen-bonded pattern. In compound (II), 5FC and 5-fluorocytosinium are linked by triple hydrogen bonds, generating two fused-ring motifs [ R 2 2 (8)]. The neutral 5FC and protonated 5-fluorocytosinum cation form a dimeric synthon [ R 2 2 (8) ring motif] via N—H...O and N—H...N hydrogen bonds. On either side of the dimeric synthon, the neutral 5FC, 5-fluorocytosinium cation, 3,5-dinitrosalicylate anion and water molecule are hydrogen bonded through N—H...O, N—H...N, N—H...O W and O W —H W ...O hydrogen bonds, forming a large ring motif [ R 10 10 (56)], leading to a three-dimensional supramolecular network. In compound (III), 2-amino-4-chloro-6-methylpyrimidine (ACP) interacts with the carboxylic acid group of 6-chloronicotinic acid via N—H...O and O—H...O hydrogen bonds, generating an R 2 2 (8) primary ring motif. Furthermore, the ACP molecules form a base pair via N—H...N hydrogen bonds. The primary motif and base pair combine to form tetrameric units, which are further connected by Cl...Cl interactions. In addition to this hydrogen-bonding interaction, compounds (I) and (III) are further enriched by π–π stacking interactions.

Abstract:Molecules bearing carboxylic acid, amide, and hydroxyl groups are ubiquitous in crystal engineering, where robust hydrogen-bonded synthons centred on these functionalities enable reliable crystal structure design. We now show that halogen bonding to the carbon π-system of such molecules, traditionally ignored in crystal engineering, permits the recognition and directional assembly of the resulting hydrogen-bonded structural subunits, leaving the archetypal hydrogen-bonded ring, ladder, and chain homosynthons intact, but repositioned in space. When applied to heteromolecular synthons, this enables rearranging more complex hydrogen-bonded motifs and the evolution of binary cocrystals into ternary ones through "latent" carbon-based recognition sites, demonstrating a rational approach to build higher-order solid-state supramolecular assemblies.
