Abstract:Organic framework materials have shown increasingly promising applications in biomedicine, such as drug delivery and release. In this work, we first synthesized a new hydroxyl-containing imine-linked two-dimensional covalent organic framework (COF) through solvothermal synthesis. Then, the imine group was converted into a benzoxazine group using a cyclization reaction. The results show that the postsynthetic modification did not change the basic framework of the original COF and did not affect the basic properties of the original COF. At the same time, the new benzoxazine group obtained by cyclization gave the COF good antibacterial activity against Escherichia coli and Staphylococcus aureus. The COF efficiency after cyclization was improved, and its antibacterial activity against both bacteria was over 90% compared with the imine-linked COF. Moreover, the benzoxazine-linked COF crystal structure and pore structures were retained, leaving the drug delivery and release functions unaffected. A benzoxazinelinked COF has never been reported because it cannot be synthesized by a direct reaction method. The work in this paper shows that the COFs that cannot be directly synthesized can be obtained through specific postsynthetic modification reactions. This means that more functional COFs can be obtained based on existing COFs, and the diversity of COF types and their potential applications can be further enriched and expanded. Abstract:Covalent organic frameworks (COFs) are an emerging class of crystalline porous polymers in which organic building blocks are covalently and topologically linked to form extended crystalline polygon structures, constituting a new platform for designing π-electronic porous materials. However, COFs are currently synthesised by a few chemical reactions, limiting the access to and exploration of new structures and properties. The development of new reaction systems that avoid such limitations to expand structural diversity is highly desired. Here we report that COFs can be synthesised via a double-stage connection that polymerises various different building blocks into crystalline polygon architectures, leading to the development of a new type of COFs with enhanced structural complexity and diversity. We show that the double-stage approach not only controls the sequence of building blocks but also allows fine engineering of pore size and shape. This strategy is widely applicable to different polymerisation systems to yield hexagonal, tetragonal and rhombus COFs with predesigned pores and π-arrays.

Abstract:In this study, a novel imine linkage covalent organic framework (COFs) was synthesized via condensation of terephthaldehyde with diethylenetriamine under solvothermal conditions to afford the corresponding COFTDETA which exhibited high thermal and chemical stability. The structure of COFTDETA was characterized by Ultraviolet-Visible spectral measurements (UV/V), Brunauer-Emmett-Teller (BET) surface area, scanning electron microscopy (SEM), Fourier transforms Infrared spectroscopy (FTIR), Thermogravimetric analyzer (TGA), Nuclear magnetic resonance (NMR) and X-ray diffraction (XRD. COFTDETA showed higher sensitivity as an antibacterial agent than tobramycin drug against different antibacterial species. The higher sensitivity is due to higher conjugation, presence of imine linkage (C=N), and free amine (NH). These experimental results were confirmed through molecular docking simulation with different kinds of proteins. Moreover, COFTDETA was optimized utilizing DFT/B3LYP/6-311(G) and HF basis set to elucidate their physical descriptor, determination of its HOMO-LUMO band gap, ESP, and (MEP) which were more correlated with biological results. Furthermore, the comparative study between experimental and theoretical results was closely related to DFT/B3LYP/6-311(G) basis set according to their 1HNMR, 13CNMR, and FT-IR.

Abstract:As the consumption of antibiotics rises, they have generated some negative impacts on organisms and the environment because they are often unable to be effectively degraded, and seeking effective detection methods is currently a challenge. Covalent–organic frameworks (COFs) are new types of crystalline porous crystals created based on the strong covalent interactions between blocked monomers, and COFs demonstrate great potential in the detection of antibiotics from aqueous solutions because of their large surface area, adjustable porosity, recyclability, and predictable structure. This review aims to present state-of-the-art insights into COFs (properties, classification, synthesis methods, and functionalization). The key mechanisms for the detection of antibiotics and the application performance of COFs in the detection of antibiotics from water are also discussed, followed by the challenges and opportunities for COFs in future research.

Abstract:Techniques beyond crystal engineering are critical for manufacturing covalent organic frameworks (COFs) and to explore them for advanced applications. However, COFs are normally obtained as insoluble, unmeltable, and thus nonprocessible microcrystalline powders. Therefore, it is a significant challenge to implement COFs into larger architectures and structural control on different length scales. Herein, a facile strategy is presented to prepare flexible COF nanofiber membranes by in-situ growth of COFs on polyacrylonitrile (PAN) nanofiber substrates via a reversible polycondensation-termination approach. The obtained PAN@COF nanofiber membranes with vertically aligned COF nanoplates combine a large functional surface with efficient mass transport, thus making it a promising adsorbent, for example, for water purification. The antibiotic pollutant ofloxacin (OFX) is removed from water with a superior absorption capacity of ≈236 mg g-1 and removal efficiency as high as 98%. The here presented in-situ growth of COFs on nanofiber membranes can be extended to various Schiff base-derived COF materials with different compositions, providing a highly efficient way to construct flexible COF-based membranes for several applications.
Targeted Synthesis of a 3D Covalent Organic Framework with Fjh Topology Exhibiting High Photoluminescence: A Robust Platform for Sensitive Detection of Furantoin

Abstract:The inappropriate use of antibiotics poses significant risks to both the environment and human health, necessitating the development of accurate and sensitive detection methods. Fluorescence detection, particularly utilizing three-dimensional (3D) covalent organic frameworks (COFs), stands out as a promising avenue. Unlike two-dimensional COFs, 3D COFs can mitigate fluorescence quenching resulting from non-radiative energy loss, primarily attributed to π-π interlayer stacking. In this study, by carefully selecting and designing the building blocks, we synthesized a 3D COF named PyTTA-TTFB-COF with fjh topology and remarkable photoluminescence. This was achieved through the combination of a 4-connected square chromophore, 1,3,6,8-Tetrakis(4-aminophenyl) pyrene, with a 3-connected triangle, 1,3,5-trimethyl-2,4,6-tris(4-formylphenyl) benzene, possessing a specific stereo conformation. Significantly, PyTTA-TTFB-COF exhibits a high photoluminescence quantum yield, notably reaching 60.1% in acetonitrile. Moreover, it displays substantial fluorescence quenching when exposed to furantoin, surpassing the performance observed with other antibiotics. Competitive experiments have confirmed that PyTTA-TTFB-COF can serve as a specific fluorescent probe for the interference-free detection of furantoin.

Abstract:Abstract The topology type and the functionalization of pores play an important role in regulating the performance of covalent organic frameworks. Herein, we designed and synthesized the covalent organic framework with hetero-environmental pores using predesigned asymmetrical dialdehyde monomer. According to the results of structural characterization, crystallinity investigation, and theoretical calculation, the hetero-environmental pores of the obtained framework are regarded as the alternant arrangement. The distinctive hetero pore structure leads the designed material to show more advantages as compared with control materials in loading both hydrophobic and hydrophilic antibiotics for wound healing. This dual-antibiotic strategy can expand the antibacterial range as compared with the single antibiotic one, and reduce the generation of drug resistance. In summary, this strategy for designing covalent organic frameworks with hetero-environmental pores can extend the structural variety and provide a pathway for improving the practical application performance of these materials.

Abstract:The development of three-dimensional (3D) architectures in COFs has been considered important owing to their prospective ad-vantages in high surface areas and plentiful active sites. Herein, we report the first example of a 3D COF with zeolitic network, namely the zeolitic organic framework (ZOF). By combining two kinds of tetrahedral building blocks with fixed or relatively free bond angles, ZOF-1 with the zeolitic crb net has been successfully synthesized. Its structure was determined by the single-crystal 3D electron dif-fraction technique. Remarkably, ZOF-1 shows high chemical stabil-ity, large pore size (up to 16 Å), and excellent specific surface area (~ 2785 m2/g), which is superior to its analogues with the same network, including traditional aluminosilicate zeolites and zeolitic imidazole frameworks. This study thus opens a new avenue to construct 3D COFs with zeolitic topologies and promotes the struc-tural diversity and practical applications of COF materials.

Abstract:Covalent organic frameworks (COFs) have attracted considerable interest over the past few years, but the available type of monomers is still limited. Herein, two novel imine-linked COFs (TaDAP and TaDA) were constructed by the condensation of 1,3,5-tris-(4-aminophenyl) triazine with 2,6-diformylpyridine (forming TaDAP) and 1,3-benzenedialdehyde (forming TaDA) under solvothermal conditions. The structures of the COFs were confirmed and characterized by solid state C-13 NMR, FT-IR, PXRD, TGA, SEM, TEM and nitrogen sorption isotherms. They appear as regular spherical particles and possess excellent thermal stability. It was also found that their dispersions emit strong fluorescence, and display quick response to certain metal ions. In particular, TaDAP can detect Fe3+ with sensitivity and selectivity. Moreover, both the COFs present excellent catalytic activity towards the Knoevenagel reaction. Accordingly, this study not only provides a new way to enrich the family of COFs, but also develops their potential applications in metal ion detection.

Abstract:Covalent organic frameworks (COFs) are an emerging class of porous crystalline polymers with well-defined structures and tunable functionalities, which have fascinating applications in a wide range of fields. However, the synthetic procedures of COFs are mainly confined to the solvothermal synthesis method which usually requires harsh experimental conditions. In this work, an effective solvent-free synthesis method to construct a series of two-dimensional (2D) COFs including imine-linked, azine-linked, β-ketoenamine-linked, which avoids the complicated solvent screening process and most of the disadvantages of solvothermal methods is developed. The crystallinity and porosity of these COFs are comparable to those prepared by traditional solvothermal routes. What's more, the advantages of the solvent-free method enable the production of gram-scale of those 2D COFs through a one-pot reaction, demonstrating high industrial application potentials.

Abstract:Covalent organic frameworks (COFs) are a new kind of functional solids featured by several fantastic structural characteristics, including a great diversity of building units and cross-linking patterns, precise integration of building blocks, and adjustable topology of porous architecture. In addition to the above features, some COF samples are constructed with high-density conjugated fragments, which have unique potential advantages in fluorescence imaging, and thus may have great potential applications in bioimaging. Herein, this article summarizes the recent progress in the design and preparation of fluorescent covalent organic frameworks. We conduct a systemic correlation investigation between the structural qualities of COF networks and biological sensors. Finally, the significant advantages, major challenges, and future opportunities of fluorescent covalent organic frameworks are discussed for the development of next-generation porous materials for sensing applications.

Abstract:Porous materials such as covalent organic frameworks (COFs) are good candidates for molecular sieves due to the chemical diversity of their building blocks, which allows fine-tuning of their chemical and physical properties by design. Tailored synthesis of inherently functional building blocks can generate framework materials with chemoresponsivity, leading to controllable functionalities such as switchable sorption and separation. Herein, we demonstrate a chemoselective, salicylideneanilines-based COF (SA-COF), which undergoes solvent-triggered tautomeric switching. This is unique compared to solid-state salicylideneanilines' counterpart, which typically requires high energy input such as photo or thermal activation to trigger the enol-keto tautomerisim and cis-trans isomerization. Accompanying the tautomerization, the ionic properties of the COF can be tuned reversibly, thus forming the basis of size-exclusion, selective ionic binding or chemoseparation in SA-COF demonstrated in this work.

Abstract:Extracting highly value-added products from natural systems with low concentrations and complex constituents is of great significance but remains a massive challenge. Herein, a series of dual-functional covalent organic frameworks with both anion exchange and pore adsorption abilities are designed and fabricated using the quaternary amine monomer doping strategy. Attributed to abundant ion exchange sites and selective pore adsorption, the COFs demonstrate high selectivity and loading capacity and fast adsorption for natural products (glycyrrhetinic acid, a high-value natural product), surpassing commercially available materials. Additionally, high-purity (>99%) glycyrrhetinic acid with a >90% extraction efficiency from natural fermentation systems is successfully achieved using a solid-phase extraction column with COFs as the separation medium. The separation column demonstrates outstanding reusability and reproducibility, providing great potential for COFs to serve as robust separation mediums for challenging natural product purification.

Abstract:Covalent organic frameworks (COFs) have great adsorption potential due to their tunable pore size and pore architecture as well as tailorable functionalities. However, the green and rapid synthesis of COFs with excellent adsorption properties remains a great challenge. Here, we presented a solvent-free and facile mechanochemical approach for the synthesis of COFs. The as-prepared COF TpBD-(SO 3 Na) 2 containing sulfonate ions exhibited good thermal and chemical stability and can serve as an effective adsorbent for selective capture of fluoroquinolone antibiotics (FQs) benefiting from the ionic interface and abundant negatively charged sites. The maximum adsorption capacities of norfloxacin, enoxacin, and ciprofloxacin were 1709.6, 1661.5, and 1362.8 mg g −1 , respectively, which outperformed those of other reported adsorbents. The adsorption mechanism was deeply explored through characterizations combined with density functional theory calculations, proving the synergistic adsorption effects of TpBD-(SO 3 Na) 2 and FQs, including π-π* interactions, electrostatic interactions, and hydrogen bonding. Moreover, a fluorescence detection platform for FQs was developed based on the fluorescence signal of sodium disulfonate TpBD-(SO 3 Na) 2 . The limit of detection of norfloxacin was 8 nmol/L with a linear range from 0.03 to 6 μmol/L. This research provides a new strategy for the green synthesis of COFs and expands their application in the synchronous detection and removal of FQs in aquatic environments.

Abstract:We designed and synthesized a novel covalent organic framework (COF) by condensation polymerization of 1,3,6,8-tetrakis(4-formylphenyl)pyrene and melamine through imine bonds (represented by Py-M-COF). The basic characterizations revealed that the Py-M-COF not only exhibited an extended π-conjugation framework, a large specific surface area (495.5 m2 g−1), big pore cavities, and nanosheet-like structure but also possessed rich functional groups, such as C˭C, C˭N, C˭O, and NH2. These features endowed the Py-M-COF with high charge carrier mobility, further improving the strong immobilization of DNA aptamer strands via π-π stacking interaction and electrostatic interaction. As such, the Py-M-COF-based electrochemical aptasensors are ultrasensitive in detecting different antibiotics, including enrofloxacin (ENR) and ampicillin (AMP), yielding extremely low detection limits of 6.07 and 0.04 fg mL−1 (S/N = 3) toward ENR and AMP, respectively, along with other excellent sensing performances. This biosensing platform based on Py-M-COF has potential applications for the sensitive detection of antibiotics or other analytes by replacing the corresponding aptamers.

Abstract:With increasing use of antibiotics, it becomes more and more important for selectively and sensitively detecting antibiotics. In this work, a new Zn(II) framework [Zn-2(L)(NDC)(HCOO)(2)]2H(2)O (1) was achieved by using organic ligands 3,3,5,5-tetra(1H-imidazol-1-yl)biphenyl (L) and 2,6-naphthalenedicarboxylic acid (H2NDC). 1 has three-dimensional (3D) framework structure and exhibits strong luminescence in the solid state as well as in the suspended acetonitrile solution. Furthermore, it was found that the emission of 1 can be quenched efficiently by trace amounts of nitrofuran antibiotics (NFs) including nitrofurazone (NZF), furazolidone (FZD) and nitrofurantoin (NFT) with detection limits of 184, 243 and 263 ppb for NZF, FZD and NFT, respectively, even in the presence of other antibiotics such as penicillin (PCL).

Abstract:Covalent organic frameworks ( COF) is a highly ordered crystalline porous polymer synthesized by dynamic covalent chemical method. Low density, large specific surface area, adjustable porosity, simple and diverse synthesis routes, designable functional units and structures, easy functionalization of surface and pore channels, and high physicochemical stability are the main characteristics of COFs. It has received extensive attention in molecular adsorption and separation, energy storage, photoelectricity, sensing, catalysis, chromatography materials, water treatment materials and biomedicine domains. This paper focuses on the recent research progress of COFs over bar based nanosystems in biomedical fields, such as biological detection and imaging, drug delivery, optical therapy and combination therapy, and finally summarize the current challenges and future development opportunities of COFs in biomedical field.

Exploration of a novel triazine-based covalent organic framework for solid-phase extraction of antibiotics
