Abstract:Several aromatic aldehydes and sodium bisulfite were mixed to prepare hydroxyl sulfonates,which were added to o-phenylenediamine or substituted o-phenylenediamine in refluxing ethanol to afford 2-substi-tuted benzimidazoles through oxidation by air directly.One of the fifteen synthesized compounds,2-(4-chlorophyenyl)-6-methoxyl-1H-benzimidazole has not been reported.All the compounds were identified by IR,1H NMR spectra and elemental analysis.The method is simple,mild,eco-friendly and requires no catalyst.

Abstract:A facile and transition-metal-free method was developed through merging the ring opening of benzoxazoles with secondary amines and N-bromosuccinimide (NBS) mediated oxidative cyclization toward the synthesis of 2-aminobenzoxazoles. NBS was selected as a powerful oxidant in the oxidative cyclization of ring-opening amidines to provide the desirable 2-aminobenzoxazoles in excellent yields (up to 94%).

Abstract:The formation of amide bonds is one of the most valuable transformations in organic synthesis. Beckmann rearrangement is a well-known method for producing secondary amides from ketoximes. This study demonstrates the rapid synthesis of ketoximes and amides in microdroplets. Many factors are found to affect the yield, such as microdroplet generation devices, temperature, catalysts, and concentrations of reactants. In particular, the temperature has a great influence on the synthesis of amide, which is demonstrated by a sharp ascendance to the yield when the temperature was increased to 45 degrees C. The best amide yield (93.3%) can be obtained by using coaxial flowing devices, a sulfonyl chloride compound as a catalyst, and heating to SS degrees C in microdroplets. The yields can reach 78.7-91.3% for benzoylaniline and 87.2-93.4% for benzophenone oximes in several seconds in microdroplets compared to 10.1-66.1% and 82.5-93.3% in several hours in the bulk phase. Apart from the dramatically decreased reaction time and enhanced reaction yields, the microdroplet synthesis is also free of severe reaction environments (anhydrous and anaerobic conditions). In addition, the synthesis in microdroplets also saves reactants and solvents and reduces the waste amounts. All of these merits indicate that the microdroplet synthesis is a high-efficiency green methodology.

Abstract:Due to their important roles in medicine and asymmetric metal catalysis, the formation of Betti bases has attracted wide interest in organic chemical community. Traditional multicomponent reaction methods for synthesizing Betti bases normally require long reaction times under harsh conditions (high temperature, microwave or ultrasonic irradiation, etc.) in the presence of various catalysts. In this study, we developed a mild, highly efficient and environmentally friendly method to synthesize Betti bases without the use of any catalysts in microdroplets. The Betti reaction was accelerated by 6.53×103 in microdroplets by comparing the measured rate constant in bulk. Fifteen Betti bases were synthesized by the microdroplet method using a variety of aldehydes, naphthols and amines with 68-98 % yields at a scaled-up amount of 1.9 g h-1 . Overall it is an attractive alternative to classic organic synthesis for the construction of Betti bases and derivatives.

Abstract:Benzimidazole scaffolds became an attractive subject due to their broad spectrum of pharmacological activities. In this work, a methodology was developed for the synthesis of N-substituted benzimidazole derivatives from benzimidazoles and α, β-unsaturated compounds (acrylonitriles, acrylate esters, phenyl vinyl sulfone) catalyzed by lipase TL IM from Thermomyces lanuginosus in continuous-flow microreactors. Investigations were conducted on reaction parameters such as solvent, substrate ratio, reaction temperature, reactant donor/acceptor structures, and reaction time. The transformation is promoted by inexpensive and readily available lipase in methanol at 45 °C for 35 min. A wide range of β-amino sulfone, β-amino nitrile, and β-amino carbonyl compounds were efficiently and selectively synthesized in high yields (76–97%). All in all, a microfluidic biocatalysis system was applied to the synthesis of N-substituted benzimidazole derivatives, and could serve as a promising fast synthesis strategy for further research to develop novel and highly potent active drugs.

Abstract:α-Acyloxy carboxamides are important multifunctional natural products that show bioactive and pharmacological activities. Traditional three-component Passerini reactions among isocyanates, aldehydes/ketones, and carboxylic acids for affording α-acyloxy carboxamides suffer from several drawbacks such as long reaction time, high reaction temperature, special reaction devices, etc. Herein, we developed a high-efficiency microdroplet method for accelerating the Passerini reactions by 3 orders of magnitude by comparing with the rate constants in bulk, achieving high-yield and gram-scale (scaling up to 1.91 g for 1 h collection) synthesis of α-acyloxy carboxamides at near room temperature. The Passerini microdroplet method shows a wide scope for a variety of benzoic acids, aryl aldehydes, and isocyanates. Moreover, the Passerini reaction was poorly conducted in aqueous microdroplets but well accelerated in acetonitrile microdroplets with at least 230 times efficiency than on-water Passerini reactions. All results proved it an attractive alternative to classic organic synthesis for the construction of α-acyloxy carboxamides and derivatives.

Abstract:In the presence of KI and (diacetoxyiodo)benzene (PIDA), primary amines bearing an alpha-hydrogen can be efficiently converted to fluoroalkylated imidazoles by reacting with fluoroalkylated alkynes and sodium azide via a cascade Michael addition/azidation/cycloamination process. KI plays a crucial role in this reaction by generating iodine(I) species to promote the cyclization. Substrate scope was explored, and the application of this protocol was demonstrated. Additionally, a plausible reaction mechanism was proposed.

Abstract:Benzimidazoles and benzothiazoles are a class of pharmacologically potential compounds, which exhibited antimicrobial, anticancer, and anti-inflammatory activities. These can be obtained by simple condensation of o -phenylenediamine or o -aminothiophenol with aromatic aldehydes. The synthetic protocol can be accomplished/improved by varying reaction parameters such as temperature, solvents, and catalysts. To develop such condensation reactions in a sustainable way, nontoxic solvents and eco- friendly catalysts are presently used. In this study, we proposed a novel and interesting strategy for obtaining diversely substituted 2-phenyl benzimidazole and 2-phenyl benzothiazole derivatives via a one-pot protocol, employing pyrrolidinium ionic liquid as a green and environmentally benign catalyst under solvent-free conditions at room temperature in an open atmosphere. The resulting products were obtained in good to excellent yields within a short reaction time (3–20 min). A plausible mechanism was also discussed. Abstract:A facile one-pot multicomponent reaction employing aromatic aldehydes, malononitrile, and phthalhydrazide in H 2 O and EtOH (1:1) mixture at 80oC has been innovated for the synthesis of 1 H -pyrazolo[1,2- b ]phthalazine-5,10-diones. This approach stands out for its simplicity, ease of execution, and high efficiency. The reaction employs 20 mol% of imidazole as an organocatalyst, enhancing its practicality and applicability. A total of 14 compounds were synthesized within 0.5 to 4 h under a greener reaction medium, with the yields of products ranging from 85–94%. The key components of this protocol are the use of H 2 O:EtOH (1:1) mixture as green solvent, recyclable reaction medium, imidazole as an organocatalyst, mild reaction conditions, easy isolation of products, good to excellent yields, and no column chromatographic purification. Using the microbial dilution method, all the synthesized compounds underwent evaluation for their antibacterial activity against a spectrum of strains, including E. coli , S. aureus , P. aeruginosa , B. subtilis , as well as antifungal activity against A. flavus and A. niger. The synthesized derivatives exhibited diverse inhibitory effects on the growth of both bacterial and fungal strains, showcasing their potential utility across various microbial targets.

Abstract:The first sequential acid-catalyzed propargylation/base-mediated aza-cycloisomerization between indolyl-benzimidazoles and propargylic alcohols is described. This protocol enables the one-pot construction of N-fused benzimidazo-β-carbolines in good yields. Abstract:A novel pyrazole synthesis approach has been developed by using Bronsted acidic ionic liquid BMImHSO(4), which acts as dual solvent-catalyst in promoting the condensation of hydrazines with various 1,3-dicarbonyl compounds at room temperature. In the reaction process, no catalysts or other organic solvents are used, and BMImHSO(4) can be readily reused without efficiency loss after simple treatment. This provides a green and mild synthetic method for the preparation of this class of heterocyclic compounds.
