Abstract:Single-particle inductively coupled plasma-mass spectrometry (sp-ICP-MS) is one of the most powerful tools in the thriving field of nanomaterial analysis. Along the same lines, single-cell ICP-MS (sc-ICP-MS) has become an invaluable tool in the study of the variances of cell populations down to a per-cell basis. Their importance and application fields have been listed numerous times, across various reports and reviews. However, not enough attention has been paid to the immense and ongoing development of the tools that are currently available to the analytical community for the acquisition, and more importantly, the treatment of single-particle and single-cell-related data. Due to the ever-increasing demands of modern research, the efficient and dependable treatment of the data has become more important than ever. In addition, the field of single-particle and single-cell analysis suffers due to a large number of approaches for the generated data-with varying levels of specificity and applicability. As a result, finding the appropriate tool or approach, or even comparing results, can be challenging. This article will attempt to bridge these gaps, by covering the evolution and current state of the tools at the disposal of sp-ICP-MS users.

Abstract:Single particle inductively coupled plasma mass spectrometry (SP-ICP-MS) refers to the use of ICP-MS as a particle counting technique. When ICP-MS measurements are performed at very high data acquisition frequencies, information about (nano)particles containing specific elements and their dissolved forms can be obtained (element mass per particle, size and number and mass concentrations). As a result of its outstanding performance, SP-ICP-MS has become a relevant technique for the analysis of complex samples containing inorganic nanoparticles. This review discusses the maturity level achieved by the technique through the methods developed for the detection, characterisation and quantification of engineered and natural (nano)particles. The application of these methods in different analytical scenarios is comprehensively reviewed and critically discussed, with special attention to their current technical and metrological limitations. The emergent applications of SP-ICP-MS in the field of nanoparticle-tagged immunoassay and hybridization methods are also reviewed.

Abstract:The content, migration and transformation of trace elements in living organisms play an important role in physiological activities. The analysis of trace elements in cells is of great significance to understand the function mechanism of trace elements in cells and organisms. Compared with conventional bulk analysis, many medical and clinical researches benefit from cell heterogeneity via single-cell analysis. Time-resolved analysis-inductively coupled plasma-mass spectrometry (TRA-ICP-MS) based strategies have the potential for both counting and quantitative assessment. It has been used to evaluate cell-to-cell difference in elemental content, cellular uptake of metal-containing drugs and nanoparticles. This review focuses on TRA-ICP-MS based methodologies for single-cell analysis over the past few decades. Recent advances in ICP-MS instrumentations (cell introduction systems and mass spectrometers), microfluidic platforms and their in-depth applications in single-cell analysis are summarized. This review also prospects future developments of TRA-ICP-MS for single-cell analysis.

Abstract:Metals have a fundamental role in microbiology, and accurate methods are needed for their identification and quantification. The inability to assess cellular heterogeneity is considered an impediment to the successful treatment of different diseases. Unlike bulk approaches, single-cell analysis allows elemental heterogeneity across genetically identical populations to be related to specific biological events and to the effectiveness of drugs. Single particle-inductively coupled plasma-mass spectrometry (SP-ICP-MS) can analyse single cells in suspension and measure this heterogeneity. Here we explore advances in instrumental design, compare mass analysers and discuss key parameters requiring optimisation. This review has identified that the effect of pre-treatment of cell suspensions and cell fixation approaches require further study and novel validation methods are needed as using bulk measurements is unsatisfactory. SP-ICP-MS has the advantage that a large number of cells can be analysed; however, it does not provide spatial information. Techniques based on laser ablation (LA) enable elemental mapping at the single-cell level, such as laser-induced breakdown spectroscopy (LIBS) and laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS). The sensitivity of commercial LIBS instruments restricts its use for sub-tissue applications; however, the capacity to analyse endogenous bulk components paired with developments in nano-LIBS technology shows great potential for cellular research. LA-ICP-MS offers high sensitivity for the direct analysis of single cells, but standardisation requires further development. The hyphenation of these trace elemental analysis techniques and their coupling with multi-omic technologies for single-cell analysis have enormous potential in answering fundamental biological questions.

Abstract:Single-particle inductively coupled plasma-mass spectrometry (SP-ICP-MS) has emerged as an important tool for the characterization of inorganic nanoparticles (NPs) in the environment. Although most SP-ICP-MS applications rely on the quadrupole ICP-MS (ICP-QMS), it is limited by the slow scanning speed of the quadrupole. Recent advancements in instrumentation have led to the development of inductively coupled plasma time-of-flight mass spectrometry (ICP-TOF-MS) which offers a viable solution. In this review, we discuss the recent advances in instrumentation and methodology of ICP-TOF-MS, followed by a detailed discussion of the applications of SP-ICP-TOFMS in analyzing NPs in the environment. SP-ICP-TOFMS has the potential to identify and quantify both anthropogenic and natural NPs in the environment, providing valuable insights into their occurrence, fate, behavior, and potential environmental risks.

Abstract:: As an emerging interdisciplinary science, metallomics aims to integrate research fields related to metals and metalloids in biological systems from a systematic perspective. Inductively coupled plasma mass spectrometry (ICP-MS) is considered one of the most versatile tools for metallomics research. This review presents a brief overview of ICP-MS and describes recent advances in ICP-MS instrumentation. Then, ICP-MS-based methods and applications are discussed, focusing on single particle analysis, single cell analysis, and spatial metallomics. With the rapid developments in instrumentation and methodology, ICP-MS-based methodologies will evolve further and play a dominant role in metallomics research.

Abstract:Laser ablation (LA) in combination with inductively coupled plasma mass spectrometry (ICP-MS) is a technically advanced micro-analytical method for direct sampling of solid materials and allows the determination of a majority of elements from the periodic table. In recent years, the technology has undergone major improvements in hardware, software and the methodology, which have led to a significant reduction of the analysis time, higher spatial resolution/image quality, better sensitivity and signal to noise ratios. Reliable and validated quantification procedures remain one of the bottlenecks in LA-ICPMS bioimaging. This review provides a comprehensive overview on different quantification strategies commonly used for bioimaging applications by LA-ICPMS. The advantages and drawbacks of existing quantification approaches in terms of analytical capabilities will be critically discussed and showcases of their application to biological samples will be presented. Recent developments and future perspectives of the field will be discussed.

Abstract:The variation among individual cells plays a significant role in many biological functions. Single-cell analysis is advantageous for gaining insight into intricate biochemical mechanisms rarely accessible when studying tissues as a whole. However, measurement on a unicellular scale is still challenging due to unicellular complex composition, minute substance quantities, and considerable differences in compound concentrations. Mass spectrometry has recently gained extensive attention in unicellular analytical fields due to its exceptional sensitivity, throughput, and compound identification abilities. At present, single-cell mass spectrometry primarily concentrates on the enhancement of ionization methods. The principal ionization approaches encompass nanoelectrospray ionization (nano-ESI), laser desorption ionization (LDI), secondary ion mass spectrometry (SIMS), and inductively coupled plasma (ICP). This article summarizes the most recent advancements in ionization techniques and explores their potential directions within the field of single-cell mass spectrometry.

Abstract:Inductively coupled plasma mass spectrometry (ICPMS) has already been demonstrated as a promising technique for metallic nanoparticle tagged bioassays due to its high sensitivity, wide dynamic linear range, and more importantly multiplex and absolute quantification ability. Besides, single nanoparticle analysis by ICPMS has also recently been applied for many metal nanoparticles. Moreover, its short data acquisition dwell times (serval hundred microseconds) lead to an extremely high signal to noise ratio for metal nanoparticles (i.e., low detection limits). This perspective focuses on single nanoparticle analysis-based ICPMS bioassays, which provide high sensitivity without any sophisticated signal amplification procedures. Herein, the recent development of single nanoparticle analysis, ICPMS instrument design, and single molecule analysis is discussed. Considering the vast types of metallic nanoparticles currently available and simultaneous multiplex detection capability of TOF-ICPMS, single nanoparticle analysis-based bioassays may open a new avenue for multiplex single molecule analysis.

Abstract:To meet the demand for multi-element/isotope analysis at the single nanoparticle (NP) or cell level, different types of inductively coupled plasma mass spectroscopy (ICP-MS) have been used to simultaneously monitor multiple mass-to-charge ratios in single-particle/cell ICP-MS (SP/SC-ICP-MS) analysis. Systematic evaluation and comparison of the performance of these techniques are urgently required. Herein, three ICP-quadrupole (Q)-MS, two ICP-time of flight (TOF)-MS, and one multi-collector (MC)-ICP-MS instruments were employed to simultaneously detect 107 Ag and 109 Ag on single Ag NPs and Ag-exposed cyanobacteria cells. The evaluation was conducted by comparing the measured event-specific 109 Ag: 107 Ag ratios with the natural ratio. Duration of NP or cell events and time resolution in the peak hopping mode were the main factors affecting the performance of ICP-Q-MS. Under the optimal condition (100 μs for both dwell time and settling time), less than 45% of the NP or cell events had a 109 Ag: 107 Ag ratio deviating <30% from the natural ratio. Most events obtained via ICP-TOF-MS were paired events with both isotopes detected. For large-size NPs and cells with high exposure levels, nearly 80% of the events had a ratio deviation within ±30%. MC-ICP-MS performed particularly well in isotope determination with all the events having a ratio deviation within ±5%. For ICP-TOF-MS and MC-ICP-MS, the signal intensity of the events was the main factor affecting the accuracy of the measured 109 Ag: 107 Ag ratios due to the counting statistics. The established methods and results provide insight on the analyses of two elements/isotopes or more on single NPs or cells. Based on the comparison of the advantages and limitations of these instruments, this study provides a critical reference for future multi-element/isotope SP/SC-ICP-MS analyses.

Abstract:In this proof-of-principle study, we present our contribution to single particle inductively coupled plasma mass spectrometry (spICPMS) developments with a novel in-house built data acquisition system with nanosecond time resolution (nsDAQ) and a matching data processing approach. The new system can continuously sample the secondary electron multiplier (SEM) detector signal and enables the detection of gold nanoparticles (AuNP) as small as 7 nm with a commercial single quadru-pole ICPMS instrument. Recording of the SEM signal by the nsDAQ is performed with a dwell time of approximately 4 ns. A tailored method was developed to process this type of transient data, which is based on determining the temporal dis-tance between detector events that is denoted as event gap (EG). We found that the inverse logarithm of EG is proportional to the particle size and the number of detector events corresponding to a particle signal distribution can be used to calibrate and determine the particle number concentration of a nanoparticle dispersion. Due to the high data acquisition frequency, a statistically significant number of data points can be obtained in under 30 s and the main measurement time limitation for analyses is merely the sample uptake time and rinsing step between analyte solutions.

Abstract:Inductively coupled-plasma mass spectrometry (ICP-MS) has transformed our knowledge on the role of trace and major elements in biology and has emerged as the most versatile technique in elemental mass spectrometry. The scope of ICP-MS has dramatically changed since its inception, and nowadays, it is a mature platform technology that is compatible with chromatographic and laser ablation (LA) systems. Over the last decades, it kept pace with various technological advances and was inspired by interdisciplinary approaches which endorsed new areas of applications. While the first part of this review was dedicated to fundamentals in ICP-MS, its hyphenated techniques and the application in biomonitoring, isotope ratio analysis, elemental speciation analysis, and elemental bioimaging, this second part will introduce relatively current directions in ICP-MS and their potential to provide novel perspectives in the medical sciences. In this context, current directions for the characterisation of novel nanomaterials which are considered for biomedical applications like drug delivery and imaging platforms will be discussed while considering different facets of ICP-MS including single event analysis and dedicated hyphenated techniques. Subsequently, immunochemistry techniques will be reviewed in their capability to expand the scope of ICP-MS enabling analysis of a large range of biomolecules alongside elements. These methods inspired mass cytometry and imaging mass cytometry and have the potential to transform diagnostics and treatment by offering new paradigms for personalised medicine. Finally, the interlacing of immunochemistry methods, single event analysis, and functional nanomaterials has opened new horizons to design novel bioassays which promise potential as assets for clinical applications and larger screening programs and will be discussed in their capabilities to detect low-level proteins and nucleic acids.

Abstract:The Single-probe is a multifunctional device that can be coupled to mass spectrometry (MS) for molecular analysis of microscale samples, such as single cells, tissue slices, and multicellular spheroids, under ambient conditions. In Single-probe single cell MS (SCMS) studies, this technique leverages direct sampling and nanoelectrospray ionization (nanoESI), enabling the analysis of cells in their native environments without the need for extensive sample preparation. The Single-probe SCMS techniques have been used for a variety of different studies at the single-cell level, including semiquantitative profiling of metabolites, quantification of target molecules, cell-cell interactions, and incorporated chemical reactions for extended molecular analysis. Due to its miniaturized design and versatility, the Single-probe has been utilized for MS imaging (MSI) of tissue sections and investigation of extracellular molecules in live single spheroids. This review summarizes recent advancements in Single-probe-based techniques and their applications, with a focus on SCMS studies, emphasizing their potential utility in advancing MS methodologies in microscale bioanalysis.
