Abstract:For the exploration of the intrinsic reactivity of two key active species in the catalytic cycle of horseradish peroxidase (HRP), Compound I (HRP-I) and Compound II (HRP-II), we generated in situ [Fe(IV) O(TMP(+.) )(2-MeIm)](+) and [Fe(IV) O(TMP)(2-MeIm)](0) (TMP=5,10,15,20-tetramesitylporphyrin; 2-MeIm=2-methylimidazole) as biomimetics for HRP-I and HRP-II, respectively. Their catalytic activities in epoxidation, hydrogen abstraction, and heteroatom oxidation reactions were studied in acetonitrile at -15 °C by utilizing rapid-scan UV/Vis spectroscopy. Comparison of the second-order rate constants measured for the direct reactions of the HRP-I and HRP-II mimics with the selected substrates clearly confirmed the outstanding oxidizing capability of the HRP-I mimic, which is significantly higher than that of HRP-II. The experimental study was supported by computational modeling (DFT calculations) of the oxidation mechanism of the selected substrates with the involvement of quartet and doublet HRP-I mimics ((2,4) Cpd I) and the closed-shell triplet spin HRP-II model ((3) Cpd II) as oxidizing species. The significantly lower activation barriers calculated for the oxidation systems involving (2,4) Cpd I than those found for (3) Cpd II are in line with the much higher oxidizing efficiency of the HRP-I mimic proven in the experimental part of the study. In addition, the DFT calculations show that all three reaction types catalyzed by HRP-I occur on the doublet spin surface in an effectively concerted manner, whereas these reactions may proceed in a stepwise mechanism with the HRP-II mimic as oxidant. However, the high desaturation or oxygen rebound barriers during CH bond activation processes by the HRP-II mimic predict a sufficient lifetime for the substrate radical formed through hydrogen abstraction. Thus, the theoretical calculations suggest that the dissociation of the substrate radical may be a more favorable pathway than desaturation or oxygen rebound processes. Importantly, depending on the electronic nature of the oxidizing species, that is, (2,4) Cpd I or (3) Cpd II, an interesting region-selective conversion phenomenon between sulfoxidation and H-atom abstraction was revealed in the course of the oxidation reaction of dimethylsulfide. The combined experimental and theoretical study on the elucidation of the intrinsic reactivity patterns of the HRP-I and HRP-II mimics provides a valuable tool for evaluating the particular role of the HRP active species in biological systems.

Abstract:The purpose of the current study was to evaluate the interaction between zinc ions and horseradish peroxidase (HRP) by ultraviolet-visible, fluorescence, circular dichroism, and FTIR spectroscopies. HRP conformation analysis revealed a noticeable decrease in α-helix from 47% for the free enzyme to 17% for HRP–Zn 2+ and a reduction in tertiary structures of the enzyme through a Zn 2+ interaction. Moreover, the fluorescence intensity of HRP was decreased significantly by Zn 2+ . The linear relationship of HRP emission data and inhibitor concentration indicated that the extinction process is linear and occurs uniformly with increasing Zn 2+ concentration. The values of K q for the zinc and HRP interaction were in the order of 10 11 L/mol, which means that HRP fluorescence was quenched by Zn 2+ through a static quenching mechanism.
The Regulatory Implications of Hydroquinone for the Multifunctional Enzyme Dehaloperoxidase-Hemoglobin from Amphitrite Ornata

Abstract:Hydroquinone (H(2)Q) has been observed to compete with the oxidation of substrates 2,4,6-tribromophenol (2,4,6-TBP) and 2,4,6-trichlorophenol (2,4,6-TCP) catalyzed by the dehalopercaidase-hemoglobin (DHP) from Amphitrite ornata in the presence of H2O2.. This competition is observed as a lag phase during which H2Q is preferentially oxidized to 1,4-benzoquinone (1,4-BQ) while totally inhibiting either 2,4,6-TBP or 2,4,6-TCP oxidation. The inhibition by H2Q is distinct from that of the native competitive inhibitor 4-bromophenol (4-BP) since H(2)Q is itself oxidized and its product 1,4-BQ is not an inhibitor. Thus, once H(2)Q is completely consumed, the inhibition is removed, and normal substrate turnover is initiated, which explains the lag phase. To probe the mechanism of lag phase, the reactions between H(2)Q and DHP were both studied both in the presence and in the absence of H2O2. The reversible reactions between ferric/oxyferrous DHP A and H(2)Q/1,4-BQ are shown to involve a proton-coupled electron transfer (PCET) mechanism, where the distal histidine His' serves as the proton acceptor. The pK(a) of the distal histidine His(55) has been determined by resonance Raman spectroscopy in order to corroborate its involvement in this mechanism. Consistent with the proposed mechanism, kinetic assays have shown that H(2)Q serves as a substrate for DHP that follows the Michaelis-Menten kinetics. Unlike H(2)Q, the product 1,4-BQ has a relatively large IC; value and therefore has negligible inhibition. This study sheds light on understanding the difference between substrate and inhibitor binding sites and regulatory implication for the peroxidase and oxygen-transporter functions in DHP. It also provides information on PCET in DHP, which is important for resolving the switching between the ferric peroxidase catalytic function and the ferrous oxygen transport function.

Abstract:Oxidation-reduction properties of horseradish peroxidase (HRP) have been investigated by using direct electrochemical methods. Two successive separated distinct one-electron processes of HRP were obtained and the related physiological processes were described. The monolayer coverage of HRP at the electrode surface is about 50 pmol/cm2. UV-Vis spectrophotometry and stable amperometry prove that the enzyme electrode possesses catalytic activity for H2O2 in the absence of a mediator and it might offer an opportunity to build the third generation of biosensors for analtes, such as H2O2, glucose and cholesterol etc.

Abstract:Horseradish peroxidase (HRP) was incorporated in dipalmitoylphosphatidic acid (DPPA) to form a film and the film was modified on pyrolytic graphite electrode. UV-Vis spectra suggested that HRP in the film could keep its secondary structure similar to the native state. A pair of stable, well-defined, and quasi-reversible cyclic voltammetric peaks was observed with the formal potential at -276.2 mV (vs. saturated calomel electrode), characteristic of heme Fe(III)/Fe(II) redox couple of HRP. The apparent heterogeneous electron transfer rate constant and other electrochemical parameters were presented. The catalytic activity of HRP in DPPA film toward oxygen, hydrogen peroxide and nitric oxide were also examined.

Abstract:We studied a possibility of the enzymatic oxidation of sulfo derivatives of di- and triarylamines with hydrogen peroxide in the presence of horseradish peroxidase (HRP). The product of the enzymatic oxidation of sodium triphenylamine-4-sulfonate (TPAS) is more stable than sodium diphenylamine-4-sulfonate. The effects of various factors, such as the nature of buffer solutions, reagent concentrations, and the presence of cationic Ir(IV) and Rh(III) complexes in the reaction medium, on the TPAS oxidation rate were studied. The kinetic regularities of the TPAS enzymatic oxidation were studied, the kinetic parameters (kcat, KM) were estimated, and an assumption was made about the process activation mechanism in the presence of catalytically active Ir(IV) and Rh(III) complexes. The reaction of the enzymatic oxidation of TPAS can be used for the determination of HRP (5.0 × 10–2 nM), hydrogen peroxide (0.6 mM), and cationic complexes of Ir(IV) (5 nM) and Rh(III) (60 nM).

Abstract:Horseradish peroxidase (HRP) mediates efficient conversion of many phenolic contaminants and thus has potential applications for pollution control. Such potentially important applications suffer however from the fact that the enzyme becomes quickly inactivated during phenol oxidation and polymerization. The work here provides the first experimental data of heme consumption and iron releases to support the hypothesis that HRP is inactivated by heme destruction. Product of heme destruction is identified using liquid chromatography with mass spectrometry. The heme macrocycle destruction involving deprivation of the heme iron and oxidation of the 4-vinyl group in heme occurs as a result of the reaction. We also demonstrated that heme consumption and iron releases resulting from HRP destruction are largely reduced in the presence of polyethylene glycol (PEG), providing the first evidence to indicate that heme destruction is effectively suppressed by co-dissolved PEG. These findings advance a better understanding of the mechanisms of HRP inactivation.

Abstract:The stability of enzymes remains a critical issue in biotechnology. Compared with the strategies for obtaining stable enzymes, chemical modification is a simple and effective technique. In the present study, chemical modification of horseradish peroxidase (HRP) was carried out with pyromellitic anhydride. HRP has achieved a prominent position in the pharmaceutical, chemical, and biotechnological industries. In this study, the effect of chemical modification on thermal stability, structure, and function of the enzyme was studied by fluorescence, circular dichroism, and absorbance measurements. The results indicated a decrease in compactness of the structure and a considerable enhancement in thermal stability of HRP below 60 °C. It seems the charge replacement and introduction of the bulky group bring about the observed structural and the functional changes.

Abstract:Biotechnology is consistently seeking improved enzyme stability. Enzymes have great properties, although their marginal stability limits their applications. Among the strategies for improving stability of the enzymes, chemical modification is a simple and effective technique. In the present study, chemical modification of horseradish peroxidase (HRP) was carried out with 2,3-dichloromaleic anhydride and 2,3-dimethylmaleic anhydride. HRP is an important heme-containing enzyme. It is widely applied in pharmacological, chemical, and medical industries. Here, thermal stability of HRP was investigated at different temperatures. In addition, the enzyme stability was evaluated in urea, DMSO, alkaline pH, and hydrogen peroxide solutions by spectroscopic techniques. Structural investigation indicated that the both anhydrides slightly decrease compactness of the enzyme structure. The results also indicated that 2,3-dichloromaleic anhydride increases thermal stability of the enzyme and its stability in urea and DMSO solutions, but 2,3-dimethylmaleic anhydride only stabilizes HRP in urea solution. Furthermore, the experiments implied that none of the modifiers are effective on the stability of HRP in extreme pH and oxidative condition. Catalytic efficiency and activation energy did not change remarkably following reaction of the enzyme with the both carboxylic anhydrides. Consequently, improvement in the stability of HRP depends on not only the type of modifier but also denaturing condition.

Abstract:Intermediate compound II and III may be formed as a result of the reaction of horseradish peroxidase with H2O2. From the UV spectra, we can see that the stability of the intermediate compounds is worse in reverse micelle than that in aqueous solution. With the increase of the water content, the stability of intermediate compounds gets better, this means that the characteristic of the water in the water pool is near that of bulk water. The results of the stopped-flow experiments show that the formation rate constant and Vinitial of intermediate compound in reverse microemulsion are bigger than that in aqueous solution. However, within die longer reaction time, the disappearance of the intermediate compound appears early in reverse microemulsion than that in aqueous solution. These results are in agreement with the result of the UV experiments.

Abstract:Horseradish peroxidase was incorporated in a kieselguhr membrane. The electron-transfer process of the enzyme was examined by cyclic voltammetry. It was observed that the electron-transfer reactivity of horseradish peroxidase was greatly enhanced, and that direct electrochemistry was accordingly feasible. Using the merits of the direct electron-transfer reactivity of horseradish peroxidase and its specific enzymatic catalysis towards hydrogen peroxide, an unmediated hydrogen peroxide biosensor was constructed. The calibration plot of this hydrogen peroxide sensor was linear in the range of 2.0 × 10 –6 mol/L–6.5 × 10 –4 mol/L. The relative standard deviation was 4.1% for 6 successive determinations at a concentration of 1.0 × 10 –4 mol/L. The detection limit was 1.0 × 10 –6 mol/L.
Mesoporous Materials Promoting Direct Electrochemistry and Electrocatalysis of Horseradish Peroxidase

Abstract:The direct electron transfer and electrocatalysis of horseradish peroxidase (HRP) immobilized on hexagonal mesoporous silicas (HMS) matrix was studied. The interaction between HRP and HMS was examined by using Fourier transform infrared spectroscopy, nitrogen adsorption isotherms and electrochemical methods. The immobilized HRP at a modified glassy carbon electrode showed a good direct electrochemical behavior, which depended on the specific properties of the HMS. Two couples of redox peaks corresponding to Fe(III) to Fe(II) conversion of the HRP intercalated in the mesopores and adsorbed on the external surface of the HMS were observed with the formal potentials of -0.315 and -0.161 V in 0.1M pH7.0 PBS, respectively. The amount of HRP intercalated in the mesopores of HMS proved to be related to the pore size. The HRP intercalated in the mesopores showed a surface controlled electrode process with a single proton transfer. The immobilized HRP displayed an excellent electrocatalytic response to the reduction of hydrogen peroxide (H2O2) without the aid of an electron mediator. The HMS provided a novel matrix for protein immobilization and direct electron transfer study of the immobilized protein.
Catalytic Promiscuity of Horseradish Peroxidase: Aerobic Oxidative Regeneration of Oxidized Nicotinamide Cofactors

Abstract:NAD(P)+ regeneration is of great importance for dehydrogenase (DH)‐catalyzed oxidations. In this work, we report horseradish peroxidase (HRP)‐catalyzed aerobic oxidation of the reduced nicotinamide cofactors, including natural and non‐natural ones, into the oxidized form at neutral and alkaline pH enabled by external amino acids (AAs). HRP showed promiscuous yet extremely low oxidase activity toward NADH at pH 7 in the absence of both AAs and mediators, as evidenced by the observation of compound III. The mechanistic study revealed that AAs as decoy molecules (likely act as acid‐base catalysts) and mediators significantly promoted the formation of compound III as well as its decay back to the ground state. The applicability of aerobic cofactor regeneration was demonstrated by the HRP total turnover number up to 88 000 as well as high yields (91‐99%) in bi‐enzymatic synthesis of carboxylic acids. The present work offers an alternative method to recycle NAD(P)+. Also, it may open up possibilities for HRP‐catalyzed aerobic oxidations.

Abstract:The premixing self-assembly solution of horseradish peroxide with poly(sodium 4-styrenesulfonate) (HRP + PSS) is made by blending HRP and polyelectrolyte PSS, and the active [(HRP + PSS)/PDDA](n) enzyme multilayer biofilm is fabricated by alternate deposition of ( HRP + PSS) and PDDA on the surface of negative ionized PET. The topography of films, the stepwise growth of layers and the activity of HRP in the film are measured with UV-Vis and AFM. HRP and PSS are assembled on the PDDA layer together from the premixing solution of ( HRP + PSS), and the ratio of HRP to PSS in every layer of (HRP + PSS) is the same. The banded structure of HRP under the PSS is observed on the PDDA layer. The Michaelis Constant of the reaction of H2O2, and 4-amino-antipyrine (4-AAP) catalyzed by HRP in biofilm is 9.7 x 10 mol(.)L and is lower than that of the reaction catalyzed by HRP in solution (1.52 x 10(-4) mol-L The affinity between H2O2, and HRP in biofilms is rather strong.

Abstract:Direct electron transfer of immobilized horseradish peroxidase on gold colloid and its application as a biosensor were investigated by using electrochemical methods. The Au colloids were associated with a cysteamine monolayer on the gold electrode surface. A pair of redox peaks attributed to the direct redox reaction of horseradish peroxidase (HRP) were observed at the HRP/Au colloid/cysteamine-modified electrode in 0.1 M phosphate buffer (pH 7.0). The surface coverage of HRP immobilized on Au colloid was about 7.6 × 10−10 mol/cm2. The sensor displayed an excellent electrocatalytic response to the reduction of H2O2 without the aid of an electron mediator. The calibration range of H2O2 was 1.4 μM to 9.2 mM with good linear relation from 1.4 μM to 2.8 mM. A detection limit of 0.58 μM was estimated at a signal-to-noise ratio of 3. The sensor showed good reproducibility for the determination of H2O2. The variation coefficients were 3.1 and 3.9% (n = 10) at 46 μM and 2.8 mM H2O2, respectively. The response showed a Michaelis–Menten behavior at higher H2O2 concentrations. The KappM value for the H2O2 sensor was found to be 2.3 mM.

Abstract:Direct electron transfer between resting horseradish peroxidase (HRP) and its oxidation states was observed at a gold mesh electrode in a spectroelectrochemical cell in the presence of DNA. The conversion between HRP and the oxidized species induced electrochemically was found to be reversible and parallel to that initiated chemically. DNA played an important role as electron carrier and promoted the electron transfer between HRP and the electrodes. Voltammetric results and CD spectra indicated an interaction between HRP and DNA. Moreover, the secondary structure of HRP was slightly disturbed upon mixing with DNA. The direct spectroelectrochemistry of HRP at a gold mesh electrode presented new information on its bioelectrochemical characteristics.

Abstract:N,N′-Dicyanomethyl-o-phenylenediamine was synthesized with a 90% yield by a reaction ofo-phenylenediamine with chloroacetonitrile in triethylamine. Our experimental results showed that it was the effective fluorogenic substrate for horseradish peroxidase (HRP) and hemin. TheKmfor the HRP system was 48 μM,and that for hemin was 1.3 μM.Properties of the substrate were evaluated from the detection limits of enzymes and H2O2. The linear ranges for the determination of HRP and hemin were 21–150 pMand 2–20 nM,respectively. The linear range for the determination of H2O2using HRP or hemin was 18–140 and 60–1000 nM,respectively. The structural elucidation of the fluorescent product using NMR and mass spectral techniques was proposed to be 1,2-dihydro-2-imido-imidazo[1,2-a] quinoxaline. Based on the product structure and earlier reports, the possible reaction mechanism of HRP and the substrate was also proposed, i.e., two steps for ring closures, one step of isomerization, and a final step of oxidative dehydrogenation.
