Insight into biofilm formation of wastewater treatment processes: Nitrogen removal performance and biological mechanisms

Abstract:Biofilm formation affects biological nitrogen (N) removal, and a sequencing batch biofilm reactor (SBBR) was set up to evaluate the changes in N removal and microbial characteristics during biofilm formation. The results indicated that the average effluent concentration of chemical oxygen demand (COD), ammonia nitrogen (NH4+-N) and total nitrogen (TN) in the SBBR were 27.48, 1.41, and 13.52 mg L-1, respectively after biofilm formation. Furthermore, this process increased microbial richness, but reduced microbial diversity. Patescibacteria, Proteobacteria, and Bacteroides were the dominant phyla that did not change after biofilm formation. After biofilm formation, Firmicutes was eliminated while Spirochaetes involved in the interspecies relationship. Biofilm increased the nitrification and denitrification relating coding genes abundance (hao, narG, narZ, nxrA, narH, narY, nxrB, napA, napB, norB, norC and nosZ), and enhanced the processes of N respiration and denitrification, carbohydrate metabolism, amino acid metabolism and membrane transport. Meanwhile, correlation analysis between genera and transcriptome reflected that Zooglea, Micropruina, Aeromonas and Tessaracoccus played essential roles in biofilm formation and N removal. The key enzyme abundance of EC:1.7.99.1, EC:1.7.2.4, and EC:1.1.1.42 of N and tricarboxylic acid (TCA) cycle increased after biofilm formation. This study can reveal the effect of biofilm formation on biological N removal and provide a theoretical foundation for the application of biofilm process.

Abstract:Bioelectrochemical enhanced nitrate removal in wastewater with high total nitrogen and low organic carbon was electrochemically investigated focusing on the relationship between biochemical and electrochemical nitrogen cycles. Under optimized external voltage of -0.6 V, apparent nitrate removal rate of bioelectrochemical denitrification was 76% higher than normal biofilm denitrification. And with the introduction of biofilm on the electrode, new reduction peak of N2O, much larger current density, and 0.4 V positively shift of on-set potential of nitrate reduction reaction were observed, suggesting a synergy of electrochemical reaction and biological reaction through enhanced electrochemical reduction of intermediate products from biological process. Oxygen reduction reaction could not be avoided during nitrogen electrochemical reduction reaction since their similar reduction potential. But it led to decrease of oxygen concentration and therefore contribute to biological denitrification. Bacteria community tests also supported a dominant bacteria which could denitrify and use external electron.

Abstract:Efficient biological denitrification process is a research hotspot in the field of wastewater treat-ment, and biological nitrification is an important unit of the said process, which can remove ammonia nitrogen from wastewater effectively. Many efficient nitrification processes such as: complete nitrification process, full dose shortcut nitrification process, half dose shortcut nitrification process, and so on, were successively exploited by scholars in China and aborad, which promoted the development of biological denitrification technology for wastewater treatment. The principles, performances and operation parameters of complete nitrification process, full dose shortcut nitrification process and half dose shortcut nitrification process were summarized, the charac-terstics and advantages of the above processes were compared, which provided references for the development and application of efficient biological nitrification process.

Abstract:Biofilms are the most favorable form of microorganism growth in nature. With the urgent demand for energy-efficient wastewater treatment today, biofilms have become a crucial tool for the implementation of new treatment technologies. Anammox-based processes are gaining global attention due to their low cost and high nitrogen removal efficiency. However, the application of anammox in mainstream wastewater treatment is still confined to laboratory settings and faces two major bottlenecks: 1) the relatively slow growth rate of anammox bacteria and 2) unstable nitrite generation. Fortunately, these issues are expected to be relieved through the integration of biofilm technologies that couple autotrophic and heterotrophic pathways. Moreover, both laboratory and full-scale evidence from recent studies highlight the great potential and versatile capability of anammox-based biofilms integrated with heterotrophic microorganisms. A solid understanding of biofilm formation, control, and optimization has thus become increasingly important for further application. This paper provides a comprehensive review of existing methods and microbial mechanisms for enhancing treatment in anammox-based biofilm systems. It critically analyses the potential roles of biofilm formation in anammox process, spatial development of microbial ecology, bioreactor control, associated challenges and perspectives in practical applications. This review is anticipated to contribute significantly to advancing the understanding of biofilm reactor design and operation in the application of new biotechnology.

Abstract:A common and long-existing operational issue of wastewater denitrification is the unexpected accumulation of nitrite (NO2-) that could suppress the activity of various microorganisms involved in biological wastewater treatment process and nitrous oxide (N2O) that could emit as a potent greenhouse gas. Recently, it has been confirmed that the accumulation of these denitrification intermediates in biological wastewater treatment process is greatly influenced by the electron competition between the four denitrification steps. However, little is known about this in biofilm systems. In this work, we applied a mathematical model that links carbon oxidation and nitrogen reduction processes through a pool of electron carriers, to assess electron competition in denitrifying biofilms. Simulations were performed comprehensively at seven combinations of electron acceptor addition scheme (i.e., simultaneous addition of one, two or three among nitrate (NO3-), NO2-, and N2O) to compare the effect of electron competition on NO3-, NO2 and N2O reduction. Overall, the effects of substrate loading, biofilm thickness and effective diffusion coefficients on electron competition are not always intuitive. Model simulations show that electron competition was intensified due to the substrate load limitation (from 120 to 20 mg COD/L) and increasing biofilm thicknesses (from 0.1 to 1.6 mm) in most cases, where electrons were prioritized to nitrite reductase because of the insufficient electron donor availability in the biofilm. In contrast, increasing effective diffusion coefficients did not pose a significant effect on electron competition and only increased electrons distributed to nitrite reductase when both NO2- and N2O are added.

Abstract:Excessive nitrogen emissions are a major contributor to water pollution, posing a threat not only to the environment but also to human health. Therefore, achieving deep denitrification of wastewater is of significant importance. Traditional biological denitrification methods have some drawbacks, including long processing times, substantial land requirements, high energy consumption, and high investment and operational costs. In contrast, the novel bio-denitrification technology reduces the traditional processing time and lowers operational and maintenance costs while improving denitrification efficiency. This technology falls within the category of environmentally friendly, low-energy deep denitrification methods. This paper introduces several innovative bio-denitrification technologies and their combinations, conducts a comparative analysis of their denitrification efficiency across various wastewater types, and concludes by outlining the future prospects for the development of these novel bio-denitrification technologies.

Abstract:There is an increasing need for application of biofilm process in the upcycling of wastewater treatment plants all around the world in recent years, yet there are few literatures on summarizing wastewater biofilm during the life cycle. In particular, there is a vacancy on characterization at various stages of biofilm and its regulation. This review provided a whole look at biofilm formation and its development, accompanied by microbial physiology, ecology, and activity, where the initialization of biofilm formation and its characterization were stressed. The new progresses on biofilm physio-ecology analysis and methods on evaluating microbial activity were summarized, while it is worth mentioning that the concept of aging biofilm was also presented. Furthermore, regulations methods of biofilm were reviewed and future research trends on biofilm control were prospected, aiming at guiding biofilm control in biofilm-based wastewater treatment.

Abstract:The presence of nitrogenous substances in wastewater discharges has attracted attention because of the role of nitrogen in eutrophication of receiving waters. Nitrogen removal is an important aspect of present day wastewater treatment processes. However, conventional nitrification-denitrification treatment is uneconomical and difficult to operate due to extremely slow nitrification and the necessity for separate nitrification and denitrification reactors. Recently, several novel aerobic denitrification processes have been developed for cost-effective biological nitrogen removal. These processes can reduce inorganic nitrate compounds to harmless nitrogen gas. Aerobic denitrification bacteria screened and identified in recent years and the nitrogen removal effects of those bacteria are summarized.

Abstract:Presently, the wastewater treatment practices can be significantly improved through the introduction of new microbial treatment technologies. To meet increasingly stringent discharge standards, new applications and control strategies for the sustainable removal of ammonium from wastewater have to be implemented. Partial nitrification to nitrite was reported to be technically feasible and economically favorable, especially when wastewater with high ammonium concentrations or low C/N ratios is treated. For successful implementation of the technology, the critical point is how to maintain partial nitrification of ammonium to nitrite. Partial nitrification can be obtained by selectively inhibiting nitrite oxidizing bacteria through appropriate regulation of the system’s DO concentration, microbial SRT, pH, temperature, substrate concentration and load, operational and aeration pattern, and inhibitor. The review addressed the microbiology, its consequences for their application, the current status regarding application, and the future developments.

Abstract:The selection of biofilm carriers is essential for enriching functional microorganisms. In this study, nonintact hollow fiber membranes were designed as immobilized biofilm carriers in a denitrification bioreactor (DNBR) for treating nitrate in the effluent of wastewater treatment plants. The DNBR performance was studied at varying hydraulic retention times of 18 to 3 h. Results showed that effluent concentrations of total nitrogen (TN) were maintained below 5 mg/L, corresponding to 75.6-84.8% TN removal. The batch tests also showed that the biofilms on nonintact membranes exhibited higher denitrification activities than the sludge flocs (i.e., detached biofilms) in the bioreactor. The 16S rRNA sequencing indicated that the denitrification performance of biofilms was attributed to the enrichment of denitrifying bacteria, particularly Azospira (relative abundance up to ca. 36.5%). Additionally, the colonization of biofilms was dominated by stochastic community assembly processes. The metagenomic analysis revealed that the biofilms harbored abundant functional genes responsible for biofilm formation, glucose metabolism, and nitrogen metabolism. Overall, the reuse of nonintact hollow fiber membranes as biofilm carriers for nitrate removal can be considered a novel approach for "treating waste with waste".

Abstract:Nitrogen pollution is an increasingly severe worldwide problem because of drainage of nitrogen-containing wastewater and intensive application of nitrogen-containing fertilizers. Denitrification, a key process in nitrogen cycles, is commonly employed for nitrogen removal in engineered wastewater treatment systems. Biological denitrification is performed by denitrifying microbes (bacteria) that use nitrate as terminal electron acceptor. Better understanding the functions of diverse microbial populations in denitrification-based wastewater treatment systems, and the interactions of these populations with operating environments, is essential for improving both treatment performance and system stability. Recent advances in “meta-omics” (e. g., genomics, transcriptomics, proteomics, metabolomics), other molecular biology tools, and microbiome analysis have greatly enhanced such understanding. This minireview summarizes recent findings regarding microbial community structure and composition, key functional microbes and their physiology, functional genes involved in nitrogen cycle, and responses of microbes and their genes to changes of environmental factors or operating parameters, in denitrification processes in wastewater treatment systems. Of particular interest are heterotrophic denitrification systems (which require alternative organic carbon sources) and the autotrophic denitrification systems (which do not require an external carbon source). Integrated microbiome and -omics approaches have great future potential for determination of optimal environmental and biotechnological parameters, novel process development, and improvement of nitrogen removal efficiency and system stability.

In Situ Probing the Effect of Potentials on the Microenvironment of Heterotrophic Denitrification Biofilm with Microelectrodes.

Abstract:Bio-electrochemical reactor provides a promising technology to remove nitrogen contaminants in water. In this study, a heterotrophic biofilm for denitrification process was developed, and stable total nitrogen removal efficiencies (>80%) were achieved. Fluorescence in situ hybridization showed that genes norB mainly transcribed in inner biofilm while genes nosZ showed similar transcription activities in the entire biofilm. The microelectrodes of pH and nitrous oxide (N2O) were applied to profile the microenvironment of denitrification biofilm. In all measurements, the microenvironment pH decreased as a function of biofilm depth. The highest N2O concentration of 90μM in biofilm was detected when the bulk solution pH was 7.3, and an applied potential of −0.2V did not decrease the yield of N2O in biofilm at pH 7.3. Nevertheless, pH 9.5 or an applied potential of −0.4V seemed not to suppress the yield of N2O in biofilm.

Abstract:The electrochemically active biofilms have been successfully developed for nitrate removal from the wastewater. The electrochemically "selected" bacteria showed higher activity than the control bacteria. Electron exchange occurred between the electrochemically active bacteria and cathode was identified. Direct electron transfer between the "selected" bacteria and cathodes may be involved in efficient denitrification besides hydrogen transfer, and would contribute to improve denitrification efficiency. The gene analysis of 16S rDNA demonstrated that there was a pronounced enrichment in bacteria of the beta-Proteobacteria with 41.93%, Uncultured bacterium clone Dok04 with 25.11% and Sphingobacteria with 6.36% of the sequences from the bacteria consortium with current acclimation. The engineering-oriented multispecies bacteria were favorable to form conductive biofilms and showed high potential for nitrate treatment. (C) 2013 Elsevier Ltd. All rights reserved.

Abstract:Partial denitrification was one of most effective ways to provide nitrite for annamox; whereas very limited research has been done to achieve nitrite accumulation in biofilm system. In this study, partial denitrification was studied in a lab-scale denitrifying biofilter (DNBF). The results showed biofilm structure variations caused the differences between nitrate specific reduction rate (NaSRR) and nitrite specific reduction rate (NiSRR), which led to nitrite accumulation in different degree at different biofilm formation phases. Hydrodynamic conditions also significantly influenced biofilm structure, nitrate and nitrite reduction activities. At the filtration velocity of 3.86 m h(1), not only biofilm structure, NaSRR and NiSRR kept relatively stable, but also 60% of nitrite accumulation and no nitrate in the effluent were achieved. Furthermore, Thauera genus bacteria, benefited for nitrite accumulation, became the dominant communities in high nitrite accumulation conditions. The partial denitrification combine with anammox in biofilter have the great potential applied in WWTPs. (C) 2017 Elsevier Ltd. All rights reserved.
