Study of Influential Elements on Nitrate Removal from Simulated Groundwater by Biofilm Electrode Reactor

Abstract:A Biofilm Electrode Reactor was operated to remove the nitrate contamination in the simulated groundwater.The effects of temperature,influent pH,hydraulic retention time(HRT)and current on the denitrification of BER system was investigated.The results showed that the reactor could be successfully started up within about 20 days;the formation of biofilm on the surface of cathode was confirmed by scanning electron microscope;nitrate removal efficiency increased with the temperature range from 13.6 ℃ to 34.0 ℃;BER has strong buffering capacity on the shock of influent pH,and nitrate removal efficiency kept above 85% from pH 6.25～8.27;the optimal HRT was 9h,at which the nitrate removal efficiency was over 85%;the nitrate removal efficiency first increased and then kept stable with the current ranged from 13 mA to 102 mA,more than 85% of nitrate can be removed under applied current(78～102 mA),and without accumulation of nitrite.The consumption of anode will be accelerated at higher current.

Abstract:Biofilm-electrode process based on autohydrogentrophic denitrification and its advantages were elaborated,with the electrode material,disposal of the process,factors influencing the treatment effect and some other aspects of the said technology emphatically analyzed.A new idea of developing a more effective and cleaner process was proposed,which using oxygen derived from inert anode electrolysis to accelerate the biological nitration.The process theory was: In order to realize the accumulation of nitrite,HRT was controlled mainly through adjusting oxygen derived from electrolysis,with aeration as the auxiliary treatment;as well as the oxygen level was controlled by adjusting the electrical current.Meanwhile,the denitrification was carried out with hydrogen derived from electrolysis as electron donor so as to realize the generation of nitrogen from nitrate under the oxygen-limited condition in BER.
Nitrate removal from drinking water by a three-dimensional biofilm-electrode reactor

Abstract:The performance of a three-dimensional biofilm-electrode reactor （3D-BER） was tested under different conditions. Results showed that nitrate removal efficiency increased when the current intensity increased from 0 mA to 100 mA. The highest nitrate removal efficiency of 73.8% was achieved at the current of 100 mA, and the nitrate concentration in the effluent was 8.27 mg· L^-1. However, with a further increase of the current intensity from 100 mA to 150 mA, the nitrate removal efficiency decreased slightly. With the current intensity increased from 0 mA to 150 mA, the nitrite generation increased in the beginning then decreased. The accumulation of nitrite was the most serious when the current was 60 mA. It was founded that the otimum temperature for denitrification was from 31 ℃ to 35 ℃ , with nitrate concentrations in effluent less than 10 mg· L^-1. A nitrate removal efficiency up to 85.5% was achieved at 35 ℃. Results showed that appropriate pH for denitrification was between 7.2 and 8.2, with nitrite concentrations in effluent lower than 1 mg· L^-1. The 3D-BER had a good pH buffer capacity. When the pH of the influent increased from 5.5 to 9.0, pH of the effluent ranged between 7.6 and 8.2, with the nitrate removal efficiency between 59.6% and 80.2%. In addition, the current intensity, temperature and pH also influenced the concentrations of ammonia nitrogen in the effluent and the removal efficiency of the total phosphorus （ TP）. Probable causes of TP removal and ammonia nitrogen generation were also discussed by comparing the performance of the 3D-BER with an electrochemical reactor.
Denitrification performance and bacterial ecological network of a reactor using biodegradable poly(3-hydroxybutyrate-co-3-hydroxyvalerate) as an electron donor for nitrate removal from aquaculture wastewater

Abstract:Nitrate accumulation is a common phenomenon in aquaculture that can lead to eutrophication of surrounding water bodies. This study used poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) as a carbon source and substrate and performed a microbial co-occurrence network ecological analysis to elucidate the denitrification processes in two packed-bed reactors with different salinities. The denitrification rate reached maximum values of 0.438 and 0.446 kg m−3 d−1 in reactor I (salinity 0 ‰) and reactor II (salinity 20 ‰), respectively. Although ammonia was formed in both systems based on dissimilation nitrate reduction to ammonia (DNRA), the concentration was very low (2.47 ± 1.99 and 2.84 ± 1.79 mg L−1); moreover, the nitrite content was average (1.01 ± 0.87 and 0.96 ± 0.86 mg L−1). These results suggested that denitrification dominated in both reactors. PHBV generally presented a stable release of DOC, although a sharp increase was observed in the start-up period of reactor II. 16S rRNA results showed that reactor I had richer microbial diversity than reactor II. Among the top ten taxa, Betaproteobacteria was the dominant class in reactor I while Gammaproteobacteria was the dominant class in reactor II. In the stable period, Thauera and Denitromonas was the most abundant genera in reactor I and reactor II, respectively. In addition, the bacterial co-occurrence network showed that reactor I had a more complex node and edge network and faster start-up time compared to reactor II; however, reactor II had a more stable nitrogen removal capacity. Higher expression of NorB and NosZ genes in reactor II indicated higher efficient denitrification in seawater system. The SEM and FTIR showed bacterial development and materials surface erosion. These findings verified the denitrification performance and niche differences between freshwater and seawater environments.

Abstract:Three-dimensional biofilm electrode reactors (3D-BERs) represent a novel technology for wastewater denitrification. Formation of mature electroactive biofilm on particle electrodes is crucial to realize successful denitrification in 3D-BERs. However, long start-up time and low electroactivity of the biofilm formed on particle electrodes limit the further application of 3D-BERs in wastewater treatment. In this work, self-assembled hybrid biofilms (SAHB) was cultivated on granular activate carbon particle electrodes of the 3D-BER by assembling nano ɑ-Fe2O3 into the biofilm. ɑ-Fe2O3 was selected due to its high affinity to bacterial outer-membrane cytochromes, an important mediator for microbial electron transfer. SAHB formed on particle electrodes were characterized and the denitrification performance of 3D-BERs was also investigated. Results indicate that nano ɑ-Fe2O3 plays positive roles in the start-up of 3D-BER, which captures more microbes into SAHB and constructs thick biofilm on particle electrodes. Special microorganisms with denitrification function related with genera of Hydrogenophaga and Opitutus are distinctively enriched in SAHB. Nano ɑ-Fe2O3 induced SAHB exhibit superior denitrification performance compared to natural biofilm. The average denitrification rate increases from 0.62 mg total nitrogen/L/h for natural biofilm to 1.73 mg total nitrogen/L/h for SAHB, mainly ascribed to accelerated nitrites reduction. Our work provides new technical solution to enhance nitrates removal in 3D-BERs and brings deep insights into application of bio-electrochemical system in wastewater treatment.

Abstract:A three-dimensional biofilm-electrode reactor (3D-BER) that combined heterotrophic and autotrophic denitrification (HAD) was developed to remove nitrate. The denitrification performance of the 3D-BER was evaluated under different experimental conditions, including current intensities (0–80 mA), COD/N ratios (0.5–5), and hydraulic retention times (2–12 h). The results showed that excessive current limited the nitrate removal efficiency. However, a longer hydraulic retention time was not required to achieve a better denitrification effect in the 3D-BER. Moreover, the nitrate could be effectively reduced over a broad range of COD/Ns (1–2.5), and its removal rate peaked at 89% at I = 40 mA, HRT = 8 h, and COD/N = 2. Although the current reduced the diversity of microorganisms in the system, it promoted the growth of dominant species. Nitrification microorganisms were enriched in the reactor, especially Thauera and Hydrogenophaga , which were crucial to the denitrification process. Thus, the combination of autotrophic denitrification and heterotrophic denitrification was promoted by the 3D-BER system to increase the efficiency of nitrogen removal.

Abstract:Agricultural waste, corncobs, and sponge iron were used to develop a self-releasing carbon biofilm electrode (SRC-BE) and utilized for denitrification experiments in three-dimensional biofilm electrode reactors (3D-BERs). Fe 2+ was released to enhance the extracellular electron transfer. Following an alkali treatment, SRC-BE were introduced into 3D-BERs of the electrode group (EG), with iron serving as the anode-cathode plate in the EG. Under a current density of 0.1 mA/cm 2 , hydraulic retention time of 2 h, and dissolved oxygen control at 0.5–1.0 mg/L, the removal rates were as follows: the highest removal rates were 71.09 % for total nitrogen, 40.26 % for NH 4 + -N, and 84.34 % for NO 3 − -N, with a stable 96.87 % removal rate for total phosphate. The effluent chemical oxygen demand in the EG remained stable at approximately 12 mg/L during the stable phase, thus reducing the risk of secondary pollution. The enrichment of electroactive microorganisms and functional genes related to C and N also verified their denitrification efficacy.

Influence of Electrode Plate Area and Arrangement of 3BER Reactor on the Nitrogen Removal

Abstract:In order to improve the nitrogen removal efficiency and effluent quality of the three-dimensional electrode biofilm process (3BER), a new designed 3BER-NC process through expanding the area of electrode plate, reducing the electrode space and changing the electrode arrangement was applied and the effect of current, influent pH and 籽( C)/籽( N) on the performance and denitrifying bacteria in system were studied in deep denitrification of the tail water from a municipal wastewater treatment plant. The research results showed that the 3BER-NC operated with an optimal current at 200 mA, which was 100 mA higher than that of 3BER. Because of its shorter electrode space, the 3BER-NC can slow down the carbon electrolysis and effluent color. Furthermore, the enlargement of electrode plate and the change of electrode position distribution of the 3 BER-NC improved the TN removal rate to 86. 45℅, which was 37. 74℅ higher than that of 3BER. The current intensity, influent pH and 籽( C )/籽( N ) had great influence on the nitrogen removal at different current and the nitrogen removal efficiency of 3 BER-NC was better than 3BER. In the nitrogen removal system of the 3BER-NC, the β-proteobacteria played a leadingrole with a joint action of heterotrophic and autotrophic denitrifying bacteria.

Abstract:<p>In this study, a woodchip biofilm electrode reactor (WBER) with woodchips embedded anode and cathode was developed, and its denitrification mechanism was analyzed by investigating the denitrification performance, organic matter change, redox environment and microbial community. The results show that the WBER with a carbon rod as anode (C-WBER) had a higher denitrification efficiency (2.58 mg NO- 3-N/(L·h)) and lower energy consumption (0.012 kWh/g NO- 3-N) at 350 mA/m<sup>2</sup>. By reducing the hydroxyl radical and dissolved oxygen concentrations, anode embedding technology effectively decreased the inhibition on microorganisms. Lignin decomposition, nitrification and aerobic denitrification were carried out in anode. Additionally, hydrogen autotrophic denitrification and heterotrophic denitrification were occurred in cathode. The WBER effectively removed nitrate and reduced the cost, providing a theoretical basis and direction for further develop BERs.</p>

Effects of Different Applied Electric Currents on Nitrate Removal Using the Biofilm-Electrode Reactor

Abstract:Nitrate enrichment in groundwater is a worldwide phenomenon, and the removal of nitrate from groundwater has raised ever-increasing concerns due to severe impacts of nitrate on human and animal health. A bioelectrochemical method for treatment of simulated nitrate-contaminated groundwater was investigated using a biofilm-electrode reactor (BER). Effects of different currents on the transformation of nitrate to nitrogen were investigated. The nitrate removal efficiency was dependent on the current. When the current was lower than 78 mA, the nitrate removal efficiency was linearly related to the current, and a portion of the nitrate was transformed to nitrite or ammonium. The maximum nitrate removal efficiency 87% was achieved at the current of 78 mA and the given operation condition. The BER could be operated efficiently at the current of 78 mA, but further increase the current did not offer significant performance promotion.
Impact of Hydraulic Retention Time and Current on the Microbial Community and Denitrification Genes in a Continuous‐flow Biofilm Electrode Reactor

Abstract:BACKGROUND A biofilm-electrode reactor (BER) was developed on the basis of hydrogenotrophic denitrification. The BER can be strengthened by adding organic matter, and the nitrate removal will be enhanced by a combination of heterotrophic and autotrophic denitrification. Denitrification in a biological system can be affected by operating parameters, and the microbial community and functional genes can be changed at the same time. In this study, the effect of hydraulic retention time (HRT) and current on the denitrification of a low chemical oxygen demand to nitrogen ratio (COD/N), saline wastewater in a continuous-flow BER was evaluated and compared to a conventional biofilm reactor (BR). RESULTS The highest extent of nitrate removal was obtained at the optimum HRT (8 h) and current (10 mA). Low levels of ammonia (0.5-4 mg L-1), produced through dissimilatory nitrate reduction to ammonia (DNRA), were observed. Quantitative real-time polymerase chain reaction analysis revealed that the proportion of nirS-type denitrifiers (3.75-8.30%) exceeded nirK-type denitrifiers (0.24-0.90%) in both BR and BER. The genera of T78, Paracoccus and Azoarcus were identified as the dominant denitrifying bacteria in the BER. The cooperative activity of nitrate, nitrite and nitrous oxide reductase is necessary to reduce nitrate to dinitrogen in the denitrification process. The highest ratio of nosZ/nir genes in the bacterial community was 0.25 when the applied current was 10 mA. CONCLUSION Compared to other denitrification reactors, the BER is more effective to treat low-COD/N, saline wastewater with short HRT and low current. (c) 2018 Society of Chemical Industry

Abstract:A dicyclic-type electrode based biofilm electrode reactor (BER) was developed for advanced nitrate removal from saline municipal wastewater. The denitrification efficiency was evaluated with a synthetic feed (NO3--N, 20 mg L-1) under different salinity and COD to nitrogen ratios (COD/N). As the salinity increased from 0% to 1.0%, the denitrification performance of both the traditional biofilm reactor (BR) and BER was inhibited; however, the BER showed better adaptation and ability to recover. The BER achieved a high nitrate removal efficiency (>= 90%) at a salinity of 1.0% and a low COD/N of 2.5 (theoretical stoichiometric 2.86 ignoring microbial growth). The abundance of Methylotenera mobilis in BR and Clostridium sticklandii in BER was higher than in the initial sludge sample used as inoculum. Likewise, the abundance of napA, nirS and nosZ genes increased as the COD/N further decreased. Under high salinity stress, the BER had a higher denitrification efficiency and the consumption of the organic carbon source (i.e., methanol) was reduced compared to BR. The cooperation between heterotrophic and autotrophic denitrifiers in the BER system provides a more efficient and feasible solution for nitrate removal from saline municipal wastewater. Abstract:The performance of three-dimensional biofilm electrode reactors (3D-BERs) in hydrogen autotrophic denitrification is determined by electroactive biofilms. Herein, a self-assembled hybrid biofilm (SAHB) is developed on granular activated carbon particle electrodes by incorporating nano alpha-Fe2O3. The influences of operating parameters such as current, influent concentration, and hydraulic retention time (HRT) on denitrification are investigated as well as the mechanisms of SAHB-improved denitrification. The results indicate that SAHB achieves a high autotrophic denitrification rate of 4.20 mg TN/L/h under 40 mA current, 100 mg/L nitrate, and 26 h HRT, outperforming the naturally formed biofilm by 23.53%. SAHB contains more microorganisms than naturally formed biofilms, revealing that nano alpha-Fe2O3 is conducive to trapping free microorganisms on particle electrodes. SAHB enriches denitrification-related genes and increases corresponding enzyme activities (particularly nitrite reductase). According to metagenomic sequencing, nano alpha-Fe2O3 addition at a current level of 40 mA appears to favor denitrification via the nirS-dependent pathway probably due to its high affinity for cytochrome C. Metagenomic binning reveals that the microorganism associated with Zoogloeaceae (Rhodocyclales order) in SAHB may play a pivotal role in hydrogen autotrophic denitrification. This work develops a novel denitrification system, furthering our understanding of hydrogen autotrophic denitrification in 3D-BERs.

Abstract:Three-dimensional biofilm electrode reactors (3D-BERs) have attracted extensive attention in recent years due to their wide application range, high efficiency and energy saving. On the basis of traditional bio-electrochemical reactor, 3D-BERs are filled with particle electrodes, also known as the third electrodes, which can not only be used as a carrier for microbial growth, but also improve the electron transfer rate of the whole system. This paper reviews the constitution, advantages and basic principles of 3D-BERs as well as current research status and progress of 3D-BERs in recent years. The selection of electrode materials, including cathode, anode and particle electrode are listed and analyzed. Different constructions of reactors, like 3D-unipolar extended reactor and coupled 3D-BERs are introduced and discussed. Various contaminants degraded by 3D-BERs including nitrogen, azo dyes, antibiotics and the others are calculated and the corresponding degradation effects are described. The influencing factors and mechanisms are also introduced. At the same time, according to the research advances of 3D-BERs, the shortcomings and weakness of this technology in the current research process are analyzed, and the future research direction of this technology is prospected. This review aims to summarize recent studies of 3DBERs in bio-electrochemical reaction and open a bright window to this booming research theme.

Abstract:Bioelectrochemical enhanced nitrate removal in wastewater with high total nitrogen and low organic carbon was electrochemically investigated focusing on the relationship between biochemical and electrochemical nitrogen cycles. Under optimized external voltage of -0.6 V, apparent nitrate removal rate of bioelectrochemical denitrification was 76% higher than normal biofilm denitrification. And with the introduction of biofilm on the electrode, new reduction peak of N2O, much larger current density, and 0.4 V positively shift of on-set potential of nitrate reduction reaction were observed, suggesting a synergy of electrochemical reaction and biological reaction through enhanced electrochemical reduction of intermediate products from biological process. Oxygen reduction reaction could not be avoided during nitrogen electrochemical reduction reaction since their similar reduction potential. But it led to decrease of oxygen concentration and therefore contribute to biological denitrification. Bacteria community tests also supported a dominant bacteria which could denitrify and use external electron.

Abstract:Nitrate-nitrogen (NO3-N) contaminating groundwater is an environmental issue in many areas, and is difficult to treat by simple processes. A bio-electrochemical reactor (BER) using copper wire and graphite plate was developed to purify the NO3-N-contaminated groundwater. The low (of 10 mA) and high (of 20 mA) electric currents were applied to the BERs, and various influent hardness levels from 20 to 80 mg/L as CaCO3 due to groundwater characteristics were supplied to clarify the total nitrogen removal efficiency and NO3-N removal mechanisms. In the BER-10, the bio-electrochemical reactions caused 85% of total nitrogen to be removed through heterotrophic and autohydrogenotrophic denitrification in the suspended sludge and biofilm. However, the chemical deposit occurring at the cathode from water hardness affected the decreasing denitrification performance; 12.6% of Mg and 8.8% of Ca elements were observed in the biofilm. The enhancement of electrochemical reactions in the BER-20 caused integrating electrochemical and bio-electrochemical reactions; the NO3-N was electrochemically reduced to NO2-N, and it was further biologically reduced to N2. A better total nitrogen removal of 95% was found; although, a larger deposit of Mg (22.8%) and Ca (10.8%) was observed. The relatively low dissolved H2 in the BER-20 confirmed that the deposit affected the decreasing gaseous H2 transfer and inhibition of autohydrogenotrophic denitrification in the suspended sludge. According to the microbial analysis, both heterotrophic and autohydrogenotrophic denitrification were obtained in the suspended sludge of both BERs; Nocadia (26.8%) was the most abundant genus in the BER-10, whereas Flavobacterium (27.1%) and Nocadia (25.0%) were the dominant genera in the BER-20.

Abstract:Bio-electrochemical system (BESs) showed a great potential for wastewater treatment. However, bottleneck problems, e. g. complex construction and high cost greatly limited the scaling-up of BES. To make BES to be an applicable wastewater treatment process, a novel up-flow bio-electrochemical reactor without membrane was developed. The cathode was placed under the anode for the reductive, toxic and refractory organic removal. In this study, the reactor was used for nitrobenzene reduction. The results showed that the efficiency of nitrobenzene reduction reached to 98% with the maximum nitrobenzene removal rate of 3.50 mol NB/m 3 TV d. The main product was aniline which could be easily biodegraded. The efficiency of aniline formation was 80%. Compared to the traditional anaerobic wastewater treatment methods, the nitrobenzene removal rate in up-flow bio-electrochemical reactor was 2 times faster. Compared to the traditional electrochemical methods for nitrobenzene removal, the cost of up-flow bio-electrochemical reactor reduced 10 times.

Abstract:The denitrification of municipal sludge was improved by combining biofilm process with the electro-chemical effect in a single novel reactor. Experiments in this reactor[electric current 60 mA, hydraulic retention time(HRTs) 6.0 h] showed that the removal of COD_ Cr, ammonia nitrogen and total nitrogen in the biofilm-electrode reactor were 2.5%, 1.2%, 14.9%, respectively, higher than those in a traditional biofilm reactor.
