Magnetic molecular imprinted polymers for in vitro controlled release and solid-phase extraction of dextromethorphan: synthesize, characterization, and application

Abstract:Herein, for the first time, the magnetic molecular imprinted polymers (MMIPs) were prepared for highly selective solid-phase extraction, and in controlled release of dextromethorphan at different pH values of 7.4 (simulated biological fluid), 1.0 (simulated gastric acid), and 6.8 (simulated intestinal fluid). The MMIPs were characterized by X-ray diffractometry, scanning electron microscopy, Fourier transform infrared spectroscopy, thermogravimetric analysis, energy-dispersive X-ray spectroscopy, and vibrating sample magnetometry. Variables affecting the performance of solid-phase extraction and sustained release of dextromethorphan are thoroughly investigated and their effects on the selectivity and efficiency of the whole process are discussed. Adsorption mechanism is fully expressed through Freundlich isotherm with a maximum adsorption capacity of 114. 8 mg/g and a pseudo-second-order kinetic model. The results of sustained release of dextromethorphan revealed that the M MIPs have a slower release rate and higher affinity toward dextromethorphan than the NMIPs owing to the selective interaction of DXM with imprinted cavities within nanoparticles. The analytical characteristics of the MMIPs-based SPE method were determined. The calibration graph was linear in the range of 0.4–200 µg L−1 with a detection limit of 0.1 µg L−1. The values of intra-day RSD% for five replicates of 20.0 and 50.0 µg L−1 of DXM were 1.8 and 2.5, respectively. Regarding the inter-day RSD%, 3.2 and 4.7% were obtained for 20 and 50 µgL−1 of DXM, respectively. The relative recoveries of DXM in spiked serum sample and pharmaceutical products were in the range 92–97%.
