Abstract:Abstract Dynamic light scattering (DLS) is a well known experimental approach uniquely suited for the characterization of small particles undergoing Brownian motion in randomly inhomogeneous turbid scattering medium, including water suspension, polymers in solutions, cells cultures, and so on. DLS is based on the illuminating of turbid medium with a coherent laser light and further analyzes the intensity fluctuations caused by the motion of the scattering particles. The DLS‐based spin‐off derivative techniques, such laser Doppler flowmetry (LDF), diffusing wave spectroscopy (DWS), laser speckle contrast imaging (LSCI), and Doppler optical coherence tomography (DOCT), are exploited widely for non‐invasive imaging of blood flow in brain, skin, muscles, and other biological tissues. The recent advancements in the DLS‐based imaging technologies in frame of their application for brain blood flow monitoring, skin perfusion measurements, and non‐invasive blood micro‐circulation characterization are overviewed. The fundamentals, breakthrough potential, and practical findings revealed by DLS‐based blood flow imaging studies, including the limitations and challenges of the approach such as movement artifacts, non‐ergodicity, and overcoming high scattering properties of studied medium, are also discussed. It is concluded that continued research and further technological advancements in DLS‐based imaging will pave the way for new exciting developments and insights into blood flow diagnostic imaging.
Static and dynamic light scattering by red blood cells: A numerical study

Abstract:Light scattering is a well-established experimental technique, which gains more and more popularity in the biological field because it offers the means for non-invasive imaging and detection. However, the interpretation of light-scattering signals remains challenging due to the complexity of most biological systems. Here, we investigate static and dynamic scattering properties of red blood cells (RBCs) using two mesoscopic hydrodynamics simulation methods-multi-particle collision dynamics and dissipative particle dynamics. Light scattering is studied for various membrane shear elasticities, bending rigidities, and RBC shapes (e.g., biconcave and stomatocyte). Simulation results from the two simulation methods show good agreement, and demonstrate that the static light scattering of a diffusing RBC is not very sensitive to the changes in membrane properties and moderate alterations in cell shapes. We also compute dynamic light scattering of a diffusing RBC, from which dynamic properties of RBCs such as diffusion coefficients can be accessed. In contrast to static light scattering, the dynamic measurements can be employed to differentiate between the biconcave and stomatocytic RBC shapes and generally allow the differentiation based on the membrane properties. Our simulation results can be used for better understanding of light scattering by RBCs and the development of new non-invasive methods for blood-flow monitoring.

Abstract:Quantitative measurements of intravascular microscopic dynamics, such as absolute blood flow velocity, shear stress and the diffusion coefficient of red blood cells (RBCs), are fundamental in understanding the blood flow behavior within the microcirculation, and for understanding why diffuse correlation spectroscopy (DCS) measurements of blood flow are dominantly sensitive to the diffusive motion of RBCs. Dynamic light scattering-optical coherence tomography (DLS-OCT) takes the advantages of using DLS to measure particle flow and diffusion within an OCT resolution-constrained three-dimensional volume, enabling the simultaneous measurements of absolute RBC velocity and diffusion coefficient with high spatial resolution. In this work, we applied DLS-OCT to measure both RBC velocity and the shear-induced diffusion coefficient within penetrating venules of the somatosensory cortex of anesthetized mice. Blood flow laminar profile measurements indicate a blunted laminar flow profile and the degree of blunting decreases with increasing vessel diameter. Measuring Capillary Flow Dynamics Using Interlaced Two-Photon Volumetric Scanning

Abstract:Two photon microscopy and optical coherence tomography (OCT) are two standard methods for measuring flow speeds of red blood cells in microvessels, particularly in animal models. However, traditional two photon microscopy lacks the depth of field to adequately capture the full volumetric complexity of the cerebral microvasculature and OCT lacks the specificity offered by fluorescent labeling. In addition, the traditional raster scanning technique utilized in both modalities requires a balance of image frame rate and field of view, which severely limits the study of RBC velocities in the microvascular network. Here, we overcome this by using a custom two photon system with an axicon based Bessel beam to obtain volumetric images of the microvascular network with fluorescent specificity. We combine this with a novel scan pattern that generates pairs of frames with short time delay sufficient for tracking red blood cell flow in capillaries. We track RBC flow speeds in 10 or more capillaries simultaneously at 1 Hz in a 237 µm × 237 µm × 120 µm volume and quantified both their spatial and temporal variability in speed. We also demonstrate the ability to track flow speed changes around stalls in capillary flow and measure to 300 µm in depth.
Cortical microvascular blood flow velocity mapping by combining dynamic light scattering optical coherence tomography and two-photon microscopy

Abstract:Significance: The accurate large-scale mapping of cerebral microvascular blood flow velocity is crucial for a better understanding of cerebral blood flow (CBF) regulation. Although optical imaging techniques enable both high-resolution microvascular angiography and fast absolute CBF velocity measurements in the mouse cortex, they usually require different imaging techniques with independent system configurations to maximize their performances. Consequently, it is still a challenge to accurately combine functional and morphological measurements to co-register CBF speed distribution from hundreds of microvessels with high-resolution microvascular angiograms. Aim: We propose a data acquisition and processing framework to co-register a large set of microvascular blood flow velocity measurements from dynamic light scattering optical coherence tomography (DLS-OCT) with the corresponding microvascular angiogram obtained using two-photon microscopy (2PM). Approach: We used DLS-OCT to first rapidly acquire a large set of microvascular velocities through a sealed cranial window in mice and then to acquire high-resolution microvascular angiograms using 2PM. The acquired data were processed in three steps: (i) 2PM angiogram coregistration with the DLS-OCT angiogram, (ii) 2PM angiogram segmentation and graphing, and (iii) mapping of the CBF velocities to the graph representation of the 2PM angiogram. Results: We implemented the developed framework on the three datasets acquired from the mice cortices to facilitate the coregistration of the large sets of DLS-OCT flow velocity measurements with 2PM angiograms. We retrieved the distributions of red blood cell velocities in arterioles, venules, and capillaries as a function of the branching order from precapillary arterioles and postcapillary venules from more than 1000 microvascular segments. Conclusions: The proposed framework may serve as a useful tool for quantitative analysis of large microvascular datasets obtained by OCT and 2PM in studies involving normal brain functioning, progression of various diseases, and numerical modeling of the oxygen advection and diffusion in the realistic microvascular networks.
Dynamic laser speckle imaging: beyond the contrast (Conference Presentation)

Abstract:Blood flow imaging is an essential part of biomedical research, particularly in the vascular and neurovascular physiology. It includes various imaging modalities, with laser speckle contrast imaging (LSCI) being one of the most extensively used tools for the rapid wide-field flow characterization. For years, the capability of LSCI to become a quantitative tool has been discussed. Being based on the contrast relation to the speckle correlation time, the method requires a robust model and its correct parametrization. Main uncertainties are (i) light scattering and particle motion regimes which define the form of the field autocorrelation function g1 and (ii) static scattering and speckle averaging effects on the intensity correlation function g2. Multi-exposure laser speckle contrast imaging and proper system calibration can solve the later issue, but in order to evaluate g1 form, one has to directly measure speckle autocorrelation. We introduce the dynamic laser speckle imaging (DLSI) as a new step in the wide-field speckle dynamics analysis. By utilizing a high-speed camera and recording backscattered light at more than 20000 frames per second we are able to measure the temporal intensity correlation (g2) in the mice cortex. We demonstrate that DLSI data can be used to estimate all parameters of the speckle autocorrelation model. By finding the best fit model for each pixel, we show that all three types of known g1 models can be found in the cortex and that the best fit model depends on the vessel size. Furthermore, we explore the commonly used model for the blood flow index and explain its deviations from the actual flow speed. We show that DLSI can be used to calibrate LSCI, thus solving contrast imaging problems and providing a lightweight quantitative tool for the blood flow imaging.

Beneath the Surface: Revealing Deep-Tissue Blood Flow in Human Subjects with Massively Parallelized Diffuse Correlation Spectroscopy

Abstract:Diffuse Correlation Spectroscopy (DCS) allows the label-free investigation of microvascular dynamics deep within living tissue. However, common implementations of DCS are currently limited to measurement depths of $\sim 1-1.5cm$, which can limit the accuracy of cerebral hemodynamics measurement. Here we present massively parallelized DCS (pDCS) using novel single photon avalanche detector (SPAD) arrays with up to 500x500 individual channels. The new SPAD array technology can boost the signal-to-noise ratio by a factor of up to 500 compared to single-pixel DCS, or by more than 15-fold compared to the most recent state-of-the-art pDCS demonstrations. Our results demonstrate the first in vivo use of this massively parallelized DCS system to measure cerebral blood flow changes at $\sim 2cm$ depth in human adults. We compared different modes of operation and applied a dual detection strategy, where a secondary SPAD array is used to simultaneously assess the superficial blood flow as a built-in reference measurement. While the blood flow in the superficial scalp tissue showed no significant change during cognitive activation, the deep pDCS measurement showed a statistically significant increase in the derived blood flow index of 8-12% when compared to the control rest state.

Abstract:We present a framework to coregister a large set of blood flow measurements from the cortical microvascular segments obtained by DLS-OCT with the microvascular angiogram obtained using TPM. By using this framework, we estimated the distributions of the mean RBC velocities in arterioles, venules, and capillaries, and as a function of the branching order from over 1,000 microvascular segments. The developed tools will help with more quantitative analysis of the large data sets obtained by OCT and TPM in the studies involving normal brain functioning, progression of various diseases, and numerical modeling of the oxygen transport in the microvascular networks.

Abstract:We describe a new light-sheet microscopy method for fast, large-scale volumetric imaging. Combining synchronized scanning illumination and oblique imaging over cleared, thick tissue sections in smooth motion, our approach achieves high-speed 3D image acquisition of an entire mouse brain within 2 hours, at a resolution capable of resolving synaptic spines. It is compatible with immunofluorescence labeling, enabling flexible cell-type specific brain mapping, and is readily scalable for large biological samples such as primate brain.

Abstract:The dynamic light scattering methods are widely used in biomedical diagnostics involving evaluation of blood flow. However, there exist some difficulties in quantitative interpretation of backscattered light signals from the viewpoint of diagnostic information. This study considers the application of the high-speed videocapillaroscopy (VCS) method that provides the direct measurement of the red blood cells (RBCs) velocity into a capillary. The VCS signal presents true oscillation nature of backscattered light caused by moving RBCs. Thus, the VCS signal can be assigned as a reference one with respect to more complicated signals like in laser Doppler flowmetry (LDF). An essential correlation between blood flow velocity oscillations in a separate human capillary and the integral perfusion estimate obtained by the LDF method has been found. The observation of blood flow by the VCS method during upper arm occlusion has shown emergence of the reverse blood flow effect in capillaries that corresponds to the biological zero signal in the LDF. The reverse blood flow effect has to be taken into account in interpretation of LDF signals.

Abstract:Dynamic light scattering (DLS) and laser speckle contrast imaging (LSCI) are closely related techniques that exploit the statistics of speckle patterns, which can be utilized to measure cerebral blood flow (CBF). Conventionally, the temporal speckle intensity auto-correlation function g2t(τ) is calculated in DLS, while the spatial speckle contrast Ks is calculated in LSCI measurements. Due to the rapid development of CMOS detection technology with increased camera frame rates while still maintaining a large number of pixels, the ensemble or spatial average of g2s(τ) as well as the temporal contrast Kt can be easily calculated and utilized to quantify CBF. Although many models have been established, a proper summary is still lacking to fully characterize DLS and LSCI measurements for spatial and temporal statistics, laser coherence properties, various motion types, etc. As a result, there are many instances where theoretical models are misused. For instance, mathematical formulas derived in the diffusive regime or for ergodic systems are sometimes applied to small animal brain measurements, e.g., mice brains, where the assumptions are not valid. Therefore, we aim to provide a review of the speckle theory for both DLS and LSCI measurements with detailed derivations from first principles, taking into account non-ergodicity, spatial and temporal statistics of speckles, scatterer motion types, and laser coherence properties. From these calculations, we elaborate on the differences between spatial and temporal averaging for DLS and LSCI measurements that are typically ignored but can result in inaccurate measurements of blood flow, particularly the spatially varying nature of the static component in g2t(τ) and Kt. We also obtained g2s(τ) maps in in vivo mouse brain measurements using high frame rate CMOS cameras which have not been demonstrated before, and compared with g2t(τ) and Ks,t. This work provides a useful guide for choosing the correct model to analyze spatial and temporal speckle statistics in in-vivo DLS and LSCI measurements.

Abstract:Abstract Large-scale imaging of brain activity with high spatio-temporal resolution is crucial for advancing our understanding of brain function. The existing neuroimaging techniques are largely limited by restricted field of view, slow imaging speed, or otherwise do not have the adequate spatial resolution to capture brain activities on a capillary and cellular level. To address these limitations, we introduce fluorescence localization microscopy aided with sparsely-labeled red blood cells for cortex-wide morphological and functional cerebral angiography with 4.9 μm spatial resolution and 1 s temporal resolution. When combined with fluorescence calcium imaging, the proposed method enables extended recordings of stimulus-evoked neuro-vascular changes in the murine brain while providing simultaneous multiparametric readings of intracellular neuronal activity, blood flow velocity/direction/volume, and vessel diameter. Owing to its simplicity and versatility, the proposed approach will become an invaluable tool for deciphering the regulation of cortical microcirculation and neurovascular coupling in health and disease.

Abstract:Fast noninvasive probing of spatially varying decorrelating events, such as cerebral blood flow beneath the human skull, is an essential task in various scientific and clinical settings. One of the primary optical techniques used is diffuse correlation spectroscopy (DCS), whose classical implementation uses a single or few single-photon detectors, resulting in poor spatial localization accuracy and relatively low temporal resolution. Here, we propose a technique termed C lassifying R apid decorrelation E vents via P arallelized single photon d E tection (CREPE), a new form of DCS that can probe and classify different decorrelating movements hidden underneath turbid volume with high sensitivity using parallelized speckle detection from a 32 × 32 pixel SPAD array. We evaluate our setup by classifying different spatiotemporal-decorrelating patterns hidden beneath a 5 mm tissue-like phantom made with rapidly decorrelating dynamic scattering media. Twelve multi-mode fibers are used to collect scattered light from different positions on the surface of the tissue phantom. To validate our setup, we generate perturbed decorrelation patterns by both a digital micromirror device (DMD) modulated at multi-kilo-hertz rates, as well as a vessel phantom containing flowing fluid. Along with a deep contrastive learning algorithm that outperforms classic unsupervised learning methods, we demonstrate our approach can accurately detect and classify different transient decorrelation events (happening in 0.1-0.4 s) underneath turbid scattering media, without any data labeling. This has the potential to be applied to non-invasively monitor deep tissue motion patterns, for example identifying normal or abnormal cerebral blood flow events, at multi-Hertz rates within a compact and static detection probe.

Abstract:Significance Characterizing blood flow by tracking individual red blood cells as they move through vessels is essential for understanding vascular function. With high spatial resolution, two-photon fluorescence microscopy is the method of choice for imaging blood flow at the cellular level. However, its application is limited to a low flow speed regimen in anesthetized animals by its slow focus scanning mechanism. Using an ultrafast scanning module, we demonstrated two-photon fluorescence imaging of blood flow at 1,000 two-dimensional frames and 1,000,000 one-dimensional line scans per second in the brains of awake mice. These ultrafast measurements enabled us to study hemodynamic and fluid mechanical regimens beyond the reach of conventional methods.

Time-domain diffuse correlation spectroscopy (TD-DCS) enables noninvasive blood flow measurements by analyzing temporal intensity fluctuations of the scattered light. With time-of-flight (TOF) resolution, TD-DCS should decompose the blood flow at different sample depths. For example, in the human head, it allows us to distinguish blood flows in the scalp, skull, or cortex. However, the tissues are typically polydisperse. So photons with a similar TOF can be scattered from structures that move at different speeds. Here, we introduce a novel approach that takes this problem into account and allows us to quantify the TOF-resolved blood flow of human tissue accurately. We apply this approach to monitor the blood flow index in the human forearm in vivo during the cuff occlusion challenge. We detect depth-dependent reactive hyperemia. Finally, we applied a controllable pressure to the human forehead in vivo to demonstrate that our approach can separate superficial from the deep blood flow. Our results can be beneficial for neuroimaging sensing applications that require short interoptode separation.

Abstract:The spinal cord (SC) is crucial for a myriad of somatosensory, autonomic signal processing, and transductions. Understanding the SC vascular structure and function thus plays an integral part in neuroscience and clinical research. However, the dense layers of myelinated ascending axons on the dorsal side inconveniently grant the SC tissue with high optical scattering property, which significantly hinders the imaging depth of the SC vasculature in vivo. Commonly used antiscattering techniques such as multiphoton fluorescence microscopy have low imaging speed and cannot capture the rapid vascular particle flow without significant motion blur. Here, advantage of the high penetration of near‐infrared (NIR)‐II fluorescence is taken to demonstrate a deep SC vascular structural image stack up to 350 µm, comparable to two‐photon microscopy. Furthermore, the red blood cells are labelled with the clinically approved NIR dye indocyanine. The combination of a fast NIR camera and indocyanine green‐red blood cells (RBCs) makes it possible to attain high‐speed 100 frame‐per‐second NIR‐II imaging to identify the corresponding changes in RBC velocity during the external hind leg stimulus. For the first time, it is established that the NIR‐II region would be a promising spectral window for SC imaging. NIR‐II fluorescence microscopy has excellent potential for clinical and basic science research on SC.

Abstract:Both the ultrastructures and dynamics of living erythrocyte membranes provide critical criteria for clinical diagnostics. However, it is challenging to simultaneously visualize these features at the single-molecule level due to the rigid photophysical requirements of different single-molecule imaging techniques. Herein, we rationally developed a far-red boron dipyrromethene membrane (BDP-Mem) probe that not only retained consistent and intensive single-molecule emission but also possessed the capability to photoswitch on cellular membranes. We also constructed a microfluidic platform for the noninvasive trapping and long-term imaging of nonadherent erythrocytes. By combining these advantageous technologies, super-resolution reconstruction and single-molecule tracking of living human RBC membranes were achieved at the molecular scale in a high-throughput fashion. Our integrated paradigm defines a quantitative approach for analyzing living RBC membranes under physiological and pathological conditions, improving imaging precisions and revealing new perspectives for future disease diagnostic approaches. ### Competing Interest Statement The authors have declared no competing interest.
Programmable scanning diffuse speckle contrast imaging of cerebral blood flow

Abstract:Significance: Cerebral blood flow (CBF) imaging is crucial for diagnosing cerebrovascular diseases. However, existing large neuroimaging techniques with high cost, low sampling rate, and poor mobility make them unsuitable for continuous and longitudinal CBF monitoring at the bedside. Aim: This study aimed to develop a low-cost, portable, programmable scanning diffuse speckle contrast imaging (PS-DSCI) technology for fast, high-density, and depth-sensitive imaging of CBF in rodents. Approach: The PS-DSCI employed a programmable digital micromirror device (DMD) for remote line-shape laser (785 nm) scanning on tissue surface and synchronized a 2D camera for capturing boundary diffuse laser speckle contrasts. New algorithms were developed to address deformations of line-shape scanning, thus minimizing CBF reconstruction artifacts. The PS-DSCI was examined in head-simulating phantoms and adult mice. Results: The PS-DSCI enables resolving Intralipid particle flow contrasts at different tissue depths. In vivo experiments in adult mice demonstrated the capability of PS-DSCI to image global/regional CBF variations induced by 8% CO2 inhalation and transient carotid artery ligations. Conclusions: Compared to conventional point scanning, the line scanning in PS-DSCI significantly increases spatiotemporal resolution. The high sampling rate of PS-DSCI is crucial for capturing rapid CBF changes while high spatial resolution is important for visualizing brain vasculature.
