Abstract:An amperometric glucose dehydrogenase biosensor is reported. It is made up of toluidine blue (TB) modified carbon paste microelectrode and enzyme film by glucose dehydrogenase immobilized on silk fibroin. In pH 7.0 NaOH-NaH 2PO 4 medium, under the concentration of nicotinamide adenine dinucleotid (NAD + ) being 1.04 × 10 -3 mol/L, the biosensor has a linear response in the range of 1.0 × 10 -4 ∼ 3.2 × 10 -3 mol/L for concentration of glucose. The response time is 20 s. The lower detection limit of microsensor is 4 × 10 -5 mol/L. The biosensor has advantages of small volume, better stability and repeat-ability.

Abstract:In this paper, we first report the construction of Nafion/glucose oxidase (GOD)/xylose dehydrogenase displayed bacteria (XDH-bacteria)/multiwalled carbon nanotubes (MWNTs) modified electrode for simultaneous voltammetric determination of d-glucose and d-xylose. The optimal conditions for the immobilized enzymes were established. Both enzymes retained their good stability and activities. In the mixture solution of d-glucose and d-xylose containing coenzyme NAD+ (the oxidized form of nicotinamide adenine dinucleotide), the Nafion/GOD/XDH-bacteria/MWNTs modified electrode exhibited quasi-reversible oxidation-reduction peak at −0.5V (vs. saturated calomel electrode, SCE) originating from the catalytic oxidation of d-glucose, and oxidation peak at +0.55V(vs. SCE) responding to the oxidation of NADH (the reduced form of nicotinamide adenine dinucleotide) by the carbon nanotubes, where NADH is the resultant product of coenzyme NAD+ involved in the catalysis of d-xylose by XDH-displayed bacteria. For the proposed biosensor, cathodic peak current at −0.5V was linear with the concentration of d-glucose within the range of 0.25–6mM with a low detection limit of 0.1mM d-glucose (S/N=3), and the anodic peak current at +0.55V was linear with the concentration of d-xylose in the range of 0.25∼4mM with a low detection limit of 0.1mM d-xylose (S/N=3). Further, d-xylose and d-glucose did not interfere with each other. 300-fold excess saccharides including d-maltose, d-galactose, d-mannose, d-sucrose, d-fructose, d-cellobiose, and 60-fold excess l-arabinose, and common interfering substances (100-fold excess ascorbic acid, dopamine, uric acid) as well as 300-fold excess d-xylitol did not affect the detection of d-glucose and d-xylose (both 1mM). Therefore, the proposed biosensor is stable, specific, reproducible, simple, rapid and cost-effective, which holds great potential in real applications.

Abstract:Glutathione (GSH) exhibits antioxidant and integrated detoxification effects, providing protection to the liver. Nevertheless, in cases of liver damage or cancerization, the GSH content experiences significant fluctuations. Consequently, GSH can serve as an effective endogenous molecule for aiding the diagnosis and pathological researches of liver diseases. In this study, a GSH-responsive conductive hydrogel was prepared for the quantitative detection of GSH. Through measuring the high-frequency impedance change of the hydrogel during the reaction, the detection of GSH in a wide range (0.1-6 mM) is realized, and the sensor has high sensitivity and good anti-interference performance.
Glucose Biosensor Based on a Platinum Electrode Modified with Rhodium Nanoparticles and with Glucose Oxidase Immobilized on Gold Nanoparticles

Abstract:We have developed an enzymatic glucose biosensor that is based on a flat platinum electrode which was covered with electrophoretically deposited rhodium (Rh) nanoparticles and then sintered to form a large surface area. The biosensor was obtained by depositing glucose oxidase (GOx), Nafion, and gold nanoparticles (AuNPs) on the Rh electrode. The electrical potential and the fractions of Nafion and GOx were optimized. The resulting biosensor has a very high sensitivity (68.1 μA mM−1 cm−2) and good linearity in the range from 0.05 to 15 mM (r = 0.989). The limit of detection is as low as 0.03 mM (at an SNR of 3). The glucose biosensor also is quite selective and is not interfered by electroactive substances including ascorbic acid, uric acid and acetaminophen. The lifespan is up to 90 days. It was applied to the determination of glucose in blood serum, and the results compare very well with those obtained with a clinical analyzer.

Abstract:To minimize background interference in electrochemical enzymatic biosensors employing electron mediators, it is essential for the electrochemical oxidation of electroactive interfering species (ISs), such as ascorbic acid (AA), to proceed slowly, and for the redox reactions between electron mediators and ISs to occur at a low rate. In this study, we introduce a novel combination of a working electrode and an electron mediator that effectively mitigates interference effects. Compared to commonly used electrodes such as Au, glassy carbon, and indium tin oxide (ITO), boron-doped diamond (BDD) electrodes demonstrate significantly lower anodic current (i.e., lower background levels) in the presence of AA. Additionally, menadione (MD) exhibits notably slower reactivity with AA compared to other electron mediators such as Ru(NH 3 ) 6 3+ , 4-amino-1-naphthol, and 1,4-naphthoquinone, primarily due to the lower formal potential of MD compared to AA. This synergistic combination of BDD electrode and MD is effectively applied in three biosensors: (i) glucose detection using electrochemical-enzymatic (EN) redox cycling, (ii) glucose detection using electrochemical-enzymatic-enzymatic (ENN) redox cycling, and (iii) lactate detection using ENN redox cycling. Our developed approach significantly outperforms the combination of ITO electrode and MD in minimizing IS interference. Glucose in artificial serum can be detected with detection limits of ~ 20 μM and ~ 3 μM in EN and ENN redox cycling, respectively. Furthermore, lactate in human serum can be detected with a detection limit of ~ 30 μM. This study demonstrates sensitive glucose and lactate detection with minimal interference, eliminating the need for (bio)chemical agents to remove interfering species.

Abstract:Glucose is one of the most important monosaccharides found in the food, as a part of more complex structures, which is a primary energy source for the brain and body. Thus, the monitoring of glucose concentration is more important in food and biological samples in order to maintain a healthy lifestyle. Herein, an electrochemical glucose biosensor was fabricated by immobilization of glucose oxidase (GOX) onto poly(3,4-ethylenedioxythiophene):4-sulfocalix [4]arene (PEDOT:SCX)/MXene modified electrode. For this purpose, firstly, PEDOT was synthesized in the presence of SCX (counterion) by the chemical oxidative method. Secondly, MXene (a 2D layered material) was synthesized by using a high-temperature furnace under a nitrogen atmosphere. After that, PEDOT:SCX/MXene (1:1) dispersion was prepared by ultrasonication which was later utilized to prepare PEDOT:SCX/MXene hybrid film. A successful formation of PEDOT:SCX/MXene film was confirmed by HR-SEM, Fourier transform infrared (FT-IR), and Raman spectroscopies. Due to the biocompatibility nature, successful immobilization of GOX was carried out onto chitosan modified PEDOT:SCX/MXene/GCE. Moreover, the electrochemical properties of PEDOT:SCX/MXene/GOX/GCE was studied through cyclic voltammetry and amperometry methods. Interestingly, a stable redox peak of FAD-GOX was observed at a formal potential of –0.435 V on PEDOT:SCX/MXene/GOX/GCE which indicated a direct electron transfer between the enzyme and the electrode surface. PEDOT:SCX/MXene/GOX/GCE also exhibited a linear response against glucose concentrations in the linear range from 0.5 to 8 mM. The effect of pH, sensors reproducibility, and repeatability of the PEDOT:SCX/MXene/GOX/GCE sensor were studied. Finally, this new biosensor was successfully applied to detect glucose in commercial fruit juice sample with satisfactory recovery.

Abstract:We developed a stable, sensitive electrochemiluminescence (ECL) biosensor based on the synthesis of a new sol-gel material with the ion-exchange capacity sol-gel to coimmobilize the Ru(bpy)(3)(2+) and enzyme. The partial sulfonated (3-mercaptopropyl)-trimethoxysilane sol-gel (PSSG) film acted as both an ion exchanger for the immobilization of Ru(bpy)(3)(2+) and a matrix to immobilize gold nanoparticles (AuNPs). The AuNPs/PSSG/Ru(bpy)(3)(2+) film modified electrode allowed sensitive the ECL detection of NADH as low as 1 nM. Such an ability of AuNPs/PSSG/Ru(bpy)(3)(2+) film to promote the electron transfer between Ru(bpy)(3)(2+) and the electrode suggested a new, promising biocompatible platform for the development of dehydrogenase-based ECL biosensors. With alcohol dehydrogenase (ADH) as a model, we then constructed an ethanol biosensor, which had a linear range of 5 microM to 5.2 mM with a detection limit of 12nM.

Abstract:Rapid and accurate detection of glucose is of great significance for diabetic management. Highly sensitive glucose sensors promise to achieve noninvasive detection technology, enabling more convenient and efficient means for large-scale screening and long-term dynamic monitoring of diabetes patients. In this work, we demonstrate a sensitive glucose electrochemical biosensor through the synergetic labelling strategy utilizing PbS colloidal quantum dots (CQDs) and Au nanospheres (AuNSs). The PbS CQDs/AuNSs/glucose oxidase (GOx) mixture could be stably immobilized on the carbon electrode surface via the one-step dip-coating method. The electrochemical biosensor employing PbS CQDs/AuNSs/GOx-modified electrode integrates the functions of specific molecule recognition, signal transduction as well as signal amplification. The sensor is capable of transducing the glucose enzyme-catalyzed reaction into significant current signals, exhibiting a good linear response in the glucose concentration range of 0.1 μM—10 mM with the limit of detection being 1.432 nM.

Abstract:A glucose biosensor was layer-by-layer assembled on a modified glassy carbon electrode (GCE) from a nanocomposite of NAD(P)+-dependent glucose dehydrogenase, aminated polyethylene glycol (mPEG), carboxylic acid-functionalized multi-wall carbon nanotubes (fMWCNTs), and ionic liquid (IL) composite functional polymers. The electrochemical electrode was denoted as NF/IL/GDH/mPEG-fMWCNTs/GCE. The composite polymer membranes were characterized by cyclic voltammetry, ultraviolet-visible spectrophotometry, electrochemical impedance spectroscopy, scanning electron microscopy, and transmission electron microscopy. The cyclic voltammogram of the modified electrode had a pair of well-defined quasi-reversible redox peaks with a formal potential of −61 mV (vs. Ag/AgCl) at a scan rate of 0.05 V s−1. The heterogeneous electron transfer constant (ks) of GDH on the composite functional polymer-modified GCE was 6.5 s−1. The biosensor could sensitively recognize and detect glucose linearly from 0.8 to 100 μM with a detection limit down to 0.46 μM (S/N = 3) and a sensitivity of 29.1 nA μM−1. The apparent Michaelis–Menten constant (Kmapp) of the modified electrode was 0.21 mM. The constructed electrochemical sensor was compared with the high-performance liquid chromatography method for the determination of glucose in commercially available glucose injections. The results demonstrated that the sensor was highly accurate and could be used for the rapid and quantitative determination of glucose concentration.

Synergy of electrocatalysis on photoelectrocatalysis and amperometric determination of trace glucose

Abstract:An amperometric method with synergistic electrocatalysis on photoelectrocatalysis was proposed, and it was used for the detection of trace glucose concentrations for the first time. Under illumination of an external xenon lamp light source, the electrocatalytic oxidation reaction of glucose on a Co3O4-modified electrode produced a synergistic effect on the photoelectrochemical reaction of glucose. The separation of electron–hole pairs generated by the photoelectrochemical reaction was promoted, and the photoelectric conversion efficiency was improved. As a result, the photoelectric current increased, and the sensitivity of the amperometric method was enhanced by about 9 times. The mechanism of synergistic electrocatalysis on photoelectrocatalysis was studied. The method can be used for the determination of glucose with a linear concentration range of 1.0 × 10−8 M to 3.5 × 10−7 M. The sensitivity was 7,753 μA cm−2 mM−1, and the detection limit was 3.6 × 10−9 M. Compared with conventional photoelectrochemical analysis methods, the chronoamperometric method has higher sensitivity and lower detection limit. The method has been used for the direct determination of trace amounts of glucose in saliva and sweat.

Abstract:This study demonstrates a facile electrochemical method for simultaneous and selective on-line measurements of glucose and lactate in the brain of freely moving rats through integration of selective electrochemical detection with in vivo microdialysis. The selective electrochemical detection is accomplished by using oxidases as the specific sensing unit and prussian blue (PB) as the electrocatalyst for the reduction and thus for the determination of H(2)O(2) generated from the oxidase-catalyzed reactions in terms of its excellent electrocatalytic activity toward H(2)O(2) reduction. The uses of "artificial peroxidase" (i.e., PB) in this work to replace "natural peroxidase" (i.e., horseradish peroxidase) used in the previous on-line electrochemical methods essentially enables the method developed here to be facile but selective for the simultaneous and on-line measurements of glucose and lactate virtually interference-free from ascorbic acid and other electroactive species coexisting in the brain. Moreover, the dual oxidase/PB-based biosensors suffer from little cross-talk and exhibit a good stability and reproducibility. The basal levels of glucose and lactate in the microdialysate from the striatum of the freely moving rats are determined to be 200 +/- 30 and 400 +/- 50 microM (n = 3), respectively. The method demonstrated here is facile but reliable and durable and may find some interesting physiological and pathological applications.
Nitrogen-doped graphene quantum dots coated with gold nanoparticles for electrochemiluminescent glucose detection using enzymatically generated hydrogen peroxide as a quencher

Abstract:Nitrogen-doped graphene quantum dots (N-GQDs) were prepared from dicyandiamide and then used as both an electrochemiluminescence (ECL) emitter and a reductant to produce gold nanoparticles (Au-N-GQDs) on their surface without using any reagent. In order to avoid resonance energy transfer, the Au-N-GQDs were stabilized with chitosan. Transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy (EDX), UV-vis spectroscopy (UV-vis) and ECL methods were used to characterize the nanocomposite. The materials was placed on a glassy carbon electrode (GCE), and the ECL signals are found to be strongly quenched by hydrogen peroxide that is enzymatically produced by oxidation of glucose. With the applied typical potential of -1.7 V, the ECL of the Au-N-GQDs modified GCE decreases linearly in the 10 nM to 5.0 μM glucose concentration range, and the lower detection limit is 3.3 nM. The influence of H2O2 to the signal has been discussed and a possible mechanism has been presented. Abstract:Glucose is a known energy source that gives insights into cellular metabolism by glycolysis, biosynthesis and glycogen for storage. Investigation of glucose uptake in immune cells requires highly expensive tags and imaging techniques, however, we developed an amine (NH 2 ) nobbled Platinum (Pt)-Silver vanadate (AgVO 3 ) sensor platform combined with 3-mercapto phenylboronic acid (MPBA) recognition element for an enzyme-free glucose sensor. Introducing the amine groups into the Pt surface protects the stability of the Pt and extends the longevity of the electrode. A slightly acidic pH-driven boronate ester formation between glucose and boronic acid mechanism and the glucose combined electronic structure of MPBA/NH 2 @Pt/AgVO 3 with its adsorption energy (−2.1 eV) has been scrutinized via density functional theory. Accelerated electron transfer ( k s = 4.69 s −1 ) driven electrochemical oxidation of glucose sensor showed the higher detection limit (LOD) of 61.3 μM and the linear dynamic range from 0.05 to 22.0 mM. Glucose uptake assay of macrophage phenotypes between the healthy and insulin-resistant types provides an in-depth understanding of how glucose impairment affects the macrophage phenotypes. We also observed the M1-type macrophages have uptaken significantly larger amounts of glucose than the others (M0 and M2).

Abstract:Accurate determination of glucose in biological samples is of great significance for healthcare monitoring and medical diagnosis. Herein we reported an economical "signal-on" glucose electrochemiluminescence (ECL) enzyme biosensor for ultrasensitive detection of glucose based on vertically-ordered mesoporous silica films (VMSF) covalently modified with a monolayer of glucose dehydrogenase (GDH). In the presence of glucose, β -nicotinamide adenine dinucleotide sodium salt (NAD + )-dependent GDH on the surface of VMSF catalyzes the oxidation of glucose and β -nicotinamide adenine dinucleotide (NADH) reduced from NAD + is generated concomitantly in situ. The access of NADH to underlying electrode surface is prohibited by ultrasmall nanopores, but it can act as a co-reactant to promote the ECL intensity of Ru(bpy) 3 2+ by the free diffusion of Ru-related species, exhibiting a unique catalytic route. By recording the ECL intensity of Ru(bpy) 3 2+ electrostatically amplified by VMSF, the present ECL enzyme biosensor allows the ultrasensitive detection of glucose substrate with a wide linear range (10 nM to 1 mM) and a rather low limit of detection (1.5 nM). Furthermore, the developed ECL enzyme biosensor is capable of anti-fouling and accurate detection of glucose in human serum and artificial sweat without tedious preprocessing, which provides a ECL enzyme sensing platform for ultrasensitive detection of a variety of substrates of interest by simply tailoring the recognition elements.

Non-Enzymatic Detection of Glucose in Neutral Solution Using PBS-Treated Electrodeposited Copper-Nickel Electrodes

Abstract:Transition metals have been explored extensively for non-enzymatic electrochemical detection of glucose. However, to enable glucose oxidation, the majority of reports require highly alkaline electrolytes which can be damaging to the sensors and hazardous to handle. In this work, we developed a non-enzymatic sensor for detection of glucose in near-neutral solution based on copper-nickel electrodes which are electrochemically modified in phosphate-buffered saline (PBS). Nickel and copper were deposited using chronopotentiometry, followed by a two-step annealing process in air (Step 1: at room temperature and Step 2: at 150 °C) and electrochemical stabilization in PBS. Morphology and chemical composition of the electrodes were characterized using scanning electron microscopy and energy-dispersive X-ray spectroscopy. Cyclic voltammetry was used to measure oxidation reaction of glucose in sodium sulfate (100 mM, pH 6.4). The PBS-Cu-Ni working electrodes enabled detection of glucose with a limit of detection (LOD) of 4.2 nM, a dynamic response from 5 nM to 20 mM, and sensitivity of 5.47 ± 0.45 μA cm-2/log10(mole.L-1) at an applied potential of 0.2 V. In addition to the ultralow LOD, the sensors are selective toward glucose in the presence of physiologically relevant concentrations of ascorbic acid and uric acid spiked in artificial saliva. The optimized PBS-Cu-Ni electrodes demonstrate better stability after seven days storage in ambient compared to the Cu-Ni electrodes without PBS treatment.

Abstract:The direct electrochemistry of glucose oxidase (GOD) adsorbed on a colloidal gold modified carbon paste electrode was investigated. The adsorbed GOD displayed a pair of redox peaks with a formal potential of −(449±1) mV in 0.1 M pH 5.0 phosphate buffer solution. The response showed a surface-controlled electrode process with an electron transfer rate constant of (38.9±5.3)/s determined in the scan rate range from 10 to 100 mV/s. GOD adsorbed on gold colloid nanoparticles maintained its bioactivity and stability. The immobilized GOD could electrocatalyze the reduction of dissolved oxygen and resulted in a great increase of the reduction peak current. Upon the addition of glucose, the reduction peak current decreased, which could be used for glucose detection with a high sensitivity (8.4 μA/mM), a linear range from 0.04 to 0.28 mM and a detection limit of 0.01 mM at a signal-to-noise ratio of 3σ. The sensor could exclude the interference of commonly coexisted uric and ascorbic acid.

Abstract:Alloy nanoparticles of the type PtxFe (where x is 1, 2 or 3) were synthesized by coreduction with sodium borohydride in the presence of carbon acting as a chemical support. The resulting nanocomposites were characterized by scanning electron microscopy and X-ray diffraction. The nanocomposite was placed on a glassy carbon electrode, and electrochemical measurements indicated an excellent catalytic activity for the oxidation of glucose even a near-neutral pH values and at a working voltage as low as 50 mV (vs. SCE). Under optimized conditions, the sensor responds to glucose in the 10.0 μM to 18.9 mM concentration range and with a 3.0 μM detection limit (at an S/N ratio of 3). Interferences by ascorbic acid, uric acid, fructose, acetamidophenol and chloride ions are negligible.Graphical AbstractNonenzymatic sensing of glucose is demonstrated at neutral pH values and low working potential using a glassy carbon electrode modified with platinum-iron alloy nanoparticles on a carbon support.

Abstract:Glucose is the principal source of energy for humans and its quantification in physiological samples can diagnose or prevent diseases. Commonly, glucose determination is based on spectrophotometric-enzymatic techniques, but at least since a decade ago, electroanalytical strategies have emerged as promising alternatives providing accuracy and precision in the determination of biomolecules. This review focuses on the development of non-enzymatic methodologies based on modified electrochemical sensors with Metal-Organic Frameworks (MOF) for glucose detection sensors in physiological samples (blood and urine). Glassy carbon electrodes (GCE), carbon paste electrodes (CPE), and screen-printed electrodes (SPE) are the main transductors modified with MOF for the electrochemical oxidation of glucose, and the maximum anodic peak current is taken to the analytical signal. The reported results demonstrated that this electroanalytical approach represents a viable alternative for fast and confident analysis of the glucose molecule.
