Abstract:Nanoparticle-mediated laser-induced breakdown (LIB) can be used for Nanoparticle synthesis, cell nanosurgery and laser-induced breakdown spectroscopy (LIBS). To investigate the photo-thermal conversion of gold nanoparticles during pulsed laser irradiation, the electron-phonon two-temperature model was established in this study. The impact of laser energy density and pulse width on the thermal conversion and morphology change of gold nanorods were investigated and compared with experimental observations. The results show that the melting threshold of gold nanorods under nanosecond laser irradiation is about twenty times that of femtosecond laser irradiation. The mechanisms of nanorod fragmentation are different between fem to and nanosecond laser irradiation: particle melting is more likely to occur in nanosecond laser irradiation, while the Coulomb explosion is highly likely to occur in femtosecond laser irradiation.

Abstract:Objective: Gold nanorods (GNRs) have emerged as versatile nanoparticles with unique properties, holding promise in various modalities of cancer treatment through drug delivery and photothermal therapy. In the rapidly evolving field of nanoparticle radiosensitization (NPRS) for cancer therapy, this study assessed the potential of gold nanorods as radiosensitizing agents by quantifying the key features of NPRS, such as secondary electron emission and dose enhancement, using Monte Carlo simulations. Approach: Employing the TOPAS track structure code, we conducted a comprehensive evaluation of the radiosensitization behavior of spherical gold nanoparticles and gold nanorods. We systematically explored the impact of nanorod geometry (in particular size and aspect ratio) and orientation on secondary electron emission and deposited energy ratio, providing validated results against previously published simulations. Main Results: Our findings demonstrate that gold nanorods exhibit comparable secondary electron emission to their spherical counterparts. Notably, nanorods with smaller surface-area-to-volume ratios (SA:V) and alignment with the incident photon beam proved to be more efficient radiosensitizing agents, showing superiority in emitted electron fluence. However, in the microscale, the deposited energy ratio (DER) was not markedly influenced by the SA:V of the nanorod. Additionally, our findings revealed that the geometry of gold nanoparticles has a more significant impact on the emission of M-shell Auger electrons (with energies below 3.5 keV) than on higher-energy electrons. Significance: This research investigated the radiosensitization properties of gold nanorods, positioning them as promising alternatives to the more conventionally studied spherical gold nanoparticles in the context of cancer research. With increasing interest in multimodal cancer therapy, our findings have the potential to contribute valuable insights into the perspective of gold nanorods as effective multipurpose agents for synergistic photothermal therapy and radiotherapy. Future directions may involve exploring alternative metallic nanorods as well as further optimizing the geometry and coating materials, opening new possibilities for more effective cancer treatments.

Synthesis of Small Gold Nanorods and Their Subsequent Functionalization with Hairpin Single Stranded DNA

Abstract:Small gold nanorods have a significantly large absorption/scattering ratio and are especially beneficial in exploiting photothermal effects, for example in photothermal therapy and remote drug release. This work systematically investigates the influence of growth conditions on the size, growth yield, and stability of small gold nanorods. The silver-assisted seed-mediated growth method was optimized to synthesize stable small gold nanorods with a high growth yield (>85%). Further study on the influence of silver ions on the growth facilitates the growth of small gold nanorods with tunable longitudinal surface plasmon resonance from 613 to 912 nm, with average dimensions of 13–25 nm in length and 5–6 nm in diameter. Moreover, the small gold nanorods were successfully functionalized with thiol-modified hairpin oligonucleotides (hpDNA) labeled with Cy5. Fluorescence intensity measurements show an increase in the presence of target DNA and an enhanced signal/background ratio when the longitudinal surface plasmon resonance of small gold nanorods overlaps with the excitation and emission wavelength of Cy5. This coincides with a reduced fluorescence lifetime of Cy5 in the hairpin structure, indicating surface plasmon resonance-enhanced energy transfer to the small gold nanorods. This study may provide insight on the synthesis and functionalization of small gold nanorods in biomedical sensing and therapy.

Abstract:The plasmon-based photothermal effect of gold nanorod (GNR) has undergone the most systematic investigation for cancer therapy in the biomedical realm. In recent years, gold nanostar (GNS) has come into sight with its attractive ability to transduce electromagnetic radiation into heat. Understanding photothermal conversion efficiency is thus becoming more important for the selection of suitable materials for photothermal therapy. In this article, we investigated systematically the photothermal conversion efficiency and the molar heating rate of GNS and GNR in three groups (S-group, M-group and L-group, representing groups of nanostructures with central extinctions at shorter, medium and longer wavelengths, respectively), to better understand the behaviour of GNS and GNR in the field of photothermal therapy. In the M-group and L-group, the photothermal conversion efficiencies of GNSs and GNRs are similar, while GNSs have a much higher molar heating rate than GNRs. Among all the samples, L-GNS has the highest molar heating rate, because of its large molar extinction coefficient. In addition, the discrete dipole approximation (DDA) was employed to simulate the optical properties of gold nanoparticles with different shapes, and the photothermal properties of GNSs and GNRs were compared experimentally and theoretically. From both the experimental and the theoretical results, M-GNS and L-GNS exhibit higher extinction efficiencies than M-GNR and L-GNR, respectively.
Design of Biopolymer-Coated Gold Nanorods as Biorelevant Photothermal Agents

Abstract:Gold nanorods (AuNRs) are emerging metallic nanoparticles utilized to generate heat for photothermal therapy (PTT) in cancer. The tunable plasmonic properties of AuNRs make them a remarkable candidate for hyperthermia. However, the cytotoxicity of AuNRs limits its biological applicability due to the existence of cetyltrimethylammonium bromide (CTAB) on the surface as a common surfactant. In this study, AuNRs are synthesized by seed-mediated growth and then the optical properties are optimized by altering AgNO3 concentration. Afterward, CTAB is replaced with biopolymers which are BSA:Dextran and BSA:Guar Gum conjugates resulting in enhanced cellular viability, enabling to use of them as biologically relevant photothermal agents. The biocompatibility of AuNRs is improved to utilize them at high concentrations for laser studies, in which similar heat generation success of CTAB- and biopolymer-coated AuNRs are shown for potential PTT applications. CTAB and biopolymer-coated AuNRs in concentrations of 0.5 and 1 mg mL-1 are irradiated under NIR light at 808 nm laser at 0.5, 0.75, and 1 W cm-2 for 300 s. The biopolymer-coated gold nanorods with different coatings preserve photothermal properties while reducing the cytotoxicity effects of CTAB and thus they are promising photothermal agents for potential PTT.

Abstract:Abstract Cancer is one of the most common incident in the world, with malignant tumors having a death rate of up to 19%. A new method of treating cancer cells effectively with minimal cytotoxicity is needed. In the field of biomedicine with unique shape-dependent optical properties, gold nanorods (GNRs) have attracted worldwide interest. These nanorods have two distinct plasmon bands. One is transverse plasmon band in the area of visible light, and the other is longitudinal band of plasmons in near infrared region. These specific characters provide promise for the design of new optically active reagents that simultaneously perform light-mediated imaging and photothermal cancer treatment. We begin our review by summarizing the latest developments in gold nanorods synthesis with a focus on seed-mediated growth method. Nanorods spontaneous self-assembly, polymer-based alignment and its applications as a novel agent for simultaneous bioimaging and photothermal cancer therapy are listed in particular.

Abstract:Gold nanorods (AuNRs) are ideal inorganic nanophotothermal agents with unique characteristics, including local surface plasmon resonance effects, easy scale preparation and functional modification, and good biocompatibility. This review summarizes several recent advances in AuNRs-based photothermal therapy (PTT) research. Functionalized AuNRs photothermal agents have optimized biocompatibility and targeting properties. The multifunctional AuNRs nanoplatform composite structure meets the requirements for synergistic effects of PTT, photoacoustic imaging, and other therapeutic methods. Photothermal therapy with AuNRs (AuNRs-PTT) is widely used to treat tumors and inflammatory diseases; its tumor-targeting, tumor metastasis inhibition, and photothermal tumor ablation abilities have remarkable curative effects. An in-depth study of AuNRs in living systems and the interactions between biological structure, nanomaterial, and near-infrared irradiation could lay the foundation for further clinical research and the broad application of AuNRs in PTT.
Size Dependence of Gold Nanorods for Efficient and Rapid Photothermal Therapy

Abstract:In recent years, gold nanomaterials have become a hot topic in photothermal tumor therapy due to their unique surface plasmon resonance characteristics. The effectiveness of photothermal therapy is highly dependent on the shape and size of gold nanoparticles. In this work, we investigate the photothermal therapeutic effects of four different sizes of gold nanorods (GNRs). The results show that the uptake of short GNRs with aspect ratios 3.3–3.5 by cells is higher than that of GNRs with aspect ratios 4–5.5. Using a laser with single pulse energy as low as 28 pJ laser for 20 s can induce the death of liver cancer cells co-cultured with short GNRs. Long GNRs required twice the energy to achieve the same therapeutic effect. The dual-temperature model is used to simulate the photothermal response of intracellular clusters irradiated by a laser. It is found that small GNRs are easier to compact because of their morphological characteristics, and the electromagnetic coupling between GNRs is better, which increases the internal field enhancement, resulting in higher local temperature. Compared with a single GNR, GNR clusters are less dependent on polarization and wavelength, which is more conducive to the flexible selection of excitation laser sources.

Seedless Synthesis of Gold Nanorods with Narrow Absorption Using Binary Surfactant Mixture

Abstract:A seedless synthetic method was developed for the preparation of high-quality gold nanorods (AuNRs) using binary surfactant mixture containing hexadecyltrimethylammonium bromide (CTAB) and sodium oleate (NaOL). The morphologies, optical properties and photothermal performances were characterized by means of transmission electron microscopy (TEM), UV-Vis-NIR spectroscopy and infrared thermal imaging system. The results indicate that uniform AuNRs can be obtained with a NaOL concentration between 8.21-11.5 mmol/L, and their longitudinal surface plasmon resonance (LSPR) wavelength can be tuned from 650 nm to 1150 nm. The AuNRs prepared with this method exhibited narrow LSPR peak,especially in the second near infrared-II region (NIR-II, lambda>1000 nm). Under the irradiation of 1064 nm laser,the temperature of AuNRs solution can increase to 67 degrees C,with a photothermal conversion efficiency of 31.5%. The AuNRs also exhibited excellent photothermal stability under laser irradiation,which can be useful for photoacoustic imaging and photothermal therapy in the second near-infrared region.
Photothermal Response of Hollow Gold Nanorods under Femtosecond Laser Irradiation

Abstract:The photothermal kinetics of hollow gold nanorod (HGNR) under femtosecond laser irradiation are studied numerically with finite-element methods and a two-temperature model. Compared with solid gold nanorod (SGNR) with the same aspect ratio (AR), the localized surface plasmon resonance (LSPR) peak of HGNR can be red-shifted to the second near-infrared window, and the absorption cross-section of HGNR can be larger than that of SGNR. In addition, under the influence of an applied numerically electromagnetic field (simulated femtosecond laser irradiation), the heat generated by HGNR makes the temperature rise of the surrounding medium faster and higher. Compared with SGNR with the same resonance wavelength, HGNR has a slightly smaller absorption cross-section but can achieve a higher temperature rise of the external medium. In addition, the laser energy, required to achieve the critical temperature for selective photothermal damage of tumor cells, is also significantly reduced. Moreover, with the same incident laser energy, the decreasing of HGNR shell thickness leads to an increase of the temperature rise of the external medium, while the change of femtosecond laser pulse width will not significantly change the temperature rise of its lattice and the external medium. In short, this study aims to provide some useful insights for the applications of HGNR in photothermal tumor therapy.

Abstract:Herein, ultrafast transient absorption spectroscopy is performed to probe the electron transfer studies between aqueous solution and gold nanorods (Au NRs). The seed-mediated growth method is used to synthesize crystalline cylindrical Au NRs having longitudinal plasmon resonance peak maximum at 825 nm. The as-synthesized Au NRs show average width and length of ∼10 ± 2 and ∼50 ± 2 nm, respectively, with an aspect ratio in the range of ∼5. The time-resolved decay profiles have been studied in a subpicosecond resolution range using pump wavelength at 410 nm excitation and probe wavelengths from visible to near-infrared region. The plasmon dynamics studies of Au NRs depend on the electron heating phenomena, coherent acoustic phonon vibration and electronic transient behavior, i.e., electron-phonon coupling, and homogenous dephasing processes. Thus, the obtained results highlighted that the ultrafast charge transfer dynamics studies in Au NRs could play an important role to elucidate their electronic, photothermal, and optical properties for molecular imaging, photothermal therapy, and optoelectronic and light-harvesting devices.

Photothermal properties of stable aggregates of gold nanorods

Abstract:Aggregation of the nanoparticles is a natural phenomena due to various biological and physical parameters but these aggregates are highly unstable. If aggregates would have been stable then it might be useful for some biomedical applications. To study the optical properties and photothermal heat generation of stable aggregates of the metal nanopartilces, we present a forced synthesis of aggregates of small gold nanorods in a mixture of Dulbecco's Modified Eagle's medium (DMEM) and Bovine Serum Albumin (BSA). We synthesised hexadecyltrimethylammonium bromide (CTAB) coated gold nanorods and then prepared the stable aggregates to study the optical characteristics of these aggregates. Absorption spectra of aggregates show the redshift compared to the monodispersive gold nanorods and confirm that the shape and size of aggregate depend on the amount of BSA in DMEM as well as on the concentration of CTAB in the stock solution of the gold nanorods suspension. The higher concentration of the BSA in DMEM and lower concentration of CTAB of monodispersive gold nanorod suspension causes greater redshift. We tuned the well defined plasmonic absorption resonance peaks of the aggregates of gold nanorods up to the second biological therapeutic window. We studied the stability of the synthesised aggregates of gold nanorods for up to one week and found that the aggregates were stable for atleast one week. A photothermal study of these aggregates was also carried out using high power broadband near-infrared light source. Aggregates were also photothermally stable which warrants their repeated use for such applications. The photothermal conversion efficiency of the these stable gold nanorod aggregates were higher than its monodispersive form of the nanorods.

Abstract:In recent years, nano-optics have had transformative impact on harnessing of energy, directing chemical reactions, and enabling novel molecular dynamics for thermodynamically intensive applications. Plasmonic nanoparticles have emerged as a tool for confining light on nanometer length scales where regions of intense electromagnetic fields can be precisely tuned for controlled surface chemistry. We demonstrate a precision pH-driven synthesis of gold nanorods with optical resonance properties that are widely tunable across the near-infrared. Through controlled electrostatic interactions, we can perform selective adsorbate molecule attachment and monitor the surface transitions through spectroscopic techniques that include ground state absorption spectrophotometry, two-dimensional X-ray absorption near-edge spectroscopy, Fourier transform infrared spectroscopy, and surface-enhanced Raman spectroscopy. We elucidate the electronic, structural, and chemical factors that contribute to plasmon-molecule dynamics at the nanoscale with broad implications to the fields of energy, photonics, and bio-inspired materials.

Selecting High-Performance Gold Nanorods for Photothermal Conversion

Abstract:In this work, we establish a new paradigm on identifying optimal arbitrarily shaped metallic nanostructures for photothermal applications. Crucial thermo-optical parameters that rule plasmonic heating are appraised, exploring a nanoparticle size-dependence approach. Our results indicate two distinct figures of merit for the optimization of metallic nanoheaters, under both non-cumulative femtosecond and continuum laser excitation. As a case study, gold nanorods are evaluated for infrared photothermal conversion in water, and the influence of the particle length and diameter are depicted. For non-cumulative femtosecond pulses, efficient photothermal conversion is observed for gold nanorods of small volumes. For continuous wave (CW) excitation at 800 nm and 1064 nm, the optimal gold nanorod dimensions (in water) are, respectively, 90 × 25nm and 150 × 30 nm. Figure of Merit (FoM) variations up to 700% were found considering structures with the same peak wavelength. The effect of collective heating is also appraised. The designing of high-performance plasmonic nanoparticles, based on quantifying FoM, allows a rational use of nanoheaters for localized photothermal applications.
Investigation of pH effect on gold nanorod synthesis

Abstract:Gold nanorods (GNRs) are routinely prepared by the seed-mediated growth method under the assistant of surfactant CTAB and it is important to understand the factors that control the formation process of GNRs. Here we investigated systematically how the pH conditions contribute to the GNR morphology and size evolution with UV-Vis absorption spectrometry and TEM. It was found that the aspect ratio and morphology of GNRs are dependent on the pH of the growth solution. Moreover, the GNR growth pathway also varies at different pH. Our results indicated that pH affected CTAB micelle's stability and reduced its absorption ability on gold surface, which changes the gold atom deposition behavior onto the surface.

Abstract:Plasmonic metal nanoparticles (NPs) have been proposed for many applications because of their fascinating optical properties related to the surface plasmon resonance (SPR) phenomenon. Due to the absorption of incident light, these objects are heated, intentionally or not, which may lead to the modification of their optical properties. In this study, the thermal variations of the absorption and scattering cross-sections of a gold NP are studied for different shapes and sizes. The results reveal that, in general, the SPR peak undergoes a redshift and its magnitude changes with increasing temperature. The maximum thermal variation of the NP optical properties is located around the SPR wavelength. Depending on the size and shape of NPs, these behaviors result in different profiles like valley-peak, valley, or peak, in the thermo-absorption and thermo-scattering spectra around the SPR wavelength. The results show that depending on the size and shape of NP, absorption and scattering cross-section thermal variation at SPR wavelength vary from − 14 × 10 −2 to 19 × 10 −2 % per Kelvin. The thermal response of the differential far-field scattering is also investigated. Once again, the maximum values of the scattered far field and its thermal variation are found around the SPR wavelength, but there is no isotropic behavior; while they are uniform in the plane perpendicular to the incident electric field, they exhibit an angular dependence in the plane including the incident wave vector and electric field.

Abstract:Transition metal dichalcogenides (TMD) coated gold nanoshells (GNSs), in addition to having low cytotoxicity and a biocompatibility value greater than graphene, exhibit strong light absorption in the near-infrared (NIR) region and high photothermal conversion efficiency. Using a quasi-static approach and bioheat equations, the optical and photothermal properties of GNSs coated with various TMDs are studied for treatment of skin cancer. Our findings show that the intensity of localized surface plasmon resonance (LSPR) peaks and their position in the extinction spectrum of nanoparticles (NPs) can be easily tuned within biological windows by varying the core radius, the gold shell thickness and the number of coating layers of the different TMDs. In order to engineer heat production at designated spatial locations of NPs, near electric field (NEF) enhancement is investigated. Moreover, the effect of laser intensity and the number of TMD layers on the temperature rise and the amount of thermal damage in skin tumor tissue and its surroundings are studied. Our results introduce GNSs with various TMD coatings as superlative nanoagents for photothermal therapy (PTT) applications.
