Abstract:Sexual fate is no longer seen as an irreversible deterministic switch set during early embryonic development but as an ongoing battle for primacy between male and female developmental trajectories. That sexual fate is not final and must be actively maintained via continuous suppression of the opposing sexual network creates the potential for flexibility into adulthood. In many fishes, sexuality is not only extremely plastic, but sex change is a usual and adaptive part of the life cycle. Sequential hermaphrodites begin life as one sex, changing sometime later to the other, and include species capable of protandrous (male-to-female), protogynous (female-to-male), or serial (bidirectional) sex change. Natural sex change involves coordinated transformations across multiple biological systems, including behavioural, anatomical, neuroendocrine, and molecular axes. We here review the biological processes underlying this amazing transformation, focussing particularly on its molecular basis, which remains poorly understood, but where new genomic technologies are significantly advancing our understanding of how sex change is initiated and progressed at the molecular level. Knowledge of how a usually committed developmental process remains plastic in sequentially hermaphroditic fishes is relevant to understanding the evolution and functioning of sexual developmental systems in vertebrates generally, as well as pathologies of sexual development in humans.

Abstract:Fishes are the only vertebrates that undergo sex change during their lifetime, but even within this group, a unique reproductive strategy is displayed by only 1.5% of the teleosts. This lability in alternating sexual fate is the result of the simultaneous suppression and activation of opposing male and female networks. Here, we provide a brief review summarizing recent advances in our understanding of the environmental cues that trigger sex change and their perception, integration, and translation into molecular cascades that convert the sex of an individual. We particularly focus on molecular events underpinning the complex behavioral and morphological transformation involved in sex change, dissecting the main molecular players and regulatory networks that shape the transformation of one sex into the opposite. We show that histological changes and molecular pathways governing gonadal reorganization are better described than the neuroendocrine basis of sex change and that, despite important advances, information is lacking for the majority of hermaphrodite species. We highlight significant gaps in our knowledge of how sex change takes place and suggest future research directions.

Abstract:Sequentially hermaphroditic fish change sex from male to female (protandry) or vice versa (protogyny), increasing their fitness by becoming highly fecund females or large dominant males, respectively. These life-history strategies present different social organizations and reproductive modes, from near-random mating in protandry, to aggregate- and harem-spawning in protogyny. Using a combination of theoretical and molecular approaches, we compared variance in reproductive success (Vk*) and effective population sizes (Ne) in several species of sex-changing fish. We observed that, regardless of the direction of sex change, individuals conform to the same overall strategy, producing more offspring and exhibiting greater Vk* in the second sex. However, protogynous species show greater Vk*, especially pronounced in haremic species, resulting in an overall reduction of Ne compared to protandrous species. Collectively and independently, our results demonstrate that the direction of sex change is a pivotal variable in predicting demographic changes and resilience in sex-changing fish, many of which sustain highly valued and vulnerable fisheries worldwide.

Abstract:Sexual dimorphism is almost ubiquitous in animals. A common pattern observed across multiple taxa involves differences in development time (sexual bimaturism) and body size (sexual size dimorphism) between conspecific males and females. Furthermore, a strict association of dimorphism at these traits has been documented in several taxa, where the sex showing shorter development time also has a smaller body size than the other sex. Growth and development are strongly dependent on environmental conditions during individual life-cycle in ectotherms, inducing considerable phenotypic plasticity. However, how phenotypic plasticity affects the association between sexual dimorphism in development time and body size remains unclear. Here, we tracked development time, body size, and body mass throughout the ontogeny of the mosquito Aedes mariae . The larval development of this species is strictly linked to Mediterranean Sea rock-pools, whose highly variable environmental conditions over minimal time frames make this organism-environment system ideal for exploring plasticity-led eco-evolutionary processes. We found differential plasticity between males and females, dissolving the link between dimorphism in development time and body size under increasing temperature and decreasing salinity conditions. These findings contrast with the current hypotheses proposed to explain the origin of the association between sexual bimaturism and sexual size dimorphism, highlighting the condition dependence of sexual dimorphism patterns and the need to consider phenotypic plasticity in future studies on their evolution.

Abstract:Studying environmental sex determination (ESD) in cichlids provides a phylogenetic and comparative approach to understand the evolution of the underlying mechanisms, their impact on the evolution of the overlying systems, and the neuroethology of life history strategies. Natural selection normally favors parents who invest equally in the development of male and female offspring, but evolution may favor deviations from this 50:50 ratio when environmental conditions produce an advantage for doing so. Many species of cichlids demonstrate ESD in response to water chemistry (temperature, pH, and oxygen concentration). The relative strengths of and the exact interactions between these factors vary between congeners, demonstrating genetic variation in sensitivity. The presence of sizable proportions of the less common sex towards the environmental extremes in most species strongly suggests the presence of some genetic sex-determining loci acting in parallel with the ESD factors. Sex determination and differentiation in these species does not seem to result in the organization of a final and irreversible sexual fate, so much as a life-long ongoing battle between competing male- and female-determining genetic and hormonal networks governed by epigenetic factors. We discuss what is and is not known about the epigenetic mechanism behind the differentiation of both gonads and sex differences in the brain. Beyond the well-studied tilapia species, the 2 best-studied dwarf cichlid systems showing ESD are the South American genus Apistogramma and the West African genus Pelvicachromis . Both species demonstrate male morphs with alternative reproductive tactics. We discuss the further neuroethology opportunities such systems provide to the study of epigenetics of alternative life history strategies and other behavioral variation.

Adaptive transgenerational plasticity (TGP) requires individuals to integrate environmental experience across multiple sources. However, few empirical studies have considered that the relative relevance of certain sources might vary across ontogeny and sexes. Here, we address this knowledge gap by studying inducible antipredator defences, one of the most convincing examples of TGP. We assessed individual and combined effects of perceived high predation risk in mothers, fathers, caring males and personal environments on the morphology of juvenile, adult male and adult female cyprinids Pimephales promelas. Parental rather than personal environmental experience determined morphological defence expression across ages and sexes, likely because parents had a longer sampling period. In juveniles and adult males, egg‐mediated environmental experience outweighed sperm‐mediated environmental experience in the induction of body shape differences, likely because eggs can transmit information beyond epigenomes. However, in adult females, where body shape responses can be interpreted as life‐history plasticity, information from egg and sperm were equally important, likely resulting from different integration mechanisms between morphological and life‐history plasticity. The importance of care‐mediated relative to gamete‐mediated variation changed between juveniles and adult males, likely because they represent short‐ and long‐term environmental experience, respectively. Instead, in adult females, both sources were again equally important, potentially owing to lag‐times of life‐history plasticity. Parental care intensity only contributed marginally to defence formation. These results highlight age‐ and sex‐specific prioritization of different environmental experiences so as to generate optimal phenotypes. Abstract:Sexual differentiation of the brain occurs in all major vertebrate lineages but is not well understood at a molecular and cellular level. Unlike most vertebrates, sex-changing fishes have the remarkable ability to change reproductive sex during adulthood in response to social stimuli, offering a unique opportunity to understand mechanisms by which the nervous system can initiate and coordinate sexual differentiation. This study explores sexual differentiation of the forebrain using single nucleus RNA-sequencing in the anemonefish , producing the first cellular atlas of a sex-changing brain. We uncover extensive sex differences in cell type-specific gene expression, relative proportions of cells, baseline neuronal excitation, and predicted inter-neuronal communication. Additionally, we identify the cholecystokinin, galanin, and estrogen systems as central molecular axes of sexual differentiation. Supported by these findings, we propose a model of neurosexual differentiation in the conserved vertebrate social decision-making network spanning multiple subtypes of neurons and glia, including neuronal subpopulations within the preoptic area that are positioned to regulate gonadal differentiation. This work deepens our understanding of sexual differentiation in the vertebrate brain and defines a rich suite of molecular and cellular pathways that differentiate during adult sex change in anemonefish.

Abstract:Environmental sex determination (ESD) occurs in divergent, phylogenetically unrelated taxa, and in some species, co-occurs with genetic sex determination (GSD) mechanisms. Although epigenetic regulation in response to environmental effects has long been proposed to be associated with ESD, a systemic analysis on epigenetic regulation of ESD is still lacking. Using half-smooth tongue sole (Cynoglossus semilaevis) as a model-a marine fish that has both ZW chromosomal GSD and temperature-dependent ESD-we investigated the role of DNA methylation in transition from GSD to ESD. Comparative analysis of the gonadal DNA methylomes of pseudomale, female, and normal male fish revealed that genes in the sex determination pathways are the major targets of substantial methylation modification during sexual reversal. Methylation modification in pseudomales is globally inherited in their ZW offspring, which can naturally develop into pseudomales without temperature incubation. Transcriptome analysis revealed that dosage compensation occurs in a restricted, methylated cytosine enriched Z chromosomal region in pseudomale testes, achieving equal expression level in normal male testes. In contrast, female-specific W chromosomal genes are suppressed in pseudomales by methylation regulation. We conclude that epigenetic regulation plays multiple crucial roles in sexual reversal of tongue sole fish. We also offer the first clues on the mechanisms behind gene dosage balancing in an organism that undergoes sexual reversal. Finally, we suggest a causal link between the bias sex chromosome assortment in the offspring of a pseudomale family and the transgenerational epigenetic inheritance of sexual reversal in tongue sole fish.

Abstract:Epigenetics plays a vital role in the interaction between living organisms and their environment by regulating biological functions and phenotypic plasticity. Considering that most aquaculture activities take place in open or natural habitats that are vulnerable to environmental changes. Promising findings from recent research conducted on various aquaculture species have provided preliminary evidence suggesting a link between epigenetic mechanisms and economically valuable characteristics. Environmental stressors, including climate changes (thermal stress, hypoxia, and water salinity), anthropogenic impacts such as (pesticides, crude oil pollution, nutritional impacts, and heavy metal) and abiotic factors (infectious diseases), can directly trigger epigenetic modifications in fish. While experiments have confirmed that many epigenetic alterations caused by environmental factors have plastic responses, some can be permanently integrated into the genome through genetic integration and promoting rapid transgenerational adaptation in fish. These environmental factors might cause irregular DNA methylation patterns in genes related to many biological events leading to organs dysfunction by inducing alterations in genes related to oxidative stress or apoptosis. Moreover, these environmental issues alter DNA/histone methylation leading to decreased reproductive competence. This review emphasizes the importance of understanding the effects of environmentally relevant issues on the epigenetic regulation of phenotypic variations in fish. The goal is to expand our knowledge of how epigenetics can either facilitate or hinder species' adaptation to these adverse conditions. Furthermore, this review outlines the areas that warrant further investigation in understanding epigenetic reactions to various environmental issues.

Abstract:The gonadal sexual fate of vertebrates is either defined by genetics or environment, or a combination of both factors. Interestingly, in sequential hermaphroditism, the animal can undergo natural sex changes from female-to-male, male-to-female, and bidirectional way throughout their lives. This change exhibits the process which shifts between oogenesis and spermatogenesis and is regarded as an ideal instance of sexual plasticity. To develop the experimental model for studying the sexual plasticity of protogynous fish, the social conditions that induce sex changes were defined in wrasse, Pseudolabrus sieboldi . When six females were kept together in a tank, the largest female became a male, whereas a similar conversion did not occur when only two females were present in a tank. A semi-gonadectomy analysis developed in the present study verified the direct relationship between gonadal sex and body coloration. In P. sieboldi , the sex change is controlled by the relative body size of an individual within a group, rather than by absolute body size. When six females were kept in smaller sized tanks, delayed sex change or unchanged individuals was observed. Overall, more than 90% of the largest females demonstrated sex change after being housed with five smaller females in different sizes of tanks ranging from 80 to 500 L. Furthermore, the experiment using a transparent barrier suggested that visual stimuli are one of the major cues to initiate sex change. Our findings on the laboratory conditions leading to the initiation of sex change in wrasse suggest the usefulness of this species as a model organism for comparative studies in molecular, cellular, and physiological mechanisms of sexual plasticity, as well as on social and reproductive behaviors.

Abstract:Most grouper species are functional protogynous hermaphrodites, but the genetic basis and the molecular mechanisms underlying the regulation of this unique reproductive strategy remain enigmatic. In this study, we reported a high‐quality chromosome‐level genome assembly of the representative orange‐spotted grouper (Epinephelus coioides). No duplication or deletion of sex differentiation related genes was found in the genome, suggesting that sex development in this grouper may be related to changes in regulatory sequences or environmental factors. Transcriptomic analyses showed that aromatase and retinoic acid are probably critical to promoting ovarian fate determination, and follicle‐stimulating hormone triggers the female‐to‐male sex change. Socially controlled sex‐change studies revealed that, in sex‐changing fish, the brain's response to social environment may be mediated by activation of phototransduction cascade and the melatonin synthesis pathway. In summary, our genomic and experimental results provide novel insights into the molecular mechanisms of sex differentiation and sex change in the protogynous groupers.

Asexuality shapes traits in a hybrid fish

Abstract:Animal morphology is influenced by several factors, including gonadal development and gametogenesis. Although their effects are well documented in male/female differentiation, much less is known about same-sex effects, such as those caused by their mode of reproduction. Here, using geometric morphometric analyses, we compare two groups of all-female triploid hybrid fish Chrosomus eos × eos-neogaeus, that differ only by their sexual and asexual reproductive strategies. We demonstrate that morphological differences arise from factors inherently associated with their mode of reproduction, with results replicated in two distinct lineages and in natural and common garden environments. Such differences provide additional insight about the costs and benefits of both reproductive strategies, which have mostly been of a demographic, population genetic, or genetic nature. In particular, these findings have important implications for the ecology of asexual organisms and contribute to the study of sex evolution by adding complexity to the paradox of sex theory.

Abstract:Phenotypic plasticity refers to the ability of an organism to alter its physiology/morphology/behavior in response to changes in environmental conditions. Although encompassing various phenomena spanning multiple levels of organization, most plastic responses seem to take place by altering gene expression and eventually altering ontogenetic trajectory in response to environmental variation. Epigenetic modifications provide a plausible link between the environment and alterations in gene expression, and the alterations in phenotype based on environmentally induced epigenetic modifications can be inherited transgenerationally. Even closely related species and populations with different genotypes may exhibit differences in the patterns and the extents of plastic responses, indicating the wide existence of plasticity genes which are independent of trait means and directly respond to environmental stimuli by triggering phenotypic changes. The ability of plasticity is not only able to affect the adaptive evolution of species significantly, but is also an outcome of evolutionary processes. Therefore, phenotypic plasticity is a potentially important molder of adaptation and evolution.
The influence of social factors on adult sex change and juvenile sexual differentiation in a diandric, protogynous epinepheline, Cephalopholis boenak (Pisces, Serranidae)

Abstract:Adult sex change and juvenile sexual differentiation in the protogynous epinepheline Cephalopholis boenak were demonstrated in captivity to be influenced by social factors. Adult sex change in C. boenak occurred in two directions, female to male and male to female. The presence or absence of a larger male plays an important role in adult female sex change; female(s) did not change sex in the presence of a larger male, but sex change occurred after the removal of the larger males in the same social groups. In male pairs, either the larger or the smaller male changed sex. Male to female sex change has not been reported previously in Cephalopholis, and only rarely in epinephelines. This is the first report of direct male sexual differentiation from juveniles (i.e. primary male development), through manipulating the number of juveniles, in a protogynous epinepheline. All isolated, single, juveniles differentiated directly as males, and male to female sex ratios did not differ significantly from 1:1 in all experimental social groups of two to four juveniles. Differentiating males grew significantly faster than differentiating females and undifferentiated juveniles during the 57-week experimental period. The role of growth rate in sex determination in C. boenak is not known but clearly plays a part in juvenile sexual differentiation and merits further investigation. Social factors influencing bi-directional adult sex change and juvenile sexual differentiation are a reflection of plasticity of sexual expression in C. boenak, in particular, and the Serranidae in general.

Abstract:Significance Environmental factors can promote phenotypic variation through alterations in the epigenome and mediate adaptation of an organism to the environment. Observations suggest the adaptation of Poecilia mexicana fish to toxic, hydrogen sulfide–rich environments in southern Mexico may, in part, be promoted through epigenetic DNA methylation alterations that became generationally stable and are inherited to subsequent generations independent of the environment. Environmental epigenetics may provide an important mechanism mediating adaptation in this species. This is an observation that the epigenome is stably inherited generationally through the germline after the removal of an environmental stressor (i.e., hydrogen sulfide) from a wild population.

Abstract:Abstract Phenotypic plasticity is critical for organismal performance and can evolve in response to natural selection. Brain morphology is often developmentally plastic, affecting animal performance in a variety of contexts. However, the degree to which the plasticity of brain morphology evolves has rarely been explored. Here, we use Trinidadian guppies (Poecilia reticulata), which are known for their repeated adaptation to high-predation (HP) and low-predation (LP) environments, to examine the evolution and plasticity of brain morphology. We exposed second-generation offspring of individuals from HP and LP sites to 2 different treatments: predation cues and conspecific social environment. Results show that LP guppies had greater plasticity in brain morphology compared to their ancestral HP population, suggesting that plasticity can evolve in response to environmentally divergent habitats. We also show sexual dimorphism in the plasticity of brain morphology, highlighting the importance of considering sex-specific variation in adaptive diversification. Overall, these results may suggest the evolution of brain morphology plasticity as an important mechanism that allows for ecological diversification and adaptation to divergent habitats.

In many cases, the occurrence of sex change is triggered by a change in the social context, such as the disappearance of the dominant individual. The orange-spotted grouper Epinephelus coioides is a typical protogynous hermaphrodite fish that changes sex from female to male and remains male throughout its life span. In this study, male-to-female sex reversal in male Epinephelus coioides was successfully induced by social isolation. The body length and mass, gonadal change, serum sex steroid hormone levels and sex-related gene expression patterns during the process of socially controlled male-to-female sex reversal in E. coioides were systematically examined. This report investigates the physiological mechanisms of the socially controlled male-to-female sex reversal process in a protogynous hermaphrodite grouper species. The results enable us to study the physiological control of sex change, not only from female to male, but also from male to female.

Abstract:Sex change in teleost fishes is commonly regulated by social factors. In species that exhibit protogynous sex change, such as the orange-spotted grouper Epinephelus coioides, when the dominant males are removed from the social group, the most dominant female initiates sex change. The aim of this study was to determine the regulatory mechanisms of socially controlled sex change in E. coioides. We investigated the seasonal variation in social behaviours and sex change throughout the reproductive cycle of E. coioides, and defined the behaviour pattern of this fish during the establishment of a dominance hierarchy. The social behaviours and sex change in this fish were affected by season, and only occurred during the prebreeding season and breeding season. Therefore, a series of sensory isolation experiments was conducted during the breeding season to determine the role of physical, visual and olfactory cues in mediating socially controlled sex change. The results demonstrated that physical interactions between individuals in the social groups were crucial for the initiation and completion of sex change, whereas visual and olfactory cues alone were insufficient in stimulating sex change in dominant females. In addition, we propose that the steroid hormones 11-ketotestosterone and cortisol are involved in regulating the initiation of socially controlled sex change.
Transgenerational effects alter the fitness consequences and genetic architecture of phenotypic plasticity and its regulatory pathways

Abstract:Parental exposure to environmental stress can influence phenotypic plasticity by offspring developing under that stressor. Transgenerational effects may also reshape natural selection on developmental plasticity by influencing its fitness consequences and expression of its genetic variation. We tested these hypotheses in the purple sea urchin , an invertebrate exposed to coastal upwelling (periods of low temperature and pH impacting biomineralization and performance). We conditioned parents and larvae to experimental upwelling and integrated RNA-seq, phenotyping of body size and biomineralization, and measured fitness-correlated traits in a quantitative genetic experiment. Larvae developing under upwelling induced widespread differential expression (DE), decreased biomineralization, and reduced body size. We detected fitness benefits for increased biomineralization and reduced size under upwelling indicative of adaptive plasticity, but only when larvae were spawned from parents exposed to upwelling. Larval DE was largely associated with adaptive phenotypic plasticity. Negative genetic correlation in DE was abundant between genes associated with adaptive plasticity. However, genetic correlations in DE associated with body size plasticity were significantly more positive in larvae from upwelling-exposed parents. These results show that transgenerational effects modify the fitness landscape and genetic architecture of phenotypic plasticity and its regulatory pathways.
