Abstract:Nitrous oxide (N2O) is an important greenhouse gas and an ozone-depleting substance. Due to the long persistence of N2O in the atmosphere, the mitigation of anthropogenic N2O emissions, which are mainly derived from microbial N2O-producing processes, including nitrification and denitrification by bacteria, archaea, and fungi, in agricultural soils, is urgently necessary. Members of mesofauna affect microbial processes by consuming microbial biomass in soil. However, how microbial consumption affects N2O emissions is largely unknown. Here, we report the significant role of fungivorous mites, the major mesofaunal group in agricultural soils, in regulating N2O production by fungi, and the results can be applied to the mitigation of N2O emissions. We found that the application of coconut husks, which is the low-value part of coconut and is commonly employed as a soil conditioner in agriculture, to soil can supply a favorable habitat for fungivorous mites due to its porous structure and thereby increase the mite abundance in agricultural fields. Because mites rapidly consume fungal N2O producers in soil, the increase in mite abundance substantially decreases the N2O emissions from soil. Our findings might provide new insight into the mechanisms of soil N2O emissions and broaden the options for the mitigation of N2O emissions.
Nitrite-dependent Anaerobic Oxidation Decreases Methane Emissions from Peatlands

Abstract:Peatlands are C-rich but N-poor ecosystems that function as an important C sink but also as significant CH4 sources and thus contribute to global warming. To confirm that N fertilization strongly reduces peatland CH4 emissions by stimulating nitrite-dependent anaerobic methane oxidation (n-damo), we investigated CH4 efflux and concentration in the soil profiles of a peatland subjected to long-term N fertilization. Contrary to previously reported increase of CH4 emissions from paddy, forest and grassland soils, N fertilization of peatland reduced CH4 efflux by 87%, especially during the hottest summer months (from 3.8 mg m(-2) h(-1) in the unfertilized control to 0.5 mg m(- 2) h(-1) in the N-fertilized site). The CH4 efflux was strongly reduced because the high organic matter content of the peat indirectly stimulated n-damo by providing CH4 (methanogens use organic matter to produce methane) and nitrite (denitrifiers use organic matter as an energy source). N fertilization reduced the CH4 concentration by 50-95% (from 7800-9500 ppm in the unfertilized control to 370-4800 ppm in the N fertilized site) in the top 30-cm soil, whereas the abundance of Methylomirabilis bacteria increased for 170% and its activity raised for 220% compared to the unfertilized control soil. Consequently, the topsoil is a hotspot of Methylomirabilis activity that use NO2- to oxidize CH4 and play an important role in reducing CH4 emissions in N-fertilized peatlands.
The Efficacy of 3,4-Dimethylpyrazole Phosphate on N2O Emissions is Linked to Niche Differentiation of Ammonia Oxidizing Archaea and Bacteria Across Four Arable Soils

Abstract:The nitrification inhibitor, 3,4-dimethylpyrazole phosphate (DMPP), can be used to reduce N2O emissions from agricultural ecosystems. However, the effectiveness of DMPP varies among soils, and this is due to both abiotic (e.g. soil properties) and biotic factors (e.g. ammonia oxidizers and denitrifier communities). Understanding the nature of these effects is necessary to improve the efficacy of DMPP, therefore encouraging wider adoption and environmental benefits. In particular, soil microbial properties associated with variation in efficacy remain largely unknown. In this study four contrasting arable soils (a grey desert soil, an alluvial paddy soil, a loess formed paddy soil, and a red soil), were characterized based on DMPP inhibition of N2O emissions and associated microbial functional guilds. DMPP significantly inhibited nitrification and N2O emissions, with an average inhibitory rate ranging from 41.7% in a red soil to 90.0% in a grey desert soil. Ammonia oxidizing bacteria (AOB) and archaea (AOA) exhibited contrasting response patterns to DMPP addition. Notably, suppression of N2O emissions by DMPP only occurred alongside fluctuations in AOB abundance. However, when AOB were inhibited, AOA abundance increased. Soil-dependent response patterns to DMPP were observed for ammonia oxidizers and denitrifiers in terms of community structure. Partial least squares path modeling (PLS-PM) found that abiotic factors, particularly pH, and biological factors such as ammonia oxidizer communities, were closely linked to N2O emissions. Our findings provide evidence that: (i) DMPP effectively inhibits nitrification through inhibiting the abundance of AOB across soil types; (ii) releasing AOA from the competition with AOB allows AOA to efficiently grow and multiply, even under high ammonium conditions; and (iii) abiotic factors play a more important role than biotic factors in soil N2O emissions.

Abstract:Nitrous oxide (N 2 O) is a potent greenhouse gas, and agricultural soils represent a major anthropogenic source. Crop residues provide nutrients for plants but also act as hotspots of N 2 O production. The hyphae of arbuscular mycorrhizal fungi (AMF) could proliferate in organic patches, utilize released N from the organic patches, and potentially mitigate N 2 O emissions. However, the effect of AMF on N 2 O emissions in degraded residue patches and the possible microbial mechanism remain uncertain. Here, a mesocosm experiment was conducted to investigate the impact of AMF ( Funneliformis mosseae ) inoculation on N 2 O emissions, availabilities of carbon and nitrogen, extracellular enzyme activities, and the abundance of key N-cycling genes in degraded residue patches. Our results showed that AMF hyphae significantly reduced N 2 O emissions from degraded residue patches. Quantitative PCR analysis of key functional genes involved in N 2 O production ( amoA , nirK , nirS ) and consumption ( nosZ ) showed that AMF significantly reduced the abundance of the bacterial amoA and nirS genes. NH 4 + , NO 3 − , total dissolved nitrogen (TDN), total nitrogen (TN), and dissolved organic carbon (DOC) contents decreased drastically in the presence of AMF. In addition, the activities of all tested extracellular enzymes were significantly decreased by AMF and positively correlated with DOC content. Multiple stepwise regression analysis demonstrated that the abundance of the nirS gene primarily influenced N 2 O emissions and was positively correlated with DOC content in degraded residue patches. Our findings indicate that AMF suppressed N 2 O producers, particularly nirS -type denitrifiers, by slowing down the release of C and N from degraded residues, thereby leading to a cascade effect on the decrease of N 2 O emissions. This study provides a promising approach to mitigate N 2 O emissions by enhancing AMF in the agroecosystems.

Abstract:Improved understanding of the microbes producing and reducing N2O spurs the invention of direct and manipulative methods to reduce N2O emissions, beyond that achieved by ‘good agronomic practice’. A core issue is the kinetics of N2O-reductase by denitrifying organisms during anoxic/hypoxic spells; the ‘hot moments’ of N2O emission. Soil pH has a pervasive effect on the Expression of the enzyme, and liming soils will reduce the N2O emission. In this review we present some novel options for mitigation of N2O emissions, beside liming. During recent years, an increasing number of strongly N2O reducing organisms have been identified, and ways to enhance their numbers and activity in soil are emerging. One suggestion is to develop inoculants for legumes that are strong N2O reducers. Another is to grow N2O reducing organisms in digestates from biogas production. These promising options need further elaboration and validation.

Meta-analysis of the effect of nitrification inhibitors on the abundance and community structure of N2O-related functional genes in agricultural soils

Abstract:Application of nitrification inhibitors (NIs) in agricultural systems is an important strategy to enhance fertilizer nitrogen use efficiency and mitigate soil nitrous oxide (N2O) emissions. Here, we conducted a global meta-analysis of 88 published studies to assess the response of N2O-related functional gene and transcript abundances, and community structure to NIs application. Application of NIs significantly reduced the abundance of ammonia-oxidizing bacteria ammonia monooxygenase (AOB amoA) genes, AOB amoA transcript and nitrite reductase (nirS and nirK) genes. The effectiveness of NIs on reducing the AOB amoA abundance was influenced by N form, soil texture, soil pH and the experimental type (field vs. laboratory). Specifically, NIs were more effective when a mixed inorganic and organic N source was applied to a medium-textured soils. The NIs effectiveness increased with increasing soil pH. The response of AOB amoA abundance to NIs application was not affected by NI type, N rate, soil moisture, soil temperature and soil organic carbon (SOC). The inhibitory effect of NIs on nirS abundance increased with increasing soil temperature. NIs decreased soil nitrifying enzyme activity (NEA) and denitrifying enzyme activity (DEA) by 34.5 % and 27.0 %, respectively, leading to an overall 63.6 % reduction of N2O emissions. Soil NEA correlated positively with the abundance and community structure of AOB amoA but not with AOA amoA. Decrease in DEA with NIs application coincided with the decreasing nirS and nirK abundances. This global-scale assessment demonstrates that the effectiveness of NIs in reducing N2O emissions was attributed to the inhibiting effects on AOB amoA, nirS and nirK genes. Our findings highlight that NIs' inhibition effects on bacterial ammonia-oxidizing community and the encode enzymes in transformation of nitrite to nitric oxide are the main mechanisms for mitigation of N fertilizer-induced N2O emissions.
Peaks of in situ N2 O emissions are influenced by N2 O-producing and reducing microbial communities across arable soils

Abstract:Agriculture is the main source of terrestrial N2 O emissions, a potent greenhouse gas and the main cause of ozone depletion. The reduction of N2 O into N2 by microorganisms carrying the nitrous oxide reductase gene (nosZ) is the only known biological process eliminating this greenhouse gas. Recent studies showed that a previously unknown clade of N2 O-reducers (nosZII) was related to the potential capacity of the soil to act as a N2 O sink. However, little is known about how this group responds to different agricultural practices. Here, we investigated how N2 O-producers and N2 O-reducers were affected by agricultural practices across a range of cropping systems in order to evaluate the consequences for N2 O emissions. The abundance of both ammonia-oxidizers and denitrifiers was quantified by real-time qPCR, and the diversity of nosZ clades was determined by 454 pyrosequencing. Denitrification and nitrification potential activities as well as in situ N2 O emissions were also assessed. Overall, greatest differences in microbial activity, diversity, and abundance were observed between sites rather than between agricultural practices at each site. To better understand the contribution of abiotic and biotic factors to the in situ N2 O emissions, we subdivided more than 59,000 field measurements into fractions from low to high rates. We found that the low N2 O emission rates were mainly explained by variation in soil properties (up to 59%), while the high rates were explained by variation in abundance and diversity of microbial communities (up to 68%). Notably, the diversity of the nosZII clade but not of the nosZI clade was important to explain the variation of in situ N2 O emissions. Altogether, these results lay the foundation for a better understanding of the response of N2 O-reducing bacteria to agricultural practices and how it may ultimately affect N2 O emissions.
Altering N2O emissions by manipulating wheat root bacterial community

Abstract:AbstractNitrous oxide (N2O) is a greenhouse gas and a potent ozone-depleting substance in the stratosphere. Agricultural soils are one of the main global sources of N2O emissions, particularly from cereal fields due to their high areal coverage. The aim of this study was to isolate N2O-reducing bacteria able to mitigate N2O emissions from the soil after inoculation. We isolated several bacteria from wheat roots that were capable of N2O reductionin vitroand studied their genetic potential and activity under different environmental conditions. Three of these isolates- all carrying the nitrous oxide reductase-encoding clade InosZ, able to reduce N2Oin vitro, and efficient colonizers of wheat roots- presented different N2O-reduction strategies when growing in the root zone, possibly due to the different conditionsin situand their metabolic preferences. Each isolate seemed to prefer to operate at different altered oxygen levels. Isolate AU243 (related toAgrobacterium/Rhizobium) could reduce both nitrate and N2O and operated better at lower oxygen levels. Isolate AU14 (related toAlcaligenes faecalis), lacking nitrate reductases, operated better under less anoxic conditions. Isolate NT128 (related toPseudomonas stutzeri) caused slightly increased N2O emissions under both anoxic and ambient conditions. These results therefore emphasize the importance of a deep understanding of soil–plant–microbe interactions when environmental application is being considered.

Abstract:Agricultural soils are an important source of nitrous oxide (N2O), a potent greenhouse gas involved in the destruction of the protective ozone layer that contributes to global warming. During N2O production, soil microorganisms play important driving and regulating roles. A few recent studies have revealed the potential effects of arbuscular mycorrhizal fungi (AMF), a widely distributed soil fungi, on controlling N2O emissions. However, how AMF regulate N2O production from soils remains poorly understood. To address the knowledge gap, we manipulated two independent soil environments, which were either allowed (AM) or prevented (NM) access by AMF hyphae in a microcosm experiment (n = 5). Soil physicochemical properties, N2O flux, the diversity of bacterial communities, and the abundance of key genes responsible for N2O production were assessed in both treatments over three months. Results showed that the presence of AMF significantly decreased N2O emissions from agricultural soils in the 1st month, and the abundance of key genes responsible for denitrification (nirK and nosZ) significantly decreased in AM treatments, indicating that the regulation of N2O emissions is transmitted by AMF-induced changes in the denitrification process. A structural equation model further revealed that AMF indirectly influenced N2O emissions by altering the abundance of N metabolism-related genes, rather than by altering soil chemical properties or the diversity of bacterial communities. Thus, we proposed a possible mechanism by which AMF can regulate denitrification activities and therefore N2O emissions from agricultural soils.

Abstract:Arbuscular mycorrhizal fungi (AMF) increase the ability of plants to obtain nitrogen (N) from the soil, and thus can affect emissions of nitrous oxide (N 2 O), a long-lived potent greenhouse gas. However, the mechanisms underlying the effects of AMF on N 2 O emissions are still poorly understood, particularly in agroecosystems with different forms of N fertilizer inputs. Utilizing a mesocosm experiment in field, we examined the effects of AMF on N 2 O emissions via their influence on maize root traits and denitrifying microorganisms under ammonia and nitrate fertilizer input using 15 N isotope tracer. Here we show that the presence of AMF alone or both maize roots and AMF increased maize biomass and their 15 N uptake, root length, root surface area, and root volume, but led to a reduction in N 2 O emissions under both N input forms. Random forest model showed that root length and surface area were the most important predictors of N 2 O emissions. Additionally, the presence of AMF reduced the ( nir K + nir S)/ nos Z ratio by increasing the relative abundance of nir S- Bradyrhizobium and Rubrivivax with ammonia input, but reducing nos Z- Azospirillum , Cupriavidus and Rhodopseudomonas under both fertilizer input. Further, N 2 O emissions were significantly and positively correlated with the nos Z-type Azospirillum , Cupriavidus and Rhodopseudomonas , but negatively correlated with the nir S-type Bradyrhizobium and Rubrivivax . These results indicate that AMF reduce N 2 O emissions by increasing root length to explore N nutrients and altering the community composition of denitrifiers, suggesting that effective management of N fertilizer forms interacting with the rhizosphere microbiome may help mitigate N 2 O emissions under future N input scenarios.

Abstract:Arbuscular mycorrhizal fungi (AMF) are key soil organisms and their extensive hyphae create a unique hyphosphere associated with microbes actively involved in N cycling. However, the underlying mechanisms how AMF and hyphae-associated microbes may cooperate to influence N 2 O emissions from "hot spot" residue patches remain unclear. Here we explored the key microbes in the hyphosphere involved in N 2 O production and consumption using amplicon and shotgun metagenomic sequencing. Chemotaxis, growth and N 2 O emissions of isolated N 2 O-reducing bacteria in response to hyphal exudates were tested using in vitro cultures and inoculation experiments.

Abstract:Arbuscular mycorrhizal fungi (AMF) play an important role in soil nitrogen cycling. However, the effect of AMF on soil N2O emission and its underlying mechanisms remain elusive. Here, we conducted a Bayesian meta-analysis to determine whether AMF has an N2O mitigation potential and what are the potential mechanisms. We found that AMF significantly decreased soil N2O emission (with an effect size of −0.90) and increased microbial biomass nitrogen and plant biomass compared to the non-AMF treatment. The effect size of soil water content and plant biomass nitrogen showed significantly positive and negative correlations with the effect size of soil N2O emission. We propose a conceptual framework with three potential mechanisms that AMF could affect soil N2O emission for further work.

Abstract:Nitrous oxide (N2O) is an important greenhouse gas (GHG) with substantial global warming potential and also contributes to ozone depletion through photochemical nitric oxide (NO) production in the stratosphere. The negative effects of N2O on climate and stratospheric ozone make N2O mitigation an international challenge. More than 60% of global N2O emissions are emitted from agricultural soils mainly due to the application of synthetic nitrogen-containing fertilizers. Thus, mitigation strategies must be developed which increase (or at least do not negatively impact) on agricultural efficiency whilst decrease the levels of N2O released. This aim is particularly important in the context of the ever expanding population and subsequent increased burden on the food chain. More than two-thirds of N2O emissions from soils can be attributed to bacterial and fungal denitrification and nitrification processes. In ammonia-oxidizing bacteria, N2O is formed through the oxidation of hydroxylamine to nitrite. In denitrifiers, nitrate is reduced to N2 via nitrite, NO and N2O production. In addition to denitrification, respiratory nitrate ammonification (also termed dissimilatory nitrate reduction to ammonium) is another important nitrate-reducing mechanism in soil, responsible for the loss of nitrate and production of N2O from reduction of NO that is formed as a by-product of the reduction process. This review will synthesize our current understanding of the environmental, regulatory and biochemical control of N2O emissions by nitrate-reducing bacteria and point to new solutions for agricultural GHG mitigation.

Abstract:The only known sink for nitrous oxide (N2O) is biochemical reduction to dinitrogen (N2) by N2O reductase (N2OR). We hypothesized that the application of N2O-reducing denitrifier-inoculated organic fertilizer could enhance soil N2O consumption while the disruption of nosZ genes could result in inactivation of N2O consumption. To test such hypotheses, a denitrifier-inoculated granular organic fertilizer was applied to both soil microcosms and fields. Of 41 denitrifier strains, 38 generated 30N2 in the end products of denitrification (30N2 and 46N2O) after the addition of Na15NO3 in culture condition, indicating their high N2O reductase activities. Of these 41 strains, 18 were screened in soil microcosms after their inoculation into the organic fertilizer, most of which were affiliated with Azospirillum and Herbaspirillum. These 18 strains were nutritionally starved to improve their survival in soil, and 14 starved and/or non-starved strains significantly decreased N2O emissions in soil microcosms. However, the N2O emission had not been decreased in soil microcosms after inoculating with a nosZ gene-disruptive strain, suggesting that N2O reductase activity might be essential for N2O consumption. Although the decrease of N2O was not significant at field scales, the application of organic fertilizer inoculated with Azospirillum sp. TSH100 and Herbaspirillum sp. UKPF54 had decreased the N2O emissions by 36.7% in Fluvisol and 23.4% in Andosol in 2014, but by 21.6% in Andosol in 2015 (H. sp. UKPF54 only). These results suggest that the application of N2O-reducing denitrifier-inoculated organic fertilizer may enhance N2O consumption or decrease N2O emissions in agricultural soils.

Abstract:Agricultural management practices can induce changes in soil aggregation structure that alter the microbial nitrous oxide (N2O) production and reduction processes occurring at the microscale, leading to large-scale consequences for N2O emissions. However, the mechanistic understanding of how organic fertilization affects these context-dependent small-scale N2O emissions and associated key nitrogen (N) cycling microbial communities is lacking. Here, denitrification gas (N2O, N2) and potential denitrification capacity N2O/(N2O + N2) were assessed by automated gas chromatography in different soil aggregates (>2 mm, 2-0.25 and <0.25 mm), while associated microbial communities were assessed by sequencing and qPCR of N2O-producing (nirK and nirS) and reducing (nosZ clade I and II) genes. The results indicated that organic fertilization reduced N2O emissions by enhancing the conversion of N2O to N2 in all aggregate sizes. Moreover, potential N2O production and reduction hotspots occurred in smaller soil aggregates, with the degree depending on organic fertilizer type and application rate. Further, significantly higher abundance and diversity of nosZ clades relative to nirK and nirS revealed complete denitrification promoted through selection of denitrifying communities at microscales favouring N2O reduction. Communities associated with high and low emission treatments form modules with specific sequence types which may be diagnostic of emission levels. Taken together, these findings suggest that organic fertilizers reduced N2O emissions through influencing soil factors and patterns of niche partitioning between N2O-producing and reducing communities within soil aggregates, and selection for communities that overall are more likely to consume than emit N2O. These findings are helpful in strengthening the ability to predict N2O emissions from agricultural soils under organic fertilization as well as contributing to the development of net-zero carbon strategies for sustainable agriculture.

Abstract:Since the development of effective N2O mitigation options is a key challenge for future agricultural practice, we studied the interactive effect of tillage systems on fertilizer-derived N2O emissions and the abundance of microbial communities involved in N2O production and reduction. Soil samples from 0–10 cm and 10–20 cm depth of reduced tillage and ploughed plots were incubated with dairy slurry (SL) and manure compost (MC) in comparison with calcium ammonium nitrate (CAN) and an unfertilized control (ZERO) for 42 days. N2O and CO2 fluxes, ammonium, nitrate, dissolved organic C, and functional gene abundances (16S rRNA gene, nirK, nirS, nosZ, bacterial and archaeal amoA) were regularly monitored. Averaged across all soil samples, N2O emissions decreased in the order CAN and SL (CAN = 748.8 ± 206.3, SL = 489.4 ± 107.2 μg kg−1) followed by MC (284.2 ± 67.3 μg kg−1) and ZERO (29.1 ± 5.9 μg kg−1). Highest cumulative N2O emissions were found in 10–20 cm of the reduced tilled soil in CAN and SL. N2O fluxes were assigned to ammonium as source in CAN and SL and correlated positively to bacterial amoA abundances. Additionally, nosZ abundances correlated negatively to N2O fluxes in the organic fertilizer treatments. Soils showed a gradient in soil organic C, 16S rRNA, nirK, and nosZ with greater amounts in the 0–10 than 10–20 cm layer. Abundances of bacterial and archaeal amoA were higher in reduced tilled soil compared to ploughed soils. The study highlights that tillage system induced biophysicochemical stratification impacts net N2O emissions within the soil profile according to N and C species added during fertilization.

Abstract:Tropical rainforest soils harbor a considerable diversity of soil fauna that contributes to emissions of N2O. Despite their ecological dominance, there is limited information available about the contribution of epigeal ant mounds to N2O emissions in these tropical soils. This study aimed to determine whether ant mounds contribute to local soil N emissions in the tropical humid rainforest. N2O emission was determined in vitro from individual live ants, ant-processed mound soils, and surrounding reference soils for two trophically distinct and abundant ant species: the leaf-cutting Atta mexicana and omnivorous Solenopsis geminata. The abundance of total bacteria, nitrifiers (AOA and AOB), and denitrifiers (nirK, nirS, and nosZ) was estimated in these soils using quantitative PCR, and their respective mineral N contents determined. There was negligible N2O emission detected from live ant individuals. However, the mound soils of both species emitted significantly greater (3-fold) amount of N2O than their respective surrounding reference soils. This emission increased significantly up to 6-fold in the presence of acetylene, indicating that, in addition to N2O, dinitrogen (N2) is also produced from these mound soils at an equivalent rate (N2O/N2 = 0.57). Functional gene abundance (nitrifiers and denitrifiers) and mineral N pools (ammonium and nitrate) were significantly greater in mound soils than in their respective reference soils. Furthermore, in the light of the measured parameters and their correlation trends, nitrification and denitrification appeared to represent the major N2O-producing microbial processes in ant mound soils. The ant mounds were estimated to contribute from 0.1 to 3.7% of the total N2O emissions of tropical rainforest soils.

Abstract:The concentration of greenhouse gases (GHGs) in the atmosphere has been increasing since the beginning of the industrial revolution. Nitrous oxide (N2O) is one of the mightiest GHGs, and agriculture is one of the main sources of N2O emissions. In this paper, we reviewed the mechanisms triggering N2O emissions and the role of agricultural practices in their mitigation. The amount of N2O produced from the soil through the combined processes of nitrification and denitrification is profoundly influenced by temperature, moisture, carbon, nitrogen and oxygen contents. These factors can be manipulated to a significant extent through field management practices, influencing N2O emission. The relationships between N2O occurrence and factors regulating it are an important premise for devising mitigation strategies. Here, we evaluated various options in the literature and found that N2O emissions can be effectively reduced by intervening on time and through the method of N supply (30–40%, with peaks up to 80%), tillage and irrigation practices (both in non-univocal way), use of amendments, such as biochar and lime (up to 80%), use of slow-release fertilizers and/or nitrification inhibitors (up to 50%), plant treatment with arbuscular mycorrhizal fungi (up to 75%), appropriate crop rotations and schemes (up to 50%), and integrated nutrient management (in a non-univocal way). In conclusion, acting on N supply (fertilizer type, dose, time, method, etc.) is the most straightforward way to achieve significant N2O reductions without compromising crop yields. However, tuning the rest of crop management (tillage, irrigation, rotation, etc.) to principles of good agricultural practices is also advisable, as it can fetch significant N2O abatement vs. the risk of unexpected rise, which can be incurred by unwary management.

Abstract:Increases in soil available nitrogen (N) influence N-cycle gene abundances and emission of nitrous oxide (N 2 O), which is primarily due to N-induced soil acidification in forest. Moreover, the extent of microbial-N saturation could control microbial activity and N 2 O emission. The contributions of N-induced alterations of microbial-N saturation and N-cycle gene abundances to N 2 O emission have rarely been quantified. Here, the mechanism underlying N 2 O emission under N additions (three chemical forms of N, i.e., NO 3 − -N, NH 4 + -N and NH 4 NO 3 –N, and each at two rates, 50 and 150 kg N ha −1 year −1 , respectively) spanning 2011–2021 was investigated in a temperate forest in Beijing. Results showed N 2 O emissions increased at both low and high N rates of all the three forms compared with control during the whole experiment. However, N 2 O emissions were lower in high rate of NH 4 NO 3 –N and NH 4 + -N treatments than the corresponding low N rates in the recent three years. Effects of N on microbial-N saturation and abundances of N-cycle genes were dependent on the N rate and form as well as experimental time. Specifically, negative effects of N on N-cycle gene abundances and positive effects of N on microbial-N saturation were demonstrated in high N rate treatments, particularly with NH 4 + addition during 2019–2021. Such effects were associated with soil acidification. A hump-backed trend between microbial-N saturation and N 2 O emissions was observed, suggesting N 2 O emissions decreased with increase of the microbial-N saturation. Furthermore, N-induced decreases in N-cycle gene abundances restrained N 2 O emissions. In particular, the nitrification process, dominated by ammonia-oxidize archaea, is critical to determination of N 2 O emissions in response to the N addition in the temperate forest. We confirmed N addition promoted soil microbial-N saturation and reduced N-cycle gene abundances, which restrained the continuous increase in N 2 O emissions. It is important for understanding the forest-N-microbe nexus under climate change.

Abstract:Nitrous oxide (N2O) is one of the most important greenhouse gases and has the potential to aggravate global warming. Arbuscular mycorrhizal (AM) fungi form mutualistic associations with most terrestrial plants, which can help plant nitrogen (N) uptake and have also been shown to reduce soil N2O emissions. However, the development and species community composition of AM fungi are influenced by warming and N deposition. To date, the mechanism by which warming, N deposition, and AM fungi interactively affect soil N2O emissions in the field is still poorly understood. We implemented elevated temperature and N addition treatments in a 5-year experiment in a semiarid temperate meadow to examine the impact of AM fungi on soil N2O fluxes under the treatment conditions. Both elevated temperature and N deposition and the combination of the two significantly increased soil N2O emissions. AM fungi reduced soil N2O emissions under elevated temperature and N deposition. The decrease in soil N2O emissions by AM fungi was lower under elevated temperature than N deposition. Under elevated temperature, the AM fungal decrease in soil N2O emissions was mainly determined by the abundance of the functional genes ammonia-oxidizing bacteria (AOB) and nosZ, while the AM fungal reduction in soil N2O emissions was mainly affected by soil microbial biomass carbon under N deposition. These results indicate that although AM fungi could reduce soil N2O emissions the pathway by which AM fungi regulate soil N2O emissions might vary under different global change contexts with elevated temperature or nitrogen deposition. Our results highlight the negative effect of AM fungi on soil N2O emissions under elevated temperature and N deposition and show that the influence of AM fungi on N2O emissions might be simultaneously determined by warming, N deposition and ecosystem types. The results of the study help to evaluate the potential role of AM fungi in reducing N2O emissions from grassland ecosystems in the context of future global change and provide a theoretical basis for sustainable grassland management.
