Abstract:206 Tribromoethanol (TBE) is a widely used anesthetic agent in embryo and reproduction research involving mice (1, 2). This drug used to be available under the trade name Avertin. It has been an extensively used anesthetic in research involving mice in many laboratories because of its rapid onset of effect, wide range of tolerance, and low mortality (3, 4). We recently used TBE to sedate and immobilize pregnant female mice for Doppler echocardiographic evaluation of mouse embryo heart function in transgenic embryos. It was found that TBE can induce transient arrhythmia in the clinically normal control mouse embryo, shortly after anesthesia is induced in the adult female. Previously, pentobarbital had been routinely used in our laboratory to sedate pregnant female mice for echocardiographic evaluation of mouse embryo heart function (5). To better define how TBE may affect cardiac function in the mouse embryo, we evaluated 51 normal embryos in seven litters longitudinally. The TBE effects were compared with those in mice of six litters, the pregnant females of which were sedated with pentobarbital (PTB) on the same days of gestation. The study reported here is of importance in the analysis of cardiac function, as well as studies of the conduction system in normal and transgenic mouse embryos. Tribromoethanol was formulated by dissolving 1 g of 2,2,2-tribromoethanol (Aldrich Chemical Co., Milwaukee, Wis.) in 1 ml of 2-methyl-2-butanol (Aldrich Chemical Co.). The TBE was stored at -208C in the dark. Before use, TBE was thawed at room temperature and diluted with sterile distilled water to make a working concentration of 25 mg/ml. The pH of TBE was monitored to rule out pH change as a possible risk factor contributing to any observed effects. The pH of TBE was stable at pH 7.0 during the period in which the study was carried out. Doppler echocardiography was carried out immediately after the pregnant female was sedated and immobilized, which usually occurred within 1 min after TBE was given. Anesthesia duration was 30 to 60 min. Pentobarbital sodium (Abbott Laboratories, North Chicago, Ill.) was diluted with 0.9% sodium chloride to a concentration of 5 mg/ml.
