Abstract:Large-scale uniform silicon nanowires (SiNWs) array was fabricated by chemical etching on n-Si(111) wafer. Silver nanoparticles (AgNPs) were loaded on their surfaces. The AgNPs on SiNWs (AgNPs@SiNWs) array exhibit strong surface-enhanced Raman effect. On the substrate surfaces, characteristic Raman signals are generated with trace amount of mouse immunoglobulin G (mIgG), goat-anti-mouse immunoglobulin G (gamIgG), and immune complexes formed from 4ng each of mIgG and gamIgG. The shifted positions and changed intensities in Raman bands indicate the occurrence of immunoreactions. This AgNPs@SiNWs array is a unique substrate for surface-enhanced Raman spectroscopy to show the immune reagents and immunoreactions at higher sensitivity.

Aptameric System for the Highly Selective and Ultrasensitive Detection of Protein in Human Serum Based on Non-Stripping Gold Nanoparticles

Abstract:A novel approach is proposed in this study for the development of an aptameric assay system for protein based on non-stripping gold nanoparticles (NPs)-triggered chemiluminescence (CL) upon target binding. The strategy chiefly depends on the formation of a sandwich-type immunocomplex among the capture antibody immobilized on the polystyrene microwells, target protein and aptamer-functionalized gold NPs. Introduction of target protein into the assay system leads to the attachment of gold NPs onto the surface of the microwells and thus the assembled gold NPs could trigger the reaction between luminol and AgNO(3) with a CL emission. Further signal amplification was achieved by a simple gold metal catalytic deposition onto the gold NPs. Such an amplified CL transduction allowed for the detection of model target IgE down to the 50 fM, which is better than most existing aptameric methods for IgE detection. This new protocol also provided a good capability in discriminating IgE from nontarget proteins such as IgG, IgA, IgM and interferon. The practical application of the proposed gold NPs-based immunoassay was successfully carried out for the determination of IgE in 35 human serum samples. Overall, the proposed assay system exhibits excellent analytical characteristics (e.g., a detection limit on the attomolar scale and a linear dynamic range of 4 orders of magnitude), and it is also straightforward to adapt this strategy to detect a spectrum of other proteins by using different aptamers. This new CL strategy might create a novel technology for developing simple biosensors in the sensitive and selective detection of target protein in a variety of clinical, environmental and biodefense applications.
Dielectrophoresis-Based Protein Enrichment for a Highly Sensitive Immunoassay Using Ag/SiO2 Nanorod Arrays

Abstract:A nanoscale insulator-based dielectrophoresis (iDEP) technique is developed for rapid enrichment of proteins and highly sensitive immunoassays. Dense arrays of nanorods (NDs) by oblique angle deposition create a super high electric field gradient of 2.6 × 1024 V2 m-3 and the concomitant strong dielectrophoresis force successfully traps small proteins at a bias as low as 5 V. 1800-fold enrichment of bovine serum albumin protein at a remarkable rate of up to 180-fold s-1 is achieved using oxide coated Ag nanorod arrays with pre-patterned sawtooth electrodes. Based on this system, an ultrasensitive immunoassay of mouse immunoglobulin G is demonstrated with a reduction in the limit of detection from 5.8 ng mL-1 (37.6 pM) down to 275.3 fg mL-1 (1.8 f M), compared with identical assays performed on glass plates. This methodology is also applied to detect a cancer biomarker prostate-specific antigen spiked in human serum with a detection limit of 2.6 ng mL-1 . This high sensitivity results from rapid biomarker enrichment and metal enhanced fluorescence through the integration of nanostructures. The concentrated proteins also accelerate binding kinetics and enable signal saturation within 1 min. Given the easy fabrication process, this nanoscale iDEP system provides a highly sensitive detection platform for point-of-care diagnostics.

Abstract:Au/Ag core-shell nanoparticles labeled by monoclonal antibody and surface-enhanced Raman scattering (SERS) probes were used for immunoassay analysis. Mouse polyclonal antibodies against hepatitis B surface antigen (mouse Anti-HBsAg PAb) were covalently immobilized on the silicon chips modified by a self-assembled monolayer of (3-amino-propyl) trimethoxysilane via glutaraldehyde (GA) activation. The Au/Ag core-shell nanoparticles were labeled by the mouse monoclonal antibody against hepatitis B surface antigen (mouse Anti-HBsAg MAb) and the probe molecule, p-mercaptobenzoic acid (MBA). Immunoreactions between the labeled Au/Ag core-shell nanoparticles and the corresponding antigens captured by the Anti-HBsAg PAb on the silicon chips were investigated by SERS.

Abstract:The identification and detection of cancer biomarkers in early stages is an important issue for the therapy of cancer. However, most methods are time-consuming and have limited sensing sensitivity and specificity. In this work, we prepared a novel plasmonic multilayered core-shell-satellite nanostructure (Au@Ag@SiO2-AuNP) consisting of a gold nanosphere with a silver coating core (Au@Ag), an ultrathin continuous silica (SiO2) shell, and a high coverage of gold nanosphere (AuNP) satellites. The Au@Ag core is a prominent surface enhanced Raman scattering (SERS) platform, and the thin SiO2 layer exhibits a long-range plasmon coupling between the Au@Ag core to the AuNP satellites, further leading to enhanced Raman scattering. Meanwhile, the outer AuNP satellites have a high biocompatibility and long-term stability. Combining the above advantages, the well-designed metallic nanoassemblies would be a promising candidate for SERS-based applications in biochemistry. For specific detection of alpha-fetoprotein (AFP), we utilized the SERS-active core-shell-satellite nanostructures modified with AFP antibody as immune probes and nitrocellulose membrane (NC) stabilized captured anti-AFP antibodies as solid substrate. To improve the detection performance, we further systematically optimized the parameters, including the silver coating thickness of the Au@Ag core and the density and size of the satellite AuNPs. Under the optimized conditions, AFP could be detected by the SERS-based sandwich immunoassay with an ultralow detection limit of 0.3 fg/mL, and the method exhibited a wide linear response from 1 fg/mL to 1 ng/mL. The limit of detection (LOD) was considerably lower than conventional methods in the literature. This work relies on the unique Au@Ag@SiO2-AuNP nanostructures as the immune probe develops a new outlook for the application of multilayered nanoassemblies and demonstrates the great potential in early tumor marker detection.

Abstract:A one-step homogenous sensitive immunoassay using surface-enhanced Raman scattering (SERS) has been developed. This strategy is based on the aggregation of Raman reporter-labeled immunogold nanoparticles induced by the immunoreaction with corresponding antigens. The aggregation of gold nanoparticles results in a SERS signal increase of the Raman reporter. Therefore, human IgG could be directly determined by measuring the Raman signal of the reporter. The process of aggregation was investigated by transmission electron microscopy (TEM) and UV–Vis absorption spectroscopy. The effects of the temperature, time, and size of gold nanoparticles on the sensitivity of the assay were examined. Using human IgG as a model protein, a wide linear dynamic range (0.1–15 μg mL−1) was reached with low detection limit (0.1 μg mL−1) under optimized assay conditions. The successful test suggests that the application of the proposed method holds promising potential for simple, fast detection of proteins in the fields of molecular biology and clinical diagnostics.

Abstract:The goal of this work is to explore the amplification effect of aptamer–gold nanoparticles (Au NPs) conjugates for ultrasensitive detection of large biomolecules by surface plasmon resonance (SPR). A novel sandwich immunoassay is designed to demonstrate the amplification effect of aptamer–Au NPs conjugates by using human immunoglobulin E (IgE) as model analyte. Human IgE, captured by immobilized goat anti-human IgE on SPR gold film, is sensitively detected by SPR spectroscopy with a lowest detection limit of 1ng/ml after anti-human IgE aptamer–Au NPs conjugates is used as amplification reagent. Meanwhile, the non-specific adsorption of aptamer–Au NPs conjugates on goat anti-human IgE is confirmed by SPR spectroscopy and then it is minimized by treating aptamer–Au NPs conjugates with 6-mercaptohexan-1-ol (MCH). These results confirm that aptamer–Au NPs conjugates is a powerful sandwich element and an excellent amplification reagent for SPR-based sandwich immunoassay.

Abstract:Few-atom gold nanoclusters (AuNCs) have been fabricated and applied in various fields owing to their remarkable optical and photophysical features. However, rational design for the antibody-mediated synthesis of fluorescent AuNCs for direct antigen-antibody reactions remains unexplored. In this work, immunoglobulin G (IgG)-functionalized AuNCs (IgG-AuNCs) were successfully prepared via a facile and fast biomineralization process. The generated IgG-AuNCs can emit intense red fluorescence with a high photoluminescence quantum yield. In addition to the strong emission, the bioactivity of IgG on the IgG-AuNCs can be retained. Surface plasmon resonance measurements suggested that IgG-AuNCs can bind to goat anti-human IgG with an affinity constant of 6.21 × 10-8 M. A simple detection method was then developed using a dot-blot immunoassay with IgG-AuNCs as fluorescent tags. Experimental results confirmed that the IgG-AuNC-based fluorescent reporters had many advantages such as low nonspecific adsorption and good photostability, offering immense potential for the development of efficient biosensors. This work can be extended to other specific antibodies to produce multifunctional AuNCs and utilized to detect and monitor targeted analytes and biological events of interest.
Ag@Au Core–Shell Porous Nanocages with Outstanding SERS Activity for Highly Sensitive SERS Immunoassay

Abstract:A highly sensitive immunoassay of biomarkers has been achieved using 4-mercaptobenzoic acid-labeled Ag@Au core–shell porous nanocage tags and α-fetoprotein immuno-sensing chips. The Ag@Au porous nanocages were uniquely synthesized by using an Ag core as a self-sacrificial template and reducing agent, where the slow reaction process led to the formation of a porous Au layer. The size of the remaining Ag core and surface roughness of the Au shell were controlled by adjusting the chloroauric acid concentration. The porous cage exhibited excellent surface-enhanced Raman spectroscopy (SERS) activity, presumably due to a synergetic interaction between newly generated hot spots in the rough Au shell and the retained SERS activity of the Ag core. Using α-fetoprotein as a model analyte for immunoassay, the SERS signal had a wide linear range of 0.20 ng mL−1 to 500.0 ng mL−1 with a detection limit of 0.12 ng mL−1. Without the need of further signal amplification, the as-prepared Ag@Au bimetallic nanocages can be directly used for highly sensitive SERS assays of other biomarkers in biomedical research, diagnostics, etc.

A Au nanoparticle and polydopamine co-modified biosensor: A strategy for in situ and label-free surface plasmon resonance immunoassays

Abstract:This article reports a surface plasmon resonance (SPR) strategy capable of label-free yet amplified in situ immunoassays for sensitive and specific detection of human IgG (hIgG), a serum marker that is important for the diagnosis of certain diseases. Primarily, a wavelength-modulated Kretschman configuration SPR analyzer was constructed, and Au film SPR biosensor chips were fabricated. Specifically, based on Au nanoparticles (AuNPs) adsorbed on the surface of the Au film, the AuNP/Au film was coated with polydopamine (PDA) to fix streptavidin (SA), and then the biotinylated antibodies were connected to the surface of the biosensor chip. The SPR analyzer was utilized for in situ real-time monitoring of hIgG. Due to the immunological recognition between the receptor and target, the surface plasmon waves produced by the attenuated total reflection were affected by the changes in the surface of the biosensor chip. The resonance wavelength (λR) of the output spectra gradually redshifted, and the redshift degrees were directly related to the target concentration. The biosensor can realize the in situ detection of hIgG, displaying satisfactory sensitivity, excellent specificity and stability. Briefly, by monitoring the shift in λR after specific binding, a new SPR immunoassay can be customized for label-free, in situ and amplified hIgG detection. The operating principle of this research could be extended as a common protocol for many other targets of interest.

Abstract:Biomolecule-functionalized Au nanoclusters (AuNCs) have drawn great interest in the electrochemiluminescence (ECL) field due to their unique optical/electrical properties, biocompatibility, and versatile bioapplication potentials. Herein, we proposed a two-in-one ECL probe of immunoglobulin G-encapsulated AuNCs (IgG-AuNCs) for the development of a high-performance ECL immunoassay (ECLIA) platform. The IgG-AuNCs were not only used as an ECL probe due to their excellent anodic ECL performance with triethylamine (TEA) as a coreactant but also used as the biorecognition element because of their well-retained bioactivity of the IgG. As a proof of concept, a new type of competitive immunosensing platform has been applied to detect IgG representing several merits of facile preparation, rapid detection, sample saving, and good analytical performance. The sensing platform exhibited a linear range of 0.5–50,000 ng/mL with a limit of detection of 0.06 ng/mL for IgG detection with high selectivity. In addition, this convenient ECLIA platform also performed well in real serum sample detection. Abstract:A surface-enhanced Raman scattering (SERS)-based sandwich type immunoassay was developed for the detection of alpha-fetoprotein (AFP) with high sensitivity, reproducibility and selectivity. In the immunoassay, an antibody immobilized nitrocellulose (NC) membrane was employed to capture AFP and antibody conjugated silica coated gold/silver core-shell nanostars (AuNS@Ag@SiO2) were used as SERS probes. AuNS@Ag was synthesized by the direct deposition of Ag on the AuNS. The Ag shell grew outward from the center towards the tips and adopted a polyhedral shape. The AuNS@Ag nanoparticles were shown to have higher SERS activity than gold nanostars (AuNSs), and their LSPR wavelengths could be finely tuned across a wide spectrum ranging from visible to near-infrared by simply adjusting the Ag shell thickness. The silica coating improved colloidal stability of the nanostructure and provided functionality for antibody conjugation. By using 4-mercaptobenzoic acid (4-MBA) as a Raman reporter molecule, the proposed SERS sensor is able to detect AFP with a low limit of detection (LOD) of 0.72 pg mL(-1) and a wide and clinically relevant linear detection range from 3 pg mL(-1) to 3 mu g mL(-1). Hence, this SERS-based immunoassay provides a strategy for the detection of trace AFP and has potential to assist in the monitoring of prognosis for hepatocellular carcinoma (HCC) patients.

Abstract:This work designed a simple, sensitive, and low-cost immunosensor for the detection of protein marker by using a carbon sphere/gold nanoparticle (CNS/AuNP) composite as an electrochemical label. The nanoscale carbon spheres, prepared with a hydrothermal method by using glucose as raw material, were used to load AuNPs for labeling antibody by electrostatic interaction, which provided a feasible pathway for electron transfer due to the remarkable conductivity. The disposable immunosensor was constructed by coating a polyethylene glycol (PEG) film on a screen-printed carbon-working electrode and then immobilizing capture antibody on the film. With a sandwich-type immunoassay format, the analyte and then the CNS/AuNP-labeled antibody were successively bound to the immunosensor. The bound AuNPs were finally electro-oxidized in 0.1?M HCl to produce AuCl4- for differential pulse voltammetric (DPV) detection. The high-loading capability of AuNPs on CNS for the sandwich-type immunorecognition led to obvious signal amplification. By using human immunoglobulin G (IgG) as model target, the DPV signal of AuNPs after electro-oxidized at optimal potential of +1.40 V for 40 s showed a wide linear dependence on the logarithm of target concentration ranging from 10 pg?mL-1 to 10 ng?mL-1. The detection limit was around 9 pg?mL-1. The immunosensor showed excellent analytical performance with cost effectivity, good fabrication reproducibility, and acceptable precision and accuracy, providing significant potential application in clinical analysis.
Naked-eye sensitive ELISA-like assay based on gold-enhanced peroxidase-like immunogold activity

Abstract:A naked-eye sensitive ELISA-like assay was developed based on gold-enhanced peroxidase-like activity of gold nanoparticles (AuNPs). Using human IgG (H-IgG) as an analytical model, goat anti-human IgG antibody (anti-IgG) adsorbed on microtiter plate and AuNPs-labeled anti-IgG acted as capture antibody and detection antibody, respectively. Because the surfaces of AuNPs were blocked by protein molecules, the peroxidase-like activity of AuNPs was almost inhibited, evaluated by the catalytic oxidation of peroxidase enzyme substrate 3,3',5,5'-tetramethylbenzidine (TMB), which could produce a bright blue color in the presence of H2O2. Fortunately, the catalytic ability of AuNPs was dramatically increased by the deposition of gold due to the formation of a new gold shell on immunogold. Under optimal reaction conditions, the colorimetric immunoassay presented a good linear relationship in the range of 0.7-100 ng/mL and the limit of detection (LOD) of 0.3 ng/mL calculated by 3σ/S for UV-vis detection, and obtained LOD of 5 ng/mL for naked-eye detection. The obtained results were competitive with conventional sandwich ELISA with the LOD of 1.6 ng/mL. Furthermore, this developed colorimetric immunoassay was successfully applied to diluted human serum and fetal bovine serum samples, and predicted a broad prospect for the use of peroxidase-like activity involving nanomaterials in bioassay and diagnostics.
Advancing Protein Detection and Analysis Based on Ag/Au PHCN for Enhanced SERS Sensitivity and Specificity in Biomolecular Diagnostics

Abstract:In the realm of disease diagnostics, particularly for conditions such as proteinuria and hemoglobinuria, the quest for a method that combines accurate, label-free detection of protein compositions and their conformational changes remains a formidable challenge. In this study, we introduce an innovative Ag/Au plasmonic hybrid coupling nanoarray (Ag/Au PHCN) architecture marked by sub-10 nm interparticle gaps. These nanoarrays, leveraging plasmonic hybrid coupling and synergistic enhancement mechanisms, create a plethora of uniform surface-enhanced Raman spectroscopy (SERS) hotspots. The Ag/Au PHCN substrates demonstrated unparalleled sensitivity in the unmarked detection of hemoglobin (HGB), bovine serum albumin (BSA), and cytochrome C (Cyt.C) in bodily fluids, incorporating the advantages of high sensitivity, high reproducibility, durability, recyclability, and biocompatibility. Notably, the detection limits for BSA and HGB are unprecedented at 0.5 and 5 ng/mL, respectively. This achievement sets a new benchmark for label-free protein detection using two-dimensional nanostructures. Crucially, the Ag/Au PHCNs possess the novel capability to discern protein conformational changes post denaturation, underscoring their potential in probing protein functionalities. Most importantly, these nanoarrays can differentiate between normal and proteinuria-affected urine samples and monitor protein content variations over time, heralding a new era in clinical diagnostics with particular relevance to proteinuria and hemoglobinuria detection.

Abstract:Surface-enhanced Raman scattering (SERS) enables rapid detection of single molecules with high specificity. However, quantitative and sensitive SERS analysis has been a challenge due to the lack of reliable SERS-active materials. In this study, we developed a quantitative SERS-based immunoassay using enzyme-guided Ag growth on Raman labeling compound (RLC)-immobilized gold nanoparticle (Au NP)-assembled silica NPs (SiO2@Au-RLC@Ag). The enzyme amplified Ag+ reduction as well as Ag growth on the RLC-immobilized Au NP-assembled silica NPs (SiO2@Au-RLC), which resulted in a significant increase in SERS signal. In the presence of target antigens such as immunoglobulinG (IgG) or prostate-specific antigen (PSA), Ab1-Antigen-Ab2 immune complex with alkaline phosphatase triggered an enzyme- catalyzed reaction to convert 2-phospho-L-ascorbic acid (2-phospho-L-AA) to ascorbic acid (AA). As produced AA reduced Ag+ to Ag, forming an Ag hot spot on the surface of SiO2@Au-RLC, which enhanced the SERS signal of SiO2@Au-RLC@Ag in a solution with a target antigen concentration. The plasmonic immunoassay for IgG detection showed a high linearity of SERS intensity in the range of 0.6 to 9.0 ng/mL with a detection limit (LOD) of 0.09 ng/mL, while an LOD of 0.006 ng/mL was obtained for PSA. The results indicate that the sensitivity of our novel SERS-based immunoassay is higher than that of conventional enzyme-based colorimetric immunoassays.

Abstract:In this paper, we combine the advantages of aptamer, nanomaterial and antibody to design an electrochemical sandwich immunoassay for the ultrasensitive detection of human immunoglobulin E (IgE) by using methylene blue (MB) as electrochemical indicator. The sandwich structure is fabricated by using goat anti-human IgE as capturing probe. Aptamer-Au nanoparticles (NPs) conjugates are used both as a sandwich amplification element as well as an accumulation reagent of MB. Once the aptamer-Au NPs conjugates specifically bind to electrode surface, MB molecules are accumulated on its surface by the specific interaction of MB with G base of aptamer-Au NPs conjugates. Therefore, with the increase of human IgE concentration, more aptamer-Au-NPs conjugates are bound, and thus, more MB molecules are accumulated. A good linear relationship is obtained for the detection of human IgE over a range of 1–10,000ng/ml with a lowest detection limit of 0.52ng/ml. In addition, by using BSA, human IgA and human IgM as contrast, the excellent specificity of this sensing system for the detection of human IgE is also demonstrated.
A Porous Au-Ag Hybrid Nanoparticle Array with Broadband Absorption and High-Density Hotspots for Stable SERS Analysis

Abstract:Constructing high-density hotspots is of crucial importance in surface enhanced Raman scattering (SERS). In this paper, we present a large-area and broadband porous Au-Ag hybrid nanoparticle array which was fabricated by an ultra-thin alumina mask (UTAM) technique incorporated with annealing and galvanic replacement techniques. Experimental results and numerical simulations demonstrated that the porous Au-Ag hybrid nanoparticle array possessed enormous hotspots for high sensitivity, uniformity, and stability in SERS analysis. A large Raman enhancement factor of 2.2 × 107 was achieved with a relative standard deviation (RSD) of 7.7%, leading to excellent reliability for Raman detection. Furthermore, this novel substrate exhibited a long shelf time in an ambient environment and promising practical applications in many SERS-based quantitative analytical and biomedical sensing techniques.

Abstract:A sensitive sandwich type immunoassay has been proposed with the detection by inductively coupled plasma mass spectrometry (ICP-MS) in a single particle mode (time resolved analysis). The signal induced by the flash of ions (197Au+) due to the ionization of single Au-nanoparticle (Au-NP) label in the plasma torch can be measured by the mass spectrometer. The frequency of the transient signals is proportional to the concentration of Au-NPs labels. Characteristics of the signals obtained from Au-NPs of 20, 45 and 80nm in diameters were discussed. The analytical figures for the determination of Au-labeled IgG using ICP-MS in conventional integral mode and single particle mode were compared in detail. Rabbit-anti-human IgG was used as a model analyte in the sandwich immunoassay. A detection limit (3σ) of 0.1ngmL−1 was obtained for rabbit-anti-human IgG after immunoreactions, with a linear range of 0.3–10ngmL−1 and a RSD of 8.1% (2.0ngmL−1). Finally, the proposed method was successfully applied to spiked rabbit-anti-human IgG samples and rabbit-anti-human serum samples. The method resulted to be a highly sensitive ICP-MS based sandwich type immunoassay.

Abstract:Immunochromatographic sensors (ICSs) are inexpensive, simple, portable, and robust, thus making ICSs commonplace in clinical diagnoses, food testing, and environmental monitoring. However, commonly used gold nanoparticles (AuNPs) ICSs have low sensitivity. Therefore, we developed highly sensitive surface enhanced Raman scattering (SERS) ICSs. To enhance the sensitivity of SERS ICSs, rough surface core–shell Au@Ag nanoparticles (RSAu@AgNPs) were prepared by coating silver on the surface of gold nanoflowers (AuNFs). Then these nanoparticles were used as SERS substrate in the SERS ICSs, after which the SERS ICSs were implemented to detect haemoglobin and heavy metal cadmium ion (Cd2+). The limit of detection (LOD) of the SERS ICSs for detecting haemoglobin was 8 ng/mL, and the linear range of the SERS ICSs was from 31.3 to 2000 ng/mL. The LOD of the SERS ICSs for detecting Cd2+ was 0.05 ng/mL and the linear analysis range was from 0.05 to 25 ng/mL. The cross reactivity of the SERS ICSs was studied and results showed that the SERS ICSs exhibited highly specific for detection of haemoglobin and Cd2+, respectively. The SERS ICSs were then used to detect haemoglobin (spiked in serum and in stool) and Cd2+ (spiked in tap water, river water, and soil leaching water), and the results showed high recovery. These characteristics indicated that SERS ICSs were ideal tools for clinical diagnosis and environmental pollution monitoring.
