Abstract:A novel and convenient procedure for synthesizing 3 ',5 '-dithiothymidine was described. In this procedure, DBU was used to form the intramolecular ring of 2,3 '-anhydrothymidine and then the thioacetic acid was used as solvent as well as the nucleophilic reagent to produce S-acetyl-3 '-thiothymidine. A very efficient deprotection step was applied to afford the target compound, which can avoid the oxidization of the thiol groups. And the key intermediate, 5 '-O-tosyl-2,3 '-anhydrothymidine, was found to have regioselectivity for different nucleophiles.

Abstract:Nucleoside reverse transcriptase inhibitors are the only drugs so far approved for the treatment of AIDS. Nucleoside 5′-phosphorothioates is relatively resistant to enzymatic transformations. Novel 2′,3′- didehydro-2′, 3′-dideoxythymidine (d4T) 5′- thiophosphoramidates have been prepared by thiophosphorochloridate chemistry. These materials were designed to act as membrane-soluble prodrugs of the bioactive free nucleotides. In vitro evaluation reveals that the compounds have a pronounced, selective anti-HIV activity in MT-4 cells. The new compounds are characterized by NMR, IR and ESI-MS. The results from NMR experiments indicate that there are anti-orientation for glycosyl grouping in compounds 5a-5e.

Abstract:ABSTRACT Thymidine analogs, including 3′-azido-3′-deoxythymidine (AZT) and 2′,3′-dideoxy-3′-deoxythymidine (D4T), are important antiretroviral agents. To exert antiretroviral activity, these analogs undergo a stepwise phosphorylation intracellularly to the active triphosphate metabolites. We previously reported that 4′-substituted D4T with an ethynyl group (i.e., 4′-ethynyl D4T) increased the anti-human immunodeficiency virus (HIV) activity and was active against multidrug-resistant HIV strains. 4′-Ethynyl D4T is a better substrate for phosphorylation by human thymidine kinase 1 than D4T is. In this report, we first studied the enzymes involved in the phosphorylation of 4′-ethynyl D4T from monophosphate to triphosphate metabolites. The 4′-ethynyl D4TMP is phosphorylated by recombinant human TMP kinase with a K m of 19 ± 4 μM and a k cat of 0.007 ± 0.001 s −1 ; the relative efficiency is about 9 and 15% of those of D4TMP and AZTMP, respectively. Several enzymes from crude cellular extracts, including nucleoside diphosphate kinase, pyruvate kinase, creatine kinase, and 3-phosphoglycerate kinase, could phosphorylate 4′-ethynyl D4T-diphosphate. The relative phosphorylation efficiencies of 4′-ethynyl D4TDP were about 3 to 25% of those of D4TDP and were generally similar to those of AZTDP. In T-lymphoid cell lines, there was a preponderant accumulation of 4′-ethynyl D4TMP, suggesting that TMP kinase could be the rate-limiting enzyme in the metabolism of 4′-ethynyl D4T. Although the same enzymes are involved in the stepwise phosphorylation of thymidine analogs, their behaviors in phosphorylating metabolites of 4′-ethynyl D4T are different from those of D4T and AZT. Qualitatively, the metabolism of 4′-ethynyl D4T is more similar to that of AZT than to that of its progenitor, D4T.
A Practical Synthesis of Stavudine(d4T) from β-Thymidine

Abstract:A practical synthesis of stavudine (d4T) was described. β-Thymidine was treated with 1.5 equiv of Ph3CCl in DMF in the presence of pyridine to form 5′-O-trityl thymidine (2) in 93% yield. The compound 2 then reacted with 2 equiv. of methylsulfonyl chloride and 3 equiv. of Et3N to produce 3′-O-methylsulfonyl-5′-O-trityl thymidine (3) in 98% yield. After the compound 3 was converted into 2,3′-anhydro-5′-O-trityl thymidine(4)in 95% yield by refluxing with equal equiv. of NaOH in ethanol, the compound 4 was then subjected to elimination with 2 equiv. of sodium methoxide in DMSO to obtain 5′-O-trityl -3′-deoxy-2′, 3′-didehydro-thymidine(5) in 96% yield. Finally, the removal of trityl group of 5 in 80% HOAc afforded(1)in 90% yield. The conversion from compound 3 to compound 5 can also be done in one step in 84% yield by treating(3)with 4 equiv. of sodium methoxide in DMSO.

Abstract:A new class of 5-halo (or methoxy)-6-alkoxy (azido or hydroxy)-5,6-dihydro-2′,3′-didehydro-3′-deoxythymidines (4–17) were investigated as potential anti-AIDS drugs. These 5,6-dihydro derivatives, which are also potential prodmgs of 2′,3′-didehydro-3′-deoxythymidine (D4T) were designed to have properties which would enhance their duration of action, lipophilicity and cephalic delivery to the central nervous system. The 5,6-dihydro derivatives of D4T (4–15), which differ in configuration at the C-5 and C-6 positions, were synthesized by the regiospecific addition of XR (X = Br, Cl, I; R = OMe, OEt, N3, OH) to the 5,6-olefinic bond of D4T. These 5,6-disubstituted-5,6-dihydro analogs of D4T are more lipophilic (P = 0.70 – 4.0 range) than D4T (P = 0.12) and are stable to E. coli thymidine phosphorylase. Regeneration of the 5,6-olefinic bond to give D4T, upon incubation of the 5-bromo- and 5-iodo-6-methoxy-5,6-dihydro derivatives (6, 7, 10, 11) with glutathione or a mouse liver soluble enzyme fraction, was extensive (50–95%). The most potent anti-HIV-1 agents, 5-iodo-6-methoxy (10, 11), 5-bromo-6-azido (14, 15) and 5-methoxy-6-hydroxy (16, 17) derivatives of D4T, exhibited anti-HIV activities comparable to D4T. Abstract:Thymidine 5'-O-(pyrophosphoryl methylphosphonate) (dTTP alpha CH3) has been chemically synthesized by condensation of thymidine 5'-O-(methylphosphonate) with pyrophosphate. This novel nucleotide, which contained an alpha-phosphorus atom as methylphosphonate, was used as a substrate of terminal deoxynucleotidyltransferase (TDTase) in the presence of oligonucleotide (5'-GCTGTATCGTCAAGGCACTC-3') as an initiator. The reaction products were separated into two components by reverse-phase high-performance liquid chromatography (RP-HPLC). These products were, after purification, digested with nuclease P1 and alkaline phosphatase followed by separation of digested products by RP-HPLC. The result showed the presence of one of the isomers of 2'-deoxycytidyl-3'-methylphosphonyl-5'-thymidine (dCpCH3T) and 2'-deoxycytidyl-3'-methylphosphonyl-5'-thymidyl-3'-methyl phosphonyl-5'-thymidin e (dCpCH3TpCH3T), respectively. Fast atom bombardment mass spectrometry of these products further supported identification of the dinucleotide and the trinucleotide. These results indicated that dTTP alpha CH3 was used as a substrate of TDTase, resulting in methylphosphonate linkages. Produced oligomers were resistant to hydrolysis by snake venom phosphodiesterase I.

Abstract:This unit describes the chemical synthesis of α-L-threofuranosyl nucleic acid (TNA) triphosphates for thymidine (T), guanosine (G), cytidine (C), and the diaminopurine (D) analog of adenosine and their incorporation into TNA oligonucleotides by enzyme-mediated polymerization of a DNA primer-template complex. Starting from suitably protected threofuranosyl nucleosides, TNA triphosphates are synthesized in a single-pot reaction and purified by ion-exchange and HPLC chromatography. Purified TNA triphosphates are diluted into stock solutions and used as substrates for the synthesis of TNA oligonucleotides. Oligonucleotide synthesis is accomplished using Therminator DNA polymerase, a commercial variant of the 9(o)N DNA polymerase bearing the A485L mutation.

Abstract:In search of potential 2,3-dideoxyguanosine (ddG) and 2,3-didehydro-2,3-dideoxyguanosine (D4G) prodrugs, a series of 6-modified ddG, D4G analogs were synthesized and evaluated for their anti-HIV activities and cytotoxities in cell-based assays. All analogs showed low cytotoxicities and some of them displayed benign anti-HIV activities. The active triphosphate forms in vivo, ddGTP and D4TTP, were also synthesized by a novel and facile "one-pot" method. The recognition of ddGTP and D4TTP by Taq, Therminater DNA polymerase and HIV reverse transcriptase (RT) incorporated in DNA/RNA strands were investigated by a non-radioactivity method and K-m were determined.

Abstract:4′-Thionucleosides (thNAs) are synthetic nucleoside analogues that have attracted attention as leads for drug discovery in oncology and virology. Here we report a de novo thNA synthesis that relies on a scalable α-fluorination and aldol reaction of α-heteroaryl acetaldehydes followed by a streamlined process involving carbonyl reduction, mesylate formation and a double displacement reaction using NaSH. We demonstrate the multigram preparation of 4′-thio-5-methyluridine and highlight the production of purine and pyrimidine thNAs as well as C2′-modified thNAs.

Abstract:Synthesis of two new 1-[(2-hydroxycthoxy)methyl]-6-(5,6,7,8-tetrahydronaphthylmethyl-1)thymine derivatives 10a-b, as potent inhibitors against a mutant type of HIV, starting from thymine, is described. In the preparation of the corresponding 10a-b from compounds 9a-b, the three-step reaction via deprotection, hydrogenolysis, and hydrogenation was carried out in a one-pot procedure.

Abstract:An efficient and novel method for synthesizing 3′,5′-dithio-2′-deoxyguanosine was described. In this method normal guanosine was used as the starting material. A very efficient procedure was used to synthesize 2́-O-tosylguanosine 1, which used 0.1eq. DBTO instead of 2eq. 1 was treated with LTBH to give 9-(2-deoxy-β-d-threo-pentofuranosyl)guanine 2. 2 could be easily turned to the target compound.

In this procedure, the key intermediate 9 was synthesized by twice S(N)2 reactions with twice 3'-configuration inversions and then it was deprotected to title compound by removing three acyl groups in one pot.

Abstract:OBJECTIVE To find a commercially useful route for the preparation of 1-(2′-bromo-2′-deoxy-3′,5′-di-O-propionyl-β-D-ribosyl)thymine. METHOD To uses thymine and tetra-O-acylribofuranose as starting materials. The process involved the following steps: silylanization, condensation, deacetylation and halogenation. RESULTS The procedure was simple and the yield of this route was higher than other synthetic routes by using cheaper reagents.CONCLUSION The improved synthetic process is practical.

Abstract:5'-hydrogenphosphonothioate derivatives of AZT, d4T, and ddI as anti-HIV prodrug candidates were synthesized in 61-76% yields under mild conditions through sequential one-pot reactions, i.e., couple of triethylammonium phosphinate with different alcohols in the presence of pivaloyl chloride (PV-Cl), following oxidation with elemental sulfur and further condensation with AZT, d4T, or ddI in the presence of PV-Cl.

Abstract:The design of novel 4'-thionucleoside analogues bearing a C2' stereogenic all-carbon quaternary center is described. The synthesis involves a highly diastereoselective Mukaiyama aldol reaction, and a diastereoselective radical-based vinyl group transfer to generate the all-carbon stereogenic C2' center, along with different approaches to control the selectivity of the N-glycosidic bond. Intramolecular SN2-like cyclization of a mixture of acyclic thioaminals provided analogues with a pyrimidine nucleobase. A kinetic bias favoring cyclization of the 1',2'-anti thioaminal furnished the desired β-D-4'-thionucleoside analogue in a 7:1 ratio. DFT calculations suggest that this kinetic resolution originates from additional steric clash in the SN2-like transition state for 1',4'-trans isomers, causing a significant decrease in their reaction rate relative to 1',4'-cis counterparts. N-glycosylation of cyclic glycosyl donors with a purine nucleobase enabled the formation of novel 2-chloroadenine 4'-thionucleoside analogues. These proprietary molecules and other derivatives are currently being evaluated both in vitro and in vivo to establish their biological profiles.

Abstract:The chemical synthesis of thiazole-4-carboxamide adenine dinucleotide (TAD), previously identified as the active anabolite of the oncolytic 2-beta-D-ribofuranosylthiazole-4-carboxamide (TR), has been achieved by three different approaches: (1) incubation of adenosine 5'-monophosphate (AMP) and 2-beta-D-ribofuranosylthiazole-4-carboxamide 5'-monophosphate (TRMP) with excess DCC in aqueous pyridine, (2) reaction of adenosine 5'-phosphoromorpholidate with TRMP in pyridine, and (3) reaction of adenosine-5'-phosphoric di-n-butylphosphinothioic anhydride with TRMP in the presence of AgNO3. While the first approach produced only traces of TAD, the last two afforded 31 and 16% yields, respectively, of isolated TAD. The synthetic material was indistinguishable from biosynthesized TAD as judged by its HPLC behavior, NMR, UV and mass spectra, enzymatic resistance to alkaline phosphatase and susceptibility to venom phosphodiesterase, IMP dehydrogenase inhibitory activity, and cytotoxicity. TAD and TR were equally effective against murine P388 leukemia when employed at equimolar doses.

Abstract:Whole cells of Escherichia coli and the thermostable bacteria, Bacillus stearothermophilus, were used for the efficient synthesis of the biologically and industrially important compound, thymidine, using 2′-deoxyinosine and thymine as substrates. In this conversion, the 2′-deoxyribose moiety of 2′-deoxyinosine was transferred to thymine by the transdeoxyglycosylation activity of these bacterial cells. For example, in the case of Bacillus stearothermophilus, the yield of pure thymidine was 56% (78% conversion). When xanthine oxidase was added to this whole cell process, the product yield increased to 68% (90% conversion). In this transformation, Bacillus stearothermophilus was used at a temperature of 55°C where the solubility of thymine is much higher than at 25°C. The bacterial cells have activity over a broad pH range (approximately 4.0 to 8.0) and the yield of product varied within this pH range with the optimum pH being at 5.2. Both bacterial cells showed a sharp decrease in activity at alkaline pHs. Cells of both bacteria can be used repeatedly without appreciable loss of activity. Thymidine was also produced from thymine and 2′-deoxyinosine using isolated purine nucleoside phos-phorylase. A dramatic increase in conversion occurred with PNPase in the presence of xanthine oxidase.

Abstract:The action of 3'-azido-3'-deoxythymidine 5'-triphosphate (N3dTTP) on DNA strand elongation catalyzed by human immunodeficiency virus type 1 reverse transcriptase was evaluated in comparison with human DNA polymerase alpha and proliferating cell nuclear antigen-independent DNA polymerase delta. Sequencing gel analysis demonstrated that the human immunodeficiency virus 1 reverse transcriptase preferentially incorporated N3dTTP into the T sites of the growing DNA strands and caused chain termination in a dose-dependent manner. This effect was observed even when the N3dTTP concentration was 0.3 microM, 100-fold less than dTTP. Studies with reverse transcriptases from avian myeloblastosis virus and Moloney murine leukemia virus showed that N3dTTP was also efficiently incorporated into DNA by these enzymes and terminated DNA strand elongation. In contrast, human DNA polymerases alpha and delta did not incorporate detectable amounts of N3dTTP into the DNA and were not inhibited by 300 microM N3dTTP. The selective incorporation of the chain-terminating nucleotide by the viral reverse transcriptases appears to be a molecular basis for the positive therapeutic index of 3'-azido-3'-deoxythymidine.
