Abstract:A method of differential spore staining utilizing Wright's stain diluted one to five in a phosphate buffer solution of pH 7.6 and following the general technic of the Dorner method is outlined. Spores are stained a deep blue while the cytoplasm of the sporangium is stained a pinkish red.

Abstract:Giemsa stain is available in most developing nations for malarial parasite detection. In the absence of Wright stain, Giemsa is also used to perform differential assessments. We are comparing Giemsa and Wright stains using buffers to determine the optimal staining protocol for differential analysis. We compared Giemsa with reverse osmosis drinking water (RO), deionized water (DI), buffer at pH 7.2, to Wright’s stain with RO and DI, using Modified‐Wright’s stain with buffer in an automated slide stainer as the control. Blood samples (n=3) were drawn from the research team and analyzed using these staining protocols. Wright’s stain with DI performed equivalent to control for white blood cell (WBC), platelets (PLT), and red blood cell (RBC). Undiluted Giemsa with DI showed good staining of WBCs and PLTs, and moderate staining of RBCs (grayish discoloration). Diluted Giemsa with buffer stained RBCs poorly (pale ghost cells) but stained WBC, and PLT well. Diluted Giemsa with DI demonstrated good staining of PLTs, poor staining of RBCs, and moderate staining of WBCs. Undiluted Giemsa with RO showed poor staining of RBCs with moderate staining of PLTs and WBCs. Diluted Giemsa with RO has poor staining of RBCs and WBCs, and moderate staining of PLTs. Wright with RO stained RBCs, WBCs, and PLTs poorly. We recommend using Wright stain with DI water for differential analysis but undiluted Giemsa with DI water is an adequate substitute. Abstract:Abstract Wright-Giemsa staining is a common procedure that is performed routinely in hematology laboratories. Consistency in intra-laboratory staining quality is essential for accurate morphological interpretation of blood smears. Although the Wright-Giemsa stain can be challenging to perform, the methods illustrated here have provided consistent, high quality stains in the Special Hematology Laboratory at the University of Minnesota for over half a century. We outline methods for collecting blood specimens, preparing the slides and performing a Wright-Giemsa stain using our combination of reagents. Various techniques that have been passed down in our laboratory for troubleshooting suboptimally stained specimens are shared as well.

Abstract:SUMMARYLife-cycle stages of the microsporidia Nosema bombycis, the pathogen causing silkworm pebrine, were separated and purified by an improved method of Percoll-gradient centrifugation. Soluble protein fractions of late sporoblasts (spore precursor cells) and mature spores were analysed by two-dimensional polyacrylamide gel electrophoresis (2D-PAGE). Protein spots were recovered from gels and analysed by mass spectrometry. The most abundant differential protein spot was identified by database search to be a hypothetical spore wall protein. Using immunoelectron microscopy, we demonstrated that HSWP5 is localized to the exospore of mature spores and renamed it as spore wall protein 5 (NbSWP5). Further spore phagocytosis assays indicated that NbSWP5 can protect spores from phagocytic uptake by cultured insect cells. This spore wall protein may function both for structural integrity and in modulating host cell invasion.

Abstract:The pollen coat, which forms on the pollen surface, consists of a lipid–protein matrix. It protects pollen from desiccation and is involved in adhesion, pollen–stigma recognition, and pollen hydration during interactions with the stigma. The classical methods used for pollen coat observation are scanning and transmission electron microscopy. In this work, we screened a collection of fluorescence dyes and identified two fluorescent brighteners FB-52 and FB-184. When they were used together with the exine-specific dye, Basic fuchsin, the pollen coat and the exine structures could be clearly visualized in the pollen of Brassica napus. This co-staining method was applied successfully in staining pollen from Fraxinus chinensis, Calystegia hederacea, and Petunia hybrida. Using this method, small pollen coat-containing cavities were detected in the outer pollen wall layer of Oryza sativa and Zea mays. We further showed these dyes are compatible with fluorescent protein markers. In the Arabidopsis thaliana transgenic line of GFP-tagged pollen coat protein GRP19, GRP19-GFP was observed to form particles at the periphery of pollen coat. This simple staining method is expected to be widely used for the studies of the palynology as well as the pollen–stigma interaction.

Abstract:A single staining solution was made by compounding it in the following order (dyes were from British Drug Houses): ethanol, 10 ml; 1% malachite green in 95% ethanol, 1 ml; distilled water, 50 ml; glycerol 25 ml; phenol, 5 gm; chloral hydrate, 5 gm; acid fuchsin 1% in water, 5 ml; orange G, 1% in water 0.5 ml; and glacial acetic acid, 1–4 ml. For best results in differentiation to give green pollen walls and red protoplasm, the staining solution should be acidified with glacial acetic acid. The amount of acid to be added depends upon thickness of the pollen walls: for very thin-walled pollen, 1 ml; for moderately thin walls, 2 ml; and for thick-walled or spiny-walled pollen, 3 ml of acid. For pollen inside non-dehiscent anthers, 4 ml of acid should be used. Staining is hastened by flaming the slide (for loose thin-walled pollen) or by immersing thick-walled pollen or anthers for 24–48 hr at 50 C. In the typical stain, aborted pollen grains are green; nonaborted, red. The method is useful for pollen inside nondehiscent anthers if these are small and not too deeply coloured naturally. The stain is very durable, especially if the coverslips are sealed with param wax. The staining solution will keep well for about a month. It is useful both for angiosperms and gymnosperm microgametes.

Abstract:A new staining method for ultrathin sections is proposed, which gives preferential contrast to certain cell structures. The tissues are fixed with aldehyde and embedded in Epon or GMA. The staining is based on the use of chelating agents. After brief prestaining with uranyl acetate, the thin sections are floated for various lengths of time on McIlvaine buffer (0.2 M or less) or on EDTA (0.2 M or less). After this treatment, poststaining with lead citrate reveals a heavy contrast of cell structures known to contain RNA, whereas deoxyribonucleoproteins have lost most or all of the stain. Several exceptions to this rule are indicated. This procedure has the character of a regressive stain, where the chelating agents seem to play the role of differentiators.

Abstract:Transmission electron microscopy was used to study the development and structure of resting spores and sporosori of Polymyxa graminis. Osmiophilic bodies generally distributed in the cytoplasm moved to the periphery as spores matured. The spore wall consisted of four or five layers, two of which extended into spines linking adjacent spores. An unstained plug in the wall, faced towards the outside of the sporosorus. Some spores germinated in situ by producing primary zoospores. Bundle-like structures in some spores may have been barley yellow mosaic virus particles.

Abstract:The Dyer, Chance, alcian blue, and tannic acid-crystal violet wall stains for bacteria were compared. All gave apparent cell wall widths which were considerably greater than those demonstrated by electron microscope methods. The exaggerated width, as seen by the light microscope, was apparently due to staining of a portion of the cytoplasmic material adjacent to the wall, and in addition, to a precipitate on the surface of the cell produced by the Dyer and the Chance stains. Heat-fixed cells on slides were shown to retain considerable height or third dimension, being about half as high as wide. Moreover, such cells had their walls flattened against the slide to form a collar-like projection around the periphery of the shrunken protoplasm. This latter effect alone was not sufficient to explain the exaggerated wall thickness shown by staining. Here we reported a new method for ultra?thin sections staining which could batch stain ultra?thin sections based on a simple equipment for storing the dye solution and batch staining. This method improved the efficiency for ultra?thin section staining and reduced the pollution of samples. Furthermore, it also protected environment as well as the operators from heavy metal toxicity.

Abstract:Summary We present here a modified protocol for safranin-alcian blue staining of paraffin-embedded plant samples. After deparaffination, the double staining uses a 2:1 mix of 1% safranin and 1% alcian blue. This method is less expensive than previously published double staining protocols as it does not require absolute ethanol. Moreover, safranin-alcian blue staining creates a range of hues that allows good cell type differentiation, it gives reproducible results for a wide variety of plant tissues and is easy to use. We hope it will be found useful both for research and teaching purposes.

Abstract:Penicillium marneffei, Histoplasma capsulatum, Mucor and Leishmania donovani can lead to penicilliosis marneffei, histoplasmosis, mucormycosis and leishmaniasis, respectively, which, to a certain extent, share similar clinical manifestations. These pathogens are approximately the same size, therefore it is relatively difficult to rapidly diagnose the diseases. The aim of the present study was to explore a novel method that attempts to rapidly identify the pathogens of these diseases. In the Wright-Giemsa staining, the four pathogens were approximately the same size and mainly existed in macrophages. The multiplying P. marneffei had two nuclei, which were on both sides of the fungus, and had light cross-walls in the middle. H. capsulatum had a purplish nucleus, which occupied between one-third and one-half of the spore. The cytoplasm was light blue. Peripheral spores were observed in the form of an empty, bright ring without color, like a capsule. Generally, Mucor were observed to have a long and lightly stained area, which could be easily confused with the Wright staining of dinuclear P. marneffei. L. donovani exhibited a deep-staining kinetoplast near the nucleus. In the Periodic Acid Schiff (PAS) staining, the pathogens of P. marneffei and H. capsulatum were distinct and stained red. Differentiation between P. marneffei and H. capsulatum relied on their modes of reproduction: P. marneffei depends on fission, when the pathogens stretch into sausage-shapes and are split by a cross-wall, while H. capsulatum depends on budding so that narrow-necked, single spores can be formed. With PAS staining, the cell walls and intracellular contents of Mucor and L. donovani were not stained, lightly stained or granulated and discontinuous. In conclusion, this method, combining PAS and Wright-Giemsa staining, is simple and rapid, and may contribute to the effective identification of the four pathogens.

Abstract:A basic dye commercially used as the brightener agent in detergents of the washings, , Calcofluor white, was found by Darken to be useful for the vital staining of several bacteria. Our study was further developed in respect to a specificity of a differential staining between animal and plant cells. Several cells of plant origin such as bacteria, Candida and Chlorophyta can be stained by the dye, but all of the tissue culture cells of animal origin so far examined can not be stained at all. When 100mcg/ml of the dye was added to the cells without receiving any fixation procedure, the plant cells can be differentiated from the animal cells, instantaeneously.Although the living bacteria without any fixation are never stained with dyes such as methylene blue, the bacteria labeled with Calcofluor white, which were able to grow and divide, were stained with a solution of methylen blue.
Nutritionally defined conditions for germination of Streptomyces viridochromogenes spores

Abstract:Spores of Streptomyces viridochromogenes were removed from the surface of solid media with glass beads and suspended in a buffer-detergent solution. Addition of yeast extract and glucose resulted in rapid loss of refractility of the spores. Appearance of germ tubes followed. Germination was accompanied by a decrease in the optical density (OD) of the suspension. The OD decrease was used as an assay for germination. A defined germination medium (DGM) comprised of L-alanine, L-glutamic acid, adenosine, para-aminobenzoic acid, and calcium and magnesium ions provided a germination rate nearly equal to that of complex media. The germination rate was essentially the same if D-alanine and D-glutamate replaced the L-isomers. The optimum pH and temperature for germination were 7.0 and 35 C. Germination was absolutely dependent on the presence of CO2. Spores harvested after growth for longer periods than the usual time (10 days) became less germinable in DGM. The same was observed for spores grown at 37 C as compared with 30 C. Spores incubated in DGM for various time periods before being transferred to a buffer solution did not continue to germinate. Spores harvested after growth of eight species of Streptomyces did not show a decrease in OD when incubated in yeast extract medium. Another strain of S. viridochromogenes did exhibit an OD decrease in the medium. Comparative properties of spores of streptomycetes, fungi, and bacilli are discussed.
Staining and discrimination of Nosema bombycis spores with Calcofluor White M2R

Abstract:The fluorescent dye Calcofluor White M2R was applied to stain and identify Nosema bombycis spores. The results showed that the spores were stained in green-blue fluorescence. With this method it is easy to distinguish microsporidian spores from virus, bacteria and fungus spores.

Abstract:Nuclear morphology studies are a reliable parameter for analysis of genotoxic properties of different agents. Such studies have been widely exploited in isolated protoplasts but seldom used in cells isolated from whole root tissue due to cell wall permeability restrictions. We present a rapid and easy protocol that will benefit the researchers in studying the nucleus stained with fluorescent dye. Here, we have modified the digestion buffer and used a permeabilization buffer to ease the entry of the fluorescent Hoechst stain through the cell wall, so that fluorescence can be observed under UV light.

Abstract:A staining technique is described which detects variations in ferrous iron content within calcite and dolomite, whilst simultaneously differentiating the dolomite from the calcite. The application of this technique to a study of dedolomitisation is pre‐ sented. Ferroan dolomite, in certain pyrite bearing limestones of the southern Jura, is shown to have been dedolomitised, that is replaced by calcite, while at the same time the pyrite content of the rock was oxidised to haematite. One of the oxidation products of the pyrite is considered to have caused the dedolomitisation.

Abstract:Bacterial spores, known for their complex and resilient structures, have been the focus of visualization using various methodologies. In this study, we applied quick-freeze and replica electron microscopy techniques, allowing observation of spores in high-contrast and three-dimensional detail. This method facilitated visualization of the spore structure with enhanced resolution and provided new insights into the spores and their germination processes. We identified and described five distinct structures: (i) hair-like structures on the spore surface, (ii) spike formation on the surface of lysozyme-treated spores, (iii) the fractured appearance of the outer spore cortex during germination, (iv) potential connections between small vesicles and the core membrane, and (v) the evolving surface structure of nascent vegetative cells during germination.

Abstract:A new method for the isolation and enumeration of streptomycete spores from soil was developed. This method makes use of a cation-exchange resin to disperse soil particles. It allowed the detection of 10 spores in 100 g of sterile soil, while ca. 10 3 could be accurately enumerated in 100 g. This method was applied to studying the fate of a marked actinophage in soil. In sterile amended and nonsterile soil, relatively high numbers of actinophages were only found during the first few days of the experiment when the host streptomycete was in the mycelial form. Later, after sporulation, lysogens could be detected in sterile amended soil and could still be found 60 days after inoculation. Although no lysogens were found in nonsterile soil, the introduced phage could still be detected in the free state after 60 days, albeit at a low titer.
