Abstract:Fabricating low-cost and efficient biofilm carriers for moving bed biofilm reactors in wastewater treatment is crucial for achieving environmental sustainability. Herein, a novel sponge biocarrier doped with NaOH-loaded biochar and nano ferrous oxalate (sponge-C2FeO4@NBC) was prepared and evaluated for nitrogenous compounds removal from recirculating aquaculture systems (RAS) wastewater by stepwise increasing ammonium nitrogen (NH4+-N) loading rates. The prepared NBC, sponge-C2FeO4@NBC, and matured biofilms were characterized using SEM, FTIR, BET, and N-2 adsorption-desorption techniques. The results reveal that the highest removal rates of NH4+-N reached 99.28 +/- 1.3% was yielded by the bioreactor filled with sponge-C2FeO4@NBC, with no obvious nitrite (NO2--N) accumulation in the final phase. The reactor packed with sponge-C2FeO4@NBC biocarrier had the highest relative abundance of functional microorganisms responsible for nitrogen metabolism than in the control reactor, confirmed from 16S rRNA gene sequencing analysis. Our study provides new insights into the newly developed biocarriers for enhancing RAS biofilters treatment performance in keeping water quality within the acceptable level for the rearing of aquatic species.

Abstract:In this study, an airlift inner-loop sequencing batch reactor using poly(butylene succinate) as the biofilm carrier and carbon source was operated under an alternant aerobic/anoxic strategy for nitrogen removal in recirculating aquaculture system. The average TAN and nitrate removal rates of 47.35±15.62gNH4-Nm(-3)d(-1) and 0.64±0.14kgNO3-Nm(-3)d(-1) were achieved with no obvious nitrite accumulation (0.70±0.76mg/L) and the dissolved organic carbon in effluents was maintained at 148.38±39.06mg/L. Besides, the activities of dissimilatory nitrate reduction to ammonium and sulfate reduction activities were successfully inhibited. The proteome KEGG analysis illustrated that ammonia might be removed through heterotrophic nitrification, while the activities of nitrate and nitrite reductases were enhanced through aeration treatment. The microbial community analysis revealed that denitrifiers of Azoarcus and Simplicispira occupied the dominate abundance which accounted for the high nitrate removal performance. Overall, this study broadened our understanding of simultaneous nitrification and denitrification using biodegradable material as biofilm carrier.

Abstract:As a novel trend, solid carbon sources are applied to act as electron donors and biofilm carrier in biological denitrification process. In this study, simultaneous nitrate and ammonium removal process in an airlift sequencing batch reactor using 3-hydroxybutyrate-co-3-hydroxyvalerate (PHBV) as carbon source and biofilm carrier under intermittent aeration conditions was established to treat effluent of synthetic marine recirculating aquaculture system. The results showed that maximum nitrate and ammonia nitrogen removal rates of 0.45 and 0.09 kg m- 3 d-1 were achieved. No significant nitrite accumulation was found during 200-day operation, while effluent dissolved organic carbon accumulation and particle size reduction significantly increased. Microbial community analysis and batch tests illuminate that the generated sludge and attached biofilm played important roles in nitrogen removal. This study demonstrates the potential mechanism for the nitrogen removal process mediated by 3-hydroxybutyrateco-3-hydroxyvalerate and provide a new idea for the alternative solutions of solid carbon sources.
Enhancement of Mariculture Wastewater Treatment Using Moving Bed Biofilm Reactors Filled with Modified Biocarriers: Characterisation, Process Performance and Microbial Community Evaluation

Abstract:This research investigated two proposed modified biofilm carriers' performances in treating recirculating aquaculture systems (RAS) wastewater under different salinities (12%o, 26%o, and 35%o) for about 92 days. Three moving bed biofilm reactors (MBBRs; R1, R2, and R3) were filled with unmodified novel sponge biocarriers (SB) served as a control, modified novel SB with ferrous oxalate (C2FeO4@SB), and modified novel SB with combined ferrous oxalate and activated carbon (C2FeO4-AC@SB), respectively. Under the highest saline condition, a significantly higher ammonia removal efficiency of 98.86 +/- 0.7% (p > 0.05) was obtained in R3, whereas R2 and R1 yielded 95.18 +/- 2.8% and 91.66 +/- 1.5%, respectively. Microbial analysis showed that Vibrio, Ruegeria, Formosa, Thalassospira, and Denitromonas were predominant genera, strictly halophilic heterotrophic nitrifying bacteria involved in nitrogen removal. In conclusion, the synergistic effects of novel sponge, C2FeO4 and AC accelerated biofilm formations and stability, subsequently enhanced the removal of ammonia from the mariculture RAS wastewater by the C2FeO4-AC@SB carriers in R3.

Abstract:Hydrodynamics played an important role in the design and operation of bioreactors for wastewater treatment. In this work, an up-flow anaerobic hybrid bioreactor built-in with fixed bio-carriers was designed and optimized using computational fluid dynamics (CFD) simulation. The results indicated that the flow regime involving with vortex and dead zone was greatly affected by the positions of water inlet and bio-carrier modules. The ideal hydraulic features were obtained when the water inlet and bio-carrier modules located 9 cm and 60 cm above the bottom of reactor. Using the optimum hybrid system for nitrogen removal from wastewater with low carbon-to-nitrogen ratio (C/N = 3), the denitrification efficiency could reach 80.9 ± 0.4%. Illumina sequencing of 16S rRNA gene amplicons revealed that the microbial community divergence occurred among the biofilm on bio-carrier, the suspended sludge phase and the inoculum. Especially, the relative abundance of denitrifying genera Denitratisoma in the biofilm of bio-carrier reaches 5.73%, 6.2 times higher than that in the suspended sludge, implying the imbedded bio-carrier was conductive to enrich the specific denitrifiers to polish the denitrification performance with low carbon source. This work provided an effective method for the design optimization of bioreactor based on CFD simulation, and developed a hybrid reactor with fixed bio-carrier for nitrogen removal from wastewater with low C/N ratio.

Abstract:A renovated suspended carrier biofilm reactor was investigated for its capability of carbon oxidation and nitrification. The carrier used was polyvinyl chloride cylinder with 2.5mm diameter and 2.5–3.0mm height. The carrier filling ratio, which is defined as the volume ratio of carrier to the whole reactor, is a key characteristic parameter for the new reactor. The operational experiments under different carrier filling ratio ranging from 10 to 75% were conducted. Meanwhile, the pollutant removal rate and behaviour of the microorganisms, such as biomass and specific oxygen utilization rate (SOUR) activity, were monitored and analyzed. The results showed that when the influent chemical oxygen demand (COD) and ammonia was about 200 and 20mg/l, hydraulic retention time of 1.0h, the optimum carrier concentration was about 50%with the average COD and ammonia removal rate about 70 and 30%, respectively.

Abstract:Benefited from the massive filling bio-carriers, the packed cage rotating biological contactors (RBCs) have better performance and application potentiality in wastewater treatment. Investigating the effects mechanism of bio-carrier filling rate is crucial for such reactors management. In this study, the pollutants removal performance, biofilms physical characteristics, and microbial communities of the biofilms under a series of bio-carrier filling rates were analyzed. The results shown, the pollutant removal rate and amount were quite different under different filling rates, and biofilms structure and microbial composition were the main factors affecting the pollutants removal performance. With the increasing filling rates, the biofilms were more mass increased (dry weight from 0.066 to 0.148 g/per carrier), thicker (from 340.30 to 850.84 μm) and lower dense (from 0.068 to 0.060 g/cm3). The microbial community composition of those biofilms was also quite different at the genus level. The effects mechanism of bio-carrier filling rate can be summarized: the filling rates affect the physical and biological characteristics of biofilms, which will further affect the microenvironment and microbial distribution in biofilms, and then determines the pollutant metabolic rate and metabolic pathway. This study will contribute to design better bio-carrier filling rate according to different wastewater treatment scenario, and promote the performance optimization of packed cage RBCs.

Abstract:Simultaneous nitrification and denitrification (SND) is a promising denitrification method. But so far, there have been few reports on the SND and denitrification mechanism of the coupled system with sponge iron (SI). In this paper, the denitrification performance of the mono-sludge system (only activated sludge), mono-carrier system (activated sludge + polyurethane foam) and coupled system (activated sludge + polyurethane foam and SI) was investigated. The study found that compared with the mono-sludge system, the TN removal efficiencies of the mono-carrier system and coupled system increased by 24% and 29.2%, respectively. The change of dissolved oxygen in the carrier of the coupled system showed that the time of forming anoxic and anaerobic micro-environment in one cycle decreased continuously until it reached zero. The nitrate-dependent ferric oxidizing bacteria Acidovorax and Aquabacterium appeared in the coupled system. In addition, it was found that the intervention of SI significantly increased the activity of membrane nitrate reductase (NAR). This study provides practical and useful technology for the treatment of domestic sewage.

A Study of the System Performance and the Microbial Community Composition of Chemical Wastewater in an AO-MBBR Treatment Process

Abstract:To improve the nitrogen removal and reduce the chemical oxygen demand (COD) of a full-scale wastewater treatment plant, two sequential batch reactor devices were used to treat chemical wastewater with biocarriers in low carbon-to-nitrogen (C/N) ratio conditions. The results showed that the addition of biocarriers to the anoxic tank reduced the average concentration of COD in the effluent from 98.1 mg/L to 80.7 mg/L and increased total nitrogen (TN) removal by 9.4%. Metagenomic sequencing was performed to study the composition and function of microbial community samples taken from anoxic sludge and anoxic-carrier biofilms in this wastewater treatment plant. The results showed that Proteobacteria and Actinobacteria were the dominant phyla in the two samples, ensuring their capability for organic matter removal. The anoxic-carrier biofilms were mainly enriched with denitrifying bacteria such as Thauera (10.7%) and Comammonas (2.2%) and the anammox bacteria Candidatus Kuenenia (0.03%). Meanwhile, the nitrogen metabolism pathway was elaborated and the abundance of the functional genes involved in the nitrogen metabolism pathway was quantified. In addition, results from qPCR showed increased copy numbers of denitrification and anammox genes in the anoxic-carrier biofilms compared to those in the anoxic sludge, further confirming the enrichment of functional bacteria.

Abstract:Biofilm is an effective simultaneous denitrification and in situ sludge reduction system, and the characteristics of different biofilm carrier have important implications for biofilm growth and in situ sludge reduction. In this study, the performance and mechanism of in situ sludge reduction were compared between FSC-SBBR and SC-SBBR with constructed by composite floating spherical carriers (FSC) and multi-faceted polyethylene suspension carriers (SC), respectively. The variation of EPS concentration indicated that the biofilm formation of FSC was faster than SC. Compared with SCSBBR, the FSC-SBBR yielded 0.16 g MLSS/g COD, almost 27.27% less sludge. The average removal rates of COD and NH4+-N were 93.39% and 96.66%, respectively, which were 5.21% and 1.43% higher than the average removal rate of SC-SBBR. Investigation of the mechanisms of sludge reduction revealed that, energy uncoupling metabolism and sludge decay were the main factors for sludge reduction inducing 43.13% and 49.65% less sludge, respectively, in FSC-SBBR. EEM fluorescence spectroscopy and SUVA analysis showed that the hydrolytic capacity of biofilm attached in FSC was stronger than those of SC, and the hydrolysis of EPS released more DOM contributed to lysis-cryptic growth metabolism. In additional, Bacteroidetes and Mizugakiibacter associated with sludge reduction were the dominant phylum and genus in FCS-SBBR. Thus, the effect of simultaneous in situ sludge reduction and pollutant removal in FSC-SBBR was better.

Abstract:Moving bed biofilm reactor (MBBR) was used for treatment of mixed domestic and industrial wastewater, in order to study the removal efficiency of organic matters and investigate the influence of volumetric loading, carrier volumetric ratio, and hydraulic retention time (HRT) on the treatment effect. It is observed that under the conditions that carrier volumetric ratio is 60%, HRT in single reactor is 6 h, and volumetric loading is 4.0-5.0 kgCOD/(m~3·d), MBBR operates steadily with good effect, and the effluent COD is 60 mg/L in average, with removal rate more than 90%.

Abstract:Aiming for the reduction of excess sludge and the simultaneous removal of organic carbon and nitrogen, a new two-stage bioreactor system constructed by combining three-phase fluidized-bed (TFB) and sludge-reduction fixed-bed bioreactor (SFB) processes with different structured porous carriers was proposed. In this system, wastewater was first treated in the TFB bioreactor, in which the organic and nitrogenous compounds were simultaneously removed, and then the excess sludge produced in the TFB and residual organic substances flowed into the SFB for further degradation of suspended solids (SS) and nutrient removal. During a 470-day continuous operation, the average COD removal ratio reached about 95% at the influent COD of 500–600mg/L. The removal ratio of T-N could reach up to 28–55% and was carried out through both microbial growth and simultaneous nitrification and denitrification (SND). The SS from TFB could be reduced in the following fixed-bed from 160mg/L to 28mg/L with two SFBs without the settling tank. Therefore, the simultaneous removal of organic carbon and nitrogen with an on-site reduction of excess sludge could be achieved in this two-stage bioreactor.
Synergic Adsorption-Biodegradation by an Advanced Carrier for Enhanced Removal of High-Strength Nitrogen and Refractory Organics.

Abstract:Coking wastewater contains not only high-strength nitrogen but also toxic biorefractory organics. This study presents simultaneous removal of high-strength quinoline, carbon, and ammonium in coking wastewater by immobilized bacterial communities composed of a heterotrophic strain Pseudomonas sp. QG6 (hereafter referred as QG6), ammonia-oxidizing bacteria (AOB), and anaerobic ammonium oxidation bacteria (anammox). The bacterial immobilization was implemented with the help of a self-designed porous cubic carrier that created structured microenvironments including an inner layer adapted for anaerobic bacteria, a middle layer suitable for coaggregation of certain aerobic and anaerobic bacteria, and an outer layer for heterotrophic bacteria. By coating functional polyurethane foam (FPUF) with iron oxide nanoparticles (IONPs), the biocarrier (IONPs-FPUF) could provide a good outer-layer barrier for absorption and selective treatment of aromatic compounds by QG6, offer a conducive environment for anammox in the inner layer, and provide a mutualistic environment for AOB in the middle layer. Consequently, simultaneous nitrification and denitrification were reached with the significant removal of up to 322 mg L-1 (98%) NH4, 311 mg L-1 (99%) NO2, and 633 mg L-1 (97%) total nitrogen (8 mg L-1 averaged NO3 concentration was recorded in the effluent), accompanied by an efficient removal of chemical oxygen demand by 3286 mg L-1 (98%) and 350 mg L-1 (100%) quinoline. This study provides an alternative way to promote synergic adsorption and biodegradation with the help of a modified biocarrier that has great potential for treatment of wastewater containing high-strength carbon, toxic organic pollutants, and nitrogen.
[Pilot-Scale Study on Treatment of Municipal Sewage by Moving-Bed Biofilm Reactor with the Hydrophobically Modified Polyurethane Cubes As Biofilm Carriers].

Abstract:The carrier is the key influencers in moving bed biofilm reactor( MBBR), in this paper, a pilot scale apparatus was set up for treating municipal wastewater using modified cubic polyurethane carriers. For MBBR, the capacity of 3-3.5 t x d(-1), hydraulic residence time of 7-8 h, under the condition of continuous feed water (COD:140-280 mg x L(-1), NH4+ -N:30-50 mg x L(-1), TN: 45-65 mg x L(-1), TP:2.5-4.0 mg x L(-1)), the speed of biofilm formation and removal effects of COD, nitrogen and phosphorus were studied. After 140 days, the results showed that the formation of biofilm on the carrier was very quickly under 24-28 degrees C, and obtained stable treatment effect about 6 days. The COD, NH4+ -N, TN, TP average removal rates were 70%, 97%, 70%, 39%, respectively. As the temperature gradually decreased to about 12 degrees C, a high NH4+ -N removal rate (97%) could still be maintained, which indicating that the modified carrier can be achieved a high nitrification rate at low temperature.

Abstract:The bioreactor performance of a moving bed biofilm reactor (MBBR) as post-treatment of expanded granular sludge bed (EGSB) effluent was investigated. Moreover, the microbial communities of the two bioreactors during different operation periods were studied. The MBBR was efficient for COD removal with the mean efficiency of 82.4%, and produced an effluent with high and stable quality against shock loading resulting from the low temperature applied to EGSB. The study indicates that the microbial community in the reactors could adapt to perturbations such as influent wastewater characteristics and operation temperature, which is beneficial to maintain efficient and stable COD removal in the combined EGSB-MBBR system. Archaeal 16S rRNA gene sequence analysis indicated the presence of Methanomethylovorans , Methanolinea , Methanoregula boonei , Methanosarcina barkeri , and Methanospirillum hungatei in the EGSB. Bacterial 16S rRNA gene sequence analysis indicated the presence of Runella limosa , Dokdonella , Sphaerotilus , Hydrogenophaga , and Pseudomonas in the MBBR. The EGSB-MBBR system established here could be used as an efficient option for organic matter removal, which holds a great potential in practical applications for nutrients (N and P) removal.

Abstract:A laboratory-scale sequencing batch reactor (SBR) and two moving bed biofilm reactors (MBBRs) with different types of biocarriers were operated to treat the effluent of chemically enhanced primary sedimentation (CEPS). Due to the low organic strength and low carbon/nitrogen ratio of the CEPS effluent, COD and NH4+-N were effectively removed by the MBBRs but not by the SBR. Of the two MBBRs, MBBR2 filled with LEVAPOR biocarrier cubes performed even better than MBBR1 filled with K3 polystyrene biocarriers. The continuous decline of the sludge concentration in the SBR and the high and stable biomass content in MBBR2 contributed to their performances. High-throughput sequencing analysis showed that the reactors had selective effects on the bacterial community structure. Principal coordinate analysis indicated the different dynamic successions in the three reactors. Network analysis showed different community composition and diversity that were highly suggestive of different bacterial interactions among the three bioreactors.

Abstract:Optimization of nitrate removal rate under low carbon-to-nitrogen ratio has always been one of the research hotspots. Biofilm reactor based on functional carrier and using interspecific synergic effect of strains provides an insight. In this study, iron-loaded corn cob was used as a functional carrier that can contribute to the cellulose degradation, iron cycling, and collaborative denitrification process of microorganisms. During biofilm reactor operation, the maximum nitrate removal efficiency was 99.30% and could reach 81.73% at no carbon source. Dissolved organic carbon and carrier characterization showed that strain ZY7 promoted the release of carbon source. The crystallinity of cellulose I and II in carrier of experimental group increased by 31.26% and decreased by 21.83%, respectively, in comparison to the control group. Microbial community showed the synergistic effect among different strains. The vitality and metabolic activity of the target microorganisms in bioreactor were increased through interspecific bacterial cooperation.
Nitrogen and Phosphorus Removal Using Fluidized-Carriers in a Full-Scale A2O Biofilm System

Abstract:A full-scale biofilm system using fluidized-carriers integrated with anaerobic-anoxic–aerobic process (treatment capacity of 3.75 × 105 m3 d−1) was used for municipal wastewater treatment. The results indicated relatively higher removal efficiencies of 86% total nitrogen (TN), 97% ammonium (NH4-N) and 97% total phosphorus (TP) were achieved, with 0.32 mg L−1 TP, 0.81 mg L−1 NH4-N and 8.07 mg L−1 TN in the effluent, which meet the Class A of Discharge standard of pollutants for municipal wastewater treatment plant (GB18918-2002) of China. The results of microbial analysis indicated that the dominant microorganisms in the suspended sludge were Proteobacteria and Bacteroidetes at phylum level and β-Proteobacter at class level. The dominant microorganism in the biofilm was Proteobacteria at phylum level, with γ-Proteobacter (17.5%), β-Proteobacter (14%) and δ-Proteobacter (13.08%) distributed at class levels. The presence of Proteobacteria and Bacteroidetes in this system may be related with the phosphorus removal. A reddish color biofilm was formed on the surface of fluidized-carriers in the anaerobic tank and showed specific anammox ability, this may be related with the dominance of 0.0278% Planctomycetaceae at family level and 0.0278% Planctomycetales at order level. Besides the denitrification effects, the possible anammox bacteria present in the anaerobic tank might have also contributed to high nitrogen removal efficiency.
Preparation of Self-Releasing Carbon Biofilm Carrier Based on Corncob and Denitrification Properties

Abstract:Wastewater with a low carbon/nitrogen (C/N) ratio is widespread and difficult to treat. The addition of an external carbon source is an effective method for treating such wastewater. Therefore, we aimed to prepare a self-releasing carbon biofilm carrier using agricultural waste (corncobs), polyvinyl alcohol, and sponge iron in various ratios to provide a carbon source that would facilitate denitrification, providing an optimal environment for microorganisms. We found that the carbon release of the MAC biofilm carrier that accumulated over 60 d was 116.139 mg of chemical oxygen demand (COD)·g−1, whereas the accumulated total nitrogen release was approximately 0 mg·(g·d)−1. The NO3−-N removal rate after 24 h reached 98.1%, whereas the theoretical use rate of the carbon source (in terms of COD) was stable at 90.34%. In addition, the sum of the abundances of the denitrifying and cellulose-degrading bacteria was 49.89%. Furthermore, biofilm carriers are used as functional carriers that contribute to cellulose degradation, a process in which sponge iron produces Fe2+ to provide electron donors and shuttles for denitrifying bacteria and forms the iron cycle, thereby inducing an increase in microbial abundance; this increase then facilitates the microbial degradation of cellulose and synergistic denitrification through interspecific bacterial cooperation. This study provides a new and effective method for enhancing the denitrification of wastewater with low C/N ratios.

Abstract:A novel suspended carrier was prepared by sticking activated carbon (AC) and magnetite (Fe 3 O 4 ) onto polypropylene slices. Although this carrier could not reverse the decreased denitrification capacity trends under anoxic conditions at an influent carbon/nitrogen (C/N) ratio of 2, it enhanced denitrification by stimulating sludge reduction and accelerating electron transfer to certain extent. The carrier stuck by mixed AC/Fe 3 O 4 exhibited better performance in terms of sludge reduction, extracellular polymeric substances (EPS) secretion, and denitrification than that merely stuck by AC and Fe 3 O 4 at an influent C/N ratio of 2. The carrier stuck by mixed AC/Fe 3 O 4 increased the total nitrogen removal efficiency by 24.6 % ± 12.5 % in a 72-h denitrification batch experiment compared to the common polypropylene carrier. Moreover, the carrier improved EPS secretion and nitrogen metabolism and promoted the growth of Trichococcus and some denitrifying genera. This study provides a reference for the treatment of low C/N ratio sewage.
