Abstract:Anemonefish association with giant sea anemone is an iconic example of mutualistic symbiosis. Living inside the sea anemone without triggering the firing of highly toxic nematocysts present at the surface of sea anemone tentacles provides a unique shelter to the fish, which in return, by its territorial aggressiveness, protects the sea anemone from predators. The mechanisms by which the fish avoids triggering nematocysts discharge remain elusive. One hypothesis proposes that absence of sialic acids might disable nematocysts discharge. Here, we verified four predictions about the role of sialic acids in anemonefish protection: (i) sialic acid levels are lower in anemonefish mucus than in non-symbiotic and sensitive damselfish mucus; (ii) this decrease is specific to mucus and not observed in other organs; (iii) during post-embryonic development the levels of sialic acids are inversely correlated with the level of protection; (iv) the levels of sialic acids are minimal in sea anemone mucus. Taken together, our results allow us to propose a general model, in which anemonefish specifically regulates the level of sialic acids in their mucus to avoid nematocysts discharge. Our analysis also highlights several genes implicated in sialic acid removal as potential targets for allowing protection. Interestingly, our results also suggest that unrelated juveniles of damselfish ( ) capable to live in proximity with giant sea anemone may use the same mechanisms. Altogether, our data suggest that clownfish use sialic acids as a Trojan horse system to downplay the defenses of the sea anemones and illustrate the convergent tinkering used by fish to allow a mutualistic association with their hosts.

Abstract:Symbiosis, a mutualistic association, involves close interactions between two or more organisms. This relationship is particularly notable in clownfish, a commercially significant marine ornamental species, being played a major role in marine aquarium trade. In the wild, clownfish form species-specific symbiotic bonds with their host sea anemones. Of course, it is a well-documented interaction, however, the underlying mechanisms remain a mystery to researchers. The current study explores the symbiotic relationship between the sebae clownfish ( Amphiprion sebae ) and its host anemone ( Stichodactyla haddoni ), as well as a non-host anemone ( Heteractis magnifica ) in captive condition. Remarkably, this study is the first documentation about the successful association of sebae clownfish with a non-host anemone in captivity. The findings could enhance the marketability of sebae clownfish in the marine aquarium trade, offering new opportunities for enthusiasts and researchers alike.

Abstract:The mutualism between clownfishes (or anemonefishes) and their giant host sea anemones are among the most immediately recognizable animal interactions on the planet and have attracted a great deal of popular and scientific attention [ - ]. However, our evolutionary understanding of this iconic symbiosis comes almost entirely from studies on clownfishes— a charismatic group of 28 described species in the genus [ ]. Adaptation to venomous sea anemones (Anthozoa: Actiniaria) provided clownfishes with novel habitat space, ultimately triggering the adaptive radiation of the group [ ]. Clownfishes diverged from their free-living ancestors 25-30 MYA with their adaptive radiation to sea anemones dating to 13.2 MYA [ , ]. Far from being mere habitat space, the host sea anemones also receive substantial benefits from hosting clownfishes, making the mutualistic and co-dependent nature of the symbiosis well established [ , ]. Yet the evolutionary consequences of mutualism with clownfishes have remained a mystery from the host perspective. Here we use bait-capture sequencing to fully resolve the evolutionary relationships among the 10 nominal species of clownfish-hosting sea anemones for the first time ( ). Using time-calibrated divergence dating analyses we calculate divergence times of less than 25 MYA for each host species, with 9 of 10 host species having divergence times within the last 13 MYA ( ). The clownfish-hosting sea anemones thus diversified coincidently with clownfishes, potentially facilitating the clownfish adaptive radiation, and providing the first strong evidence for co-evolutionary patterns in this iconic partnership.

Abstract:The hermit crab-sea anemone (HCA) partnership is a common facultative mutualism. The anemone protects its host from predators such as cephalopods and shell crushing crabs by its stinging cells. The crab provides the anemone with a hard substrate and increased access to oxygenated water and food and in some cases with protection from predators. These partnerships are extremely diverse and complex. This overview addresses issues related to formation of the associations, the early history of these partnerships, placement of anemones on the hermit crab shell, intra and interspecific competition amongst hermit crabs over sea anemones, costs and benefits for crab and anemone from being associated and the evolution of these partnerships based on molecular phylogenies using nuclear and mitochondrial markers.

Abstract:The symbiosis between clownfishes (or anemonefishes) and their host sea anemones ranks among the most recognizable animal interactions on the planet. Found on coral reef habitats across the Indian and Pacific Oceans, 28 recognized species of clownfishes adaptively radiated from a common ancestor to live obligately with only 10 nominal species of host sea anemones. Are the host sea anemones truly less diverse than clownfishes? Did the symbiosis with clownfishes trigger a reciprocal adaptive radiation in sea anemones, or minimally, a co-evolutionary response to the mutualism? To address these questions, we combined fine- and broad-scale biogeographic sampling with multiple independent genomic datasets for the bubble-tip sea anemone, —the most common clownfish host anemone throughout the Indo-West Pacific. Fine-scale sampling and restriction site associated DNA sequencing (RADseq) throughout the Japanese Archipelago revealed three highly divergent cryptic species: two of which co-occur throughout the Ryukyu Islands and can be differentiated by the clownfish species they host. Remarkably, broader biogeographic sampling and bait-capture sequencing reveals that this pattern is not simply the result of local ecological processes unique to Japan, but part of a deeper evolutionary signal where some species of serve as host to the generalist clownfish species and others serve as host to the specialist clownfish . In total, we delimit at least five cryptic species in that have diversified within the last five million years. The rapid diversification of combined with functional ecological and phenotypic differentiation supports the hypothesis that this may represent an adaptive radiation in response to mutualism with clownfishes. Our data indicate that clownfishes are not merely settling in locally available hosts but recruiting to specialized host lineages with which they have co-evolved. These findings have important implications for understanding how the clownfish-sea anemone symbiosis has evolved and will shape future research agendas on this iconic model system.

Abstract:The fauna of the anemonefishes (Pomacentridae, Amphiprioninae) and their host sea anemones , inhabiting the detached coral reef of the Hon Rom Island, has been first described. Changes in faunas of these groups over 2003–2018 were traced. The species (qualitative) composition of these groups did not change over the period indicated above. It includes four anemonefish species: Amphiprion clarkii , A. frenatus, A. perideraion , and A. sandaracinos and six sea anemone species symbiotic with them: Entacmaea quadricolor , Stichodactyla mertensii , Heteractis aurora , H. crispa , H. magnifica , and H. malu . Four of these host sea anemone species ( S. mertensii , H. magnifica , E. quadricolor , and H. crispa ) belong to the principal host sea anemones of the reef anemonefishes, i.e. ensure the long-term existence of spawning pairs of symbiont fishes . However, quantitative changes in the faunas of the study groups have occurred. Two host sea anemone species ( S. mertensii and H. magnifica ) and mature anemonefishes inhabiting them were recorded each year in the observation period. The number of the sea anemones S. mertensii reduced by two thirds to 2018, compared to that in 2003–2005, and 100% of adult A. sandaracinos disappeared, while the number of adult A. clarkii within the study sea anemone species reduced by one third. The number of sea anemones H. magnifica reduced by 20% until 2018, compared to that in 2003–2010; 40% of adult A. perideraion disappeared. At least a threefold decrease in the pairs number of the anemonefish A. frenatus within the sea anemones E. quadricolor was observed; pairs of the anemonefish A. clarkii replaced them. It has been shown that there is no displacement of spawning pairs of A. sandaracinos , A. perideraion , and A. frenatus , respectively, from their host sea anemones S. mertensii , H. magnifica , and E. quadricolor , but there is replacement of these anemonefish species in the fish-free sea anemones by another anemonefish species— A. clarkii .

Abstract:Coral reef growth and recovery from bleaching depend on the onset and physiological maintenance of cnidarian-dinoflagellate symbiosis. Many cnidarian hosts can acquire new symbionts (family Symbiodiniaceae) from the environment. Symbiodiniaceae can be found at significant densities within other hosts and lesser densities as free-living populations on macroalgae, in sediments, and in the water column. The mechanisms underlining host selectivity towards symbionts are still under investigation, and even less is known about the timing and community composition when hosts uptake symbionts in their natural environments. This project studied symbiont diversity in environmental reservoirs and the progression of symbiont acquisition in bleached hosts. We first characterized various Symbiodiniaceae reservoirs in an estuarine habitat of Florida Bay at Key Largo, Florida. We then deployed bleached Exaiptasia sp. anemones in the habitat and recorded their recovery for three weeks, assessing symbiont density through cell counting and diversity via next-generation sequencing (NGS). We found that environmental reservoirs exhibit higher symbiont diversity but much lower densities than in hospite living reservoirs, highlighting the potential for healthy hosts as symbiont donors. We showed that the anemones can recover from bleaching using their residual leftover symbiont cells but recover 40% faster when also uptaking new symbionts from their environment, including non-homologous strains. We also report that symbiont shuffling can be observed in disturbed anemones in as little as three days, with decreasing symbiont density but higher symbiont diversity. Our data can guide future studies towards understanding small-scale dynamics and symbiont acquisition for cnidarians recovering from bleaching.

Abstract:The mutualistic symbiosis between anthozoans and intra-gastrodermal dinoflagellates of the family Symbiodiniaceae is the functional basis of all coral reef ecosystems, with the latter providing up to 95% of their fixed photosynthate to their hosts in exchange for nutrients. However, recent studies of sponges, jellyfish, and anemones have revealed the potential for this mutualistic relationship to shift to parasitism under stressful conditions. Over a period of eight weeks, we compared the physiological conditions of both inoculated and aposymbiotic anemones ( Exaiptasia pallida ) that were either fed or starved. By the sixth week, both fed groups of anemones were significantly larger than their starved counterparts. Moreover, inoculated and starved anemones tended to disintegrate into “tissue balls” within eight weeks, and 25% of the samples died; in contrast, starved aposymbiotic anemones required six months to form tissue balls, and no anemones from this group died. Our results show that the dinoflagellates within inoculated anemones may have posed a fatal metabolic burden on their hosts during starvation; this may be because of the need to prioritize their own metabolism and nourishment at the expense of their hosts. Collectively, our study reveals the potential of this dynamic symbiotic association to shift away from mutualism during food-deprived conditions.

Abstract:Symbiotic relationships are common in marine ecosystems, especially between cnidarians and crustaceans. This study explored the association of the arrow crab Stenorhynchus lanceolatus with different hosts in the Canary Islands, focusing on the relationship with the sea anemone Telmatactis cricoides . Field surveys and laboratory experiments were conducted to assess crab preference among potential hosts (anemones and sea urchins), association rates and behaviours of crabs interacting with the anemone. Field results revealed similar prevalence on anemones and sea urchins, and that most crabs associated to the anemone were solitary females. However, under laboratory conditions, sea urchins were observed to be preferred as hosts, but association with anemones were more stable in time, although its colonization period was slower, likely due to their toxicity and lack of environmental cues. Additionally, crab intraspecific competition seemed to favour the association with the anemone. This study showed the mutualistic and facultative nature of the relationship between S. lanceolatus and T. cricoides, advancing our understanding of benthic ecosystems dynamics

Self/Non-Self Recognition Affects Cnida Discharge and Tentacle Contraction in the Sea Anemone Haliplanella luciae

Abstract:Certain species of sea anemone live in tightly packed communities, among clonemates and non-clonemates. Competition for space leads to intraspecific and interspecific aggressive interactions among anemones. The initial aggressive interactions appear to involve reciprocal discharge of cnidae triggered by contact with non-self feeding tentacles. We asked whether molecules contained in anemone-derived mucus constituted an important cue alone or in combination with cell surface molecules in stimulating aggressive or avoidance behaviors. In this study, we found that self and non-self stimuli differentially influenced two effector systems: cnida discharge and tentacle contraction. Interspecific mucus enhanced nematocyst discharge by 44% and spirocyst discharge by 90%, as compared to baseline discharge obtained in seawater alone. Conspecific stimuli accompanying touch inhibited specific tentacle contractions occurring on the far side of anemones relative to the site of contact. The greatest tentacle contractions occurred with exposure to interspecific mucus and tissue. Thus, several receptor systems are involved that integrate chemical and mechanical cues in order to initiate appropriate and graded effector responses during competition for space.
Venomics Reveals the Venom Complexity of Sea Anemone Heteractis magnifica

Abstract:The venoms of various sea anemones are rich in diverse toxins, which usually play a dual role in capturing prey and deterring predators. However, the complex components of such venoms have not been well known yet. Here, venomics of integrating transcriptomic and proteomic technologies was applied for the first time to identify putative protein and peptide toxins from different tissues of the representative sea anemone, Heteractis magnifica. The transcriptomic analysis of H. magnifica identified 728 putative toxin sequences, including 442 and 381 from the tentacles and the column, respectively, and they were assigned to 68 gene superfamilies. The proteomic analysis confirmed 101 protein and peptide toxins in the venom, including 91 in the tentacles and 39 in the column. The integrated venomics also confirmed that some toxins such as the ShK-like peptides and defensins are co-expressed in both the tentacles and the column. Meanwhile, a homology analysis was conducted to predict the three-dimensional structures and potential activity of seven representative toxins. Altogether, this venomics study revealed the venom complexity of H. magnifica, which will help deepen our understanding of cnidarian toxins, thereby supporting the in-depth development of valuable marine drugs.

Advances In Marine Symbiotic Cyanobacteria

Abstract:Marine microbial symbionts represent a hotspot in the field of marine microbiology. Marine plants and animals, such as sponge, sea squirt, worm, and algae host symbiotic cyanobacteria with great diversity. Most of the symbiotic cyanobacteria are host-specific and can be transmitted directly from parent to offspring. Symbiotic cyanobacteria play an important role in nitrogen fixation, nutrition and energy transfer and are possible true producers of bioactive marine natural products. Though diverse cyanobacteria have been revealed by culture-independent methods, the isolation and culture of symbiotic cyanobacteria is a challenge. In this chapter, the advances in diversity, transmission, symbiotic relationship with the host, isolation and natural products of marine symbiotic cyanobacteria are reviewed.

Abstract:Abstract The deep sea is one of the most extensive ecosystems on earth. Organisms living there survive in an extremely harsh environment, and their mitochondrial energy metabolism might be a result of evolution. As one of the most important organelles, mitochondria generate energy through energy metabolism and play an important role in almost all biological activities. In this study, the mitogenome of a deep‐sea sea anemone (Bolocera sp.) was sequenced and characterized. Like other metazoans, it contained 13 energy pathway protein‐coding genes and two ribosomal RNAs. However, it also exhibited some unique features: just two transfer RNA genes, two group I introns, two transposon‐like noncanonical open reading frames (ORFs), and a control region‐like (CR‐like) element. All of the mitochondrial genes were coded by the same strand (the H‐strand). The genetic order and orientation were identical to those of most sequenced actiniarians. Phylogenetic analyses showed that this species was closely related to Bolocera tuediae. Positive selection analysis showed that three residues (31 L and 42 N in ATP6, 570 S in ND5) of Bolocera sp. were positively selected sites. By comparing these features with those of shallow sea anemone species, we deduced that these novel gene features may influence the activity of mitochondrial genes. This study may provide some clues regarding the adaptation of Bolocera sp. to the deep‐sea environment.
A New Species of Sea Whip Gorgonian-Associated Zoantharian (Cnidaria: Anthozoa: Hexacorallia: Parazoanthidae) from the Ryukyu Islands, Japan, with Subgeneric Subdivision of Genus Umimayanthus

Abstract:Symbioses between invertebrates are common in the ocean although usually the diversity and specificity of their interactions are not well understood. Parazoanthidae (Cnidaria: Anthozoa: Zoantharia) is one of the most diverse zoantharian families in terms of numbers of genera and species. Species in this family are commonly associated with various other invertebrates that they utilize as their substrate. Previous studies have re-organized the taxonomy of Parazoanthidae and revealed a strong specificity between many parazoanthid species and genera and their substrates. However, our understanding of the species diversity of Parazoanthidae is far from complete, as parazoanthids are often overlooked in sampling surveys. In this study, we establish three subgenera under the genus Umimayanthus Montenegro, Sinniger, and Reimer, 2015; the nominotypical Umimayanthus, Paraumimayanthus subgen nov., and Gorgoniazoanthus subgen. nov., based on the finding of a new species, Umimayanthus (Gorgoniazoanthus) kanabou sp. nov., associated with the sea-whip gorgonian Ellisella sp. from approximately 30 m depth in shallow mesophotic coral reef communities in Oura Bay on Okinawajima Island and in Oshima Strait near Amami-Oshima Island, in the Ryukyu Islands, southern Japan. We additionally report on gastropods and crustaceans observed in association with U. kanabou, and these species are thought to potentially prey upon the zoantharians or on gorgonian polyps. Umimayanthus kanabou is phylogenetically closely related to congeneric sponge-associated Umimayanthus spp., further supporting the recent hypothesis that substrate preferences may change during the evolutionary history of zoantharians.

Abstract:Widespread non-native species tend to demonstrate an apparent lack of selectivity in habitat requirements, feeding regimes, and reproductive needs, while displaying a tendency to thrive in human-modified habitats. The high phenotypic plasticity typical of sessile, substrate-attached marine species may enhance their chances of survival and spread in a new region. Anthropogenic activities have changed marine habitats over a wide range of phenomena, including water temperature, community species composition, and the types of available substrates, creating new physical and biotic regimes that may contribute to the potential for successful species introduction. Here we examine ten species of sea anemones that have been introduced outside of their native range, and elucidate specific characteristics that are common among globally introduced sea anemones. Various life history strategies enable these species to survive and flourish through transport, introduction, establishment and spread, leading to the successful colonization of a new geographic area. Considering life history strategies and weighing of vector potential, we suggest conditions that facilitate introduction of these species, and identify species of sea anemones that may be introduced in the future in the face of changing climate and increased anthropogenic activities.

In situ observation on two 'elusive' rhodaliid siphonophores (Cnidaria; Hydrozoa; Siphonophora) from the Red Sea, including a potential new species within the genus Archangelopsis

Abstract:Rhodaliids, a specific family of siphonophores inhabiting the benthic zone, have remained enigmatic due to their rarity and elusive nature. These unique organisms, primarily found in open ocean habitats, exhibit distinctive features and are characterized by complex structures. During the Red Sea Decade Expedition, two rhodaliid specimens were collected at the sea bed at water depths of 438 and 495 meters. Regardless of challenges in specimen preservation, detailed morphological analysis revealed unique characteristics. Genetic analysis, employing the 16S rRNA marker, revealed one specimen closely related to Thermopalia taraxaca , while the taxonomic traits of this specimen suggested the identification of Archangelopsis jagoa . While most of the morphological features of the second specimen were also very similar to A. jagoa , some differences in coloration suggest the discovery of either an unusual colour variant or a potential new species within this genus. Notably, this study reports the first sequencing of A. jagoa . This species demonstrated a capacity to thrive in low-oxygen environments, challenging conventional assumptions about their habitat requirements. Despite difficulties in specimen handling and genetic analysis limitations due to a lack of comprehensive data, this research sheds light on the elusive world of benthic rhodaliids.

Abstract:Symbiotic cnidarians such as corals and anemones form highly productive and biodiverse coral reef ecosystems in nutrient-poor ocean environments, a phenomenon known as Darwin's paradox. Resolving this paradox requires elucidating the molecular bases of efficient nutrient distribution and recycling in the cnidarian-dinoflagellate symbiosis. Using the sea anemone Aiptasia, we show that during symbiosis, the increased availability of glucose and the presence of the algae jointly induce the coordinated up-regulation and relocalization of glucose and ammonium transporters. These molecular responses are critical to support symbiont functioning and organism-wide nitrogen assimilation through glutamine synthetase/glutamate synthase-mediated amino acid biosynthesis. Our results reveal crucial aspects of the molecular mechanisms underlying nitrogen conservation and recycling in these organisms that allow them to thrive in the nitrogen-poor ocean environments.
