Abstract:Aluminum cookware are widely used in many parts of the world. Data is increasing on the leaching of toxic metals from aluminum cookware into food and drink. In the present study, cytogenotoxicity of water boiled in three different aluminum pots (new, 3-year-old, and 6-year-old) in onion root tip's dividing cells was evaluated using the Allium cepa assay. The concentrations of Pb, As, Cd, Cu, Ni, and Al in the samples were also analyzed. Onion bulbs were grown in the boiled water samples, while tap water served as the control. Cytological and genetic analyses were carried out after 48 h, while analysis of inhibition of root length was carried out after 72 h. The results showed a significant (p < 0.05) cell proliferation and root growth inhibition compared with the control, which is dependent on the duration of use of the aluminum pots. The boiled water samples also caused modification of the root morphology as well as chromosomal aberrations which include sticky chromosomes, anaphase bridge, and disturbed spindle. The highest cytogenotoxicity was observed in the 6-year-old aluminum pot and the least in the new aluminum pot. Pb, As, Cd, Cu, Ni, and Al analyzed in the samples, with the highest concentrations in the 6-year-old aluminum pot, were believed to be responsible for the cytogenotoxicity observed in the A. cepa assay. The data of this study are indications that the aluminum pot–boiled water contains substances with the potential to be cytotoxic and cause mutations in somatic cells of A. cepa.

Abstract:Objective:Aluminum toxicity in acidic soil is a serious problem worldwide,which results in poor quality and big loss of wheat produc- tion.Therefore,studies on the molecular mechanism of Aluminum tolerence has become one of the important tasks in wheat stress resistance re- search.Methods:In this study,hexaploid wheats Al-sensitive cultivar Scout66,Al-tolerant cultivar Atlas66 and an elite wheat cultivar in He- bei province EM12 were analyzed embryo induction frequency and changes in cell ultra-structure under different concentrations of Aluminum ion stress by tissue culture method and transmission electron microscopy (TEM) method respectively.Results:The results showed that Al stress re- duced the embryo induction frequency and restrained the differentiation of embryo (immature embryo induction reduced from 94.0% to 25.3%; mature embryo induction reduced from 75.0% to 17.3% );Al stress badly destroyed the ultra-structure of root tip cells,the nucleus changed uneven,the numbers of vacuole and mitochondria increased but crista of mitochondria changed unclear and almost disappeared.Conclusion: Based on the above-result,the conclusion can be drawn out that Al damages the cell ultra-structure especially in nucleus and mitochondria, which implies that the damage of cell ultra-structure is probably the key factor in Al inhibition of cell differentiation and division.
Studies on the in Vivo and in Vitro Mutagenicity and the Lipid Peroxidation of Chlorinated Surface (drinking) Water in Rats and Metabolically Competent Human Cells

Abstract:In the present study, DNA damaging and mutagenic effects of chlorinated drinking water (CDW) extracts obtained from polluted raw water resources were examined in metabolically competent human Hep G2 hepatoma cells using the in vitro micronucleus assay and the single cell gel electrophoresis (SCGE, comet assay). Additionally, the in vivo induction of micronuclei (MN) was studied in polychromatic erythrocytes (PCEs) derived from bone marrow of CDW-treated Wistar rats. Furthermore, we examined the influence of CDW on the lipid peroxidation (LpO) in blood, liver, kidney and testicle of rats. The results demonstrated significant increases of micronucleated PCEs in the bone marrow of rats fed with relatively low CDW doses (33.3ml/kg body weight per day). Similar effects, i.e. increases of MN frequencies, were found in Hep G2 hepatoma cells after CDW treatment (41 MN/1000 binucleated cells (BNCs) for 167ml CDW) in comparison to the vehicle control (24 MN/1000 BNC). Additionally, DNA damages caused by CDW were observed in the comet assay. As a product of LpO, the levels of malondialdehyde (MDA) were significantly enhanced almost in all animals and organs tested after CDW treatment. In livers and serum of rats dose-dependent increases of MDA were observed. The data indicated that extracts from CDW obtained from polluted raw water were able to cause oxidative damages and to induce various biological effects in mammalian cells in vivo and in vitro, i.e. clastogenicity and/or aneugenicity, DNA strand breaks and/or alkali-labile damages. The consistency of the results among the various biological systems and endpoints led to the conclusion that the consumption of chlorinated drinking water obtained from polluted raw water may enhance the body burden with mutagenic and/or carcinogenic substances and therefore, means a potential genetic hazard for human health.
Inhibition Of Cell Growth Caused By Aluminum Toxicity Results From Aluminum-Induced Cell Death In Barley Suspension Cells

Abstract:The inhibition of cell growth caused by aluminum (Al) toxicity was studied in barley (Hordeum vulgare L., cv. Zaoshu 3 from our laboratory) suspension cells in this paper. The growth of suspension cells in 0.1-5.0 mM Al treatment groups was obviously inhibited in 24-h Al treatments and was not recovered during 24-h recovery culture without Al. On the average, 46.70% or 83.36% of suspension cells lost viability respectively during 24-h Al treatments or 24-h recovery culture by the detection of trypan blue staining, The agarose gel electrophoresis shows that DNA ladders were induced after 24-h Al (0.1-5.0 mM) treatments. Al toxicity significantly inhibits cell division and promotes chromosomal variation, but the mitotic index (MI) of suspension cells is so low that the inhibition of Al toxicity on cell division does not cause the significant inhibition of cell growth after short-term exposure to Al. These results suggest that Al-induced inhibition of cell growth mainly results from cell death occurring during Al treatments or recovery culture, which can occur possibly via programmed cell death (PCD)-related signal transduction pathway.

Abstract:Objective To explore the effects of chemical reaction of aluminum salt in water solution on developmental toxicity.Methods Inhibiting actions on embryo growth and development,and organ morphogenetic differentiation in rats induced by newly prepared and 11.5-month stored aluminum sulfate solution were observed and compared using whole embryo culture method in rats.Results Both of the two observed solutions showed a certain embryo toxicity and teratogenicity.The toxicity of stored solution decreased significantly compared with that of newly prepared solution.At the same concentration of aluminum,the total Brown's morphogenetic score reflecting the every index of embryo growth and development and organ morphogenetic differentiation,the incidence rates of terata and the number of dead embryos were significant lower in rats exposed to stored solution compared with those exposed to newly prepared solution,especially the morbidity of embryos.Conclusion The complexation reaction between aluminum ions (Al3+) and water molecules in stored aluminum sulfate solution resulted in the changes of existing state of Al3+ and its decreasing developmental toxicity to rats.
Effect of drinking water disinfection by-products in human peripheral blood lymphocytes and sperm

Abstract:Background: Drinking water disinfection by-products (DBPs) are generated by the chemical disinfection of water and may pose hazards to public health. Two major classes of DBPs are found in finished drinking water: haloacetic acids (HAAs) and trihalomethanes (THMs). HAAs are formed following disinfection with chlorine, which reacts with iodide and bromide in the water. Previously the HAAs were shown to be cytotoxic, genotoxic, mutagenic, teratogenic and carcinogenic. Objectives: To determine the effect of HAAs in human somatic and germ cells and whether oxidative stress is involved in genotoxic action. In the present study both somatic and germ cells have been examined as peripheral blood lymphocytes and sperm. Methods: The effects of three HAA compounds: iodoacetic acid (IAA), bromoacetic acid (BAA) and chloroacetic acid (CAA) were investigated. After determining appropriate concentration responses, oxygen radical involvement with the antioxidants, butylated hydroxanisole (BHA) and the enzyme catalase, were investigated in the single cell gel electrophoresis (Comet) assay under alkaline conditions, >pH 13 and the micronucleus assay. Results: In the Comet assay, BHA and catalase were able to reduce DNA damage in each cell type compared to HAA alone. In the micronucleus assay, micronuclei (MNi) were found in peripheral lymphocytes exposed to all three HAAs and catalase and BHA were in general, able to reduce MNi induction, suggesting oxygen radicals play a role in both assays. Conclusion: These observations are of concern to public health since both human somatic and germ cells show similar genotoxic responses.

Aluminum contamination of food during culinary preparation: Case study with aluminum foil and consumers' preferences

Abstract:The aim of the work was to estimate the degree of aluminum leakage from aluminum foil during baking process of selected food/meals. The experiment included 11 different types of food (Atlantic salmon Salmo salar, mackerel Scomber scombrus, duck breasts, cheese Hermelín, tomato, paprika, Carlsbad dumplings, pork roast, pork neck, chicken breasts, and chicken thighs) baked both marinated and not marinated. The aluminum content was measured by AAS and ICP/MS methods. The highest aluminum increase was observed in the samples of marinated Salmo salar (41.86 ± 0.56 mg/kg), Scomber scombrus (49.34 ± 0.44 mg/kg), and duck breast (117.26 ± 1.37 g/kg). The research was also supported by the survey that consisted of 784 respondents with different sociodemographic characteristics. The study clearly showed the occurrence of aluminum contamination of food when it is prepared by baking in aluminum foil. It cannot be concluded that aluminum leakage will occur with each type of food. The aluminum contents found among investigated samples are not alarming, though the increase was measured up to 40 times. On the other hand, revealed aluminum contents can represent a risk for younger/smaller children and for individuals with diagnosed certain ailments.
[The Investigation of DNA Double-Strand Breaks of Lymphocytes in Workers Exposed to Lead and DNA Double-Strand Breaks of Human Lymphocytes Induced by Lead in Vitro].

Abstract:OBJECTIVE To study DNA double-strand breaks of human peripheral lymphocytes exposed to lead with flow cytometry (FCM). METHODS The lymphocytes were obtained from 36 workers occupationally exposed to lead and 70 residents without occupational exposure to lead. DNA double-strand breaks were detected by flow cytometer assay. The lymphocytes from health people were incubated with lead at different doses and time, FCM assay was used to detect DNA double-strand breaks. RESULTS DNA double-strand breaks and fluorescence intensity of high exposed group and low exposed group were 41.76% ± 28.57%, 9.90 ± 3.35 and 33.18% ± 30.64%, 9.39 ± 4.83, respectively, which were significantly higher than those (0.28% ± 0.28% and 6.95 ± 2.93) of control group (P<0.05). The results of in vitro experiment indicated that DNA double-strand breaks of lymphocytes exposed to Pb at the dose of 125.0, 250.0, 500.0 µmol/L for 1 and 2 h were significantly different from those of the negative control group and positive control group (P<0.01). DNA double-strand breaks increased at beginning and then decreased with lead doses. CONCLUSION Lead can induce DNA double-strand breaks, γH2AX detected using flow cytometer assay can be used to measure the DSBs of DNA in large samples.

Abstract:Background: Acrylamide (AA) is a water-soluble toxic chemical that is considered one of the most important food contaminants. Furthermore, AA is considered a major public health risk.Methods: This study was designed to evaluate the effects of AA on cytotoxicity, oxidative damage and genotoxicity in human lymphocytes and also to evaluate the protective effects of the chrysin (CH). Lymphocytes after isolation from the blood were treated with AA (50 µM), AA (50 µM) plus CH (10, 25, 50 µM) and CH (50 µM), and parameters such as cell viability, mitochondrial and lysosomal damage, as well as oxidative damage to DNA were examined.Results: The results showed that CH was able to reduce cytotoxicity, reactive oxygen species (ROS) levels, lipid peroxidation (LPO) level, collapse in mitochondrial membrane potential (MMP) and oxidative damage of DNA caused by AA in human lymphocytes. Also, co-treatment of the AA-exposed human lymphocytes with CH increases the glutathione (GSH) levels.Conclusion: Results suggest that CH (10, 25, 50 µM) shows a protective role in AA-induced cytotoxicity, oxidative stress, mitochondrial damage and DNA oxidative damage.

Abstract:The effect of Zr2+ irradiation on microstructure and corrosion in 360 °C lithiated water with 0.01 mol/L LiOH was investigated on a 90Nb-10Zr binary alloy prepared by referring to the β-Nb phase in zirconium alloys. The results revealed that the irradiation induced a ZrO2 film on the specimen surface and increased dislocations in matrix. After corrosion, oxide scales with a three-layer structure were formed regardless of irradiation. However, the scale formed on the irradiated surface was relatively thin presumably due to the ZrO2 film. A hypothesis for the corrosion mechanism of β-Nb phase in lithiated water upon irradiation was proposed.

Revisiting the effect of boiling on halogenated disinfection byproducts, total organic halogen, and cytotoxicity in simulated tap water

Abstract:Boiling is a widely adopted household tap water treatment method because of its ability to inactivate chlorine-resistant pathogenic bacteria, and to reduce certain groups of disinfection byproducts (DBPs). From a more comprehensive point of view, this study revisited the effect of boiling on four groups of 17 aliphatic DBPs and six groups of 44 aromatic DBPs in both simulated chlorinated and chloraminated tap water samples, with a special focus on the changes of total organic halogen (TOX) and cytotoxicity. Results showed that the concentrations of aliphatic DBPs substantially decreased during boiling via volatilization (trihalomethanes and chloral hydrate) and hydrolysis (haloacetamides) in chlorinated and chloraminated tap water samples. The concentrations of aromatic DBPs during boiling generally followed an increasing trend due to decarboxylation of polycarboxylic precursors in chlorinated tap water samples, and showed a first increasing and then decreasing trend in chloraminated tap water samples. A sharp decreasing of TOX occurred in the heating process of tap water samples from room temperature to 100 °C, and a relatively gentle decreasing was kept in the prolonged boiling process till 5 min. The most abundant DBP group in the tap water samples without boiling was trihalomethanes, and was replaced by haloacetic acids with boiling for 5 min. Continuous boiling for 5 min substantially reduced the cytotoxicity of chlorinated and chloraminated water samples by 52.6% and 21.3%, respectively. Reduction of cytotoxicity matched well with the reduction of TOCl (r = 0.907, P < 0.01), TOBr (r = 0.885, P < 0.01) and TOX (r = 0.905, P < 0.01), suggesting that the cytotoxicity reduction during boiling was mainly ascribed to the reduction of halogenated DBPs. Therefore, boiling of tap water to 100 °C was strongly recommended to reduce the potential health risks induced by tap water ingestion.
[Study on DNA Strand Breaks in Workers Exposed to Soluble Chromate].

Abstract:OBJECTIVE:To explore the biological effective markers, we investigated DNA strand breaks in peripheral lymphocytes from occupational population with broad ranges of soluble chromate exposure.METHODS:We conducted a cross-sectional study in the chromate exposed workers employed at a chromate factory in a district of Jinan, Shandong Province. The studied population contained 114 workers from different processes of the chromate plants, in addition, 30 farmers in the countryside about one hundred kilometers away from the factory, without exposure to chromate were matched with the exposed subjects by age, gender and smoking status being identified as a control group. Personal information on age, chromate exposure, medical history (including acute infection and medicine usage), smoking habit and alcohol consumption was obtained by questionnaire. DNA strand breaks in lymphocytes were detected by single-cell gel electrophoresis assay (comet assay) and the DNA damaged degree was evaluated by the score weighted by comet type. The air concentration of chromate was determined by individual sampling for 8 hours per day as shift work and chromium was assayed by atomic absorption spectrometry. The chromium content in the erythrocytes from peripheral blood was determined by graphite furnace atomic absorption spectrometry. The data were analyzed by SPSS10.0 software for statistical significance.RESULTS:(1) The results showed that the score for DNA strand breaks in lymphocytes were 54.52 +/- 23.51 in the exposed group, which was significantly higher than those in the control group (24.70 +/- 11.84) (P < 0.01). (2) The degree of DNA strand breaks in lymphocytes was increased in a dose-dependent manner ranging from 0 microg/m(3) to 106.00 microg/m(3). (3) Correlation analysis showed that there was a significant positive correlation between airborne chromate concentration and the degree of DNA strand break in lymphocytes (P < 0.01). (4) By multiple regression analysis, it was found that the airborne concentrations, chromium contents in red blood cells and smoking habits were factors which might affect the degree of DNA breaks.CONCLUSION:Our findings suggest that DNA strand break in lymphocytes should be an effective biomarker for occupational chromate-exposed population and be applied in biological monitoring and health risk assessment for occupational chromate-exposed population.

Abstract:Hydrogen energy is gaining attention as a non-polluting source. Aluminum’s reaction with water for propulsion and hydrogen production is widely used, but its nanoscale characteristics are unexplored. This study investigated aluminum hydrolysis in water vapor by ReaxFF-MD, examining effects of temperature and electric field. The oxidation of the aluminum surface is accompanied by aluminum dissolution, hydrogen generation and H3O+ formation, Formed hydrides and oxides were AlH3 and Al2O3. Hydrogen disappearance at 700 K and 900 K was due to conversion to AlH3. The diffusion of hydrogen and oxygen atoms in the Z direction was enhanced at increasing temperatures, Outer aluminum atoms preferentially reacted with oxygen to form Al2O3, while inner atoms reacted with hydrogen to form AlH3, the loosely packed outer oxide layer contains inclusions of AlH3. Studies at room temperature under different electric fields showed −Z field promoted the reaction while + Z field inhibited it, attributed to different electric field strengths’ effect on OH– ion electron-sharing Coulomb equilibrium, resulting in varying rates of hydroxide polar covalent bond cleavage. Our results clarify the atomic-scale effects of temperature and electric field on the aluminum-water reaction, providing guidance for optimizing experimental conditions and enhancing the efficiency of hydrogen production.

Impact of Cookware Types on Leaking Heavy Metals into Food based on pH Values, and their Potential Impact on the Health Status of a Saudi Sample using a Survey Study Design

Abstract:This study aimed to measure the leaked metal concentrations in cooked food using three types of cookware (stainless steel (18/10), cast iron, and aluminum). Two types of vegetables were used based on their pH values, tomatoes (pH 4.30-4.90) and zucchini (pH 9), as acidic and basic food materials. In addition, a survey study was performed on a randomly chosen Saudi sample to investigate the impact of their income, age, and awareness level in choosing between the different cookware available in the market, as well as to investigate the impact of the used cookware on their health status. The results showed that cooking acidic food (e.g., tomatoes) in aluminum pots can lead to an increase in the concentrations of leaked metals into the food such as leaking copper (0.27mg/kg), iron (3.048mg/kg), aluminum (0.91mg/kg) and magnesium (95.13 mg/kg) but no change in the concentrations of leaked metals into the food such as Chromium and Nickle (<0.4mg/kg). Results revealed that the only cookware that had the least leaked metals into the acidic food was cast iron. For the basic food (e.g., zucchini), results showed that zucchini turned into an acidic medium after cooking which increased the leaking of different metals into the cooked zucchini. The only cookware that had the least leaked metals into the zucchini was the stainless steel (18/10). The leaked metals into the food may affect the health; the results of this study’s survey showed a potential correlation between using cast iron and stainless steel with the incidence of diabetes, arthritis, and heart diseases. Results also showed that some individuals are aware of choosing between the different cookware available in the market based on the health impact. However, their income affected their choices significantly regardless of their awareness. In conclusion, it can be recommended based on the results of this study that cast iron is more preferable to be used for cooking acidic food, whereas aluminum is not preferable for cooking basic foods.
Effect of Sodium Benzoate Preservative on Micronucleus Induction, Chromosome Break, and Ala40Thr Superoxide Dismutase Gene Mutation in Lymphocytes

Abstract:Sodium benzoate is food preservative that inhibits microbial growth. The effects of sodium benzoate preservative on micronucleus induction, chromosome break, and Ala40Thr superoxide dismutase gene mutation in lymphocytes were studied. Sodium benzoate concentrations of 0.5, 1.0, 1.5, and 2.0 mg/mL were treated in lymphocyte cell line for 24 and 48 hrs, respectively. Micronucleus test, standard chromosome culture technique, PCR, and automated sequencing technique were done to detect micronucleus, chromosome break, and gene mutation. The results showed that, at 24- and 48-hour. incubation time, sodium benzoate concentrations of 1.0, 1.5, and 2.0 mg/mL increased micronucleus formation when comparing with the control group ( P < 0.05 ). At 24- and 48-hour. incubation time, sodium benzoate concentrations of 2.0 mg/mL increased chromosome break when comparing with the control group ( P < 0.05 ). Sodium benzoate did not cause Ala40Thr (GCG → ACG) in superoxide dismutase gene. Sodium benzoate had the mutagenic and cytotoxic toxicity in lymphocytes caused by micronucleus formation and chromosome break.

Abstract:Aluminum poisoning has been reported in some parts of the world. It is one of the global health problems that affect many organs. Aluminum is widely used daily by humans and industries. Residues of aluminum compounds can be found in drinking water, food, air, medicine, deodorants, cosmetics, packaging, many appliances and equipment, buildings, transportation industries, and aerospace engineering. Exposure to high levels of aluminum compounds leads to aluminum poisoning. Aluminum poisoning has complex and multidimensional effects, such as disruption or inhibition of enzymes activities, changing protein synthesis, nucleic acid function, and cell membrane permeability, preventing DNA repair, altering the stability of DNA organization, inhibition of the protein phosphatase 2A (PP2A) activity, increasing reactive oxygen species (ROS) production, inducing oxidative stress, decreasing activity of antioxidant enzymes, altering cellular iron homeostasis, and changing NF-kB, p53, and JNK pathway leading to apoptosis. Aluminum poisoning can affect blood content, musculoskeletal system, kidney, liver, and respiratory and nervous system, and the extent of poisoning can be diagnosed by assaying aluminum compounds in blood, urine, hair, nails, and sweat. Chelator agents such as deferoxamine (DFO) are used in the case of aluminum poisoning. Besides, combination therapies are recommended.

Toxicity of Metal Ions Released from a Fixed Orthodontic Appliance to Gastrointestinal Tract Cell Lines

Abstract:The mechanism of toxicity and cellular response to metal ions present in the environment is still a very current area of research. In this work, which is a continuation of the study of the toxicity of metal ions released by fixed orthodontic appliances, eluates of archwires, brackets, ligatures, and bands are used to test the prooxidant effect, cytotoxicity, and genotoxicity on cell lines of the gastrointestinal tract. Eluates obtained after three immersion periods (3, 7, and 14 days) and with known amounts and types of metal ions were used. Four cell lines—CAL 27 (human tongue), Hep-G2 (liver), AGS (stomach) and CaCo-2 (colon)—were treated with each type of eluate at four concentrations (0.1×, 0.5×, 1.0×, and 2.0×) for 24 h. Most eluates had toxic effects on CAL 27 cells over the entire concentration range regardless of exposure time, while CaCo-2 proved to be the most resistant. In AGS and Hep-G2 cells, all samples tested induced free radical formation, with the highest concentration (2×) causing a decrease in free radicals formed compared to the lowest concentrations. Eluates containing Cr, Mn, and Al showed a slight pro-oxidant effect on DNA (on plasmid φX-174 RF I) and slight genotoxicity (comet assay), but these effects are not so great that the human body could not "resist" them. Statistical analysis of data on chemical composition, cytotoxicity, ROS, genotoxicity, and prooxidative DNA damage shows the influence of metal ions present in some eluates on the toxicity obtained. Fe and Ni are responsible for the production of ROS, while Mn and Cr have a great influence on hydroxyl radicals, which cause single-strand breaks in supercoiled plasmid DNA in addition to the production of ROS. On the other hand, Fe, Cr, Mn, and Al are responsible for the cytotoxic effect of the studied eluates. The obtained results confirm that this type of research is useful and brings us closer to more accurate in vivo conditions.

Abstract:Background: High-dose radiation altering the genetic material in patients' bone marrow cells can lead to hematopoietic radiation syndrome. Accordingly, the presence of radiation protections agents is critical to preventing these adverse effects. Objective: This study aimed to evaluate the radioprotection of the exclusive or combination effect of resveratrol and crocin extracts at various concentrations on irradiated human lymphocytes. Material and methods: In this experimental study, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method was used to evaluate the cell viability in pre-treatment with resveratrol, crocin, or a combination of both, using a concentration range of 5 to 4800 μM / ml in 24 h. The chromosomal aberration test was employed to determine the aberration frequency in 48 h. This study was performed on human peripheral blood lymphocytes treated with 2 Gy radiation and reliability of measurements performed by the triplicate repeat. Results: MTT results showed that the groups treated with either resveratrol or crocin at concentrations of 5 to 4800 µM had no significant reduction in cell viability. The cytogenetic analysis of irradiated lymphocytes with 2 Gy X-rays revealed a reduction in the frequency of dicentric chromosomes in all treated groups in contrast with the control group. The most significant reduction occurred in those treated with a single agent at the concentration of 100 µM and a combined drug at the concentration of 50 µM. Conclusion: The combination of resveratrol and crocin is considered a potential radioprotector and prophylactic for patients before radiation therapy.
