Abstract:Introduction: Previous studies found that vitamin D receptor (VDR) TaqI, BsmI, FokI and ApaI gene polymorphisms are associated with several inflammatory diseases. However, the relationship between VDR gene polymorphisms and chronic spontaneous urticaria (CSU) is not clear. Aim: The purpose of our study was to explore the relationship between the polymorphism of VDR and the incidence of chronic spontaneous urticaria in the Chinese Han population. Meanwhile, the vitamin D levels in patients with chronic spontaneous urticaria were also detected and the effects of VDR gene polymorphism on vitamin D levels were detected. Material and methods: The genotypes of four VDR polymorphisms (TaqI, BsmI, ApaI, and FokI) were studied using allele-specific PCR analysis in 90 CSU patients and 90 healthy controls. Results: Compared to the control group, the mutant allele (C) of FokI were more common in patients with CSU (57.2% vs. 45%, p = 0.020, odds ratio (OR) = 0.612, 95% confidence interval (CI): 0.403-0.928). We found that serum vitamin D levels were significantly lower in CSU patients than in healthy controls (p = 0.023). However, the effect of VDR gene polymorphism on vitamin D levels was not found in patients of CSU. Conclusions: We first reported the effect of VDR gene FokI (rs2228570) polymorphism on the incidence of chronic spontaneous urticaria in the Chinese Han population.

Abstract:Although chronic spontaneous urticaria (CSU) is a common disease, genome-wide association studies (GWASs) of CSU are lacking. We aimed to identify susceptibility single-nucleotide polymorphism (SNPs) by performing to our knowledge previously unreported GWAS in Chinese Han adults with CSU. The discovery cohort included 430 CSU cases and 482 healthy controls. The GWAS findings were validated in 800 CSU cases and 900 healthy controls. Genetic, functional enrichment and bioinformatic analyses of genome-wide significant SNPs were performed to assess association between CSU and autoimmunity or atopy. Five genome-wide significant SNPs were identified: rs434124/LILRA3, rs61986182/IGHG1/2, rs73075571/TDGF1, rs9378141/HLA-G and rs3789612/PTPN22. The first 4 SNPs were in linkage disequilibrium with autoimmune-related diseases-associated SNPs and were cis expression quantitative trait loci in immune cells. The 5 SNPs-annotated genes were significantly enriched in immune processes. Higher polygenic risk scores, allele frequencies of rs3789612*T, rs9378141*C and rs73075571*G were significantly associated with autoimmune-related CSU phenotypes including positive anti-thyroglobulin IgG, positive anti-FcεRIα IgG, total IgE < 40 IU/mL and positive anti-thyroid peroxidase IgG, not with atopic or allergic sensitized CSU phenotypes. This GWAS of CSU identifies 5 risk loci and reveals that CSU shares genetic overlap with autoimmune diseases and that genetic factors predisposing to CSU mainly manifest through associations with autoimmune traits.

Abstract:Background: Ulcerative colitis (UC) is characterized by chronic inflammation of the colon and rectum as a result of an exaggerated T-cell response. Cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) is a T cell-restricted surface molecule induced with TCR or CD28 activation. There is evidence for genetic involvement of CTLA-4 in several autoimmune diseases, with the focus on the possible role of genetic variation of the CTLA-4 locus. The aim of this study was to investigate CTLA-4 gene polyrnorphisrns in patients with UC in a Chinese population with Han nationality.Methods: The C - 318T polymorphism in the promoter region and A+49G polymorphism in exon I of the CTLA-4 gene were studied by a polymerase chain reaction-sequence-specific primer method. We studied 82 unrelated patients with UC and 204 healthy controls in a Chinese population with Han nationality.Results: The frequency of the haplotype 2,3 (- 318C+49G/-318T+ 49A) was 26% in patients with UC and 41% in healthy controls (Fisher exact test P = 0.0147, odds ratio = 0.4918, 95% confidence interval: 0.2784 similar to 0.8688), but this significance disappeared when Bonferoni correction was applied. No other significant differences in the distribution of allele and genotype frequencies were observed between C-318T and A+49G gene polymorphisms and UC in the Chinese Han population.Conclusion: The C-318T and A+49G polymorphisms of the CTLA-4 gene were not associated with UC in Chinese Han patients.
Association of TLR4 gene polymorphisms with childhood Henoch–Schönlein purpura in a Chinese population

Abstract:Recent studies demonstrated that aberrant activation of Toll-like receptor (TLR) 4 was involved in the pathogenesis of Henoch–Schönlein purpura (HSP). In this study, we evaluated the association between TLR4 gene polymorphisms and the risk of childhood HSP in a Chinese population. A total of 175 HSP patients and 186 controls were recruited in this case–control study. Three single-nucleotide polymorphisms of the TLR4 gene (rs1927914, rs10759932 and rs1927907) were genotyped using the matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and Sequenom MassARRAY system. Our results revealed that a significantly reduced risk for HSP was associated with the G allele (OR = 0.71; p = 0.023) and G/G genotype (OR = 0.49; p = 0.021) of rs1927914. We also showed that rs1927914 variant decreased the risk of HSP in recessive inheritance model (OR = 0.55; p = 0.035, G/G vs A/A + A/G). In addition, we observed that a significantly decreased frequency of the haplotype GTC (rs1927914-rs10759932-rs1927907) in HSP patients compared with controls (OR = 0.56; p = 0.028). Our data suggested that TLR 4 rs1927914 polymorphism was associated with the decreased susceptibility to HSP in the Chinese children.
Association of CD14 Gene Polymorphism with Atopic Diseases in Chinese Han Ethnic Group Children

Abstract:OBJECTIVE:To investigate the distribution characteristics of the single nucleotide polymorphisms (SNPs) of the human CD14 gene in Chinese Han ethnic group children in Wenzhou, and their association with atopic diseases.METHODS:Totally 113 cases were recruited in atopic disease group who met the following criteria: 2 - 12 years old, clinically diagnosed as asthma or allergic rhinitis or atopic dermatitis, elevation of serum total IgE levels and serum specific IgE. Sixty-seven healthy children were enrolled in control group. The related regions of CD14 gene were sequenced to identify and characterize the SNPs, and plasma TIgE and SIgE were detected by immunoassay system and uniCAP system, respectively. The frequency of genotypes and alleles between two groups, as well as the levels of IgE in different genotypes, were compared.RESULTS:CD14/-159 SNP was present in Han ethnic group population of Wenzhou. The frequency of each genotype was 57.0% (TT), 28.0% (TC), 15.0% (CC) in normal children, and 46.9% (TT), 35.4% (TC), 17.7% (CC) in atopic children. No significant difference was found in the distribution of CD14/-159 polymorphism between atopic children and healthy control (chi(2) = 1.918, P > 0.05) according to Hardy-Weinberg principle statistics. There were no significant difference in frequency of each genotype between boys and girls. No significant difference was found in the total plasma IgE levels among groups of TT genotypes [(2520 +/- 460) IU/L], TC genotypes [(2400 +/- 460) IU/L] and CC genotype [(2500 +/- 460) IU/L] (F = 0.807, P > 0.05).CONCLUSION:CD14/-159 SNP is present in Han ethnic group children in Wenzhou, and other SNP in CD14 gene was not found. TT genotype was the primary genotype in CD14/-159 SNP in the children studied. No relationship between CD14/-159 SNP and atopic disease or serum total IgE level was found.
Genetic Susceptibility to Ulcerative Colitis in the Chinese Han Ethnic Population: Association with Tnf Polymorphisms

Abstract:Background Tumor necrosis factor alpha (TNF alpha) is an important proinflammatory cytokine that has been implicated in the pathogenesis of inflammatory bowel disease (IBD). Recent studies have evaluated the role of TNF promoter polymorphisms in IBD, whereas the data are inconsistent. Trans-racial mapping in an ethnically distinct but homogenous population may help clarify these associations. We investigate the association between TNF promoter polymorphisms and susceptibility to ulcerative colitis (UC) in the Chinese Han ethnic population.Methods We studied 110 unrelated UC patients and 292 healthy controls from Zhejiang Province, China. Genotyping for 6 common TNF promoter polymorphisms (TNF -1031T/C, -863C/A, -857C/T, -380G/A, -308G/A, -238G/A) was carried out by polymerase chain reaction sequence-specific primers (PCR-SSP).Results TNF-308A was associated with disease (allele frequency patients 14.6% vs controls 8.9%, P=0.02). TNF -857T was increased in patients but without statistical significance (allele frequency 17.3% vs 12.2%, P=0.06). Haplotype analysis revealed 6 haplotypes including two (H5 and H3), which contained TNF -308A. H5 was associated with disease (haplotype frequency patients -12.3% vs controls 7.5%, P=0.03). Of note the rare haplotype H3 has not previously been identified in Caucasian populations. Homozygosity for the haplotype H4 comprising the common alleles at each TNF promoter single-nucleotide polymorphism (SNP) was negatively associated with disease (patients vs controls 24.5% vs 34.9%, P < 0.05).Conclusions We report the association with TNF -308A polymorphisms in Chinese patients with ulcerative colitis. The functional study in Chinese Han ethnic population is now required.
Validation of Susceptibility Loci for Vitiligo Identified by GWAS in the Chinese Han Population

Abstract:Forty-nine susceptible loci have been reported to be significantly associated with vitiligo by genome-wide association studies (GWASs) in European-derived whites. To date, some of these reported susceptibility loci have not yet been validated in the Chinese Han population. The purpose of this study was to examine whether the 16 reported susceptible loci in European-derived whites were associated with vitiligo in the Chinese Han population. Imputation was performed using our previous GWAS dataset by IMPUTE v2.2.2. The 16 imputed top single-nucleotide polymorphisms (SNPs) with suggestive signals, together with the reported SNPs, were genotyped in a total of 2581 patients and 2579 controls by the Sequenom MassARRAY system. PLINK 2.0 software was used to perform association analysis. The dbSNP database, HaploReg, and eQTL data were adopted to annotate the biological function of the SNPs. Finally, four SNPs from three loci were significantly associated with vitiligo, including rs3747517 (P = 1.29 × 10–3, OR = 0.87) in 2q24.2, rs4807000 (P = 7.78 × 10–24, OR = 0.66) and rs6510827 (P = 3.65 × 10–5, OR = 1.19) in 19p13.3, and rs4822024 (P = 6.37 × 10–10, OR = 0.67) in 22q13.2. According to the dbSNP database, rs3747517 is a missense variant of IFIH1, rs4807000 and rs6510827 are located in TICAM1, and rs4822024 is located 6 kb upstream of TEF. Further bioinformatics analysis by HaploReg and eQTL found that rs4807000, rs6510827, and rs4822024 are involved in regulating gene expression. Our study revealed the strong association of 2q24.2 (rs3747517), 19p13.3 (rs4807000, rs6510827), and 22q13.2 (rs4822024) with the risk of vitiligo in the Chinese Han population, which implicates common factors for vitiligo across different ethnicities, and helps expand the understanding of the genetic basis of this disease.
Lack of Association Between Interleukin-8 Gene +781 C/T Polymorphism and Henoch-Schönlein Purpura in Childhood.

Abstract:Henoch-Schönlein purpura (HSP), a common allergic hemorrhagic disease, occurs frequently in children affecting kidney, joint and skin. While interleukin-8 (IL-8) plays an important role in inflammation, the association between IL-8 gene +781 C/T polymorphism and HSP remains unclear. Interleukin-8, an important chemokine related to the initiation and amplification of acute inflammatory responses, has been reported to be involved in the pathogenesis of some autoimmune and inflammatory diseases. In this study, we aimed to investigate whether IL-8 gene +781 C/T (rs2227306) polymorphism has an influence on susceptibility and clinical manifestations of patients to HSP. This hospital-based case-control study comprised 192 patients with HSP and 202 healthy controls. The genotypes of IL-8 gene +781 C/T polymorphism were identified using PCR-TaqMan method. All genotype frequencies of both groups (patients and controls) conformed to the Hardy-Weinberg equilibrium. No significant differences in allele or genotype frequencies of IL-8 gene +781 C/T polymorphism were observed between patients with HSP and controls (p=0.98, χ2=0.000 and p=0.49, χ2=1.432, respectively). When patients were stratified for the presence of joint, gastrointestinal and renal manifestations, genotype frequencies of IL-8 gene polymorphism were found no statistically significant differences (p>0.05). Our findings do not support that IL-8 gene +781 C/T polymorphism has an effect on the susceptibility to HSP in Chinese children.
[Interactions Between Susceptible Genes and Risk Environmental Factors in Chinese Females with Systemic Lupus Erythematosus].

Abstract:OBJECTIVETo explore the interactions of gene polymorphisms of cytotoxic T lymphocyte antigen 4 (CTLA-4) and programmed cell death 1 (PDCD-1) with risk environmental factors in individuals with systemic lupus erythematosus (SLE) from Han nationality female population in South of Changjiang River region of China.METHODSWith a case-only design, a total of 258 cases were enrolled in this study, and single nucleotide polymorphisms (SNPs) of the PDCD-1 and CTLA-4 genes were determined by means of PCR-RFLP. With the aid of Poisson loglinear mode, interactions between gene-gene and gene-environment were fitted under the dominant, recessive, additive and multiple models, respectively.RESULTSIt was found that interaction existed between GG genotype of PD1.6 and UV history under separate inherent models of the recessive mode (OR = 3.714, 95%CI: 1.235 - 11.179) and additive mode (OR = 3.199, 95%CI: 1.023 - 10.004). For CTLA-4 locus, there existed interactions between TT/TC genotype of -1722T-->C and UV history under the dominant model (OR = 4.874, 95%CI: 1.119 - 21.242), and interaction between T allele and UV history was also found under the multiple model (OR = 1.470, 95%CI: 1.047 - 2.065). While, under the additive mode for CTLA-4, it was found that interactions existed between TT genotype of -1722T-->C and UV history (OR = 4.744, 95%CI: 1.037 - 21.737), as well as between TC genotype of -1722T-->C and UV history (OR = 4.973, 95%CI: 1.110 - 22.287).CONCLUSIONThe interactions between UV history and polymorphisms of CTLA-4 and PDCD-1 gene for SLE were observed, which indicates that there may be association of their interactions with the development of SLE in Han nationality females population in the south regions of Changjiang River in China.
Analysis of Association of Cytotoxic T Lymphocyte-Associated Antigen-4 Gene Promoter Polymorphisms with the Clinical Feathers of Systemic Lupus Erythematosus in Han Nationality Population in South of the Changjiang River

Abstract:Objective To explore the association between polymorphisms within cytotoxic T lymphocyte-associated antigen-4(CTLA-4) gene promoter and systemic lupus erythematosus(SLE) phenotypes in Han nationality population in South of the Changjiang River. Methods One hundred and twenty female cases were enrolled with the aim of case only design,and the genotype was determined by PCR-RFLP.Then,we calculated χ2 and ORs for related study. Results Analysis of association between the 2 SNPs and organ damage indicated no significant relationship(P0.05).The-1722T/T genotype frequency in patients with positive anti-SSA was higher than that of patients with negative anti-SSA(P=0.020,OR=5.26,95%CI:1.30-21.32).The-318C/T and T/T combined group in patients with positive anti-RNP antibody had lower frequency than that of patients with negative anti-RNP antibody(P=0.016,OR=0.16,95%CI:0.04-0.71).Meanwhile,there was a linkage disequilibrium between the-1722TC and-318CT,so we analyzed the association between haplotypes and clinical feathers.In patients with positive anti-SSA,there was a relative high proportion in T-C haplotype(P=0.014,OR=1.90,95%CI:1.14-3.18),and a less one in C-C haplotype(P=0.028,OR=0.55,95%CI:0.32-0.94).Patients with positive anti-RNP antibody was found to have a lower frequency of T-T haplotype compared to patients with negative anti-RNP antibody(P=0.029,OR=0.20,95%CI:0.05-0.85). Conclusions In Han nationality population in South of the Changjiang River,-1722TC allele may play an important role in the production of anti-SSA,and-318CT allele may play an important role in the production of anti-RNP antibody.
Association Of Ctla4 Gene Polymorphisms With Susceptibility And Pathology Correlation To Pulmonary Tuberculosis In Southern Han Chinese

Abstract:The cytotoxic T lymphocyte antigen-4 (CTLA4) gene is a key negative regulator of the T lymphocyte immune response. It has been found that CTLA4 + 49A>G (rs231775), + 6230G>A (rs3087243), and 11430G>A (rs11571319) polymorphisms are associated with susceptibility to many autoimmune diseases, and can down-regulate the inhibition of cellular immune response of CTLA4. Three SNPs in CTLA4 were genotyped by using the PCR and DNA sequencing methods in order to reveal the susceptibility and pathology correlation to pulmonary tuberculosis in Southern Han Chinese. We found that the frequency of CTLA4 + 49AG genotype in the pulmonary tuberculosis patients (38.42%) was significantly lower than that of the healthy controls (49.77%), (P-cor=0.038, OR 0.653, 95% CI 0.436-0.978). But, no associations were found between the other 2 SNPs (+ 6230G>A, 11430G>A) and tuberculosis (P>0.05). Haplotype analysis showed that the frequency of haplotype AGG in the healthy controls group (6.9%) was significantly higher than the pulmonary tuberculosis patients group (1.4%), (global P=0.005, P-cor=0.0002, OR 0.183, 95% CI 0.072-0.468). In addition, haplotype GGA was found to be significantly related to tuberculosis with double lung lesion rather than single lung lesion (P-cor=0.042). This study is the first to report that genetic variants in the CTLA4 gene can be associated with pulmonary tuberculosis in Southern Han Chinese, and CTLA4 + 49AG genotype as well as haplotype AGG may reduce the risk of being infected with pulmonary tuberculosis. The GGA haplotype was related to tuberculosis with double lung lesion, which provides a new experimental basis to clarify the pathogenesis of pulmonary tuberculosis.

Abstract:Interleukin-10 (IL-10), one of the anti-inflammatory cytokines, plays a major role in the pathogenesis of Henoch-Schönlein purpura (HSP). In present study, we investigated the association between genetic variation in IL-10 gene and susceptibility to HSP in a Chinese childhood population. Considering the overlapping clinical manifestations during the course of disease, the relation between IL-10 gene polymorphisms and HSP clinical heterogeneity was also assessed. We analyzed three IL-10 tag single nucleotide polymorphisms (SNPs; rs3021094, rs3790622, and rs1800872) using the Sequenom MassARRAY system by means of matrix-assisted laser desorption ionization-time of flight mass spectrometry method in 182 patients with HSP and 202 healthy controls. For the frequency of alleles, genotypes, and haplotypes of IL-10 SNPs, no significant differences were observed between HSP patients and controls. In addition, we did not find any association of IL-10 gene polymorphisms with the clinical manifestations of HSP. Our results suggest that genetic variation in IL-10 gene is unlikely to confer susceptibility to HSP in Chinese children.
Cytotoxic T Lymphocyte-Associated Antigen-4 Gene Promoter Polymorphisms in Patients with Systemic Lupus Erythematosus in Southern Chinese Population

Abstract:Purpose To assess the association between polymorphisms within Cytotoxic T lymphocyte-associated antigen-4 (CTLA-4) gene promoter and the morbidity of systemic lupus erythematosus(SLE) in Chinese people. Methods A case-control led study was performed for CTLA-4 polymorphisms, -1722 T〉C and -318 C〉T in the promoter region in 97 patients with SLE and 200 healthy individuals. We examined the CTLA-4 genotype of these subjects by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results There was a significant difference in -1722 T〉C genotype frequencies between the two groups (P=0.002). Carrier frequencies of the -1722 T allele in the promoter were significantly increased in SLE patients compared with those in controls (odds ratio=1.94, 95% confidence interval:1.34-2.80, P=0.000). There was no difference in -318 C〉T genetype and allele frequencies between the two groups. However, there was a linkage disequilibrium between the -1722 T〉C and -318 C〉T. There was a significant difference in CTLA-4 haplotype distribution between the two groups (χ2=12.64,P=0.005), which the relative high proportion was T-C haplotype (P=0.001) and the less one was C-C haplotype (P=0.002) in SLE group, compared to control group. Conclusions The -1722 T〉C in the promoter of CTLA-4 was determined to be susceptible to SLE in southern Chinese population. Although the result showed that -318 C〉T was not responsible for SLE, the T-C haplotype might be a susceptible haplotype and C-C haplotype a protective one in southern Chinese population.

Abstract:Background: Several studies have stated that TNF-α participates in the pathogenesis of scleritis, but also in several systemic autoimmune diseases and vasculitis, of which some are associated with scleritis. Earlier GWAS and SNP studies have confirmed that multiple SNPs of TNF related genes are associated with many immune-mediated disorders. The purpose of this study was to examine the association of TNF related gene polymorphisms with scleritis in Chinese Han. A case-control study was carried out in 556 non-infectious scleritis cases and 742 normal controls. A total of 28 single-nucleotide polymorphisms (SNPs) were genotyped by the iPLEXGold genotyping assay. Results: No significant correlations were seen between the individual SNPs in the TNF related genes and scleritis. Haplotype analysis showed a significantly decreased frequency of a TNFAIP3 TGT haplotype (order of SNPs: rs9494885, rs3799491, rs2230926) (Pc = 0.021, OR = 0.717, 95% CI = 0.563-0.913) and a significantly increased frequency of a TNFSF4 GT haplotype (order of SNPs: rs3850641, rs704840) (Pc = 0.004, OR = 1.691, 95% CI = 1.205-2.372) and TNFSF15 CCC haplotype (order of SNPs: rs6478106, rs3810936, rs7865494) (Pc = 0.012, OR = 1.662, 95% CI = 1.168-2.363) in patients with scleritis as compared with healthy volunteers. Conclusions: This study reveals that a TGT haplotype in TNFAIP3 may be a protective factor for the development of scleritis and that a GT haplotype in TNFSF4 and a CCC haplotype in TNFSF15 may be risk factors for scleritis in Chinese Han.
Ctla4 Polymorphisms Are Associated With Severe Pulmonary Tuberculosis In A Chinese Han Population

Abstract:The cytotoxic T lymphocyte antigen‐4 (CTLA4) is a key negative regulator in T lymphocyte activation. Polymorphisms on CTLA4 are associated with susceptibility to many autoimmune diseases, resulting in down‐regulation of the inhibition of cellular immune response. The present study aimed to investigate potential associations of CTLA4 +49A>;G (rs231775), +6230G>;A (rs3087243), and 11430G>;A (rs11571319) polymorphisms with pulmonary tuberculosis (PTB) in a Chinese Han population. A case‐control study was performed, and the three SNPs were genotyped by PCR and DNA sequencing methods. We found the frequency of CTLA4 +49AG genotype in 274 PTB cases was significantly lower than that of the 265 controls ( P cor =0.025, 0R 0.613, 95% CI 0.401–0.937). However, no associations were found between the other 2 SNPs ( P >;0.05). After divided PTB cases into two groups: with or without cavities, we found a novel relation of +6230AG genotype to cavities ( P cor =0.046, 0R 2.677, 95% CI 1.029–6.965). Lately haplotype analysis showed that haplotype AGG was significantly associated with healthy controls (P=0.002, OR 0.286, 95% CI 0.124–0.657), while haplotype AAG was related to PTB with cavities ( P =0.047, OR 2.053, 95% CI 1.000–4.215). Our results suggest the CTLA4 polymorphisms can be associated with PTB in a Chinese Han population, and +49AG may reduce the risk of being PTB, while +6230AG may contribute to cavities formation. Abstract:OBJECTIVE:To investigate the relationship of gene polymorphisms of programmed cell death 1 gene (PDCD1) and ultraviolet history with systemic lupus erythematosus (SLE) among the Han population in the southern region of yangtze river in China. METHODS:With a case control design, a total of 159 SLE cases and 159 controls were enrolled in this study, and single nucleotide polymorphisms (SNPs) of the PDCD1 gene were determined by PCR-restriction fragment length polymorphism (RFLP). With the aid of the logistic regression model, the effect of gene polymorphism, environmental factor and the interaction between gene and environment were fitted under the recessive, dominant, additive and codominant mode, respectively. RESULTS:Three models were screened as the optimal models under the additive mode and one model under the dominant mode, according to the lowest value of Akaike's Information Criteria (AIC). After the control of age and gender, it was found that the frequency of ultraviolet exposure was higher in cases than in controls with significant difference under all models (P<0.05). For the haplotypes composed of the alleles of PD1.2, PD1.5 and PD1.6, there was significantly higher frequency of G-T-A haplotype (0.1196 vs 0.0363) and lower frequency of A-C-A haplotype (0.4746 vs 0.5399) in cases than that in controls (P<0.05) under the additive mode, and the G-T-A haplotype was associated with an increased risk for SLE (OR=4.319), while A-C-A haplotype was shown as a protective factor for SLE (OR=0.571). Moreover, interaction between A-C-G haplotype and ultraviolet exposure, which was related to an increased risk for SLE (beta5=1.182, Z=2.2898, P<0.05, OR=3.261), was also found under this mode. Additionally, the frequency of G-C-G haplotype was higher in cases than that in controls (0.1287 vs 0.0361) under the dominant mode with statistically significant difference (P<0.05, OR=4.332). CONCLUSION:Authors' results indicate that ultraviolet exposure, G-T-A or G-C-G haplotype and interaction between A-C-G and ultraviolet exposure may be associated with genetic susceptibility to SLE in Han population in the southern region of yangtze river in China under certain genetic modes.

Abstract:Abstract The majority of patients who experience cutaneous adverse drug reactions (cADRs) concurrently receive multiple medications, meaning that the causative drug remains unidentified. We explored the association between human leukocyte antigen (HLA) alleles and cADRs, regardless of the allergenic drug, to investigate whether different drug-induced cADRs were associated with the same or similar risk alleles in a Han Chinese population. We genotyped a sample of 146 cADR patients and 230 population controls from the same hospital and systematically analyzed the association between HLA Class I genes and cADRs. The carrier frequency of HLA-B∗46:01 in cADR patients was found to be significantly higher than that in population controls (P = .0021, odds ratio [OR] = 2.18, 95% confidence interval [CI]: 1.33–2.58). Subgroup analysis showed that HLA-B∗46:01 was significantly associated with urticaria and erythema multiforme (P = .0077, OR = 2.53, 95% CI: 1.30–4.91; and P = .0049, OR = 2.77, 95% CI: 1.39–5.50, respectively). Furthermore, a significant association was also detected between HLA-A∗02:01 and erythema multiforme (P = .0038, OR = 2.65, 95% CI: 1.31–5.33). This study is the first to demonstrate that HLA-B∗46:01 is a risk allele for cADRs in a Han Chinese population, indicating that screening for HLA-B∗46:01 prior to the administration of medication may predict the risk of developing cADRs.

Abstract:Immunoglobulin (Ig) E and IgG anti-thyroid autoantibodies (AAbs) play important roles in the immunopathogenesis of chronic spontaneous urticaria (CSU). To date, association of IgE and IgG AAbs with Chinese CSU patients has not been fully investigated. We aimed to explore prevalence rates of IgE and IgG AAbs in Chinese CSU patients and their association with clinical and laboratory parameters. Serum IgE and IgG AAbs against thyroid peroxidase (TPO) and thyroglobulin (TG), total IgE (tIgE) and specific IgEs were measured using enzyme-linked immunosorbent assay, chemiluminescence microparticle immunoassay and immunoblotting. Meta-analyses and literature review were conducted. The meta-analyses indicated that CSU cases were 4.98, 6.90 and 6.68 times more likely to have positive anti-TPO IgE, anti-TPO IgG and anti-TG IgG (all P < 0.001) compared with controls, respectively, and revealed a positive correlation between the prevalence rates of anti-TPO IgE and anti-TPO IgG (r = 0.53, P = 0.025). A total of 1,100 Chinese Han adult CSU patients and 1,100 ethnicity-, age- and sex-matched healthy controls were recruited from 15 centers. Prevalence rates of anti-TPO IgE, anti-TPO IgG, anti-TG IgE or anti-TG IgG in the patients were all significantly higher than those in the controls. Significant correlations were observed between prevalence rates of anti-TPO IgE and anti-TPO IgG (r = 0.297, P < 0.001) as well as between those of anti-TG IgE and anti-TG IgG in the patients (r = 0.137, P < 0.001). Patients with anti-TPO IgE or anti-TPO IgG had significantly lower tIgE levels (P < 0.001). Positive anti-TPO IgE, positive anti-TPO IgG and tIgE < 40 IU/mL were independent predictors of antihistamine-refractory cases. In conclusion, the prevalence rates of IgE and IgG AAbs in Chinese CSU patients are significantly elevated and reciprocally correlated. This study verifies the results of previous case-control studies of CSU patients from other populations and ethnicities.

Abstract:Objectives: Interleukin-33 (IL-33) was one of the members of IL-1 family, and it was reported that the single nucleotide polymorphisms (SNPs) in the IL-33 gene was contribute to the susceptibility to immune related diseases, including rheumatoid arthritis, asthma, Behcet's disease, systemic lupus erythematosus and ankylosing spondylitis. However, the potential association between SNPs in IL-33 gene and Henoch-Schonlein purpura (HSP) was not explored.Methods: We designed a case-control study, a total of 181 HSP patients and 202 healthy pediatric controls were included, which aimed to study the relationship between the SNPs in the IL-33 gene (rs1929992, rs7044343) and HSP with a Chinese pediatric cohort.Results: There were no evidence for the association of rs1929992 and rs7044343 polymorphism in IL-33 gene with pediatric HSP patients (C versus T, P = 0.622, OR= 0.931, 95% CI: 0.7 -1.238; TT versus CC, P = 0.742, OR= 0.908, 95% CI: 0.513 -1.609; CT versus CC, P = 0.158, OR= 0.714, 95% CI: 0.446 1.141 and Cversus T,P= 0.920, OR= 0.986, 95% CI: 0.742 -1.309; TT versus CC,P= 0.939, OR= 0.978, 95% CI: 0.549 -1.741; CT versus CC,P = 0.416, OR= 0.817, 95% CI: 0.501-1.331, respectively). Moreover, association studies were performed on the correlation between IL-33 gene polymorphisms and HSP nephritis patients as well as patients with gastrointestinal manifestation and joint involvement. However, there were no significant association regarding the distribution of allele and genotype frequencies between HSP patients with different system involvement and healthy controls either.Conclusions: The present findings indicated that both of rs1929992 and rs7044343 in gene IL-33 polymorphism were not related to the susceptibility to HSP and the different system involvement of HSP patients.
