Abstract:We investigated whether TESPA1 gene polymorphisms were associated with increased risk of developing ankylosing spondylitis (AS). We also studied whether TESPA1 gene interacts with environmental factors. A total of 494 patients with AS and 478 matched healthy controls were genotyped for four SNPs (rs1801876, rs2171497, rs4758994, and rs997173) in the TESPA1 gene. We found no evidence of association between these SNPs and AS susceptibility, and between their haplotypes and the disease. But, patients with rs1801876 GA, GG, and AA genotypes had significantly different Bath Ankylosing Spondylitis Functional Index (BASFI) scores ( p = 0.023). There were significantly different visual analogue scale (VAS) night pain assessment scores ( p = 0.040) and BASFI scores ( p = 0.023) among different genotypes at rs2171497 locus. There were also significantly different chest expansion scores ( p = 0.042) and Bath Ankylosing Spondylitis Disease Activity Index (BASDAI) scores ( p = 0.014) among different genotypes at rs997173 locus. For multiple testing, Bonferroni correction was performed. After Bonferroni correction, clinical characteristics of these three loci showed association between different genotype groups. These findings indicated that the TESPA1 gene is not involved in AS genetic predisposition in the Han Chinese population; however, it may play an important role in the clinical characteristics of AS.
Genetic Susceptibility to Ulcerative Colitis in the Chinese Han Ethnic Population: Association with Tnf Polymorphisms

Abstract:Background Tumor necrosis factor alpha (TNF alpha) is an important proinflammatory cytokine that has been implicated in the pathogenesis of inflammatory bowel disease (IBD). Recent studies have evaluated the role of TNF promoter polymorphisms in IBD, whereas the data are inconsistent. Trans-racial mapping in an ethnically distinct but homogenous population may help clarify these associations. We investigate the association between TNF promoter polymorphisms and susceptibility to ulcerative colitis (UC) in the Chinese Han ethnic population.Methods We studied 110 unrelated UC patients and 292 healthy controls from Zhejiang Province, China. Genotyping for 6 common TNF promoter polymorphisms (TNF -1031T/C, -863C/A, -857C/T, -380G/A, -308G/A, -238G/A) was carried out by polymerase chain reaction sequence-specific primers (PCR-SSP).Results TNF-308A was associated with disease (allele frequency patients 14.6% vs controls 8.9%, P=0.02). TNF -857T was increased in patients but without statistical significance (allele frequency 17.3% vs 12.2%, P=0.06). Haplotype analysis revealed 6 haplotypes including two (H5 and H3), which contained TNF -308A. H5 was associated with disease (haplotype frequency patients -12.3% vs controls 7.5%, P=0.03). Of note the rare haplotype H3 has not previously been identified in Caucasian populations. Homozygosity for the haplotype H4 comprising the common alleles at each TNF promoter single-nucleotide polymorphism (SNP) was negatively associated with disease (patients vs controls 24.5% vs 34.9%, P < 0.05).Conclusions We report the association with TNF -308A polymorphisms in Chinese patients with ulcerative colitis. The functional study in Chinese Han ethnic population is now required.

Abstract:Objective To investigate the association between the single nucleotide polymorphism( SNP) of Tumor necrosis factor superfamily member 15( TNFSF15) and Crohn's disease( CD) of han nationality in the place of Zhejiang. Methods Extract the peripheral venous blood of 42 CD patients and 49healthy persons followed by extracting DNA. Design specific primers of target gene segments of the three polymorphism sites rs3810936,rs6478109,rs7848647,which are located in TNFSF15. Amplify target gene segments of DNA specimens using PCR followed by sequencing them. At last,analyze the relationship among allele polymorphism and susceptibility and subtype of disease. Results The TNFSF15 SNP( rs3810936,rs6478109,rs7848647) showed no significant in allele frequency between the control and CD groups. In CD group and control group, both haplotype B1( cc-gg-cc) carring the three risk alleles of rs3810936,rs6478109,rs7848647( 38. 1% and 26. 5%,χ2= 1. 393,P = 0. 238) and haplotype A1( tt-aa-tt) carring the opposite alleles of three SNPs( 14. 3% and 12. 2%,χ2= 0. 082,P = 0. 774) showed no significant association with CD. According to multiple regression analysis,these three polymorphisms of TNFSF15 have no significant association with clinical subtypes involved of CD in China,but there is relationship between rs3810936and rs6478109( r = 0. 802,P < 0. 01),rs3810936 and rs7848647( r = 0. 793,P < 0. 01),rs6478109 and rs7848647( r = 0. 948,P < 0. 01). Conclusions SNP( rs3810936,rs6478109,rs7848647) of TNFSF 15showed no significant association with CD in Chinese Han population,and have no significant association with clinical subtypes,it may have racial and regional difference.

Abstract:Objective: Genome-wide association studies of rheumatoid arthritis (RA) in Caucasian populations have identified polymorphisms in and near the tumor necrosis factor α (TNF-α)-induced protein 3 gene (TNFAIP3).This study aimed to investigate the association between the critical polymorphisms in the TNFAIP3 gene and the risk of RA in the Chinese population.Material and methods: A hospital-based case-control study of 344 RA patients and 344 matched controls was conducted.After informed consent was obtained, each person donated 2 ml of blood for biomarker testing.Three polymorphisms were determined by the TaqMan SNP genotyping method.The statistical analyses were performed primarily with SPSS (v13.0).Results: This study showed that individuals with the rs2230926 TG genotype had an increased risk of RA compared with those carrying the TT genotype (OR = 1.74, 95% CI: 1.12-2.72,p = 0.015) and the combined TG/GG genotypes were also associated with an increased risk of RA (OR = 1.75, 95% CI: 1.13-2.72,p = 0.012).Individuals carrying the rs10499194 CT heterozygous genotype had a 1.88-fold increased risk of RA compared with those with the homozygous wild genotype CC (95% CI: 1.05-3.36,p = 0.034).Subset analysis stratified to gender showed that significant associations were present in the female population, but not in the male population.Haplotype analysis showed that the T-A-C and G-G-C haplotypes were associated with increased risks of RA (ORs were 2. 46 and 2.27, p values were 0.002 and 0.002, respectively).Conclusions: The rs10499194 and rs2230926 polymorphisms in the TNFAIP3 gene region may be genetic risk modifiers for RA in the Chinese population.

Abstract:Tumour necrosis factor-α (TNF-α), an important proinflammatory cytokine, exerts a variety of physiological and pathogenic effects that lead to tissue destruction. Studies on the association of TNF-α genetic polymorphisms with systemic lupus erythematosus (SLE) have yielded inconclusive results. We investigated the association of TNF-α genetic polymorphisms (−1031T/C, −863C/A, −857T/C, −308A/G and +489A/G) with SLE in Taiwanese patients and controls. Our results indicate that 1) the frequency of the A-allele at −863 position was significantly higher in SLE patients (odds ratio = 1.46; 95% CI = 1.02–2.08); 2) the frequency of the A-allele at +489 position was significantly higher in SLE patients (odds ratio = 1.79; 95% CI = 1.21–2.65); 3) the AA or GA genotype frequencies at +489 position were significantly increased in SLE patients (AA genotype: odds ratio = 11.20; 95% CI = 1.36–92.55; GA genotype: odds ratio = 1.63; 95% CI = 1.03–2.58); 4) no significant association of TNF-α haplotypic distributions was observed, except for the haplotypes TCCGA, CACGA and CCCGG; and 5) the genotype frequency of the polymorphisms at −1031 was significantly different in patients with antinuclear antibodies ( P = 0.022). The allele and genotype frequencies of the polymorphisms at −863 were not significantly different. The genotype frequency of the polymorphisms at −857 was significantly different in patients with haematological disorder ( P = 0.025). The frequency of A allele of the polymorphisms at −308 was significantly increased in patients with malar rash ( P = 0.033), discoid rash ( P = 0.023), photosensitivity ( P = 0.037), oral ulcers ( P = 0.002) and serositis ( P = 0.029). The genotype frequency of the polymorphisms at +489 was significantly different in patients with discoid rash and photosensitivity (data not shown; discoid rash, P = 0.031; photosensitivity, P = 0.044). These results suggest that TNF-α genetic polymorphisms contribute to SLE susceptibility in the Taiwanese population.

Abstract:Previous studies have shown associations of polymorphisms in the tumor necrosis factor (TNF) receptor super family member 1A ( TNFRSF1A ) gene with several groups of inflammatory and autoimmune related diseases, but associations of TNFRSF1A polymorphisms with autoimmune thyroid diseases (AITD), mainly including two sub-types of Hashimoto's thyroiditis (HT) and Graves' disease (GD), in the Chinese Han population is unclear. A case-control study of 1812 subjects (965 AITD patients and 847 unrelated healthy controls) was conducted to assess AITD associations with five single nucleotide polymorphisms (SNPs), including rs4149576, rs4149577, rs4149570, rs1800693, and rs767455 in the TNFRSF1A gene locus. Genotyping was performed and evaluated using the platform of ligase detection reaction. No significant difference was observed in the allele and genotype frequencies between HT or GD patients and controls in any of the five SNPs in the TNFRSF1A gene (all p values >0.05). However, a moderate association of rs4149570 with HT was found after adjusting for age and gender [odds ratio (OR)=1.40, p=0.03]. No obvious difference was found in the haplotype distribution of any of the five SNPs in the TNFRSF1A gene between the AITD patients and controls. These data suggest that these five SNPs in the TNFRSF1A gene are not associated with AITD in the Chinese Han population, but rs4149570 shows a weak association with HT after adjusting for gender and age.

Abstract:Systemic sclerosis (SSc) is a complex disease involving multiple genetic factors. An association of the IRF5 polymorphism with SSc was reported in Caucasian populations of Europe and North America, as well as in Japanese populations. The present study aimed to examine whether the SSc-associated SNP rs2004640 of IRF5 gene confer susceptibility to SSc and clinical features of SSc in a Han Chinese population. A Han Chinese cohort consisting of 424 SSc patients and 502 healthy controls were examined in the study. TaqMan assays were carried out to examine the SNP. Exact p-values were obtained (Fisher's test) from 2x2 tables of allele counts and disease status. SSc patients of Han Chinese showed increased homozygous TT genotype of the rs2004640 (p = 0.027, odds ratio (OR) = 1.4, CI =1.03-1.93), which was significantly associated with pulmonary fibrosis of SSc and ATA-positive SSc of Han Chinese. The lcSSc and ACA-positive SSc of Han Chinese appeared also in association with the increased T allele frequency. However, the Chinese dcSSc did not show any association with the rs2004640. The results were consistent with previous reports in other ethnic populations in supporting the notion that polymorphisms of IRF5 may play an important role in susceptibility to SSc.

Abstract:There are reports of IL-1 complex gene polymorphisms in ankylosing spondylitis (AS; MIM 106300), but the results have been inconsistent among populations. Moreover, few studies examine the association between IL-1 complex gene polymorphisms and clinical symptoms of AS patients. We investigated polymorphisms of IL-1 complex with AS in the Chinese Han population in this study. Chinese Han AS patients and ethnically matched healthy controls were genotyped for five single nucleotide polymorphisms (IL1beta+3953, beta-511, F10.3, RN.4, RN.6/1) and the IL1RN.VNTR of IL-1 gene cluster. Allele, Genotype and haplotype frequencies were compared between cases and controls by SHEsis software. The frequency of allele C of the marker IL1F10.3 was significantly increased in AS patients versus controls [p = 0.001, odds ratio (OR) = 1.54, 95% confidence interval (CI) = 1.19-1.20; p = 0.002, respectively]. Strong linkage disequilibrium was identified between IL1B-511, IL1B+3953 and RN4 in both patients and healthy controls (D' > 0.95). Haplotypes of pairs of these markers (6) were also significantly associated with AS. The strongest associations observed was between allele combination B-511-T/B+3953-C/F10.3-C/RN4-T/RN2VNTR-1/RN6.1-C and AS (p = 3.32 x 10(-5), OR = 4.41, 95% CI=2.1-9.3). Clinical manifestation showed week association between RN2VNTR A2 allele and risk of peripheral arthritis (OR = 0.2, 95% CI = 0.07-0.91). The IL-1 gene cluster is associated with AS in Chinese population. This finding provides strong statistical support for the previously observed relationship and indicates possible association between clinical manifestation and genetic factor.

Abstract:The aim of this study was to investigate the association of single nucleotide polymorphisms (SNPs) in tumor necrosis factor alpha ( TNFα ) promoter with rheumatoid arthritis (RA) in Chinese Han population. Six SNPs at positions −238, −244, −308, −376, −857, −863 within TNFα promoter were genotyped in 367 unrelated RA patients and 271 healthy controls, using direct DNA sequencing method. Allelic, genotypic, and haplotypic associations of these SNPs with RA were examined. The frequencies of TNFα −308A allele and the haplotype GACC (in order of −238, −308, −857, −863) were significantly lower in RA patients when compared with healthy controls ( P = 0.0046, P = 0.0045, respectively) but TNFα −308 polymorphism was not related to the clinical manifestations of RA patients. These results implied that TNFα gene itself or a gene in linkage disequilibrium with it might be associated with RA in Han population of Eastern China.

Abstract:Systemic lupus erythematosus (SLE) is an autoimmune disease with heterogeneous clinical manifestations influenced by genetic and environmental factors. Five novel susceptibility genes (TNIP1, SLC15A4, ETS1, RasGRP3 and IKZF1) for SLE have been identified in a recent genome-wide association study of a Chinese Han population. This study investigated their relationships with disease subphenotypes, including renal nephritis, photosensitivity, antinuclear antibody (ANA), age at diagnosis, malar rash, discoid rash, immunological disorder, oral ulcer, hematological disorder, neurological disorder, serositis, arthritis and vasculitis. Significant associations were found for the single nucleotide polymorphism rs10036748 of TNIP1 with photosensitivity (odds ratio (OR) = 0.87, p = 0.01) and vasculitis (OR = 1.18, p = 0.04); rs10847697 of SLC15A4 with discoid rash (OR = 1.18, p = 0.02); rs6590330 of ETS1 with SLE of age at diagnosis <20 years (OR = 1.24, p = 8.91 × 10-5); rs13385731 of RasGRP3 with malar rash (OR = 1.20, p = 0.01), discoid rash (OR = 0.78, p = 0.02) and ANA (OR = 0.72, p = 0.004); rs4917014 of IKZF1 with renal nephritis (OR = 1.13, p = 0.02) and malar rash (OR = 0.83, p = 0.00038), respectively. [Association analysis of seven single nucleotide polymorphisms identified by genome-wide association study with age-related macular degeneration among ethnic Han Chinese population]

Abstract:Objective: To assess the association of 7 single nucleotide polymorphisms (SNPs) including rs13278062 (TNFRSF10A), rs3750846 (ARMS2-HTRA1), rs429358 (APOE), rs5817082 (CEPT), rs2043085 (LIPC), rs1626340 (TGFBR1), and rs8135665 (SLC16A8) identified through genome-wide association study (GWAS) with age-related macular degeneration (AMD) among ethnic Han Chinese from Sichuan, China. Methods: A cohort of 576 AMD patients and 572 healthy controls were enrolled in a case-control study. The SNPs were genotyped by a Mass array MALDI-TOF System. On the premise that the genotype distribution of each SNP locus in both groups satisfied Hardy-Weinberg equilibrium, the genetic pattern was analyzed and the scores of allele and genotype frequencies ware compared. Results: There was a significant association between TNFRSF10A rs13278062 and AMD under the heterozygous model (P = 0.000, OR = 1.529, 95%CI = 1.196-1.954) and the dominant model (P = 0.002, OR = 1.459, 95%CI = 1.154-1.865), suggesting that subjects carrying rs13278062GT and rs13278062TT + GT are more likely to develop the AMD, whereas no significant difference was observed for rs13278062 under other models. No association was detected with the other six SNPs and AMD under various genetic models. Conclusion: This case-control association study has indicated that TNFRSF10A rs13278062 is associated with AMD under the heterozygous and dominant models, suggesting that the TNFRSF10A variant may be involved in the development of AMD among ethnic Han Chinese population.
Polymorphisms of the TNF Gene and Three Susceptibility Loci Are Associated with Crohn's Disease and Perianal Fistula Crohn's Disease: A Study among the Han Population from South China

Abstract:BACKGROUND Although 90 susceptibility loci of Crohn's disease (CD) have been confirmed in the Asian population, susceptibility genes for perianal fistula of CD (pCD) in this population remain unknown. This study explored susceptibility genes for CD and pCD in the Han population from South China. MATERIAL AND METHODS In total, 490 patients diagnosed with CD between July 2012 and June 2016 at the Sixth Affiliated Hospital of Sun Yat-sen University were included and divided into the CD group (n=240) and the pCD group (n=250). The healthy control group was composed of 260 volunteers. Peripheral blood samples were taken, and single nucleotide polymorphism (SNP) locus sequencing was used to screen for susceptibility loci. SNPs were sequenced using matrix-assisted laser desorption ionization time-of-flight mass spectrometry. RESULTS Nine SNPs in TNFSF1 on chromosome 9 were associated with CD. Among them, the rs6478106 locus is a risk locus for CD. The distribution frequency of the T allele of the rs6478106 SNP was significantly different between cases and controls (32.49% versus 18.27%, P<0.001). Rs72553867, located in the IRGM gene on chromosome 5, rs4409764, located in the NKX2-3 gene on chromosome 10, and rs3731772, located in the AOX1 gene on chromosome 2, were susceptibility factors for pCD. Nine SNPs located in TNFSF15 on chromosome 9 were related to CD in Han individuals from Southern China. CONCLUSIONS The rs6478106 T allele is associated with the risk of CD in the investigated population. SNPs rs72553867 (IRGM gene), rs4409764 (NKX2-3 gene), and rs3731772 (AOX1 gene) increase the risk of pCD.

Abstract:Rheumatoid arthritis (RA) is a complicated rheumatic autoimmune disease. Lectin, galactoside-binding soluble, 2 (LGALS2), LGALS3 and LGALS9, three members of the galectin family, play potential roles in autoimmune diseases, including RA. However, association of genetic polymorphisms of LGALS2, LGALS3 and LGALS9 with RA risk in a Southern Chinese Han population has not been elucidated. A case-control study was conducted herein, including 500 RA patients and 650 healthy individuals of Southern Chinese Han origin. Twelve single nucleotide polymorphisms (SNPs), including rs7291467 for the LGALS2 gene, rs4644, rs4652, rs1009977, rs2274273 and rs17128183 for the LGALS3 gene, and rs4795835, rs3763959, rs4239242, rs3751093, rs732222 and rs4794976 for the LGALS9 gene, were genotyped. Polymorphisms were genotyped using the KASP method. Frequencies of rs1009977 genotype TG and rs3751093 genotype GA of LGALS3 gene were significantly different between RA patients and healthy controls (P=0.049, P=0.033). Allele T and genotypes TT and TT+TG of rs4794976 for LGALS9 gene were significantly correlated with RA risk (P=0.017, P=0.012, P=0.041). Subgroup analysis revealed that rs1009977, rs2274273 and rs17128183 polymorphisms of LGALS3 gene and rs4795835 polymorphism of LGALS9 gene were correlated with several RA clinical manifestations (all P<0.05). In addition, haplotype GCGTT showed an increased risk for RA (OR=1.216, 95% CI: 1.028-1.438, P=0.023), whereas haplotype GCGTG showed a reduced risk for RA susceptibility (OR=0.779, 95% CI: 0.625-0.971, P=0.026). In conclusion, LGALS3 and LGALS9 gene polymorphisms may associate with RA predisposition in a Southern Chinese Han population.

Abstract:Objective This work is aimed to investigate the possible association of dendritic cell immunoreceptor (DCIR) with rheumatoid arthritis (RA) susceptibility in Chinese Han population.Methods A total of 523 patients with RA and 510 healthy controls were genotyped for single-nucleotide polymorphism (SNP) rs2377422 and rs10840759.Association analyses were performed on the whole data set and on RA subsets based on the status of anti-cyclic citrullinated peptide antibody (CCP) in RA patients.Finally,we carried out the association analysis of rs2377422 with DCIR mRNA expression in RA patients.Statistical analysis used in this study included X2 test,Logistic regression,and Mann-Whitney U test.Results DCIR rs2377422 was found significantly associated with RA (allele analysis: OR 1.26; 95％CI 1.06～1.51,P=0.005; genotype analysis CC vs TT+TC: OR 1.34; 95％CI 1.18～2.06,P=0.004).Following stratification for anti-CCP antibody status,association of ra2377422 with anti-CCP-positive RA was observed (allele analysis: OR 1.22,95％CI 0.99～1.48,P=0.055).In contrast,the SNP rs2377422 was found specifically susceptible to anti-CCP-negative RA (allele analysis: OR 1.46; 95％CI 1.10～1.93,P=0.0091; genotype analysis CC vs TT+TC: OR 1.58;95％CI 1.01～2.47,P=0.043),despite loss of power in the analysis.DCIR gene transcription quantification analysis further proved the dominant effect of rs2480256 CC genotype on DCIR mRNA expression levels in RA patients (CC vs TT+TC: 0.429±0.069 vs 0.238±-0.023,U=1861,P=0.0015).Conclusion The study provides evidence for the association between DCIR rs2377422 and RA,particularly with anti-CCP-negative RA in Chinese Han populations.
Association Of Tnf-Alpha Genetic Polymorphisms With Hepatocellular Carcinoma Susceptibility: A Case-Control Study In A Han Chinese Population

Abstract:The single nucleotide polymorphisms (SNPs) within the tumor necrosis factor-alpha (TNF-alpha) gene promoter region have been reported to be associated with susceptibility to various types of cancers. A case-control study (126 hepatocellular carcinoma [HCC] patients and 126 normal controls) was conducted to elucidate their possible association with the risk of hepatitis B virus (HBV)-related HCC in a Han Chinese population. TNF-alpha polymorphisms -1031T/C, -863C/A, -857C/T, -308G/A, and -238G/A were genotyped by polymerase chain reaction (PCR) and direct DNA sequencing. Disease associations were analyzed by the chi-square test or Fisher's exact test. When analyzed by overall groups, no significant differences in genotype and allele distributions were observed between the control and cases. However, stratified analysis according to sex showed that the frequency of the homozygous C allele of the -857 polymorphism was lower in female cases than in female controls (62.9% vs. 88.9%, p = 0.026). In addition, further haplotype analysis revealed that the TCCGA (-1031/-863/-857/-308/-238) was more frequent in controls than cases (p = 0.018; odds ratio = 0.266; 95% confidence interval, 0.083-0.857). These results indicated that the TNF-alpha-857C/T polymorphism may modify HBV-related HCC risk among women, and the haplotype TCCGA (-1031/-863/-857/-308/-238) may account for a decreased susceptibility to HCC development in the Han Chinese population. Additional studies in patients with different ethnic backgrounds are needed to validate these finding and to further explore the genetic pathogenesis of HBV-related HCC.

Abstract:Objective Several studies have reported STAT4 polymorphism is strongly associated with increased susceptibility to rheumatoid arthritis (RA). However, a study from China showed no association between STAT4 and RA susceptibility in a Chinese Han subpopulation. Since the northern Hans are known to be genetically different from the southern Hans, the aim of this study was to investigate the association of STAT4 polymorphism with RA in a large cohort of a northern Chinese Han subpopulation. Methods 640 RA patients and 662 healthy controls were enrolled. DNA samples were genotyped for STAT4 rs7574865 by direct sequencing. The association of single nucleotide polymorphism (SNP) rs7574865 with RA susceptibility was calculated and the relationship between rs7574865 polymorphism and RA subgroups stratified by clinical features was estimated. Results We confirmed a significant association of STAT4 rs7574865 polymorphism with RA susceptibility in northern Chinese Han population. The frequency of the minor T allele in RA was significantly higher than in healthy controls (35.2% vs. 31.1%; P=0.029, OR 1.2 [95% CI 1.02-1.41]). There was also a significant difference in the distribution of the genotypes of SNP rs7574865 between RA patients and healthy controls (P=0.02). Stratification analyses showed no associations between the genetic risk and clinical/serologic features, but a potential high frequency of TT genotype in a rheumatoid factor-negative subgroup, although it did not reach statistical significance (P=0.084, OR 2.01 [95% CI 0.91-4.45]). Conclusion STAT4 rs7574865 is significantly associated with RA susceptibility in northern Chinese Han subpopulations. The genetic differences of Han subpopulations should be considered when genetic susceptibility for diseases is studied.

Abstract:Scleroderma is an autoimmune fibrosing disorder that can be further subclassified as localized scleroderma (LSc) and systemic sclerosis (SSc). LSc is characterized by sclerotic and pigmented skin lesions, while SSc is a more generalized disorder of the connective tissue involving a number of organs. SSc is characterized by the thickening of dermal collagen bundles, fibrosis, and vascular abnormalities in the visceral organs.<a class="ejp-citation-link js-ejp-citation-link">[1]</a> Despite the differences in their morphologic features and clinical presentation, these two diseases do share some characteristics including endothelial cell dysfunction, T helper 2 (Th2) cell dominance during immune activation and excess fibrosis of the skin with similar pathologic findings, leading to the hypothesis that SSc and LSc share an underlying mechanism of pathogensis. Currently, multiple lines of evidence suggest that genetic factors may contribute to SSc susceptibility.<a class="ejp-citation-link js-ejp-citation-link">[2]</a> Human leukocyte antigen (HLA) genes are closely linked to SSc susceptibility, with studies describing some correlation between SSc and HLA-B, HLA-C, HLA-DRA, HLA-DRB1, HLA-DRB5, HLA-DQA1, HLA-DQB1, HLA-DMB, HLA-DOA, HLA-DPA1, HLA-DPB1, and HLA-DPB2.<a class="ejp-citation-link js-ejp-citation-link">[2]</a> A large number of non-HLA genes have also been reported to be associated with the development of SSc, these include connective tissue growth factor gene (TGF), signal transducer and activator of transcription gene (STAT4) amongst others [Supplementary Table 1, <a href="http://links.lww.com/CM9/A294">http://links.lww.com/CM9/A294</a>]. However, only some of these results could be verified in additional studies and similar studies in the Chinese population remain extremely limited. Interestingly, genes like interleukin 1 (IL-1), ras homolog family member B (RHOB), family with sequence similarity 167-member A-BLK proto-oncogene (FAM167A-BLK), STAT4, interferon regulatory factor 5 gene (IRF5), HLA-DPB1, and keratin1 (KRT1) have all been linked to SSc susceptibility in the Chinese population.<a class="ejp-citation-link js-ejp-citation-link">[3]</a> Although it has been reported that LSc tends to exhibit some form of familial inheritance, there have been no clearly identified heritable traits for this disease. Additionally, no study, to our knowledge, has investigated the common genetic factors for LSc and SSc. Thus, it would be of significant interest to examine the susceptibility of LSc and determine whether there are any correlations between the genetic profiles of SSc and LSc. This study aimed to identify the genetic profile of LSc patients and any shared genetic features of SSc and LSc in the Chinese population. Furthermore, this study evaluated the occurance of previously described SSc loci in the Chinese population and compared their frequency with those reported in other ethnic communities.

Association of Tnfaip3 Polymorphism with Rheumatic Heart Disease in Chinese Han Population

Abstract:In a pair-matched case-control study (239 versus 478) conducted in Chinese Han population, we investigated the association between tumor necrosis factor-alpha-induced protein 3 (TNFAIP3) gene, tumor necrosis factor receptor-associated factor 1 (TRAF1) gene, complement component 5 (C5) gene, and rheumatic heart disease (RHD). We observed no association with RHD for the five tagging single nucleotide polymorphisms (tSNP) in the C5 gene, the three tSNPs in the TNFAIP3 gene, or the two tSNPs in the TRAF1 gene. However, we determined that the tSNP, rs582757, located at intron_5 of the TNFAIP3 gene, associated with RHD in Chinese Han population. Both the distribution of genotype and allele frequencies differed significantly between case and control subjects (p = 0.001 and p = 0.0004, respectively). The minor C allele reduced the risk of RHD with a per-allele odds ratio of 0.57 (0.42-0.78) for the additive model in univariate analysis (p = 0.000). Under a dominant model, CC/CT carriers had a 0.54-fold reduced risk of RHD (95% confidence interval 0.38-0.75, p = 0.000) than TT carriers. Therefore, we report a new genetic variant (rs582757) in the TNFAIP3 gene that associated with the prevalence of RHD in Chinese Han population. Further genetic and functional studies are required to identify the etiological variants in linkage disequilibrium with this polymorphism.
