Abstract:Although adipose stem cell-conditioned medium (ASC-CM) has demonstrated the effect of promoting the cutaneous wound healing, the mechanism for this response on the effector cells (e.g., dermal fibroblasts) during the process remains to be determined. In this study, we aim to investigate the types and contents of cytokines in ASC-CM and the effects of some kinds of common cytokines in ASC-CM, such as EGF, PDGF-AA, VEGF, and bFGF, on dermal fibroblasts proliferation and migration in wound healing process. Results showed that these four cytokines had high concentrations in ASC-CM. The migration of skin fibroblasts could be significantly stimulated by VEGF, bFGF, and PDGF-AA, and the proliferation could be significantly stimulated by bFGF and EGF in ASC-CM. Additionally, ASC-CM had more obvious promoting effect on fibroblasts proliferation and migration than single cytokine. These observations suggested that ASC-CM played an important role in the cutaneous injury partly by the synergistic actions of several cytokines in promoting dermal fibroblasts proliferation and migration, and ASC-CM was more adaptive than each single cytokine to be applied in promoting the wound healing.

Effect of the Conditioned Medium of ADSCs on Fibroblasts

Abstract:Objective To isolate and culture adipose-derived stem cells (ADSCs),and study the effect of the conditioned medium of ADSCs (ADSC-CM) on fibroblasts. Methods ADSCs was isolated from human adipose tissue and cultured. Then the conditioned medium of ADSCs was used to culture fibroblasts. Cells counting by the MTT method was to determined the proliferation. Annexin V/PI was used to investigate the apoptosis rate of fibroblasts; the cell migration was measured by in intro wound-healing assay. Results ADSCs was derived by culture. Compared with those in control group,the proliferation and migration in ADSC-CM group were significantly higher than that in control group (P0.05 or P0.01); and the apoptosis rate of fibroblasts in ADSC-CM group was also found to lower than that in control group (P0.05). Conclusion ADSC-CM could obviously promote fibroblasts on proliferation; migration and suppress apoptosis.

Abstract:OBJECTIVE:To review the biochemical characteristics, application progress, and prospects of the adipose-derived stem cells (ADSCs).METHODS:The recent original experimental and clinical literature about ADSCs was extensively reviewed and analyzed.RESULTS:ADSCs can be readily harvested in large numbers from adipose tissue with properties of stable proliferation and potential differentiation in vitro. Significant progress of ADSCs is made in the animal experiment and the clinical application. It has been widely used in the clinical treatment of cardiovascular disease, metabolic disease, encephalopathy, and tissue engineering repair.CONCLUSION:ADSCs have gradually replaced bone marrow mesenchymal stem cells and become the focused hot spot of regenerative medicine and stem cells.
Effects of conditioned medium of human ADSCs on HaCaT cells

Abstract:AIM:To isolate and culture adipose-derived stem cells(ADSCs) and to study the effects of cytokines secreted by ADSCs on HaCaT cells. METHODS: ADSCs were isolated from human adipose tissues and cultured for 3 d. ELISA was used to detect the secretion level of VEGF and HGF. The conditioned medium of ADSCs(ADSC-CM) was then used to culture HaCaT cells. MTT method was used to determine the proliferation and Annexin V/PI was used to examine the apoptosis rate of HaCaT cells. The cell migration was measured by in vitro wound-healing assay.RESULTS: ADSCs were isolated and cultured successfully. The amount of VEGF and HGF was (287±47) ng/L and (577±85) ng/L, respectively. The A490 nm values of HaCaT cells in 30% ADSC-CM group and 50% ADSC-CM group were significantly higher than those in control group(P<0.05 or P<0.01). The apoptosis rate of HaCaT cells in experiment group was obviously lower than that in control group(P<0.05). The migration distance of HaCaT cells in experiment was significantly longer than that in control group at 36 and 48 h(P<0.05 or P<0.01). CONCLUSION: ADSCs can secrete cytokines and ADSC-CM promotes proliferation and migration and suppresses apoptosis of HaCaT cells.

Abstract:Adult stem cells have been extensively investigated for tissue repair therapies. Adipose-derived stem cells (ASCs) were shown to improve wound healing by promoting re-epithelialization and vascularization as well as modulating the inflammatory immune response. In this study, we used ex vivo human skin cultured in a six-well plate with trans-well inserts as a model for superficial wounds. Standardized wounds were created and treated with allogeneic ASCs, ASCs conditioned medium (ASC-CM), or cell culture medium (DMEM) supplemented with fetal calf serum (FCS). Skin viability (XTT test), histology (hematoxylin and eosin, H and E), β-catenin expression as well as inflammatory mediators and growth factors were monitored over 12 days of skin culture. We observed only a moderate time-dependent decrease in skin metabolic activity while skin morphology was preserved, and re-epithelialization occurred at the wound edges. An increase in β-catenin expression was observed in the newly formed epithelia, especially in the samples treated with ASC-CM. In general, increased growth factors and inflammatory mediators, e.g., hepatocytes growth factor (HGF), platelet-derived growth factor subunit AA (PDGF-AA), IL-1α, IL-7, TNF-α, and IL-10, were observed over the incubation time. Interestingly, different expression profiles were observed for the different treatments. Samples treated with ASC-CM significantly increased the levels of inflammatory cytokines and PDGF-AA with respect to control, whereas the treatment with ASCs in DMEM with 10% FCS resulted in significantly increased levels of fibroblast growth factor-basic (FGF-basic) and moderate increases of immunomodulatory cytokines. These results confirm that the wound microenvironment can influence the type of mediators secreted by ASCs and the mode as to how they improve the wound healing process. Comparative investigations with pre-activated ASCs will elucidate further aspects of the wound healing mechanism and improve the protocols of ACS application.

Abstract:Objective To observe the effects of different culture conditions on growth state of human adipose tissue-derived stem cells (hADSCs).Methods The subcutaneous adipose tissue was collected surgically.Primary hADSCs were isolated subsequently by the methods of stirring digestion within 1％ collagenase type Ⅰ.The cells were divided into three groups and cultured in high-glucose DMEM,high-glucose DMEM containing basic fibroblast growth factor (bFGF) and mesenchymal stem cell media (MSCM)respectively.The expression levels of CD31,CD34 and Stro-1 were detected by using immumofluorescence and flow cytometry.The growth state of cells in the three groups was evaluated.Proliferation of cells in each group was evaluated after culture in different culture conditions.Results hADSCs were successfully isolated through stirring digestion within collagenase Ⅰ.The positive expression rate of CD31,CD34 and Stro-I in hADSCs was 0.9％,2.9％ and 56.7％ respectively in DMEM,1.3％,2.5％ and 73.5％ in DMEM containing bFGF,and 0.7％,2.4％ and 84.5％ in MSCM,respectively.The cell doubling time in DMEM,DMEM containing bFGF and MSCM was 93.7 h,64.5 h and 49.1 h respectively.MSCM and high-glucose DMEM containing bFGF could significantly promote the proliferation of cells in each group as compared with high-glucose DMEM (P ＜ 0.05).Conclusion A great number of hADSCs could be obtained in a short time when cultured in MSCM and high-glucose DMEM containing bFGF,which can suffice for the cell source for tissue engineering.

Abstract:Stem cells possess the ability to differentiate into different lineages and the ability to self-renew, thus representing an excellent tool for regenerative medicine. They can be isolated from different tissues, including the adipose tissue. Adipose tissue and human adipose-derived stem cells (hADSCs) are privileged candidates for regenerative medicine procedures or other plastic reconstructive surgeries. The cellular environment is able to influence the fate of stem cells residing in the tissue. In a previous study, we exposed hADSCs to an exhausted medium of a breast cancer cell line (MCF-7) recovered at different days (4, 7, and 10 days). In the same paper, we inferred that the medium was able to influence the behaviour of stem cells. Considering these results, in the present study, we evaluated the expression of the major genes related to adipogenic and osteogenic differentiation. To confirm the gene expression data, oil red and alizarin red colorimetric assays were performed. Lastly, we evaluated the expression of miRNAs influencing the differentiation process and the proliferation rate, maintaining a proliferative state. The data obtained confirmed that cells exposed to the medium maintained a stem and proliferative state that could lead to a risky proliferative phenotype.

Abstract:Objective: To explore the effects of adipose-derived stem cell released exosomes (ADSC-Exos) on the proliferation, migration, and tube-like differentiation of human umbilical vein endothelial cells (HUVECs). Methods: Adipose tissue voluntarily donated by liposuction patients was obtained. The ADSCs were harvested by enzyme digestion and identified by flow cytometry and adipogenic induction. The ADSC-Exos were extracted from the supernatant of the 3rd generation ADSCs and the morphology was observed by transmission electron microscopy. The surface proteins (Alix and CD63) were detected by Western blot. The nanoparticle tracking analyzer NanoSight was used to analyze the size distribution of ADSC-Exos. After co-culture of PKH26 fluorescently labeled ADSC-Exos with HUVECs, confocal microscopy had been used to observe whether ADSC-Exos could absorbed by HUVECs. ADSC-Exos and HUVECs were co-cultured for 1, 2, 3, 4, and 5 days. The effect of ADSC-Exos on the proliferation of HUVECs was detected by cell counting kit 8 (CCK-8) assay. The expression of VEGF protein in the supernatant of HUVECs with or without ADSC-Exos had been detected by ELISA after 12 hours. Transwell migration assay was used to detect the effect of ADSC-Exos on the migration ability of HUVECs. The effect of ADSC-Exos on the tubular structure formation of HUVECs was observed by Matrigel experiments in vitro. The formation of subcutaneous tubular structure in vivo was observed in BALB/c male nude mice via the injection of HUVECs and Matrigel with or without ADSC-Exos. After 2 weeks, the neovascularization in Matrigel was measured and mean blood vessel density (MVD) was calculated. The above experiments were all controlled by the same amount of PBS. Results: After identification, the cultured cells were consistent with the characteristics of ADSCs. ADSC-Exos were circular or elliptical membranous vesicle with uniform morphology under transmission electron microscopy, and expresses the signature proteins Alix and CD63 with particle size ranging from 30 to 200 nm. Confocal microscopy results showed that ADSC-Exos could be absorbed by HUVECs. The CCK-8 analysis showed that the cell proliferation of the experimental group was better than that of the control group at each time point ( P<0.05). The result of Transwell showed that the trans-membrane migration cells in the experimental group were significantly more than that in the control group ( t=9.534, P=0.000). In vitro, Matrigel tube-forming experiment showed that the number of tube-like structures in the experimental group was significantly higher than that of the control group ( t=15.910, P=0.000). In vivo, the MVD of the experimental group was significantly higher than that of the control group ( t=16.710, P=0.000). The ELISA assay showed that the expression of VEGF protein in the supernatant of the experimental group was significantly higher than that of the control group ( t=21.470, P=0.000). Conclusion: ADSC-Exos can promote proliferation, migration, and tube-like structure formation of HUVECs, suggesting that ADSC-Exos can promote angiogenesis in vitro and in vivo.
Biological effects of paracrine from insulin stimulated adipose-derived stem cells(ADSC) on human vascular endothelial cells

Abstract:Objective To study the biological effects of the paracrine from ADSC after being stimulated by insulin on vascular endothelial cells. Methods(1) ADSC was isolated from human adipose tissue and cultured in vitro. The third generation cells were collected and divided into insulin group (1, cultured with serum-free DMEM containing 1 × 10-7 mol/L insulin) and control group (C, cultured with serum-free DMEM) according to the random number table, with 6 slots in each group. Three days later, ADSC culture medium (ADSC-CM) was collected for determination of levels of vascular endothelial growth factor (VEGF) and hepatocyte growth fator (HGF) by ELISA. (2) Human umbilical vein endothelial cells (HUVEC) were cultured to the third generation, and they were cultured with special nutrient solution and divided into ADSC-CM with insulin stimulation group (AI) , ADSC-CM without insulin stimulation group (AC), insulin group(I, with same concentration as above), blank control group (BC) according to the random number table. Three days later, proliferation of HUVEC was determined with MTT method(with expression of absorbance value). Another two samples of HUVEC were respectively divided into 4 groups as above for determination of apoptosis rate with Annexin V/FITC double-staining 12 hours after culture, and H UVEC migration with scratch adhesion test at post scratch hour (PSH) 12, 24, 36, 48. Data were processed with t test.Results(1) Compared with those in C group [(287 ± 47) , (577 ± 84) pg/mL, respectively] , the secretion levels of VEGF and HGF in I group [(643 ±64) , (930 ±68) pg/mL, respectively] were significantly increased(with t value respectively 18. 869, 18. 475, P values all below 0.05). (2) The absorbance value of HUVEC in AI and AC groups was 0. 847 ± 0. 042, 0. 798 ± 0.022, respectively, which were higher than that in I and BC groups [0. 665 ± 0. 028(with t value respectively 4. 579, 3. 732) , 0. 674 ± 0.031(with t value respectively 3. 761, 4. 073) , P values all below 0. 01] , and that in AI group was higher than that in AC group(t =2.576, P ＜0.05). The apoptosis rates of HUVEC in AI and AC groups [(5.8±1.9)%,(9.0 ± 2.0)%, respectively] were obviously lower as compared with that in I and BC groups [(30.4 ±6. 0) %(with t value respectively 12. 891, 10.417),(31.4 ± 7.4) %(with t value respectively 11. 474,9. 783), P values all below 0. 05], and that in AC group was higher than that in AI group(t = 8. 548, P ＜0. 05). The distance of migration of HUVEC in AI and AC groups were greater than that in I and BC groups at PSH 36, 48, and that in AI group was greater as compared with that in AC group(with t value respectively 4.076, 4. 573, P values all below 0. 05). Conclusions Paracrine from ADSC after being stimulated by insulin can promote proliferation and migration of HUVEC, and suppress its apoptosis, and it is beneficial for tissue vascularization.

Abstract:BACKGROUND: Adipose-derived stem cells (ASCs) are mesenchymal stem cells that have recently been applied to tissue repair and regeneration. Keratinocytes and dermal fibroblasts play key roles in cutaneous wound healing.OBJECTIVE: We investigated the paracrine effects of ASCs on HaCaT cells (i.e., immortalized human keratinocytes) and human dermal fibroblasts to explore the mechanism of the effects of ASCs on cutaneous wound healing.METHODS: HaCaT cells and primary cultured human dermal fibroblasts were treated with 50% conditioned medium of ASCs (ASC-CM). Viability, in vitro wound healing, and fibroblast-populated collagen lattice contraction assays were conducted, and reverse transcription-polymerase chain reaction (RT-PCR) for the type I procollagen α1 chain gene was performed.RESULTS: The proliferation of HaCaT cells and fibroblasts was increased by ASC-CM in the viability assay. ASC-CM promoted in vitro wound healing of HaCaT cells and increased the contraction of the fibroblast-populated collagen lattice. RT-PCR showed that the transcription of the type I procollagen α1 chain gene in fibroblasts was upregulated by ASC-CM.CONCLUSION: The stimulatory effect of ASC on cutaneous wound healing may be partially mediated by paracrine effects of ASCs on other skin cells. Application of ASCs or ASC-derived molecules could be an innovative therapeutic approach in the treatment of chronic wounds and other conditions.

Abstract:BACKGROUND AND OBJECTIVES: The application of adipose derived stem cells (ADSCs) in skin repair has attracted much attention nowadays. Epidermal growth factor (EGF) participates in the progress of skin proliferation, differentiation and so forth. We aimed to explore the role of EGF in the proliferation, invasion, migration and transdifferentiation into epidermal cell phenotypes of ADSCs.METHODS AND RESULTS: ADSCs were extracted from adipose tissues from patient. Immunophenotyping was determined by flow cytometry. Overexpressed EGF or siEGF was transfected by lentiviruses. EGF was determined by enzyme linked immunosorbent assay (ELISA) or western blot. ADSCs and HaCaT cells were co-cultured by Transwell chambers. Conditioned medium (CM) was obtained from cultured HaCaT cells and used for the culturing of ADSCs. Cell viability was tested by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Invasion rate was measured by Transwell invasion assay and migration rate by wound healing test. mRNA and protein levels were measured by qPCR and western blot respectively. The extracted cells from adipose tissues were identified as ADSCs by morphology and immunophenotyping. The expression of EGF was up or down regulated constantly in HaCaT cell line after transfection. EGF overexpression upregulated the proliferation, migration and invasion rates of ADSCs, and EGF expression regulated the expression of cytokeratin-19 (CK19) and integrin-<i>β</i> as well.CONCLUSIONS: EGF could be served as a stimulus to promote the proliferation, migration, and invasion as well as the transdifferentiation into epidermal stem cell immunophenotyping of ADSCs. The results showed that EGF had a promising effect on the repair of skin wound.

[VEGF Overespression Promotes the Proliferation and Differentiation of Human Adipose-Derived Stem Cells].

Abstract:Objective To investigate the effect of vascular endothelial growth factor (VEGF) overexpression on the phenotype, proliferation and multilineage differentiation ability of human adipose-derived stem cells (hADSCs). Methods Passage-2 hADSCs were transfected with pIRES2-EGFP-VEGF plasmid using DOTAP liposomal technique, whereas hADSCs in control group were transfected in absence of plasmid. The success of transfection was confirmed by immunofluorescent cytochemistry. Phenotype differences between the two groups were detected using flow cytometry. Proliferation ability was assessed using MTT assay. Adipogenic and osteogenic differentiation were induced and confirmed using Oil Red O staining and alizarin red staining. Results The transfected hADSCs expressed EGFP and VEGF, whereas no EGFP was observed in the untransfected hADSCs. Both groups of hADSCs showed high expressions of CD29, CD44, CD90, and low expressions of CD31 and CD45. Compared with the untransfected hADSCs, the transfected hADSCs exhibited significantly increased proliferation ability as well as the number of lipid droplets, calcium nodules, and the stained area. Conclusion VEGF overexpression promotes proliferation and multilineage differentiation potential of hADSCs without evident phenotype changes.
Influence of micorocarrier suspension culture system on the proliferation and differentiation of human adipose derived stem cells

Abstract:Aim: To observe the interventional effects of micorocarriers suspension culture system on the adult adipose derived stem cells and the ability of differentiation of the cultured cells. Methods: The experiment was carried out in the Tissue Engineering Research Center, Institute of Basic Medical Science, Academy of Military Medical Sciences from September to December 2004. Cytodex3 microcarriers served as suspension culture system provided condition of wall-adhesion for the wall-adhesion dependent adipose derived stem cells, the concentration was 5 g/L, and the cells were cultured statically in the 12-well culture plate. The cells were inoculated with a density of 1 × 108 L-1 in both cultural systems, and the cell proliferation was observed respectively; The cellular phenotype and the ability of differentiation of the cells cultured with the suspension culture system were detected. Results: Nine days after microcarrier suspension culture system, the density of viable cells was the maximal (1.60 × 109 L-1), which was observed on the 7th day of static culture (3.38 × 108 L-1). Under the condition of microcarrier suspension culture, the adipose derived stem cells grew more productively, and the production of cell was higher, which were superior to those under static culture. Eleven days after microcarrier suspension culture, adipose derived stem cells still maintains the characteristics of multi-directional differentiation. Conclusion: The application of suspension culture system and microcarrier culture technique is good for the rapid growth of large scale; After cell amplification, the cellular phenotype and ability of differentiation are still maintained. The obtained adipose derived stem cells can be used as the seeding cells in the experimental study of tissue engineering and in clinic.
[Effects of mouse adipose-derived stem cell conditioned medium on the apoptosis of keratinocytes induced by thermal injury in vitro]

Abstract:Objective: To investigate the effects of mouse adipose-derived stem cell conditioned medium (ADSC-CM) on apoptosis of keratinocytes (human epithelial cell line HaCaT) induced by thermal injury in vitro. Methods: (1) Adipose-derived stem cells (ADSCs) from inguinal adipose tissue of 5 healthy BALB/c mice were isolated, cultured, and purified by collagenase digestion in vitro. The 3rd passage of cells were collected for morphologic observation, detection of expressions of surface markers CD31, CD34, CD45, CD90, and CD105 with flow cytometer, and identification of adipogenic and osteogenic differentiation. (2) HaCaT cells were incubated in water at 51.5 °C for 35 seconds to reproduce thermal injury model, and then the apoptosis rate was detected immediately after injury by flow cytometer. (3) Thermally injured HaCaT cells were divided into routine culture group (RC, cultured with DMEM containing 10% FBS), serum-free group (cultured with serum-free DMEM), 50%ADSC-CM group (cultured with DMEM containing 50%ADSC-CM), and 100%ADSC-CM group (cultured with 100%ADSC-CM) according to the random number table. After 24 hours, apoptosis of HaCaT cells was observed by acridine orange-ethidium bromide (AO-EB) staining; apoptotic rate was determined by flow cytometer; the mRNA and protein levels of Bcl-2 and caspase-3 were respectively determined by real-time fluorescent quantitative RT-PCR technique and Western blotting (protein level was denoted as gray value); the cell cycles were determined by flow cytometer. All above experiments were repeated for 3 times. Data were processed with one-way analysis of variance and LSD- t test. Results: (1) The 3rd passage of cells proliferated well showing fusiform shape similar to fibroblasts. The positive expression rates of CD31, CD34, and CD45 were less than 10.0%, while those of CD90 and CD105 were above 90.0%. The cells could differentiate into adipocytes and osteoblasts. They were identified as ADSCs. (2) Immediately after injury, apoptotic rate of HaCaT cell was (9.8 ± 0.4)%. (3) The number of apoptotic cells was significantly higher in serum-free group than in the other three groups with AO-EB staining. The apoptotic rate of serum-free group [(8.1 ± 1.2)%] was significantly higher than that of 50%ADSC-CM group [(6.0 ± 0.8)%], group RC [(4.6 ± 0.8)%], or 100%ADSC-CM group [(3.1 ± 0.4)%], with t values respectively 3.02, 4.96, 6.60, P values below 0.01. There was no statistically significant difference in apoptotic rate between group RC and 100% ADSC-CM group (t = 1.50, P > 0.05), while statistically significant difference was found between 100% ADSC-CM group and 50%ADSC-CM group (t = 10.21, P < 0.01). (4) The mRNA level of Bcl-2 of serum-free group (0.34 ± 0.08) was significantly lower than that of group RC, 50%ADSC-CM group, and 100%ADSC-CM group (0.98 ± 0.04, 0.77 ± 0.05, 1.06 ± 0.04, with t values respectively 12.87, 8.07, 14.11, P values below 0.01). Compared with that of 100%ADSC-CM group, the mRNA level of Bcl-2 of group RC was slightly decreased (t = 0.08, P > 0.05) and that of 50%ADSC-CM group was significantly decreased (t = 8.08, P < 0.01). (5) The mRNA level of caspase-3 of serum-free group (1.15 ± 0.05) was obviously higher than that of 50%ADSC-CM group (0.72 ± 0.11), group RC (0.41 ± 0.03), or 100%ADSC-CM group (0.38 ± 0.11), with t values respectively 6.93, 13.97, 22.79, P values below 0.01. Compared with 100%ADSC-CM group, the mRNA level of caspase-3 was slightly increased in group RC (t = 0.05, P > 0.05) and significantly increased in 50%ADSC-CM group (t = 4.77, P < 0.01). (6) The protein level of Bcl-2 was significantly lower in serum-free group (0.93 ± 0.04) than in group RC, 50%ADSC-CM group, and 100%ADSC-CM group (1.74 ± 0.06, 1.32 ± 0.05, 1.90 ± 0.04, with t values respectively 20.45, 11.15, 31.38, P values below 0.01). Compared with that of 100%ADSC-CM group, the protein level of Bcl-2 of group RC was slightly decreased (t = 1.33, P > 0.05), but that of 50%ADSC-CM group was obviously decreased (t = 17.30, P < 0.01). (7) The protein level of caspase-3 was obviously higher in serum-free group (0.63 ± 0.08) than in 50%ADSC-CM group, group RC, and 100%ADSC-CM group (0.46 ± 0.03, 0.29 ± 0.08, 0.21 ± 0.03, with t values respectively 3.28, 5.05, 8.46, P values below 0.01). Compared with that of 100%ADSC-CM group, the protein level of caspase-3 of group RC was slightly increased (t = 0.08, P > 0.05), but that of 50%ADSC-CM group was significantly increased (t = 3.52, P < 0.05). (8) Compared with that of serum-free group, the percentage of cells in G2/M phase of each of the other 3 groups was significantly decreased (with t values respectively 6.88, 4.08, 7.28, P < 0.05 or P < 0.01). Compared with that in serum-free group, the percentage of cells in S phase was significantly increased in group RC and 100% ADSC-CM group (with t values respectively 2.67 and 2.40, P values below 0.05). There was no statistically significant difference in the percentage of cells in G0/G1 phase among all groups (F = 0.70, P > 0.05). Conclusions: 100% xenogeneic ADSC-CM can suppress apoptosis of HaCaT cells induced by thermal injury through regulating the expression of Bcl-2 and caspase-3, and accelerate cell cycle progression by ameliorating the retardation of cell growth in G2/M phase, and all these effects may give rise to some potential in the treatment of burn wounds at early stage.

Abstract:To isolate and culture adipose-derived stem cells (ADSCs), and to study the effects of the conditioned medium of ADSCs (ADSC-CM) treated with insulin on HaCaT cells.ADSCs were isolated from adipose tissue donated by the patient receiving abdominal surgery and were cultured. The concentration of ADSCs at passage 3 was adjusted to 5 x 10(4) cells/mL. The cells were divided into 2 groups: group A in which the cells were incubated in 1 x 10(-7) mol/L insulin for 3 days, and group B in which the cells were not treated with insulin. ADSC-CM in each group was collected 3 days after culture, then levels of VEGF and hepatocyte growth factor (HGF). HaCaT cells were cultured and the cells at passage 4 were divided into 4 groups: group A1, 0.5 mL 2% FBS and 0.5 mL ADSC-CM from group A; group B1, 0.5 mL 2% FBS and 0.5 mL ADSC-CM from group B; group C1, 1 mL 2% FBS of 1 x 10(-7) mol/L insulin; group D1, 1 mL 2% FBS. Proliferation of HaCaT cells was detected by MTT method 3 days after culture, apoptosis rate of HaCaT cells was measured by Annexin V-FITC double staining 12 hours after culture, and the migration ability was measured by in vitro wound-healing assay 0, 12, 24, 36 and 48 hours after culture.The level of VEGF in groups A and B was (643.28 +/- 63.57) and (286.52 +/- 46.68) pg/mL, respectively, and the level of HGF in groups A and B was (929.95 +/- 67.52) and (576.61 +/- 84.29) pg/mL, respectively, suggesting differences were significant between two groups (P < 0.05). Cell proliferation detection showed the absorbance value of HaCaT cells in group A1, B1, C1 and D1 was 0.881 +/- 0.039, 0.804 +/- 0.041, 0.663 +/- 0.027 and 0.652 +/- 0.042, respectively, suggesting there was significant difference between groups A1 and B1 and groups C1 and D1 (P < 0.01), group A1 was significantly higher than group B1 (P < 0.05). The apoptosis rate of HaCaT cells in groups A1, B1, C1 and D1 was 5.23% +/- 1.98%, 8.82% +/- 2.59%, 31.70% +/- 8.85% and 29.60% +/- 8.41%, respectively, indicating there was significant difference between groups A1 and B1 and groups C1 and D1 (P < 0.05), group B1 was significantly higher than group A1 (P < 0.05). The migration distance of HaCaT cells in groups A1, B1, C1 and D1 at 36 hours was (0.184 6 +/- 0.019 2), (0.159 8 +/- 0.029 4), (0.059 2 +/- 0.017 6) and (0.058 2 +/- 0.012 3) mm, respectively, whereas at 48 hours, it was (0.231 8 +/- 0.1740), (0.205 1 +/- 0.012 1), (0.079 2 +/- 0.008 1) and (0.078 4 +/- 0.011 7) mm, respectively, suggesting there were significant differences between groups A1 and B1 and groups C1 and D1 at 36 and 48 hours (P < 0.01), group A1 was significantly higher than group B1 (P < 0.05) at 36 and 48 hours, no significant difference was evident at other time points (P > 0.05).ADSCs treated with insulin can significantly promote the proliferation and the migration of HaCaT cells and inhibit their apoptosis.

Abstract:Adipose-derived stem cells (ADSCs) are a subset of mesenchymal stem cells (MSCs), which have promised a vast therapeutic potential in tissue regeneration. Recent studies have demonstrated that combining stem cells with mechanical stretch may strengthen the efficacy of regenerative therapies. However, the exact influences of mechanical stretch on MSCs still remain inconclusive. In this study, human ADSCs (hADSCs) were applied cyclic stretch stimulation under an in vitro stretching model for designated duration. We found that mechanical stretch significantly promoted the proliferation, adhesion and migration of hADSCs, suppressing cellular apoptosis and increasing the production of pro-healing cytokines. For differentiation of hADSCs, mechanical stretch inhibited adipogenesis, but enhanced osteogenesis. Long-term stretch could promote ageing of hADSCs, but did not alter the cell size and typical immunophenotypic characteristics. Furthermore, we revealed that PI3K/AKT and MAPK pathways might participate in the effects of mechanical stretch on the biological characteristics of hADSCs. Taken together, mechanical stretch is an effective strategy for enhancing stem cell behaviour and regulating stem cell fate. The synergy between hADSCs and mechanical stretch would most likely facilitate tissue regeneration and promote the development of stem cell therapy.
Immune regulatory effect of human adipose-derived mesenchymal stem cells

Abstract:BACKGROUND: We know little about the immune regulatory effect of adipose-derived mesenchymal stem cel s. OBJECTIVE: To investigate the influence of adult adipose-derived mesenchymal stem cel s on lymphocyte proliferation, cel s subsets and the level of cel factors in vitro, in order to understand the immune regulatory effect. METHODS: Adipose-derived mesenchymal stem cel s were isolated from adult adipose tissue. The phenotype was identified with the flow cytometry. Adipose-derived mesenchymal stem cel s were induced into osteoblasts and myocardial cel s, respectively, and identified with the immunocytochemistry staining. The human peripheral blood lymphocytes were separated and cultured with adipose-derived mesenchymal stem cel s according to different concentrations (1:1, 1:10, 1:100), and added the plant haemagglutinin for simulation. The control group was set up, and after 3 days, the culture fluid was col ected. RESULTS AND CONCLUSION: The absorbance value of adipose-derived mesenchymal stem cel s in the treatment group after co-cultured with lymphocytes with three different concentrations was significantly lower than that in the control group, and 1:1 concentration had the lowest absorbance value (P < 0.05); the proportion of CD4+CD25+ Tregs in the treatment groups was significantly higher than that in the control group, and highest in the 1:1 concentration group. The cel factors of interleukin-2, interleukin-4 and γ-interferon were obviously reduced in the treatment groups, while the interleukin-10 level was increased, and showed a certain concentration-dependent manner (P < 0.05). Adipose-derived mesenchymal stem cel s can significantly suppress the proliferation of lymphocyte in vitro. The inhibition effect may relate with the increasing of CD4+CD25+ Tregs subsets, and relate with the changing of cytokine secretion of lymphocyte.

Abstract:Objective To investigate the possibility of adipose derived stem cells ( ADSCs) for wound healing by detecting cellular phenotype conversion of ADSCs into endothelial cells (ECs). Methods ADSCs were isolated and cultured from adipose tissue derived from SD rats (n=8), and maintained in Dulbecco's modified Eagle's medium (DMEM) containing 10% fetal bovine serum (FBS) in vitro. The marker antigen of P3 ADSCs was detected by analysis CD49d and CD106 antigens expression using flow cytometry, and the multipotential differentiation of P3 ADSCs were identified by specific medium inducing to differentiate into osteoblasts and adipocytes. And then, the ADSCs were cultured and induced for 3 days by condition culture medium (containing 30% superior of homogenating rat blood vessels in 10%FBS DMEM) as experimental group, and were cultured by 10% FBS DMEM as control group, and the expression of CD34 and von Willebrand factor (vWF) in ADSCs were analyzed by flow cytometry. Results Flow cytometry analysis showed that the expression of CD49d and CD106 in ADSCs were positive(98.32 ±0.37)% and negative(1.67 ±0.61)% , respectively. The multipotential differentiation experiment demonstrated that the cultured P3 ADSCs can be induced to differentiate into osteoblasts and adipocytes in vitro. The positive rate of CD34 and vWF were (77.14 ± 0.76) % and (75.46 ± 0.37) % in condition medium group, higher than (1.38 ±0.31)% and (1.70±0.23)% in 10% FBS DMEM control group, respectively (P<0.01). Conclusion The ADSCs can be induced to differentiated into ECs, suggesting that ADSCs have potential to take part in wound repair and angiogenesis.
