EFFECTS OF HUMAN ADIPOSE-DERIVED STEM CELLS ON SURVIVAL OF SUBCUTANEOUS VASCULAR NET SKIN GRAFT IN RATS

Abstract:Objective To study the influence of human adipose-derived stem cells(ADSCs) on survival of subcutaneous vascular net skin graft in rats and its mechanism.Methods ADSCs were cultured in vitro,assayed by flow cytometry and labeled with EdU(5-Ethynyl-2'-deoxyuridine).Skin graft areas were designed on the middle back of the rats.The rats in experimental group(n=10) were injected with labeled ADSCs PBS,and those in the control group(n=10) only PBS.The rats were killed seven days after operation.Survival rate of the skin grafts was compared between the two groups.The skin grafts were stained with HE for investigating microvessel count under a light microscope;the expressions of CD31 and vascular endothelial growth factors(VEGF) were detected by using immunohistochemical staining,and the distribution and differentiation of ADSCs in transplanted flaps were labeled with EdU and detected.Results The survival rate of transplanted flaps,the number of microvessel in a unit area,and the expression of VEGF in the experimental group were higher than the control,being significant of the differences between the two groups(t=3.870-14.729,P<0.05).EdU-positive cells differentiated into part of vascular endothelial cells in experimental group while no EdU-positive cells were detected in the control.Conclusion Locally transplanted ADSCs can be differentiated into vascular endothelial cells,promote secretion of VEGF,and motivate the survival of the grafts.
